Nine alkaloids (acridine, aristolochic acid, atropine, berberine, caffeine, nicotine, scopolamine, sparteine, and strychnine) were evaluated as feeding deterrents for gypsy moth larvae (Lymantria dispar (L.); Lepidoptera: Lymantriidae). Our aim was to determine and compare the taste threshold concentrations, as well as the ED50 values, of the nine alkaloids to determine their potency as feeding deterrents. The alkaloids were applied to disks cut from red oak leaves (Quercus rubra) (L.), a plant species highly favored by larvae of this polyphagous insect species. We used two-choice feeding bioassays to test a broad range of biologically relevant alkaloid concentrations spanning five logarithmic steps. We observed increasing feeding deterrent responses for all the alkaloids tested and found that the alkaloids tested exhibited different deterrency threshold concentrations ranging from 0.1 to 10 mM. In conclusion, it appears that this generalist insect species bears a relatively high sensitivity to these alkaloids, which confirms behavioral observations that it avoids foliage containing alkaloids. Bioassay studies were conducted to investigate the influence of Dimilin (diflubenzuron), a chitinsynthetase inhibitor used for insecticidal control of the gypsy moth, Lymantria dispar, on the development and viability of a microsporidian pathogen of L. dispar. Before or after an infection with a Nosema species, L. dispar larvae were fed Dimilin in sublethal dosages. Dimilin fed to L. dispar larvae at 0.65 ng/cm2 diet surface resulted in a total larval mortality of 53%. Although the microsporidian infection alone did not cause high mortality rates (9%), mortality increased to 96% when L. dispar larvae were inoculated with both Dimilin and Nosema spores. When Dimilin was fed to the larvae 24 h before or 6 days after inoculation with the microsporidium, the number of mature spores produced was significantly reduced. When Dimilin was fed to the larvae 24 h after microsporidian inoculation, the number of spores produced was not significantly reduced. Spores that were produced in larvae after Dimilin had been ingested with the diet were less infectious than spores produced in control larvae; the experimental infection rate decreased from 94% when spores obtained from control larvae were used, to 48 or 10% when spores obtained from larvae fed Dimilin 24 h or 6 days after Nosema inoculation, respectively, were used. Mature microsporidian spores washed in Dimilin solution prior to oral inoculation, however, were as infectious as spores stored in liquid nitrogen. We have shown that Dimilin interferes with the establishment of the parasite in its host. In addition, when Nosema sp. succeeds in infecting the L. dispar host despite treatment with Dimilin, the microsporidium does not develop optimally and spore production is reduced.  相似文献

Information is given concerning two standard buffer solutions suitable as pH references in 30, 40, and 50 mass% dimethyl sulfoxide (DMSO)/H2O mixed solvents at subzero temperatures from −20 to 0 °C, with the intention of establishing a pH (designated pH*) scale. The two buffers selected were the ampholytes N,N-bis(2-hydroxyethyl)-2-aminoethane sulfonic acid (“bes”) and N-tris(hydroxymethyl)methylglycine (“tricine”), and the reference standard consisted of equal molal quantities of the buffer and its respective sodium salt. The assignment of pH* values was based on measurements of the emf of cells without liquid junction of the type: Pt;H2(g,1 atm) ¦Bes, Na Besate, NaCl ¦ AgCl;Ag and Pt;H2(g,1 atm) ¦Tricine, Na Tricinate, NaCl ¦AgCl;Ag and the pH* was derived from a determination of K2, the equilibrium constant for the dissociation process (Buffer)±/ai (Buffer) + H+.  相似文献

Calosoma sycophanta L. adults were fed either gypsy moth (Lymantria dispar L.) larvae or split grapes for set periods of time while their reproduction was monitored. Few female beetles reproduced unless fed gypsy moth larvae during the first week after they ended hibernation. Even females initially fed grapes that were later fed larvae had reduced reproduction. The implications these results have for relationships between beetle and gypsy moth populations are discussed.  相似文献

Summary Wire cages with different-sized meshes were placed on trunks and around leaves at different heights in oak trees and in forest litter. Gypsy moth, Lymantria dispar, instars II–V tethered with threads were placed in each cage (instars II–III only in leaf cages) as well as outside the cages. Predation of larvae decreased from near ground to mid-crown in trees and was highest in litter and very low on leaves. Predation in litter was not strongly related to cage type, suggesting that small, invertebrate predators were active there, but IV–V instars on trunks were mainly killed by relatively large predators, probably forest mice and shrews. Influences of the time of day and weather on predation were evaluated by observing tethered V–VI instars in litter and on tree trunks hourly. Ants, particularly carpenter ants (Camponotus ferrugineus) and Formica sp., and probably vertebrates, were conspicuous predators in the litter. Ants were most active at lower relative humidities, while other predators were apparently not influenced by humidity. No daily activity rhythms of predation were noted. Invertebrates appear to be important predators of larvae only in the litter whereas vertebrates, such as forest mice and shrews, also readily attack larvae on tree trunks.  相似文献

The gut microbiota plays an important role in pheromone production, pesticide degradation, vitamin synthesis, and pathogen prevention in the host animal. Therefore, similar to gut morphology and digestive enzyme activity, the gut microbiota may also get altered under plant defensive compound-induced stress. To test this hypothesis, Dendrolimus superans larvae were fed either aconitine- or nicotine-treated fresh leaves of Larix gmelinii, and Lymantria dispar larvae were fed either aconitine- or nicotine-treated fresh leaves of Salix matsudana. Subsequently, the larvae were sampled 72hr after diet administration and DNA extracted from larval enteric canals were employed for gut microbial 16S ribosomal RNA gene sequencing (338 F and 806 R primers). The sequence analysis revealed that dietary nicotine and aconitine influenced the dominant bacteria in the larval gut and determined their abundance. Moreover, the effect of either aconitine or nicotine on D. superans and L. dispar larvae had a greater dependence on insect species than on secondary plant metabolites. These findings further our understanding of the interaction between herbivores and host plants and the coevolution of plants and insects.  相似文献

Collaborative research was conducted at the INRA Research Centers to assess the microbial control potential of Beauveria bassiana- and Lecanicillium lecanii-based formulations against whiteflies in protected crops under Mediterranean conditions. Four series of small-scale glasshouse trials were performed in 1999 and 2000 in southern France. Two applications at 4–5 day intervals of Naturalis-L and Mycotal were conducted on young larvae of the greenhouse whitefly, Trialeurodes vaporariorum, at rates recommended by the manufacturers. Because of the expectation that environmental conditions prevailing in Mediterranean greenhouse crops may lead to greater climatic constraints for mycoinsecticide efficacy than in more temperate areas, manipulation of the greenhouse climate has been used to aim at optimizing mycoinsecticide efficacy. The climatic management strategy was mainly based on closing the ridge vents 2 h more at night-time in so-called “humid” glasshouse compartment than in a “dry” one. Thus, the daily period at high humidity (>90% RH) was two or three times longer in the “humid” compartment than in the “dry” one. In spite of this differential, mycoinsecticide treatments reduced numbers of surviving whitefly larvae by >85% in the “humid” compartment as expected as favorable, as well as in the “dry” compartment, expected as unfavorable. The results indicated clearly that both B. bassiana- and L. lecanii-based mycoinsecticides have a strong potential for microbial control of whitefly larvae infesting tomato crops at moderate ambient humidity in Mediterranean glasshouses. Our investigations provided strong arguments for explaining these unexpected results. The RH conditions prevailing in the targeted insect habitat should be greatly disconnected from that of the ambient glasshouse air. We suggest that strategies of mycoinsecticide optimization against phyllophagous insects in protected crops have to take into account factors acting on the leaf transpiration activity.  相似文献

Effects of exposure to maize pollen of event Bt176 (cultivar “Navares”) on the larvae of the European common swallowtail (Papilio machaon L.) were studied in the laboratory. First instar larvae were exposed to different pollen densities applied to leaf disks of Pastinaca sativa L. for 48 h. Pollen densities applied in this study were in the range recorded from the field. Larvae which were exposed to higher Bt maize pollen densities consumed more pollen and had a lower survival rate. The LD50 with regard to larvae surviving to adulthood was 13.72 pollen grains consumed by first-instar larva. Uptake of Bt maize pollen led to a reduced plant consumption, to a lower body weight, and to a longer development time of larvae. Effects on pupal weight and duration of the pupal period were present but less pronounced and smaller than effects on larvae. Larvae having consumed Bt-maize pollen as first instars had a lower body weight as adult females and smaller forewings as adult males. We conclude that possible effects of Bt maize on European butterflies and moths must be evaluated more rigorously before Bt maize should be cultivated over large areas.  相似文献

We examined the effects of CO2-mediated changes in the foliar chemistry of paper birch (Betula papyrifera) and white pine (Pinus strobus) on performance of the gypsy moth (Lymantria dispar). Trees were grown under ambient or enriched CO2 conditions, and foliage was subjected to plant chemical assays and insect bioassays. Enriched CO2 atmospheres reduced foliar nitrogen levels and increased condensed tannin levels in birch but not in pine. Foliar carbohydrate concentrations were not markedly altered by CO2 environment. Gypsy moth performance was significantly affected by CO2 level, species, and the CO2 x species interaction. Under elevated CO2 conditions, growth was reduced for larvae fed birch, while development was prolonged for larvae fed pine. Although gypsy moths performed better overall on birch than pine, birch-fed larvae were influenced more by CO2-mediated changes in host quality.  相似文献

We have determined the sequence of an oligonucleotide from the large ribosomal subunit RNA of Staphylococcus aureus whose methylation renders the organism resistant to erythromycin and other antibiotics (the “MLS” phenotype). Analysis of RNase A digests of [3H]methyl-, 32P-labeled RNA yielded the sequence GG · m26A · AAGACp, where m26A is an N6-dimethylated adenosine residue that in sensitive cells is unmethylated. Comparison with homologous sequences recently reported for Saccharomyces cerevesiae mitochondria indicates that an A to G mutation in this latter system mimics dimethylation in St. aureus with regard to functional consequences.  相似文献

Cyanobacteria (photoautotrophic prokariota) have potential for the control of pathogenic bacteria and fungi. The effect of intra and extracellular products from cyanobacterial strains on the growth of fungi isolated from “wood blue stain,” was tested. Extracellular products were obtained by concentration and sterilization of the culture medium where cyanobacteria were grown. Cyanobacterial substances promoted or inhibited fungal growth according to the fungal and cyanobacterial strains tested. Extracellular products from Nostoc muscorum 79a and the methanolic extract from Microchaete tenera 84b biomass inhibited growth of Sphaeropsis sapinea 2157 (64.7 and 775.6%, respectively). Extracellular products of Nostoc piscinale 59 and biomass methanolic extract from N. muscorum 79a produced the highest growth promotion of Trichoderma boningii 452 (105.0%) and T. viride 993 (136.7%). Extracellular products of the heterotrophic lactic acid bacterium Streptococcus termophilus were also tested and strongly inhibited (64–92%) all the fungal strains. The tested fungi have different sensitivity to the bioactive substances present in the biomass and/or the culture medium of the studied cyanobacteria and lactic acid bacterium. N. muscorum 79a, M. tenera 84b, and S. termophilus have potential to control the wood blue stain fungi by a friendly environmental alternative.  相似文献

The effects of different diatom species and types of substrates in combination with 0.45 μM GABA on the metamorphosis of Haliotis asinina larvae were tested. Diatom slurry elicited the best metamorphic response followed by Amphora sp., Amphora + Nitzschia and Nitzschia cf. frustulum in that order. With regards to substrate types, roughened plexiglass seemed to be the most preferred while fibrocement the least preferred surface. Overall, diatom slurry grown on plexiglass surface promoted the greatest number of metamorphosed H. asinina postlarvae. For economic considerations and practical reasons, chemical inducers like gamma-aminobutyric acid (GABA), should be used singly or separately from other settlement-inducing cues, such as the “substrate-diatom” complex.  相似文献

“Red tide” events are frequent and periodical in Bahía de Mazatlán, Sinaloa, México. Yet, the ones observed from 4 February to 4 June 2000, showed some distinctive features: First, the dinoflagellates Prorocentrum balticum (85%), P. mexicanum (5%), and Ceratium furca (5%), dominated the composition of the blooms; Second, the average cell abundance by date was 1.3×106 cells l−1, with a range of 3.5×103 to 24,500 × 103 cells l−1, well above previous records; Third, the temperature registered at 10–20 m deep was unusually cold (19 °C), below the normal average of 21.5 °C observed over the last 10 years. Salinity was high (35.9 psu) and showed very little influence on the water density gradient. A mean thermal stratification index (TSI), of 3.4, with a maximum of 7 °C, was observed throughout the period, in spite of a weak upwelling activity and intermittent strong NW winds which were unable to break up water column stratification. Temperature fluctuations at the surface and at the bottom layers showed a 30-day periodicity, suggesting some association with the lunar cycle. To explain the characteristics of the “red tides” registered in Bahía de Mazatlán during the winter–spring period of year 2000, it is proposed that the temperature and density stratification, stabilized further by internal waves that compensated for the weak upwelling activity and provided the necessary nutrients to the surface layer, favored the persistence and intensity of the harmful algal bloom events then observed.  相似文献

Mutant frequency “saturation” in Arabidopsis' “main” inflorescence after fast neutron or X-ray treatment of seeds was observed using cell populations with heterogeneous radiation-sensitivities. Mutant frequencies were determined in (1) morphologically normal and conspicuous plants, (2) main and lateral inflorescences, and (3) different M1-fertility classes. At moderate radiation doses (47 Gy fast neutrons and 233 Gy X-rays) significant differences in mutant frequencies between different M1-fertility classes were observed. This suggests that cell populations with heterogeneous sensitivities were studied. No significant differences were observed either between the mutant frequencies in main and lateral inflorescences or between the ones in inflorescences of morphologically normal and conspicuous plants. This suggests that these inflorescences were formed by cell populations with similar but heterogeneous sensitivities.  相似文献

A water-soluble glucuronan “protuberic acid”, [α]d22 −83.6° and purified from Kobayashia Nipponica, and its physicochemical properties were investigated.The purified protuberic acid was homogeneous as shown by zone electrophoresis, gel filtration over Sepharose 4B, and ultracentrifugation. The sedimentation coefficient was 1.8 S and its intrinsic viscosity was 1.1 dl/g. By gel filtration the molecular weight was estimated to be about 170 000. The results of periodate oxidation, methylation analysis, and partial acid hydrolysis indicated that this acidic polysaccharide has a linear structure of mainly 1,4-linkages and containing an acid-labile linkage. We examined toxicity of acephate to third-instar gypsy moth, Lymantria dispar (L.) (Lepidoptera: Lymantriidae), under different conditions of administration method, availability of food to larvae during bioassay, host plant, and activity of detoxifying enzymes. Larvae that had been fed field-collected foliage of white alder (Alnus rhombifolia Nutt.) were less susceptible 48 h after treatment with topically applied acephate if they were allowed to continue feeding on foliage during the bioassay period (LD50= 60.6 μg/g larva ) than if they were not (LD50= 13.5 μg/g larva ). All surviving larvae were replaced on their original food plant after the 48-h bioassay; of these, 14.4% of the larvae not fed during treatment died before pupation, compared with 1.3% of the larvae fed alder during treatment. The LD50 obtained for topically treated larvae reared and treated on Douglas-fir, Pseudotsuga menziesii (Mirb.) Franco, (51.1 μg/g larva) was comparable to that obtained for larvae fed alder (60.0 μg/g larva) throughout treatment. Larvae treated orally with acephate, however, were slightly more susceptible when reared on Douglas-fir (LC50, 20.3 ppm ) than when reared on alder (LC50, 27.0 ppm ). Post-treatment mortality in orally treated larvae was 10.3% in those fed alder and 9.5% in those fed Douglas-fir. Higher cytochrome P-450 activities in larvae reared on Douglas-fir apparently did not enhance tolerance to acephate. Both sexes of orally treated larvae took significantly longer to pupate than did controls on both foliage types, as did topically treated males fed Douglas-fir. Pupal weight generally was slightly, but not always significantly, higher in treated than untreated larvae under all dietary and treatment regimes.  相似文献

The universe of cellular forms has received scarce attention by mainstream neo-Darwinian views. The possibility that a fundamental trait of biological order may consist upon, or be guided by, developmental processes not completely amenable to natural selection was more akin to previous epochs of biological thought, i.e. the “bauplan” discussion. Thirty years ago, however, Lynn and Tucker studied the biological mechanisms responsible for defining organelles position inside cells. The fact that differentiated structures performing a specific function within the eukaryotic cell (i.e. mitochondrion, vacuole, or chloroplast) were occupying specific positions in the protoplasm was the observational and experimental support of the ‘morphogenetic field’ notion at the cellular level. In the present paper we study the morphogenetic field evolution yielding from an initial population of undifferentiated cells to diversified unicellular organisms as well as specialized eukaryotic cell types. The cells are represented as Julia sets and Pickover biomorphs, simulating the effect of Darwinian natural selection with a simple genetic algorithm. The morphogenetic field “defines” the locations where cells are differentiated or sub-cellular components (or organelles) become organized. It may be realized by different possibilities, one of them by diffusing chemicals along the Turing model. We found that Pickover cells show a higher diversity of size and form than those populations evolved as Julia sets. Another novelty is the way that cellular organelles and cell nucleus fill in the cell, always in dependence on the previous cell definition as Julia set or Pickover biomorph. Our findings support the existence of specific attractors representing the functional and stable form of a differentiated cell—genuine cellular bauplans. The configuration of the morphogenetic field is “attracted” towards one or another attractor depending on the environmental influences as modeled by a particular fitness function. The model promotes the classical discussions of D’Arcy Thompson and the more recent views of Waddington, Goodwin and others that consider organisms as dynamical systems that evolve through a ‘master plan’ of transformations, amenable to natural selection. Intriguingly, the model also connects with current developments on mechanobiology, highlighting the informational–developmental role that cytoskeletons may play.  相似文献

From March through April 1998, a massive “red tide” occurred in coastal waters of south China, including Hong Kong. The “red tide” rapidly killed various species of caged fish and affected coral fishes, killing a few of them, and caused great economic loss and ecological damage. Samples collected from a permanent station located in Port Shelter revealed a new dinoflagellate species, Karenia digitata which was suspected to be the causative species of this “red tide”. Species composition and abundance analysis revealed that an algal bloom persisted in Port Shelter during this entire period. Diatoms and dinoflagellates were the two main groups which dominated the phytoplankton and, in general, when there was an increase in the density of diatoms there was a decline in the density of dinoflagellates, and vice versa. The suspected “red tide” causative species together with other dinoflagellate species started to bloom in late February and reached their highest density on 18 March, when fish kills were first reported at Crooked Island, a semi-enclosed bay to the northeast of Hong Kong. During a 16-week period, dinoflagellate species dominated three times, and coincided with low wind speeds. Constant salinity and a continuing increase in sea surface water temperature suggested warm water intrusion into Hong Kong’s coastal waters during this “red tide” bloom period. Various nutrient elements, e.g. NH4-N, total Kjeldahl nitrogen (TKN), and PO4-P were high at the beginning of the bloom but experienced a sharp decrease thereafter. It is suggested that this early 1998 massive “red tide” in Hong Kong waters might have been triggered by a synchronous appearance of optimal climatic, nutritional and hydrographic conditions.  相似文献

Gypsy moth (Lymantria dispar) larvae displayed marked developmental resistance within an instar to L. dispar M nucleopolyhedrovirus (LdMNPV) regardless of the route of infection (oral or intrahemocoelic) in a previous study, indicating that in gypsy moth, this resistance has a systemic component. In this study, gypsy moth larvae challenged with the Amsacta moorei entomopoxvirus (AMEV) showed developmental resistance within the fourth instar to oral, but not intrahemocoelic, inoculation. In general, gypsy moth is considered refractory to oral challenge with AMEV, but in this study, 43% mortality occurred in newly molted fourth instars fed a dose of 5×106 large spheroids of AMEV; large spheroids were found to be more infectious than small spheroids when separated by a sucrose gradient. Developmental resistance within the fourth instar was reflected by a 2-fold reduction in mortality (18%–21%) with 5×106 large spheroids in larvae orally challenged at 24, 48 or 72 h post-molt. Fourth instars were highly sensitive to intrahemocoelic challenge with AMEV; 1PFU produced approximately 80% mortality regardless of age within the instar. These results indicate that in gypsy moth, systemic developmental resistance may be specific to LdMNPV, reflecting a co-evolutionary relationship between the baculovirus and its host.  相似文献
