Ornithine decarboxylase (ODC) forms a stable complex with its antizyme (Az), a non-competitive protein inhibitor of ODC. The complex formation of ODC with Az occurs very rapidly and is dissociated by high salt concentrations e.g., 10% ammonium sulfate. When ODC and Az were mixed in the presence of increasing concentrations of Mg2+, a relief of ODC inhibition by Az was obtained. Complete relief of inhibition occurred at 2.0 mM of MgCl2. Other bivalent cations Ca2+, Ba2+, Co2+, Mn2+, Zn2+ as well as the monocations Na+ and K+ caused similar effect. The polyamines putrescine, spermidine and spermine also caused relief of the in vitro inhibition of ODC by Az. Therefore, the in vivo inactivation of ODC by forming the ODC-Az complex is dependent on the intracellular amounts of salt and polyamines.  相似文献

Complex formation with alkali and alkaline earth metal ions of cyclic octapeptides, cyclo(Phe-Pro)4, cyclo(Leu-Pro)4, and cyclo[Lys(Z)-Pro]4 was investigated in relation to conformation. In an alcohol solution, cyclo(Phe-Pro)4 did not form complexes. However, cyclo(Leu-Pro)4 and cyclo[Lys(Z)-Pro]4 formed complexes selectively with Ba2+ and Ca2+ ions. Changing the solvent from alcohol to acetonitrile, the complexation behavior was very different. In acetonitrile, cyclo(Phe-Pro)4 was found to form a complex with Ba2+, and CD spectra of cyclo(Leu-Pro)4 and cyclo[Lys(Z)-Pro]4 changed sharply on complexation with K+. Rate constants of the complex formation between the cyclic octapeptides and metal salts were in the range of 0.7–12 L mol?1 min?1 in an alcohol solution. One of the two types of complex formation in acetonitrile was much faster than that in an alcohol solution.  相似文献

Conductance and equilibrium dialysis studies are reported for the aqueous systems (native calf thymus) DNA-CaCl2 and DNA-MgCl2 at various pH values and ionic strengths at 25 °C. Discontinuities occur in the conductance curves at mole ratios of Ca2+ and Mg2+ to nucleate phosphorus of 0.125, 0.30, and 0.50. The dialysis results show the formation of complexes of stoichiometry 0.50 and 1.00 mol Ca2+ or Mg2+/mol nucleate phosphorus (2:1 and 1:1 complexes), the latter only in neutral or alkaline solutions, in agreement with the conductance discontinuity at 0.50. The other discontinuities may be due to preferential binding in the formation of the 2:1 complex. Binding constants for the 2:1 complexes are evaluated. Absorption-temperature profiles have been determined for “native” and dialysed DNA in the presence of NaCl, CaCl2, and MgCl2. For dialysed DNA at 26 ° C and 260 nm the decrease of absorbance with increased salt concentration was halted for MgCl2 and CaCl2 at a concentration corresponding to the formation of the 2:1 complex. The absorbance of “native” DNA did not decrease. Tm and the reciprocal of the hypochromic rise (1h) increased linearly with log (salt concn). Values of Tm were the same at 230, 260, and 280 nm, but h was greater at 230 and 280 than at 260 nm, which may be due to the existence of alternating blocks of (A + T) and (G ? C) pairs. Cortical vesicles (CV) possess components critical to the mechanism of exocytosis. The homotypic fusion of CV centrifuged or settled into contact has a sigmoidal Ca2+ activity curve comparable to exocytosis (CV–PM fusion). Here we show that Sr2+ and Ba2+ also trigger CV–CV fusion, and agents affecting different steps of exocytotic fusion block Ca2+, Sr2+, and Ba2+-triggered CV–CV fusion. The maximal number of active fusion complexes per vesicle, <n\>Max, was quantified by NEM inhibition of fusion, showing that CV–CV fusion satisfies many criteria of a mathematical analysis developed for exocytosis. Both <n\>Max and the Ca2+ sensitivity of fusion complex activation were comparable to that determined for CV–PM fusion. Using Ca2+-induced SNARE complex disruption, we have analyzed the relationship between membrane fusion (CV–CV and CV–PM) and the SNARE complex. Fusion and complex disruption have different sensitivities to Ca2+, Sr2+, and Ba2+, the complex remains Ca2+- sensitive on fusion-incompetent CV, and disruption does not correlate with the quantified activation of fusion complexes. Under conditions which disrupt the SNARE complex, CV on the PM remain docked and fusion competent, and isolated CV still dock and fuse, but with a markedly reduced Ca2+ sensitivity. Thus, in this system, neither the formation, presence, nor disruption of the SNARE complex is essential to the Ca2+-triggered fusion of exocytotic membranes. Therefore the SNARE complex alone cannot be the universal minimal fusion machine for intracellular fusion. We suggest that this complex modulates the Ca2+ sensitivity of fusion.  相似文献

Abstract— The formation of a complex between myelin basic protein and S-100 protein was detected from the change in migration of S-100 protein on immunoelectrophoresis. A degree of specificity for the interaction was shown by two observations: (1) two other pure acidic proteins. III-III-2 and bovine serum albumin, did not show it and (2) complex formation was dependent on specific ions, either Ca2+ (10 mM) or Mn2+ (1 mM). Mg2+, Ba2+, and Li+ had no effect. Non-specific interactions between S-100 protein and other basic molecules (histones. polylysine) are not dependent on specific ions such as Ca2+ and Mn2+. The complex was stable at physiological salt concentrations and contained 3 mol of basic protein per mol of S-100 protein. Complex formation was also detected from the alteration of migration rate of S-100 protein in polyacrylamide gels. Serological activity (complement-fixation) of S-100 protein with anti-S-100 serum was reduced in the complex by 30%.  相似文献

The fungus Geotrichum candidum was selected from isolates of oil-mill waste as a potent lipase producer. Factors affecting lipase production by the fungus G. candidum in yeast-extract-peptone medium have been optimized by using a Box–Behnken design with seven variables to identify the significant correlation between effects of these variables in the production of the enzyme lipase. The experimental values were found to be in accordance with the predicted values, the correlation coefficient is 0.9957. It was observed that the variables days (6), pH (7.0), temperature (30 °C), carbon (1.25%), nitrogen (2.0%), Tween (1.0%) and salt concentrations (0.5 mM) were the optimum conditions for maximum lipase production (87.7 LU/ml). The enzyme was purified to homogeneity with an apparent molecular mass of 32 kDa by SDS-PAGE. The optimum pH at 40 °C was 7.0 and the optimum temperature at pH 7.0 was 40 °C. The enzyme was stable within a pH range of 6.5 to 8.5 at 30 °C for 24 h. The enzyme activity was strongly inhibited by AgNO3, NiCl2, HgCl2, and EDTA. However, the presence of Ca2+ and Ba2+ ions enhanced the activity of the enzyme.  相似文献

The salen‐type ligand prepared with (R,R) diphenylethan‐1,2‐diamine and salicylaldehyde provides stable and inert complexes KLnL2 upon simple reaction with lanthanide halides or pseudohalides LnX3 (Ln = Tb3+‐Lu3+; X = Cl? or TfO?) of its potassium salt. All the complexes were completely characterized through nuclear magnetic resonance (NMR), electronic circular dichroism (ECD) in the UV and some (Er3+, Tm3+, Yb3+) also with Near‐IR ECD (NIR‐ECD) and luminescence (Tb3+, Tm3+). Careful analysis of the NMR shifts demonstrated that the complexes are isostructural in solution and afforded an accurate geometry. This was further confirmed by means of Density Functional Theory (DFT) optimization of the Lu3+ complex, and by comparing the ligand‐centered experimental and time‐dependent TD‐DFT computed UV‐ECD spectra. As final validation, we used the NIR‐ECD spectrum of the Yb3+ derivative calculated by means of Richardson's equations. In the research described here we prepared a novel, modified polystyrene (PS) with iminoether as the complexing agent for Ba2+. Most heavy metals cause environmental, atmospheric pollution.[1–2] They cause consequence for humans health and aquatic life due to their toxicity. They become strongly toxic by mixing with different environmental elements and their removal from contaminated water is very important. The structure of all modified polystyrene such as nitrated polystyrene (PS−NO2), aminated polystyrene (PS−NH2), aminated polystyrene with imidate group (PS−NH−Im) and the complex with barium metal (PS−NH−Im/Ba2+) were analyzed by Fourier transform infrared spectroscopy (FT-IR), and the formation of N-2-Benzimidazolyl iminoether grafted PS was proved. The thermal stability and structure of the polystyrene and modified polystyrene were studied by differential thermal analysis (DTA) and X-ray diffractometry (XRD), respectively. The elemental analysis was used for the determination of the chemical composition of the modified PS. The grafted polystyrene was used in order to adsorb barium from wastewater with an acceptable cost before the wastewater distribution in the environment. The impedance analysis of the polystyrene complex PS−NH−Im/Ba2+ indicated an activated thermal conduction mechanism.The conductivity analysis of the complex with barium metal PS−NH−Im/Ba2+ was studied; the activation energy was deduced from an Arrhenius plot and corresponded to, , suggesting PS−NH−Im/Ba2+ was a proton-type of semiconductor.  相似文献

The conductance of black lipid membranes in the presence of 2,4,6-trinitrophenol (or 2,4-dinitrophenol) is considerably enhanced, if the cation carriers valinomycin, enniatin B or nonactin are added. The effect is, however, largely independent of the cation concentration and is identical for the cations Li+, Na+ and Ba2+. This finding, as well as the sign and magnitude of the diffusion potential in the presence of a gradient of picrate are consistent with the assumption that the transport of picrate anions is facilitated by the above-mentioned macrocyclic compounds, but that cations are not directly involved. A model is suggested which, based on the generation of mobile defect structures by the incorporation of large molecules, allows one to explain facilitated transport without the assumption of stable chemical bonds between a carrier and its transported substrate.If K+ is present in the aqueous phase, the conductance is largely determined by the permeation of the cation complexes of valinomycin and nonactin. The conductance is, however, increased by adsorption of picrate anions to the membrane surface. The negative surface potential generated by the adsorption layer seems to be responsible for the saturation of the conductance at high picrate concentrations in the absence of valinomycin and nonactin.  相似文献

The effect of membrane potential on the activity of the ATP-dependent Ca2+ pump of isolated canine ventricular sarcolemmal vesicles were investigated. The membrane potential was controlled by the intravesicular and extravesicular concentration of K+, and the initial rates of Ca2+ uptake both in the presence and the absence of valinomycin were determined. The rate of Ca2+ uptake was stimulated by a inside-negative potential induced in the presence of valinomycin. The valinomycin-dependent stimulation was enhanced by the addition of K+ channel blocker, tetraethylammonium ion or Ba2+. The electrogenicity of cardiac sarcolemmal ATP-dependent Ca2+ pump is suggested from the increase of Ca2+ uptake by negative potential induced by valinomycin.  相似文献

GeCl4 and meso-tetraarylporphyrins (H2TAPP) react in chloroform solvent for formation sandwich intermediate sitting-atop (i-SAT) complexes, [GeCl4(H2TAP)2]. The various spectral data (1H NMR, 13C NMR, UV-Vis, FT-IR and elemental analysis) were used for characterization of the i-SAT complexes. In the sandwich complexes, the pyrrole rings of two porphyrin macrocycles are tilted up and down and act as electron donors by lone pairs of pyrrolic nitrogens to germanium center of GeCl4. The spectral results of 1H NMR and FT-IR showed that in the i-SAT complexes, the hydrogen atoms of two pyrrolic nitrogens remained still on the porphyrin macrocycles.  相似文献

 The acidity constants of methyl phosphoric acid, CH3OPO(OH)2, and orthophosphoric acid, HOPO(OH)2, and the stability constants of the 1 : 1 complexes formed between Mg2+, Ca2+, Sr2+, Ba2+, Mn2+, Co2+, Ni2+, Cu2+, Zn2+, or Cd2+ and methyl phosphate, CH3OPO3 2–, or hydrogen phosphate, HOPO3 2–, were determined by potentiometric pH titration in aqueous solution (25  °C;I = 0.1 M, NaNO3). On the basis of previously established log K versus pK a straight-line plots for the complexes of simple phosphate monoesters and phosphonate derivatives, R-PO3 2–, where R is a noncoordinating residue, it is shown that the stability of the M(CH3OPO3) complexes is solely determined (as one might expect) by the basicity of the –PO3 2– residue. It is emphasized that the mentioned reference lines may also be used to reveal increased complex stabilities, for example, for certain complexes formed with 8-quinolyl phosphate the occurrence of 7-membered chelates can be proven in this way; the same procedure is also applicable to complexes of nucleotides, etc. The M(HOPO3) complexes are slightly more stable (on average by 0.08 log unit) than it is expected from the basicity of HPO4 2–; this observation is attributed to a more effective solvation, including hydrogen bonding, than is possible with CH3OPO3 2– species. Selective deposition of BaSO4 in the tight junctions (TJs) of frog skins led to profound and reversible functional alterations of these structures, as revealed by changes of tissue conductance (G), clamping current (I), and fluxes of extracellular markers (sulfate (JSO 4) and sucrose (JSUC)). Experiments were performed with nominally Ca2+ -free simple salt solutions on the apical side (usually KCl) and Na2SO4-Ringer on the inner side of skins. The deposition of BaSO4 in the TJs was obtained by diffusion and/or migration through the paracellular path of Ba2+ from the apical solution and SO 4 2– from the inner solution. A brief presence (2 to 6 min) of apical Ba2+ (Ba2+ pulse) is followed (i.e., when Ba2+ is removed from the apical fluid) by a large increase of G, I, JSO 4 and JSUC, above pre-Ba2+ levels. These attain a steady state within 15 to 30 min (overshoot phase), characterizing a conspicuous increase of the paracellular permeability. During the overshoot phase, a second Ba2+ pulse blocks the paracellular route while apical Ba2+ is present, leading to a new and larger overshoot when the Ba2+ pulse is terminated. Addition of apical Ca2+ triggers the resealing of the TJs, resulting in a full recovery of G, I, JSO 4 and JSUC. This Ca2+ -induced recovery persists when apical Ca2+ is removed. The presence of a normal Ca2+ concentration in the inner bathing Ringer does not induce the recovery process. Tissues remain viable after being submitted to the Ba2+ treatment and the subsequent overshoot. Experiments performed in the urinary bladder of Rana catesbeiana and skins and urinary bladders of Bufo marinus indicate that Ba2+ effect can also be elicited in these tissues. The above results seem to report general properties of the TJs. The fungus Cunninghamella verticillata was selected from isolates of oil-mill waste as a potent lipase producer as determined by the Rhodamine-B plate method. The lipase was purified from C. verticillata by ammonium sulphate fractionation, ion exchange chromatography and gel filtration. The purified enzyme was formed from a monomeric protein with molecular masses of 49 and 42 kDa by SDS–PAGE and gel filtration, respectively. The optimum pH at 40 °C was 7.5 and the optimum temperature at pH 7.5 was 40 °C. The enzyme was stable between a pH range of 7.5 and 9.0 at 30 °C for 24 h. The enzyme activity was strongly inhibited by AgNO3, NiCl2, HgCl2, CdCl2 and EDTA. However, the presence of Ca2+, Mn2+ and Ba2+ ions enhanced the activity of the enzyme. The activity of purified lipase with respect to pH, temperature and salt concentration was optimized using a Box–Behnken design experiment. A polynomial regression model used in analysing this data, showed a significant lack of fitness. Therefore, quadratic terms were incorporated in the regression model through variables. Maximum lipase activity (100%) was observed with 2 mM CaCl2, (pH 7.5) at a temperature of 40 °C. Regression co-efficient correlation was calculated as 0.9956.  相似文献

An 1H NMR (nuclear magnetic resonance) spectroscopic structural analysis of Cd2+ complexes formed with the pentapeptide phytochelatin, (NH3)+−(ψ-Glu-Cys)2−Gly−COO−(PC2), at a pH of 7.5 showed that the two thiol groups of the Cys residues and either the carbonyl or amide group of the peptide bond between Glu1 and Cys1 act as possible donor groups in the complexes at Cd2+/PC2 ratios up to 0.4. As the ratio increases, the carboxylate group of Glu2 and either the carbonyl or amide group of the peptide bond between Cys1 and Glu2 comes to serve as a donor group. The manner in which Cd2+ forms complexes with PC2 is distinctly different from Zn2+ and might account for the role of phytochelatin in Cd2+ detoxification. Electron absorption spectrometry demonstrated that the Cd2+ complexes are coordinated in a tetrahedral fashion by four thiol groups and that several sulfur atoms might bridge Cd2+ ions, resulting in the formation of polynuclear complexes. This contrasts with Zn2+ complex formation, which consists exclusively of a 1:1 complex.  相似文献

Metal complexes of thiamine pyrophosphate (TPP) of the general formula [M2(TPPH)2Cl2].4H2O (M =Zn2+, Cd2+) were isolated from methanolic solutions and characterized by elemental analysis, FT-IR, and multinuclear NMR spectroscopies. The data provide evidence for the bonding of the metals to the N(1') atom of the pyrimidine ring and to the pyrophosphate group. The stability constant measurements of TPP and 2-(α-hydroxyethyl)thiamine pyrophosphate (HETPP) metal complexes in aqueous solution imply the formation of dimeric complex species similar to the isolated solid products. They indicate also that HETPP forms more stable metal complexes than does TPP. To evaluate the coenzyme action of TPP and HETPP metal complexes, enzymic studies have been done using pyruvate decarboxylase apoenzyme. TPP metal complexes do not bind to the apoenzyme, unlike the Zn(II)-HETPP complex which can act as coenzyme. Ba2+ block of large conductance Ca2+-activated K+ channels was studied in patches of membrane excised from cultures of rat skeletal muscle using the patch clamp technique. Under conditions in which a blocking Ba2+ ion would dissociate to the external solution (150 mM N-methyl-d-glucamine+ o, 500 mM K+ i, 10 μM Ba2+ i, +30 mV, and 100 μM Ca2+ i to fully activate the channel), Ba2+ blocks with a mean duration of ∼2 s occurred, on average, once every ∼100 ms of channel open time. Of these Ba2+ blocks, 78% terminated with a single step in the current to the fully open level and 22% terminated with a transition to a subconductance level at ∼0.26 of the fully open level (preopening) before stepping to the fully open level. Only one apparent preclosing was observed in ∼10,000 Ba2+ blocks. Thus, the preopenings represent Ba2+-induced time-irreversible subconductance gating. The fraction of Ba2+ blocks terminating with a preopening and the duration of preopenings (exponentially distributed, mean = 0.75 ms) appeared independent of changes in [Ba2+]i or membrane potential. The fractional conductance of the preopenings increased from 0.24 at +10 mV to 0.39 at +90 mV. In contrast, the average subconductance level during normal gating in the absence of Ba2+ was independent of membrane potential, suggesting different mechanisms for preopenings and normal subconductance levels. Preopenings were also observed with 10 mM Ba2+ o and no added Ba2+ i. Adding K+, Rb+, or Na+ to the external solution decreased the fraction of Ba2+ blocks with preopenings, with K+ and Rb+ being more effective than Na+. These results are consistent with models in which the blocking Ba2+ ion either induces a preopening gate, and then dissociates to the external solution, or moves to a site located on the external side of the Ba2+ blocking site and acts directly as the preopening gate.  相似文献

Based on equilibrium binding studies, as well as on kinetic investigations, two types of interactions of Cu2+ ions with native DNA at low ionic strength could be characterized, namely, a nondenaturing and a denaturing complex formation. During a fast nondenaturing complex formation at low relative ligand concentrations and at low temperatures, different binding sites at the DNA bases become occupied by the metal ions. This type of interaction includes chelate formation of Cu2+ ions with atoms N(7) of purine bases and the oxygens of the corresponding phosphate groups, chelation between atoms N(7) and O of C(6) of the guanine bases, as well as the formation of specific intestrand crosslink complexes at adjacent G°C pairs of the sequence dGpC. CD spectra of the resulting nondenatured complex (DNA–Cu2+)nat may be interpreted in terms of a conformational change of DNA from the B-form to a C-like form on ligand binding. A slow cooperative denaturing complex formation occurs at increased copper concentrations and/or at increased temperatures. The uv absorption and CD spectra of the resulting complex, (DNA–Cu2+)denat, indicate DNA denaturation during this type of interaction. Such a conclusion is confirmed by microcalorimetric measurements, which show that the reaction consumes nearly the same amount of heat as acid denaturation of DNA. From these and the kinetic results, the following mechanism for the denaturing action of the ligands is suggested: binding of Cu2+ ions to atoms N(3) of the cytosine bases takes place when the cytosines come to the outside of the double helix as a result of statistical fluctuations. After the completion of the binding process, the bases cannot return to their initial positions, and thus local denaturation at the G·C pairs is brought about. The probability of the necessary fluctuations occurring is increased by chelation of Cu2+ ions between atoms N(7) and O of C(6) of the guanine bases during nondenaturing complex formation, which loosens one of the hydrogen bonds within the G·C pairs, as well as by raising the temperature. The implications of the new binding model, which comprises both the sequence-specific interstand crosslinks and the described mechanism of denaturing complex formation, are discussed and some predictions are made. The model is also used to explain the different renaturation properties of the denatured complexes of Cu2+, Cd2+, and Zn2+ ions with DNA. In temperature-jump experiments with the nondenatured complex (DNA–Cu2+)nat, a specific kinetic effect is observed, namely, the appearance of a lag in the response to the perturbation. The resulting sigmoidal shape of the kinetic curves is considered to be a consequence of the necessity of disrupting a certain number of the crosslinks existing in the nondenatured complex before the local unwinding of the binding regions (a main step of denaturing complex formation) may proceed.  相似文献

The interaction of antibiotic valinomycin with manganese (II) has been studied using circular dichroism, electron spin resonance and infrared techniques. Results show that Mn(II) forms complexes with valinomycin in both 2:1 (valinomycin-ion-valinomycin sandwich) and 1:1 (equimolar) stoichiometries. The 1:1 type observed here is very different from the well known K+-valinomycin bracelet conformation.  相似文献

A cold-active alkaline amylase producer Bacillus subtilis N8 was isolated from soil samples. Amylase synthesis optimally occurred at 15°C and pH 10.0 on agar plates containing starch. The molecular weight of the enzyme was found to be 205?kDa by performing SDS-PAGE. While the enzyme exhibited the highest activity at 25°C and pH 8.0, it was highly stable in alkaline media (pH 8.0–12.0) and retained 96% of its original activity at low temperatures (10–40°C) for 24?hr. While the amylase activity increased in the presence of β-mercaptoethanol (103%); Ba2+, Ca2+, Na+, Zn2+, Mn2+, H2O2, and Triton X-100 slightly inhibited the activity. The enzyme showed resistance to some denaturants: such as SDS, EDTA, and urea (52, 65, and 42%, respectively). N8 α-amylase displayed the maximum remaining activity of 56% with 3% NaCl. The major final products of starch were glucose, maltose, and maltose-derived oligosaccharides. This novel cold-active α-amylase has the potential to be used in the industries of detergent and food, bioremediation process and production of prebiotics.  相似文献
