Malaria remains a major health burden especially for the developing countries. Despite concerted efforts at using the current control tools, such as bed nets, anti malarial drugs and vector control measures, the disease is accountable for close to a million deaths annually. Vaccines have been proposed as a necessary addition to the armamentarium that could work towards elimination and eventual eradication of malaria in view of their historical significance in combating infectious diseases. However, because malaria vaccines would work differently depending on the targeted parasite stage, this review addresses the potential impact various malaria vaccine types could have on transmission. Further, because of the wide variation in the epidemiology of malaria across the endemic regions, this paper proposes that the ideal approach to malaria control ought to be tailor-made depending on the specific context. Finally, it suggests that although it is highly desirable to anticipate and aim for malaria elimination and eventual eradication, many affected regions should prioritize reduction of mortality and morbidity before aspiring for elimination.  相似文献

RNA interference (RNAi) is widely used in Caenorhabditis elegans to identify gene function and has been adapted as a high-throughput screening method to identify genes involved in essential processes. The technique has been applied to parasitic nematodes with variable success and we believe that inconsistent outcomes preclude its use as a robust screen with which to identify potential control targets. In this article, key issues that require clarification are discussed, including the mode of delivery of double-stranded RNA to the parasite, the developmental stage targeted and, perhaps of most importance, whether the RNAi pathway (as defined by studies in C. elegans) is fully functional in some parasitic nematodes.  相似文献

We describe here the tunability of the HNN experiment to obtain certain residue specific peak patterns in the spectra of (15N, 13C) labeled proteins. This is achieved by tuning a band-selective 180° pulse on the carbon channel in the pulse sequence, whereby one can tamper with the Cα–Cβ coupling evolutions for the different residues. Specifically, we generate distinctive peak patterns for serine and threonine and their neighbors in the different planes of the three dimensional spectrum. These provide useful anchor points during sequential assignment of backbone resonances. The performance of this experiment, referred to as HNN-ST here, is demonstrated using two proteins, one properly folded and the other completely denatured. With the availability of high field spectrometers, techniques such as TROSY, and ever increasing sensitivities in the probes, this experiment with its large number of check points has a great potential for rapid and unambiguous backbone resonance assignment in large proteins.  相似文献

Mass extinctions of varying magnitude prune the continuous diversification predicted by Darwinian evolutionary processes. They are caused by events that are too rare to become adaptatively accommodated. Their effects depend not only on the nature and magnitude of the triggering event but also on the state of the biosphere at the particular time. This is most clearly shown by the existence of Golden Ages preceding all Phanerozoic mass extinctions. These coincide with greenhouse periods, in which doomed clades gave rise to heteromorphs, deviating in strange ways from established bauplans. When critically examined, the seemingly ‘decadent’ morphologies of Schindewolf's ‘typolytic stages’ turn out to have been highly functional. The paradoxical link between adaptive peaks and evolutionary failure can now be explained. Specialisation tends to increase vulnerability (1) by narrowing niches and (2) by the retention of clade-specific conservative features that happen to become fatal Achilles’ Heels for entire clades in the face of a particular perturbation. Following extinctions, the availability of open niches favoured relatively rapid diversification of more innovative clades and their rise to ecological dominance (Schindewolf's ‘typogenetic stage’). Although the long-term changes can be observed only in the fossil record, Golden Biotopes in the present biosphere show that the Darwinian process may also be promoted by ecological isolation. As a result, clade histories do resemble individual biographies, but for ecological rather than orthogenetic reasons. This insight may help us to deal with the present mass extinction caused by our own species.  相似文献

Hundreds of studies have assessed variation in the degree to which people experience disgust toward substances associated with pathogens, but little is known about the mechanistic sources of this variation. The current investigation uses olfactory perception and threshold methods to test whether it is apparent at the cue-detection level, at the cue-interpretation level, or both. It further tests whether relations between disgust sensitivity and olfactory perception are specific to odors associated with pathogens. Two studies (N's = 119 and 160) of individuals sampled from a Dutch university each revealed that pathogen disgust sensitivity relates to valence perceptions of odors found in pathogen sources, but not to valence perceptions of odors not associated with pathogens, nor to intensity perceptions of odors of either type. Study 2, which also assessed olfactory thresholds via a three-alternative forced-choice staircase method, did not reveal a relation between pathogen disgust sensitivity and the ability to detect an odor associated with pathogens, nor an odor not associated with pathogens. In total, results are consistent with the idea that pathogen disgust sensitivity relates to how olfactory pathogen cues are interpreted after detection, but not necessarily to the ability to detect such cues.  相似文献
