We conducted phylogenetic analyses of the sect. Caprini and its closely related sections within Astragalus. Analyses of a combined dataset including nrDNA ETS and three cpDNA markers using maximum parsimony and Bayesian inference from 44 species of sect. Caprini and its allied taxa yielded congruent relationships among several major lineages. These results largely disagree with previously recognized taxonomic groups, most notably in the following ways: (1) subsects. Caprini and Purpurascentes of sect. Caprini are not natural groups; (2) sects. Alopecuroidei and Laxiflori are nested within sect. Astragalus; and (3) subsect. Chronopus constitutes a separate phylogenetic lineage. Representatives of sects. Astragalus, Alopecuroidei, and Laxiflori share a common ancestor with that of sect. Caprini. Our studies indicate that Astragalus annularis is an outlier species for the genus Astragalus and sect. Caraganella is the first-diverging clade within the genus Astragalus. Results of these analyses are supported by morphology and suggest the need for new taxonomic delimitations, which are forthcoming. Key morphological characters were mapped onto the phylogenetic tree and discussed.  相似文献

Astragalus is with nearly 3000 described species the largest genus of flowering plants. So far analyses of pollen characters have only been conducted for a few species of the groups within the genus. Here we analyse pollen grains of 22 species representative for Astragalus section Hymenostegis using scanning electron microscopy. We found the basic shape of the pollen grains to be oblate-spheroidal and apertures to be tricolpate as for other eudicots. The sculpturing pattern of the exine is micro-reticulate. Pollen grains show low morphological variation among different species of this section, but differences occur between sections of the genus. We conclude that the vast morphological differentiation that occurred during the rapid radiation of section Hymenostegis was not accompanied by comparable differentiation in pollen morphology.  相似文献

采用ISSR分子标记技术对蒙古高原黄耆属属下部分争议类群的亲缘关系进行了研究。用11条ISSR引物对8个种的45份材料进行扩增,共获得条带188条,其中多态性条带178条,多态性条带百分率达94.7%。用NTSYSpc2.10e软件对相关数据进行UPGMA聚类分析,在相似性系数为0.73处,45份材料被划分为7大类群,其中,科布尔黄耆(Astragalus koburensis)与乳白花黄耆(A.galactites)聚为一支,表明前者可能仅是后者的生态型,应当处理为后者的异名;不同产区的糙叶黄耆(A.scaberrimus)和变异黄耆(A.variabilis)分别聚为一支,其各自的形态变异得以区分,但均应属于种内变异的范畴;莲山黄耆(A.leansanicus)与长管萼黄耆(A.limprichtii)分别聚为一支,支持将二者处理为两个相互独立的种。在相似性系数为0.66处,45份材料又被划分为4大类,类群的划分与黄耆属属下"组"的划分接近,但又存有差别。PCoA分析的结果与UPGMA聚类分析基本一致。相关分析结果表明,以组为单位划分黄耆属植物具有合理性,但部分组的设立以及部分种的归属仍需调整,准确地揭示黄耆属植物间的系统进化关系,还需要更多的材料采用多种分子标记方法。  相似文献

The subgenus Allium section Allium includes economically important species, such as garlic and leek, as well as other polyploid minor crops. Phylogenetic studies within this section, with a focus on horticultural groups within A. ampeloprasum, were performed on 31 accessions of 17 species using the nuclear ribosomal DNA internal transcribed spacer (ITS) region and the chloroplast trnL-F and trnD-T regions. The results confirmed the monophyly of section Allium. Four main clades were identified on all ITS analyses but the relationships among those and the remaining species studied within section Allium remained unresolved. Trees based on cpDNA recovered two major clades and a topology only partly congruent with that of the ITS tree. Intra-individual polymorphism of the ITS region proved useful in tracking putative parent species of polyploid taxa. The allopolyploid origin of great headed garlic (GHG), A. iranicum and A. polyanthum was confirmed. No signs of hybridization in leek or kurrat were detected but possible introgression events were identified in pearl onion and bulbous leek. Although GHG is often used as a garlic substitute, molecular analysis revealed only a distant relationship with garlic. We also clarified the previous incorrect classification of cultivated forms within A. ampeloprasum, by showing that leek, kurrat, pearl onion, and bulbous leek should be considered separately from GHG.  相似文献

Saponins are widely distributed among plant kingdom and possess a wide range of pharmacological properties. Genus Astragalus is the largest in the Fabaceae family, comprising more than 2,200 species spread over the Globe. Astragalus species have been used in traditional medicine of many countries as well as in the modern one. The main use in the folk medicine of the species is due to their saponin content. The paper is focused on the above mentioned group of compounds. Acipenser oxyrinchus is considered extirpated in Europe, but numerous breeding populations still exist on the Atlantic coast of North America. An adult female A. oxyrinchus, 2500 mm total length and 120 kg wet weight, was accidentally fished on 24 November 2010 near the coast of Gijón, Asturias, Spain. The fish was identified by its morphological pattern as well as by mitochondrial and nuclear DNA analyses. Because the sturgeon was found far away from any known breeding area, it was considered a stray or vagrant specimen. It certainly has a natural origin, but its eventual birthplace could not be determined. Because its current occurrence was unknown in southwestern Europe until now, the species is not cataloged or protected in this area. Therefore, the residual European stocks of A. oxyrinchus ought to be listed as Critically Endangered (CR) according to the IUCN categories. Likewise, it is imperative for southwestern European countries with an historic or recent occurrence of A. oxyrinchus to protect the species through domestic and international legislation. The present sympatric occurrence of A. sturio and A. oxyrinchus raises new challenges about key questions, such as the species selection for restoration program in European countries. Accurate monitoring is mandatory to obtain appropriate information for an assessment of the current occurrence of A. oxyrinchus in southwestern Europe.  相似文献

We use chloroplast DNA sequencing to examine aspects of the pre-European Māori cultivation of an endemic New Zealand root crop, Arthropodium cirratum (rengarenga). Researching the early stages of domestication is not possible for the majority of crops, because their cultivation began many thousands of years ago and/or they have been substantially altered by modern breeding methods. We found high levels of genetic variation and structuring characterised the natural distribution of A. cirratum, while the translocated populations only retained low levels of this diversity, indicating a strong bottleneck even at the early stages of this species’ cultivation. The high structuring detected at four chloroplast loci within the natural A. cirratum range enabled the putative source(s) of the translocated populations to be identified as most likely located in the eastern Bay of Plenty/East Cape region. The high structuring within A. cirratum also has implications for the conservation of genetic diversity within this species, which has undergone recent declines in both its natural and translocated ranges.  相似文献

The Lebanese mountain range is an important zone of plant species richness and endemism where the genus Astragalus constitutes a principal component of plant biodiversity. Most of endemic Astragalus taxa, living in mountains and arid zones of Mounts Lebanon and anti-Lebanon, are characterized by a cushion, spiny vegetative form, named “tragacanthic”, which is a remarkable example of vegetative convergence evolution. Because of determination difficulties, taxonomic uncertainties, and discrepancy in the number of taxa listed according to authors, new data are hardly needed to improve systematics of Astragalus and to investigate the role of the Lebanese mountain range as refugia of biodiversity. Before this study only two values on the genome size of Astragalus were reported in the literature and no previous molecular studies had been carried out on Astragalus genus in Lebanon. We examined the utility of rDNA ITS molecular markers to distinguish Astragalus species of Lebanese mountain range and the variation range of their genome size. The main results revealed a striking diversity in Lebanese Astragalus species with the emphasis of a huge variation of genome sizes, an important inter-specific chromosome polymorphism and the existence of a high phylogenetic diversity. The strict endemic species of the Lebanese mountains are positioned throughout the phylogeny. These results confirm that the Lebanon and anti-Lebanon mounts constitute a third diversity center for Astragalus and that high altitude areas are important refugia of plant biodiversity despite centuries of exploitation by humans.  相似文献

Nine recently described Aspergillus species and four known species in section Versicolores were tested for their ability to produce sterigmatocystin on two liquid media, Czapek w/20 % Sucrose Broth and Yeast Extract Broth grown in the dark for 1 week at 25 °C. Detection and quantification of ST were performed by reversed-phase liquid chromatography coupled with electrospray ionization ion trap mass spectrometry. Limit of detection was 3 ng/mL and limit of quantification was 10 ng/mL. Nine newly described Aspergillus species from various substrates, A. amoenus, A. creber, A. cvjetkovicii, A. fructus, A. jensenii, A. puulaauensis, A. subversicolor, A. tennesseensis and A. venenatus in section Versicolores were found to produce sterigmatocystin. Production was confirmed in recently collected isolates of A. protuberus and A. versicolor. A. austroafricanus and A. tabacinus did not produce sterigmatocystin.  相似文献

Aspergillus terreus is emerging as an etiologic agent of invasive aspergillosis in immunocompromised individuals in several medical centers in the world. Infections due to A. terreus are of concern due to its resistance to amphotericin B, in vivo and in vitro, resulting in poor response to antifungal therapy and high mortality. Herein we examined a large collection of molecularly characterized, geographically diverse A. terreus isolates (n = 140) from clinical and environmental sources in India for the occurrence of cryptic A. terreus species. The population structure of the Indian A. terreus isolates and their association with those outside India was determined using microsatellite based typing (STR) technique and Amplified Fragment Length Polymorphism analysis (AFLP). Additionally, in vitro antifungal susceptibility of A. terreus isolates was determined against 7 antifungals. Sequence analyses of the calmodulin locus identified the recently described cryptic species A. hortai, comprising 1.4% of Aspergillus section Terrei isolates cultured from cases of aspergilloma and probable invasive aspergillosis not reported previously. All the nine markers used for STR typing of A. terreus species complex proved to be highly polymorphic. The presence of high genetic diversity revealing 75 distinct genotypes among 101 Indian A. terreus isolates was similar to the marked heterogeneity noticed in the 47 global A. terreus population exhibiting 38 unique genotypes mainly among isolates from North America and Europe. Also, AFLP analysis showed distinct banding patterns for genotypically diverse A. terreus isolates. Furthermore, no correlation between a particular genotype and amphotericin B susceptibility was observed. Overall, 8% of the A. terreus isolates exhibited low MICs of amphotericin B. All the echinocandins and azoles (voriconazole, posaconazole and isavuconazole) demonstrated high potency against all the isolates. The study emphasizes the need of molecular characterization of A. terreus species complex isolates to better understand the ecology, acquisition and transmission of this species.  相似文献

Amblyceps waikhomi sp. nov. is described from the Nongkon stream which drains into the Noa Dehing River, a tributary of the Brahmaputra River, in Arunachal Pradesh, India. The new species can be distinguished from congeners (except A. torrentis) in having a deeper body depth at anus. It further differs from congeners (except A. mangois and A. serratum) in having fewer vertebrae, from A. mangois in lacking (vs. having) strongly-developed projections on the proximal lepidotrichia of the median caudal-fin rays, and in having a longer, wider, and deeper head; and from A. serratum in having a posteriorly smooth (vs. with 4–5 serrations) pectoral spine, and unequal jaw length (lower jaw longer and weakly-projecting anteriorly vs. equal upper and lower jaws). It additionally differs from A. murraystuarti, A. torrentis, A. apangi, A. laticeps, and A. cerinum in having a deeply forked (vs. emarginate or truncate) caudal fin. This species is the seventh amblycipitid species known to occur in the Ganga-Brahmaputra River system.  相似文献

Peanut, Arachis hypogaea L., is a protein-rich species consumed worldwide. A key improvement to peanut culture involves the development of cultivars that resist fungal diseases such as rust, leaf spot and scab. Over three years, we evaluated fungal resistance under field conditions of 43 wild accessions and three interspecific hybrids of the genus Arachis, as well as six A. hypogaea genotypes. In the first year, we evaluated resistance to early and late leaf spot, rust and scab. In the second and third years, we evaluated the 18 wild species with the best resistance scores and control cultivar IAC Caiapó for resistance to leaf spot and rust. All wild accessions displayed greater resistance than A. hypogaea but differed in their degree of resistance, even within the same species. We found accessions with as good as or better resistance than A. cardenasii, including: A. stenosperma (V15076 and Sv 3712), A. kuhlmannii (V 6413), A. kempff-mercadoi (V 13250), A. hoehnei (KG 30006), and A. helodes (V 6325). Amphidiploids and hybrids of A. hypogaea behaved similarly to wild species. An additional four accessions deserve further evaluation: A. magna (V 13751 and KG 30097) and A. gregoryi (V 14767 and V 14957). Although they did not display as strong resistance as the accessions cited above, they belong to the B genome type that is crucial to resistance gene introgression and pyramidization in A. hypogaea.  相似文献

Amanita caesareoides is a sister species of Amanita caesarea, also known as Caesar’s mushroom and one of the most desirable edible mycorrhizal mushrooms. However, cultivation of Caesar’s mushrooms has not yet been successful due to the difficulties involved in establishing pure cultures. In this study, we established pure cultures of four Asian Caesar’s mushroom species, i.e., A. caesareoides, Amanita javanica, Amanita esculenta, and Amanita similis, which were identified by sequence analysis of their rDNA internal transcribed spacer (ITS) region. Five selected isolates in A. caesareoides, A. javanica, and A. esculenta were tested for ectomycorrhizal syntheses with axenic Pinus densiflora seedlings in vitro. Ectomycorrhizal tips of each fungal isolate tested were observed on pine lateral roots within 5 months of inoculation. Seventeen pine seedlings that formed ectomycorrhizas in vitro with these three Amanita species were acclimatized under non-sterile conditions. Seven months following acclimatization, ectomycorrhizal colonization by A. caesareoides was observed on newly grown root tips, which was confirmed by polymerase chain reaction restriction fragment length polymorphism analysis of the fungal rDNA ITS region. Two other Amanita species also survived during ectomycorrhizal acclimatization. These results suggest that the cultivation of A. caesareoides and its relatives can be attempted through mycorrhizal synthesis using P. densiflora as a host. This is the first report of in vitro mycorrhization of Asian Caesar’s mushrooms and their acclimatization under non-sterile conditions.  相似文献

Fatty acid (FAs) and RAPD profiles were used to examine phenotypic and genetic relationships between eight Astragalus species including Astragalus maximus Willd. var. maximus, Astragalus coadunatus Hub. Mor. & Chamb., Astragalus kurdicus Boiss. var. kurdicus, Astragalus lagurus Willd, Astragalus christianus L., Astragalus cicer L., Astragalus atrocarpus Champ & Matthews and Astragalus onobrychioides Bieb., which were wildly growing in eastern Anatolia region of Turkey. All of the eight Astragalus species tested in this study were separated based on the presence and composition of 45 different FAs. Four of the Astragalus species including A. coadunatus, A. lagurus, A. christianus, and A. atrocarpus were rich in terms of FA contents containing at least 22–31 different FAs. The relative proportions of two fatty acids, 16:0, and 18:1:ω8c were higher in these four Astragalus species. The remaining species have limited number of FAs with unique FAMEs profiles. Six of the 10 decamer primers examined were selected to find out genetic polymorphism in Astragalus species. A total of 98 polymorphic bands were observed, ranging in size from 250 bp to 3000 bp. The RAPD results suggested that A. atrocarpus, A. onobrychioides and A. kurdicus are closely related and completely different from the other species. Six genetically distinct groups were found among the species of Astragalus. High genetic variations among Astragalus species growing wildly in eastern Anatolia region of Turkey may imply the differences in their origins. The results in the present study suggested that both RAPD and FA analyses are useful for differentiation of Astragalus species.  相似文献

Orchid bees compose an exclusive Neotropical pollinators group, with bright body coloration. Several of those species build their own nests, while others are reported as nest cleptoparasites. Here, the objective was to evaluate whether the inclusion of a strong biotic interaction, such as the presence of a host species, improved the ability of species distribution models (SDMs) to predict the geographic range of the cleptoparasite species. The target species were Aglae caerulea and its host species Eulaema nigrita. Additionally, since A. caerulea is more frequently found in the Amazon rather than the Cerrado areas, a secondary objective was to evaluate whether this species is increasing or decreasing its distribution given South American past and current climatic conditions. SDMs methods (Maxent and Bioclim), in addition with current and past South American climatic conditions, as well as the occurrences for A. caerulea and E. nigrita were used to generate the distribution models. The distribution of A. caerulea was generated with and without the inclusion of the distribution of E. nigrita as a predictor variable. The results indicate A. caerulea was barely affected by past climatic conditions and the populations from the Cerrado savanna could be at least 21,000 years old (the last glacial maximum), as well as the Amazonian ones. On the other hand, in this study, the inclusion of the host-cleptoparasite interaction complex did not statistically improve the quality of the produced models, which means that the geographic range of this cleptoparasite species is mainly constrained by climate and not by the presence of the host species. Nonetheless, this could also be caused by unknown complexes of other Euglossini hosts with A. caerulea, which still are still needed to be described by science.  相似文献

Alloteuthis is a group of small, slender loliginid squids of minor fisheries importance. There are three nominal Alloteuthis species—A. media (Linnaeus), A. subulata (Lamarck) and A. africana Adam. Two of these species (A. media and A. subulata) have largely overlapping ranges in the Mediterranean and northeastern Atlantic, while A. africana is found along the west coast of Africa. Despite the low level of species diversity, Alloteuthis taxonomy and systematics are confused, and assignment of specimens to species can be difficult. To clarify Alloteuthis systematics, we gathered morphometric data and DNA sequence data from two mitochondrial loci and a nuclear locus from Alloteuthis specimens collected from several localities. Analyses of the morphometric data suggest that head width is the main variable allowing separation of A. africana from the other two species, and central club sucker size separates A. media from A. subulata. One easily diagnosable character often used to distinguish Alloteuthis species—relative fin length—appears to be of little taxonomic value. Only three specimens assignable to A. subulata both morphologically and genetically were found, all from the Adriatic; possible reasons for this apparent rarity are discussed. Gene tree parsimony and coalescent-based methods were used to estimate species relationships from the molecular data, and both supported a sister-species relationship between A. media and A. subulata. Analyses of molecular variation (AMOVA's) revealed significant genetic differentiation between Atlantic and Mediterranean A. media. This study highlights the importance of 1) sampling multiple individuals, locations and loci for species-level phylogenetic studies, 2) using morphometric analyses to reveal taxonomically meaningful morphological characters and 3) accounting for the stochastic nature of the coalescent process when estimating species phylogenies for closely related taxa.  相似文献

Polyploid species possess more than two sets of chromosomes and may show high gene redundancy, hybrid vigor, and masking of deleterious alleles compared to their parent species. Following this, it is hypothesized that this makes them better at adapting to novel environments than their parent species, possibly due to phenotypic plasticity. The allopolyploid Arabidopsis suecica and its parent species A. arenosa and A. thaliana were chosen as a model system to investigate relationships between phenotypic plasticity, fitness, and genetic variation. Particularly, we test if A. suecica is more plastic, show higher genetic diversity, and/or have higher fitness than its parent species. Wild Norwegian populations of each species were analyzed for phenotypic responses to differences in availability of nutrient, water, and light, while genetic diversity was assessed through analysis of AFLP markers. Arabidopsis arenosa showed a higher level of phenotypic plasticity and higher levels of genetic diversity than the two other species, probably related to its outbreeding reproduction strategy. Furthermore, a general positive relationship between genetic diversity and phenotypic plasticity was found. Low genetic diversity was found in the inbreeding A. thaliana. Geographic spacing of populations might explain the clear genetic structure in A. arenosa, while the lack of structure in A. suecica could be due to coherent populations. Fitness measured as allocation of resources to reproduction, pointed toward A. arenosa having lower fitness under poor environmental conditions. Arabidopsis suecica, on the other hand, showed tendencies toward keeping up fitness under different environmental conditions.  相似文献
