As wild organisms adapt to the laboratory environment, they become less relevant as biological models. It has been suggested that a commonly used S. cerevisiae strain has rapidly accumulated mutations in the lab. Oxygen uptake rates in cultured rat hepatocytes 总被引:5,自引:0,他引:5
One potential treatment of acute liver failure involves the use of an extracorporeal device composed of functional hepatocytes. A major issue in the design of such a large-scale device is providing the hepatocytes with a sufficient supply of oxygen and other nutrients. In this study, we have designed and characterized a simple perfusion system hepatocytes using this system. The OUR of hepatocytes was determined during the first day after seeding on a single collagen gel and during the long-term stable culture after the addition of a top layer of collagen. The OUR increased to 20.7 +/- 0.57 pmol/sec/mug DNA during the first 13 hours of culture on a single collagen gel, while during the next 11 hours, the OUR declined to 10.6 +/- 1.5 pmol/sec/mug DNA. In parallel with the increase in OUR during the first 10 hours, we observed significant cell spreading, suggesting that the oxygen supply to the cells may be critical for the spreading and adaptation of the anchorage-dependent hepatocytes following isolation. Addition of a top layer of collagen to hepatocyte cultures for 24 hours of culture on a single collagen layer resulted in a stable OUR for 15 days. The volatile organic compound composition (VOCs) of printing press air wasfound to contain mostly ethanol, but also ethyl acetate, 1-propanol, 2-propanol,1-methoxy-2-propanol and 3-ethoxy-1-propanol. A pilot plant bioscrubber inoculatedwith a mixed microbial population was constructed on-site. The bioscrubber wasable to treat the polluted gas efficiently. It, however, suffered from strong wall growthand blockages in the column. The efficiencies of the pilot plant and a bioreactor iscompared. The yeasts Candida guilliermondii and Saccharomyces cerevisiae known to tolerate ethanol were selected instead of mixed population to avoid the wall growth and blockages in the bioreactor. The removal of the VOCs both individually and as a complex mixture was tested in a microcultivation system and in continuous chemostat cultures with and without cell recycling. The Candida yeast could use all the compounds as a carbon source while growth of S. cerevisiae was markedly slower on the methoxylated and ethoxylated propanols. Best total removal of the VOCs was 99% and achieved by C. guilliermondii. The only compound that was not totally removable in the chemostat experiment with C. guilliermondii was 1-methoxy-2-propanol. In laboratory scale the total and volumetric removal of VOCs by C. guilliermondii was more efficient compared to the pilot plant encouraging to scale up and applying the yeast bioreactor to real field conditions. 相似文献

For recombinant protein production in E. coli fed-batch cultures, post-induction conditions have great influence in the quantity and quality of the product. The present paper covers the effect of different factors affecting the cellular environment in recombinant aldolase (rhamnulose-1-phosphate aldolase, RhuA) production. An operational mode employing an exponential addition profile for constant specific growth rate has been analyzed, in order to understand and define possible modifications with influence on post-induction cellular behavior. A constant addition profile has been demonstrated to render higher specific aldolase production than the exponential addition profile, probably due to a more constant environment for the cells. On the other hand, amino acid (leucine) supplementation has proven to increase protein quality in terms of activity units (U) per unit mass of RhuA (U mg?1 RhuA), alleviating metabolic overload. Based on the above, a production process was set up and scaled up to pilot plant. Resulting production was double that of a standard laboratory operation, 45,000 U L?1, and almost all the protein retained the 6xHis-tag with the highest quality, 11.3 U mg?1 RhuA. 相似文献

The biology, and hence the mass culture, of Dunaliella viridis closely follows that of Dunaliella salina, which is successfully mass cultured for the production of -carotene. Both algae can grow at extremely high salinities and light intensities. They co-exist in the coastal salt lake, Hutt Lagoon, Western Australia. In contrast to D. salina, D. viridis does not accumulate large amounts of -carotene, producing only up to 0.7% of mixed carotenoids (lutein, zeaxathin, other oxygenated carotenoids and -carotene), compared to D. salina's ca 10% dry wt of mainly -carotene. However, in laboratory experiments, D. viridisgrew much faster and to much higher cell densities than D. salina, and attained levels of mixed carotenoids similar to those of D. salina (ca 13 mg L–1 carotenoid). Preliminary experiments in outdoor ponds were much less promising. Harvesting by chemical flocculation was as effective as with D. salina, but extraction of carotenoids directly into vegetable oil proved inefficient. When incorporated into feed, caretonoids derived from D. viridis pigmented hen eggs acceptably. Extrapolating from laboratory results, and using costing derived from D. salina technology, the cost of production of mixed oxygenated carotenoids from D. viridis was similar to that for the production of -carotene from D. salina, at ca $A500 kg–1. 相似文献

1999年 4~ 6月 ,采用室内实验生态学方法对栉孔扇贝的耗氧率和排氨率进行了研究 .结果表明 ,在适宜的温度范围内 ,栉孔扇贝的耗氧率和排氨率均与温度成正比 ,而与体重呈负相关关系 .在实验室温度 (8~ 2 8℃ )条件下 ,栉孔扇贝的耗氧率为 0 .48~ 9.0 9mg·g-1·h-1,排氨率为 0 .0 5~ 1 0 1mg·g-1·h-1.其中耗氧率在 2 3℃时达到最高值 ,2 8℃时开始下降 ,而排氨率则呈持续升高趋势 .栉孔扇贝的日常代谢明显高于标准代谢 ,耗氧率和排氨率平均值分别提高约 35 .8%和 75 .9% . 相似文献

采用室内实验生态学方法研究了不同栖息水温和不同溶解氧水平下处于标准代谢状态的西施舌耗氧率与排氨率,并测定了窒息点.结果表明,在25 ℃时,水中DO≥3.11±0.15 mg·L-1时,西施舌的耗氧率和排氨率分别为0.7±0.05 mg·g-1·h-1和2.56±0.05 μmol·g-1·h-1,处于相对稳定状态;当DO低于此值则代谢出现异常,耗氧率随DO下降而下降,直到窒息为止,其窒息点为1.22±0.06 mg·L-1,而排氨率也呈直线下降,但排氨停止滞后于耗氧停止.耗氧率与栖息水温呈二次线型关系:OCR=-0.0027T2+0.1367T-0.9557,R2=0.972;水温为25.3 ℃时,西施舌的耗氧率达到最大,为0.77 mg·g-1·h-1.处于适温状态(15 ℃和20 ℃)的O/N值要高于低温(10 ℃)和高温(25 ℃和30 ℃)时的O/N值,西施舌在适宜条件下更多地依赖于脂肪供能维持标准代谢,而在环境不适时则更多地调用机体的蛋白质来维持生理代谢需要. 相似文献

The reported rates of thermal 13-cis to all-trans isomerization of the protonated Schiff base of retinal (PSBR) in solution and in bacteriorhodopsin (BR) are shown to be correlated with the red shift in the absorption maximum of the chromophore, though the linear fit is different for BR and for a model PSBR in solution. Because the red shift in the absorption has been previously shown to be correlated with π-electron delocalization in the chromophore, this suggests that the thermal isomerization rate is largely regulated by the amount of double bond character in the chromophore. Because the linear fit of isomerization rates with absorption maxima is different for BR and the model PSBR, specific interactions of the protein with the chromophore must also be a factor in determining thermal isomerization rates in BR. A model of the later steps in the photocycle of BR is presented in which the 13-cis to all-trans thermal isomerization occurs during the O intermediate. 相似文献
