Catumaxomab, a monoclonal bispecific trifunctional antibody, was approved in the european Union in April 2009 for the intraperitoneal treatment of patients with malignant ascites. The marketing authorization holder Fresenius Biotech GmbH developed catumaxomab (Removab®) together with its partner TRiOn Pharma GmbH, Germany. it is the first substance worldwide with a regulatory label for the treatment of malignant ascites due to epithelial carcinomas. Since the peritoneum is of mesothelial origin and therefore lacks epCAM expression, the intraperitoneal administration of catumaxomab is an attractive targeted immunotherapeutic approach. Catumaxomab is able to destroy epCAM positive tumor cells in the peritoneal cavity known as the main cause of malignant ascites. in addition, catumaxomab is a potential therapeutic option for several primary tumors since the epCAM molecule is expressed on the majority of epithelial carcinomas. Peptide therapeutics have played a notable role in medical practice since the advent of insulin therapy in the 1920s. Over 60 peptide drugs are approved in the United States and other major markets, and peptides continue to enter clinical development at a steady pace. Peptide drug discovery has diversified beyond its traditional focus on endogenous human peptides to include a broader range of structures identified from other natural sources or through medicinal chemistry efforts. We maintain a comprehensive dataset on peptides that have entered human clinical studies that includes over 150 peptides in active development today. Here we provide an overview of the peptide therapeutic landscape, including historical perspectives, molecular characteristics, regulatory benchmarks, and a therapeutic area breakdown.  相似文献

The Kabat Database was initially started in 1970 to determine the combining site of antibodies based on the available amino acid sequences. The precise delineation of complementarity determining regions (CDR) of both light and heavy chains provides the first example of how properly aligned sequences can be used to derive structural and functional information of biological macromolecules. This knowledge has subsequently been applied to the construction of artificial antibodies with prescribed specificities, and to many other studies. The Kabat database now includes nucleotide sequences, sequences of T cell receptors for antigens (TCR), major histocompatibility complex (MHC) class I and II molecules, and other proteins of immunological interest. While new sequences are continually added into this database, we have undertaken the task of developing more analytical methods to study the information content of this collection of aligned sequences. Flowers, the reproductive structures of the approximately 400 000 extant species of flowering plants, exist in a tremendous range of forms and sizes, mainly due to developmental differences involving the number, arrangement, size and form of the floral organs of which they consist. However, this tremendous diversity is underpinned by a surprisingly robust basic floral structure in which a central group of carpels forms on an axis of determinate growth, almost invariably surrounded by two successive zones containing stamens and perianth organs, respectively. Over the last 25 years, remarkable progress has been achieved in describing the molecular mechanisms that control almost all aspects of flower development, from the phase change that initiates flowering to the final production of fruits and seeds. However, this work has been performed almost exclusively in a small number of eudicot model species, chief among which is Arabidopsis thaliana. Studies of flower development must now be extended to a much wider phylogenetic range of flowering plants and, indeed, to their closest living relatives, the gymnosperms. Studies of further, more wide-ranging models should provide insights that, for various reasons, cannot be obtained by studying the major existing models alone. The use of further models should also help to explain how the first flowering plants evolved from an unknown, although presumably gymnosperm-like ancestor, and rapidly diversified to become the largest major plant group and to dominate the terrestrial flora. The benefits for society of a thorough understanding of flower development are self-evident, as human life depends to a large extent on flowering plants and on the fruits and seeds they produce. In this preface to the Special Issue, we introduce eleven articles on flower development, representing work in both established and further models, including gymnosperms. We also present some of our own views on current trends and future directions of the flower development field.  相似文献

This paper outlines some of the basic ideas of the orienting response (OR) that have developed from the classical writings of E.N. Sokolov, in particular the effects of stimulus novelty, intensity, and significance upon the OR, and predictions about these effects on a range of physiological measures traditionally associated with the OR. Such measures include the GSR, respiration, heart rate, vascular responses, EEG, and pupil diameter. Unfortunately, many of the predictions of classical OR theory do not hold up when such a fine-grain analysis is undertaken. Possible conceptualizations of the discrepancies between Sokolovian predictions and empirical data are considered--should we accept such discrepancies as merely reflecting the imperfect nature of many OR indices, or seek other regularizing principles? Preliminary Process Theory has been proposed as one alternative account of the existing data base, and similarities and differences between it and Sokolovian theory are explored. The need for further investigation of such problems, in the very foundations of OR theory, appears to be of fundamental importance to the future status of the OR. An appendix provides a discussion between Barry and Sokolov on some of these issues.  相似文献

Fundamental research in enzymology have remarkably developed during the past thirty years. However, the actual application of enzymes has been rather limited for several technical and economical reasons. Such limiting factors are now being overcome year after year. Immobilized enzyme and cell technology might be a kind of break-through in this field. From the industrial view-point, enzyme technology is now coming into its real developmental stage. Enzyme technology has recently been finding several directions for its development. These directions may be classified as follows : (1) new industrial catalysts, (2) Tools for food production and processing, (3) pharmaceutical uses, (4) analytical and measurement tools, (5) aids for screening new physiologically active substances, and (6) aids for creation of new sources of energy and raw materials. The authors wish to demonstrate such directions of enzyme technology using actual examples studied and/or operated mainly in Japan.  相似文献

As originally conceived, central neurotransmitters operated uniformly, exciting or inhibiting postsynaptic targets by receptors that activated passive ionic conductances. As the list of transmitter substances and their actions expanded, concepts of transmitter actions have broadened and grown more complex to include a variety of intramembranous and intracytoplasmic second messengers that can regulate both active and passive ionic conductances. Present-day research directions center on further expansion of the lists of identified transmitter candidates, and on the more precise characterization of their sites and mechanisms of receptor regulation and transduction. Current research is also illuminating the means by which neurotransmitters act in a coordinated fashion to regulate common synaptic targets. Future directions will likely include new forms of interneuronal, intraneuronal, and glial signals, including lipids, steroids, and as-yet-undiscovered superfamilies of peptides and receptors. Although recent advances in understanding specific transmitters have been achieved largely through in vitro electrophysiological analyses, it is hoped that future research will recast these events in the context of the intact functioning brain. Neurotransmitters are likely to remain a productive focus of future research.  相似文献

Citrus is one of the most important commercial and nutritional fruit crops in the world, hence it needs to be improved to cater to the diverse needs of consumers and crop breeders. Genetic manipulation through conventional techniques in this genus is invariably a difficult task for plant breeders as it poses various biological limitations comprising long juvenile period, high heterozygosity, sexual incompatibility, nucellar polyembryony and large plant size that greatly hinder cultivar improvement. Hence, several attempts were made to improve Citrus sps. by using various in vitro techniques. Citrus sps are widely known for their recalcitrance to transformation and subsequent rooting, but constant research has led to the establishment of improved protocols to ensure the production of uniformly transformed plants, albeit with relatively low efficiency, depending upon the genotype. Genetic modification through Agrobacterium-mediated transformation has emerged as an important tool for introducing agronomically important genes into Citrus sps. Somatic hybridization has been applied to overcome self and cross-incompatibility barriers and generated inter-specific and inter-generic hybrids. Encouraging results have been achieved through transgenics for resistance against viruses and bacteria, thereby augmenting the yield and quality of the fruit. Now, when major transformation and regeneration protocols have sufficiently been standardized for important cultivars, ongoing citrus research focuses mainly on incorporating such genes in citrus genotypes that can combat different biotic and abiotic stresses. 性系统是被子植物繁育系统的核心, 决定着植物种群的遗传特征、进化方向与速度, 在种群动态、群落结构及生态系统的构建与维持中具有重要意义。本文回顾了被子植物性系统的发展历程及研究方向, 总结了近30年基于性系统研究的前沿科学问题, 包括性系统的多样性和进化、与其他功能性状的生态关联、沿环境梯度的分布格局及变化规律、与群落物种共存机制和群落动态的关系及其对干扰的响应。尽管有关植物性系统的研究已经延伸到生态学领域的诸多方面, 有力地推动了各方面的发展, 但仍有很多值得关注和需要着重研究的方向和问题。本文对未来基于植物性系统的研究方向等提出了展望, 并指出, 在当前全球气候变化背景下, 性系统可作为重要的功能性状应用于指导生物多样性保护和生态系统管理政策的制定。  相似文献

Endocrine disruptors: present issues, future directions   总被引:12,自引:0,他引:12  
A variety of natural products and synthetic chemicals, known collectively as endocrine-disrupting compounds (EDCs), mimic or interfere with the mechanisms that govern vertebrate reproductive development and function. At present, research has focused on (i) the morphological and functional consequences of EDCs; (ii) identifying and determining the relative potencies of synthetic and steroidal compounds that have endocrine-disrupting effects; (iii) the mechanism of action of EDCs at the molecular level; and (iv) the recognition that in "real life," contamination usually reflects mixtures of EDCs. Future research must examine (i) the interactive nature of EDCs, particularly whether the threshold concept as developed in traditional toxicological research applies to these chemicals; (ii) when and how EDCs act at the physiological level, particularly how they may organize the neural substrates of reproductive physiology and behavior; (iii) the various effects these compounds have on different species, individuals, and even tissues; and (iv) how adaptations may evolve in natural populations with continued exposure to EDCs. Several predictions are offered that reflect these new perspectives. Specifically, (i) the threshold assumption will be found not to apply to EDCs because they mimic the actions of endogenous molecules (e.g., estrogen) critical to development; hence, the threshold is automatically exceeded with exposure. (ii) Behavior can compound and magnify the effects of EDCs over successive generations; that is, bioaccumulated EDCs inherited from the mother not only influence the morphological and physiological development of the offspring but also the offsprings' reproductive behavior as adults. This adult behavior, in turn, can have further consequences on the sexual development of their own young. (iii) The sensitivity of a species or an individual to a compound is related to species (individual)-typical concentrations of circulating gonadal steroid hormones. Related to this is the recent finding that alternate forms of the putative receptors are differentially distributed, thereby contributing to the different effects that have been observed. (iv) Except in extraordinary situations, populations often continue to exist in contaminated sites. One possible explanation for this observation that needs to be considered is that animals can rapidly adapt to the nature and level of contamination in their environment. It is unlikely that successive generations coincidentally become insensitive to gonadal steroid hormones fundamentally important as biological regulators of development and reproduction. Rather, adaptive alterations in the genes that encode steroid receptors may occur with chronic exposure to EDCs, allowing the sex hormone receptor to discriminate natural steroids from EDCs.  相似文献

A state-of-the-art report is presented on recent progress in selected areas of myodynamics, but also on problems that severely hamper the further development of the discipline. Significant advances have been made in elucidating the force-producing interaction between actin and the myosin-S1-subunit, including the localization of the most probable molecular site of power stroke initiation. Concerning the architecture of the myostructures, strong experimental evidence has accumulated for numerous intra-, inter-, and extramuscular pathways for lateral force transmission in addition to the serial sarcomere-to-sarcomere myotendinous path.

It is shown that contemporary muscle models are inadequate in most respects and lag far behind the requirements an appropriate myodynamic model should fulfil. A similar comment applies to the current approaches designed to solve the myoskeletal indeterminacy problem. These formulations neglect myodynamic properties and do not allow for the implementation of biologically realistic objective functions. The solutions currently obtained are highly unsatisfactory. New research directions to rectify these situations are suggested, also with regard to the identification of subject-specific myodynamic parameters.  相似文献

Many adhesion and signaling molecules critical for development, as well as surface markers implicated in diseases ranging from cancer to influenza, contain oligosaccharides that modify their functions. Inside a cell, complex glycosylation pathways assemble these oligosaccharides and attach them to proteins and lipids as they traffic to the cell surface. Until recently, practical technologies to manipulate glycosylation have lagged unlike the molecular biologic and genetic methods available to intervene in nucleic acid and protein biochemistry; now, metabolic oligosaccharide engineering shows promise for manipulating glycosylation. In this methodology, exogenously-supplied non-natural sugars intercept biosynthetic pathways and exploit the remarkable ability of many of the enzymes involved in glycosylation to process metabolites with slightly altered chemical structures. To date, non-natural forms of sialic acid, GalNAc, GlcNAc, and fucose have been incorporated into glycoconjugates that appear on the cell surface; in addition O-GlcNAc protein modification involved in intracellular signaling has been tagged with modified forms of this sugar. Reactive functional groups, including ketones, azides, and thiols, have been incorporated into glycoconjugates and thereby provide chemical 'tags' that can be used for diverse purposes ranging from drug delivery to new modes of carbohydrate-based cell adhesion that can be used to control stem cell destiny. Finally, strategies for further engineering non-natural sugars to improve their pharmacological properties and provide complementary biological activities, such as addition of short chain fatty acids, are discussed in this article.  相似文献

Biotic resistance, the ability of communities to resist exotic invasions, has long attracted interest in the research and management communities. However, inconsistencies exist in various biotic resistance studies and less is known about the current status and knowledge gaps of biotic resistance in forest ecosystems. In this paper, we provide a brief review of the history and mechanisms of the biotic resistance hypothesis, and summarize the central topics and knowledge gaps related to biotic resistance with a special emphasis on forest ecosystems. Overall, although the amount of research efforts on biotic resistance in forest ecosystems has increased since the mid-2000s, aspects such as resistance to exotic pests and pathogens remain understudied. In addition, we synthesize ecological and statistical explanations of observed inconsistencies and provide suggestions for future research directions. Some of the observed inconsistencies on biotic resistance can be attributed to (1) the interactive or additive effects of other ecological processes and (2) the statistical artifacts of modifiable areal unit problem. With the advancement of new statistical knowledge and tools, along with availability of big data, biotic resistance research can be greatly improved with the simultaneous consideration of key ecological processes, the attention to various scales involved, and the addition of understudied systems.  相似文献

There is a large body of research reporting high rates of psychotic disorders among many migrant and minority ethnic groups, particularly in Northern Europe. In the context of increasing migration and consequent cultural diversity in many places worldwide, these findings are a major social and public health concern. In this paper, we take stock of the current state of the art, reviewing evidence on variations in rates of psychoses and putative explanations, including relevant theories and models. We discuss in particular: a) the wide variation in reported rates of psychotic disorders by ethnic group, and b) the evidence implicating social risks to explain this variation, at ecological and individual levels. We go on to set out our proposed socio‐developmental model, that posits greater exposure to systemic social risks over the life course, particularly those involving threat, hostility and violence, to explain high rates of psychoses in some migrant and minority ethnic groups. Based on this analysis, the challenge of addressing this social and public health issue needs to be met at multiple levels, including social policy, community initiatives, and mental health service reform.  相似文献

This theme issue has highlighted the links between sociality, health and fitness in a broad range of organisms, and with approaches that include field and captive studies of animals, comparative and meta-analyses, theoretical modelling and clinical and psychological studies of humans. In this concluding chapter, we synthesize the results of these diverse studies into some of the key concepts discussed in this issue, focusing on risks of infectious disease through social contact, the effects of competition in groups on susceptibility to disease, and the integration of sociality into research on life-history trade-offs. Interestingly, the studies in this issue both support pre-existing hypotheses, and in other ways challenge those hypotheses. We focus on unexpected results, including a lack of association between ectoparasites and fitness and weak results from a meta-analysis of the links between dominance rank and immune function, and place these results in a broader context. We also review relevant topics that were not covered fully in this theme issue, including self-medication and sickness behaviours, society-level defences against infectious disease, sexual selection, evolutionary medicine, implications for conservation biology and selective pressures on parasite traits. We conclude by identifying general open questions to stimulate and guide future research on the links between sociality, health and fitness.  相似文献
