Background

Children receiving Total Body Irradiation (TBI) in preparation for Hematopoietic Stem Cell Transplantation (HSCT) are at risk for Growth Hormone Deficiency (GHD), which sometimes severely compromises their Final Height (FH). To better represent the impact of such therapies on growth we apply a mathematical model, which accounts both for the gompertzian-like growth trend and the hormone-related ‘spurts’, and evaluate how the parameter values estimated on the children undergoing TBI differ from those of the matched normal population.

Methods

25 patients long-term childhood lymphoblastic and myeloid acute leukaemia survivors followed at Pediatric Onco-Hematology, Stem Cell Transplantation and Cellular Therapy Division, Regina Margherita Children’s Hospital (Turin, Italy) were retrospectively analysed for assessing the influence of TBI on their longitudinal growth and for validating a new method to estimate the GH therapy effects. Six were treated with GH therapy after a GHD diagnosis.

Results

We show that when TBI was performed before puberty overall growth and pubertal duration were significantly impaired, but such growth limitations were completely reverted in the small sample (6 over 25) of children who underwent GH replacement therapies.

Conclusion

Since in principle the model could account for any additional growth ‘spurt’ induced by therapy, it may become a useful ‘simulation’ tool for paediatricians for comparing the predicted therapy effectiveness depending on its timing and dosage.

Division of labour in Solenopsis invicta follows a familiar pattern: younger, smaller ants tend toward brood care while older, larger ants tend toward foraging. However, long-term observations of marked individuals reveal that length of nursing and foraging ‘careers’ and the age of transition between these activities vary considerably between and within size groups, and are related to length of life. Experiments with entire colonies show that larger ants are more likely than smaller ants to forage for insect prey. There are two main worker castes, ‘nurses’ and ‘foragers’, whose members span a wide age-size range, and a large ‘reserve’ subcaste, heterogeneous in age, size, and behaviour: reserves may nurse, forage, store liquid food, or relay food from nurses to foragers. The proportion of ants engaged in foraging decreases with colony size because many ants in large colonies are not exposed to recruitment signals.  相似文献

I have been invited by the board of the French Society of Cell Biology (SBCF) to write a text around my presentation in the Symposium ‘A day at the Cell Centre’, held at the Curie Institute on May 17, 2019, and organized by four of my former students, namely Juliette Azimzadeh, Nathalie Delgehyr, Matthieu Piel and Manuel Théry. We conducted an experiment in the Beijing Milu Park to study the social behavior of male Père David's deer, and related social behavior to social position and serum testosterone level of the stags during rut. We classified the stags into three rank classes according to their rutting behavior: ‘harem master’, ‘challenger’ and ‘bachelor’. We monitored the behaviors of four ‘harem masters’, five ‘challengers’ and five ‘bachelors’, and analyzed serum testosterone levels in blood samples of those 14 stags using radioimmunoassay. We defined the effectiveness value, E = A/T, to assess the effectiveness of herding or mating attempts made by stags (‘T’ represents the frequency of herding or mating attempts made by a stag and ‘A’ represents the frequency of herding or mating attempts accepted by hinds). We found that: (1) the ‘harem masters’ and the ‘challengers’ displayed more frequent rut and locomotive behaviors but fewer ingestion behaviors than the ‘bachelors’; (2) serum testosterone levels in the ‘harem masters’ and the ‘challengers’ were higher than that in the ‘bachelors’; (3) effectiveness value of herding attempts differed significantly between the three types of stags, being highest in the ‘harem masters’ and the lowest in the ‘bachelors’; and (4) effectiveness value of mating attempts was significantly greater for the ‘harem masters’ than for the ‘challengers’. We conclude that: (1) reproductive behavior of the Père David's deer stags is strongly associated with social rank; (2) social roles of Père David's deer stags during the rut are related to the testosterone secretion; and (3) rank class affects the mating opportunity of the stags.  相似文献

Cell competition where ‘loser’ cells are eliminated by neighbors with higher fitness is a widespread phenomenon in development. However, a growing body of evidence argues cells with somatic mutations compete with their wild type counterparts in the earliest stages of cancer development. Recent studies have begun to shed light on the molecular and cellular mechanisms that alter the competitiveness of cells carrying somatic mutations in adult tissues. Cells with a ‘winner’ phenotype create clones which may expand into extensive fields of mutant cells within normal appearing epithelium, favoring the accumulation of further genetic alterations and the evolution of cancer. Here we focus on how mutations which disrupt the Notch signaling pathway confer a ‘super competitor’ status on cells in squamous epithelia and consider the broader implications for cancer evolution.  相似文献

A short review presented deals with the history of biochemistry development in the western regions of Ukraine. Two principal biochemical schools were founded here by J. Parnas (1884-1949) and S. Gzhytskiy (1900-1976). While most of the students and collaborators of Prof. J. Parnas left for Poland and other western states, those ones of Prof. S. Gzhytskiy stayed in Lviv and other scientific centers of Ukraine. In 1979 Prof. S. Kusen (one of Gzhytskiy's former students and collaborators) and Prof. G. Shavlovsky headed two scientific departments founded in Lviv at O. V. Palladin Institute of Biochemistry. This event could be considered as the beginning of modern biochemistry development in the western regions of Ukraine. Since 1992 in Lviv there exists the Division of Regulatory Cell Systems of O. V. Palladin Institute of Biochemistry of the National Academy of Sciences of Ukraine headed since 1995 by Prof. R. Stoika. Four Departments work in the structure of this Division: 1) the Department of Biochemistry of Cell Differentiation headed in 1979-1997 by S. Kusen and since 1997 by L. Drobot; 2) the Department of Regulation of Cell Proliferation created in 1993 and headed by R. Stoika; 3) the Department of Biochemical Genetics created in 1988 and headed by A. Sibirny; 4) the Department of Regulation of Synthesis of Low Molecular Compounds headed in 1979-1996 by G. Shavlovsky and since 1996 by D. Fedorovych. Division of Regulatory Cell Systems is presently the leading scientific center in Ukraine in the study of the biochemical mechanisms of proliferation, differentiation and apoptosis of normal and tumour cells and in the development of effective biotechnological processes for obtaining the biologically active substances using yeast. Numerous publications of its collaborators in the high impact factor scientific magazines as well as the realisation of the international grants confirm this statement. Taking into account the high level of scientific research and availability of highly skilled scientists at the Division in 1999 the Presidium of the National Academy of Sciences of Ukraine took a resolution to transform the Division into the Institute of Cell Biology of the National Academy of Sciences of Ukraine, which was founded in 2000 on the basis of the Division.  相似文献

Expansins are wall‐loosening proteins that promote the extension of primary cell walls without the hydrolysis of major structural components. Previously, proteins from the EXPA (α–expansin) family were found to loosen eudicot cell walls but to be less effective on grass cell walls, whereas the reverse pattern was found for EXPB (β–expansin) proteins obtained from grass pollen. To understand the evolutionary and structural bases for the selectivity of EXPB action, we assessed the extension (creep) response of cell walls from diverse monocot families to EXPA and EXPB treatments. Cell walls from Cyperaceae and Juncaceae (families closely related to grasses) displayed a typical grass response (‘β–response’). Walls from more distant monocots, including some species that share with grasses high levels of arabinoxylan, responded preferentially to α–expansins (‘α–response’), behaving in this regard like eudicots. An expansin with selective activity for grass cell walls was detected in Cyperaceae pollen, coinciding with the expression of genes from the divergent EXPB–I branch that includes grass pollen β–expansins. The evolutionary origin of this branch was located within Poales on the basis of phylogenetic analyses and its association with the ‘sigma’ whole‐genome duplication. Accelerated evolution in this branch has remodeled the protein surface in contact with the substrate, potentially for binding highly substituted arabinoxylan. We propose that the evolution of the divergent EXPB–I group made a fundamental change in the target and mechanism of wall loosening in the grass lineage possible, involving a new structural role for xylans and the expansins that target them.  相似文献

This paper presents the results of floral spectral studies on 1275 flowers from India, Brazil, Israel, Germany, and Norway. Floral spectral reflectance from 400 to 700 nm (nm) was used to quantitatively represent ‘human-perceived’ color of flowers in Red, Green, Blue color space. Floral spectral reflectance from 350 to 600 nm was used to discern and objectively represent ‘insect pollinator-perceived’ flower colors in color hexagon. We leverage the advantage offered by ‘quantified human perception’ provided by ‘human-perceived’ floral colors to represent the distribution of floral hues and uncover the relationship between the composition of incoming solar radiation and predominant ‘human-perceived’ floral colors at the tropics and the higher latitudes. Further, the observed species-level mutual exclusivity of ‘insect pollinator-perceived’ floral colors is stated as chromatic exclusivity hypothesis. We compare ‘human-perceived’ and ‘insect pollinator-perceived’ floral colors at Trivandrum (India) and provide a physical explanation for short and long ‘wavelength triads’ of insect pollinator and human visual sensitivity respectively.  相似文献

Colorectal cancer is the second most common cancer in women and third most common cancer in men. Cell signaling alterations in colon cancer, especially in aggressive metastatic tumors, require further investigations. The present study aims to compare the expression pattern of proteins associated with cell signaling in paired tumor and non-tumor samples of patients with colon cancer, as well as to define the cluster of proteins to differentiate patients with non-metastatic (Dukes’ grade B) and metastatic (Dukes’ grade C&D) colon cancer. Frozen tumor and non-tumor samples were collected after tumor resection from 19 patients with colon cancer. The Panorama? Antibody Microarray-Cell Signaling kits were used for the analyses. The expression ratios of paired tumor/non-tumor samples were calculated for the each protein. We employed R packages ‘samr’, ‘gplots’, ‘supclust’ (pelora, wilma algorithms), ‘glmnet’ for the differential expression analysis, supervised clustering and penalized logistic regression. Significance analysis of microarrays revealed 9 significantly up-regulated proteins, including protein kinase C gamma, c-Myc, MDM2, pan cytokeratin, and 1 significantly down-regulated protein (GAP1) in tumoral mucosa. Pan-cytokeratin and APP were up-regulated in tumor versus non-tumor tissue, and were selected in the predictive cluster to discriminate colon cancer type. Higher levels of S-100b and phospho-Tau-pSer199/202 were confirmed as the predictors of non-metastatic colon cancer by all employed regression/clustering methods. Deregulated proteins in colon cancer are involved in oncogenic signal transduction, cell cycle control, and regulation of cytoskeleton/transport. Further studies are needed to validate potential protein markers of colon cancer development and metastatic progression.  相似文献

Massive efforts to sequence cancer genomes have compiled an impressive catalogue of cancer mutations, revealing the recurrent exploitation of a handful of ‘hallmark cancer pathways’. However, unraveling how sets of mutated proteins in these and other pathways hijack pro-proliferative signaling networks and dictate therapeutic responsiveness remains challenging. Here, we show that cancer driver protein–protein interactions are enriched for additional cancer drivers, highlighting the power of physical interaction maps to explain known, as well as uncover new, disease-promoting pathway interrelationships. We hypothesize that by systematically mapping the protein–protein and genetic interactions in cancer—thereby creating Cancer Cell Maps—we will create resources against which to contextualize a patient’s mutations into perturbed pathways/complexes and thereby specify a matching targeted therapeutic cocktail.  相似文献

在大田条件下研究了两种品质类型花生(Arachis hypogaea)品质形成的动态差异及其子叶细胞超微结构的差异。结果表明, 高蛋白品种‘XB023’的蛋白质含量在籽仁发育前期较高油品种‘鲁花9号’低, 后期显著高于‘鲁花9号’, 且成熟期籽仁8种必需氨基酸组分含量均高于‘鲁花9号’, 其中谷氨酸、赖氨酸和亮氨酸含量差异极显著; ‘XB023’脂肪含量在籽仁发育期一直低于‘鲁花9号’。‘XB023’各时期的籽仁可溶性糖含量和油酸/亚油酸(O/L)值均显著低于‘鲁花9号’。两品种在果针入土10天时子叶细胞即形成淀粉粒、脂体和蛋白体, 随后脂体、蛋白体的数量不断增加, 淀粉粒先增大后逐渐缩小解体。‘XB023’的脂体达到最大的时间早于‘鲁花9号’, 而‘鲁花9号’的脂体快速积累的时间比‘XB023’长。两品种蛋白体大小都在果针入土40天时达到最大值, ‘XB023’的蛋白体在籽仁发育后期数量增加较快。高蛋白品种较高的蛋白质含量由其子叶细胞中较大蛋白体的大小和较多的蛋白体数量决定, 而高油品种较高的脂肪含量是由其较多的脂体数量决定。  相似文献
