The objectives of this study were to determine: (1) if the onset of winter hair growth (anagen) in mink could be delayed or inhibited by elevating endogenous PRL concentrations; (2) if bilaterally adrenalectomy (ADX)-induced winter anagen occurs concomitantly with a reduction in serum PRL concentrations, and (3) if exogenous dehydroepiandrosterone (DHEA), an adrenal steroid or Delta(5)-DIOL (a peripherally produced metabolite of DHEA), would delay or inhibit the onset of winter anagen. During early July, while in the resting (telogen) stage of the hair growth cycle, mink were treated with slow release implants containing haloperidol (HAL, a dopaminergic antagonist), melatonin (MEL), deoxycorticosterone (DOC), DHEA and Delta(5)-DIOL. In addition, mink were ADX'd and supplemented with DOC and DHEA. MEL reduced PRL levels to basal levels and induced winter anagen 7 weeks earlier than controls. Surprisingly, HAL initiated winter anagen 7 weeks earlier than controls (P < 0.05), although serum PRL levels were not different between the two groups. Mink that were ADX'd or ADX + DHEA-treated exhibited winter anagen 6 weeks earlier than controls (P < 0.05), but serum PRL concentrations were not different between the three groups. The administration of DHEA or Delta(5)-DIOL to mink with intact adrenals had no effect on the time of onset of winter anagen or serum PRL levels. Our findings suggest that a reduction in circulating PRL levels is not essential for onset of winter anagen in the mink and that the apparent inhibitory effects of the adrenal glands on initiation of winter anagen is not mediated through DHEA or its metabolite Delta(5)-DIOL. Mapping of QTL affecting fur quality traits (guard hair length, guard hair thickness, density of wool, surface of the fur and quality) and skin length was performed in a three‐generation mink population (F2 design). In the parental generation, Nordic Brown mink were crossed reciprocally with American Black short nap mink. In all, 1082 mink encompassing three generations were used for the analyses. The mink were genotyped for 104 microsatellites covering all 14 autosomes. The QTL analyses were performed by least‐square regression implemented in gridqtl software. Genetic and phenotypic correlations and heritabilities were estimated using the average information‐restricted maximum‐likelihood method. Evidence was found for QTL affecting fur quality traits on nine autosomes. QTL were detected for guard hair thickness on chromosomes 1, 2, 3, 6 and 13; for guard hair length on chromosomes 2, 3 and 6; for wool density on chromosomes 6 and 13; for surface on chromosomes 7, 12 and 13; for quality on chromosomes 6, 7, 11 and 13; and for skin length on chromosomes 7 and 9. Proximity of locations of QTL for guard hair length, guard hair thickness and for wool density and quality suggests that some of the traits are in part under the influence of the same genes. Traits under the influence of QTL at close or identical positions also were traits that were strongly genotypically correlated. Based on the results of correlation analyses, the most important single traits influencing the quality were found to be density of wool, guard hair thickness and appearance of the surface.  相似文献

Temperate and boreal mammals undergo seasonal changes in pelage that facilitate thermoregulation in winter and summer. We investigated photoperiodic influences on pelage characteristics of male Siberian and Syrian hamsters. Fur density (mg fur/cm2 skin) was measured by weighing the shavings of fur patches removed from the dorsal and ventral surfaces of hamsters maintained in long days (LDs) or transferred to short days (SDs). Patches were reshaved 3 wk later to assess fur regrowth (mg regrown fur/cm2 skin). Fur density was greater in SD than in LD Siberian hamsters after 11 wk of differential phototreatment. The onset of increased fur density in SDs was accompanied by a transient increase in fur regrowth (11-14 wk on the dorsal surface and 7-10 and 11-14 wk on the ventral surface), suggestive of a seasonal molting process. Fur density, body mass, and pelage color of Siberian hamsters returned to values characteristic of LD males after a similar duration of prolonged (>27 wk) SD treatment and appear to be regulated by a similar or common interval-timing mechanism. In Syrian hamsters, dorsal fur density, fur regrowth, and hair lengths were greater in SD than in LD males. Castration increased and testosterone (T) treatment decreased dorsal and ventral fur regrowth in LD and SD hamsters, but the effects of T manipulations on fur density were limited to a decrease in dorsal fur density after T treatment. Decreased circulating T in SDs likely contributes to the seasonal molt of male hamsters by increasing the rate of fur growth during the transition to the winter pelage.  相似文献

The role of the adrenal gland in suppressing the onset of maternal behavior in virgin female rats was investigated. Virgin female rats were either ovariectomized, adrenalectomized, or subjected to the combined operation and tested for the induction of maternal behavior by being exposed to young pups. Females that were both ovariectomized and adrenalectomized 4 weeks prior to testing exhibited significantly shorter latencies to the onset of maternal behavior than that of females subjected to either ovariectomy or adrenalectomy alone. Replacement of either estrogen or progesterone in the group that was both adrenalectomized and ovariectomized resulted in a prolongation of the average latency to become maternal. The results indicated that both estrogen and progesterone can act to inhibit the onset of maternal behavior and that the adrenal gland may act to suppress the onset of such behavior by supplying an extra-ovarian source of endogenous progesterone and estrogen.  相似文献

The effect of glucocorticoids on the diurnal rhythm of rat liver 3-hydroxy-3-methyl-glutaryl-CoA reductase (HMGR) has been controversial. Also, diurnal variation of D site-binding protein (DBP) has been suggested to be under a negative control of glucocorticoids. Here we have re-evaluated the effects of adrenal hormones on these rhythms at the level of gene expression. Sham-operated and bilaterally adrenalectomized rats were killed at 4-hr intervals and total RNA from each liver was subjected to Northern blot analysis. Diurnal variation patterns of HMGR and DBP mRNA levels in adrenalectomized rats were substantially identical to those in sham-operated rats, although DBP mRNA levels in adrenalectomized rats were slightly more abundant than in control rats. HMGR mRNA levels in adrenalectomized rats in the dark period were insensitive to a single injection of adrenal hormones, whereas the augmented levels of DBP mRNA in these animals were returned to the control levels by this treatment, indicating that glucocorticoids are prone to decrease the amplitude of variation in the DBP gene expression. The present results suggest that adrenal hormones are not critical for the generation of diurnal rhythms of these mRNAs.  相似文献

Anise oil, anethole, and several related compounds inhibit the deposition of glycogen in the livers of fasted, adrenalectomized rats by adrenal cortical extract. This effect is reversed by increased levels of adrenal extract.Anethole blocks the effect of adrenal cortical hormones in reducing insulin sensitivity of adrenalectomized rats.Adrenal hypertropy of unilaterally adrenalectomized rats is increased by anethole.It is postulated that these compounds compete for an active carbohydrate intermediate formed during glycogenesis.The possible role of β-glucuronidase is also pointed out.  相似文献

The influence of the adrenal gland and photoperiod on the adult female hamster estrous cycle was investigated. Hamsters were exposed to 14 hours of light per day and estrous cycles were monitored. Normal cycles were characterized by a copious discharge every 4th day. Hamsters were then adrenalectomized, adrenalectomized and blinded, or blinded only. Adrenalectomized/blinded hamsters were given a .2% sodium saccharine-1%(w/v) sodium chloride solution to drink which promotes long-term survival in most adrenalectomized hamsters. With blinding alone, it took from 18 to 62 days for cessation of estrous cycles. Cycles ceased in a mean of 36.2 days in blinded/adrenalectomized hamsters. After adrenalectomy only, 3 hamsters continued to show 4-day estrous cycles for at least 9 weeks in response to the long days. 6 adrenalectomized animals died due to adrenal insufficiency but displayed several 4-day estrous cycles. It is suggested that in the hamster, adrenal hormones are nonessential for the long-term expression of estrous cyclicity. However, further research is needed to determine whether adrenal hormones are capable of modulating photoperiodic time measurement or ovarian regression.  相似文献

It has been reported that the adrenal gland is essential to the development of obesity if Ay/a Yellow obese mice (Hausberger and Hausberger 1960). Since the actual body composition data to support this report has not been published, we attempted to duplicate this observation by adrenalectomy of the Yellow mice before the onset of obesity. Two groups of Yellow mice (Ay/a) and normal mice (a/a) were either sham operated or adrenalectomized at two months of age and at four months of age. Body weight was monitored until body gain had stopped. At that time the animals were sacrified and checked for completeness of adrenal removal. Body composition of dry matter, fat, protein, and ash was determined. Adrenalectomy caused a reduction of body fat of 33% and 30% in both the a/a Control and the Ay/a Yellow mice, respectively. The adrenalectomized Yellow mice were still fatter than adrenalectomized Controls (25.3% fat vs. 10.6% fat). Yellow and control adrenalectomized mice showed similar depression in growth rate. These data suggest that while the adrenal gland is essential for complete expression of the genetic potential for fat deposition, it may not be necessary for partial expression indicating a secondary role in the development of obesity in the Ay/a Yellow mouse.  相似文献

The present study investigated the effects of dietary copper (Cu) on growth performance and fur quality in growing-furring minks. One hundred and five standard dark female minks were randomly assigned to seven groups with the following dietary treatments: basal diet with no supplemental Cu (control) and basal diet supplemented with either 6, 12, 24, 48, 96 or 192 mg/kg Cu from copper sulphate, respectively. Our data showed that final body weight (P?=?0.033), daily gain (P?=?0.029) and fat digestibility (P?=?0.0006) responded to increasing levels of Cu. The activity of glutamic-oxalacetic transaminase (GOT) and glutamic-pyruvic transaminase (GPT) in serum increased (linear and quadratic, P?<?0.05) as Cu increased in the diet. Increasing Cu improved total protein (TP) and albumin (ALB) (quadratic, P?<?0.05). The level of ceruloplasmin (CER) responded in a linear (P?<?0.0001) and quadratic (P?<?0.0001) form with increasing level of Cu. Colour intensity of those minks pelted suggested that relatively high levels of supplemental Cu have a beneficial effect on intensifying hair colour of dark mink but did not affect leather thickness. Liver Cu and plasma Cu concentrations of the mink linearly (P?<?0.0001) responded to increasing levels of Cu. Our results indicate that growing-furring mink can efficiently utilize added dietary fat and that Cu plays an important role in the digestion of dietary fat in growing-furring mink, and supplemental dietary Cu in growing-furring mink promotes fat digestion and improve hair colour.  相似文献

Moulting cycles     
Moulting in mammals is a cyclic phenomenon which often occurs in a wave–like pattern. The moult cycle depends upon an inherent rhythm of activity in the hair follicles, which may be modified by systemic factors. In laboratory rodents a number of hormones affect the timing of the moult, as well as affecting the amount of hair produced and the loss of club hairs.
In Microtus agrestis a seasonal moult results in a sparse coat with coarse hairs in summer and a dense coat with fine hairs in winter. The moult appears to be adjusted to the environment by way of the endocrine system, with adrenal and thyroid hormones, as well as sex hormones, involved in the regulation. The importance of such adaptive coat changes are discussed.  相似文献

The influence of l ‐homoarginine on the heat‐induced aggregation of three model proteins, i.e. porcine, mink, and human growth hormones was investigated by circular dichroism spectroscopy. It was found that the effect of l ‐homoarginine as an analogue of arginine depends on the concentration of the additive as well as the protein itself. l ‐Homoarginine increased the onset temperature of heat‐induced aggregation of both porcine and mink growth hormones. However, the formation of human growth hormone aggregates was increased at low concentrations of l ‐homoarginine. Only at higher concentrations of the additive was the onset temperature of human growth hormone aggregation found to increase. Additional experiments of human growth hormone melting in the presence of histidine, lysine, and sodium chloride were performed. The effect of lysine was similar as in the presence of l ‐homoarginine. It follows that in protein formulations low concentrations of amino acids should be used with some precaution. At low concentration of additive, depending on the charge of both protein and amino acid used, the promotion of aggregation of unfolding intermediates may occur. © 2015 American Institute of Chemical Engineers Biotechnol. Adrenal hormones were studied as possible triggering substances of the synthesis of acute-phase reactants in rats. alpha-Macrofoetoprotein, which rises sharply in concentration during inflammation, was used to monitor the acute-phase reaction. In normal rats glucocorticoids and catecholamines induce alpha-macrofoetoprotein synthesis; glucocorticoids only increase alpha-macrofoetoprotein to moderate levels in plasma, but catecholamines enhance alpha-macrofoetoprotein synthesis to very high levels, comparable with those observed in the post-injury phase. However, catecholamines in vivo also activate the adrenal cortex, suggesting a synergistic effect of both kinds of adrenal hormones. Our study showed that in adrenalectomized rats, the effect of catecholamines on alpha-macrofoetoprotein synthesis is greatly diminished, whereas the moderate effect of glucocorticoids remains. Combination of glucocorticoids and catecholamines induces extremely high alpha-macrofoetoprotein levels in both adrenalectomized and normal rats. With crossed immunoelectrophoresis it was shown that other acute-phase reactants, such as haptoglobin and alpha 1-major acute-phase protein, are affected differently by the hormones. Contrary to glucocorticoids, catecholamines give a pattern comparable with that found after surgical injury.  相似文献

研究表明紫貂和水貂被毛的形态高度分化,可分为针毛,中间针毛和绒毛3类。针毛笔直,远端(大约3/4处)皮质高度发育,有良好的耐磨性。中间针毛比针毛短、细,大约有3个弯曲,且髓质含量最高。绒毛最短、细,大约有5个弯曲,髓质也较发达。 扫描电镜下的毛小皮形态可归纳为波纹形,披针形,菱形和倒三角形4种。光镜下的髓质形态可归纳为单柱组型,多柱组型和网型3种。比较分析的结果表明:毛小皮和髓质形态在这两种动物间的差别较明显,具有种的特异性、可作为哺乳动物分类的参考指标。  相似文献

1. Melatonin and glucocorticoids are known to affect the immune response in an opposite mode. The probability for an interaction between these hormones in the thymus gland has been investigated in rats following chronic administration of exogenous melatonin and long-term exposure to variable levels of circulating glucocorticoids. 2. Daily melatonin administration was shown to affect the properties of corticosterone and progestin receptors in the thymus in the presence of normal and increased systemic corticosterone concentrations, but not in adrenalectomized animals. 3. In intact rats melatonin caused a marked increase in the affinity and a decrease in the density of thymic receptors for adrenal steroids. Following corticosterone overdosage, simultaneously with melatonin treatment, a decrease in receptor affinity and a relative increase in the number of binding sites was observed. 4. The results suggest that steroid hormone receptors in the thymus might be considered as a target site for the interaction between melatonin and adrenal steroids in the modulation of the immune response.  相似文献

The immune system is considered to be an energetically expensive component of an individual's life history. Investment in the immune system can depend on the environment that an individual finds itself in. The American mink, Neovison vison, exists in the natural environment and on fur farms. The natural environment and mink farm differ in many ways, such as wild mink being exposed to many parasites that are less prevalent and less abundant in the domestic environment because of veterinary care. We collected free‐ranging mink from commercial trappers and domestic mink from fur farmers in Ontario and Nova Scotia, Canada, and examined relative spleen mass. Wild male mink had larger spleens than domestic mink in Nova Scotia, with a similar trend in Ontario. Female mink that escaped from farms (feral) in Nova Scotia had significantly larger spleens than their domestic counterparts on the farms. Both of these results are consistent with the prediction that the natural environment contains parasites and pathogens that require enhanced investment in the immune system. In Nova Scotia, females had larger spleens than males, whether considering wild or domestic populations. Finally, wild mink showed greater condition dependence of spleen mass than domestic populations. Further investigations should include experimental approaches such as providing veterinary care to wild populations to assess the effects of parasites and pathogens on the immune system. © 2012 The Linnean Society of London, Biological Journal of the Linnean Society, 2012, 107 , 624–631.  相似文献

We studied a group housing system as an alternative to the traditional pair housing of juvenile mink. The focus was on both the welfare and production of mink. The pairs were housed in standard mink cages, whereas the groups were in row cage systems consisting of three standard mink cages connected to each other. The welfare of the mink was evaluated by behavioural observations (stereotypies and social contacts), evaluation of the incidence of scars assumed to be caused by biting, and adrenal function (serum cortisol level after adrenocorticotropic hormone (ACTH) administration and adrenal mass). Feed consumption, pelt length, quality and price were used for comparing the two housing systems from the economic point of view. Although the incidence of scars showed that there might have been more aggressive behaviour among the group-housed than among the pair-housed mink, this was not observed unambiguously in behavioural observations, and, at least, aggression did not cause mortality or serious injuries to the animals as has been observed in some earlier studies. In addition, the housing system did not affect pelt size, and, although the quality of the pelts was slightly lower in the group than in pair-housed mink, there was only a tendency for lower pelt prices. The lower pelt prices in the group-housed mink might even be partially compensated for by the group-housed mink eating 10% to 20% less in the late autumn, due to thermoregulatory benefits, than their pair-housed conspecifics. The results on the frequency of stereotypic behaviour (but not adrenal function) suggest that the group-housed animals were possibly less stressed than the pair-housed animals. Group housing of juvenile farmed mink in a row cage system cannot be recommended before the effects on welfare and production are clarified in further studies.  相似文献

The urinary bladder of Bufo marinus has been shown to excrete H+ and NH4+ and this excretion is increased by metabolic acidosis. The involvement of the adrenal gland and its steroid secretions in the adaptation for increased acid and ammonia excretion by the bladder was tested during the course of this study. Groups of toads were adrenalectomized and maintained in chronic NH4Cl-induced acidosis. Three other groups of toads were adrenalectomized and put in acidosis but repleted with 2.5 mg/day of either cortisol (CT), dexamethasone (Dexa), or deoxycorticosterone acetate (DOCA). All control groups were sham-operated. The bladders were excised after 3 days and mounted between 2-ml Lucite chambers. Net H+ and NH4+ fluxes into the mucosal media were measured and reported in units of nanomoles per 100 mg bladder per minute. In control acidotic toads H+ excretion was 20.1 +/- 2.0 and the adrenalectomized nonreplete group H+ excretion was 14.2 +/- 1.87 (P less than 0.04). For the same groups NH4+ excretion was 2.90 +/- 0.26 for the controls and 1.38 +/- 0.19 for the adrenalectomized (P less than 0.001). The H+ excretion in CT-, Dexa-, and DOCA-repleted toads was not significantly different from the control group. NH4+ excretion, however, showed a 55% decrease (P less than 0.001) in the CT group, and a 45% decrease (P less than 0.05) in the Dexa group. The NH4+ excretion in the DOCA repleted group was significantly different from the control group. Therefore, we conclude that the adrenal gland plays a role in the adaptive increase of H+ and NH4+ excretion by the urinary bladder in acidosis through the secretion of steroid hormones. The increase in NH4+ excretion appears to be a mineralocorticoid-stimulated process. We were not able to determine in this study if the steroid hormones had an exacting regulatory role or one of a permissive role over H+ and NH4+ excretion in the toad urinary bladder.  相似文献

The concentrations of catecholamine and indoleamine metabolites were measured in intact and adrenalectomized mice to determine whether adrenal hormones mediate or modulate the stress-induced responses. Thirty minutes of footshock resulted in significant increases of the ratios of the dopamine (DA) catabolite, dihydroxyphenylacetic acid (DOPAC), to DA in prefrontal cortex, nucleus accumbens, striatum, hypothalamus, and brainstem, and of homovanillic (HVA)/DA ratios in nucleus accumbens, striatum, amygdala, and hypothalamus. Ratios of 3-methoxy-4-hydroxyphenylethyleneglycol to norepinephrine (NE) were also increased in prefrontal cortex, nucleus accumbens, septum, amygdala, hypothalamus, hippocampus, and brainstem. The concentration of NE was decreased in amygdala. 5-Hydroxyindoleacetic acid (5-HIAA)/5-hydroxytryptamine (5-HT, serotonin) ratios and free tryptophan were also increased in every brain region. Very similar data were obtained from mice restrained for 30 min. Adrenalectomy resulted in increased HVA/DA ratios in prefrontal cortex and striatum, and 5-HIAA/5-HT in septum. The stress-related changes were largely similar in adrenalectomized mice. Significant interactions between adrenalectomy and footshock treatment occurred in prefrontal cortical DOPAC/DA and hypothalamic NE which was depleted only in adrenalectomized mice, suggesting tendencies for these measures to be more responsive in adrenalectomized mice. Corticosterone administration (0.5-2.0 mg/kg s.c.) which resulted in plasma concentrations in the physiological range did not alter the concentrations of the cerebral metabolites measured in any region. We conclude that adrenal hormones do not mediate cerebral catecholamine or indoleamine metabolism in stress, although adrenalectomy may affect HVA and 5-HIAA metabolism, and there was a tendency for catecholamines to be more sensitive to stress in adrenalectomized animals.  相似文献

Water-restricted rats exhibit a rapid decrease in plasma corticosterone after drinking. The present study examined the effect of restriction-induced drinking on plasma aldosterone and plasma clearance of corticosterone. Rats were water restricted for 6-7 days and then killed before or 15 min after water administration; plasma and adrenal hormones were assayed. Plasma and adrenal corticosterone decreased after drinking without a change in plasma corticosteroid-binding globulin; plasma ACTH decreased or did not change. In contrast, plasma aldosterone did not change or increased after drinking; plasma renin activity was elevated by water restriction and increased further after drinking. In another experiment, rats were adrenalectomized, and corticosterone and aldosterone were replaced with pellets and osmotic minipumps, respectively. Rats were water restricted and killed. There was a small decrease in plasma corticosterone but no change in aldosterone after drinking in adrenalectomized animals. These data suggest that changes in plasma steroids after restriction-induced drinking result from zone-specific responses of the adrenal to known secretagogues, with minimal contribution from increased plasma clearance.  相似文献
