Chlorotic and green needles from Norway spruce (Picea abies L.) trees were sampled in the Calcareous Bavarian Alps in winter. The needles were used for analysis of the mineral and pigment contents, the levels of antioxidants (ascorbate, glutathione), and the activities of protective enzymes (superoxide dismutase, catalase, ascorbate peroxidase, monodehydroascorbate radical reductase, dehydroascorbate reductase, glutathione reductase). In addition, the activities of two respiratory enzymes (glucose-6-phosphate dehydrogenase, NAD-malate dehydrogenase), which might provide the NADPH necessary for functioning of the antioxidative system, were determined. We found that chlorotic needles were severely manganese deficient (3 to 6 micrograms Mn per gram dry weight as compared with up to 190 micrograms Mn per gram dry weight in green needles) but had a similar dry weight to fresh weight ratio, had a similar protein content, and showed no evidence for enhanced lipid peroxidation as compared with green needles. In chlorotic needles, the level of total ascorbate and the activities of superoxide dismutase, monodehydroascorbate radical reductase, NAD-malate dehydrogenase, and glucose-6-phosphate dehydrogenase were significantly increased, whereas the levels of ascorbate peroxidase, dehydroascorbate reductase, glutathione reductase, and glutathione were not affected. The ratio of ascorbate to dehydroascorbate was similar in both green and chlorotic needles. These results suggest that in spruce needles monodehydroascorbate radical reductase is the key enzyme involved in maintaining ascorbate in its reduced state. Acidic and basic peroxidases, termed as POD-A and POD-B, wereisolated from the apoplastic space of spruce (Picea abies, L.)needles and purified by acetone precipitation and anion exchangechromatography to apparent homogeneity. The molecular massesof POD-A and POD-B were 39.6 and 29.0 kDa, respectively. ThepH optimum of both isozymes ranged from 4.5 to 6. The apparentKm values of POD-A and POD-B were 460 and 210 µM for coniferylalcohol. Both isozymes acted also as NADH oxidases with apparentKm-values of 103 µM (POD-A) and 70 µM (POD-B). NAD+but not NADH was found in the apoplastic space of lignifyingneedles. Based on the lignification rate, the contents and kineticproperties of PODs, NADH oxidation by POD is not the major sourceof H2O2 required for lignin polymerisation. Cell wall-associated peroxidases (EC 1.11.1.7
[EC]
) were extractedfrom the current year's needles of Norway spruce trees (Piceaabies L.) in two fractions, namely soluble apoplastic peroxidasesand covalently wall-bound peroxidases. Peroxidase activitieswere determined with two substrates: coniferyl alcohol, whichis important for lignification, and NADH, which is necessaryfor the production of H2O2. Coniferyl alcohol peroxidase activitywas detected in both the soluble apoplastic fraction and thewall-bound fraction, whereas NADH oxidase activity was foundonly in the soluble apoplastic fraction. Net oxidation of coniferylalcohol and NADH was inhibited by ascorbate, which reduced theoxidized intermediates of the peroxidase- and oxidase-catalyzedreactions. Since ascorbate itself was oxidized in these reactions,the inhibition was not persistent and it was released once theascorbate present in the assay mixture had been oxidized. Ascorbatedelayed the oxidation of NADH 10-fold more efficiently thanthe oxidation of coniferyl alcohol. Although the level and theredox state of apoplastic ascorbate were lower in lignifyingneedles than in mature needles, the concentration, which was1.17 mM in apoplastic washing fluids, was sufficiently highto inhibit peroxidase activity in vitro. These results suggestthat peroxidases can catalyze lignification only if local differencesexist in the concentration of reduced ascorbate between lignifyingand non-lignifying tissues. The activity of superoxide dismutase was investigated in needles of spruce trees. To obtain maximum activity, needles were homogenized in the presence of Triton X-100 and polyvinylpyrrolidone. Superoxide dismutase activity was measured in dialyzed extracts with a modified epinephrine assay (HP Misra, I Fridovich [1972] J Biol Chem 247: 3170-3175) at pH 10.2. The extracts contained 70 to 120 units of superoxide dismutase per milligram protein. One unit of superoxide dismutase was completely inhibited in the presence of 20 micromolar NaCN. On native polyacrylamide gels three electromorphs were visualized after staining for activity. All three species were sensitive to CN− and H2O2 and were therefore assumed to be Cu/Zn-superoxide dismutases. Superoxide dismutase activity was dependent on the age of the needles and declined by approximately 25% within 3 to 4 years. 相似文献

Changes in superoxide dismutase (SOD; EC 1.15.1.1

) activityand isozymes were investigated in different developmental stagesof Norway spruce (Picea abies L.). Spruce seeds, seedlings grownin a climate chamber and foliar buds from field-grown treescontained two CuZn-SODs comigrating with SODs I and II previouslyidentified as the chloroplastic and cytosolic SODs in spruceneedles [Krniger et al. (1992) Plant Physiol. 100: 334]. Inaddition one Mn-SOD (SOD III) was identified by insensitivityto cyanide and H2O2. Highest total SOD activities were detectedin buds before bud break and in germinating seeds. In seedsand foliar buds SOD II was the major isozyme, whereas SOD Iwas dominant in mature needles. SOD III was present in all developmentalstages of the seedlings, but disappeared in field-grown treesduring bud break and reappeared at the end of summer in matureneedles. Purification of Two Superoxide Dismutase Isozymes and Their Subcellular Localization in Needles and Roots of Norway Spruce (Picea abies L.) Trees 下载免费PDF全文
Two isozymes of superoxide dismutase (SOD; EC 1.15.1.1) were purified from Norway spruce (Picea abies L.) needles to apparent electrophoretic homogeneity. Purification factors were 354 for SOD I and 265 for SOD II. The native molecular mass of both purified enzymes was approximately 33 kD, as determined by gel filtration. The subunit molecular weights, as estimated by sodium dodecyl sulfate polyacrylamide gel electrophoresis, were 20,000 for SOD I and 16,000 for SOD II in the presence of 2-mercaptoethanol, and 15,800 and 15,000, respectively, in its absence. These results indicate that the native enzymes were homodimers whose subunits contained intrachain disulfide bonds. Isoelectric points determined by nondenaturing isoelectric focusing were 4.5 and 5.5 for SOD I and II, respectively. NH2-terminal sequence analysis of the first 22 to 23 amino acids revealed 70 to 75% sequence identity with chloroplastic CuZn SODs from other plant species for SOD I, and 75% sequence identity with the cytosolic CuZn SOD from Scots pine for SOD II. Callus has been induced on needle and bud explants from 2, 6 and 26 year old Picea abies (L.) Karst, trees. Seventy seven percent of the needle explants from the youngest material was productive compared to 0.2% for the old tree. New embryogenic sections appeared in the non-embryogenic callus derived from 2, 6 and 26 year old trees after prolonged culture and was promoted by the use of an established embryogenic callus line in a nurse culture system.Abbreviations EDTA

Hydrogen peroxide (H2O2) scavenging systems of spruce (Picea abies) needles were investigated in both extracts obtained from the extracellular space and extracts of total needles. As assessed by the lack of activity of symplastic marker enzymes, the extracellular washing fluid was free from intracellular contaminations. In the extracellular washing fluid ascorbate, glutathione, cysteine, and high specific activities of guaiacol peroxidases were observed. Guaiacol peroxidases in the extracellular washing fluid and needle homogenates had the same catalytic properties, i.e. temperature optimum at 50°C, pH optimum in the range of pH 5 to 6 and low affinity for guaiacol (apparent Km = 40 millimolar) and H2O2 (apparent Km = 1-3 millimolar). Needle homogenates contained ascorbate peroxidase, dehydroascorbate reductase, monodehydroascorbate reductase, glutathione reductase, and catalase, but not glutathione peroxidase activity. None of these activities was detected in the extracellular washing fluid. Ascorbate and glutathione related enzymes were freeze sensitive; ascorbate peroxidase was labile in the absence of ascorbate. The significance of extracellular antioxidants for the detoxification of injurious oxygen species is discussed. 相似文献

Individual needles of 4-year-old spruce trees (Picea abies L.)were exposed to 35S-GSH via the cut surface of the needle tipfor 23 h at different times during the growing season.From the previous year's source needle, 35S-GSH was exportedinto the twig and translocated predominantly towards the twigapex. Independent from the growing season, transport towardsthe stem did not exceed 20% of the radioactivity recovered fromthe tissue samples. In July, the new needles appeared to bemetabolic sinks for reduced sulphur as indicated by the higherpercentages of both soluble and insoluble 35S in this twig section,as compared to the previous years' needles. By use of methanol extraction, two different consecutive partition procedures, repeated polyvinylpyrrolidone column chromatography, silicic acid partition column chromatography and the dwarf rice, lettuce, and barley half-seed bioassays, several gibberellin-like substances were detected in elongating shoots of Picea abies (L.) Karst. No significant differences in the content of gibberellin-like substances could be detected between juvenile and mature trees. The ability of plants to track seasonal changes is largely dependent on genes assigned to the photoperiod pathway, and variation in those genes is thereby important for adaptation to local day length conditions. Extensive physiological data in several temperate conifer species suggest that populations are adapted to local light conditions, but data on the genes underlying this adaptation are more limited. Here we present nucleotide diversity data from 19 genes putatively involved in photoperiodic response in Norway spruce (Picea abies). Based on similarity to model plants the genes were grouped into three categories according to their presumed position in the photoperiod pathway: photoreceptors, circadian clock genes, and downstream targets. An HKA (Hudson, Kreitman and Aquade) test showed a significant excess of diversity at photoreceptor genes, but no departure from neutrality at circadian genes and downstream targets. Departures from neutrality were also tested with Tajima''s D and Fay and Wu''s H statistics under three demographic scenarios: the standard neutral model, a population expansion model, and a more complex population split model. Only one gene, the circadian clock gene PaPRR3 with a highly positive Tajima''s D value, deviates significantly from all tested demographic scenarios. As the PaPRR3 gene harbours multiple non-synonymous variants it appears as an excellent candidate gene for control of photoperiod response in Norway spruce. 相似文献

Gibberellins A1 (GA1), A3 and A9 were identified from extracts of shoots of 6-month old Norway spruce (Picea abies) seedlings by the use of sequential reverse and normal phase high performance liquid chromatography (HPLC), bioassay, radioimmunoassay (RIA) and combined gas chromatography-mass spectrometry (GC-MS). Summary Embryogenic callus developed in 55% of the mature embryo explants of Norway spruce (Picea abies L.) growing on a LP medium minus the amino acids and sugars (except sucrose). This is the highest reported yield of embryogenic callus from mature embryos of P. abies that has ever been reported. Callus induction from either the middle or the end of the hypocotyl of the embryos began after 2–3 weeks. Three types of calli were recovered: (a) globular, (b) light green-compact, (c) white mucilaginous. Only the white mucilaginous calli were embryogenic. The globular and light green-compact calli never become embryogenic, even after several subcultures. The development of somatic embryos was accomplished on half-strength macro-elements of NSIII medium containing 1 M -naphthaleneacetic acid, 1 M abscisic acid, and 3% sucrose. The addition of 10–7
M buthionine sulfoximine to the medium increased the development of somatic embryos by three fold. These results suggest that there is a great potential for increasing the frequency and development of somatic embryos in P. abies. Seasonal changes of ascorbate peroxidase and monodehydroascorbateradical reductase activities were studied in foliar tissuesof Norway spruce (Picea abies L.). In mature needles, APX activitiesdid not show seasonal fluctuations and were similar to thosefound in resting buds. Monodehydroascorbate radical reductaseactivity was higher in needles than in buds and higher in winterthan in summer. Maximum activities of both enzymes were foundbefore bud break and minimum activities in newly formed needles.When spruce seedlings were exposed to an artifical frost eventof 5°C for one night in spring, ascorbate peroxidaseactivity declined in young needles before the onset of visibleinjury but corresponding to a sudden upsurge in lipid peroxidation.After one week, some shoots showed severe symptoms of injury,some were slightly injured and others did not show any visibleinjury. In lethally injured needles, antioxidative protection(ascorbate peroxidase, monodehydroascorbate radical reductase,glutathione reductase, glutathione, ascorbate, superoxide dismutase)had collapsed. Surviving needles showed a coordinated increasein all components of the antioxidative system suggesting anefficient induction of defense systems. However, enhanced protectionwas observed only transiently. In fall, needles that had beenexposed to frost in spring contained significantly less antioxidantsthan unstressed needles indicating that unseasonal frost causedmemory effects. The long-term impact of elevated concentration of CO2 on assimilation activity of sun-exposed (E) versus shaded (S) foliage was investigated in a Norway spruce stand [Picea abies (L.) Karst, age 14 years] after three years of cultivation in two domes with adjustable windows (DAW). One DAW was supplied with ambient air [AC, ca. 350 µmol(CO2) mol–1) and the second with elevated CO2 concentration [EC = AC plus 350 µmol(CO2) mol–1]. The pronounced vertical profile of the photosynthetic photon flux density (PPFD) led to the typical differentiation of the photosynthetic apparatus between the shaded and sun needles. Namely, photon-saturated values of maximal net photosynthetic rate (P
Nmax) and apparent quantum yield () were significantly higher/lower for E-needles as compared with the S-ones. The prolonged exposure to EC was responsible for the apparent assimilatory activity stimulation observed mainly in deeply shaded needles. The degree of this stimulation decreases in the order: S-needles dense part > S-needles sparse part > E-needles dense part > E-needles sparse part. In exposed needles some signals on a manifestation of the acclimation depression of the photosynthetic activity were found. The long-term effect of EC was responsible for the decrease of nitrogen content of needles and for its smoother gradient between E- and S-needles. Young seedlings of Picea abies Karst, grown in nutrient solution were treated with the growth retardants Amo-1618, B-995, and CCC. These were added to the nutrient medium. B-995 and CCC retarded root and shoot growth in the concentrations 100, 10, and 1 mg/l. Growth was almost entirely inhibited by 300 mg/l, obviously due to toxicity. The effects of Amo-1618 were similar but more varying. GA counteracted the effects of all the retardants on shoot growth, but not on root growth. 相似文献

Summary Dark respiration and photosynthetic carbon dioxide refixation in purple and green Picea abies cones were investigated from budbreak to cone maturity. The rate of dark respiration per unit dry weight and CO2 refixation capacity decreased during cone maturation. At the beginning of the growing season, photosynthetic CO2 refixation could reduce the amount of CO2 released by respiration in green and purple cones by 50% and 40%, respectively. The seasonal performance of the components of the cone carbon balance was calculated using information on the seasonal course of respiration, refixation capacity and the light response curves of cone photosynthesis, as well as the actual light and temperature regime in the field. The daily gain of CO2 refixation reached 28%–34% of respiration in green and 22%–26% in purple cones during the first month of their growth, but decreased later in the season. Over the entire growth period refixation reduced carbon costs of cone production in both cone colour polymorphs by 16%–17%. 相似文献

A manganese-containing superoxide dismutase (SOD; EC 1.15.1.1

)was purified to electrophoretic homogeneity from seeds of Norwayspruce (Picea abies L.). The apparent molecular mass of thepurified enzyme was 86 kDa, as determined by gel filtration.The subunit molecular mass, estimated by SDS-polyacrylamidegel electrophoresis, was 22 kDa both in the presence and inthe absence of 2-mercaptoethanol. Thus, the native enzyme isa homotetramer with subunits that were not linked by disulfidebonds. The isoelectric point of this Mn-SOD was 5.5. The specificactivity of the Mn-SOD was strongly pH-dependent and was 400units per nmol SOD at pH 7.8 and 30 units per nmol SOD at pH10.4. The first 25 amino acid residues in the amino terminalregion of spruce Mn-SOD exhibited a high degree of sequencehomology to those of Mn-SODs from other organisms.
