Using immunohistochemistry and SDS-PAGE, we studied the myosin heavy chain (MyHC) composition and fibre type distribution of hindlimb muscles of hopping and non-hopping Australian marsupials. We showed that hindlimb muscles of a bandicoot (Isoodon obesulus, order Peramelomorphia) and a small macropodoid, the brushtail bettong (Bettongia penicillata) expressed four MyHCs, slow, 2a, 2x and 2b, and had the corresponding fibre types as other macropods reported earlier. The fastest and most powerful 2b fibres predominated in most bettong hindlimb muscles, but were absent in the gastrocnemius and the flexor digitorum profundus, which are involved in elastic strain energy saving during hopping. The gastrocnemius of four large macropodids also showed little or no 2b MyHC, whereas this isoform was abundant in their tibialis anterior, which is not involved in elastic energy saving. In contrast, 2b MyHC predominated in the gastrocnemius of four non-hopping marsupials. These results suggest that absence of 2b fibres may be a general feature of macropodoid muscles involved in elastic energy saving. Large eutherians except llamas and pigs also have no 2b fibres. We hypothesize that 2x and 2a fibres perform better than 2b fibres in the storage and recovery of kinetic energy during locomotion in both marsupials and eutherians.  相似文献

Bacteriophages were observed in forestomach contents from three species of Australian macropodoid marsupials possessing a foregut fermentative digestion: the eastern grey kangaroo (Macropus giganteus), the eastern wallaroo (Macropus robustus robustus), and the rufous bettong (Aepyprymnus rufescens). Forty-six morphologically distinct phage types, representing the families Myoviridae, Siphoviridae, and Podoviridae, were identified. The range of forms varied between host species. The greatest diversity of phage types was found in forestomach contents of the wallaroo, and few phage types were recorded from the rufous bettongs. It is concluded that macropodoid marsupials, in common with their eutherian counterparts, possess diverse populations of bacteriophages in their fermentative forestomachs.  相似文献

Myosin heavy chains (MyHCs) and fibre types in the masseter muscle of seven species of Australian marsupials (brushtail and ringtail possums, bettong, bandicoot, dunnart, two species of antechinuses) spanning three orders were studied by native myosin electrophoresis, SDS-PAGE, immunoblotting and immunohistochemistry. We found only two fibre types in the masseter muscles of these animals: (1) masticatory fibres expressing masticatory MyHC, and (2) hybrid α/β fibres that co-express α-cardiac and β-cardiac MyHCs. Masticatory fibres predominate in most species, being appropriate for predation or for chewing tough vegetable matter. The relative abundance of α/β fibres decreased from 60% to 0 in the order: ringtail possum > brushtail possum > bettong > bandicoot > dunnart/antechinus. These variations in masseter fibre type are correlated with decreasing amounts of vegetable matter in the diets of these animals. The results are in contrast to earlier work on masseter fibres of macropodids that expressed α-cardiac MyHC almost homogeneously. The fact that the bettong (Family: Potoroidae), which belong to the same marsupial superfamily (Macropodoidea) as kangaroos and wallabies (Family: Macropodidae), has not specialized in the exclusive expression of α-cardiac MyHC as members of the latter family suggests that this specialization was of recent phylogenetic origin (30 million years before present).  相似文献

The amino acid sequence was established by N-terminal sequence analysis of high-performance liquid chromatography purified intact protein and peptides isolated from chemical and enzymatic digests of the protein. The protein contains 158 residues including four cysteines. The sequence comparison with the tamar wallaby (Macropus eugenii) late lactation protein shows only five differences. The protein is identified as a new member of a novel late lactation protein family present in the milk of marsupials.  相似文献

We report the first isolation and sequencing of genomic BAC clones containing the marsupial milk protein genes Whey Acidic Protein (WAP) and Early Lactation Protein (ELP). The stripe-faced dunnart WAPgene sequence contained five exons, the middle three of which code for the WAPmotifs and four disulphide core domains which characterize WAP. The dunnart ELPgene sequence contained three exons encoding a protein with a Kunitz motif common to serine protease inhibitors. Fluorescence in situ hybridization located the WAPgene to chromosome 1p in the stripe-faced dunnart, and the ELPgene to 2q. Northern blot analysis of lactating mammary tissue of the closely related fat-tailed dunnart has shown asynchronous expression of these milk protein genes. ELPwas expressed at only the earlier phase of lactation and WAPonly at the later phase of lactation, in contrast to beta-lactoglobulin (BLG) and alpha-lactalbumin (ALA) genes, which were expressed in both phases of lactation. This asynchronous expression during the lactation cycle in the fat-tailed dunnart is similar to other marsupials and it probably represents a pattern that is ancestral to Australian marsupials.  相似文献

The Tasmanian bettong (Bettongia gaimardi, a marsupial) is a rat-kangaroo that increases nonshivering thermogenesis (NST) in response to norepinephrine (NE). This study attempted to assess whether brown adipose tissue (BAT), a specialized thermogenic effector, is involved in NST in the bettong. Regulatory NST, indicated by resting oxygen consumption (Vo2) of the whole body, was measured under conscious conditions at 20 degrees C with various stimuli: cold (4 degrees -5 degrees C) or warm (25 degrees C) acclimation, NE injection, and the beta3-adrenoceptor agonist (BRL) 37344. In line with the functional studies in vivo, the presence of BAT was evaluated by examining the expression of the uncoupling protein 1 (UCP1) with both rat cDNA and oligonucleotide probes. Both NE and BRL 37344 significantly stimulated NST in the bettong. After cold acclimation of the animals (at 4 degrees -5 degrees C for 2 wk), the resting Vo2 was increased by 15% and the thermogenic effect of NE was enhanced; warm-acclimated animals showed a slightly depressed response. However, no expression of UCP1 was detected in bettongs either before or after cold exposure (2 wk). These data suggest that the observed NST in the marsupial bettong is not attributable to BAT.  相似文献

BackgroundThe carbohydrate fraction of mammalian milk is constituted of lactose and oligosaccharides, most of which contain a lactose unit at their reducing ends. Although lactose is the predominant saccharide in the milk of most eutherians, oligosaccharides significantly predominate over lactose in the milk of monotremes and marsupials.Scope of reviewThis review describes the most likely process by which lactose and milk oligosaccharides were acquired during the evolution of mammals and the mechanisms by which these saccharides are digested and absorbed by the suckling neonates.Major conclusionsDuring the evolution of mammals, c-type lysozyme evolved to α-lactalbumin. This permitted the biosynthesis of lactose by modulating the substrate specificity of β4galactosyltransferase 1, thus enabling the concomitant biosynthesis of milk oligosaccharides through the activities of several glycosyltransferases using lactose as an acceptor. In most eutherian mammals the digestion of lactose to glucose and galactose is achieved through the action of intestinal lactase (β-galactosidase), which is located within the small intestinal brush border. This enzyme, however, is absent in neonatal monotremes and macropod marsupials. It has therefore been proposed that in these species the absorption of milk oligosaccharides is achieved by pinocytosis or endocytosis, after which digestion occurs through the actions of several lysosomal acid glycosidases. This process would enable the milk oligosaccharides of monotremes and marsupials to be utilized as a significant energy source for the suckling neonates.General significanceThe evolution and significance of milk oligosaccharides is discussed in relation to the evolution of mammals.  相似文献

1. Twenty one sequential milk samples from an American marsupial (Monodelphis domestica) were analyzed. 2. The solids fraction was relatively low initially (10% w/w) and then increased to about 30% at mid-lactation where it remained until lactation ceased. 3. During the first 50 days of lactation the relative proportions of protein, fat and carbohydrate were 36, 28 and 34%, respectively, of the solids fraction. 4. Sodium and magnesium concentrations were constant; however levels of potassium and magnesium varied substantially. 5. The milk of M. domestica exhibits a similar pattern of quantitative and qualitative changes to that seen in Australian marsupials.  相似文献

The woylie or brush-tailed bettong (Bettongia penicillata) is a medium-sized native Australian marsupial that has undergone a dramatic decline in numbers in recent years. Trypanosome parasites have been identified in the woylie but little is known about the prevalence and clinical impact of other haemoprotozoan parasites in these marsupials. In the present study, the occurrence and molecular phylogeny of a piroplasm was studied in woylies from six different sites in Western Australia (WA). Blood samples were screened by PCR at the 18S rRNA locus and 80.4% (123/153) of the blood samples were positive for piroplasm DNA. Sequence and phylogenetic analysis of 12 of these positives identified them as Theileria penicillata, and sequencing of cloned PCR products indicated that no other species of Theileria were present. Infected woylies had a lower body weight but microscopic evaluation of the blood films indicated that T. penicillata did not appear to cause red cell injury or anaemia. Further studies are required to determine the clinical significance of T. penicillata in woylies.  相似文献

beta-Lactoglobulin has been isolated in the milk of the Eastern Grey Kangaroo (Macropus giganteus). This is the first time this protein has been reported to be in the milk of marsupials. The complete amino-acid sequence has been determined by spinning cup and pulsed liquid phase microsequencing of the protein and peptides after enzymatic or cyanogen bromide cleavages. The 155-residue protein is the shortest beta-lactoglobulin so far sequenced. When the kangaroo protein is included in a comparison of the members of the beta-lactoglobulin family, the percentage of residues common to all members is reduced from 33% to 13%. Despite the large number of accumulated amino-acid exchanges the protein exists as a dimer and shows higher homology to the usually very conservative dimeric, ruminant beta-lactoglobulins than to the monomeric protein from monogastrics. Half-cystine residues that form disulphide bridges are conserved. The Eastern Grey Kangaroo beta-lactoglobulin possesses significant homology in several characteristic segments thought to be important for a functional trait common to the beta-lactoglobulin family and retinol-binding proteins. Structural similarity to the retinol-binding protein is indicated by 22% of identical residues. Homology to the beta-lactoglobulins and retinol-binding proteins, the binding site and possible function based on comparative structural studies are discussed.  相似文献

The activities of galactokinase, hexose-1-phosphate uridylyl transferase and UDPglucose 4-epimerase in homogenates of livers of two adult and 20 suckling tammar wallabies aged from 6 to 50 weeks were investigated. The activities of all three enzymes were high until 24-30 weeks post partum, after which they declined to low levels. The activities of the three liver enzymes were high in pouch young of six other species of marsupial. Comparison of the activities of the three liver enzymes in suckling tammar wallabies with those in suckling rats showed no difference between the two species in regard to galactokinase and uridylyl transferase, but the UDPglucose 4-epimerase activity in tammar wallabies was approximately double than found in rats. This may be related to the high galactose content of tammar wallaby milk compared with rat milk. In suckling tammar wallabies, the liver had higher enzyme activities than other tissues studied. It is concluded that, contrary to the suggestion of Stephens et al. (1975), pouch young marsupials are not deficient in their ability to metabolize galactose.  相似文献

Milk was obtained from three captive servals. The average nutrient content was 158.3+/-44.4 g protein; 152.6+/-62.3 g fat; and 68.7+/-31.4 g lactose per kg milk. The protein fraction respectively consisted of 117.7+/-44.8 g caseins per kg milk and of 40.6+/-6.7 g whey proteins per kg milk. Electrophoresis and identification of protein bands showed a similar migrating sequence of proteins as seen in cheetah and cat milk, with small differences in the beta-caseins. The lipid fraction contains 313.3+/-18.8 g saturated and 338.6+/-11.9 g mono unsaturated fatty acids per kg milk fat respectively. The high content of 292.4+/-24.9 g kg(-1) milk fat of polyunsaturated fatty acids is due to a high content in linolenic acid. No short chain fatty acids, but substantial levels of uneven carbon chain fatty acids were observed. In general, serval milk has a higher protein and fat content than that of the domestic cat and cheetah, and a lower content of unsaturated fatty acids than that of the domestic cat.  相似文献

Erythrocytic galactokinase and/or galactose-1-phosphate uridyl transferase activity were low in many species of marsupials. However, cataract formation was observed only in pouch-young members of these species when reared on cow's milk. The galactose tolerance of young kangaroos was found to be greatly impaired, but improved rapidly and markedly at the stage of which the definitive structure of the ruminant type of stomach as in adults is formed. The combination of high absorption of galactose and low levels of galactokinase and/or transferase thus appears to determine the predisposition of pouch-young marsupials to galactose cataractogenesis.  相似文献

The milk constituents of Dasyurus viverrinus, a carnivorous marsupial, exhibited major quantitative and qualitative changes during the course of lactation. The milk produced in the early stages of lactation was dilute, about 13-16% (w/w) solids before 3 weeks with carbohydrate representing the major fraction. In the latter stages of lactation the milk was concentrated, around 30% solids, and lipid was the predominant fraction. Palmitic acid was the major fatty acid present in early-stage milk but oleic acid became predominant in milk after 10 weeks post-partum. The changes in milk composition in D. viverrinus were similar to those described for the milks of herbivorous marsupials which therefore suggests that this pattern may be uniform throughout the Marsupialia.  相似文献

Milk samples were obtained at regular intervals throughout lactation from northern brown bandicoots, Isoodon macrourus, in captivity. Total concentration of milk solids was initially 7% (w/w) and increased linearly to 45% (w/w) by 55 days. Carbohydrate, lipid and protein concentrations increased from about 2% (w/w) to about 7-8% (w/w) at 30 days. Thereafter they diverged, with lipid increasing to between 25-30% (w/w) at 56 days, protein reaching maximal values of 10-15% (w/w) at just over 40 days and carbohydrate gradually declining to about 5% (w/w) at 56 days before a rapid fall to 1-2% (w/w) at the completion of lactation. The milk of the bandicoot exhibits a similar pattern of change during the course of lactation to that shown by other marsupials.  相似文献

1. 1H NMR spectra were acquired from whole plasma, intact erythrocytes, and ultrafiltrates of erythrocytes from nine native and eight introduced (domestic) Australian animals; single-pulse, spin-echo and 2-dimensional spectra were obtained. The aim was to detect and at least semi-quantify metabolites in the samples and compare the profiles amongst the species. 2. The Australian natives that were studied were all marsupials: greater brown bandicoot; bettong; eastern grey kangaroo; red kangaroo; koala; possum; red necked pademelon; Tammar wallaby; and wombat. The introduced mammals that were studied were: cat; cattle; dog; goat; horse; pig; rabbit; and sheep. 3. Because of the range of habitats and diets amongst the animals, it was postulated that the concentrations of the common metabolites in the blood would show marked differences and that there would also be some metabolites that were peculiar to a given animal. There were several major differences in the spectra: in the spectra of plasma, the glycoprotein and lipoprotein resonances showed the largest inter-species variation, whereas the most dramatic finding from the spectra of erythrocytes was a very high concentration of lysine in the cells from the Tammar wallaby.  相似文献

An examination of movement paths, foraging patterns and habitat use of an endangered mycophagous marsupial, the northern bettong (Bettongia tropica), was undertaken in fire‐prone forest in north‐eastern Australia before and after experimentally induced fires. Fungal biomass remaining at bettong foraging points was similar across the study area prior to burning, but increased significantly on burnt ground during the period after fire. After burning, significantly more bettongs chose to forage in burnt habitat and those that did experienced higher probabilities of truffle recovery. Using data from spool‐and‐line tracking, observed movement patterns of bettongs were compared with those expected from a simple null model of animal movement (a correlated random walk). Analysis of mean‐squared displacement revealed that 22% of observations fell beyond the model’s 95% prediction interval. Further analysis revealed the reasons for the model’s inadequacy: bettongs exhibited area‐restricted search behaviour by taking significantly more frequent and more acute turns immediately prior to and following recovery of hypogeous fungi (truffles), and by taking significantly more frequent and more acute turns following any other foraging activity. In general ecological terms, the results indicate a flexible response by the northern bettong to habitat alteration and increased food availability brought about by low intensity fires.  相似文献

The objective of the present study was to determine the effects of rumen-protected choline (RPC) supplementation on body condition, milk production and milk choline content during the periparturient period. Thirty-two Holstein cows were allocated into two groups (RPC group - with RPC supplementation, and control group - without RPC supplementation) 28 days before the expected calving. Cows were fed the experimental diet from 21 days before expected calving until 60 days of lactation. The daily diet of the RPC group contained 100 g of RPC from 21 days before calving until calving and 200 g RPC after calving for 60 days of lactation, which provided 25 g and 50 g per day choline, respectively. Body condition was scored on days -21, 7, 35 and 60 relative to calving. Milk production was measured at every milking; milk fat, protein and choline content were determined on days 7, 35 and 60 of lactation. Body condition was not affected by RPC supplementation. Milk yield was 4.4 kg higher for the group of cows receiving supplementary choline during the 60 days experimental period and 4% fat-corrected milk production was also increased by 2.5 kg/day. Milk fat content was not altered by treatment, but fat yield was increased by 0.10 kg/day as a consequence of higher milk yield in the RPC-treated group. Milk protein content tended to increase by RPC supplementation and a 0.18 kg/day significant improvement of protein yield was detected. Milk choline content increased in both groups after calving as the lactating period advanced. However, milk choline content and choline yield were significantly higher in the RPC group than in the control group. The improved milk choline and choline yield provide evidence that some of the applied RPC escaped ruminal degradation, was absorbed from the small intestine and improved the choline supply of the cows and contributed to the changes of production variables.  相似文献
