Effects of azadirachtin, a triterpenoid extracted from neem seed, Azadirachta indica A. Juss., were similar to those of insect growth regulators against the immature stages of the born fly, Haematobia irritans (L.), the stable fly, Stomoxys calcitrans (L.), and the house fly, Musca domestica L. When an ethanolic extract of ground seed was blended into cow manure, LC50 and LC90's for larval horn flies were 0.096 and 0.133 ppm azadirachtin, respectively. An emulsifiable concentrate (EC) had an LC50 for larval horn flies of 0.151 ppm and an LC90 of 0.268 ppm. For larval stable flies, the EC formulation had an LC50 of 7.7 ppm and an LC90 of 18.7 ppm azadirachtin in manure. Against larval house flies, the LC50 and LC90 were 10.5 and 20.2 ppm, respectively. When the EC formulation was administered orally to cattle at a rate of greater than or equal to 0.03 mg azadirachtin per kg of body weight per day or when ground neem seed was given as a daily supplement of greater than or equal to 10 mg seed per kg body weight, horn fly development in the manure was almost completely inhibited. In contrast, ground seed mixed in cattle feed at the rate of 100-400 mg seed per kg of body weight per day caused less than 50% inhibition of stable flies in the manure.  相似文献

The human genome is divided into isochores, large stretches (>300 kb) of genomic DNA with more or less consistent GC content. Mutational/neutralist and selectionist models have been put forward to explain their existence. A major criticism of the mutational models is that they cannot account for the higher GC content at fourfold-redundant silent sites within exons (GC4) than in flanking introns (GCi). Indeed, it has been asserted that it is hard to envisage a mutational bias explanation, as it is difficult to see how repair enzymes might act differently in exons and their flanking introns. However, this rejection, we note, ignores the effects of transposable elements (TEs), which are a major component of introns and tend to cause them to have a GC content different from (usually lower than) that dictated by point mutational processes alone. As TEs tend not to insert at the extremities of introns, this model predicts that GC content at the extremities of introns should be more like that at GC4 than are the intronic interiors. This we show to be true. The model also correctly predicts that small introns should have a composition more like that at GC4 than large introns. We conclude that the logic of the previous rejection of neutralist models is unsafe.  相似文献

cDNAs for alcohol dehydrogenase (ADH) isozymes were cloned and sequenced from two tephritid fruit flies, the medfly Ceratitis capitata and the olive fly Bactrocera oleae. Because of the high sequence divergence compared with the Drosophila sequences, the medfly cDNAs were cloned using sequence information from the purified proteins, and the olive fly cDNAs were cloned by functional complementation in yeast. The medfly peptide sequences are about 83% identical to each other, and the corresponding mRNAs have the tissue distribution shown by the corresponding isozymes, ADH-1 and ADH-2. The olive fly peptide sequence is more closely related to medfly ADH-2. The tephritid ADHs share less than 40% sequence identity with Drosophila ADH and ADH-related genes but are >57% identical to the ADH of the flesh fly Sarcophaga peregrina, a more distantly related species. To explain this unexpected finding, it is proposed that the ADH: genes of the family Drosophilidae may not be orthologous to the ADH: genes of the other two families, Tephritidae and Sarcophagidae.  相似文献

The neotropical warble fly, Dermatobia hominis (Fig. 1), has plagued neotropical America since preColombian times, and has become an economically important pest causing substantial losses to the meat, milk and leather industries from northern Mexico down to northern Argentina. Its life cycle (Box 1) is astonishingly complex, requiring another insect as a phoretic carrier of its eggs to the skin of its mammal hosts. ABSTRACT. A rapid decline in receptivity of mated female Glossina morsitans morsitans Westwood is shown to depend on both physical and chemical stimuli associated with copulation. Radiolabelling revealed the transfer of substances from the male to the haemolymph of the female during copulation. Implantation of male tissues or their injection as homogenates into virgin females showed the chemical stimulus to come from the male accessory glands. Receptivity decreased in females mated to males with ejaculatory ducts severed or testes removed and also in females which had a glass bead inserted into their uterus and/or the tip of their abdomen covered with wax, suggesting that a physical stimulus inducing refractoriness is provided by distension of the uterus and/or stimulation of their terminalial setae. Exposing virgin females to daily short matings in which no male materials were transferred, confirmed this. Receptivity also declined slowly with age in unexposed virgin females. Transfusion of haemolymph from mated females (up to 11 days old) into virgins did not indicate the existence of a haemolymph-borne ovulation-inducing factor; apparently only physical stimuli from mating are involved in the induction of ovulation, and somehow prime the ovarian tissue so that it responds appropriately later when the egg has matured. Whether the stimulus is transmitted to the ovary neurally or hormonally is unknown.  相似文献

1. Insoluble and "soluble" forms of cytochrome c oxidase were prepared from house flies by methods described for mammalian tissue. A 4.5-fold purification was accomplished by preparing the enzyme in insoluble form (as compared with a crude water brei). The oxidase in "soluble" form was purified 20.0-fold. 2. The absorption spectrum of the "soluble" preparation from flies was similar to that from mammalian tissue, and indicated the presence of cytochromes a + a(3) and b. In contrast with the corresponding mammalian enzymes these components were not separable by the existing techniques. 3. The Soret peak seen at 440 mmicro and the alpha peak at 600 mmicro were due. primarily to cytochromes a(3) and a, respectively. 4. The oxidase activity of whole house flies was comparable with that of rat abdominal muscle. In both organisms, this activity was inhibited by cyanide to approximately the same extent. 5. The optimum buffer concentration for oxidase activity, as measured spectrophotometrically, was approximately 0.02 to 0.06 M. This is in agreement with results obtained for mammalian enzyme, as measured both manometrically and spectrophotometrically. 6. The present study has demonstrated the following components of the cytochrome system in house flies: b, c, a, and a(3).  相似文献

No-choice tests were conducted to determine whether fruit of southern highbush blueberry, Vaccinium corymbosum L., hybrids are hosts for three invasive tephritid fruit flies in Hawaii. Fruit of various blueberry cultivars was exposed to gravid female flies of Bactrocera dorsalis Hendel (oriental fruit fly), Ceratitis capitata (Wiedemann) (Mediterranean fruit fly), or Bactrocera cucurbitae Coquillet (melon fly) in screen cages outdoors for 6 h and then held on sand in the laboratory for 2 wk for pupal development and adult emergence. Each of the 15 blueberry cultivars tested were infested by oriental fruit fly and Mediterranean fruit fly, confirming that these fruit flies will oviposit on blueberry fruit and that blueberry is a suitable host for fly development. However, there was significant cultivar variation in susceptibility to fruit fly infestation. For oriental fruit fly, 'Sapphire' fruit produced an average of 1.42 puparia per g, twice as high as that of the next most susceptible cultivar 'Emerald' (0.70 puparia per g). 'Legacy', 'Biloxi', and 'Spring High' were least susceptible to infestation, producing only 0.20-0.25 oriental fruit fly puparia per g of fruit. For Mediterranean fruit fly, 'Blue Crisp' produced 0.50 puparia per g of fruit, whereas 'Sharpblue' produced only 0.03 puparia per g of fruit. Blueberry was a marginal host for melon fly. This information will aid in development of pest management recommendations for blueberry cultivars as planting of low-chill cultivars expands to areas with subtropical and tropical fruit flies. Planting of fruit fly resistant cultivars may result in lower infestation levels and less crop loss.  相似文献

Recent studies of gene expression in the developing fruitfly leg support a model – Meinhardt's Boundary Model – which seems to contradict the prevailing paradigm for pattern formation in the imaginal discs of Drosophila – the Polar Coordinate Model. Reasoning from geometric first principles, this article examines the strengths and weaknesses of these hypotheses, plus some baffling phenomena that neither model can comfortably explain. The deeper question at issue is: how does the fly's genome encode the three-dimensional anatomy of the adult? Does it demarcate territories and boundaries (as in a geopolitical map) and then use those boundaries and their points of intersection as a scaffolding on which to erect the anatomy (the Boundary Model)? Or does it assign cellular fates within a relatively seamless coordinate system (the Polar Coordinate Model)? The existence of hybrid Cartesian-polar models shows that the alternatives may not be so clear-cut: a single organ might utilize different systems that are spatially superimposed or temporally sequential.  相似文献
