Inputs of CH(4) from sediments, including methane seeps on the continental margin and methane-rich mud volcanoes on the abyssal plain, make the Black Sea the world's largest surface water reservoir of dissolved methane and drive a high rate of aerobic and anaerobic oxidation of methane in the water column. Here we present the first combined organic geochemical and molecular ecology data on a water column profile of the western Black Sea. We show that aerobic methanotrophs type I are responsible for methane oxidation in the oxic water column and ANME-1- and ANME-2-related organisms for anaerobic methane oxidation. The occurrence of methanotrophs type I cells in the anoxic zone suggests that inactive cells settle to deeper waters. Molecular and biomarker results suggest that a clear distinction between the occurrence of ANME-1- and ANME-2-related lineages exists, i.e. ANME-1-related organisms are responsible for anaerobic methane oxidation below 600 m water depth, whereas ANME-2-related organisms are responsible for this process in the anoxic water column above approximately 600 m water depth.  相似文献

We tested new strategies for the isolation of abundant bacteria from coastal North Sea surface waters, which included reducing by several orders of magnitude the concentrations of inorganic N and P compounds in a synthetic seawater medium. Agar plates were resampled over 37 days, and slowly growing colonies were allowed to develop by repeatedly removing all newly formed colonies. A fivefold increase of colonies was observed on plates with reduced nutrient levels, and the phylogenetic composition of the culture collection changed over time, towards members of the Roseobacter lineage and other alpha-proteobacteria. Novel gamma-proteobacteria from a previously uncultured but cosmopolitan lineage (NOR5) formed colonies only after 12 days of plate incubation. A time series of German Bight surface waters (January to December 1998) was screened by fluorescence in situ hybridization (FISH) with isolate-specific and general probes. During spring and early summer, a prominent fraction of FISH-detectable bacteria (mean, 51%) were affiliated with the Cytophaga-Flavobacterium group (CF) of the Bacteroidetes. One Cytophaga sp. lineage with cultured representatives formed almost 20% of the CF group. Members of the Roseobacter cluster constituted approximately 50% of alpha-proteobacteria, but none of the Roseobacter-related isolates formed populations of >1% in the environment. Thus, the readily culturable members of this clade are probably not representative of Roseobacter species that are common in the water column. In contrast, members of NOR5 were found at high abundances (>10(5) cells ml(-1)) in the summer plankton. Some abundant pelagic bacteria are apparently able to form colonies on solid media, but appropriate isolation techniques for different species need to be developed.  相似文献

Intact core tetraether membrane lipids of marine planktonic Crenarchaeota were quantified in water column-suspended particulate matter obtained from four depth intervals ( approximately 70, 500, 1,000 and 1,500 m) at seven stations in the northwestern Arabian Sea to investigate the distribution of the organisms at various depths. Maximum concentrations generally occurred at 500 m, near the top of the oxygen minimum zone, and the concentrations at this depth were, in most cases, slightly higher than those in surface waters. In contrast, lipids derived from eukaryotes (cholesterol) and from eukaryotes and bacteria (fatty acids) were at their highest concentrations in surface waters. This indicates that these crenarchaeotes are not restricted to the photic zone of the ocean, which is consistent with the results of recent molecular biological studies. Since the Arabian Sea has a strong oxygen minimum zone between 100 and 1,000 m, with minimum oxygen levels of <1 microM, the abundance of crenarchaeotal membrane lipids at 500 m suggests that planktonic Crenarchaeota are probably facultative anaerobes. The cell numbers we calculated from the concentrations of membrane lipids are similar to those reported for the Central Pacific Ocean, supporting the recent estimation of M. B. Karner, E. F. DeLong, and D. M. Karl ( Nature 409:507-510, 2001) that the world's oceans contain ca. 10(28) cells of planktonic Crenarchaeota.  相似文献

During the late Pleistocene, sapropels (layers of organic-carbon rich sediment) formed throughout the entire Eastern Mediterranean Basin in close association with glacial/interglacial transitions. The current theory for the mechanism of sapropel formation involves a density stratification of the water column, due to the invasion of a large quantity of low-saline water, which resulted in oxygen depletion of the bottom waters. Most workers believe that this low-salinity water was glacial meltwater that entered the Mediterranean via the Black Sea and a series of interconnected glacial lakes, but the suggestion also has been made that the freshwater originated from the Nile River. In this study the oxygen isotope values of planktonic foraminifera,Globigerinoides ruber, have been examined in six gravity cores and one piston core from the southern Levantine Basin, and compared with the oxygen isotope records ofG. ruber from other areas of the Eastern Mediterranean. This study deals mainly with the latest sapropel which was deposited approximately 7000 to 9000 years ago. Results indicate that Nile discharge probably does reduce salinities somewhat in the immediate area surrounding the mouth of the Nile, but this water is rapidly mixed with the highly saline waters of the easternmost Mediterranean.Using a mixing equation and surface water salinity limitations, an approximate oxygen isotope balance of surface waters was calculated for the time of latest sapropel deposition. This calculation shows that neither Nile River discharge nor Black Sea input (nor both together) are large enough to account for the large-scale oxygen isotope depletion associated with latest sapropel deposition in the Eastern Mediterranean. This suggests that part of the isotopic change at Termination I is probably due to increased surface water salinities during the last glacial maximum. In addition, evidence from the timing of sapropel 1 deposition and the dissolved oxygen balance indicates that deposition of the latest sapropel is associated with increased surface water production of biogenic material, as much as three times higher than that of present day.  相似文献

In the course of cruises by FRV Scotia to study the ecology of O-group gadoids in the northern North Sea in October 1977 and November 1978, the fish fauna of the upper 1 m of the water column were sampled with an improvised gear. The most abundant species were O-group rocklings and clupeids and three spined sticklebacks. The frequency of occurrence of the last named and their relatively high abundance (up to 99 individuals in a 30 min haul) suggests that sticklebacks are more widely distributed throughout the open North Sea than is presently supposed. Young rocklings may amount to a considerable biomass in the near-surface waters.  相似文献

Studies of microbial communities in areas of the world where permanent marine water column oxyclines exist suggest they are “hotspots” of microbial activity, and that these water features and the anoxic waters below them are inhabited by diverse protist taxa, including ciliates. These communities have minimal taxonomic overlap with those in overlying oxic water columns. Some ciliate taxa have been detected in multiple locations where these stable water column oxyclines exist; however, differences in such factors as hydrochemistry in the habitats that have been studied suggest local selection for distinct communities. We compare published data on ciliate communities from studies of deep marine water column oxyclines in Caricao Basin, Venezuela, and the Black Sea, with data from coastal, shallower oxycline waters in Framvaren and Mariager fjords, and from several deep‐sea hypersaline anoxic basins in the Eastern Mediterranean Sea. Putative symbioses between Bacteria, Archaea, and ciliates observed along these oxyclines suggests a strategy of cooperative metabolism for survival that includes chemosynthetic autotrophy and exchanges of metabolic intermediates or end products between hosts and their prokaryotic partners.  相似文献

Meristic variation among stocks of greater lizardfish Saurida tumbil through the western coasts of the Arabian Gulf and Sea of Oman was examined using meristic characters. Statistical analysis of meristic traits proposed that there is constrained migration of populations of greater lizardfish along the western coast of the Arabian Gulf and Sea of Oman. Overlapping of the two samples from the northern part of the Arabian Gulf (Iraq-Kuwait waters), three samples from the middle region of the Arabian Gulf (Bahrain-Qatar-Saudi Arabia) and two samples from the southern part of the Arabian Gulf/Sea of Oman (United Arab Emirates–Sultanate of Oman) suggested that there are three self-recruiting populations in the studied area. Inspection of the role of each meristic trait variable to Canonical discriminant analysis showed that changes among samples appeared to be linked with the pattern of distribution of water temperature and configuration of current in both the Arabian Gulf and Sea of Oman areas.  相似文献

Populations of the colonial hydroid Obelia geniculata in the White Sea reproduce asexually by frustule formation. Young medusae appear in the plankton during July and August. The number of medusae rarely exceeds 36 per m3, and the average number varies every year from 0.4 to 10 per m3. The size of medusae is smaller than reported from other regions. The umbrella of the largest recorded medusa was only 0.57 mm in diameter and the specimen had just 35 tentacles. Only a few mature medusae were found during the study. The colonies in the White Sea are epiphytic and grow only on laminarian thalli. At the beginning of July there are no colonies on thalli from the upper subtidal zone. By the end of August, colonies of O.␣geniculata had increased in density to 30 per m2. Hydroid recruitment was attributed to active frustule production by colonies living below that zone. The frustules detach from the stems of the hydroids and are found in plankton. Production of frustules on branches occurs continuously during colony growth until water temperatures climb above 0 °C. We found that water temperature in this Arctic environment is generally too low for medusa maturation and planula development in the species. Propagation by frustule formation is the principal means of reproduction in Obelia geniculata within the White Sea, and this phenomenon accounts for the species being a dominant epiphyte on laminarian thalli there.  相似文献

Distributions of prokaryotic cell size and morphology were studied in different areas of the Mediterranean Sea by using image analysis on samples collected from surface down to bathypelagic layers (max depth 4,900?m) in the Southern Tyrrhenian, Southern Adriatic and Eastern Mediterranean Seas. Distribution of cell size of prokaryotes in marine ecosystem is very often not considered, which makes our study first in the context of prokaryotic ecology. In the deep Mediterranean layers, an usually-not-considered form of carbon sequestration through prokaryotic cells has been highlighted, which is consistent with an increase in cell size with the depth of the water column. A wide range in prokaryotic cell volumes was observed (between 0.045 and 0.566?μm3). Increase in cell size with depth was opposed to cell abundance distribution. Our results from microscopic observations were confirmed by the increasing HNA/LNA ratio (HNA, cells with high nucleic acid content; LNA, cells with low nucleic acid content) along the water column. Implications of our results on the increasing cell size with depth are in the fact that the quantitative estimation of prokaryotic biomass changes along the water column and the amount of carbon sequestered in the deep biota is enhanced.  相似文献

An oligonucleotide primer, NITRO821R, targeting the 16S rRNA gene of unicellular cyanobacterial N2 fixers was developed based on newly derived sequences from Crocosphaera sp. strain WH 8501 and Cyanothece sp. strains WH 8902 and WH 8904 as well as several previously described sequences of Cyanothece sp. and sequences of intracellular cyanobacterial symbionts of the marine diatom Climacodium frauenfeldianum. This oligonucleotide is specific for the targeted organisms, which represent a well-defined phylogenetic lineage, and can detect as few as 50 cells in a standard PCR when it is used as a reverse primer together with the cyanobacterium- and plastid-specific forward primer CYA359F (U. Nubel, F. Garcia-Pichel, and G. Muyzer, Appl. Environ. Microbiol. 63:3327-3332, 1997). Use of this primer pair in the PCR allowed analysis of the distribution of marine unicellular cyanobacterial diazotrophs along a transect following the 67 degrees E meridian from Victoria, Seychelles, to Muscat, Oman (0.5 degrees S to 26 degrees N) in the Arabian Sea. These organisms were found to be preferentially located in warm (>29 degrees C) oligotrophic subsurface waters between 0 and 7 degrees N, but they were also found at a station north of Oman at 26 degrees N, 56 degrees 35'E, where similar water column conditions prevailed. Slightly cooler oligotrophic waters (<29 degrees C) did not contain these organisms or the numbers were considerably reduced, suggesting that temperature is a key factor in dictating the abundance of this unicellular cyanobacterial diazotroph lineage in marine environments.  相似文献

In laboratory experiments, bacterioplankton were incubated under different nutrient conditions, and the percentage of bacteria exhibiting a polysaccharidic capsule (capsulated bacteria) and that of CTC (cyanotetrazolium chloride)-positive and therefore metabolically highly active bacteria were determined. In these seawater cultures amended with nutrients more than 95% of the CTC-positive cells exhibited a capsule. During two cruises, one to the North Atlantic and one to the North Sea, we investigated the distribution of capsulated bacteria throughout the water column. Capsulated bacteria were generally more abundant in eutrophic surface waters than in deeper layers or more oligotrophic regions. In the upper 100 m of the North Atlantic, about 6–14% of the total bacterioplankton community was capsulated, while in the layers below 100 m depth, 97% of the bacteria lacked a visible capsule. The percentage of capsulated bacteria correlated with bacterial abundance and production, and chlorophyll a concentration. Also, the bioavailability of DOC (dissolved organic carbon), estimated by the ratio between bacterial production and DOC concentration, significantly correlated with the percentage of capsulated bacteria. In the North Sea, the contribution of capsulated bacteria to the total number of bacteria decreased from the surface (3 m depth) to the near-bottom (25–35 m) layers from 20% to 14% capsulated bacteria. In the nearshore area of the North Sea, about 27% of the bacteria exhibited a capsule. Overall, a pronounced decrease in the contribution of capsulated bacteria to the total bacterial abundance was detectable from the eutrophic coastal environment to the open North Atlantic. Using this epifluorescence-based technique to enumerate capsulated bacterioplankton thus allowed us to routinely assess the number of capsulated bacteria even in the oceanic water column. Based on the data obtained in this study we conclude that almost all metabolically highly active bacteria exhibit a capsule, but also some of the metabolically less active cells express a polysaccharide capsule detectable with this method.  相似文献

Information on possible effects of pollution on fish life is reviewed with special emphasis to the North Sea. Concentrations of heavy metals are high in onshore waters, sediments and organisms. Offshore regions are considered to be not endangered. In estuaries concentrations of certain heavy metals are often surpassing thresholds, which have been experimentally demonstrated to produce sublethal effects on marine organisms. Also mussel culture areas had to be closed due to excess of cadmium contamination. Mercury in fillets of cod in the southern North Sea sometimes reaches 0.3 ppm representing 1/3 of the legal limit. Reductions of inputs are necessary due to the bioaccumulative and toxic potential of heavy metals. Organochlorine contamination of water, sediments and organisms is higher in the southern than in the northern North Sea. In offshore areas clearly elevated levels of PCB's, which are the dominating organochlorine compound in marine organisms, have been analyzed. At present only a minor fraction of total organochlorine residues accumulated in marine biota is analyzed. Margins between experimentally derived lethal levels and those occurring in marine biota are narrow suggesting that sublethal effects might occur. One of the special concerns in the context of organochlorine contamination has to be seen in the potential of these substances affecting the reproductive capacity of marine organisms. Reduction measures are urgently necessary and should include substances with bioaccumulative and toxicological potential, which are not presently included in Marine Monitoring Programmes. Impact of oil pollution is considered to be a regional problem, although especially in estuaries concentrations in the water column might be high. Input through oil platforms has led to local reductions in benthic communities. Measures have to be taken, to further reduce these inputs also in view to avoid contamination of fish in the vicinity of oil fields. Oxygen depletion in German and Danish coastal waters has been shown to be a severe degradation in coastal water quality resulting in significant impact on fish populations. Indications for large-scale avoidance reactions of low DO-areas and mortalities of fish and benthos organisms have been demonstrated. It is suspected that the low DO-situations are related to increased nutrients input and high phytoplankton production. Improvement of the situation can only be expected, if input of nutrients is reduced. Reasons for the occurrence of toxic plankton blooms are not understood. An intensified research is necessary, to avoid damage to fisheries and mariculture. Although activity of radionucleids in North Sea waters is increased due to input of radioactive material from La Hague and Sellafield reprocessing units, the present levels of activity are not considered to be a threat for marine life. The same holds for thermal pollution, which is, depending on the recipient area, expected to have only local effects. Sewage sludge dumping and discharge of untreated sewage introduce significant amounts of accompanying heavy metals, which can be expected to have a negative influence on the total metal budget of recipient waters. Nutrient input through sewage will exhibit only local effects. Dumping of wastes from titanium dioxide industry are suspected to be causally linked to the occurrence of increased prevalences of certain diseases of dab in the German Bight. Present management strategies should take account of the fact that through dumping of waste from titanium dioxide production significant quantities of heavy metal are introduced into the southern North Sea. Also the incineration of chemical wastes at sea should be reduced due to uncertainties concerning quantities and toxicity of incineration products and their potential impact on marine life. It is concluded that clear evidence exists for pollution impact in estuarine regions. Most of the major estuaries at the North Sea coast show benthos reductions, which can be related to water quality and it has to be assumed that areas of a sublethal effect are located in the vicinity of those areas of well demonstrated effects. They are of unknown size and attempts have to be undertaken to quantify areas of sublethal impact. The occurrence of fish diseases is interpreted as an expression of degraded water quality. Some of the diseases detected can produce mortality, thereby having an as yet unquantified impact on fish populations. The presence of fish diseases also reduces the marketability of catches. It cannot be excluded that changes in species composition of fish populations in German Wadden Sea areas are related to pollution. Concentrations of organochlorine substances in fishes of the southern North Sea give rise to the concern that their reproductive capability might be impaired. Future work has to be directed to this problem. Impact of pollution on populations of marine organisms has so far not been demonstrated.  相似文献

Since the first North Sea Conference (1984, Bremen), all countries bordering the North Sea made commitments to reduce discharge of hazardous substances into the North Sea. From Belgium and The Netherlands, large reductions (upto 90) in heavy metal emissions from land-based sources have been reported between 1985 and 2000. Recently, some studies in the Western Scheldt estuary (WS) have shown that total metal concentration in the water, sediments and suspended particles have decreased compared to levels in the 70s. However, data on aquatic organisms is still very limited and it is therefore difficult to confirm whether the reductions in pollution input and generally improving water quality in the WS have a corresponding impact on the levels of heavy metals in aquatic organisms. The current study measured metal concentrations in the soft tissues of mussels, Mytilus edulis (known to be good indicators of environmental metal pollution) during the period 1996–2002. Spatial (salinity and pollution gradients), temporal and seasonal variations were also studied. Results showed a down-stream decreasing trend for the metals studied (Cd, Co, Cr, Cu, Fe, Mn, Ni, Pb and Zn) during all sampling campaigns. There was also a significant seasonal effect on tissue metal concentrations, with a peak observed around spring in both WS and the nearby less polluted Eastern Scheldt (ES). On temporal trends, a clear drop of metals in mussels was observed in the early 80s coinciding with the start of the efforts to reduce chemical pollution input into the North Sea. Since those early reductions, metal concentrations in mussels generally remained unchanged upto mid 90s. However, in recent times metal concentration in mussels have increased significantly, for example Cd in 2002 was almost 10 times the values in 1983 and similar to levels seen during the peak in the 70s. Other metals also increased in the 90s also reaching levels seen in the 70s. As there is no indication of recent increase in metal input into the estuary, we suggest that increased metal concentrations observed in mussels in recent years especially in the upper estuary are most likely a result of changes in physical and chemical speciation and metal bioavailability. Such changes may be caused by changes in some water quality parameters in the estuary (i.e. increased dissolved oxygen, concentration of organic matter), resulting in conditions that favour releases of sediment-bound metals into the water column. The relationship between metal content and season showed very similar annual profiles in the polluted WS and less polluted ES. Thus, seasonal variations in metal concentrations appear to be largely controlled by biological processes, while total body burdens are dependent on environmental levels and bioavailability.  相似文献

Several blooms of Microcystis aeruginosa have been observed in the Patos Lagoon estuary during the last fifteen years without a proper investigation of their ecological importance or possible toxicity. The present study has identified and quantified the presence of cyanobacteria in the Patos Lagoon estuary, particularly of M. aeruginosa. During this survey, identification and quantification of the main phytoplankton groups were done in relation to geographical distribution in the estuary. The presence of M. aeruginosa colonies in the estuarine region confirmed their superficial distribution throughout the estuarine waters during twelve months with a maximum of 1, 3.10(6) cells. L-1 in December, 1994 and a minimum of 1, 5.10(5) cells. L-1 in August, 1995 and also confirmed that M. aeruginosa originated from waters in the north of the estuary. The period of the highest cell and colonies densities was coincident with high chlorophyll-a levels in surface waters. Toxicity of M. aeruginosa bloom material was determined by bioassay and concentrations of hepatotoxins microcystins were identified by HPLC-DAD. M. aeruginosa blooms were considered highly toxic, presenting a 24 h-LD50 lower than 100 mg.Kg-1 b.w. and a toxin content higher than 1 microgram.mg-1 d.w. Several microcystin variants were found in the extracts with microcystin-LR predominating.  相似文献

Atlantic cod bearing pseudobranchial tumours were investigated from both North Sea and Baltic Sea areas. Light and electron microscopic studies revealed a tumorous structure previously described in cod from Atlantic and Pacific waters. Additionally, cysts, multinucleated cells and early stages of mitosis are described. These findings support the hypothesis that pseudobranchial tumours are instigated by a protozoan infection causing xenotumours associated with the pseudobranch organ. The cells contained in these lesions showed similarities to amoebae.  相似文献

The bacterial community composition of marine surface sediments originating from various regions of the Eastern Mediterranean Sea (12 sampling sites) was compared by parallel use of three fingerprinting methods: analysis of 16S rRNA gene fragment heterogeneity by denaturing gradient electrophoresis (DGGE), terminal restriction fragment length polymorphism (T-RFLP), and analysis of phospholipid-linked fatty acid composition (PLFA). Sampling sites were located at variable depths (30–2860 m; water column depth above the sediments) and the sediments differed greatly also in their degree of petroleum contamination (0.4–18 μg g−1), organic carbon (0.38–1.5%), and chlorophyll a content (0.01–7.7 μg g−1). Despite a high degree of correlation between the three different community fingerprint methods, some major differences were observed. DGGE banding patterns showed a significant separation of sediment communities from the northern, more productive waters of the Thermaikos Gulf and the oligotrophic waters of the Cretan, S. Ionian, and Levantine Sea. T-RFLP analysis clearly separated the communities of deep sediments (>1494 m depth) from their shallow (<617 m) counterparts. PLFA analysis grouped a shallow station from the productive waters of the north with the deep oligotrophic sediments from the Ionian and Levantine Sea, with low concentrations of PLFAs, and hence low microbial biomass, as the common denominator. The degree of petroleum contamination was not significantly correlated to the apparent composition of the microbial communities for any of the three methods, whereas organic carbon content and sediment chlorophyll a were important in this regard.  相似文献

We compared the detection of bacteria and archaea in the coastal North Sea and at Monterey Bay, Calif., after fluorescence in situ hybridization (FISH) either with rRNA-targeted oligonucleotide probes monolabeled with the cyanin dye Cy3 (oligoFISH) or with fluorescein-labeled polyribonucleotide probes (polyFISH). During an annual cycle in German Bight surface waters, the percentages of bacteria visualized by polyFISH (annual mean, 77% of total counts) were significantly higher than those detected by oligoFISH (53%). The fraction of total bacteria visualized by oligoFISH declined during winter, whereas cell numbers determined by polyFISH remained constant throughout the year. Depth profiles from Monterey Bay showed large differences in the fraction of bacterial cells visualized by polyFISH and oligoFISH in the deeper water layers irrespective of the season. Image-analyzed microscopy indicated that the superior detection of cells by polyFISH with fluorescein-labeled probes in bacterioplankton samples was less a consequence of higher absolute fluorescence intensities but was rather related to quasi-linear bleaching dynamics and to a higher signal-to-background ratio. The relative abundances of archaea in North Sea and Monterey Bay spring samples as determined by oligoFISH were on average higher than those determined by polyFISH. However, simultaneous hybridizations with oligonucleotide probes for bacteria and archaea suggested that the oligoFISH probe ARCH915 unspecifically stained a population of bacteria. Using either FISH technique, blooms of archaea were observed in North Sea surface waters during the spring and summer months. Marine group II archaea (Euryarchaeota) reached >30% of total picoplankton abundances, as determined by polyFISH. We suggest that studies of pelagic microbial community structure using oligoFISH with monolabeled probes should focus on environments that yield detections > or =70% of total cell counts, e.g., coastal surface waters during spring and summer.  相似文献
