In 1971 the Grevelingen estuary was embanked. In the newly created lake Grevelingen the tidal movements stopped and a few thousand ha of sandflats fell permanently dry. Ca 40% of the surface of those flats was immediately afterwards sown with rye and other grasses to prevent wind erosion. This fixation of a rather uniform environment resulted in a monotonous vegetation cover. Grazing with domestic animals is now applied as management practice to create more environmental variation and thereby a higher species diversity.The present study gives the results of eleven years of comparing vegetation development under various management practices, including non-interference in the spontaneous and sown vegetation. Sequential vegetation mapping, repeated inventories of selected areas and studies in permannent plots are the main techniques used.On the unsown shore zones interesting vegetation types are developing where species diversity is higher than in the sown areas. In the shore zones not only a faster succession occurred compared with the sown areas, but also a shifting of environmental gradients, e.g. in moisture and salinity conditions, encouraged vegetation changes. After 10 yr grazed areas had a higher number of species than ungrazed equivalent areas. The results also indicated that grazing slows down the establishment of (tall) woody species and shrub development.Nomenclature of angiosperms and syntaxa follows Heukels & van Ooststroom (1977) and Westhoff & den Held (1975), respectively.The authors are indebted to Dr W. Data are presented to evaluate further the abundance/biomass comparison method for the detection of pollution disturbance in marine benthic communities. The results suggest that this technique cannot be used in estuarine intertidal communities. The grossly polluted configurations appeared under unpolluted conditions, while undisturbed patterns were found under polluted conditions. Summary We have determined the molecular characteristics of -thalassemia in 12 HbH subjects from Taiwan by restriction endonuclease mapping with -and -specific probes. We have found four types of defects in the -thalassemia-2 genetic determinant: 3.7 type I; 4.2; CS; and T. All HbH subjects carried the ——SEA genotype in the -thalassemia-1 determinant. At least two different subtypes of ——SEA genotype were observed in this study.  相似文献

Morniga M is a jacalin-related and mannose-specific lectin isolated from the bark of the mulberry (Morus nigra). In order to understand the function and application of this novel lectin, the binding property of Morniga M was studied in detail using an enzyme-linked lectinosorbent assay and lectin-glycan inhibition assay with extended glycan/ligand collection. From the results, it was found that the di-, tri-, and oligomannosyl structural units of N-glycans such as those of the bovine 1-acid glycoprotein (gp) and lactoferrin were the most active gps, but not the O-glycans or polysaccharides including mannan from yeast. The binding affinity of Morniga M for ligands can be ranked in decreasing order as follows: gps carrying multiple N-glycans with oligomannosyl residues >> N-glycopeptide with a single trimannosyl core > Tri-Man oligomer [Man1 6(Man 1 3) Man], Penta-Man oligomer [Man1 6(Man1 3)Man1 6(Man1 3) Man] Man 1 2, 3 or 6 Man > Man > GlcNAc, Glc >> L-Fuc, Gal, GalNAc (inactive), demonstrating the unique specificity of this lectin that may not only assist in our understanding of cell surface carbohydrate ligand-lectin recognition, but also provide informative guidelines for the application of this structural probe in biotechnological and clinical regimens, especially in the detection and purification of N-linked glycans.  相似文献

The inside-out vesicles of plasma membranes were isolated from pumpkin stem cells, and the kinetics of sucrose efflux induced by the K+-diffusion potential (D) was studied by measuring light transmission. Two phases differing in their rates and duration were identified in D-dependent changes of light transmission. The increase in Delevated the rate and magnitude of the fast phase in light transmission changes but did not markedly affect the rate of the slow phase. These two phases also differed in their sensitivity to inhibitors and to changes in sucrose concentration in the external medium. Measurements of Dduring sucrose transport by means of the fluorescence probe dis-C3-(5) revealed differences in the magnitude of Dand its stability in vesicles loaded with sucrose and mannitol, as well as under the action of inhibitors. The two-phase dependence of sucrose efflux from vesicles on the applied diffusion potential is discussed in the context of modern concepts on the functioning of sucrose carriers in the membranes.  相似文献

Primary structure of -chain of pigeon is presented. It was determined by amino acid sequence analysis of intact -chain and its peptides obtained by the enzymatic and chemical cleavage. Comparison of amino acid sequence of the chain with other available data shows 14 Ile, 61 Lys, and 113 Ile as residues specific to pigeon. Cationic derivatives of deoxycholic acid with N,N-dimethylenediamine, -aminocaproic acid, and pyridine as polar heads were synthesized. The cationic groups were linked to 3- and 12-hydroxy groups of the steroid moiety through ester or urethane bonds. Liposomal formulations of the compounds synthesized may be used for gene delivery in cells.  相似文献

EnteropathogenicEscherichia coli (EPEC), first described in the 1940's and 1950's, remain an important cause of severe infantile diarrhoea in many parts of the developing world. EPEC do not produce enterotoxins and are not invasive; instead their virulence depends upon exploitation of host cell signalling pathways and the host cell cytoskeleton both as a means of colonizing mucosal surfaces of the small intestine and causing diarrhoea. Following initial mucosal attachment, EPEC secrete signalling proteins and expresss a surface adhesin, intimin, to produce attaching & effacing lesions in the enterocyte brush border membrane characterised by localised destruction of brush border microvilli, intimate bacterial adhesion and cytoskeletal reorganisation and accretion beneath attached bacteria. The pathophysiology of EPEC diarrhoea is also complex and probably results from a combination of epithelial cell responses including both electrolyte secretion and structural damage.  相似文献

Crossing experiments were carried out between the stiped-egg form collected from Sapporo (43°04N) and Machida (35°33N), and the stipeless-egg form ofPanonychus akitanus Ehara from Tomakomai (42°37N) and Imagane (42°34N). Intra-form crosses gave a high proportion of female progeny, but inter-form crosses only male progeny, suggesting that fertilization did not take place. The number of eggs laid during the first 10 days of the oviposition period was greater in intra-form crosses than in inter-form crosses (P<0.001). The latter, except for the cross between the Sapporo and the Imagane populations, had similar egg production to virgin females (P<0.05).Mated females showed a greater daily oviposition rate than virgin females. However, the latter drastically increased their oviposition rate after mating with their sons. The two forms are thus reproductively isolated.  相似文献

Summary This study concerns the characterization of chromosomes with hybrid genes for Hb Lepore-Washington (44 chromosomes), for Hb Lepore-Baltimore (5 chromosomes), for Hb P-Nilotic (8 chromosomes), and for Hb Kenya (7 chromosomes) by determining a relatively large number of restriction enzyme polymorphism. Two, and possibly three, different Hb Lepore-Washington chromosomes were identified by specific haplotypes, while the haplotype of the Hb Lepore-Baltimore chromosome had its own characteristic pattern. A likely conclusion is that the crossovers leading to the formation of these chromosomes have occurred as independent events within the populations. Chromosomes with the -Lepore-Washington hybrid gene maintained specific characteristies (such as increased Hb F levels in heterozygotes, and high or low G values in this Hb F) which have been observed in normal individuals with chromosomes having comparable haplotypes. Only one haplotype was observed for each of the chromosomes carrying either the -P-Nilotic hybrid gene or the A hybrid gene of Hb Kenya.  相似文献

Summary The flux from the biomedical channel at TRIUMF increases with increasing channel momentum, while the contaminating electron flux decreases. Since the electrons appear to result from conversion of the high energy-rays produced by 0 decay in the production target, the electron contamination can be reduced further by target configurations which minimize gamma conversion.The attenuation of beams by in-flight interactions was found to decrease from an initial value of 1.67 ± 0.02 % per g/cm2 at zero depth to 1.48 ± 0.02 % per g/cm2 near the stopping peak.The inactivation of cultured CHO cells by an extended-peak dose distribution has been measured using the gel technique. The survival data have been fitted by a model which characterizes the physical quality of the dose profile by means of measured star densities. This model provides a convenient method of analysis for large sets of survival data and may be useful for prediction of the biological effect of new dose distributions.The RBE value for 50% survival measured at the centre of a 7 cm extended peak was found to be approximately 1.4, in reasonable agreement with recent values obtained at LAMPF and SIN.  相似文献

Detailed karyological surveys of the ant Myrmecia pilosula species group, which is characterized by the lowest chromosome number in higher organisms (2n=2), were attempted. We revealed that this species has developed highly complicated chromosomal polymorphisms. Their chromosome numbers are in the range 2n=2, 3, and 4, and six polymorphic chromosomes are involved, i.e., two for chromosome 1 (denoted as SM1 and ST1), three for chromosome 2 (A2, A2, and M2), and M(1+2) for the 2n=2 karyotype. We suggested that these chromosomes were induced from a pseudo-acrocentric (A 1 M ) and A2 as follows: (1) A 1 M SM1 or ST1 by two independent pericentric inversions; (2) A2A2M2 by chromosomal gap insertion and centromere shift; and (3) ST1+A2M(1+2) by telomere fusion, where (3) means that the 2n=2 karyotype was derived secondarily from a 2n=4 karyotype. It is a noteworthy finding that active nucleolus organizer (NOR) sites, in terms of silver staining, are tightly linked with the centromere in this species, and that both the centromere and NOR of A2 were inactivated after the telomere fusion.  相似文献

Both freely suspended cells and immobilized cultures of Spirulina platensis, a blue-green alga, biotransformed exogenously fed codeine, an opium alkaloid, to morphine. The external addition of codeine to the culture medium did not affect the growth of S. platensis. Immobilization of Spirulina in a calcium alginate gel matrix was optimized by using 2% (w/v) sodium alginate and reducing the concentration of nutrients of Zarrouk's medium, which caused destabilization of the calcium alginate gel. The accumulation of morphine increased gradually and reached maxima of 330g 100ml–1 culture at 105h in freely suspended and 351g 100ml–1 at 96h in immobilized Spirulina cultures. Accumulation of morphine was detected only in the medium, whereas cells did not show accumulation. The immobilized Spirulina cultures showed marginally higher conversion of codeine to morphine over freely suspended cultures.  相似文献

Summary 3-Aminoglycoside phosphotransferase [APH(3)] enzymes are a group responsible for resistance to the antibiotics kanamycin (Km) and neomycin (Nm) in bacteria. Escherichia coli ECT24, originally isolated from a meat sample, harboured an 83-kb conjugative R-plasmid (pRPJ24) that carries transferable resistance to Km and Nm. Plasmid pRPJ24 was transferred by conjugation to Enterobacter cloacae 94R, which was used as the source of plasmid DNA in development of a probe for the Km-resistance determinant. Random cloning of BamHI and HindIII double-digest restriction fragments of pRPJ24 in the pUC18 vector plasmid produced clones resistant to both Nm and Km carrying a 1.9-kb DNA insert. Southern hybridization of pRPJ24 cloned chimeric plasmid DNA (pKPJ94) showed homology with the APH(3)II gene from transposon Tn5. A PstI digest of pKPJ94 produced a 920-bp fragment which hybridized with the APH(3)II structural gene, and was used as a DNA probe for the APH(3)II subclass gene. A 980-bp BamHI fragment from plasmid pGH54 carrying the APH(3)I gene from transposon Tn903 was used as a subclass I probe. Total DNA from 206 randomly screened Km-resistant Enterobacteriaceae isolates from raw ground beef and chicken meat samples were examined for the occurrence of APH(3) subclass I and II using non-radioactively-labelled DNA probes. Thirty-six percent and 60% of the isolates examined carried subclass I and II resistances, respectively, in the isolates from chicken meat samples. The corresponding values for bacterial strains from raw ground beef samples were 51% and 72%, respectively. Four percent of the resistant bacterial isolates from chicken samples did not display homology to either probe. This value was 28% for bacterial isolates from ground beef. Species-rich floating fen ecosystems in former turf ponds in the western part of The Netherlands are subject to nitrogen enrichment because of high atmospheric N deposition (50 kg ha–1,Y–1,). and supply of polluted river water in dry summer periods. Further, some fens have become more influenced by rain water because downward seepage to the groundwater has increased due to hydrological alterations. This paper describes changes in plant biomass production by comparing seasonal maximum biomass values for 15 fen sites determined with standard procedures in 1981 and 1988. Fen sites in different polders showed different species composition, which is related to differences in hydrology and history of fen management among the polders. The mid-succession fens (type 1) which are characteristically N-limited have shown a biomass increase in spite of the annual mowing regime, which shows that these fens are becoming enriched with nitrogen. There are indications that the role of phosphorus as a limiting factor increases in these fens, and that a shift of N-limited towards P-limited phanerogam growth occurs. This may bring these fens eventually in the late-succession stage, as presently found in Het Hol (type 2). The fens in this stage are P-limited and have a different species composition. It was concluded that the mesotrophic fens in the Vechtplassen area, characterized by a species-rich vegetation, can only persist in their eutrophicated environment if they are located in a groundwater discharge area and if they are annually harvested in the summer. If all fens in the area, will eventually become P-limited it is expected that the species composition will change to a more uniform late-succession vegetation type.  相似文献

Recent research has shown that extrafloral nectar secretion by plants, which also attracts ants, is a defense inducible by herbivory damage. Our research addresses the question of whether plants can manipulate the honeydew secretions of homopterans as an inducible defense in response to herbivory in the same manner as extrafloral nectaries. We investigated changes in honeydew composition and excretion rate by the facultatively ant-attended aphid Chaitophorus saliniger Shinji (Homoptera: Aphididae) depending on the presence or absence of herbivory by caterpillars (Clostera anastomosis L.; Lepidoptera: Notodontidae) on their host plant, the willow Salix gilgiana Seemen. Our results found no evidence to suggest that herbivory damage to the aphids host plant causes significant changes in aphid population growth, honeydew droplet volume, volume of honeydew excreted per aphid per hour, or composition of three abundant sugars in aphid honeydew as a result of herbivory damage to the aphids host plant, suggesting that, in this particular system, the plant is not manipulating the aphids honeydew output for its own benefit.  相似文献

Summary An immunoelectronmicroscopic method for the specific localization of neurohypophyseal hormones was developed in neurohypophyses of Wistar and Brattleboro rats, the latter strain being homozygous for diabetes insipidus. If the proper precautions were omitted, a marked cross reactivity between antivasopressin and antioxytocin preparations was found. Cross reaction of an antivasopressin plasma with oxytocin, at a dilution of less than 11600, resulted in electron density of all granules within neurosecretory fibres of the Brattleboro and Wistar neurohypophyses. However, this cross reactivity could be eliminated either by sufficient dilution of the antiplasma, or by its purification. Purification of the antibodies was performed by absorption to agarose beads coated with the cross reacting component. Upon incubation with antivasopressin (diluted unpurified 11600 or purified 180) and unpurified antioxytocin (1400) plasma, sections of a Wistar neurohypophysis revealed two types of neurosecretory fibres, containing either electron dense or lucent granules. Oxytocin and vasopressin containing neurosecretory fibres were found as clusters in the neurohypophysis. The specificity of both unpurified antivasopressin (11600) and antioxytocin (1400) plasma was confirmed on serial sections of a Wistar neurohypophysis, alternately incubated with the solutions of the two antibodies.These data prove that the one-cell-one-hormone hypothesis holds true for the hypothalamic-neurohypophyseal system.The authors wish to thank Dr. L.A. Sponge secondary metabolites have been investigated to find potential lead compounds for the development of commercially interesting products. During the Camellia project entitled Environmentally compatible antifouling coatings for the protection of ships, water systems, fish cages and other immersed structures against aquatic growth, several analogues of terpenes containing isocyano, thioisocyano, thiocyano and formamide functionalities were synthesized and evaluated for their antifouling activity against the settlement of the barnacle Balanus amphitrite. The best activities were obtained with N-formylated alkylamines the length of the carbon chain of which is comprised between C-11 and C-14. In the aromatic series, the isocyano derivatives showed high antifouling activities. But they were too toxic against many microorganisms to be incorporated in paint formulations. During the Symbiosponge project entitled Biology of sponge natural products the methanolic extract of about 230 sponges were submitted to bioassays guided fractionation to isolate bioactive compounds. Several cytostatic sesquiterpene quinones and tryptamine-derived alkaloids were isolated from 4 sponges of the genus Hyrtios. A new 4-sphingenine analogue and a novel polyacetylenic derivative were found to be responsible for the bioactivities of the methanolic extracts of Haliclona vansoesti and Callyspongia pseudoreticulata, respectively.  相似文献

Extraocular muscles (EOM) are typically spared in Duchenne muscular dystrophy. We hypothesized that this might be due to different patterns of utrophin expression. The expression of utrophin was examined in EOM of normal cats using immunohistochemical methods and Western blot. For detecting acetylcholine receptors (AChR), we used -bungarotoxin. Surprisingly, -bungarotoxin failed to stain the AChR and no expression of utrophin could be detected at the neuromuscular junctions. Our study could indicate that the expression of utrophin is dependent on the structure of the AChR.  相似文献
