Stem base rot of Dieffenbachia maculata caused by Phytophthora mexicana During the last years a stem rot of Dieffenbachia maculata (Lodd.) G. Don has been observed in greenhouses of several commercial nurseries, located in different parts of the Federal Republic of Germany. The disease partly caused heavy losses. From all diseased plant parts (stem base, petiole and root) the same fungus could be isolated. It was diagnosed as Phytophthora mexicana Hots, et Hart., which is closely related to P. palmivora (Butl.) Butl. Apparently the former species is recorded for the first time as the causal organism of a Dieffenbachia stem rot and also for the first time in Europe. It is supposed that the fungus was imported to Germany via the Netherlands because all plants had been bought there, which diseased later.  相似文献

Biochemical profiles on API Rapid CH* strips and protein profiles on polyacrylamide gels in the presence of sodium dodecyl sulfate were used to distinguish two strains of the entomopathogenic fungusBeauveria bassiana (Balsamo) Vuillemin, ARSEF 2991 and ATCC 44860. Next, the toxicity of these two strains was determined at concentrations of 102, 104, 106 and 108 blastospores/ml on larvae of the Colorado potato beetleLeptinotarsa decemlineata Say (Coleoptera: Chrysomelidae) and of its predator, the spotted ladybird beetle,Coleomegilla maculata lengi Timberlake (Coleoptera: Coccinellidae). Both strains were highly toxic toL. decemlineata larvae. However, the two strains exhibited different levels of toxicity forC. maculata larvae: ARSEF 2991 was toxic, whereas ATCC 44860 caused little coccinellid larval mortality.

The objective of this work was to study the role of the phytochromes (phy) B, D and E in the thermoperiodic control of elongation and flowering time in Arabidopsis thaliana. WT, and phyB, phyD and phyE single mutants, and phyB phyD and phyB phyE double mutants, were grown under day/night temperatures (DT/NT) of 12/22°C, 17/17°C or 22/12°C (negative, zero and positive DIF, respectively) for inflorescence stem length measurements, and under DT/NT 17/25°C or 25/17°C (negative and positive DIF, respectively) for leaf morphology and flowering time measurements. In WT final length of the stem, petiole and leaf blade were longer under positive DIF compared to negative DIF. The temperature effect was stronger in the leaf petiole than the stem, whereas only a slight change was seen in the leaf blade length direction and none in the width direction. The temperature effect on stem and petiole elongation was reduced or nearly eliminated in the genotypes lacking phyB, while a phyD or a phyE mutation had no influence or a slightly positive influence on the temperature effect, respectively. These results suggest that phyB, and not phyD or phyE, is needed for a complete thermoperiodic control of elongation growth in A. thaliana. For all genotypes tested, plants flowered earlier at negative DIF than positive DIF, suggesting that none of the three phytochromes B, D, or E is needed for a thermoperiodic control of flowering time in A. thaliana.  相似文献

The disease control efficacy of quarantine heat treatments developed for fruit fly disinfestation in mangoes cv. Kensington Pride was evaluated in this study. Heat was applied using high humidity (>95% r.h.) hot air (HHHA) at temperatures ranging from 47–49°C. Anthracnose, caused by Colletotrichum gloeosporioides, was well controlled in mangoes heated to a core temperature of 46°C, 47°C or 48°C for 24, 10 or 8 min respectively, prior to ripening at 23°C for 16 days. Stem end rot, caused by Dothiorella dominicana and Lasiodiplodia theobromae, was not satisfactorily controlled by these treatments. In a subsequent experiment, fruit were immersed in a hot benomyl (0.5 g a.i. litre“1 at 52°C for 5 min) or unheated prochloraz (0.25 ml a.i. litre1 at 28°C for 30 s) dip before or after the application of HHHA (core temperature of 47°C for 10 min). During storage at 23°C for 15 days, the incidence of stem end rot was reduced by HHHA alone, although immersion in hot benomyl either before or after HHHA treatment greatly improved stem end rot control. HHHA treatment (core temperature of 46.5°C for 10 min) alone reduced the incidence of anthracnose in mangoes stored at 13°C for 14 days prior to ripening at 22°C, although a combination treatment consisting of HHHA and either hot benomyl or unheated prochloraz gave complete control of anthracnose under these storage conditions. HHHA treatment alone gave no control of stem end rot in mangoes stored at 13°C prior to ripening at 22°C. A supplementary hot benomyl treatment was required for acceptable control of this disease in cool-stored mangoes. The development of yellow skin colour in fruit was accelerated by HHHA treatment.  相似文献

To clarify the effects of peripheral herbal plants onGlehnia littoralis growth in coastal sand dunes, the morphology of their aboveground portions was surveyed in five communities:Carex kobomugi, Calystegia soldanella, Ischaemum anthephoroides, Oenothera biennis, andElymus mollis. Correlation coefficients (CC) were generally significant at the 1% level between community properties [total aboveground biomass (B) and height (H) of dominant species per unit area] and those ofG. littoralis [leaf number (Nl), petiole angle (Anp), petiole length (Lp), petiole weight (Wp), Lp/Wp, Lp/weight of leaf blade (Wb), Wp/total weight (Wt), specific leaf area (SLA), stem length (Ls), and Ls/weight of stem (Ws)J The exceptions were among four pairings: B and NI, B and Wt, H and Nl, and H and Wt. Of the two community properties, biomass had the greatest association with leaf properties while H was most closely related to those of the stems. Petiole angle increased along with leaf order, from 0° to 42° for the C.kobomugi community, from 5° to 55° forCalystegia soldanella, from 49° to 74° forI. anthephoroides, from 54° to 80° forO. biennis, and from 75° to 85° forE. mollis. In all communities, the properties of Wp, SLA, and Wb increased up to the third or fourth leaf, but then decreased; the exception was for Lp/Wp, which was the reverse. Leaf order of the largest one moved from first position to third as either B or H increased in a community.  相似文献

Spontaneous and induced blocking and unblocking of phloem transport   总被引:1,自引:1,他引:0  
Abstract Steady-state labelling with 11CO2 was used to observe the blocking of phloem transport, induced by chilling short regions of stems or petioles of velvetleaf (Abutilon theophrasti Medic.) or cotton (Gossipium hirsutum L.). The abruptness of these blockages was evidenced by sharp decreases in 11C activity below, and increases above a 2 to 3 cm region cooled from 28°C to 18 or 13°C for periods as short as 2 min. Abrupt unblocking of transport in velvetleaf occurred a few minutes after rewarming, as evidenced by a sharp rise and and overshoot in 11C activity. Recovery of transport in cotton was more prolonged and was marked by occasional spontaneous blocking and unblocking of transport at various points along the petiole or stem, not necessarily in the cooled region. Similar spontaneous events were often observed in undisturbed cotton plants, but only rarely in velvetleaf.  相似文献

Ralstonia solanacearum (RS) race 1 biovar 4 (R1bv4), causal agent of bacterial wilt in vegetable sweet potato (VSP), is often latent in VSP vines and is important in introduction of the pathogen to newly planted fields. In this study, the effects of biological and environmental factors on the distribution and multiplication of R1bv4 in VSP tissues were examined. Based on stem-injection inoculation, the R1bv4strain of NC01 could cause 49.0% and 33.0% wilting on VSP cultivars TN71 and WS, respectively. The populations of NC01 in diseased TN71 and WS were 108–109 cfu/g tissue at 28th day after inoculation. On the other hand, the R1bv4 could not cause symptom in cultivars of TN57 and VSPSL-1 vine and the NC01 was confined to near the injection sites. Temperature tests indicated that NC01 could cause 28.0% and 14.0% wilting on cultivar TN71 at 28 and 20°C, respectively. Moreover, the populations of NC01in diseased plants were 1.6 × 109 and 7.9 × 108 cfu/g tissue at 28 and 20°C, respectively. The distribution of NC01 in VSP stem indicated that the isolation frequency of NC01 was lower than 31.0% in terminal shoots or erect stems and 45.0 to 100.0% in creeping stem after 8 wks planted in infested soil (106 cfu/g soil). The results demonstrated that terminal shoots or erect stems were not common carrier for transmitting R1bv4. Furthermore, two R1bv4 strains, NC01 and HsinT01, were examined the ability for latent infection on cv. TN71. The results revealed that NC01 and HsinT01 showed different ability of latent infection on cultivar TN71. NC01 had lower percentage (46.8% and 45.1%) than HsinT01 (93.4% and 75.3%) at 20 and 28°C.  相似文献

The reference strains of Enterococcus seriolicida (ATCC 49156T) (T = type strain) and of Lactococcus garvieae (ATCC 43921T) and 30 field strains of Gram-positive cocci isolated from diseased rainbow trout in Italy were found to be phenotypically (API 20 STREPT and API 50 CH) and genetically (DNA-DNA hybridization) similar. The high DNA-DNA homologies (70–100%) and the low ΔTm(e) (less than 1.1°C) among these strains showed that Enterococcus seriolicida and Lactococcus garvieae are synonyms, describing a single bacterial species. E. seriolicida strains should be classified as L. garvieae, which must be considered as a major pathogen of freshwater and salt water fish with a world-wide distribution.  相似文献

API 50 CH fermentation profiles of 45 Lactobacillus, one Atopobium, and three Weissella strains incubated at 30°C and 37°C were evaluated. Atopobium uli and ten species of Lactobacillus showed stable patterns despite the change in temperature. The rest of the type strains showed discrepancy between the two incubation temperatures: 18 strains lost, 12 additionally fermented another sugar, and 7 others fermented a different one in lieu. The variation was maximum in L. delbrueckii subsp. delbrueckii. L. malefermentans failed to ferment any of the substrates at 37°C. Majority of the food and plant-associated strains (mainly heterofermenters) retained distinctive traits at 30°C, while most of the animal-associated strains (mostly homofermenters) did so at 37°C. No general trend was observed; 30°C appeared to promote heterofermentation, while 37°C favored homofermenters. Use of API 50 CH profiles for taxonomic purpose in most lactobacilli appears reproducible if a specific temperature for a species is strictly followed. Highland bananas are an important staple food in East Africa, but there is little information on their physiology and growth patterns. This makes it difficult to identify opportunities for yield improvement. We studied allometric relationships by evaluating different phenological stages of highland banana growth for use in growth assessment, understanding banana crop physiology and yield prediction. Pared corms of uniform size (cv. Kisansa) were planted in a pest‐free field in Kawanda (central Uganda), supplied with fertilizers and irrigated during dry periods. In addition, tissue‐cultured plants (cv. Kisansa) were planted in an adjacent field and in Ntungamo (southwest Uganda), with various nutrient addition treatments (of N, P, K, Mg, S, Zn, B and Mo). Plant height, girth at base, number of functional leaves and phenological stages were monitored monthly. Destructive sampling allowed derivation of allometric relationships to describe leaf area and biomass distribution in plants throughout the growth cycle. Individual leaf area was estimated as LA (m2) = length (m) × maximum lamina width (m) × 0.68. Total plant leaf area (TLA) was estimated as the product of the measured middle leaf area (MLA) and the number of functional leaves. MLA was estimated as MLA (m2) = ?0.404 + 0.381 height (m) + 0.411 girth (m). A light extinction coefficient (k = 0.7) was estimated from photosynthetically active radiation measurements in a 1.0 m grid over the entire day. The dominant dry matter (DM) sinks changed from leaves at 1118 °C days (47% of DM) and 1518 °C days (46% of DM), to the stem at 2125 °C days (43% of DM) and 3383 °C days (58% of DM), and finally to the bunch at harvest (4326 °C days) with 53% of DM. The allometric relationship between above‐ground biomass (AGB in kg DM) and girth (cm) during the vegetative phase followed a power function, AGB = 0.0001 (girth)2.35 (R2 = 0.99), but followed exponential functions at flowering, AGB = 0.325 e0.036(girth) (R2 = 0.79) and at harvest, AGB = 0.069 e0.068(girth) (R2 = 0.96). Girth at flowering was a good parameter for predicting yields with R2 = 0.7 (cv. Mbwazirume) and R2 = 0.57 (cv. Kisansa) obtained between actual and predicted bunch weights. This article shows that allometric relationship can be derived and used to assess biomass production and for developing banana growth models, which can help breeders and agronomists to further exploit the crop's potential.  相似文献

Seeking new yeast strains having the ability to protect apple fruits against blue mould for a long time under different storage conditions was the main goal of this work. Based on the in vitro test, yeast strains KKUY0017 and KKUY0051 were selected as the most effective antagonists against Penicillium expansum. Sequencing of 26S rDNA of both yeasts confirmed that the identity of KKUY0017 and KKUY0051 was Cryptococcus albidus and Wickerhamomyces anomalus, respectively. The two strains protected the apple fruits from the blue mould disease under a wide range of temperature (5–30°C); however, W. anomalus KKUY0051 was more effective. At 25°C, W. anomalus KKUY0051 involved in the reduction of disease severity and disease incidence of blue mould by 56.49% and 57.78%, respectively. When either of the two yeasts was applied in concentration of 108 or 109 cells/mL, the maximum reduction in disease severity and disease incidence was achieved. Under cold storage (5°C), both yeast strains succeeded to protect the apple fruits free from the infection up to 24 days. Electron micrograph showed a fit attachment between the cells of C. albidus KKUY0017 and the fungal hyphae leading to the degrading of the hyphae; however, W. anomalus killed the fungal hyphae without direct attachment to them. Gas chromatography–mass spectrometry analysis of the cell-free extract of W. anomalus KKUY0051 revealed the presence of toxic compounds such as the nitrophenol derivatives. The results support the assumption that the main mode of action of this yeast is by killer toxins. We conclude that application of these yeasts under cold storage condition could keep the apple fruits free from blue mould infection for a long time.  相似文献

Pathogenicity of Sirodesmin-deficient Mutants of Phoma lingam   总被引:1,自引:0,他引:1  
Sirodesmin-deficient mutants were produced from aggressive isolates of Phoma lingam by UV-mutagenesis. The mutation rates were 7.1 × 10-3 for mutants expressing a 100-fold reduction of sirodesmin PL in liquid cultures and 1.8 × 10-3 for mutants with a 1000-fold reduction. The low ability of the mutants to produce sirodesmins was also apparent in planta after infection of cotyledons or the stem basis of Brassica napus. Growth rates under in vitro conditions, colony morphology, mating types and the formation of pycnidia and asci were not affected by the mutation. The mutants had the same ability as the wild types to infect cotyledons of the susceptible B. napus cv. Lirabon and to cause a typical grey-green tissue collapse. On the stem basis of the cv. Cobra, however, mutants caused significantly smaller lesions. These results indicate that phytotoxic sirodesmins produced by P. lingam have no basic role in pathogenesis and apparently are not involved in the virulence on cotyledons. However, it cannot be excluded that the compounds act as virulence factors on the stem base of B. napus.  相似文献

Eighty-five strains of bacteria were screened for selection of microorganisms suitable for industrial production of polynucleotides. Among these bacteria, Achromobacter sp. KR 170-4 (ATCC 21942) was found to be rich in polynucleotide Phosphorylase (PNPase) in its “salt-shockate” as compared with the other strains tested. PNPase was purified about 50-fold from the “salt-shockate” of Achromobacter sp. KR 170-4, and properties of the enzyme were elucidated. Optimal pH for reaction was 10.1. Stable pH range at 37°C was between pH 6.5 and 10.5. Optimal temperatures were 46°C for polymerization of ADP or IDP, and 43°C for CDP or UDP. The enzyme was stable below 55°C at pH 9.2. The enzyme required Mn2+ rather than Mg2+ unlike the other PNPases reported. Optimal concentration of Mn2+ was 6 mM.  相似文献

When potato seed tubers (Solanum tuberosum cv. Pentland Javelin) were stored in darkness or diffuse daylight at 9°C and transferred at intervals to conditions suitable for sprouting, their capacity for sprout growth was unaffected by the presence or absence of light during previous storage. When similar tubers were stored at 10°C, 18°C or 25°C, sprout growth commenced earliest at 25°C, but the date was unaffected by fluorescent light. It was concluded that light did not affect the length of the dormant period, but only the rate of sprout elongation after that period had ceased. When tubers with growing sprouts at 10°C or 18°C were transferred from darkness into fluorescent light, sprout growth virtually ceased. Transfer from light into darkness resulted in immediate sprout growth, at a rate comparable with tubers stored continuously in the dark. Tubers of three Peruvian cultivars, stored in farm-scale diffuse-daylight stores, grew progressively shorter sprouts with increasing daily exposure to light from 30 min to 12 h. Storage of cv. Wilja under 21 Wm-2 (total) of white fluorescent light for 10 h per day maintained the sprouts at the same length as ten times this light intensity for 1 h per day. In a subsequent experiment with cv. Bintje the 10 h, low-intensity light regime gave slightly shorter sprouts. It appeared that the total light energy falling on the tubers was the dominant factor controlling sprout growth.  相似文献

Maize (Zea mays L.) seedlings of two cultivars (cv. Bastion adapted to W. Europe, and cv. Batan 8686 adapted to the highlands of Mexico), raised in a glasshouse (19–25 °C), were transferred to 4.5 or 9 °C at photon flux density (PPFD) of 950 μmol m−2 s−1 with 10-h photoperiod for 58 h and then allowed to recover at 22 °C for 16 h (14 h dark and 2 h at PPFD of 180 μmol m−2 s−1). The ultrastructural responses after 4 h or 26 h at 4.5 °C were the disappearance of starch grains in the bundle sheath chloroplasts and the contraction of intrathylakoid spaces in stromal thylakoids of the mesophyll chloroplasts. At this time, bundle sheath chloroplasts of cv. Batan 8686 formed peripheral reticulum. Prolonged stress at 4.5 °C (50 h) caused plastid swelling and the dilation of intrathylakoid spaces, mainly in mesophyll chloroplasts. Bundle sheath chloroplasts of cv. Batan 8686 seedlings appeared well preserved in shape and structure. Batan 8686 had also higher net photosynthetic rates during chilling and recovery than Bastion. Extended leaf photobleaching developed during the recovery period after chilling at 4.5 °C. This was associated with collapsed chloroplast envelopes, disintegrated chloroplasts and very poor staining.  相似文献

Abstract Isolated leaves of Plectranthus fruticosus were grown in cubic plastic cuvettes, and were supplied via their cut petioles with nutrient solution and indole-3-acetic acid (10?6m ). Holes bored in the cuvette walls allowed the petioles to be oriented at approximately 60°, 90° or 120° to the vertical. Growth of the leaves initially oriented at angles of 60° and 90°, which simulated the situation in the intact plant, did not result in epinastic bending of the petiole. Inversion of the leaves (adaxial surface of the petiole downwards) and orientation of the adaxial/abaxial surfaces of the horizontal petiole parallel to the gravity vector, however, yielded strong epinastic bending of the petioles. In the latter case, this bending was not in the direction of the gravity vector (evidence for point (iii), below). Furthermore, epinastic bending occurred, when the isolated leaves were rotated on a clinostat (petioles parallel to the rotation axis or inclined to the rotation axis at an angle of 30°; 3 r.p.m.). Since a possible influence of the shoot was excluded, it is concluded that (i) perception and response are restricted to the leaf, (ii) gravity alone is sufficient to induce epinasty, (iii) a gravitropic component of the response can probably be excluded. The clinostat induced epinasty may not have been caused by nullifying the effect of gravity but due to continuous gravistimulation of the leaf.  相似文献

A collection of Pseudomonas strains was isolated in different regions of Tunisia in the period 2016–2017 from the fruits and leaves of Citrus sinensis cv. ‘Valencia Late’ and Citrus limon cv. ‘Eureka’ plants with symptoms of blast and black pit disease. A phylogenetic analysis of the housekeeping gene rpoD was used for strain identification at the species level. The results demonstrated the affiliation of these strains with the genus Pseudomonas and revealed the presence of 11 strains representing two putative new species in two monophyletic branches. These strains were analyzed morphologically and genotypically by multilocus sequence analyses of the rpoD, gyrB and 16S rRNA (rrs) gene sequences, and their phenotypic characteristics by API 20NE and Biolog GEN III. Plant pathogenic properties were confirmed on fruits and detached leaves of C. limon cv. ‘Eureka’. Fatty acids and WC MALDI-TOF MS major protein profiles were determined. The genomes of both representatives were sequenced. The average nucleotide index and genome-to-genome distance from KC12T and E10BT are below the cut-off established for a described species. These results support the conclusion that the strains KC12T, KC17, KC20, KC22, KC24A, KC25 and KC26 represent a novel species of Pseudomonas, for which the name of Pseudomonas kairouanensis is proposed. The type strain is KC12T (=CECT9766 and CFBP 8662). The strains E10BT, E10AB, E10CB1 and Iy3BA represent another novel species of Pseudomonas for which the name of Pseudomonas nabeulensis is proposed; the type strain is E10BT (=CECT9765 and CFBP 8661).  相似文献

Six commercial carnation cultivars were inoculated with Fusarium oxysporum f. sp. dianthi race 2, and grown under three different temperature regimes. Colonization by the pathogen and development of wilt symptoms were assessed at intervals up to 40 days. No symptoms and very little colonization were seen in any of the cultivars at 14–15°C. At a temperature of 22°C, the cultivars were clearly differentiated into three groups: those with resistance, partial resistance or susceptibility to the pathogen depending on the severity of symptoms and the extent of fungal colonization. Symptom severity was associated with the extent of colonization. This differentiation was not seen at 26°C, when all cultivars except the most resistant, cv.‘Carrier 929′, rapidly became diseased and died by 23 days after inoculation. ‘Carrier 929’ also showed some wilt symptoms at this temperature and was colonized throughout the height of the stem after 40 days. The pathogen caused disease at 26°C by a combination of vascular wilting and stem base and root rotting. Fungal colonization was assayed at 22°C by the dilution plate/homogenization method and by estimation of fungal chitin in a highly resistant (‘Carrier 929′) and in a highly susceptible (‘Red Baron’) cultivar. Both methods of assay gave similar results. In ‘Red Baron’, colonization increased slowly up to 20 days after inoculation then progressed rapidly, closely following the development of severe wilt symptoms. In ‘Carrier 929’, colonization remained very low. The low level of fungal biomass in ‘Carrier 929’ compared with ‘Red Baron’ indicated that the former cultivar showed true resistance as opposed to tolerance to the disease.  相似文献

Summary The infection of root hairs and nodulation ofT. subterraneum L. cv. Cranmore byRhizobium trifolii strain TA1 was extremely sensitive to root temperature. Within the range 11°–19°C infection was progressively delayed as root temperature decreased but at 7°C no root hairs were infected by 40 days. At 7°C lateral root formation was stimulated in the root zone where nodules formed at 19°C.  相似文献
