采用单一碳源回收菌群的方法与ERIC-PCR方法相结合,检测水稻(Oryza sativa L.)根际施用转基因微生物E4(Enterobacteria cloacae E4)后,其根际微生物的群落结构和多样性的变化,进而推测转基因微生物E4在田间施用的安全性。结果表明:转基因微生物E4施用到水稻根际后,水稻根际的代谢指纹图谱和。DNA指纹图谱都发生了改变,采用Sollthelxi blotting检测显示:E4成为根际的优势菌,这对植物生长有利,应该不会造成不利的影响。  相似文献

The beneficial effects of probiotic Enterococcus spp. in different hosts, such as mice and humans, have previously been reported in several studies. However, studies of large domestic animals, as well as challenge studies with pathogenic microorganisms, are very rare. Here, we investigated the influence of oral treatment of pigs with the probiotic bacterium Enterococcus faecium NCIMB 10415 on Salmonella enterica serovar Typhimurium DT104 infections in weaning piglets. Clinical symptoms, fecal excretion, the organ distribution of Salmonella, and the humoral immune response (immunoglobulin G [IgG], IgM, and IgA levels) in serum were examined. A pool of 89 piglets was randomly divided into probiotic and control groups. The probiotic group received a feed supplement containing E. faecium starting on day 14 postpartum prior to challenge with Salmonella serovar Typhimurium DT104 at 28 days postpartum. After challenge with Salmonella serovar Typhimurium DT104, piglets in both groups showed no severe clinical signs of salmonellosis. However, fecal excretion and colonization of Salmonella in organs were significantly greater in piglets fed E. faecium. Likewise, the humoral immune response against Salmonella (serum IgM and IgA levels) was significantly greater in the probiotic group animals than in control animals. The results of this study suggest that E. faecium NCIMB 10415 treatment enhanced the course of infection in weaning piglets challenged with Salmonella serovar Typhimurium DT104. However, the probiotic treatment also appeared to result in greater production of specific antibodies against Salmonella serovar Typhimurium DT104.The problem of increasing microbial resistance to antibiotics resulting from years of overuse and the resulting ban on the use of antibiotics in animal production have led to increased interest in alternatives to antibiotics in animal production. In recent years, probiotic bacteria have been considered as an alternative means of reducing pathogen loads in animal breeding and production units. However, while a number of studies have focused on the mode of action of probiotics, the mode of action these bacteria is not fully understood yet.A recent interdisciplinary research study of the modes of action of probiotics in swine showed that Enterococcus faecium NCIMB 10415 reduced the pathogenic bacterial load of healthy piglets (20, 26, 30, 36). In vitro studies further demonstrated that this E. faecium probiotic strain decreased the rate of invasion of a porcine intestinal epithelial cell line by Salmonella enterica serovar Typhimurium. To determine whether probiotics also provide a measure of protection during infections, experimental challenge studies with pathogenic bacteria at a defined infectious dose and under comparable conditions seem to be necessary. Field studies could be more representative of the real situation; however, the infection pressure is too low and difficult to define, and systematic sampling cannot be done.Studies of larger domestic and production animals are rare. Most such studies deal with the mode of action of probiotics in the healthy host, and only a few studies have investigated the mode of action in the context of infections with pathogenic bacteria, such as Salmonella.In a related study, weaned piglets were fed a mixture of five probiotic strains (one Pediococcus strain and four Lactobacillus strains) and challenged with Salmonella serovar Typhimurium (7). In that study, reduced incidence, severity, and duration of diarrhea and a reduced microbiological load of Salmonella were observed. Fedorka-Cray et al. (11) observed reduced numbers of Salmonella bacteria in cecal contents and at the ileocolic junction in S. enterica serovar Choleraesuis-challenged weaning piglets fed a competitive exclusion culture. In vitro investigations showed that Enterococcus strains have inhibitory effects on the growth of S. enterica serovar Enteritidis, and these effects were explained by both enterotoxin and nonenterotoxin factors (37). Other studies showed that E. faecium may be beneficial to the adhesion and colonization of Clostridium jejuni in the canine intestine (29) and reduced the rate of carryover infections with obligate intracellular pathogens from infected sows in piglets (26). E. faecium has also been shown to influence the composition of the bacterial community in the avian, swine, and canine gastrointestinal tracts (25, 29, 36).Infections with S. enterica are some of the most important sources of human gastroenteritis (39). In Germany, 52,563 human salmonellosis cases were reported in 2006 (http://www3.rki.de/SurvStat). The consumption of contaminated pork and pork products was found to be associated with 20% of human salmonellosis cases in Germany (33), indicating the importance of meat or meat products as a potential source of infection for consumers. Salmonella serovar Typhimurium, especially phage type DT104, is the Salmonella serotype most frequently isolated from pork (27), and it is of particular concern because of its acquisition of multiple antibiotic resistance (1, 38).In this study, we investigated the effect of E. faecium NCIMB 10415 on the infection dynamics of Salmonella serovar Typhimurium DT104, fecal shedding, and the patterns of Salmonella distribution in internal organs, as well as on the humoral immune response to Salmonella in weaning piglets. To the best of our knowledge, this is the first experimental study of the mode of action of a probiotic strain of E. faecium in which dissemination to different internal organs was investigated using weaned piglets experimentally infected with Salmonella.  相似文献

The antimicrobial activity of phenyl-thiazolylurea-sulfonamides against Staphylococcus aureus PheRS are dependent upon phenylalanine levels in the extracellular fluids. Inhibitor efficacy in animal models of infection is substantially diminished by dietary phenylalanine intake, thereby reducing the perceived clinical utility of this inhibitor class. The search for novel antibacterial compounds against Gram-negative pathogens led to a re-evaluation of this phenomenon, which is shown here to be unique to S. aureus. Inhibition of macromolecular syntheses and characterization of novel resistance mutations in Escherichia coli demonstrate that antimicrobial activity of phenyl-thiazolylurea-sulfonamides is mediated by PheRS inhibition, validating this enzyme as a viable drug discovery target for Gram-negative pathogens. A search for novel inhibitors of PheRS yielded three novel chemical starting points. NMR studies were used to confirm direct target engagement for phenylalanine-competitive hits. The crystallographic structure of Pseudomonas aeruginosa PheRS defined the binding modes of these hits and revealed an auxiliary hydrophobic pocket that is positioned adjacent to the phenylalanine binding site. Three viable inhibitor-resistant mutants were mapped to this pocket, suggesting that this region is a potential liability for drug discovery.  相似文献

目的:构建一株表达TNF-α Fab'抗体的大肠杆菌工程菌,并设计一种高效实用的策略以促进大肠杆菌周质空间的可溶性Fab'抗体表达。方法:首先,通过更换不同表达载体,改变轻链和重链顺序,更换信号肽,共表达分子伴侣(Skp)、二硫键合成酶(Dsbc)、肽基辅氨酰顺反异构酶(PPIB)、二硫键异构酶(hPDI)、核酸酶(Nuclease),以评估对Fab'抗体表达量的改善。其次,纯化表达的Fab'抗体。通过周质提取、Q阴离子交换柱净化、苯基柱捕获、Protein L柱亲和三步纯化方案得到高纯度的Fab'抗体。最终将纯化后的Fab'抗体进行亲和力测定。结果:提高正确组装的Fab'抗体表达量的策略有——将目的蛋白构建至pET-30a载体;重链在前、轻链在后;轻、重链采用相异的信号肽;共表达hPDI。周质提取液中的Fab'抗体浓度达到588.0mg/L提取液,纯化后产量可达28.2mg/L发酵液,总回收率为32.0%,纯度为90.9%。Fab'抗体亲和力为(5.8±3.0)×10-9mol/L,体外细胞学活性IC50为(5.2±2.4)×10-11mol/L。结论:通过大肠杆菌工程菌分子构建方式的优化,得到了一株高效表达可溶性Fab'抗体的工程菌株,为可溶性小分子抗体的规模化生产奠定了研究基础。  相似文献

Parts 1 and 2 have shown that the rate of production of bacterial cellulose stagnates because of the limitation of substrate supply and a wall effect, which hinders the removal of the product from the active cell zone. This paper demonstrates, how both of these problems can be eliminated in a novel bioreactor, where the substrates (mainly glucose and oxygen) are both fed directly to the surface of the product cellulose. This involves the generation of an aerosol spray of glucose and its even distribution to the living bacteria on the medium‐air interface. The apparatus was built and operated up to eight weeks with a constant rate of cellulose production. The aerosol system provides the basis for an economic production of bacterial cellulose in surface culture.  相似文献

目的:探究高迁移率族蛋白1(HMGB1)拮抗剂BoxA尾静脉注射对细菌性脑膜炎(BM)大鼠模型的临床体征改善和炎症抑制作用。方法:除外正常对照的雄性Sprague-Dawley(SD)大鼠设为对照组(n=20),另取60只大鼠行脑室立体定向注射20μL大肠杆菌Escherichia coli (DH5α1×10~7CFU/m L)建立BM模型,之后随机分为两组(各组n=30),一组尾静脉注射HMGB1拮抗剂BoxA,即BoxA组;一组麻醉后进行尾静脉注射无菌磷酸盐(PBS),即Vehicle组。造模3 d后,对各组大鼠的临床指标以及病理生理参数(颅内压和脑脊液白细胞(WBC)计数)进行评估,使用酶联免疫吸附实验(ELISA)法检测血清中HMGB1的相对含量,使用伊文思蓝染色观察血脑屏障(BBB)通透性,使用免疫荧光染色检测大脑皮层炎症因子(IL-1β和TNF-α)的表达水平。结果:相比Control组,Vehicle组临床指标,颅内压,WBC计数以及血清HMGB1含量明显提升(P0.05);而BoxA组相比Vehicle组,以上改变有部分减少(P0.05)。另外,Vehicle组较Control组EB渗漏增加且炎症因子(IL-1β和TNF-α)表达水平增高(P0.05)。与Vehicle组相比,BoxA组的这些变化亦被部分调节(P0.05)。结论:HMGB1抑制剂BoxA尾静脉注射能够下调HMGB1表达水平并同时缓解细菌性脑膜炎大鼠的临床症状和炎症反应。  相似文献

The Escherichia coli O9a and O8 polymannose O-polysaccharides (O-PSs) serve as model systems for the biosynthesis of bacterial polysaccharides by ATP-binding cassette transporter-dependent pathways. Both O-PSs contain a conserved primer-adaptor domain at the reducing terminus and a serotype-specific repeat unit domain. The repeat unit domain is polymerized by the serotype-specific WbdA mannosyltransferase. In serotype O9a, WbdA is a bifunctional α-(1→2)-, α-(1→3)-mannosyltransferase, and its counterpart in serotype O8 is trifunctional (α-(1→2), α-(1→3), and β-(1→2)). Little is known about the detailed structures or mechanisms of action of the WbdA polymerases, and here we establish that they are multidomain enzymes. WbdAO9a contains two separable and functionally active domains, whereas WbdAO8 possesses three. In WbdCO9a and WbdBO9a, substitution of the first Glu of the EX7E motif had detrimental effects on the enzyme activity, whereas substitution of the second had no significant effect on activity in vivo. Mutation of the Glu residues in the EX7E motif of the N-terminal WbdAO9a domain resulted in WbdA variants unable to synthesize O-PS. In contrast, mutation of the Glu residues in the motif of the C-terminal WbdAO9a domain generated an enzyme capable of synthesizing an altered O-PS repeat unit consisting of only α-(1→2) linkages. In vitro assays with synthetic acceptors unequivocally confirmed that the N-terminal domain of WbdAO9a possesses α-(1→2)-mannosyltransferase activity. Together, these studies form a framework for detailed structure-function studies on individual domains and a strategy applicable for dissection and analysis of other multidomain glycosyltransferases.  相似文献

AIMS: To determine the fate of the enteric indicator organism, Escherichia coli, in sewage sludge (biosolids)-amended agricultural soil in relation to soil type and moisture status under controlled conditions. METHODS AND RESULTS: We enumerated Escherichia coli in soil by membrane filtration and most probable number techniques. The background concentration of E. coli was higher in sandy loam than in silty clay soil. E. coli numbers increased in soil following addition of dewatered, mesophilic anaerobically digested sludge. Escherichia coli declined to a small extent with time in both moist and air-dried unamended control soils, although decay was only highly significant (P < 0.001) in moist sandy loam (T(90) = 100 days). Removal rates were high in sludge-treated moist soil (T(90) = 20 days), but were significantly reduced in amended air-dried soil. CONCLUSIONS: Slow removal of E. coli in air-dried soil as against their rapid decay in moist soil after sludge application indicated that the soil biota are involved in pathogen reduction processes in sludge-amended soil. SIGNIFICANCE AND IMPACT OF THE STUDY: Soil ecological mechanisms are implicated as having a critical role in the fate of enteric organisms introduced into temperate agricultural soil in sewage sludge.  相似文献

肠毒素大肠杆菌F18(ECF18)是引起仔猪断奶后水肿和腹泻病的主要病原菌,a1—岩藻糖转移酶基因(FUT1)是ECF18侵染猪小肠的受体蛋白候选基因。通过采用PCR—RFLP方法检测了5个西方商业猪种以及21个中国地方猪种(群)1458个个体在FUT1基因开放阅读框架的307核苷酸位点的G-A点突变(M307^G-A)遗传变异。结果表明:5个外来猪种以及中国地方猪种中的临高猪在该FUT1基因位点存在多态性,其他中国地方猪种均表现为极端的单态分布,只有易感的GG基因型,没有多态性。由此提示:1)如果猪FUT1 M307^G-A点突变是决定猪小肠上ECF18受体表达与否的关键因素,则绝大部分中国地方猪种均不具备抵抗ECF18的遗传基础,这除了表明ECF18抗性基因有可能起源于西方猪种外,同时也表明对中国地方猪种中在这个位点惟一存在多态性的海南临高猪的品种资源保存具有非常重要的意义。2)一般而言,在中国的养猪生产实践中,中国地方猪种的仔猪抗水肿与腹泻病能力普遍强于外来猪种,研究的结果提示有必要对中国地方猪种所具备的上述遗传抗性做更深入的研究,寻找、定位其相应的QTL或/和抗性基因。  相似文献

A study of mycorrhizal activity was conducted in a mature Norway spruce ((Picea abies) [L.] Karst.) stand subjected to soil treatments of liming and acidification for six years (Höglwald research project). Samples were collected five times during one growing season using a soil corer. All the turgescent, not shriveled mycorrhizal tips were sorted out and identified on the fungal species level as far as yet possible. The proportion of each mycorrhizal type on the plots was calculated. The results revealed a shift in the mycorrhizal communities caused by both acid rain and liming. Data are in agreement with the findings of the more comprehensive study on the mycorrhizal communities carried out by another research group on the same plots (Taylor et al., submitted)The activity of the predominant types of mycorrhizas, Piceirhiza gelatinosa, Piceirhiza nigra, Russula ochroleuca-P. abies, Tuber puberulum-P. abies, Tylospora sp.-P. abies, Xerocomus badius-P. abies, was investigated by staining hand sectioned tips with FDA and their fluorescence. Different FDA-hydrolysing activities of the mycorrihizal types had been found in a previous-year study on the same plots and were confirmed during the second year. The proportion of the different stages of activity of the mycorrhizal tissues was calculated on the type level and in connection to the soil treatments. X. badius-P. abies and R. ochroleuca-P. abies displayed the most active fungal tissues and proportion increased on the acidic plots while Tuber puberulum-P. abies and Piceirhiza nigra were the most active types and occurred in higher proportion after liming. Thus, the overall activities of the mycorrhizas were only slightly changed by the treatments. In addition to the mycorrhizal effect acidification reduced while liming enhanced the meristematic activity of the short root tips. The same tendency was found by studying root production on the same plots (Hahn and Marschner, 1998). Although nearly 3000 mycorrhizal tips were studied, the data are still limited, allowing no statistical validation. This is, however, the first investigation connecting overall activity of the mycorrihizal tissues with the proportion of the mycorrhizal types as influenced both by alterations of the forest soil caused by acid rain and liming. The results are interesting and reasonable but further investments are necessary to validate the general conclusions.  相似文献

The provisioning parameters, breeding success, adult mass, andsurvival of yellow-nosed albatrosses were studied over 7 successiveyears at Amsterdam Island, southern Indian Ocean. We examinedthe ability of this long-lived seabird to adjust its breedingeffort under different environmental conditions and the fitnessconsequences in terms of survival and quality of offspring produced. Provisioning rate and adult mass varied extensivelybetween years, and the lowest and highest values were associatedwith sea surface temperature anomalies. When waters aroundthe island were colder, adults were in good condition and broughtlarge meals at short intervals, whereas warmer waters resultedin lower provisioning rates, lower adult mass, and lighter chicksat fledging. Adult survival and fledging success were not affectedby sea surface temperature anomalies. Yellow-nosed albatrossesappear to be unable to adjust their breeding effort every season,and their differential breeding investment probably primarilyreflects different levels of food availability. Yellow-nosedalbatrosses are able to regulate their provisioning behavior according to the nutritional status of their chick only whenconditions are favorable. Birds appear to invest primarilyin their own future maintenance rather than in provisioning.They have a wide safety margin in body mass that limits mortalityrisks during good years as well as during poor years. However,during unfavorable seasons adults continue to provision chicksthat have a poor prospect of survival to breeding, withoutadditional survival costs for the parents. Favorable seasonstherefore have a high value in terms of fitness because ofthe high quality of the chick produced. We suggest that understandinghow long-lived animals optimize their provisioning behaviorand lifetime reproduction can only be achieved through studiesencompassing several contrasted seasons.  相似文献

Reproductive success of the cooperative breeding Seychelleswarbler (Acrocephalus sechellensis) increases with age. Thisage effect is not due to differential survival or increasedreproductive effort, but to accumulated helping and breedingexperience. In their first year of breeding, reproductive performanceof inexperienced warblers with neither helping nor breedingexperience was significandy lower than that of warblers of thesame age with either previous helping or breeding experience.Reproductive performance was the same for primiparae with helpingexperience and for birds with breeding experience. Female primiparaewith helping experience or breeding experience built betternests and spent more time incubating than inexperienced females,which led to increased hatching success. Male primiparae withhelping experience or males with breeding experience guardedthe clutch better than inexperienced males, which led to reducedegg predation. Even-aged warblers with different previous experienceswere transferred to unoccupied islands, where birds startedbreeding immediately in high-quality territories. The experimentshowed that birds with helping experience produced their firstfledgling as fast as experienced breeders, and significandyfaster than inexperienced birds. Breeding performance did notimprove further with experience after the first successful breedingattempt. Only birds with previous breeding experience who pairedwith inexperienced birds, were likely to change mate. The otherpair combinations remained stable. Thus, primiparous birds withhelping experience have greater lifetime reproductive successthan inexperienced primiparae of the same age. This experimentshows that helping behavior has not only been selected for inthe context of promoting an individual's indirect fitness, butalso in the context of gaining helping experience which translatesinto improved reproductive success when a helper becomes a breeder.[Behav Ecol 7: 326-333 (1996)]  相似文献

AIMS: The work is intended to achieve optimum culture conditions of alpha-galactosidase production by a mutant strain Penicillium sp. in solid-state fermentation (SSF). METHODS AND RESULTS: Certain fermentation parameters involving incubation temperature, moisture content, initial pH value, inoculum and load size of medium, and incubation time were investigated separately. The optimal temperature and moisture level for alpha-galactosidase biosynthesis was found to be 30 degrees C and 50%, respectively. The range of pH 5.5-6.5 was favourable. About 40-50 g of medium in 250-ml flask and inoculum over 1.0 x 10(6) spores were suitable for enzyme production. Seventy-five hours of incubation was enough for maximum alpha-galactosidase production. Substrate as wheat bran supplemented with soyabean meal and beet pulp markedly improved the enzyme yield in trays. CONCLUSIONS: Under optimum culture conditions, the alpha-galactosidase activity from Penicillium sp. MAFIC-6 indicated 185.2 U g(-1) in tray of SSF. SIGNIFICANT AND IMPACT OF THE STUDY: The process on alpha-galactosidase production in laboratory scale may have a potentiality of scaling-up.  相似文献

Allo-chimerism and clonal elimination of self antigen (Ag) (Ia + Mls-1a) reactive Vβ6+ T cells were analyzed and compared between allogeneic bone marrow (BM) chimeras reconstituted with BM cells which had been treated with anti-Thy-1 monoclonal antibody (mAb) plus complement (C) (T chimeras) and BM chimeras which had been reconstituted with BM cells pretreated with anti-Thy-1 mAb alone (T+ chimeras). When lethally irradiated AKR (Mls-1a) mice were reconstituted with BM cells from B10 or B10 H-2 congenic mice, both T+ and T chimeras were entirely free of signs of graft-versus-host reaction (GVHR). However, complete replacement of the AKR lymphoid tissues by donor BM cells was accomplished at an early stage in T+ chimeras but not in T chimeras. On the other hand, clonal elimination of Vβ6+ T cells reactive to the recipient Ag (Mls-1a) was abolished in T+ chimeras but successfully induced in T chimeras. The Vβ6+ T cells not eliminated in T+ chimeras showed depressed responses against Mls-1a antigens. The findings herein demonstrate that T cells which contaminate a BM inoculum survive in recipient mice after treatment with anti-Thy-1 mAb without C in vitro followed by BMT. The surviving T cells have been estimated to represent fewer than 0.5% of the BM cells inoculated. These cells appear to accelerate the full replacement of recipient lymphoid tissues by donor cells. Furthermore, the T cells which survive in the marrow inoculum influence eventually the development of a tolerant state in the T cell repertoire of the donor.  相似文献

CO and NO are small toxic gaseous molecules that play pivotal roles in biology as gasotransmitters. During bacterial infection, NO, produced by the host via the inducible NO synthase, exerts critical antibacterial effects while CO, generated by heme oxygenases, enhances phagocytosis of macrophages. In Escherichia coli, other bacteria and fungi, the flavohemoglobin Hmp is the most important detoxification mechanism converting NO and O2 to the ion nitrate (NO3). The protoheme of Hmp binds not only O2 and NO, but also CO so that this ligand is expected to be an inhibitor of NO detoxification in vivo and in vitro. CORM-3 (Ru(CO)3Cl(glycinate)) is a metal carbonyl compound extensively used and recently shown to have potent antibacterial properties. In this study, attenuation of the NO resistance of E. coli by CORM-3 is demonstrated in vivo. However, polarographic measurements showed that CO gas, but not CORM-3, produced inhibition of the NO detoxification activity of Hmp in vitro. Nevertheless, CO release from CORM-3 in the presence of soluble cellular compounds is demonstrated by formation of carboxy-Hmp. We show that the inability of CORM-3 to inhibit the activity of purified Hmp is due to slow release of CO in protein solutions alone i.e. when sodium dithionite, widely used in previous studies of CO release from CORM-3, is excluded. Finally, we measure intracellular CO released from CORM-3 by following the formation of carboxy-Hmp in respiring cells. CORM-3 is a tool to explore the concerted effects of CO and NO in vivo.  相似文献

The growth of Hansenula polymorpha and Kloeckera sp. 2201 with a mixture of glucose and methanol (38.8%/61.2%, w/w) and the regulation of the methanol dissimilating enzymes alcohol oxidase, catalase, formaldehyde dehydrogenase and formate dehydrogenase were studied in chemostat culture, as a function of the dilution rate. Both organisms utilized and assimilated glucose and methanol simultaneously up to dilution rates of 0.30 h-1 (H. polymorpha) and 0.26h-1, respectively (Kloeckera sp. 2201) which significantly exceeded max found for the two yeasts with methanol as the only source of carbon. At higher dilution rates methanol utilisation ceased and only glucose was assimilated. Over the whole range of mixed-substrate growth both carbon sources were assimilated with the same efficiency as during growth with glucose or methanol alone.In cultures of H. polymorpha, however, the growth yield for glucose was lowered by the unmetabolized methanol at high dilution rates. During growth on both carbon sources the repression of the synthesis of all catabolic methanol enzymes which is normally caused by glucose was overcome by the inductive effect of the simultaneously fed methanol. In both organisms the synthesis of alcohol oxidase was found to be regulated differently as compared to catalase, formaldehyde and formate dehydrogenase. Whereas increasing repression of the synthesis of alcohol oxidase was found with increasing dilution rates as indicated by gradually decreasing specific activities of this enzyme in cell-free extracts, the specific activities of this enzyme in cell-free extracts, the specific activities of catalase and the dehydrogenases increased with increasing growth rates until repression started. Abstract. The effects of pheromone concentration and ambient temperature on male gypsy moth, Lymantria dispar (L.) (Lepidoptera), flight responses to pheromone were investigated in a wind tunnel. As the pheromone dose increased from 10 ng to 1000 ng, males flew at progressively slower airspeeds and ground speeds, and reduced their wingbeat frequencies. Furthermore, the moths steered significantly smaller course angles as the pheromone concentration increased, indicating that they were adopting a more upwind heading. The overall width of the flight tracks also decreased when males flew in more concentrated pheromone plumes. Estimation of plume dimensions using a male wing-fanning assay showed that as pheromone dosage increased, the resultant active spaces became wider, indicating that an inverse relationship existed between the dimensions of the time-averaged plume and the width of track reversals and that most turns were initiated within the plume. When males were flown at cool (20°C) and warm (26°C) ambient temperatures but to equivalent pheromone emission rates, they exhibited higher airspeeds and ground speeds at the higher temperature but steered larger course angles. Track widths, and length of track legs were, however, similar at the two temperatures. The mean turning frequency was nearly the same (c. 4 turns/s) across all the concentrations and temperatures tested even though the moths' thoracic temperature differed by 5°C when the ambient temperature was varied.  相似文献

Protonation of the Ca2+ ligands of the SR Ca2+-ATPase (SERCA1a) was studied by a combination of rapid scan FTIR spectroscopy and electrostatic calculations. With FTIR spectroscopy, we investigated the pH dependence of CO bands of the Ca2+-free phosphoenzyme (E2P) and obtained direct experimental evidence for the protonation of carboxyl groups upon Ca2+ release. At least three of the infrared signals from protonated carboxyl groups of E2P are pH dependent with pKa values near 8.3: a band at 1758 cm−1 characteristic of nonhydrogen-bonded carbonyl groups, a shoulder at 1720 cm−1, and part of a band at 1710 cm−1, both characteristic of hydrogen-bonded carbonyl groups. The bands are thus assigned to H+ binding residues, some of which are involved in H+ countertransport. At pH 9, bands at 1743 and 1710 cm−1 remain which we do not attribute to Ca2+/H+ exchange. We also obtained evidence for a pH-dependent conformational change in β-sheet or turn structures of the ATPase. With MCCE on the E2P analog E2(), we assigned infrared bands to specific residues and analyzed whether or not the carbonyl groups of the acidic Ca2+ ligands are hydrogen bonded. The carbonyl groups of Glu771, Asp800, and Glu908 were found to be hydrogen bonded and will thus contribute to the lower wave number bands. The carbonyl group of some side-chain conformations of Asp800 is left without a hydrogen-bonding partner; they will therefore contribute to the higher wave number band.  相似文献
