The gorgonian Leptogorgia sarmentosa contains three new steroids, (20S)-20-hydroxycholestane-3,16-dione (1), (16S, 20S)-16,20-dihydroxycholestan-3-one (2), and (20S)-20-hydroxycholest-1-ene-3,16-dione (3) together with a known related compound (4). Their structures were defined by spectroscopic analysis. The new steroids exhibited significant cytotoxicity against four tumor cell lines (ED50 = 1 microg/ml).  相似文献

A series of new polyoxygenated steroid derivatives with various steroid skeleton moieties were synthesized. Antitumor activity of the compounds against three tumor cell lines (Breast cancer MCF7, lung cancer NCI and oral cancer KB) were evaluated. Compounds with aromatic A ring of this series exhibited the most potent cytotoxicities in all tested cells. The absence of OH at C-16 or lack of cholesterol like side chain at C-20 in the steroid skeleton apparently result in decreased cytotoxicity. The compound became inactive when the side chain contains double bond at C-24-C-25. When hydroxyl group at C-3 was protected no cytotoxicities against MCF7 and NCI and considerable low cytotoxicity against KB cell lines were observed.  相似文献

Abstract

Leptogorgia sarmentosa (Cnidaria: Octocorallia) is a common soft bottom-gravel gorgonian in the northwestern Mediterranean Sea. This study examined the annual reproductive cycle of a L. sarmentosa. population at a depth of 30 m, and further determined the sex ratio, the intra-colonial gonadal output variability and the age at first reproduction. L. sarmentosa was gonochoric and the population studied was characterized by a sex ratio slightly biased towards female colonies (1 male:1.6 female). Secondary branches had higher polyp fertility (number of gonads per polyp) compared to primary branches, but because of the number of polyps on the former, most of the sexual effort was actually contributed by the primary branches, as in other gorgonian species. Male colonies became fertile above a height of 21 cm, but female colonies had gonads even in the 1–10 cm height class. Spawning occurred between late August and early September, although there was no direct observation of broadcast spawning or surface brooder strategy. The maximum gonad size was 550 μm in males and 500 μm in females, with a mean diameter of 270 μm for spermaries, and 190 μm for eggs. The number of gonads per polyp prior to spawning reached 4–5 spermaries polyp!1 in male colonies, and 3–4 eggs polyp!1 in female colonies. Spawning did not appear to be synchronized with the full moon. L. sarmentosa appeared to have a different reproductive strategy to other Mediterranean gorgonians (living on hard bottom substrata), probably because of its different habitat preferences and biological (morphology, diet, prey capture rates, growth, biochemical levels) features.  相似文献

The gorgonian coral Leptogorgia virgulata contains thyroxine, or a thyroxine-like substance, referred to here as G-T(4). The G-T(4) levels were significantly higher in colonies collected in the summer vs. winter months. Using immunocytochemical techniques, G-T(4) was localized in the axis, polyp epithelium, and within the electron dense bodies of scleroblasts (spicule-forming cells), as well as on the periphery of spicules. G-T(4) was also localized in the mesoglea between closely adjacent scleroblasts. The effects of exogenous T(4) on the uptake of Ca(45) was determined in spicule, tissue and axis fractions of L. virgulata. The uptake of Ca(45) increased in T(4) treated spicules but decreased in the tissue fraction for all time periods tested. The uptake of Ca(45) into axes was not affected by exogenous T(4) until day 10 of the study. These data suggest that G-T(4) may function in the process of spicule formation. 1,25-dihydroxyvitamin D apparently is synthesized via ultraviolet radiation. Colonies deprived of ultraviolet radiation had significantly more 'irregular' spicules than colonies maintained in ultraviolet radiation. Exposure to sunlight therefore may be associated with the process of normal spicule formation.  相似文献

Heterosteroids remain interesting due to their potential biological activities. This prompted us to synthesize novel thia steroids possessing the heteroatom in the A-ring. We set out to describe a new and versatile method for preparing 3-thia steroids from cholic acid via a selective oxidation of one hydroxyl group, a Baeyer-Villiger oxidation and a photolysis as the key steps. The characteristic 1H and 13C NMR spectroscopic features of the synthesized compounds are reported.  相似文献

Along the northeastern Gulf of Mexico coast, Leptogorgia hebes is the predominant gorgonian at two inner shelf (22 and 27 m) sites, while Leptogorgia virgulata dominates a third shallow (1–2m), inshore site. The axial skeleton of both species is composed of concentric rings which appeared to exhibit annual periodicity. Although age structure of the populations differed between shelf sites, neither colony growth rates, estimated from direct measurements of ring widths plotted on Walford graphs, nor overall branching complexity, determined from bifurcation ratios (Rb), differed between the shelf populations. Larger individuals at the 27m shelf site were simply older than those at the shallower (22m) shelf site. Walford plots of growth increments revealed colony growth of the shallow inshore species to be similar to that of the offshore species.  相似文献

In this study, we analyzed the response of the temperate, shallow-water gorgonian, Leptogorgia virgulata, to temperature stress. Proteins were pulse labeled with (35)S-methionine/cysteine for 1 h to 2 h at 22 degrees C (control), or 38 degrees C, or for 4 h at 12.5 degrees C. Heat shock induced synthesis of unique proteins of 112, 89, and 74 kDa, with 102, 98 and 56 kDa proteins present in the control as well. Cold shock from 22 degrees C-12.5 degrees C induced the synthesis of a 25 kDa protein, with a 44 kDa protein present in the control as well. Control samples expressed unique proteins of 38, and 33 kDa. Non-radioactive proteins expressed under the same conditions as above, as well as natural field conditions, were tested for reactivity with antibodies to heat shock proteins (HSPs). HSP60 was the major protein found in L. virgulata. Although HSP47, HSP60, and HSP104 were present in all samples, the expression of HSP60 was enhanced in heat stressed colonies, while HSP47 and HSP104 expression were greatest in cold shocked samples. Inducible HSP70 was expressed in cold-shocked, heat-shocked, and field samples. Constitutively expressed HSP70 was absent from all samples. The expression of HSP90 was limited to heat shocked colonies. The expression of both HSP70 and HSP104 suggests that the organism may also develop a stress tolerance response.  相似文献

  • 1.1. Longitudinally split or completely regenerated branch tips from Leplogorgia virgulata show no differences in calcium uptake between control and ouabain treatments. This indicates that there is no ouabain sensitive Na+, K+-ATPase involved in calcium uptake.
  • 2.2. The tissue fractions of both regenerated and split branch tips show, at certain times, higher calcium uptake than control fractions. In the spicule fractions of these tips calcium uptake decreases in vandate treated specimens.
  • 3.3. Pulse-chase experiments show an initial rapid release of calcium from the tips into surrounding seawater.
  • 4.4. The results may suggest the presence of outwardly directed calcium pumps on the basal/lateral and apical plasma membranes of the epithelial cells. Outwardly directed calcium pumps may also be envisaged on the cell membranes of scleroblasts. In addition, pumps may move calcium into specific organelles of the scleroblasts en route to the spicule forming vacuoles.
  • 5.5. These pumps are likely to be Ca2+-ATPase.

Chemical investigation on gorgonian coral Subergorgia suberosa collected from the South China Sea led to the isolation and identification of eight 17-acetyl pregnane steroids (18), of which 1, 6 and 7 were obtained for the first time from this species, as well as from the genus Subergorgia. Compounds 1 and 7 are new natural products. Compared with other species in the genus Subergorgia, pregnane steroids with 17-acetyl group (18) were only achieved from S. suberosa, suggesting that the 17-acetyl pregnane steroids might be characteristic constituents of S. suberosa, and may be useful as chemotaxonomic markers for the species S. suberosa.  相似文献

Two new pentahydroxylated steroids, (3β,12β,16β,23E)-cholesta-5,23-diene-3,12,16,20,25-pentaol (1) and (3β,12β,16β)-cholesta-5,25(26)-diene-3,12,16,20,24-pentaol (2), were isolated from the EtOH extract of the South China Sea gorgonian Subergorgia suberosa. In addition, four known steroids were detected. The structures of these compounds were established by spectroscopic methods and comparison of their data with those of the related known compounds. Two new polyhydroxysteroids isihippurols A (1) and B (2) were isolated from the MeOH extract of the gorgonian Isis hippuris in addition to nine known steroids namely gorgosterol, 3alpha-acetoxy-24-methyl 11beta,18; 18,20beta; 22,25-triepoxy-5alpha-furostane, hippurin-1, 22-epi-hippuristanol, 22-epi-hippurin-1, 3-acetyl-2-deacetyl-22-epi-hippurin-1, 2-deacetylhippurin-1, 3beta, 7alpha, 11alpha-trihydroxygorgost-5-ene-12beta-acetate and 2-deacetyl-22-epi-hippurin-1. The structures of 1 and 2 have been determined as 1alpha, 3beta, 5alpha, 6beta, 11alpha-pentahydroxygorgosta-12beta-monoacetate and 1alpha, 3beta, 11alpha-trihydroxygorgosta-5alpha, 6alpha-epoxy-12beta-monoacetate respectively by spectral analysis and chemical correlation.  相似文献

Spicules of the gorgonian Leptogorgia virgulata possess an insoluble matrix fraction that is predominantly collagenous in summer months. This collagenous component is largely absent in winter months. Using an antibody directed against the 140 kD collagenous protein (CP) of the insoluble matrix, immuno-gold labelling was employed to localized this protein at the transmission electron-microscopy level throughout the year, and in different areas of the gorgonian colonies. Within the tip regions, the 140 kD CP varied throughout the year in the spicules, electron-dense bodies (EDBs) of scleroblasts, polyp vesicles, desmocytes and axes. In the mid and base regions, the 140 kD CP varied throughout the year in the spicules, EDBs and lysosomes of scleroblasts, desmocytes and axes. This variation in the location and density of the label suggests a dynamic annual cycling of the collagenous component of the insoluble matrix. EDBs may transport a collagenous component of the matrix to the spicule-forming vacuole. A component of the 140 kD CP may be transported and/or degraded by polyp vesicles and lysosomes, respectively. The pattern of labelling of the axial region suggests that translocation and storage of a component of the collagenous protein may occur. Environmental factors may be responsible for the triggering of matrix cycling.  相似文献

A new 8-hydroxy briarane diterpenoid (compound 1) with antifungal activity was isolated along with known compounds (2-5) from the gorgonian coral Junceellajuncea. On the basis of spectroscopic data (1D and 2D NMR, MS), the structure of the compound 1 was established as (1S,2S,8S,9S,10S,11R,12R,14S,17R)-11,20-epoxy-14-(3-methylbutanoyl)-2,9,12-triacetoxy-8-hydroxybriar-5(16)-en-18,7-olide. Compound 1 exhibited significant activity against fungi (Aspergillus niger, Candida albicans and Penicillium notatum) but the other compounds (2-5) did not exhibit the activity against fungi.  相似文献

Protoapigenone, a natural flavonoid possessing an unusual p-quinol moiety on its B-ring, is a novel prospective anticancer agent with low toxicity that is currently in development. The first economical, one-step synthesis of protoapigenone from apigenin is described on up to gram scale. 13 new 1'-O-alkylflavone analogs were also synthesized, either from apigenin or β-naphthoflavone. The in vitro cytotoxic activity of each compound was tested on six human cancer cell lines (HepG2, Hep3B, Ca9-22, A549, MCF-7 and MDA-MB-231). In the case of 1'-O-alkyl-protoapigenone derivatives, structure-activity relationships were found depending on the side-chain, and protoapigenone 1'-O-butyl ether was found to exert significantly stronger activity against three of the cell lines (Hep3B, MCF-7 and MDA-MB-231) than its non-substituted analog, protoapigenone itself. In contrast to this, all β-naphthoflavone derivatives bearing the same pharmacophore on their B-ring showed decreased cytotoxic activities when substituted with an O-alkyl side-chain at position 1', comparing to that of the non-substituted compound.  相似文献

Mesogligorgia scotiae gen. nov., sp. nov. is described and illustrated from a colony collected in the Scotia Sea, 2,201–2,213 m in depth, on the ANDEEP-I cruise. The new taxon is placed in the family Plexauridae because of: 1) the presence of a horny axis with a cross-chambered central core and numerous loculi, 2) retractile polyps in calyces with distinct spicular components, and 3) armed polyps with large sclerites with a poorly- developed collaret and eight well-developed points. The irregularly distributed sclerites running along the axis, into a thick mesogloeal coenenchyme, and the elongated spindles with irregular ends are the most distinctive characters of the newly proposed genus.  相似文献

Summary The route of calcium transport to the sites of spicule formation in the gorgonian Leptogorgia virgulata has been examined by the use of 45Ca as a tracer in light- and electron-microscopic autoradiography. From 1 to 15 min after the 15-min incubation the tracer accumulates in the axis. After 15 min there is a movement of label out of the axis largely to the peripheral region of the axis, the axial epithelium, and the mesoglea. By 60 min much of the label is in the spicules reaching its maximum level at 120 min. When calcium enters the scleroblast from the mesoglea, it appears to be transported to the spicule by electron-dense bodies. There does not appear to be a simultaneous release of all ionic calcium from the axis, but rather a continuously increasing efflux which levels off at 60–120 min. Not all of the calcium reaching the axis will traverse it en route to spicules; instead, a portion of it apparently precipitates as an amorphous compound.Contribution No. 550; Belle W. Summary The sequence of the synthesis and transport of the organic matrix of spicules has been elucidated in the gorgonian Leptogorgia virgulata by use of 3H-aspartic acid as the tracer in electron-microscopic autoradiography. The entire process of matrix synthesis and transport takes approximately 2 h. It seems that the protein moiety of the organic matrix is synthesized in the RER prior to 5 min following the initial 10 min incubation in the tracer. At the 5 min chase the label is moving from the RER to the Golgi complexes where the carbohydrate moiety of the matrix is presumed to be synthesized. At the 5 to 15 min chases the label is transported out of the Golgi complexes via Golgi vesicles. This phase continues for 30 min. From 60 to 120 min the 3H-aspartic acid moves to the spicules. After 120 min the majority of the label has moved into the spicules. Silver grain counts over both multivesicular and electron-dense bodies remain at relatively low and constant levels over 4 h indicating that neither organelle is involved in the synthesis and transport of the organic matrix.Contribution No 512; Belle W. A library of extracts established from hundreds of marine organisms was screened by a cytotoxicity test. The active organic extract of an Okinawan marine sponge of the genus Dysidea was subjected to bioassay-guided fractionation to give three new polyoxygenated steroids dysideasterols F-H (1-3), together with two known related compounds (4 and 5). Their structures were confirmed by NMR and mass spectroscopic analyses. A characteristic structural feature of 2, 4 and 5 is an allylic epoxide, whereas this epoxide undergoes ring-opening by a neighbouring hydroxyl group to give a tetrahydrofuran ring in 1 and 3. All compounds 1-5 exhibited a similar cytotoxic effect with IC50 values of 0.15-0.3 μM against human epidermoid carcinoma A431 cells, demonstrating that the allylic epoxide moiety was not responsible for this cytotoxic effect.  相似文献
