Concentrations of adrenal steroid metabolites in feces are routinely used to assess the welfare of animals that are the subject of conservation efforts. The assumption that low and declining corticoid concentrations indicate the absence of stress and acclimation, respectively, is often made without experimental support or wild-animal comparisons, although intrinsic control of adrenal steroids might occur even under ongoing stress and distress. We adopted the capture and 11-week captivity of 18 black ( Diceros bicornis : 11 males, seven females) and 52 white ( Ceratotherium simum : 22 males, 30 females) rhinoceros as an experimental test of the relationship between corticoid concentrations and stress (translocation) and measured for suppressed gonad function as an indicator of distress – the biological cost of cumulative stressors. Fecal samples collected from the rectum at capture and during captivity were stored frozen and their corticoid, and androgen (in males) or progestin (in females), concentrations determined by radioimmunoassay. Corticoid profiles followed the expected pattern of being two to five times pre-capture levels (ng g−1: black rhino: female 24.5±3.7, male 23.9±2.2; white rhino: female 16.3±1.6, male 12.3±2.4) for up to 17 days after capture and declined with time in captivity. Black rhinoceros and male white rhinoceros corticoids declined below pre-capture values and were associated with suppressed levels of androgens and progestins with increased time in captivity. Declining corticoids could not be interpreted as acclimation or the absence of stressors, without also measuring for distress in African rhinoceros. White rhinoceros female corticoid values remained elevated, although their gonad steroid levels were also suppressed. We discuss our findings for the management of rhinoceros in the wild and captivity.  相似文献

We compared the efficiency of succinylcholine chloride, xylazine hydrochloride and carfentanil/xylazine mixtures in immobilizing 364 free-ranging moose (Alces alces) between 1987 and 1997 in Québec (Canada). With succinylcholine chloride (0.070, 0.062, 0.051 mg/kg of estimated body weight for calves, juveniles and adults), 63% of the 252 immobilization attempts led to complete immobilization and marking, whereas 7% of the darted animals died of respiratory paralysis during handling. The moose took an average of 13 min to lay down after darting (down time). Injection of xylazine (3.67-4.22 mg/kg) permitted sedation (the animal laid down but got up again when approached) or complete immobilization in 78% of the 40 darted adult moose, the mean down time being 8.7 min. No mortality was noted with this drug but 58% of the marked animals were only sedated. The use of RX821002A (0.058 mg/kg) as an antagonist, permitted a mean recovery time of 2.8 min after intravenous injection. With the carfentanil/xylazine mixtures (0.0071 and 0.181 mg/kg), 96% of the immobilization trials (n = 72) led to complete (88%) or partial (8%) immobilization, but 6% of the moose died several days after capture. The mean down time was 6.6 min, and injection of naltrexone (0.709 mg/kg) antagonized the effect of the immobilizing agent within 3.7 min. The respiratory rate was higher (P < 0.05) among moose immobilized with xylazine (35/min) than among those immobilized with carfentanil/xylazine mixtures (19/min) but this variation could be related to a longer pursuit time (z = 3.60; P < 0.01) and higher stress levels during handling. Rectal temperature also was higher with xylazine but the difference was small (39.7 vs. 39.3, P = 0.03) and did not differ significantly between the sexes (P > 0.05). Considering loss of materials and helicopter flight time due to non-successful marking trials, carfentanil/xylazine mixtures were the least expensive ($333 Cdn/animal).  相似文献

The main factors that affect early survival, physical damage and stress reactions of sardine Sardina pilchardus after live capture and introduction to captivity were examined. A total of 2800 sardines were captured alive from commercial purse seiners in five trials off southern Portugal and monitored for 4 weeks in aquaculture tanks. Survival rates varied considerably between trials (from <20 to >80% after a month), with most deaths occurring in the first 5 days. Sardine early survival was affected by factors related to conditions at sea (catch composition, sea temperature and transportation density), during introduction to captivity (magnitude of thermal shock, land transportation duration and use of antibacterial treatment) and, possibly, their interaction. Physical damage was related to the probability of dying, with fish that died during the first week showing significantly higher scale loss and larger caudal fin erosion that those that were alive in the same period. For all stress variables measured (blood haematocrit, cortisol, glucose and ions in the plasma), the most extreme values were attained during introduction to captivity or in the first hours after. After 2 weeks in captivity, most variables had returned to levels close to those observed at the onset of purse-seine fishing, suggesting that maintenance conditions were adequate to permit a rapid recovery from fishing and transport stress.  相似文献

The tendency of juvenile and adult field-trapped Norway rats (Rattus norvegicus) to exhibit self-induced food deprivation in response to changes in their living environment was studied after different periods of residency in the laboratory environment. Self-imposed food deprivation resulting from the forced occupation of a novel environment (activity wheels) was greater for those animals tested immediately upon capture and after 25 weeks in captivity than for rats in captivity for 4 weeks. The deprivation response of juveniles was less than for adults immediately following capture but after 25 weeks in captivity the responses of the two age groups were similar. Deprivation responses following reintroduction to the home cage environment diminished over time in captivity. Wheel-running by adults was greatest immediately following capture whereas juveniles continued to show relatively high levels of wheel-running at 4 weeks in captivity. The results are discussed in terms of habituation to the laboratory environment and time-dependent changes in response to novelty.  相似文献

Researchers use the 13-lined ground squirrel for studies of hibernation biochemistry and physiology, as well as for modeling a variety of potential biomedical applications of hibernation physiology. It is currently necessary to capture research specimens from the wild; this presents a host of unknown variables, not least of which is the stress of captivity. Moreover, many investigators are unfamiliar with the husbandry of this species. The authors describe practical methods for their capture, year-round care (including hibernation), captive mating, and rearing of the young. These practices will allow the researcher to better standardize his or her population of research animals, optimizing the use of this interesting model organism.  相似文献

The bearded dragon (Pogona barbata: Agamidae) is a diurnal, oviparous, multi-clutching lizard from Australia. We examined plasma hormonal responses to capture in males and females during the spring breeding season. Corticosterone concentrations at capture (0 h; < or =3 min after capture) were low (males: 1.81+/-0.63 ng/ml; females 2. 23+/-0.47 ng/ml) and within sexes were unrelated to the time of the day, snout-vent length or, in females, reproductive condition (vitellogenic, gravid, assumed spent). Corticosterone concentrations at capture were significantly and inversely correlated with body condition in males, but not in females. Unexpectedly, neither sex showed significant changes in mean concentrations of corticosterone at 3.5 or 24 h after capture compared with 0 h values. Corticosterone concentrations at 3.5 h after capture did not differ between dragons bled at capture or not. Concentrations of progesterone in both the sexes did not change between 0 h and 3.5 or 24 h after capture. Testosterone concentrations in males at capture were moderate (10.1+/-2.2 ng/ml), and unchanged at 3.5 h after capture. The adrenocortical axis of adult bearded dragons in the breeding season seems remarkably unresponsive to capture compared with many other reptiles. Low adrenocortical responses to capture may be a feature of reptiles known to adjust well to captivity.  相似文献

During a 2-year study on wild boar ecology in a semiagricultural area of Northern Apennines (central Italy), 47 animals were caught with traps and subsequently immobilized with chemical restrain based on a 1:1 mixture of xylazine and tiletamine/zolazepam (1 ml contains: 50 mg of xylazine, 25 mg of tiletamine, and 25 mg of zolazepam). Effective chemical immobilization was observed in 55% of the caught wild boars, and a high capture mortality rate occurred (in 10.6% of wild boars). A posteriori analysis, by means of a logistic regression model, was performed to assess potential factors associated with wild boar chemical immobilization failure. Underdosage and increase in the number of animals caught together significantly increase the wild boar chemical restrain failure, while the capture mortality appears to be positively related to the sole trap size alone. The stress developed during the capture plays a relevant role influencing the outcomes of the anesthesia. Thus, the use of alternative handling or catching procedures with the aim to reduce stress in captured animals could maximize the safety and the efficiency of the wild boar chemical immobilization.  相似文献

Reproductively active female red gurnard Chelidonichthys kumu were captured on long-lines, and placed in confinement tanks for 24, 48, 72 and 96 h to examine the effect of capture and confinement on reproductive parameters (experiment I). Plasma cortisol at the time of capture was elevated to levels typical of stressed fish in other species (53–125 ng ml−1). Final plasma cortisol levels in red gurnard confined for any length of time were not significantly different from one another (ranging from 17 to 43 ng ml−1), indicating that fish were chronically stressed when held in captivity for up to 96 h after capture. When initial and final plasma cortisol levels were compared within confinement groups, cortisol decreased significantly after 24 and 96 h of confinement indicating that some acclimation to captivity may have occurred. In contrast, plasma 17β-estradiol (E2) and testosterone (T) levels decreased significantly to levels comparable to those in post-spawned fish, after any period of confinement, and remained low throughout the experiment. Another group of fish was captured and confined in the same manner as experiment I but subjected to repeated blood sampling every 24 h, until 96 h post-capture. In these fish, plasma cortisol levels decreased significantly from 127 ng ml−1 after 24 h confinement and thereafter showed no change (25–45 ng ml−1). Plasma E2 decreased significantly after 72 h of confinement while plasma T showed no change from levels at capture. Increased amounts of follicular atresia were found in vitellogenic oocytes of fish confined for longer periods of time in experiment I, indicating that capture and confinement stress affect reproduction negatively in captive wild fish.  相似文献

Fifteen male white-tailed deer (Odocoileus virginianus) were administered xylazine hydrochloride (1 mg/kg BW i.m.), xylazine hydrochloride (1 mg/kg i.m.) followed by caffeine sodium benzoate (10 mg/kg i.m.), xylazine hydrochloride (0.5 mg/kg i.m.) and ketamine hydrochloride (4.5 mg/kg i.m.), and xylazine hydrochloride (1 mg/kg i.m.) followed by yohimbine hydrochloride (0.125 mg/kg i.m.), in a Latin Square design. Mean dry matter intake (DMI) for 4 days pre-treatment was compared to each of 4 days post-treatment. A significant (P less than 0.01) decrease in DMI was found only on the first day following treatment for each of the four drug combinations. The percent decreases in DMI on the first 24-hr period after immobilization were: xylazine hydrochloride 47%, xylazine hydrochloride/caffeine sodium benzoate 36%, xylazine hydrochloride/yohimbine hydrochloride 36%, and xylazine hydrochloride/ketamine hydrochloride 31%. The xylazine hydrochloride/ketamine hydrochloride combination was found to be insufficient to adequately sedate the deer. The use of caffeine or yohimbine hydrochloride is recommended to reduce recumbency time, but offers no improvement in xylazine hydrochloride-induced anorexia.  相似文献

Understanding the changes in genetic variance which may occur as populations move from nature into captivity has been considered important when populations in captivity are used as models of wild ones. However, the inherent significance of these changes has not previously been appreciated in a conservation context: are the methods aimed at founding captive populations with gene diversity representative of natural populations likely also to capture representative quantitative genetic variation? Here, I investigate changes in heritability and a less traditional measure, evolvability, between nature and captivity for the large milkweed bug, Oncopeltus fasciatus, to address this question. Founders were collected from a 100-km transect across the north-eastern US, and five traits (wing colour, pronotum colour, wing length, early fecundity and later fecundity) were recorded for founders and for their offspring during two generations in captivity. Analyses reveal significant heritable variation for some life history and morphological traits in both environments, with comparable absolute levels of evolvability across all traits (0-30%). Randomization tests show that while changes in heritability and total phenotypic variance were highly variable, additive genetic variance and evolvability remained stable across the environmental transition in the three morphological traits (changing 1-2% or less), while they declined significantly in the two life-history traits (5-8%). Although it is unclear whether the declines were due to selection or gene-by-environment interactions (or both), such declines do not appear inevitable: captive populations with small numbers of founders may contain substantial amounts of the evolvability found in nature, at least for some traits.  相似文献

Haptoglobin (Hp) is an acute-phase protein synthesized in the liver that circulates at elevated concentrations in response to tissue damage caused by inflammation, infection, and trauma. As part of a larger study, sera Hp concentrations were measured in temporarily captive (n = 21) and free-range (n = 38) western stock juvenile Steller sea lions (Eumetopias jubatus) sampled from 2003 to 2006. Baseline Hp concentration at time of capture was 133.3 +/- 17.4 mg/dl. Temporarily captive animals exhibited a 3.2-fold increase in Hp concentrations during the first 4 wk of captivity, followed by a return to entry levels by week 5. Haptoglobin levels were not influenced by age, season, or parasite load. There was a significant positive correlation between Hp concentrations and white blood cell count (P < 0.001) and globulin levels (P < 0.001) and a negative correlation to red blood cell count and hematocrit (P < 0.001 for both). There was no correlation between Hp levels and platelet count (P = 0.095) or hemoglobin (P = 0.457). Routine blubber biopsies collected under gas anesthesia did not produce a measurable Hp response. One animal with a large abscess had an Hp spike of 1,006.0 mg/dl that returned to entry levels after treatment. In conclusion, serum Hp levels correlate to the stable clinical health status observed during captivity, with moderate Hp response during capture and initial acclimation to captivity and acute response to inflammation and infection.  相似文献

Hematologic and serum chemistry values were determined for 20 adult captive European wildcats (Felis silvestris) in Lleida (Catalonia, Spain). Seven wildcats (4 females and 3 males) were captured in the wild and 13 (4 females and 9 males) were born and raised in captivity. Samples were collected between September and December from 1993 to 1998. Blood was obtained by jugular venipuncture after administration of either ketamine and xylazine or ketamine and medetomidine. Females had significantly higher mean eosinophil counts, albumin concentration, (and A/G ratio) and lower mature neutrophil counts, although these differences were not clinically relevant. Results for many of the blood parameters fall within the reference range for domestic cats. Mean values for alanine aminotransferase, aspartate aminotransferase, creatine phosphokinase, and lactate dehydrogenase activities as well as blood urea nitrogen, glucose, and sodium concentrations were higher than the upper limit of the reference range for domestic cats.  相似文献

Telazol–xylazine and ketamine–xylazine are versatile and safe drug combinations that are used frequently for chemical immobilization of cervids. Although neither combination consistently offers rapid induction and recovery, we hypothesized that a combination of Telazol, ketamine, and xylazine (TKX) would provide a safe and effective alternative for immobilization of white-tailed deer (Odocoileus virginianus). During a 2-stage study, we evaluated the effectiveness of yohimbine and tolazoline as alpha2-adrenergic antagonists (2005–2006), and characterized the factors that affected chemical immobilization of male deer with a targeted dose of telazol (2.20 mg/kg), ketamine (1.76 mg/kg), and xylazine (0.44 mg/kg), using explosive-charged darts (2007–2010). During the first stage, we randomly assigned deer to antagonist treatments, including a control group that did not receive an antagonist (n = 8), a tolazoline (4 mg/kg) treatment (n = 16), and a yohimbine (0.11 mg/kg) treatment (n = 15). Recovery times were longer (P = 0.0013) for control (150.6 ± 21.7 min) and yohimbine (74.5 ± 13.1 min), compared with tolazoline (12.5 ± 12.3 min). Tolazoline resulted in faster and more complete recovery compared with the frequent incomplete antagonism and ataxia observed with yohimbine. During the second stage, 56 immobilization events (2007–2010) with TKX yielded a mean induction time of 7.8 minutes (SE = 0.44). Repeated-measures analyses indicated that induction and recovery were affected by body weight, with larger males taking longer to become recumbent (P = 0.08), but they recovered more rapidly (P = 0.003) following administration of tolazoline. Physiological parameters we measured under anesthesia were within normal ranges for white-tailed deer; however, initial temperature was higher (β = −0.86) for younger males (P = 0.014). Final physiological parameters were closely related to initial measurements, with rectal temperature being the most preserved (β = 0.90); heart and respiration rate declined (β < 0.60) during anesthesia. Our results indicate that TKX may be useful for chemically immobilizing white-tailed deer, and we recommend tolazoline as an antagonist for xylazine. © 2012 The Wildlife Society.  相似文献

As the only obligatorily predatory primates, tarsiers are notoriously difficult to keep successfully in captivity. Here we report empirical and experimental results from a 5-year study of behavior and life history in captive Tarsius bancanus. Four reproducing adult tarsiers used space nonrandomly, preferring small-diameter vertical or near-vertical locomotor substrates at midlevel enclosure heights (1.2–2.1 m) for sleeping, scanning, and prey capture. The tarsiers were completely nocturnal, and spent 78% of the scotophase scanning, 13% sleeping, and 9% in prey capture and other activities. Only live crickets were eaten; prey capture rates were highest in the first hour after waking, but overall activity rates were highest later in the scotophase. Adult males and nonpregnant or lactating females ingested approximately 44.7–49.7 kJ/day. Growing and lactating individuals ingested approximately 84.4–94.1 KJ/day. An energetically conservative, sit-and-wait predatory strategy was employed, in which 88% of capture attempts were successful. Most successful prey captures involved reaching for, or leaping from, 90° or 60° supports in a horizontal or downward direction onto prey less than 0.6 m away. Virtually all prey captures were in arboreal locations, despite much higher densities of crickets on enclosure floors. Prey capture rates during the first hour of the nocturnal activity period were positively correlated with arboreal cricket densities. At constant arboreal cricket densities, capture rates were negatively correlated with ambient light intensity, with optimum levels for prey capture ranging from 0.1 to 2.0 Lux. In terms of social behavior, these T. bancanus were nongregarious. Females enforced interindividual spacing by chasing and displacing males. Chase/displacement rates increased significantly during late pregnancy and lactation, apparently in an attempt to keep males from harassing infants. There was no direct male parental care. Infants were precocial at birth, and grew at a rate of 0.35–0.5 g/day, until nutritional weaning at approximately 60 days of age. October 2001 to January 2002, captive free-ranging white-tailed deer (Odocoileus virginianus) were immobilized with a combination of carfentanil citrate and xylazine hydrochloride. From this study, we selected a dose of carfentanil/xylazine for the purpose of comparing immobilization parameters and physiologic effects with those of a combination of tiletamine and zolazepam (Telazol) and xylazine. Animals were initially given intramuscular injections of 10 mg xylazine and one of four doses of carfentanil (i.e., 0.5, 1.0, 1.5, and 2.0 mg). A carfentanil dose of 1.2 mg (x +/- SD = 23.5 +/- 3.2 microg/kg) and 10 mg xylazine (0.2 +/- 0.03 mg/kg) were selected, based on induction times and previously published reports, to compare with a combination of 230 mg of Telazol (4.5 +/- 0.6 mg/kg) and 120 mg xylazine (2.3 +/- 0.3 mg/kg). Time to first observable drug effects and to induction were significantly longer for deer treated with carfentanil/xylazine than with Telazol/xylazine (P < 0.01). Hyperthermia was common in deer immobilized with carfentanil/xylazine, but heart rate, respiration rate, and hemoglobin saturation were within acceptable levels. Degree of anesthesia of deer immobilized with Telazol/xylazine was superior to deer immobilized with carfentanil/xylazine. The combination of 120 mg of naltrexone hydrochloride and 6.5 mg of yohimbine hydrochloride provided rapid and complete reversal (1.9 +/- 1.1 min) of carfentanil/xylazine immobilization. Animals immobilized with Telazol/xylazine had long recovery times with occasional resedation after antagonism with 6.5 mg of yohimbine. The combination of carfentanil and xylazine at the doses tested did not provide reliable induction or immobilization of white-tailel (leer even though drug reversal was rapid and safe using naltrexone and yohimbine.  相似文献

In vivo imaging of rats represents an important tool for outcome evaluation in research on stroke, brain trauma, and other neurologic diseases. Since sedation of animals is necessary to avoid artifacts, a mixture of ketamine and xylazine is frequently used for anesthesia. We assessed the suitable dosage of narcotics and its correlation to severe respiratory adverse events in 269 cases of ketamine/xylazine anesthesia in male Wistar rats for performance of magnetic resonance imaging after middle cerebral artery occlusion (MCAO) or sham surgery. Anesthesia depth was not measured. Anesthesia was efficacious in avoiding movement artifacts during imaging. Necessary dosage was lower if rodents were subjected to MCAO instead of sham surgery, if body weight was below baseline, and if time since surgery was short. If anesthesia was induced during the first 2 days after surgery in animals with body weight loss, necessary dose rates were 27% below doses required for rats more than 10 days post-surgery with body weight above baseline (91.4/8.3 versus 125.1/11.3 mg of ketamine/xylazine/kg). A dose adaptation scale for the prediction of necessary dose rates was developed. Apnea developed in 3.3% of all animals. Use of ketamine/xylazine anesthesia for imaging procedures is feasible and safe, though it is associated with a small risk of respiratory arrest. In case of apnea, inspiration can be provoked by a puff of air into the rat's pelt. If unsuccessful, endotracheal intubation and mechanical ventilation are needed until spontaneous breathing is restored or xylazine effects are antagonized.  相似文献

Red foxes (Vulpes vulpes) were immobilized with one of the following drug combinations: ketamine/xylazine (n = 22), ketamine/promazine (n = 35), ketamine/midazolam (n = 13), or tiletamine/zolazepam (n = 22). Foxes given ketamine/xylazine had the shortest induction and longest recovery times relative to other drug combinations, whereas foxes given ketamine/midazolam had the longest induction times. Recommended doses for the various combinations are given. Foxes given ketamine/xylazine were given either 0.1, 0.2, 0.4 mg/kg yohimbine, or saline 40 min after anesthetic induction. Administration of yohimbine significantly shortened arousal and recovery times relative to control values (P less than 0.001).  相似文献

This study examined adrenal and gonadal responses to capture and to longer term captivity in males of the scincid lizard, Egernia whitii. Animals subjected to acute capture stress in the field exhibited a rapid rise in plasma corticosterone concentrations that had not attenuated by 4 h after capture; plasma testosterone concentrations decreased significantly over this period. Animals returned to captive conditions in the laboratory showed little change in plasma corticosterone concentrations by 1 week, but concentrations were significantly lower at 4 weeks. Plasma testosterone did not vary significantly during this period. Sampling over a diel period indicated that plasma corticosterone concentrations vary little over the daylight hours; however, there was a significant decrease between the samples taken at 17:00 h and 21:00 h. These results suggest that, as in other species, the acute adrenal response to capture stress may confound assessment of other physiological parameters, although if experiments are carried out during daylight hours, time of sampling should have little influence on plasma corticosterone concentrations. The results also suggest that males of E. whitii require at least a week to adapt to captivity, although further studies investigating different captive conditions are warranted for this social species.  相似文献

Baltic salmon brood fish were investigated for the presence of Flavobacterium psychrophilum in the kidney, spleen, brain and sexual products (ovarian fluid, unfertilised eggs and milt). Samples for bacteriology were taken at capture, when the fish were ascending their native river to spawn, and after a period of captivity in indoor pools, at stripping. During captivity, abnormal wiggling behaviour was recorded in some of the fish. Bacterial samples were taken to determine if F. psychrophilum had any role in the aetiology of the condition. Furthermore, the presence of F. psychrophilum on egg surfaces during incubation was investigated. F. psychrophilum was isolated from internal organs and/or sexual products in 7 out of 50 (14.0%) fish sampled at capture and 63 out of 272 (23.2%) fish sampled at stripping. The bacteria was isolated from either spleen or gonads in 2 out of 19 (10.5%) fish with abnormal wiggling behaviour but no bacteria was isolated from the brain. No F. psychrophilum was isolated from eggs at the eyed stage. Just before hatching, the bacterium was isolated from 5 out of 15 (33.3%) family groups. The present study shows that Baltic salmon brood fish are carriers of F. psychrophilum during their spawning migration. The presence of the bacteria in sexual products from both females and males indicates that transmission from the brood fish to the offspring should be considered an important route of infection.  相似文献

Sixteen captive female red deer were successfully anesthetized to surgically implant a telemetry system. The deer were immobilized with (mean±SD) 1.79±0.29 mg/kg xylazine and 1.79±0.29 mg/kg tiletamine/zolazepam given intramuscularly with a dart gun. Anesthesia was maintained for 69±2 min using a total intravenous protocol with a catheter placed in the jugular vein. Group X received xylazine (0.5±0.055 mg/kg/hr) and group D, detomidine (2±0.22 μg/kg/hr), both in combination with ketamine (2±0.02 mg/kg/hr) and midazolam (0.03±0.0033 mg/kg/hr), as a constant rate infusion. Anesthesia was reversed with 0.09±0.01 mg/kg atipamezole and 8.7±1.21 μg/kg sarmazenil given intravenously in both groups. These drug combinations provided smooth induction, stable anesthesia for surgery, and rapid recovery. Respiratory depression and mild hypoxemia were seen, and we, therefore, recommend using supplemental intranasal oxygen.  相似文献
