The GTP-binding subunit of transducin (Tα) activates the cGMP phosphodiesterase (PDE) of bovine retinal rods by relieving the constraint imposed by the inhibitory subunit PDEγ. We have isolated and characterized the complex Tα.GTPγS-PDEγ formed when Tα is activated by the nonhydrolyzable analog GTPγS. Sedimentation and light-scattering techniques demonstrate that, in contrast to free Tγ.GTPγS, which is soluble, the Tα.GTPγS-PDEγ complex, as well as Tα.GTP-PDEγ, is membrane bound at cytosolic ionic strength. It is eluted from the membrane at low ionic strength as a monomeric and 1:1 stoichiometric complex. The relative affinities of PDEγ for PDEαβ and for Tα.GTP are discussed.  相似文献

The N-formyl-L-methionyl-L-leucyl-L-phenylalanine (fMLP)-OMe (1) analogues for-Thp-Leu-Ain-OMe (2), for-Thp-Leu-Phe-OMe (3), for-Met-Leu-Ain-OMe (4), for-Met-Delta(z)Leu-Phe-OMe (5), for-Met-Lys-Phe-For-Met-Lys-Phe (6), for-Met-Leu-Pheol-COMe (7), and for-Nle-Leu-Phe-OMe (8) have been studied. Some of these have been found selective towards the activation of different biological responses of human neutrophils. In particular, peptides 2 and 3, which evoke only chemotaxis, are ineffective in enhancing inositol phosphate, as well as cyclic AMP (cAMP) levels. On the contrary, analogues 5 and 7, which induce superoxide anion production and degranulation, but not chemotaxis, significantly increase the levels of the two intracellular messengers, as is the case of the full agonists 1 and 6. The Ca(2+) ionophore A23187 also activates phospholipase C (PLC) and increases the nucleotide levels; when tested in combination with peptide 1 or 5, a supra-additive enhancement of cAMP concentration is obtained. The PLC blocker, U-73122, inhibits the formylpeptide-induced inositol phosphate formation, as well as cAMP increase. Moreover, this drug drastically reduces superoxide anion release triggered by 1 or 5, whereas it inhibits to a much lesser extent neutrophil chemotaxis induced by 1 or 2. Our results suggest that: (i) PLC stimulation is involved in cAMP enhancement by formylpeptides; (ii) the activation of PLC by formylpeptides, in conditions of increased Ca(2+) influx, induces a supra-additive enhancement of the nucleotide; (iii) the inability of pure chemoattractants to significantly alter the PLC activity or cAMP level, differently from full agonists or peptides specific in inducing superoxide anion release, appears as a general property. Thus, the activation of neutrophil PLC seems essential for superoxide anion release, but less involved in the chemotactic response.  相似文献

Purmorphamine is a purine analog and it affects osteogenesis, in part, through the modulation of Sonic Hedgehog signaling pathway. However, the underlying mechanism has remained elusive. Previously, we identified two purmorphamine derivatives that modulate the expression of osteogenic markers includingRunx2, Dlx2 andHes1. However, the extents of osteogenesis varied depending on the assay employed, and this necessitates the identification of reliable markers. Here, we report that the expression of osteocalcin was most sensitive to the treatment of purmorphamine derivatives among genes we analyzed by RT-PCR. Purmorphamine derivatives differentially modulated the expression of early and late osteogenic markers; alkaline phosphatase and osteocalcin gene respectively. In addition, the expression of osteocalcin is also modulated differentially by purmorphamine derivatives over time. Although JNK activity is reported to be necessary for osteocalcin expression, its requirement varied among purmorphamine derivatives. Also the overall cytotoxic response to purmorphamine derivatives appeared slightly different.  相似文献

Regulation of cyclic GMP binding to retinal rod membranes by calcium   总被引:1,自引:0,他引:1  
The apparently cooperative binding of 8-(5-thioacetamidofluorescein)-cGMP (SAF-cGMP) to cGMP-binding sites of the rod outer segments is regulated by Ca2+ in the 0.1-1 microM activity range. High Ca2+ reduces, and low Ca2+ increases the affinity of SAF-cGMP binding. This regulation involves only intrinsic membrane components. It is proposed that an allosteric regulation of cGMP binding by Ca2+ can contribute to photoreceptor potential adaptation.  相似文献

During static conditions the nociceptive reflex is known to vary as a function of, for example, the stimulus position, stimulus intensity, and muscle contraction. The aim of the present human study was to investigate whether the reflex and the corresponding perception of pain are modulated by cyclic movements of the limb involved. Reflexes, evoked by nociceptive electric stimulation of the sural nerve, were recorded from the biceps femoris and the rectus femoris muscles in eight volunteers. Four different experiments were performed to compare the nociceptive reflex and pain score elicited during active isometric/dynamic flexion/extension of the knee joint. The amplitudes of the reflexes were largest for the dynamic conditions. The reflexes, evoked during dynamic extension and isometric contraction of the rectus femoris muscle, had the shortest latencies but the recordings from the biceps femoris muscle were larger than from the rectus femoris muscle. Knee joint angle recordings showed that the largest angle variations occurred for the dynamic conditions and were only marginally disturbed for the isometric conditions. A given stimulus intensity evoked the highest pain intensity during isometric contractions. This indicates that there would seem to be no causal relationship between the size of the nociceptive reflex and the pain intensity.  相似文献

Antibiotic resistance has reached alarming levels in many clinically-relevant human pathogens, and there is an increasing clinical need for new antibiotics active on drug-resistant Gram-negative pathogens who rapidly evolve towards pandrug resistance phenotypes. Here, we report on two related classes of guanidinic compounds endowed with antibacterial activity. The two best compounds (9a and 13d) exhibited the most potent antibacterial activity with MIC values ranging 0.12–8 μg/ml with most tested pathogens, including both Gram-positive and Gram-negative bacteria. Interestingly, MIC values were not affected (1–8 μg/ml) when measured using recent clinical isolates with various antibiotic resistance determinants. The results reported herein identify guazatine derivatives as an interesting starting point for the optimization of a potentially novel class of antibacterial agents.  相似文献

Acetylcholine esterase (AChE) activity is lowered in vitiliginous skin. The AChE activity in 52 cases of vitiligo during repigmentation and depigmentation has been observed in this study. The cases with marginal dendritic melanocytes show that AChE is negative in these cells during depigmentation but positive on repigmentation. There is little variation in activity in the cases showing nondendritic marginal melanocytes. Acetylcholine (ACh) has an inhibitory effect on dopa oxidase activity in both types of marginal melanocytes in vitiligo. ACh modulates pigment production by the melanocytes, its role being inhibitory. From the present results, it is evident that a fall in AChE activity in the melanocytes leads to greater inhibition by ACh aggravating the process of depigmentation in vitiligo.  相似文献

Modulation of suppressor cell activity by thymosin was evaluated in vivo in a murine tumor system, and in vitro on human lymphocytes. We showed that splenocytes from tumor bearing mice were able to enhance tumor growth in a syngeneic system. This enhancement was T dependent and disappeared by Thymosin treatment. A decrease of Tumor size associated with injection of bone marrow cells treated by thymosin was observed. In the human system we showed that ConA stimulated lymphocytes were able to suppress the response of normal lymphocytes to PHA, PWM and ConA, and in MLC. This effect was significantly blocked in presence of thymosin fraction 5.  相似文献

In order to identify the cGMP-sensitive ion channel protein in frog rod outer segments (ROS), we analyzed cGMP binding proteins in the ROS by means of photoaffinity labeling with [3H]cGMP. We found four cGMP binding proteins with molecular weights (Mws) of 250K, 100K, 92K, and 53K. The 250K protein was an integral-membrane protein, which we named cG-Protein, (cG stands for cGMP). The cGMP-binding to cG-Protein was slightly increased by CaCl2. cG-Protein has a carbohydrate moiety. The amount of cG-Protein per single rod outer segment was estimated to be 9.0 x 10(6) molecules. Light-dependent phosphorylation of cG-Protein with [gamma-32P]ATP was observed. The 100K and 92K proteins were peripheral-membrane proteins, corresponding to cGMP phosphodiesterase. The 53K protein was a soluble protein. Incorporation of a membrane protein fraction of frog ROS into a planar lipid bilayer resulted in the appearance of at least three kinds of ion channel activities; two of them were related to cGMP. The possibility that cG-Protein is the cGMP-sensitive ion channel in vivo is discussed.  相似文献

First discovered in the sensory epithelium of the visual and olfactory systems, cyclic nucleotide-gated (CNG) ion channels have now been found in tissues throughout the body. Native rod CNG channels are tetramers composed of homologous, but distinct, alpha- and beta-subunits. The goal of this study was to develop a novel method for targeting covalent attachment of cGMP to individual subunit types. Toward this goal, we have found that treatment of membrane patches expressing rod alpha-subunit channels with sulfhydryl-reactive derivatives of cGMP resulted in irreversible activation. The persistent currents were sensitive to block by both Mg(2+) and tetracaine. Pretreatment of the patch with the sulfhydryl-blocking reagents N-ethylmaleimide (NEM) and bis-dithionitrobenzoic acid (DTNB) prevented covalent activation; the effect of DTNB was reversed by reduction with DTT. Furthermore, the process of covalent activation was dramatically slowed by the presence of an excess of 8-Br-cGMP. These results suggested that covalent activation resulted from the tethering of cGMP near the channel's ligand-binding sites by reaction with an endogenous cysteine. The alpha-subunit of the rod channel contains seven cysteine residues, and we set out to determine the site of attachment by site-directed mutagenesis. Surprisingly, irreversible activation was not abolished by elimination of all seven cysteine residues. This result suggests that the site of attachment is on a tightly associated protein, rather than on the channel protein itself. To further investigate these results, we treated patches containing irreversibly activated channels with 100 microg/mL trypsin and discovered two modes of covalent activation. One type developed rapidly and was removed by trypsin treatment, and the second developed slowly and was resistant to trypsin treatment. Both types of covalent activation were present in all mutants tested and were also present when CNG channels were expressed in HEK-293 cells. These results suggest that CNG channel subunits may associate with endogenous proteins when they are expressed in heterologous systems.  相似文献

The effect of lignin on cytotoxicity, mutagenicity and SOS response induced by 4-nitroquinoline-N-oxide (4NQO), 3-(5-nitro-2-furyl)acrylic acid (5NFAA), 2-nitrofluorene (2NF) as well as hydrogen peroxide was investigated in bacterial assay systems, i.e. the Ames test with Salmonella typhimurium TA98, TA100, TA102 and the SOS chromotest with Escherichia coli PQ37. Lignin preparations obtained from beech wood significantly decreased the mutagenicity induced by 4NQO, 2NF and H(2)O(2). In the case of mutagenicity induced by 5NFAA the effect was lower. Antimutagenic properties of lignin samples tested were shown also by SOS chromotest where lignin inhibited the ability of both 4NQO and H(2)O(2) to induce the SOS response. Derivatives of lignin including those from soft and hard wood, as well as from annual plants differ in their efficiency to inhibit the induction of the SOS response. The modified lignins isolated from beech and spruce wood exhibit a high level of protection. Lignins from annual plants-corn cobs and straw-only marginally evoked an antimutagenic response, but their effect was increased by hydrothermic treatment of both annual plants. The results obtained indicate the prospective utilization of lignin preparations as additive in chemo-prevention. The antimutagenic effect of lignin samples varies with the method of isolation and modification, as well as with the genetic origin of the lignin.  相似文献

Reprogramming energy metabolism from oxidative phosphorylation to aerobic glycolysis, a common feature of human cancer, is associated with a relative acidic tumor microenvironment which can sometimes be further accentuated by hypoxia operating within most solid tumors. We found that alteration of extracellular pH induces marked and rapid changes of autophagic activity. Interestingly, acidic and basic conditions induced completely opposite effect on autophagy, with its activity suppressed at lower pH whereas stimulated at higher pH. Gene knockdown experiments indicated that pH induced-autophagy requires Beclin 1, Vps34 and Atg5, key components of the autophagy pathway. Of note, an acidic condition not only inhibits the basal but also blocks the starvation-induced autophagy activity. Significantly, examination of different areas of tumor mass revealed a lower autophagic activity within the inner region than the outer region. These findings have important implications on the connections between autophagy and cancer as well as a wide range of other physiological and pathological processes.  相似文献
