Energy dispersive X-ray fluorescence (ED-XRF) technique has been used for the determination of trace element profile during different developmental stages of somatic embryogenic callus of an economically important medicinal plant, Plantago ovata Forssk. Somatic embryogenesis is a plant tissue culture-based technique, which is used for plant regeneration and crop improvement. In the present investigation, elemental content was analysed using ED-XRF technique during different developmental stages and also determine the effect of additives—casein hydrolysate and coconut water on the trace elemental profile of embryogenic callus tissue of P. ovata. Subsequent experiments showed significant alteration in the concentration of K, Ca, Mn, Fe, Zn, Cu, Br, and Sr in both the embryogenic and non-embryogenic callus. Higher K, Ca, Fe, Cu, and Zn accumulation was in embryogenic tissue stage compared to other stages, suggesting these elements are crucial for successful embryogenesis. The results suggest that this information could be useful for formulating a media for in vitro embryo induction of P. ovata.  相似文献

The elemental composition of rabbit liver was determined by the PIXE and micro-PIXE methods. The mean concentrations of P, S, Cl, K, Fe, Cu, Zn, and Rb measured by both methods were similar. The latter method also allowed for localization of elements within lobule territory. It has been found that some elements are more prevalent in the veins (Cl, Fe) and others in the liver parenchyma (P, Cu, Zn). Moreover, Zn showed the characteristic intralobular distribution. Some methodological aspects of microbeam application to biological materials were also discussed.  相似文献

Heavy metals are known to induce Fe chlorosis in different plantspecies. Heavy-metal-induced chlorosis is generally correlatedwith low plant Fe contents, suggesting effects of heavy metalson Fe mobilization and uptake. Under Fe-deficient conditions,dicotyledonous plants enhance root Fe(lll) reductase activity,thus increasing the capacity to reduce Fe(MI) to Fe(ll), theform in which roots absorb Fe. We studied the effect of severalheavy metals (Mn, Pb, Zn, Mo, Ni, Cu, and Cd) on the inductionof enhanced root Fe(lll) reductase by 11-d-old Fe-deficientcucumber [Cucumis sativus L. cv. Ashley). The effect of theseheavy metals on the function of the induced Fe(lll) reductasewas also investigated. Results showed that some heavy metalscan inhibit both the induction and function of root Fe(lll)reductase. Ni, at 20//M, and Cu and Cd, at 5 fiM concentrationor higher, severely inhibited the induction of root Fe(lll)reductase while Mn, Pb, Zn, and Mo had little effect, even atconcentrations higher than 20 //M. The Cu, Fe and Zn contents in isolated cytochrome c oxidase preparations from heart, liver, diaphragm or kidney of bovine, pig and rat was measured by proton-induced X-ray emission (PIXE). The average Cu/2Fe ratio was 2.73 and Zn/2Fe ratio 0.98. Correspondingly a Cu/Zn ratio of 2.76 was found. Dialysis of the bovine heart enzyme against increasing EDTA concentrations up to 30 mM did not change this result. It is concluded that all isozymes of mammalian cytochrome c oxidase contain 3 Cu, 2 Fe and 1 Zn per monomeric catalytic unit.  相似文献

Proton-induced X-ray emission (PIXE) method was employed to study the concentration of heavy elements in betel leaves, betel nuts, and mineral lime consumed in Bangladesh. The samples were collected from different parts of Bangladesh and analyzed by the thicktarget external beam technique of the PIXE method. The samples were exposed to the proton beam as 1-mm thick pellets and irradiated with 2.0-MeV protons having 20-nA beam intensity. The concentration of some 15 elements (K, Ca, Ti, V, Cr, Mn, Fe, Ni, Cu, Zn, Se, Br, Rb, Sr, and Pb) was measured by comparison with a calibration curve constructed from the NBS orchard leaf standard SRM 1571. The validity of the procedure has been established by comparative measurements of Cu and Zn with atomic absorption spectrophotometry. The significance of the results is discussed in view of their implications in health and disease.  相似文献

A particle-induced X-ray emission (PIXE) analysis method is presented, which allows measurement of eight elements (i.e., K, Ca, Mn, Fe, Cu, Zn, Se, and Rb) in human brain samples of only a few mg dry weight. The precision and accuracy of the method were investigated by analyzing animal brain matter with both PIXE and instrumental neutron activation analysis (INAA). The method was applied to measure the 8 elements in 46 different regions of 3 human brains. The sections analyzed originated from either the left or the right cerebral hemisphere, brain stem, and cerebellum. For one of the brains, sections were also analyzed from 26 corresponding regions of both hemispheres. For all elements, similar concentrations were found in the corresponding areas of the left and right sides of the brain. The concentrations (in μg/g dry weight) of the elements K, Fe, Cu, Zn, Se, and Rb were consistently higher in cortical structures than in white matter. Deep nuclei and brain stem, which have a mixed composition, showed intermediate values for K, Zn, Se, and Rb. A hierarchical cluster analysis indicated that the various brain regions clustered into two large groups, one comprising gray and mixed matter regions and the other, white and mixed matter brain areas.  相似文献

Eight elements (i.e. K, Ca, Mn, Fe, Cu, Zn, Se, and Rb) were measured in 50 different regions of 12 normal human brains by particle-induced X-ray emission (PIXE) analysis. The dry weight concentrations of K, Fe, Cu, Zn, Se, and Rb were consistently higher for gray than for white matter areas. The K, Zn and Se concentrations for the regions of mixed composition and, to some extent, also the Rb concentrations, were intermediate between the gray and white matter values, and they tended to decrease with decreasing neuron density. The mean dry weight concentrations of K, Ca, Zn, Se, and Rb in the various brain regions were highly correlated with the mean wet-to-dry weight ratios of these regions. For Mn, Fe, and Cu, however, such a correlation was not observed, and these elements exhibited elevated levels in several structures of the basal ganglia. For K, Fe, and Se the concentrations seemed to change with age. A hierarchical cluster analysis indicated that the structures clustered into two large groups, one comprising gray and mixed matter regions, the other white and mixed matter areas. Brain structures involved in the same physiological function or morphologically similar regions often conglomerated in a single subcluster.  相似文献

The influence of Fe nutrition on the distribution of the heavy metals Fe, Mn, Zn, and Cu and of the heavy metal chelators nicotianamine (NA) and citrate in 6 different shoot and 3 different root parts and in xylem exudate of a NA-containing tomato wild type and its NA-less mutant was investigated. Under the same Fe supply the mutant showed higher Fe, Mn, and Zn concentrations in all organs investigated, with exception of the shoot apex. The Cu concentration in the mutant was only in root parts higher than in the wild type but much lower in leaves. Analyses of xylem exudate showed that Fe, Mn, and Zn were readily translocated by both genotypes from the roots to the shoot at all levels of Fe supply, whereas in the absence of NA, Cu was only poorly transported. Citrate as main Fe chelator in the xylem was present in high concentrations in xylem exudate of the wild type under low Fe supply but in the mutant also at 10 M FeEDTA. NA occurred in xylem exudate of the wild type in concentrations high enough to chelate heavy metal ions.Generally, high Fe supply induced a decrease of Mn, Cu, and Zn concentrations in all organs of the wild type whereas high concentrations were observed in most cases under Fe deficiency. A positive correlation between Fe supply and NA concentration existed only in the shoot apex and in the xylem exudate of wild type plants. From the correlation between Cu and NA translocation and from the high stability constant of the NA-Cu-complex (log K=18.6) it is concluded that NA is a chelator for Cu in the xylem, whereas the translocation of Fe, Mn, and Zn is independent of NA.  相似文献

Hair trace element (TE) (Cr, Mn, Fe, Zn, Cu, Br, Rb, Sr, Pb) levels from 336 healthy subjects were measured by the Proton-Induced X-ray Emission (PIXE) method. The subjects were divided in three groups: 157 full-term neonates (75 male and 82 female), 86 children (41 male and 45 female) ages 6 to 11 yr, and 93 adolescents (51 male and 42 female) 11 to 16 yr old. Cu, Zn, Cr, and Br show an increase from birth to 8 yr and then decrease. Fe, Mn, and Sr strongly decrease up to 8 yr and then remain almost stable. Sex differences are present in Fe, Zn, and Br of children and in Cu, Cr, and Br of adolescents.  相似文献

Concentrations of 8 trace elements (Fe, Cu, Zn, Se, Br, Pb, Rb, and Sr) at different lactation time were measured by the PIXE multi-elemental technique. Time dependence and interelement correlations were studied. A total of 200 milk samples from 32 lactating mothers were supplied from 2 to 120 d after delivery of 26 full-term and 6 preterm infants. All elements showed a lognormal frequency-distribution. The Fe, Cu, Zn, and Se contents in preterm milk were found to be somewhat different with respect to full-term milk. Cu, Zn, Se, Br, Pb, and Rb concentrations declined with lactation time, both in pre- and full-term samples. Sr and Fe contents did not show any change with time. Detailed analysis of data by partial correlation and multiple regression methods was performed. No substantial differences between preterm and full-term samples were found in the results of partial correlation analysis. Cu and Zn were found to be correlated with lactation time, whereas the measured time dependence for the other elements has to be attributed to the effect of the existing interelement correlation. All the measured elements appeared to be correlated with at least one other element. In particular, Se was inversely correlated with Zn and directly with Cu. The zinc and copper contents in milk can therefore depend on the variation in the mother selenium intake.  相似文献

Airborne metal deposition in the major urban and the industrial districts of Kocaeli was monitored using Xanthoria parietina lichen specimen as a biomonitoring organism. Lichen samples were analyzed for Al, As, Co, Cd, Cu, Fe, Hg, Mn, Ni, Pb, Ti, Tl, V and Zn contents to determine the relationship between the potential pollutant sources in the region and the degree of airborne metal deposition. Results showed that airborne metal deposition in the Kocaeli province was widespread and environmental alteration was serious near the industrial facilities. Mean metal concentrations of lichen samples in the industrial district (Dilovası) of Kocaeli were two to seven folds higher than those in the urban districts of Kocaeli: Mn (7), Pb–Cd–Zn (6), Fe–Ni–Cu (3) and Al–Co–Ti–Hg–As–V (2). Environmental alteration in Dilovası region was severe in terms of all metals analyzed. Cluster analysis showed that metal industry (iron–steel, aluminum, zinc) in Dilovası, fossil fuel combustion processes related to the industry and power plant and heavy traffic contributed significantly to the metal emission in Dilovası region. Airborne metal deposition in the urban districts of Kocaeli was high especially around the coal-fired cement plant in Hereke and petroleum refinery in Körfez. Fossil fuel combustion and traffic emission were among the important sources of airborne metals in the urban–suburban districts.  相似文献

copper, zinc, iron, and other nine trace elements (TEs) in 30 patients with hepatocellular carcinoma (HCC) were determined from the HCC itself and the surrounding liver parenchyma (SLP) using particle induced X-ray emission (PIXE). The result showed that the level of Zn, Fe, Co, Ca, Mn, Rb, and Cl in HCC significantly differed from that in the SLP (P<0.01), while no notable change has been observed in content of Cu (P=0.9). However, in relation to tumor size, an imbalance Cu distribution was observed. The content of Cu present in small HCC (<35 mm in average diameter) was greater than that in SLP (P<0.4), and when HCC progressed to large one (>35 mm in average diameter), hepatic Cu was no longer accumulated. Other elements such as Zn and Fe, showed no change in concentration relative to tumor size. The significance of the critical HCC size where a change from accumulation to loss in Cu contents is seen is also discussed.  相似文献

Proton-induced X-ray emission (PIXE) analysis was employed to determine the concentrations of six elements in the plasma and erythrocytes of 18 cancer subjects (15 males and 3 females) with neoplastic disorders and in 70 controls (35 males and 35 females). It was found that the concentrations of Br, K, and Zn were significantly elevated in the erythrocytes of the cancer subjects compared to the controls, whereas the concentration of Fe was significantly depressed, but with no difference observed in the concentration of Ca. In the plasma, the concentrations of Br, Cu, Ca, and K were significantly elevated, whereas the concentrations of Fe and Zn were found to be significantly depressed compared to the controls.  相似文献

Changes in blood parameters and plasma elements of carp exposed for 14 days to diluted sewage were studied. The levels of elements in small amounts of plasma samples were also determined by PIXE analysis. Carp exposed to diluted sewage increased ammonia, glucose, Mg, Cu and Br, and decreased protein, K, Fe and Zn in plasma. It is concluded that sewage exposure causes considerable stress responses even with sufficient DO in water. PIXE for multielement analysis was also found to be quite useful even for small amounts of samples.  相似文献

于2016年7至10月采用电感耦合等离子体发射光谱法(ICP-OES),测定了内蒙古包头南海子湿地繁殖期过后的白琵鹭(Platalea leucorodia)、苍鹭(Ardea cinerea)和夜鹭(Nycticorax nycticorax)3种鹭鸟初级飞羽及环境因子(水、土壤、食物)中As、Cd、Cr、Cu、Ni、Pb、Zn、Fe、Mn、Hg 10种重金属的含量,采用单因素方差分析方法比较了不同鹭鸟种类羽毛重金属含量差异,并通过生物富集系数及Pearson相关性检验分析了羽毛与环境因子间重金属含量之间的关系,以揭示包头南海子湿地环境中重金属污染现状及生物富集特征。结果表明:(1)被检测的10种重金属中,As、Cd、Cr、Cu、Pb、Zn、Hg 7种元素在湿地环境中均已超标,尤其土壤中Fe、Zn、Cu已达到重度污染的程度。(2)不同重金属元素在鹭鸟羽毛中的含量存在差异,其中Fe元素在白琵鹭羽毛中的含量水平最高(388.77 mg/kg),Cd元素在夜鹭羽毛中的含量最低(0.12 mg/kg)。在鹭鸟羽毛中重金属含量由高至低的顺序分别为,白琵鹭Fe、Zn、Mn、Cu、Hg、Cr、Ni、Pb、As、Cd,苍鹭Zn、Fe、Cu、Cr、Ni、As、Mn、Hg、Pb、Cd,夜鹭Zn、Fe、Mn、Cu、Ni、Pb、Hg、Cr、As、Cd。除Pb和Cd元素外,其他8种元素含量在3种鹭鸟羽毛中的含量种间差异显著。(3)相关分析表明,鹭鸟羽毛中的重金属含量与环境因子中的重金属含量显著相关且呈现富集特征,为此可作为监测当地环境污染的指示性材料。  相似文献

研究了南京城市土壤重金属含量、来源及其与土壤性质的关系。结果表明,南京城市土壤中,Fe、Ni、Co、V污染不明显,但受到了不同程度的Mn、Cr、Cu、Zn、Pb污染,其中:Pb污染非常严重;重金属在土壤剖面分布没有规律性;Fe、Ni、Co、V元素主要来源于原土壤物质,Cu、Zn、Pb、Cr元素主要来源于人为输入,Mn可能在不同的土壤中来源不同;Fe、Cr、Ni、Co、V元素含量之间均呈极显著正相关,Cu、Zn、Pb、Cr元素含量之间均呈极显著正相关。Fe、Co、V、Ni含量与粘粒含量、CEC呈极显著正相关;Cu、Zn、Pb含量与粘粒含量呈极显著负相关;Cu、Zn、Pb、Cr含量与有机碳呈极显著正相关,Pb含量与pH呈极显著正相关。  相似文献

Summary YoungPinus radiata D. Don were fertilized with copper oxychloride and copper sulfate on a random block basis with four replicates. Roots, needles, stems and shoot apices were analyzed separately for Cu, Fe, Mn and Zn contents at different times from fertilization. The results showed a rapid plant copper absorption, no effects being found on the Fe, Mn and Zn content. The dynamic internal distribution of copper among different plant parts is shown. The treatments applied did not cause any phenotypical alteration nor phytotoxicity in the plants, and no significant effect of the fertilizer chemicals on each other.  相似文献
