Transgenic plants offer many advantages, including low cost of production (by elimination of fermenters), storage and transportation; heat stability; and absence of human pathogens. When therapeutic proteins are orally delivered, plant cells protect antigens in the stomach through bioencapsulation and eliminate the need for expensive purification and sterile injections, in addition to development of both systemic and mucosal immunity. Chloroplast genetic engineering offers several advantages, including high levels of transgene expression, transgene containment via maternal inheritance and multi-gene expression in a single transformation event. Hyper-expression of vaccine antigens against cholera, tetanus, anthrax, plague or canine parvovirus (4-31% of total soluble protein, tsp) in transgenic chloroplasts (leaves) or non-green plastids (carrots, tomato), as well as the availability of antibiotic-free selectable markers or the ability to excise selectable marker genes, facilitate oral delivery. Hyper-expression of several therapeutic proteins, including human serum albumin (11.1% tsp), somatotropin (7% tsp), interferon-gamma (6% tsp), anti-microbial peptide (21.5% tsp), facilitates efficient and economic purification. Also, the presence of chaperones and enzymes in chloroplasts facilitate assembly of complex multi-subunit proteins and correct folding of human blood proteins with proper disulfide bonds. Functionality of chloroplast-derived vaccine antigens and therapeutic proteins has been demonstrated by several assays, including the macrophage lysis assay, GM1-ganglioside binding assay, protection of HeLa cells or human lung carcinoma cells against encephalomyocarditis virus, systemic immune response, protection against pathogen challenge, and growth or inhibition of cell cultures. Thus, transgenic chloroplasts are ideal bioreactors for production of functional human and animal therapeutic proteins in an environmentally friendly manner.  相似文献

The drought tolerance of the commercial apple ( Malus domestica Borkh.) rootstocks M9, M26, M27 and MM111, and some new selections from the rootstock breeding programme at HRI-East Malling (AR69-7, AR295-6, AR360-19, AR486-1 and AR628-2), was assessed using potted, glasshouse-grown, unworked rootstocks. After an initial period of growth under well-watered conditions the amount of irrigation was gradually reduced, for some treatments, to simulate natural drying in the soil. At the end of a six-month growth period, the rootstocks were harvested and the production of dry matter and its partitioning to various plant parts determined. The rootstocks exhibited large differences in shoot and root dry matter, and root length but not all the rootstocks showed declines in root mass or length in response to the droughting treatment. The dwarfing rootstocks tended to have smaller amounts of both coarse (>2 mm diameter) and fine roots (<2 mm diameter), than the more vigorous rootstocks. Irrespective of rootstock or irrigation treatment there was a close linear relationship between coarse and fine root. There was also no change in the length/weight relationship for fine roots irrespective of rootstock or irrigation treatment, i.e. 42 m of fine root weighed 1 g dry weight. In some cases the amount of root produced could be directly correlated with the rootstock known potential to control scion vigour, but this was not true for all the rootstocks examined. The absence of this relationship was particularly evident in some of the new selections of rootstock. The possible causes for these differences, compared with commercially used rootstocks, is discussed in relation to the origin and parentage of the rootstock selections. Despite this lack of a root length/vigour relationship, the amount of dry matter partitioned to shoot growth reflected the rootstocks' known vigour. The different responses of these rootstocks to drought are discussed along with their implications for understanding the mechanisms by which rootstocks are thought to dwarf scion shoots.  相似文献

During the past two decades, antibodies, antibody derivatives and vaccines have been developed for therapeutic and diagnostic applications in human and veterinary medicine. Numerous species of dicot and monocot plants have been genetically modified to produce antibodies or vaccines, and a number of diverse transformation methods and strategies to enhance the accumulation of the pharmaceutical proteins are now available. Veterinary applications are the specific focus of this article, in particular for pathogenic viruses, bacteria and eukaryotic parasites. We focus on the advantages and remaining challenges of plant-based therapeutic proteins for veterinary applications with emphasis on expression platforms, technologies and economic considerations.  相似文献

Background

Highly pathogenic influenza viruses pose a constant threat which could lead to a global pandemic. Vaccination remains the principal measure to reduce morbidity and mortality from such pandemics. The availability and surging demand for pandemic vaccines needs to be addressed in the preparedness plans. This study presents an improved high-yield manufacturing process for the inactivated influenza H5N1 vaccines using Madin-Darby canine kidney (MDCK) cells grown in a serum-free (SF) medium microcarrier cell culture system.

Principal Finding

The current study has evaluated the performance of cell adaptation switched from serum-containing (SC) medium to several commercial SF media. The selected SF medium was further evaluated in various bioreactor culture systems for process scale-up evaluation. No significant difference was found in the cell growth in different sizes of bioreactors studied. In the 7.5 L bioreactor runs, the cell concentration reached to 2.3×106 cells/mL after 5 days. The maximum virus titers of 1024 Hemagglutinin (HA) units/50 µL and 7.1±0.3×108 pfu/mL were obtained after 3 days infection. The concentration of HA antigen as determined by SRID was found to be 14.1 µg/mL which was higher than those obtained from the SC medium. A mouse immunogenicity study showed that the formalin-inactivated purified SF vaccine candidate formulated with alum adjuvant could induce protective level of virus neutralization titers similar to those obtained from the SC medium. In addition, the H5N1 viruses produced from either SC or SF media showed the same antigenic reactivity with the NIBRG14 standard antisera.

Conclusions

The advantages of this SF cell-based manufacturing process could reduce the animal serum contamination, the cost and lot-to-lot variation of SC medium production. This study provides useful information to manufacturers that are planning to use SF medium for cell-based influenza vaccine production.  相似文献

GSK Biologicals has been involved in the production of Polio vaccine since the early start of Polio vaccination, beginning with the first generation of Inactivated Polio Vaccine (IPV). Over time, the company has developed solid industrial experience and knowledge that significantly contributes today to the quality of our Polio vaccines. GSK Biologicals' current IPV is now routinely produced according to the process defined by Van Wezel (RIVM) in the late seventies, using Vero cells and micro-carrier technology in bioreactors. In addition to compliance with current requirements (World Health Organization, European Pharmacopoeia, Code of Federal Regulations USA), the quality of the routine vaccine is guaranteed by numerous additional data related to the characterization, to the consistency, and to the validation of the process and the testing. This supplementary data package will allow, for instance, for the application of the in vitro potency testing for routine release instead of the in vivo testing. The present views on the Polio vaccine strategy for the post eradication era have portrayed a very limited role for the current IPV. The main reasons relate to post-eradication bio-containment needs and to production capacity and costs. A reevaluation of the classic approach taken to the use of the current IPV produced from wild type polio strains positions this vaccine as a real alternative to other strategies, allowing us to take advantage of the excellent performance of IPV over many years.  相似文献

Summary A large amount of high-molecular-mass anthocyanin pigment was produced by fermentation of malt grains without steaming, in which the malts were used as raw materials. The formation of a purple pigment from green malt was the most prominent from all malt grains tested. The precursor of the purple pigment was presumed to be a polyphenol (proanthocyanidin). When malt was used as the raw material, it allowed the production of pigment without using the glucoamylase preparation that is used for the fermentation of barley bran. In addition, the pigment was produced even from dry malt that had been treated with heat (80°C).  相似文献

Maize plants were grown singly in pots in a population of 3.7 plants m-2, and 98 days after sowing (shortly after flowering) were arranged into three populations, 1.27 (W), 3.70 (M), and 6.15 (C) plants m-2, respectively. The plants were harvested 131 days after sowing, about 4 wk before the normal time of maturity, because some plants at the closest spacing began dying. Increment of dry weight between days 98 and 131 was 373, 243, and 108 g/plant and grain dry weight at harvest was 218, 220 and 195 g/plant in populations W, M and C, respectively. Thus, dry weight of parts of the plant other than grain increased in population W, remained about the same in population M, and decreased in population C between days 98 and 131. Number of kernels per plant was c. 540 in all three populations. On day 98, immediately after rearrangement, leaf area index (L) was 2.0, 5.8 and 9.7 in populations W, M and C, respectively. By day 131 L had decreased to 6.7 in population C, but had hardly changed in the other two populations. Crop growth rate between days 98 and 131 was 101, 191 and 141 g m-2 wk-1, and grain dry wt per unit of land on day 131 was 277, 816 and 1196 g m-2, in populations W, M and C, respectively.  相似文献

This study investigates the production of alginate microcapsules, which have been coated with the polysaccharide chitosan, and evaluates some of their properties with the intention of improving the gastrointestinal viability of a probiotic ( Bifidobacterium breve ) by encapsulation in this system. The microcapsules were dried by a variety of methods, and the most suitable was chosen. The work described in this Article is the first report detailing the effects of drying on the properties of these microcapsules and the viability of the bacteria within relative to wet microcapsules. The pH range over which chitosan and alginate form polyelectrolyte complexes was explored by spectrophotometry, and this extended into swelling studies on the microcapsules over a range of pHs associated with the gastrointestinal tract. It was shown that chitosan stabilizes the alginate microcapsules at pHs above 3, extending the stability of the capsules under these conditions. The effect of chitosan exposure time on the coating thickness was investigated for the first time by confocal laser scanning microscopy, and its penetration into the alginate matrix was shown to be particularly slow. Coating with chitosan was found to increase the survival of B. breve in simulated gastric fluid as well as prolong its release upon exposure to intestinal pH.  相似文献

Very limited information regarding fine-root growth and production of tropical dry forests is available. Fine roots and small roots are defined as rootlets with diameters < 1 mm and 1.1 to 5 mm, respectively. Live and dead fine-and small-root mass fluctuations were studied over one year by means of soil core analyses in the deciduous dry forest of Chamela, Mexico, at 19° 30, 2 km inland from the Pacific Ocean. By means of systematically varying the distance of soil core extraction points from tree stems, it was shown that random core collection is justified. A difference between fine-root biomass on south and north facing slopes was documented, although this difference was significant only during the rainy season. The live/dead ratio of fine roots was highest during the rainy period. The annual fine-root production for 1989 was estimated at 4.23 Mg ha-1 by summing significant fine-root biomass changes between sampling dates. This value is higher than most of the comparable data from other ecosystems.  相似文献
