Context: Stomach ulcers are the common gastrointestinal disorders worldwide.

Objective: This study aimed to investigate the therapeutic impact of Pulicaria crispa aerial parts ethanol extract against gastric ulcer in rats.

Materials and methods: Ulcer was induced by one oral dose of ethanol (0.5?ml/100g body weight) on 24?hours empty stomach, then the plant extract (500?mg/kg b.wt.) was orally administered daily for one week. Ranitidine (100?mg/kg b.wt.); as a reference drug was evaluated. Stomach acidity and volume, as well as lesion counts were measured. Levels of malondialdehyde (MDA), glutathione (GSH) and superoxide dismutase (SOD) were estimated. Assay of different marker enzymes; succinate dehydrogenase (SDH), lactate dehydrogenase (LDH), glucose-6-phosphatase (G-6-Pase), acid phosphatase (AP) and 5′-nucleotidase (5′NT) were determined. Interlukin-10 (IL-10), intracellular adhesion molecule-1 (ICAM-1) and tumor necrosis factor alpha (TNF-α) were also determined. Stomach histopathological assessment was detected.

Results: Gastric ulcer showed drastic changes in oxidative stress, cell organelles and inflammatory markers. These biomarkers served as good tools to identify the presence of gastric ulcer. Treatment with P. crispa recorded amelioration in most parameters exceeding the auto healing effect.

Conclusion: Healing potency of P. crispa is possibly related to its content of glycosides, coumarins, flavonoids, tannins, sterols and triterpenes.  相似文献

为了探讨乙醇性胃粘膜损伤和牛磺酸对其保护作用的机制,将40只SD大鼠分为对照组、乙醇损伤组和牛磺酸保护组,观察其胃粘膜中内皮素(ET)、一氧化氮合成酶(NOS)、生长抑素(SS)和血管活性肠肽(VIP)的变化。结果如下:(1)随着乙醇作用时间延长、粘膜的损伤加重,胃粘膜内ET含量显著上升,而NOS、SS和VIP含量显著下降;(2)牛磺酸可显著减轻乙醇性胃粘膜损伤,抑制ET释放,增加NOS活性和SS、VIP的含量。上述结果表示,ET、NOS、SS和VIP的变化参与了乙醇性胃粘膜损伤的病理生理过程;牛磺酸对乙醇性胃粘膜损伤具有保护作用,可能与其调节胃粘膜内ET、NOS、SS和VIP的释放与活性有关。  相似文献

The proopiomelanocortin-derived tridecapeptide alpha-melanocyte-stimulating hormone (alpha-MSH) is a neuropeptide that exerts broad anti-inflammatory actions in mammals. This study aimed to investigate the effect of alpha-MSH on ethanol-induced gastric ulcer in rats and to evaluate the involvement of endogenous somatostatin in the actions of the peptide. The rats received 1 mL 75% ethanol or saline orally. alpha-MSH was given (25 micro g/rat; i.p.) alone or following the somatostatin antagonist cyclo-(7-aminoheptanoyl-PH-E-d-Trp-Lys-THR) (10 microM/kg; i.p.) administration. Gastric lesions were scored macroscopically and microscopically following decapitation at 30 min after ethanol challenge. Gastric malondialdehyde (MDA) level, myeloperoxidase (MPO) activity and mast cell counts were assessed. Ethanol-induced gastric hemorrhagic lesions were characterized by increased gastric MDA level, MPO activity and mast cell counts. alpha-MSH treatment decreased the extent of tissue injury and reversed tissue MDA level, MPO activity and mast cell counts. The effect of the peptide on the severity of gastric lesions, MDA level and MPO activity was reversed by the somatostatin antagonist. In conclusion, alpha-MSH is beneficial in a rat model of gastric ulcer via mechanisms which partly involve the endogenous somatostatin.  相似文献

采用高效液相色谱分离、串联四极杆-飞行时间质谱正离子模式检测‘红肉脐橙’和‘清家脐橙’果实黄皮层、白皮层、囊衣和汁胞中的主要类黄酮。根据保留时间、精确质荷比、二级质谱以及标准品化合物验证,确定了脐橙不同组织中含量较高的甜橙黄酮、川陈皮素等13种类黄酮。依据峰面积比较相对含量,认为脐橙黄皮层中类黄酮含量丰富,白皮层和囊衣中类黄酮含量次之,汁胞中类黄酮相对含量较少。黄皮层中以甜橙黄酮、川陈皮素和橘皮素等多甲氧基黄酮为主,而白皮层、囊衣和汁胞中的类黄酮以橙皮苷、柚皮苷为主。脐橙相同组织如白皮层、囊衣和汁胞中类黄酮的相对含量在品种间无显著差异,但橙皮苷、3,5,6,7,3',4'-六甲氧基黄酮和橘皮素在两品种的黄皮层中相对含量差异显著。研究结果为进一步研究和综合利用脐橙的活性物质提供了科学依据。  相似文献

Although Helicobacter pylori (Hp) plays an important role in the pathogenesis of chronic gastritis and gastric ulcer, little is known about the probable mechanisms of these types of gastrointestinal damage. To determine the precise mechanisms involved in ulcer formation, immune responses in patients with gastric ulcer (GUP) caused by Hp infection (Hp+) were compared with those of other gastritis patients (GP). The sensitivity and proliferation of peripheral blood mononuclear cells (PBMNCs) obtained from patients were evaluated by 3‐(4,5‐dimethylthiazol‐2‐yl)‐2,5‐diphenyltetrazolium bromide (MTT) assay against exposure with complex Hp crude antigen (HPCA) and mitogen (phytohemagglutinin, PHA). Production of inflammatory cytokines, including interleukin (IL)‐1β and IL‐8, in serum and supernatants of PBMNCs were then measured by ELISA. It was found that, after stimulation with PHA, both IL‐8 and IL‐1β concentrations in sera and supernatants as well as proliferation and sensitivity were statistically greater in GUP Hp+ than GP Hp?. Furthermore, HPCA inhibited the proliferation of PBMNCs dose‐dependently; however, it stimulated IL‐8 and IL‐1β production in supernatants of mononuclear cells. Therefore, the up‐regulated concentrations of IL‐8 and IL‐1β may have been caused by increase in the size of mononuclear cell subpopulations or in their cytokine secretory activity, indicating the greatest cell responsiveness in GUP Hp+ patients. These results suggest that tissue damage and ulcers occur in patients who produce more IL‐8 and IL‐1β than patients who do not develop ulcers; the former consequently have more activated immune cells at the site of infection. Therefore, both host responses and Hp virulence factors may be involved in the development of gastric ulcers.

Two Diaporthe species isolated from fruit of Citrus sinensis in China were characterized based on morphology and multilocus phylogeny of ITS, tef1, and tub2 gene sequences. The phylogeny indicated that the two species match Diaporthe taoicola and D. siamensis. A critical examination of phenotypic characteristics confirmed the phylogenetic results. Diaporthe taoicola was morphologically characterized by producing Alpha conidia with tapering toward both ends. Meanwhile, D. siamensis produced cylindrical or ellipsoidal Alpha conidia with two oil drops. Pathogenicity tests revealed that both species were pathogenic to fruit of C. sinensis. To our knowledge, the two species were firstly reported on Citrus sinensis in China.  相似文献

We investigated the gastroprotective effect of apricot kernel oil on ethanol induced gastric ulcer in rats. Male Wistar albino rats were divided into control, ethanol and apricot kernel oil + ethanol groups. The fatty acid composition of apricot kernel oil was determined using GC-MS. A gastric ulcer index was defined as the area percentage of the gastric mucosa consisting of ulcerated tissue. Gastric tissue was investigated by TUNEL staining for apoptosis, immunohistochemical iNOS staining, measurement of gastric IL-10 and IL-6 expression by ELISA and assays of catalase, malondialdehyde and superoxide dismutase. The ethanol group exhibited a higher gastric ulcer score, increased IL-6 level, increased number of inducible nitric oxide synthase-positive and TUNEL positive cells, and a higher MDA level compared to the control group. The apricot kernel oil + ethanol group exhibited significantly fewer gastric lesions compared to the ethanol group. Apricot kernel oil protects rat gastric mucosa against ethanol induced injury by its anti-inflammatory, anti-oxidative and anti-apoptotic effects, and might be useful for reducing the severity of gastric ulcers.  相似文献

MYB类蛋白是一类与植物防卫反应有关的转录因子家族,本研究利用构建的甜橙健株与感染黄龙病的病株差减(SSH)文库,采用RACE技术克隆了一个MYB类基因的cDNA全长序列,命名为CsMYB(GenBank登录号为HQ841074)。柑橘CsMYB基因的cDNA全长为1 306 bp,生物信息学分析显示,该基因包括一个909 bp的完整开放读码框以及一个典型的26 bp poly-A,编码302个氨基酸,分子量为32.97 kD,等电点为8.5。同时,还有MYB类基因的保守特征区域,即在N端有两个典型的MYB DNA结合域:R2和R3。实时荧光定量PCR分析表明,柑橘CsMYB基因受到黄龙病菌侵染后的不同时期表达量不同,伴随着黄龙病病程的变化呈现不同的表达变化。推测CsMYB基因是一个转录因子,可能参与柑橘对黄龙病菌的防御反应过程。  相似文献

Background. The recently reported OMNIUM and ASTRONAUT NSAID ulcer prevention trials using omeprazole to prevent endoscopic ulcer recurrence among chronic NSAID users suggested superiority over misoprostol or ranitidine. Aim. To test the hypothesis the results from the OMNIUM and ASTRONAUT studies would not be generalizible as ulcer healing and ulcer recurrence would differ in relation to Helicobacter pylori status. Methods. The data regarding H. pylori status were made available by AstraZenca allowing separate analysis of the outcome of those with NASID ulcers (i.e. without H. pylori infection) and those NSAID use was complicated with the presence of an active H. pylori infection. Results. Reanalysis confirmed that omeprazole was superior to placebo for the prevention of ulcer recurrence in chronic NSAID users. However, overall omeprazole was not significantly better than the subtherapeutic dose (400 µg/day) of misoprostol (14.5% vs. 19.6%, respectively, p = .93); 400 µg of misoprostol was actually superior to omeprazole for the prevention of gastric ulcers among those NSAID ulcers (8.2% vs. 16.6% for misoprostol and omeprazole, respectively; p < .05). Omeprazole was also not statistically different from misoprostol for gastric ulcer prevention in those whose NSAID use was complicated by an active H. pylori infection. Omeprazole was not significantly different from 300 mg of ranitidine for the prevention of NSAID gastric ulcers (14.6% vs. 11.6%, respectively, p = .56). Duodenal ulcers were over represented among H. pylori infected NSAID users and duodenal ulcer prevention was more sensitive to acid suppression than gastric ulcer. Conclusion. The OMNIUM and ASTRONAUT trials may have provided an unrealistic sense of security regarding the effectiveness of omeprazole for protection against ulcer recurrence in chronic NSAID users.  相似文献

采用营养液培养的方法,对缺硫脐橙叶片的光合特性进行了研究。结果表明,在缺硫情况下,脐橙叶片的净光合速率(Pn)、光呼吸速率(Pr)、光合色素含量、可溶性蛋白质含量、初始荧光(Fo)、光化学效率(Fv/Fm)、最大荧光(Fm)和电子传递速率(ETR)显著下降,而光呼吸/光合比(Pr/Pn)显著升高。缺硫脐橙植株的光合能力降低,可能是叶绿体发育不全或特性功能蛋白含量不足所致。  相似文献

通过连续两年田间试验,比较纽荷尔脐橙高接在两个不同基砧/中间砧组合上的产量表现,并探讨其原因。结果表明,红桔/锦橙组合投产显著地晚于枳/国庆一号温州蜜柑组合(对照);其晚实原因在于:红桔/锦橙组合比对照枝条加长生长旺、加粗生长弱、枝条较直立,树冠郁闭;红桔/锦橙组合树体光合能力受抑制;红桔/锦橙组合叶片可溶性糖含量低于对照,而淀粉含量高于对照;红桔/锦橙组合叶片N、P含量比对照低,Ca含量、C/N水平比对照高。  相似文献

目的:观察埃索美拉唑联合瑞巴派特、阿莫西林、克拉霉素治疗幽门螺杆菌阳性胃溃疡临床疗效。方法:60例确诊的Hp阳性胃溃疡患者随机分为对照组(30例)和治疗组(30例),其中对照组患者给予奥美拉唑+阿莫西林+克拉霉素三联法治疗,实验组给予埃索美拉唑+瑞巴派特+阿莫西林+克拉霉素四联法治疗。观察比较两组患者临床症状缓解情况,溃疡愈合率、Hp根除率及溃疡复发率。结果:①经过治疗,所有患者腹痛、腹胀、反酸、暧气等临床症状积分均显著降低(P<0.01),且治疗组下降程度大于对照组,两组间差异有统计学意义(P<0.05)。②治疗组患者痊愈率为60.00%、总有效率为93.33%,明显高于对照组痊愈率(43.33%)和总有效率(80.00%),两组间差异有统计学意义(P<0.05)。③治疗组S2期获得率、溃疡愈合率和Hp根除率分别为93.33%、96.67%和93.33%,显著高于对照组60.00%的S2期获得率、70.00%的愈合率和83.33%的根除率(P<0.01或0.05)。④随访1年后,治疗组患者溃疡复发率为11.54%,与对照组32.00%的复发率比较差异有显著性(P<0.05)。结论:四联疗法治疗幽门螺杆菌阳性胃溃疡可有效缓解患者临床症状,提高溃疡愈合质量,根除Hp感染,减少复发,效果优于三联疗法。  相似文献

The methods of GC with fused silica capillary column, its Kovats retention index of GC and GC-MS-DS etc. We were used to study the aromatic compounds of the peel essential oil of Citrus sinensis (L.) Osbeck and aromatic concentrates of fruit flesh. 29 chemical constituents from the peels and 40 chemical constituents from the fruit flesh were identified. The trace constituents α-sinensal 0.01% and β-sinensal 0.01% are the special aroma ones for Citrus sinensis. At the same time, the changes flavor components in Citrus sinensis peels and fruit flesh during storage were also explored.  相似文献

Four-step protocol was established for the in vitro regeneration of Citrus sinensis cv. Tarocco somatic embryos that were morphologically similar to small somatic embryos in vivo. The regeneration procedure comprises a mechanical destruction of embryogenic culture to obtain proembryogenic cell masses (PEMs) (step 1) followed by culturing on three different media (steps 2–4). The approach developed allows in vitro simulating somatic embryogenesis by dividing this process into three partially independent steps: PEM → globular somatic embryo → heart-shaped somatic embryo → somatic embryo with developed cotyledons. The highest frequency of morphogenetic stage transition was 64, 40, and 26%, respectively. It was shown that the first step (PEM → globular embryo) was associated with the formation of heterogeneous population of spherical bodies 50–500 μm in diameter, among which about 40% were somatic embryos at globular stage. The scheme is offered of alternative pathways for the development of spherical bodies in vitro, and interrelations between their sizes and ability to direct morphogenesis are discussed.  相似文献

锦橙(Citrussinensis(L.)Osbeckcv.Jincneng)珠心愈伤组织悬浮细胞通过γ射线诱变与高浓度羟脯氨酸离体选择的方法,获得了抗性细胞系,并再生植株。抗性细胞系及其再生植株比供体(对照)的抗寒性分别提高1.4℃和24℃,连续3年测定结果表明再生植株抗寒性的增强是稳定的。其叶片总DNA的RAPD分析说明供体与变异植株可能在DNA水平上存在细微的差异,冷驯化进一步加强抗性细胞系与再生植株在抗氧化酶SOD、过氧化氢酶(CAT)活性和脯氨酸(Pro)、丙二醛含量上的差异。变异系抗性增强的生理基础涉及Pro代谢外的其他途径,抗氧化酶活性的提高与其抗寒性的增强有关。  相似文献

Stress gastric ulcer is a serious complication, but the mechanism involved is not fully clarified. It is well known that mucosal cell apoptosis plays a crucial role in the pathogenesis of gastric ulceration. Recent studies have shown that endoplasmic reticulum (ER) stress is an important pathway leading to cellular apoptosis. To investigate the role of ER stress in the pathogenesis of stress gastric ulcer, we studied the alteration in the expression of ER stress markers GRP78 (glucose-regulated protein 78) and caspase-12 (an ER stress-specific proapoptotic molecule) and their relations with gastric mucosal apoptosis during development of stress gastric lesions in the water-immersion and restraint stress (WRS) model in rats. Rats developed severe gastric lesions after 6 h of WRS. Typical apoptosis was observed at the edge cells of WRS induced gastric lesions. Western blot analysis showed that GRP78 and activated caspase-12 were over-expressed in the gastric tissues of WRS rats. Immunohistochemical analysis demonstrated that increased GRP78 and caspase-12 were distributed only under the lesions. In addition, dithiothreitol and tunicamycin (ER stress inducers), which increased the expression of GRP78 and activated caspase-12, caused gastric mucosal injury and mucosal cell apoptosis in vitro. These findings suggest that ER stress might be involved in the development of stress gastric ulcer through an apoptotic mechanism.  相似文献

Pistils and various fruit explants of Citrus limon L. Burm. f. and Citrus sinensis L. Osbeck were cultivated in vitro . Basal medium as well as medium supplemented with IAA, GA3 or benzylaminopurine, supported growth of all explants for more than one year. Pistils did not enlarge considerably, but gave rise to active callus growth; callus proliferation and viability was enhanced by all hormones. Culture of fruit slice explants resulted, in addition to peel hypertrophy and callus proliferation, in a marked growth of two distinct types of juice vesicles. The growth of juice vesicle explants was promoted by all three growth hormones. – It is suggested that the successfully prolonged in vitro culture of various fruit explants, and especially of juice vesicles, may aid in studies of fruit development and physiology.  相似文献

Citrus sinensis chromosomes, although small in size, present a remarkable differentiation of bands with the fluorochromes CMA and DAPI. These bands suggest that some heteromorphisms are fixed in this species. To investigate the extension of these heteromorphisms, ten cultivars of C. sinenesis were analysed with CMA/DAPI staining and, in some of them, the 18S–5.8S–25S rRNA and 5S rRNA genes were located by in situ hybridization. CMA/DAPI staining showed exactly the same CMA+/DAPI banding pattern for all cultivars. In situ hybridization revealed three 18S–5.8S–25S rRNA gene sites, two proximally located on two similar chromosomes and one terminally located on a third non-related chromosome. Two 5S rRNA gene sites were observed in this species, with one located proximal to the telomeric 18S–5.8S–25S rDNA site. Both cytological approaches revealed an invariable, heterozygotic karyotype among sweet orange cultivars. Based on these data, the putative hybrid origin of the species is discussed.
