Electroantennogram (EAG) responses were recorded from apterous virginoparae of the vetch aphidMegoura viciae Buckton (Homoptera, Aphididae) to more than eighty volatile compounds in order to investigate its sensory ability to perceive plant odours. The response profile ofM. viciae reveals a differential sensitivity for the array of plant volatiles tested. The whole group of general green leaf volatiles is very stimulatory. In addition to (E)-2-hexenal, the following compounds of this group elicit large EAG responses: (E)-2-heptenal, 1-octenol-3, hexyl acetate, (Z)-3-hexenyl acetate, hexanol-1, hexanal, 2-heptanone and 3-octanone. Relatively large EAGs are also produced by 4-methoxybenzaldehyde (p-anisaldehyde), hexanonitrile, heptanonitrile, 1,6-hexanedithiol, butyl isothiocyanate, 4-pentenyl isothiocyanate, (−)-(1S)-β-pinene, (+)-(S)-carvone, (−)-(R)-carvone, α-terpineol, linalool and citronellal. The nitriles are the most effective of all plant volatiles tested. Structure-activity relationships occur in various groups of chemicals and members of the green leaf volatiles, benzaldehydes, isothiocyanates and monoterpenes are ranked accordingly. In the group of green leaf volatiles, aliphatic aldehydes are more stimulating than the corresponding alcohols. EAG responses to series of saturated aliphatic alcohols and aldehydes reveal that C6 and C7 compounds are the most stimulatory. Dose-response curves show that the rank order of EAG response amplitudes hardly changes at lower dosages. It is concluded thatM. viciae perceives general plant volatiles as well as more-specific components, such as nitriles and isothiocyanates, associated with the odour blends of non-host plant species.

Abstract A Y-tube olfactometer and a still-air olfactometer were developed to determine the attractiveness of several host plants for the vine weevil ( Otiorhynchus sulcatus (F.); Coleoptera: Curculionidae). Odours of weevil-damaged yew ( Taxus baccata ) and spindle trees ( Euonymus fortunei ) are attractive to the vine weevil, but Rhododendron and strawberry ( Fragaria  ×  ananassa ) are not. Undamaged Euonymus is attractive to the weevils in springtime but not in late summer. When clean air or undamaged Euonymus is the alternative, weevils strongly prefer weevil-damaged Euonymus foliage, and this preference is retained throughout the year. Hence, plant damage plays a role in attraction of the vine weevil. In contrast to the permanent attractiveness of weevil-damaged Euonymus , mechanically damaged plants gradually lose the attractiveness that they have early in the growing season. This suggests that emission of volatiles, produced by the plants in response to weevil damage, is important for attraction of the weevils because the weevils may use these plant odours to find suitable food plants throughout the season. Apart from weevil-damage-related plant volatiles, green leaf volatiles must also play a significant role, as indicated by the fact that weevils prefer: early season, undamaged Euonymus over clean air; early season, mechanically damaged Euonymus over undamaged Euonymus ; and, throughout the season, had no preference when mechanically damaged Euonymus is tested against weevil-damaged Euonymus . Thus, monitoring traps may be developed by the use of green leaf volatiles and/or herbivore-induced volatiles, as attractants.

Drosophila suzukii Matsumura, an endemic pest in southeast Asia, has invaded Europe and the U.S.A. Unlike most of its closely related sibling species, the serrated ovipositor of D. suzukii permits ovipositing in undamaged fresh fruits. In the present study, volatiles are identified from host plants that are potentially involved in D. suzukii host recognition and oviposition behaviour. It is shown that mated females are attracted to volatiles emitted from intact fruits. The antennally‐active suite of compounds released from the fresh fruits is identified by gas chromatography coupled with electroantennographic detection, as well as gas chromatography‐mass spectrometry. In olfactometer bioassays, mated females are significantly attracted to an electroantennographically active volatile, isoamyl acetate, when tested at 10 µg of synthetic compound in a rubber septa, which has a release rate comparable to that of fresh fruits. In addition, a genomic survey shows that D. suzukii not only possesses the full repertoire of genes encoding odorant receptors activated by isoamyl acetate in D. melanogaster, but also that one of the genes, OR67a, is represented by five duplicated copies. These results indicate that D. suzukii uses olfactory cues to select oviposition sites. The identification of volatiles emitted by host fruits that attract D. suzukii may aid in the development of a selective and efficient synthetic lure for monitoring this pest. As a close relative of Drosophila melanogaster, D. suzukii provides a unique opportunity for understanding the physiological mechanisms involved in the shift of this species from use of rotten to ripe fruits for oviposition.

Heterorhabditis megidis (UK211) was applied against black vine weevil (BVW, Otiorhynchus sulcatus) in potted plants in a polyethylene (2002) or glasshouse (2003) and in field planted strawberries (2003). Both potted and field strawberries were artificially infested with BVW larvae. In a 2002 pot planting in the polyethylene house, a single drench application of 25,000 H. megidis infective juveniles per plant in 50 ml of water in mid September, reduced the number of BVW larvae to 1.8/20 plants. A second application in early October gave a reduction of 0.2/20 plants and in the third application, the following March no live weevils were recovered, compared to the control which had 8.2 larvae/20 plants. In a 2003 pot planting in a glasshouse, similar treatments gave a reduction of 5.2, 5.4 and 0.8 larvae/20 plants, respectively, compared to the control where 26.2 larvae/20 plants were recovered. In an artificially, BVW infested field trial, similar treatments gave a reduction to 2.2 larvae/20 plants in the single September treatment, and 2 larvae/20 plants in the single October treatment. The double (September and October) application reduced BVW larvae further to 1.6/20 plants and the triple (September, October and April) application to 0.4 larvae/20 plants, compared to the control where four larvae corresponded to every 20 plants. There was, therefore, little difference between the single and double autumn treatments indoors or in the field, and it mattered little whether the single application in the field was made in September or October under the conditions of 2003. Early spring application gave a significant reduction in BVW in each of the three experiments.

Electroantennogram (EAG) recordings from the antennae of a highly specialized phytophagous species — the oak flea weevil, Rhynchaenus quercus L. — revealed two general properties of its antennal olfactory system: (1) a high and to some extent selective sensitivity to chemotaxonomically nonspecific compounds, namely the so-called general green leaf volatiles as well as to geraniol and linalool, (2) a lack of marked differences in the general excitation level caused by stimulation with vapours from leaves of both host plants and non-host plants. It was also observed, that the shapes of EAG's evoked by vapours from cut leaves and general green leaf volatiles differed from those elicited by corresponding concentrations of geraniol and linalool, where recovery was distinctly slower. The results of EAG tests suggest that the general green leaf volatile complex is important in host-odour perception by the oak flea weevil. A possible function of these compounds in host-plant odour discrimination is discussed.

The vine weevil Otiorhynchus sulcatus is a parthenogenetic reproducing species which forages for suitable host plants at night, but is found congregated in dark places during the day. Frass of this weevil species is suspected to contain attractive compounds that are host‐plant related. Using a still‐air olfactometer, we tested adult vine weevils at night for their behavioural response to odours from conspecifics, feeding on a mixture of spindle tree (Euonymus fortunei) and yew (Taxus baccata), and to a sexually reproducing related species (Otiorhynchus salicicola), feeding on a mixture of ivy (Hedera helix) and cherry laurel (Prunus laurocerasus). Their attraction to conspecifics and O. salicicola appeared to be related to frass production. Freshly collected frass from O. sulcatus and from O. salicicola males and females was attractive. Prunus laurocerasus and H. helix have not been observed to be hosts of the vine weevil in the field. However, our tests showed that the vine weevil was attracted to mechanically damaged leaves of both plant species, whereas undamaged leaves were not attractive. Only undamaged young unfolding leaves of H. helix were also attractive. The attraction to odours from mechanically damaged host and non‐host plants suggested the involvement of compounds that are commonly found in many plant species. The involvement of plant compounds and/or aggregation pheromones in attraction to frass of the vine weevil and frass of the related weevil species O. salicicola is discussed.

Plants release hundreds of volatiles that are important in the interaction with herbivorous animals, but which odorants are detected by which species? In this study, single receptor neurons on the antenna of the oligophagous strawberry blossom weevil Anthonomus rubi were screened for sensitivity to naturally produced plant compounds by the use of gas chromatography linked to electrophysiological recordings from single cells. The narrow tuning of the neurons was demonstrated by responses solely to a few structurally related sesquiterpenes, aromatics or monoterpene hydrocarbons out of hundreds of plant constituents tested. We present five olfactory receptor neuron types, identified according to one primary odorant i.e. the compound to which the neurons are most sensitive. These odorants, (-)-germacrene D, (-)-beta-caryophyllene, methyl salicylate, E-beta-ocimene and (3E)-4,8-dimethyl-1,3,7-nonatriene, present in the intact strawberry plant, are induced in higher amounts by weevil feeding. This suggests that these compounds can provide information about the presence of conspecifics. We used protocols especially designed to allow comparison with previously investigated species. Striking similarities, but also differences, are demonstrated between receptor neuron specificity in the strawberry weevil and moths.

The effect of five commercial potting media, peat, bark, coir, and peat blended with 10% and 20% compost green waste (CGW) on the virulence of six commercially available entomopathogenic nematodes (EPN), Heterorhabditis bacteriophora UWS1, Heterorhabditis megidis, Heterorhabditis downesi, Steinernema feltiae, Steinernema carpocapsae, and Steinernema kraussei was tested against third-instar black vine weevil (BVW), Otiorhynchus sulcatus. Media type was shown to significantly affect EPN virulence. Heterorhabditis species caused 100% larval mortality in all media whereas Steinernema species caused 100% larval mortality only in the peat blended with 20% CGW. A later experiment investigated the effect of potting media on the virulence of EPN species against BVW by comparing the vertical dispersal of EPN in the presence and absence of BVW larva. Media type significantly influenced EPN dispersal. Dispersal of H. bacteriophora was higher than H. megidis, H. downesi, or S. kraussei in all media, whereas, S. feltiae and S. carpocapsae dispersal was much reduced and restricted to peat blended with 20% CGW and coir, respectively. In the absence of larvae, most of the EPN species remained in the same segment they were applied in, suggesting that the larvae responded to host volatile cues. Greenhouse trials were conducted to evaluate the efficacy of most virulent strain, H. bacteriophora in conditions more representative of those in the field, using 2.5 × 109 infective juveniles/ha. The efficacy of H. bacteriophora UWS1 against third-instar BVW was 100% in peat, and peat blended with 10% and 20% CGW but only 70% in bark and coir, 2 weeks after application. These studies suggest that potting media significantly affects the efficacy and dispersal of EPN for BVW control.

Odours of female Zabrotes subfasciatus Boh, elicit strong electroantennogram (EAG) responses in their sexual partners. Virgin female insects used in experiments were of different ages and given seeds, or not, of the host plant, Phaseolus vulgaris L. The odours of females which did not get host plant seeds, do not elicit EAG responses in virgin males, until they are at least 6 days old. On the other hand, the release of the sex pheromone as perceived by the male antennae, occurs earlier in the life of females when they are in contact with the host plant's seeds. This suggests that the sexual attractiveness of the females of this specialist insect is probably closely correlated with vitellogenesis.

Action potentials from individual olfactory cells and receptor potentials were recorded from antennae of female cabbage white butterflies, Pieris brassicae, using a new ‘surface-contact’ recording technique. Stimuli used were plant volatiles and conspecific egg odours. Most cells were activated by some stimuli and inhibited by others. Hence, the ‘odour spectra’ of most cells included activating as well as inhibiting substances. In addition, the ‘response profile’ of a given stimulus contained both positively and negatively responding cells.Cluster analysis of the odour spectra revealed the existence of moderately separated clusters of olfactory receptors. No groups of cells specifically tuned to odours of eggs or cabbage leaves were found. Analysis of the response profiles did not show a clear clustering of stimuli. The results suggested that the membranes of different receptor cells contain several types of acceptor sites in various proportions. No correlation between EAG and receptor potential amplitude or spike frequency was found. It was concluded that the EAG may be useful in comparing the relative receptor sensitivities only when chemically related compounds are involved.

Olfactory responses of two species of grasshoppers to plant odours
Electroantennogram (EAG) responses were recorded from two species of oedipodine grasshoppers, Oedaleus decorus asiaticus L. (graminivorous) and Angaracris barabensis Pall. (forbivorous), to volatiles emitted by chopped leaves of ten plant species. Male O. d. asiaticus showed much stronger EAG responses than conspecific females and both sexes of A. barabensis. Sexual differences in EAG responses correspond to different numbers of antennal sensilla of both sexes and to certain behavioural and morphological factors as well. The overall EAG response profiles of the two grasshopper species to the ten plant odours were similar. However, adaptation to host odour might have occurred because of differences in their feeding habits. Females of the graminivorous O. d. asiaticus possess a significantly higher olfactory sensitivity for poaceous plant species than A. barabensis, while the forbivorous A. barabensis showed significantly higher EAG responses to Allium senescens (Liliaceae), and a tendency of high responses to composite plant species in comparison with O. d. asiaticus.

Chemical trails of the hosts (footprints) are important cues for the host searching behaviour of egg parasitoids of the family Scelionidae. The present study aims to determine the influence of the footprints of three neotropical stink bugs (Euchistus heros, Dichelops melacanthus and Nezara viridula) on the foraging behaviour of two parasitoids, Trissolcus basalis and Telenomus podisi, as well as a possible selective response to fooprints of their preferred hosts. Accordingly, Tr. basalis and Te. podisi females are allowed to forage on open arenas where E. heros, D. melacanthus or N. viridula had walked or on open arenas that had been treated with samples of an extract from each stink bug's footprints. Hexane extracts of stink bug footprints are obtained from solvent-washed Petri dishes where insects were allowed to walk for 24 h, and these extracts were analyzed by gas chromatography-flame ionization detection and gas chromatography-mass spectrometry. Each parasitoid responds selectively to the footprints of their preferred host (Tr. basalis to footprints of N. viridula and Te. podisi to footprints of E. heros). Twenty-six compounds comprising of C18 to C35 (saturated and unsaturated) and methyl branched hydrocarbons were identified in extracts of E. heros, D. melacanthus and N. viridula, respectively. There are significant differences in the total amount of the compounds identified in the footprint stink bug's extracts and also a difference in the amounts of individual compounds. In addition, the behavioural assays showed that footprints of stink bugs are stimuli that are used by egg parasitoids to search, discriminate and selectively locate their preferred host.

Abstract 1 The relationship between reproductive performance and preference for potential host plants of the vine weevil is investigated, as shown in tests on contact (or feeding) preference, presented herein, and tests on olfactory preference, published elsewhere. 2 Assessment of reproductive performance shows that the host‐plant range of the adult vine weevil Otiorhynchus sulcatus in Europe is limited to one gymnosperm genus (Taxus sp.) and a broad range of angiosperm plants in two subclasses of the Dicotyledonae, namely Dilleniidae and Rosidae. The successful reproduction on very distantly related plant taxa suggests that the original weevil‐ and plant‐habitat has mediated the current host‐plant range of the vine weevil. 3 Contact‐preference tests with equally suitable hosts, such as Aronia, Fragaria, Euonymus and Taxus, and one less suitable host, Humulus, indicate a mismatch between contact preference and performance and, as far as olfactory preferences are known, these match neither the contact preferences nor the performance. This mismatch may arise because (i) host plant species offered do not occur in weevil habitat in Europe (e.g. Aronia and the cultivated Fragaria come from North America) and (ii) predation (or disease) risks differ among host plants, thereby altering effective reproductive performance. 4 With respect to performance on novel hosts (Thuja, Prunus) and bad hosts (Rhododendron), some between‐individual variation is found within a single population, suggesting that local populations harbour (possibly genetic) variation for adaptation to new hosts. How this variation is maintained in the face of strong selection pressures on local populations of flightless and thelytokous weevils, is an important question for understanding the broad host plant range in the vine weevil.

Abstract:  The weevil Scyphophorus acupunctatus Gyllenhal (Col., Curculionidae) is the most important insect pest of cultivated agaves in Mexico. The objective of this study was to search for potential attractants for this weevil species from the freshly cut fleshy leaves of healthy Agave tequilana Weber var. Blue plants. Combined gas chromatography-electroantennography (GC-EAD) analysis of A. tequilana volatile extracts showed that seven peaks elicited antennal responses from males and females. Five of these peaks were identified by gas chromatography-mass spectrometry (GC-MS) as o -xylene, α -pinene, 3-carene, γ -terpinene and linalool. Electroantennogram (EAG) records of both sexes to 1-, 10- and 100- μ g-stimulus load of these five compounds showed that there was a dose effect on the EAG responses for linalool, 3-carene and α -pinene, but not for γ -terpinene and o -xylene. There were no sexual differences between male and female EAG responses to any of the compounds tested. Y-tube olfactometer bioassays of the compounds showed that males and females were attracted to α -pinene, 3-carene, γ -terpinene and/or linalool at doses of 1 and 10  μ g, while weevils were repelled by linalool at a dose of 100  μ g.

Electroantennogram responses of aphid nymphs to plant volatiles
Abstract. Electroantennogram (EAG) responses of immature aphids were investigated for 30 plant volatile compounds in third‐ and fourth‐stadium nymphs of the black bean aphid, Aphis fabae. The nymphs were destined to develop into adult alate (winged) virginoparae. The EAG response profiles were similar to those previously reported in the adults. Among the compounds tested, hexanonitrile elicited the largest EAG responses in both nymphal stadia, corresponding to previously reported results with adults. Six‐carbon aliphatic compounds showed relatively higher EAG activities in the nymphs but, in contrast, (E)‐2‐hexenal, benzaldehyde, α‐pinene and β‐pinene, and citronellal elicited relatively smaller EAG responses in nymphs than adults. Although overall EAG response profiles were similar between the third and the fourth stadia for the majority of the volatiles, four aldehyde compounds, hexanal (E)‐2‐heptenal, 2‐hydroxybenzaldehyde and citronellal, showed relatively higher EAG activities in the third than in the fourth stadium. The present study indicates that aphid nymphs possess a functional olfactory receptor system before the antennae are fully developed morphologically and physiologically.

The sexual and host‐related behaviours of the fruit fly Anastrepha obliqua Macquart (Diptera: Tephritidae) are mediated by volatile compounds. However, whether the physiological state of this species affects its antennal and behavioural responses to semiochemicals is unknown. The effects of age, mating status, diet and the topical application of methoprene, a Juvenile hormone analogue (JHA), on the antennal sensitivity of this tephritid fruit fly species to selected male [(Z)‐3‐nonenol] and host fruit volatiles (ethyl benzoate, ethyl hexanoate, ethyl butyrate and trans‐β‐ocimene) are investigated using electroantennography (EAG). Overall, (Z)‐3‐nonenol and ethyl benzoate elicit the highest EAG responses in both sexes. Flies of both sexes aged 1, 5 and 10 days old show higher EAG responses to the tested compounds compared with flies aged 20 days old. Virgin females and males show higher EAG responses to volatile compounds than mated flies. Females and males fed with sugar plus protein show higher antennal responses to volatiles compared with flies fed sugar or protein alone. Flies of both sexes treated with methoprene show higher antennal responses than flies treated with acetone (control). These results suggest that the peripheral olfactory system in A. obliqua is modulated by the physiological state of the flies.
