Experiments were performed to optimize the macronutrients concentrations for in vitro rooting of Ceratonia siliqua micropropagated shoots. Several dilutions of Murashige and Skoog (MS) medium were tested: full-strength MS, half-strength MS ( MS), and MS + full N. The frequency of in vitro rooting was enhanced when the MS was used (50 % rooted shoots). Mature leaves from 20 – 30 year-old carob trees and from 2 year-old micropropagated plants were collected and the concentrations of macronutrients (N, P, K, Ca, Mg) assessed. Based on the mineral composition of the leaves a new medium was formulated and compared with the previous ones showing an increment of the rooting frequency to 80 %. Moreover, shoots rooted in the new medium did not show symptoms of apical necrosis that occurred in the other tested media.  相似文献

Summary The use of several S1 individuals to represent an S0 individual permits the use of a Design II mating scheme for plants with only one pistillate flower per plant. Estimates of additive (V A ) and dominance (V D ) variance from this mating scheme will be biased upwards, when a small number (10) of individuals of each S1 line are used. This bias can be computed, and the additive and dominance estimates can be corrected. Of particular interest is the observation that the additive genetic variance contributes to bias in estimates of V D . When S0 plants are non inbred and their selfedprogeny (S1 lines) are used to represent them in developing families for use in the Design II, where m1 is the number of individuals used to represent an S1 line in developing half sib-families and m2 is the number of individuals used to represent the S1 line in making up full sib-families. For example, in a 3×3 Design II, with about 10 individuals used to represent each S1 line in each cross, m2 = 10 and m1 = 30. The electrochemical potential differences for potassium, between excised barley (Hordeum vulgare L.) roots and external media containing 0.05 mM KCl+0.5 mM CaSO4, were determined over a 4-h period during which initially low-K+ roots accumulated K+ by pretreatment in 50 mM KCl plus 0.5 mM CaCl2. This pretreatment resulted in increased internal [K+], decreased K+ influx (as measured from 0.05 mM KCl+0.5 mM CaSO4) and decreased values of . These observations indicate that the decline of K+ influx associated with increased internal K+ concentration cannot be accounted for by passive adjustment to the electrochemical gradient for this ion.  相似文献

The effects of these salts on the helix–coil transition of rA–rU and rI–rC helices were examined. Increasing the size of the hydrophobic substituents, R1, R2, and R3 lowers the degree of stabilization of the helical structure. The disproportionation reaction, 2rA–rU→rA–rU2 + rA occurs readily with salts II and III, especially when the substituents, R1, R2, and R3 are small, i.e., H or Me. Spermine is found to stabilize the rA–rU2 and rI–rC helices to approximately the same extent; however, large differences between the degree of stabilization of rA–rU2 and rI-rC helices are observed when the substituents R1, R2, and R3 are large hydrophobic groups. Similar results are also obtained for the spermidine series. Finally, differences in the interactions of the salts II and III with rA–rU2 and rI–rC helices suggest that the latter helix is denser.  相似文献

Summary The effects of ambient temperature (T a) on ventilation and gas exchange in chukar partridges (Alectoris chukar) were determined after acclimation to low and high altitute (LA and HA; 340 and 3,800 m, respectively).At both LA and HA, oxygen consumption ( ) increased with decreasingT a atT a from 20 to –20°C. AtT a of 35 to 40°C, increased above thermoneutral values at HA but remained constant and minimal at LA. Water loss rates increased rapidly atT a>30°C at both altitudes as birds began to pant. Ventilation rates (f) during panting were 5-to 23-fold greater than the minimalf at thermoneutralT a.Increased atT a below thermoneutrality was supported by increased minute volume (V i) at both altitudes. The change inV i was primarily a function of changing tidal volume (V t), althoughf increased slightly asT a declined. Summary Rosy finches (Leucosticte arctoa) breed at altitudes above 3500 m in eastern California. House finches (Carpodacus mexicanus) belong to the same subfamily (Carduelinae), but breed at much lower elevations. Oxygen consumption ( ) and ventilatory parameters of these two species were measured over a wide range of ambient temperatures (T a) at low altitude (LA; 150 m) and at high altitude (HA; 3800 m).Minimal nighttime 's of rosy finches and house finches at LA (T a=30°C) were close to allometrically predicted values for passerine birds. Summary The energy requirements of Adélie penguin (Pygoscelis adeliae) chicks were analysed with respect to body mass (W, 0.145–3.35 kg, n=36) and various forms of activity (lying, standing, minor activity, locomotion, walking on a treadmill). Direct respirometry was used to measure O2 consumption ( ) and CO2 production. Heart rate (HR, bpm) was recorded from the ECG obtained by both externally attached electrodes and implantable HR-transmitters. The parameters measured were not affected by hand-rearing of the chicks or by implanting transmitters. HR measured in the laboratory and in the field were comparable. Oxygen uptake ranged from in lying chicks to at maximal activity, RQ=0.76. Metabolic rate in small wild chicks (0.14–0.38 kg) was not affected by time of day, nor was their feeding frequency in the colony (Dec 20–21). Regressions of HR on were highly significant (p< 0.0001) in transmitter implanted chicks (n=4), and two relationships are proposed for the pooled data, one for minor activities ( ), and one for walking ( ). Zusammenfassung Bei Röntgenbestrahlung von L-Leucin·HCl bei Zimmertemperatur wird das >C-H-Proton unter Bildung des Radikals (CH3)2 C-CH2-CHNH3 +-COOH abgetrennt. Das von acht -Protonen stammende ESR-Spektrum wurde wegen nur schwacher Anisotropie der -Protonen-Hfs auch im polykristallinen Zustand vollständig aufgelöst, und es wurden folgende isotrope Aufspaltungen ermittelt: bei Messung bei Zimmertemperatur = 23.5±1 Gauß für sechs äquivalente CH3-Protonen und =8,0±1 Gauß und = 38,8±1 Gauß für die zwei Methylenprotonen, bei Messung bei 77 °K entsprechend = 22,6±1 Gauß =8,8±1 Gauß und =45,2±1 Gauß (Temperaturabhängigkeit der CH2-Protonenkopplung). Das bei Zimmertemperatur beobachtete Radikal wird bereits durch Bestrahlung bei 77 °K gebildet.
ESR-studies on the structure of radicals in X-irradiated L-leucine hydrochloride
Summary The structure of radicals in X-irradiated polycrystalline L-leucine·HCl has been studied by ESR. At room temperature the radical (CH3)2 > C-CH2-CHNH 3 + -COOH is formed by abstraction of the >C-H-proton. The isotropic part of hyperfine interaction with eight -protons has been measured even in polycrystalline state with following values: at 300 °K =23,5±1 gauss for six equivalent CH3-protons and =8.0±1 gauss and =38.8±1 gauss for two CH2-protons, at 77 °K =22.6±1 gauss, =8.8±1 gauss and =45-2±1 gauss respectively (the coupling of the CH2-protons is dependent from temperature). This radical is present already following irradiation at 77 °K.

Summary The cellular mechanism of K-stimulated Cl transport in locust hindgut was studied using double-barrelled ionsensitive microelectrodes and electrophysiological techniques. Steady-state net electrochemical potentials for Cl and K and the conductances of apical and basal membranes and paracellular pathway were determined under control conditions, during exposure to 1mm cAMP, and following ion substitutions. Under control open-circuit conditions, intracellular Cl activity (a Cl c ) was 3.5 times that predicted for passive equilibrium across the apical membrane. The net electrochemical potential opposing Cl entry from the mucosal side increased by 50% during cAMP stimulation of transepithelial Cl absorption whereas the net electrochemical potential favoring Cl exit across the basal membrane was unchanged. No correlation was observed between and the net electrochemical potential across the apical membrane for Na. The net electrochemical potential favoring K entry across the apical membrane was negligible underI sc conditions when Cl transport rate was approximately 10 eq cm–2 hr–1. Locust rectal cells showed electrical and dye coupling. The results also indicate that most transepithelial diffusion of ions is transcellular and that epithelial tightness effectively increases during exposure to cAMP becauseR a andR b both decrease, by 80% whileR j is unchanged. The cAMP-induced R b was abolished in Cl-free saline whereas R a was insensitive to Cl removal, but was blocked by removing K from the saline. Based on these findings, our model for Cl absorption in locust hindgut features i) an active entry step for Cl at the apical membrane which is stimulated by cAMP and by low levels of K on the mucosal side, but is not energized by or a large cAMP-stimulated Cl conductance in the basal membrane and a similar cAMP-stimulated K conductance in the apical membrane. cAMP dose-response curves are similar for the stimulation of active Cl absorption and Cl-independent (i. e. K) conductance, indicating that cAMP exerts dual control over active Cl transport and counter-ion permeability.  相似文献

Methanogenic archaea are strictly anaerobic organisms that derive their metabolic energy from the conversion of a restricted number of substrates to methane. H2+CO2 and formate are converted to CH4 via the CO2-reducing pathway, while methanol and methylamines are metabolized by the methylotrophic pathway. A limited number of methanogenic organisms utilize acetate by the aceticlastic pathway. Redox reactions involved in these processes are partly catalyzed by membrane-bound enzyme systems that generate or, in the case of endergonic reactions, use electrochemical ion gradients. The H2:heterodisulfide oxidoreductase, the F420H2:heterodisulfide oxidoreductase and the CO:heterodisulfide oxidoreductase, are novel systems that generate a proton motive force by redox-potential-driven H+ translocation. The methyltetrahydromethanopterin:coenzyme M methyltransferase is a unique, reversible sodium ion pump that couples methyl transfer with the transport of Na+ across the cytoplasmic membrane. Formylmethanofuran dehydrogenase is a reversible ion pump that catalyzes formylation and deformylation, of methanofuran. In summary, the pathways are coupled to the generation of an electrochemical sodium ion gradient and an electrochemical proton gradient. Both ion gradients are used directly for ATP synthesis via membrane integral ATP synthases. Summary Oxygen consumption rates ( ) and a number of possible anaerobic end products were determined forLimaria fragilis at rest, and following active swimming. increased up to 8-fold (=4) during swimming. Swimming did not change the concentrations ofl-lactate, alanine or arginine phosphate in the single striated fast adductor muscle. Summary Metal coordination confers an extraordinary structural stability to the ferrichromes which, independent of their variable amino acid composition, results in a basically unperturbed conformation for all the homologous peptides in the series. The proton magnetic resonance (pmr) characteristics for Al3+ analogues (alumichromes) reflect this conformational isomorphism in usual solvents so that single site substitutions are clearly recognized in the pmr spectra. Thus, the substitution of glycine byl-alanine orl-serine introduce new resonances characteristic of the sidechains and alter the pattern of the amide NH pmr region in that doublets substitute for glycyl triplets at the same site. Since for glycine- andl-serine-containing alumichromes the resonances have already been identified, it is possible to unequivocally establish the primary structure of the twol-alanyl homologues ferrichrome C ( ) and sake colorant A ( ) on the basis of the comparative pmr spectra of their Al3+ analogues, namely, alumichrome C and alumisake. The resonance assignment, and hence the site occupancy, is substantiated by the temperature coefficients of the NH chemical shifts, rates of1H-2H exchange and homonuclear proton spin decoupling experiments centered on the NH spectral region. Occupancy of site 1 by a glycine residue is observed for all known ferrichromes, which serves to conserve a hairpin turn. This method of obtaining sequence information should prove of general use for other systems of homologous polypeptides, provided their conformations are not affected by the residue substitutions.  相似文献

Summary Intracellular C1, K and Na activities (a Cl i ,a k i anda Na i ) and transmucosal membrane potential (E m) in epithelial cells ofNecturus gallbladder were measured at different external Na concentrations ([Na]o), with liquid ion-exchanger and conventional microelectrodes. Bladders were mounted in a divided chamber at 23°C between identical HCO3-free Ringer solutions containing 5mm K. The pH was 7.2. Tris was substituted for Na. Measurements were made under steady-state conditions as determined by the constancy of the transepithelial potential difference. Both,a Cl i anda Na i increased in a saturable fashion with [Na]o.E m did not change significantly. Average values (±sem) under normal conditions ([Na]o=100mm) fora Cl i ,a Na i andE m were 16.8±0.8mm (n=9), 9.7±0.6mm (n=10) and –52.6±0.6 mV (n=26), respectively. In Na-free mediaa Cl i declined to its equilibrium value.a K i (96±2mm;n=7) did not change when [Na]o was varied between 100 and 10mm but decreased to 80±3mm (n=4) in Na-free media.Transmembrane electrochemical potential differences, , for Cl and Na were calculated at four different [Na]o levels. A highly significant linear relation between and was found, indicating that Cl and Na transport are energetically linked. The results support the view that the energy necessary for intracellular Cl accumulation is derived from the simultaneous dissipation of the chemical potential gradient of Na across the apical membrane and that the coupled entry mechanism is electroneutral.  相似文献

Methanobacterium thermoautotrophicum was grown in continuous culture in a fermenter gassed with H2 and CO2 as sole carbon and energy sources, and in a medium which contained either NH4Cl or gaseous N2 as nitrogen source. Growth was possible with N2. Steady states were obtained at various gas flow rates with NH4Cl and with and the maintenance coefficient varied with the gas input and with the nitrogen source. A year mark and recapture study of aquatic Dytiscidae (Coleoptera) in a group of seven semi-permanent and temporary ponds in Cheshire (UK) is described. Pond water levels showed strong seasonal fluctuations with most ponds drying and refilling on several occasions over the study period. Sampling and labelling of aquatic Dytiscidae was carried out at monthly intervals. Beetles were labelled by fixing numbered waterproof paper labels to the elytra with cyanoacrylate `Superglue'. A total of 780 beetles were marked and released of which 11.5% were subsequently recaptured over varying time intervals of one month to eighteen months. The six species marked (Agabus bipustulatus (Linnaeus, 1767), Acilius sulcatus (Linnaeus, 1758), Colymbetes fuscus (Linnaeus, 1758), Dytiscus marginalis (Linnaeus, 1758), Ilybius ater (De Geer, 1774) and Ilybius fuliginosus (Fabricius, 1792)) are all known to be highly flight mobile. Colymbetes fuscus did not move between ponds as a result of ponds drying up in the summer. The behaviour of Dytiscus marginalis and Acilius sulcatus was found to be in broad agreement with previous accounts of dispersal from over-wintering ponds into temporary ponds in the spring with movement back to permanent ponds in the summer as the temporary ponds dry up. In contrast, Agabus bipustulatus did not migrate when ponds dried up in the summer but remained in terrestrial vegetation in damp pond basins for several months after open water had disappeared, only migrating if the basin dried out completely. Both species of Ilybius were captured and marked in very low numbers and no recaptures were made. As more ponds became temporary over the years of the study Agabus bipustulatus persisted while the other five species declined or were lost altogether.  相似文献

Summary Certain sections ofJosiah Willard Gibbs's thermodynamics papers might be applicable to biological equilibrium and growth, normal or abnormal.Gibbs added terms i dm i to the differential of the internal energy d=td–pd, (t=temperature,p=pressure,=entropy,=volume) where is the potential of substancem i , to provide for chemical as well as thermal and mechanical equilibrium. In this article a further generalization is suggested, to include biological equilibrium by adding to de terms of the form GdN, the variableN being the number of cells, where is a growth potential that measures exactly the resistance toward spontaneous growth. The functionG, like i is intensive in nature (i.e. depends on intensive variables only) except for a conversion factor ,M=m i , affording possible insight into why incipient abnormal growth is often independent of the number of cells. Useful applications might follow from identities between , or and or respectively. The following new function is studied, , a natural generalization of theGibbs free energy function , the possibility of measuring it electrically, and comparison of its role with that of for the possible experimental determination ofG. Gibbs's necessary and sufficient conditions for heterogeneous equilibrium ofn components inm phases are generalized and also modified to include broader restraining conditions like ,j=1,f,n, the > being characteristic of only living cellular phases. Careful appraisal of the term biological stability is followed by new criteria for stability, instability, and limits of stability, (neutral equilibrium) in terms of derivatives ofG, with possible medical applications. Three different sections of Gibbs's works tend to indicate that, for a biological phase, lower pressure usually increases its stability. The equation , where =surface tension,p, p = pressures,r, r=radii of curvature, is applied to possible control of tissue growth at interfaces. Methods of altering the equilibrum between three phasesA, B, C by varying the interfacial tensions AB , BC , AC , using relations like AB < AC + BC for stability of theA, B interface, suggest different means for shifting biological equilibrium between normal and abnormal cells through the introduction of new third phases at the interface. Summary In pot experiments with barley, mustard, leek, lettuce and spinach, and in a field experiment with 30 cultivars of barley uptakes of K, Mg, Ca, Na and N were studied at varying concentrations and activities of these cations in the soil solution.The sum of macro cations (K, Mg, Ca, Na) in meq per 100 g aerial plant parts were independent of the chemical composition of the soil solution, but dependent on plant species and on the N concentration in the plant.The ratios of mean net inflows of Mg, Ca and K into plants and corresponding cation activity ratios (aMg/aCa and ) in the soil solution were linearly related and highly correlated under conditions in which growth rate and/or rate of incorporation into new tissues constituted the rate determining step of cation uptake. Consequently, mean net inflows of K, Mg and Ca were independent of ion concentration and ion activity of K, Mg or Ca in the soil solution under the conditions of constant activity ratio.The results agree with the concept that plants have a finite cation uptake capacity, and that plants are in a equilibrium-like state with the activities of K, Mg, and Ca ions in the soil solution. The results indicate that both ratios and content of exchangeable cations should be considered in our evaluation of soil test data.  相似文献

The present investigation examined the relationship between CO2 sensitivity [at rest (S R) and during exercise (S E)] and the ventilatory response to exercise in ten elderly (61–79 years) and ten younger (17–26 years) subjects. The gradient of the relationship between minute ventilation and CO2 production ( E/ CO2) of the elderly subjects was greater than that of the younger subjects [mean (SEM); 32.8 (1.6) vs 27.3 (0.4); P<0.01]. At rest, S R was lower for the elderly than for the younger group [10.77 (1.72) vs 16.95 (2.13) 1 · min–1 · kPa–1; 1.44 (0.23) vs 2.26 (0.28) 1 · min–1 · mmHg–1; P<0.05], but S E was not significantly different between the two groups [17.85 (2.49) vs 19.17 (1.62) l · min–1 · kPa–1; 2.38 (0.33) vs 2.56 (0.21) 1 · min–1 · mmHg–1]. There were significant correlations between both S R and S E, and E/ CO2 (P<0.05; P<0.001) for the younger group, bot none for the elderly. The absence of a correlation for the elderly supports the suggestion that E/ CO2 is not an appropriate index of the ventilatory response to exercise for elderly humans.  相似文献

The effect of exercise training on heart rate variability (HRV) and improvements in peak oxygen consumption ( peak) was examined in sedentary middle-aged men. The HRV and absolute and relative peak of training (n = 19) and control (n = 15) subjects were assessed before and after a 24-session moderate intensity exercise training programme. Results indicated that with exercise training there was a significantly increased absolute and relative peak (P < 0.005) for the training group (12% and 11% respectively) with no increase for the control group. The training group also displayed a significant reduction in resting heart rate; however, HRV remained unchanged. The trained subjects were further categorized into high (n = 5) and low (n = 5) HRV groups and changes in peak were compared. Improvements in both absolute and relative peak were significantly greater (P > 0.005) in the high HRV group (17% and 20% respectively) compared to the low HRV group (6% and 1% respectively). The groups did not differ in mean age, pretraining oxygen consumption, or resting heart rate. These results would seem to suggest that a short aerobic training programme does not alter HRV in middle-aged men. Individual differences in HRV, however, may be associated with peak response to aerobic training.  相似文献

Summary The paper deals with the distribution of certain classes of algae in two fishponds of Hyderabad — India. Amongst all the ecological factors discussed in this regard, it is emphasized that Pearsall's basic ratio of water (Na + K/Ca + Mg) plays a distinctly important role. It is concluded that (a) if the basic ratio fluctuates between .027 and 1.2, Diatoms dominate the algal population, (b) if it works out between 1.2 and 2.0, green algae and Diatoms contribute more or less equal percentages to the bulk of phytoplankton and (c) if it is higher than 2.0, there is very little chance of Diatoms becoming dominant as the green algae, particularly Chlorococcales and Desmids, are in abundance. It is also noted that very high values of the basic ratio (3.7–5.5) do not help in increasing the percentage of green algae any further in the population.  相似文献
