Desert wadis are widespread in the Arabian Peninsula and play a vital role in the ecology of the region; nevertheless, these ecosystems are among the least studied. Various types of microbial mats are predominant in wadis, but information on their bacterial diversity and spatial distribution is very scarce. We investigated bacterial diversity, pigments and lipid composition of ten mats located at the down-, mid- and upstream of a desert wadi in Oman. Direct microscopy revealed the existence of different unicellular and filamentous cyanobacteria, with the dominance of the heterocystous genera Calothrix and Scytonema. The majority of MiSeq 16S rRNA sequences (44-76%) were affiliated to Cyanobacteria and Proteobacteria. While Alphaproteobacteria was the most dominant proteobacterial class (10 to 48% of total sequences), Gamma- and Deltaproteobacteria were subdominant. Cluster analysis showed that the mats’ bacterial communities at the different locations along the wadi were different and shared less than 60% of their operational taxonomic units (OTUs). Chlorophyll a and scytonemin were the most predominant pigments in all mats. Different saturated, branched and mono- and poly-unsaturated fatty acids were detected in all mats, with C16 and C18 compounds as most dominant. The detected pigments and fatty acids indicate a major role of cyanobacteria in the wadi mats and the adaptation of microorganisms therein to the harsh wadi environment. Detection of diadinoxanthin and fucoxanthin confirmed the presence of diatoms. We conclude that microbial mats are important elements in wadi ecosystems and exist in a great variety of structure and community composition.  相似文献

Under the influence of alkyl hydroxybenzene (C6-AHB) added to cell suspensions at concentrations of (1–5) × 10–3M, the cells of Saccharomyces cerevisiae, Micrococcus luteus, and Thioalkalivibrio versutusunderwent dramatic changes in the ultrastructural organization of cell membranes, cytoplasm, and inclusions. In yeast suspension, the first changes were observed after 15 min in the structure of pocket-like invaginations in the cytoplasmic membrane (CM): they were shortened and thickened. In the subsequent 30 to 60 min, CM ruptures were formed in the regions devoid of intramembrane protein particles and in the pocket-like invaginations. After 24 h, complete disintegration of the intracellular membrane structures and conglomeration of the ribosomal part of the cytoplasm occurred. Similar changes were observed on the exposure of gram-positive and gram-negative bacteria to AHB. However, the cell wall in all the microorganisms studied was not destroyed, and in Micrococcus luteusit was even thickened. These mummified forms were preserved as morphologically intact but nonviable cells for more than three years of observations. By their ultrastructural characteristics, these mummified forms of microorganisms were similar to the fossilized microorganisms discovered by us in fibrous kerite. The concept of micromummies was formulated. AHB are supposed to play an important role in the process of fossilization of microorganisms in nature.  相似文献

The ultrastructure of the stigma and associated flagellar-microtubular systems in Dinobryon cylindricum var. alpinum is described in detail and compared with observations on comparable photo-kinetic systems in other chrysophycean organisms. The chloroplastidic stigma of D. cylindricum var. alpinum is shown to lie in a particular positional relationship to the flagellar swelling in the anterior furrow and to several other organelles, to consist of a monolayer of c. 40 pigmented granules, each c. 250–500 nm diameter, arranged in a definite pattern, and to be overlain by several membrane systems. Other cytoplasmic pigmented bodies with dense crystalline contents surrounded by a single “unit membrane” aggregate near the anterior furrow on the side opposite the stigma. The swelling on the proximal portion of the smooth flagellum is separated from the plasmalemma of the anterior furrow by a nearly constant distance of 75–100 nm, has a multilamellate substructure that is linked by fine radiating interconnections to the axoneme doublets, and is connected to the plasmalemma by a system of fibrillar interconnections. A transitional helix in the basal body region is described as similar to structures reported in other chrysophycean flagellates. A striated rhizoplast with a periodicity of c. 90 nm extends from basal body I to the nuclear envelope. A seven-stranded microtubular root extends from the same basal body. Other fibrous and microtubular root systems are also described. The inter-relationships and possible functions of the aforementioned structures are discussed.  相似文献

Microbial communities growing in the bed of the alkaline, sulfide hot spring Bol'sherechenskii (the Baikal rift area) were studied over many years (1986–2001). The effluent water temperature ranged from 72 to 74°C, pH was from 9.25 to 9.8, and sulfide content was from 12 to 13.4 mg/ml. Simultaneous effects of several extreme factors restrict the spread of phototrophic microorganisms. Visible microbial mat appears with a decrease in the temperature to 62°C and in sulfide content to 5.9 mg/l. Cyanobacteria predominated in all biological zones of the microbial mat. The filamentous cyanobacteria of the genus Phormidium are the major mat-forming organisms, whereas unicellular cyanobacteria and the filamentous green bacterium Chloroflexus aurantiacus are minor components of the phototrophic communities. No cyanobacteria of the species Mastigocladus laminosus, typical of neutral and subacid springs, were identified. Seventeen species of both anoxygenic phototrophic bacteria and cyanobacteria were isolated from the microbial mats, most of which exhibited optimum growth at 20 to 45°C. The anoxygenic phototrophs were neutrophiles with pH optimum at about 7. The cyanobacteria were the most adapted to the alkaline conditions in the spring. Their optimum growth was observed at pH 8.5–9.0. As determined by the in situ radioisotope method, the optimal growth and decomposition rates were observed at 40–32°C, which is 10–15°C lower than the same parameter in the sulfide-deficient Octopus Spring (Yellowstone, United States). The maximum chlorophyll a concentration was 555 mg/m2 at 40°C. The total rate of photosynthesis in the mats reached 1.3 g C/m2 per day. The maximum rate of dark fixation of carbon dioxide in the microbial mats was 0.806 g C/m2 per day. The maximum rate of sulfate reduction comprised 0.367 g S/m2 per day at 40°C. The rate of methanogenesis did not exceed 1.188 g C/m2 per day. The role of methanogenesis in the terminal decomposition of the organic matter was insignificant. Methane formation consumed 100 times less organic matter than sulfate reduction.  相似文献

A laboratory model of a cyano-bacterial mat with mineral layers of carbonates was used to examine the dynamics of the transformation of calcium-magnesium carbonate under the conditions of a soda lake. The activity of various organisms of the cyanobacterial community results in conditions under which the Ca-Mg carbonate precipitate undergoes changes. The crystal lattice of the initial carbonate is restructured; its mineralogical composition changes depending on the conditions of the mat. In magnesium calcites, which are formed under such low-temperature conditions, a rudimentary cation adjustment can occur with the formation of dolomite domains. These experiments confirm the hypothesis that the dolomite found in stromatolites is of a secondary origin and can be formed in the course of transformation of Ca-Mg carbonates under alkaline conditions in an alkaliphilic cyanobacterial community.  相似文献

Cephalic sense organs of Heterodera glycines males were examined in detail by electron microscopy. Each amphid basically consists of an amphidial gland, a nerve bundle, and an amphidial duct. The amphidial gland consists of a microvillous region, and laterally is closely associated with a large secretory cell. The nerve bundle penetrates the microvillous region, and further anteriorly individual nerve processes (dendrites) separate from one another, thus forming a sensilla pouch which is enveloped by the microvillous region of the gland. Anterior to the pouch, the cilia-like dendrites converge as they enter and eventually terminate in the amphidial duct. Heterodera glvcines males have the innervation basis for a full complement of sixteen papillae, although surface manifestations are present for only six minute inner labial papillae. In addition, four outer labial and four cephalic receptors terminate beneath the surface, and another two dendrite pairs end further posteriorly beneath the basal plate of the cephalic framework. Papillary receptors which terminate beneath the surface are probably mechanoreceptive, whereas inner labial papillae have pore-like openings to the exterior and may be chemoreceptive. Amphids and papillae of H. glycines are fundamentally similar to those of Meloidogyne incognita, although certain striking differences exist.  相似文献

Annual and perennial cyanobacterial mats from streams on Signy Island, Antarctica, show similar areal concentrations of chlorophyll-a and areal rates of photosynthesis. Maximum rates of photosynthesis were temperature dependant over the range 0–14 C, with a Q10 of approximately 2.5. Rates of photosynthesis per unit chlorophyll-a were comparable to other Antarctic mat communities but low compared to phytoplankton from upstream lakes. Areal rates of photosynthesis were however much higher than for phytoplankton. Low chlorophyll-specific rates of photosynthesis are interpreted as the effect of self shading within the mats. It is hypothesised that these mats rapidly attenuate incoming radiation and that photosynthesis in most of the mat is effectively light-limited. This situation is likely to occur in all thick periphyton films.  相似文献

Encephalitozoon hellem is a new human microsporidian isolated from corneal biopsies and conjunctival scrapings of three AIDS patients and cultured in Madin Darby canine kidney (MDCK) cells. Encephalitozoon hellem and Encephalitozoon cuniculi display different protein profiles with sodium dodecyl sulfate-polyacrylamide gel electrophoresis and unique antibody binding patterns with murine antisera against Western blots of each organism. Developmental stages of E. hellem in culture are similar to E. cuniculi. Meronts are 1.3–2.7 μm in diameter, develop within a parasitophorous vacuole adjacent to the vacuolar membrane, divide by binary fission, and contain one or two discrete nuclei. Sporonts measure 2 × 3 μm, separate from the vacuolar membrane, and have a thickened outer membrane. Sporoblasts display a tri-layered wall and possess the earliest recognized polar filaments. Mature spores measure 1 × 1.5 μm and are more electron-dense than other stages. Each spore contains a single nucleus, a polar tubule with four to nine coils, thin electron-dense exospore and thick, electron-lucent endospore. Although E. hellem and E. cuniculi differ biochemically and immunologically, their fine structure and development are indistinguishable.  相似文献

Photosynthetic activity, the content of various photosynthetic pigments, and the chloroplast ultrastructure were examined in the leaves of cucumber (Cucumis sativus L.) and pea (Pisum sativum L.) plants of different ages grown under red light (600–700 nm, 100 W/m2). In pea leaves tolerant to red-light irradiation, chloroplast ultrastructure did not essentially change. In the first true leaves of cucumber plants susceptible to red-light irradiation, we observed a considerable increase in the number and size of plastoglobules, the appearance of chloroplasts lacking grana or containing only infrequent grana, and stromal thylakoids. In the upper leaves of 22-day-old cucumber plants, the chloroplast structure was essentially similar to that of the control chloroplasts in white light, and we therefore suppose that these plants have acclimated to red light.  相似文献

Amphids, and the cephalic and labial papillae of Meloidogyne incognita males were examined in detail by electron microscopy. Each amphid basically consists of an amphidial gland, a nerve bundle and an amphidial duct. The gland is a broad microvillous organ with a narrow anterior process, which is closely associated with the amphidial duct. A posterior process of the gland contains secretory organelles and proceeds along the esophagus with the lateral cephalic nerve bundle. The nerve bundle penetrates the broad portion of the gland and, subsequently, individual nerve processes (dendrites) separate from one another, thus forming the sensilla pouch which is enveloped by the gland. Anterior to the pouch, the dendrites converge as they enter and eventually terminate in the amphidial duct. The external opening of the duct is a broad slit which separates the cheek, the outermost part of the lateral lip, from the remainder of the lip region. M. incognita males have six inner labial papillae and four outer cephalic papillae which are each innervated by two and one cilia, respectively. In labial papillae, the cilia appear to terminate at the base of a pore opening, whereas in cephalic papillae each cilium terminates beneath the labial cuticle.  相似文献

Nostoc commune Vaucher (a cyanobacterium) is a very conspicuous terrestrial primary producer in Victoria Land, continental Antarctica. Because polar ecosystems are considered to be especially sensitive to environmental changes, understanding the environmental constraints on net carbon (C) fixation by N. commune is necessary to determine the effects of environmental changes on the ecological functioning of ice‐free areas of the continent. A model describing net C fixation in terrestrial populations of N. commune in an Antarctic dry valley was constructed using field and laboratory measurements in which N. commune colonies were exposed to different combinations of incident irradiance (400–700 nm), temperature, and degree of desiccation. For desiccated N. commune mats with water content ≤ 30% saturation, net C fixation was highly variable between replicates and could not be modelled. However, for colonies at > 30% saturation, rates of net C fixation and dark respiration depended strongly on irradiance and temperature. Net C fixation reached a maximum rate of 21.6 μg C m− 2 s− 1 at irradiance of approximately 250 μmol m− 2 s− 1 and the optimum temperature of 20.5 °C. Agreement between predicted short‐term net C fixation and field and laboratory measurements allowed estimation of total seasonal fixation, using previously published environmental data. Annual net C fixation was estimated in the range 14.5–21.0 g C fixed m− 2Nostoc mat, depending on year/season. Estimates for different seasons correlated with thermal time (accumulated hours above 0 °C during the year) rather than irradiance, in contrast to communities in local lacustrine environments, where irradiance is the main driver of primary productivity. In the terrestrial habitat, N. commune appears to compromise between an ability to capitalize on short periods of higher temperature and efficient utilization of lower irradiance at low temperature. The relationship between thermal time and net annual C fixation by N. commune is strongly linear.  相似文献

ABSTRACT. The first ultrastructural study of the actinosporean genus Triactinomyxon was carried out on Triactinomyxon legeri from the intestinal epithelium of Tubifex tubifex. The developmental cycle starts with bi- and uninucleate cells. We propose that these cells may be an early proliferative phase of the cycle and may unite to give rise to the four-cell stage, initiating pansporoblast formation. Valvogenic cells transform in the long stylus and anchor-like projections of the spore. In the capsulogenic cells, the primordium of the polar capsules originates as a simple, dense, club-shaped structure not observed in other actinosporeans. In all other respects, actinosporean ultrastructure follows more or less similar patterns. Comparison of actinosporean and myxosporean species gives evidence of considerable structural similarity, exemplified in both classes by the occurrence of cell junctions in their multicellular spores, identical polar capsules and their morphogenesis, cell-in-cell condition, pansporoblast formation, and presence of dense bodies (sporoplasmosomes) primarily in the sporoplasm. This unity of patterns speaks in favor of the postulated actinosporean-myxosporean transformation, which warrants further study.  相似文献

Several studies have shown that the fatty acid composition of circulating lipids reflects dietary fat intake, in turn being related to health status. The fatty acid composition of plasma lipids is therefore an important parameter in studies on dietary interventions. The aim of our study was to develop a rapid and inexpensive method for the analysis of circulating fatty acids applicable to large population groups. Drops of blood collected from fingertips have been directly subjected to transmethylation for gas chromatography analysis. This new method, validated for reproducibility, has been compared with the conventional method, based on withdrawal of blood from the antecubital vein followed by lipid extraction, and identical data have been obtained with the two techniques. Observed and predicted differences between blood and plasma fatty acids are related to the contribution of circulating cell membranes in blood. Finally the application of the methods to samples from 100 healthy subjects and the assessed correlation between dietary habits and blood fatty acid profiles demonstrate the validity of the new method and its applicability to nutritional and epidemiological studies.  相似文献

Females of Anopheles albimanus mosquito are known to prefer floating cyanobacterial mats over open water for oviposition. We used Solid Phase MicroExtraction (SPME) devices on-site to trap substances volatilized from these two environments, followed by analysis by GC-MS (gas chromatograph-mass spectrometer). In enclosed headspace field microcosms, an unidentified C-15 aliphatic alcohol was persistently found over cyanobacterial mats as compared to open water. Based on this finding, we conducted oviposition experiments using a commercially available compound, n-pentadecanol, close in molecular weight and mass spectral pattern to the unknown C-15 aliphatic alcohol. The results indicate a tendency of female mosquito to oviposit more eggs in containers with the tested chemical as compared to water and blank.  相似文献

The purpose of this study was to characterize Ni- and Zn-sulfides precipitated in sulfate-reducing bacterial cultures. Fe-free media containing 58 mM SO 4 2? were amended with Ni and Zn chloride followed by inoculation. Precipitates were sampled from cultures after two weeks of incubation at 22, 45, and 60 ° C. Abiotic controls were prepared by reacting bacteria-free liquid media with Na 2 S solutions under otherwise identical conditions. Precipitates were collected anaerobically, freeze-dried and analyzed by x-ray diffraction (XRD), scanning electron microscopy, and for total Ni, Zn, and S. In Ni-containing media, biogenic sulfide precipitates were mostly heazelwoodite (Ni 3 S 2 ), whereas abiotic precipitates were mixed heazelwoodite and vaesite (NiS 2 ). The biogenic Ni-precipitates were better crystalline than the corresponding abiotic samples. Sphalerite (ZnS) was identified by XRD in precipitates sampled from Zn-containing media. Scanning electron microscopy revealed disordered morphological features for the sulfides, which occurred mostly as aggregates of fine particles in biogenic samples, whereas abiotic precipitates contained more plate- and needle-like structures.  相似文献

The fine structure of the sapropelic ciliate Saprodinium dentatum is described based on phase-contrast microscopy, silver-staining techniques, cryo-fracture scanning electron microscopy, and thin sections. The study concentrates on a detailed analysis of the somatic cortex and the oral ciliature of this highly asymmetric, laterally compressed ciliate. The cell shape is dominated by a number of site-specific spines and the curving course of 10 somatic kineties (SK 1–10). The SK, composed of dikinetids, show an intrakinety differentiation that seems characteristic for other odontostomes as well. The anterior segment of the SK is mostly ciliated, followed by a non-ciliated segment in which the kinetosomes lack all typical fiber systems. Except for SK 4–6, the posterior segment is ciliated again, forming the spine kinetics associated with particular caudal spines. The anterior segment of SK 3 through SK 7 form the frontal band, which together with the two frontal kineties constitutes the main locomotory organelle for a ciliate that creeps on the substratum. A short kinety with inverse polarity, not seen in earlier light microscopical studies, was observed near the oral spine. We made particular effort to find a logical explanation for the observed association of the SK with the various caudal spines. The oral ciliature consists of nine adoral organelles located in a tripartite oral cavity. The absence of a paroral ciliature together with the position of the cytostome anterior to the adoral organelles may be the result of rotational movement of the oral apparatus during the evolution of these bizarre ciliates. Results are discussed with special reference to the phylogenetic relationship of the Odontostomatida to the Heterotrichida and no conclusive answer was found in this first electron microscopical study of an odontostomatid ciliate.  相似文献

The plasma-borne long-chain free fatty acids (FFA) enter skeletal muscle cells. Upon entering they are oxidized or esterified and a fraction remains free (non-esterified). The data on free fatty acids in skeletal muscles remain highly controversial. Furthermore, the composition of individual fatty acids in various lipid fractions including free fatty acids, monoglyceride and diglyceride in muscles has not been characterized. Also data on the composition of fatty acids esterified into muscle triglycerides and phospholipids are incomplete. The present study was undertaken to examine a composition of fatty acids in lipid fractions of different skeletal muscle types. For this purpose, samples of the rat soleus, red and white portions of gastrocnemius were excised, trimmed of visible fat and fascias and immediately frozen in liquid nitrogen. Samples were then pulverized and, lipids were extracted and fractionated by thin-layer chromatography. Individual long-chain fatty acids in different fractions were identified, characterized and quantitated by gas-liquid chromatography. FFA composition in the plasma was also determined. The total FFA content in the soleus, red and white gastrocnemius was 69.1 ± 10.8, 49.0 ± 13.6 and 22.7 ± 8.6 nmol/g, respectively. Palmitic and oleic acids were the major fatty acids in the muscles FFA fraction. Monoglyceride fraction of each muscle contained palmitic, stearic and linoleic acid as the major fatty acids, Diglyceride fraction contained mostly palmitic and oleic acid whereas triglyceride fraction mostly palmitic and linoleic acid.. The fraction of phospholipids was composed mostly of palmitic and linoleic acid but contained also considerable percentage of archidonic acid. Total plasma FFA/muscle FFA ratio depended on a muscle type and was: 2.4 in the soleus, 3.5 in the red and 7.4 in the white gastrocnemius. This assured transport of FFA to the myocytes. However, there were great differences in the ratio between particular FFA within the same muscle as well between the muscles. It indicates that individual FFA are either selectively transported from the plasma to the muscles or selectively used within the myocytes or both.  相似文献

1. SJL/J mice were maintained on semipurified diets which differed in the ratio of polyunsaturated/saturated fatty acid content (P/S). Exposure was from conception and was maintained for periods ranging from 6 to 34 weeks. 2. Neural cell cultures were prepared from dorsal root ganglia (DRG). After 6 and 20 days of culture, neuronal electric membrane properties were determined quantitatively by intracellular recording. 3. A number of significant differences were observed for the two dietary conditions. DRG from mice on the low-P/s diet had an increase in the rate of fall of both phases of repolarization which, in conjunction with the reduced action potential overshoot, led to a reduced action potential duration. This shift to shorter-duration action potentials was accompanied by a shift to more monophasic falling phases. The low-P/S neurons also exhibited a decreased afterhyperpolarization, decreased specific membrane resistance, and decreased membrane electrical time constant compared to high-P/S neurons. 4. It was concluded that the P/S ratio in the diet can have a significant effect on the electric properties of neurons. The high-P/S neurons tended to have action potentials with biphasic repolarizations and longer durations. In contrast, the low-P/S neurons tended to have action potentials with monophasic repolarizations and shorter durations. Moreover, the known ionic dependence of these two types of action potentials suggested that the low-P/S diet resulted in action potentials with a more exclusive Na dependence, while the high-P/S diet resulted in action potentials with both Na and Ca dependence.  相似文献

Tuatara (Sphenodon) are rare reptiles endemic to New Zealand. Wild tuatara on Stephens Island (study population) prey on insects as well as the eggs and chicks of a small nesting seabird, the fairy prion (Pachyptila turtur). Tuatara in captivity (zoos) are fed diets containing different insects and lacking seabirds. We compared the fatty acid composition of major dietary items and plasma of wild and captive tuatara. Fairy prions (eaten by tuatara in the wild) were rich in C20 and C22 polyunsaturated fatty acids (PUFA), especially the n-3 fatty acids eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA). In contrast, items from the diet of captive tuatara contained no C20 and C22 PUFA and were higher in medium-chain and less unsaturated fatty acids. Plasma from wild tuatara was higher in n-3 PUFA [including alpha-linoleic acid (C18:3n-3), EPA and DHA], and generally lower in oleic acid (C18:1) and palmitic acid (C16:0), than plasma from captive tuatara in the various fractions (phospholipid, triacylglycerol, cholesterol ester and free fatty acids). Plasma from wild adult tuatara showed strong seasonal variation in fatty acid composition, reflecting seasonal consumption of fairy prions.
