Serum samples were collected 1–3 times weekly from two Baird's tapirs (Tapirus bairdii) for 6 months in 1987–1988, and for more than 3 consecutive years beginning in 1989 to characterize hormone patterns during the estrous cycle and pregnancy. Based on serum progesterone concentrations, mean (±SEM) duration of the estrous cycle (n = 20) was 30.8 ± 2.6 days (range, 25–38 days) with a luteal phase length of 18.1 ± 0.4 days (range, 15–20 days). Mean peak serum progesterone concentrations during the luteal phase were 1.35 ± 0.16 ng/ml, and nadir concentrations were 0.19 ± 0.03 ng/ml during the interluteal period. Distinct surges of estradiol preceded luteal phase progesterone increases in most (14/20) cycles. Gestation length was 392 ± 4 days for three complete pregnancies. Mean serum progesterone concentrations increased throughout gestation and were 1.83 ± 0.13, 2.73 ± 0.13, and 4.30 ± 0.16 ng/ml during early, mid- and late gestation, respectively. Serum estradiol concentrations began to rise during mid-gestation, increasing dramatically during the last week of pregnancy. Patterns of serum estriol and estrone secretion during pregnancy were similar to that observed for estradiol. In contrast to progesterone and estrogens, serum cortisol concentrations were unchanged during pregnancy or parturition. Females resumed cycling 16.2 ± 2.0 days after parturition (n = 4) and, on two occasions, females became pregnant during the first postpartum estrus. These data suggest that the tapir cycles at approximately monthly intervals and that increases in serum progesterone are indicative of luteal activity. The interluteal period is relatively long, comprising approximately 40% of the estrous cycle. During gestation, progesterone concentrations are increased above luteal phase levels, and there is evidence of increased estrogen production during late gestation. The absence of increased cortisol secretion at the end of gestation suggests that this steroid does not play a major role in initiating parturition in this species. © 1994 Wiley-Liss, Inc.  相似文献

Immunoreactive pregnanediol-3-glucuronide was assessed in daily urine samples from five female okapis. Indexed by creatinine (CR), the resultant profiles accurately reflect ovulation and pregnancy in this species. The infertile cycle was approximately 14.5 days by this method with the preovulatory levels averaging less than 1 ng/mg CR (N = 88) for approximately eight days (N = 28). Postovulatory levels averaged 24 ng/mg CR (N = 55) in one female and 12 ng/mg CR (N = 27) in another for approximately six days (N = 26). Pregnancy is confidently detected by three weeks postcoition by elevated pregnanediol levels that remain significantly higher that luteal phase levels until weeks before parturition.  相似文献

Details of the endocrinology of reproduction in the genus Callithrix are known only for the common marmoset, C. jacchus. This paper presents the patterns of urinary pregnandiol-3-glucuronide (PdG), urinary estrone conjugates (E1C), and gonadotropin excretion throughout the reproductive cycle of Wied's black tufted-ear marmoset (C. kuhli) as determined via steroid conjugate enzyme immunoassays (EIA) and gonadotropin radioimmunoassays (RIA). Postpartum ovulation occurred at 13.6 ± 1.2 days after parturition (n = 12) and was characterized by low PdG and E1C concentrations accompanied by a spike in luteinizing hormone (LH)/chorionic gonadotropin (CG) concentration. After conception, PdG concentrations increased dramatically until they dropped to periovulatory concentrations in the third trimester of pregnancy. Mean PdG concentrations in the first and second trimesters (33.7 ± 8.4 and 39.0 ± 10.9 μg/mg creatinine, respectively) were three times that of third trimester concentrations (11.7 ± 1.4 μg/mg Cr; n = 8). Urinary concentrations of E1C rose more gradually during pregnancy and remained higher prepartum than urinary concentrations of PdG. Urinary gonadotropin concentrations also increased after conception (first trimester concentrations = 24.5 ± 4.5 ng/mg Cr) and continued to increase in the second trimester (51.4 ± 7.6 ng/mg Cr), until they finally decreased in the third trimester (mean = 7.9 ± 1.4 ng/mg Cr; n = 8). The interbirth interval was 156.3 ± 2.9 days (n = 6), with a gestation of 143.1 ± 1.6 days (n = 8). Nonconceptive cycle length was 24.9 ± 0.6 days (n = 4). The reproductive physiology of nine Galápagos tortoises (Geochelone nigra) was studied from February 1988 to May 1989. The study encompassed the annual reproductive cycle to include complete mating and nesting sequences. Male (n = 4) and female (n = 5) seasonal reproductive changes were determined throughout the study with endocrine analysis and ultrasonographic examinations. Males displayed a prenuptial rise in serum testosterone (x― ±SE = 6.62 ± 0.92 ng/ml in August) during which gonadal maturation and spermatogenesis are thought to occur. The male reproductive cycle appears consistent with the prenuptial spermatogenic pattern exhibited by other tropical turtles. In the females, testosterone rose during the mating period (x― ± SE = 499.3 ± 124.6 pg/ml in October) prior to ovulation and is probably related to receptivity in the females. Progesterone was more variable, but also peaked during the mating period (x― ± SE = 1,017.2 ± 220.6 pg/ml in October) and appears related to ovulation. Estradiol rose several months prior to mating (x― ± SE = 75.5 ± 11.9 pg/ml in July) and was correlated with increased serum calcium levels. This increase in estradiol is thought to stimulate vitellogenesis several months prior to mating. Nesting occurred from November 1988 to April 1989, during which six clutches were laid. A study was undertaken to evaluate the effects of using an oral progestagen for synchronisation of parturition in the sow. Multiparous Landrace X Large White sows were fed 20 mg allyl trenbolone daily from day 110 to 115 of gestation then 15 mg on day 116 (Group R; N = 12); untreated sows of similar background served as controls (Group C; N = 9). Blood samples, taken at 8-h intervals from day 110 of gestation to onset of parturition, then every 4 h until parturition was complete, were assayed for plasma levels of progesterone, unconjugated oestrone, cortisol and prolactin. Duration of farrowing, incidence of stillbirths and individual piglet birth weights were recorded. Five Group R sows farrowed during the period of progestagen administration, while for the remaining 7, the mean interval from last progestagen treatment to emergence of first piglet was 31.6 ±5.5 h. Gestation length, duration of parturition, and mean interval between successive births all were longer in Group R than in Group C (116.5 ± 0.34 compared with 115.0 ± 0.52 days, P < 0.01; 9.73 ± 1.98 compared with 3.08 ± 0.70 h, P < 0.01; and 53.4 ± 10.2 compared with 16.2 ± 2.4 min., P < 0.01, respectively). No significant treatment differences were apparent for litter size at birth, proportion stillborn or piglet birth weights. Profile analysis showed that plasma progesterone levels in Group R were lower (P < 0.05) during the 30 h prefarrowing, suggesting a longer mean interval between functional luteolysis and parturition in these animals. In both Groups plasma levels of unconjugated oestrone rose in the prefarrowing period, the levels being higher (P < 0.05) in Group R Peak oestrone levels occurred at the commencement of, and had declined to low levels by the completion of, farrowing. Cortisol levels exhibited a pattern similar to that of oestrone, although peak levels at parturition were lower in Group R (P < 0.01). Plasma prolactin levels in the 24 h prepartum rose faster and reached higher levels (P < 0.05) in Group C than Group R, but the difference was no longer apparent subsequent to first piglet emergence. It is concluded that the use of this progestagen to delay parturition upset the synchronisation of endocrine events at farrowing, resulting in an increased duration of parturition.  相似文献

Until now, unlike their relatives, rhinos and horses tapirs have received considerably less attention in studies about communication. Therefore, it was the aim of this study to test which stimuli contain acoustical information for tapirs. For this purpose, the reactions of tapirs on acoustical stimuli (playback of different animal voices) were examined. Research visits took place at the zoos of Berlin, Dortmund, Heidelberg, Munich, Nuremberg and Osnabrück during the year 2005. A total of 20 individuals, thereof 8 (4.4) Malayan tapirs (Tapirus indicus) and 12 (4.8) Lowland tapirs (Tapirus terrestris) attended the experiments. The playback experiments showed that tapirs distinguish between the voices of different animal species. The results point to the conclusion that the reactions of the tapirs relate to phylogeny. The most intense interest was taken in their own species followed by the closely related ones.  相似文献

Ischemia of rat intestine was induced in vivo by occlusion of the superior mesenteric artery (SMA) for 15 min. Sodium salicylate, 100 mg/kg, given IP, 30 min prior to the ischemic event served as a specific trap for hydroxyl radicals and provided direct evidence for the involvement of free radicals during the ischemic insult. Portions of the bowel were sequentially isolated and removed. The hydroxylation products. dihyd-roxybenzoic acid (DHBA) derivatives were isolated, identified and qunatified by HPLC coupled with electrochemical detection (ECD). The level of 2,5-DHBA (Mean ± SE, ng/g tissue) in the preischemic bowel (N = 21) was 241.8 ± 10.0. It rose significantly to 313.3 ± 15.5 in the ischemic specimen (p = 0.0129) and remained unchanged in the reperfusion period (322.8 ± 15.5). The histological examination correlated well with these levels: mild villi damage in the ischemic period with no further damage in the reperfusion period.  相似文献

Although numbers of giant anteaters within North American facilities have been steadily increasing for the last 15 years, the population now exhibits an unstable age distribution with genetically valuable individuals nearing reproductive senescence. Contributing to this issue is the Association of Zoos and Aquariums (AZA) described lack of standardization of breeding introduction practices and high risk of female injury occurring during such pairings. This report describes the development of a successful breeding protocol at Cleveland Metroparks Zoo based on hormone monitoring and efficient communication between science and animal management teams that minimizes risk of female injury. By training a female giant anteater for urine sample and body weight data collection, staff members accurately predicted estrus, and timed breeding introductions to facilitate positive interactions between the male and female. Such training also allowed for careful monitoring of two pregnancies through parturition (169–184 days from breeding) and post‐partum return to estrus (114–129 days from parturition). Urinary hormone monitoring revealed a sharp progestogen increase averaging >five‐fold over basal levels (0.52 ± 0.05 ng/mg creatinine) which was sustained throughout the second half of pregnancy. Mean regular estrous cycle length (n = 14 cycles), was calculated as 46.17 ± 1.39 days, measured as days between estrogen peaks of mean concentration 2.27 ± 0.19 ng/mg creatinine. This report summarizes impressive collaborative efforts among multiple zoological departments to achieve extensive hormonal and body weight monitoring from a female giant anteater, adding valuable information on reproductive parameters, and specifics for novel hormone assay techniques.  相似文献

The Malayan, or Asian, tapir (Tapirus indicus) has a diminishing wild population and is becoming more common in captivity as zoos attempt to manage sustainable ex situ populations. Tapirs can be relatively easy to maintain and breed, but captive animals appear to suffer from reduced activity budgets, obesity, and poor public image. A questionnaire‐based survey was designed and sent specifically to 10 collections around the world that exhibit Malayan tapirs, with the aim of assessing husbandry regimes to determine prevalence of standardized practices as well as highlighting any key differences, and to showcase good practice, thus providing information beneficial to those maintaining this species in their zoo. Twenty‐five animals were included in the survey from collections across four continents. The research's major conclusions show differing dietary make‐up, with a lack of forage provision, contrasting with a diverse array of enrichment protocols used. Significant differences were noted between zoos for total amount of food offered (P = 0.000) as well as ratios of forage to concentrate pellet offered (P = 0.004). Comparing food offered to male and female tapirs with published requirements for an “average” of either gender shows not all zoos providing the amount suggested in husbandry guidelines. Intelligently designed and original enrichment was provided to all animals but differences between zoos were noted in the application and “usefulness” of enrichment for individual tapir. Overall, animals are benefiting from enrichment but welfare could be further improved via consistent feeding of ad libitum forage and regular use of browse as a constituent part of daily rations. A Sumatran rhinoceros with a history of early pregnancy loss was supplemented with a synthetic progestin, altrenogest (Regu‐Mate®), and delivered a healthy, full‐term calf 475 days after mating. Serum hormone concentrations were measured throughout gestation, and ultrasonography was used to monitor embryo/fetal growth and viability. The embryonic vesicle growth curve was characterized by three phases: rapid expansion, plateau, and a final rapid expansion, and was similar to that in the domestic horse. Fetal sex was determined by ultrasound on day 73 of gestation. After day 80 of gestation, transabdominal examinations were more useful than rectal examinations for imaging the fetus. Serum progesterone concentrations remained at luteal levels (1.5±0.5 ng/ml) for the first 2 months of pregnancy, and then they gradually increased. However, progesterone decreased almost to luteal levels during the fifth month before it increased again, and eventually reached peak concentrations (13.3±1.9 ng/ml) shortly before parturition. Relaxin concentrations remained basal (≤0.5 ng/ml) for the first half of the pregnancy, increased to 2.7±1.2 ng/ml and stabilized until 2 weeks before parturition, when relaxin spiked to unusually high concentrations (800–1300 ng/ml). Prolactin concentrations were at baseline (7.2±1.7 ng/ml) throughout most of the gestation, but rose markedly 2 weeks before parturition, reaching concentrations as high as 75 ng/ml. Attempts to measure serum estrogen concentrations were unsuccessful. These data represent the first attempt to characterize pregnancy in the critically endangered Sumatran rhinoceros, a species that heretofore had not successfully reproduced in captivity for 112 years. Background Animals in captivity should receive adequate sunlight exposure for sufficient generation of vitamin D [25(OH)D]. In the present study, 25(OH)D serum levels of 84 Callithrix penicillata primates were evaluated. Objectives To determine 25(OH)D levels of those animals; to evaluate the influence of gender and period of sunlight exposure on their 25(OH)D levels. Methods Three groups were evaluated: group 1 (n = 29) on free sunlight exposure; group 2 (n = 34) on partial sunlight exposure; group 3 (n = 21) without sunlight exposure. Results The obtained 25(OH)D values were: group 1, 121.2 ± 33.3 ng/ml; group 2, 115.2 ± 32.2 ng/ml; group 3, 53.3 ± 10.4 ng/ml. Significant statistical differences were obtained between groups 1 and 3 (p < 0.001) and groups 2 and 3 (p < 0.001); no statistical difference was found between genders. Conclusion Direct sunlight exposure is essential for 25(OH)D sufficiency and it is proposed that the 25(OH)D normal range for captive Callithrix penicillata would be from 104.8 to 137.1 ng/ml (CI = 95%).  相似文献

Urinary estrone conjugates were measured directly by radioimmunoassay (RIA) in 20 pregnancies from preconception diestrus to day 78 of pregnancy. High performance liquid chromatography separation defined estrone sulfate (ES) as the predominant immunoreactive peak which accounted for 94% to 97% of the total immunoreactivity after chromatography. Diestrous values indexed by creatinine were 0.15 ± 0.07 micrograms/mg Cr, x ± SEM as compared to estrous values which rose to 0.47 ± 0.14 micrograms/mg Cr, x ± SEM. Urinary ES concentrations significantly increased (P = 0.0001 in pregnant mares from day 35 to day 47 (1.21 ± 0.12 micrograms/mg Cr) as compared to day 25 to day 34 (0.27 ± 0.01 micrograms/mg Cr). Measurement of urinary ES may provide an alternate or augmentive method of pregnancy diagnosis in the domestic mare.  相似文献

Objective: Recent evidence suggests that diets high in calcium and dairy products are associated with lower body weight, particularly lower body fat levels. The purpose of this study was to compare weight and body fat loss on a calorie-restricted, low-dairy (CR) vs. high-dairy (CR+D) diet. Research Methods and Procedures: Fifty-four subjects (BMI 30 ± 2.5 kg/m2, 45 ± 6.6 years, 4 men) were randomly assigned to calorie-restricted (−500 kcal/d) low-dairy calcium (n = 29; ∼1 serving dairy/d, 500 mg/d calcium) or high-dairy calcium (n = 25; 3 to 4 servings dairy/d, 1200 to 1400 mg/d calcium) diets for 12 months. Main outcome measures included change in weight (kilograms) and body fat (percentage). Results: There were no significant differences between groups at baseline. At 12 months, weight and body fat loss were not significantly different. Subjects in the CR vs. CR+D conditions lost 9.6 ± 6.5 vs. 10.8 ± 5.9 kg (p = 0.56) and 9.0 ± 3.8 vs. 10.1 ± 3.6 kg body fat (p = 0.37). Discussion: These findings suggest that a high-dairy calcium diet does not substantially improve weight loss beyond what can be achieved in a behavioral intervention.  相似文献

Hormonal profiles during postpartum estrus, time of conception, and pregnancy were determined in urine samples from six cotton-top tamarins (Saguinus oedipus oedipus). Noninvasive collection techniques permitted daily sampling throughout lactation and pregnancy. Urinary estrone (E1), estradiol (E2), and both bioactive and immunoreactive luteinizing hormone/chorionic gonadotropin (LH/CG) measures revealed an interval of 19 ± 2.07 (S E M) days between parturition and the postpartum ovulatory LH peak. An increase in both E1 and E2 was seen prior to the LH peak; however, E1 and E2 continued to increase to their highest concentrations after the LH peak. Since postpartum ovulations resulted in pregnancy, neither postpartum estrus nor conception was suppressed by lactation. The length of gestation (measured from the LH peak to parturition) was 183.7 ± 1.14 (S E M) days, which is at least 30 days longer than that previously reported for other callitrichid species. Both E1 and E2 reached their maximum levels during midpregnancy but showed a rapid decline at parturition. Gestational levels of CG were first detectable approximately 20 days after the LH peak and continued to be elevated for approximately 80 days. The Sub-Human Primate Tube Test (SHPTT) for pregnancy did not detect the LH Peak and was less sensitive than other methods in detecting CG. Two RIA methods and a bioassay technique could not distinguish between LH and CG. We concluded that monitoring both estrogen and LH concentration was needed to determine when ovulation occurs in the cotton-top tamarin, since peak values of estrogen are seen after the ovulatory LH peak. Also, these tamarins were pregnant the majority of the time, indicating an unusually high fertility rate in contrast to most noncallitrichid primate species.  相似文献

Direct measurements of urinary immunoreactive estrone conjugates (E1C) and pregnanediol glucuronide (PdG), were applied to monitoring the ovarian cycle (n = 9) and pregnancy (3 full term pregnancies, 2 mid-term abortions) in Saguinus fuscicollis. During the ovarian cycle, urinary E1C concentrations revealed a high degree of day-to-day variability and appeared to be uninformative in reflecting cyclic ovarian function. In contrast, PdG was a reliable indicator of ovarian cyclicity with excretion patterns corresponding well with plasma progesterone profiles. Luteal phase PdG concentrations were on average 4–7–fold higher than corresponding follicular phase values. On the basis of changes in circulating progesterone, a mean cycle length of 25.7 ±1.0 days with an average follicular phase of 7.1 ± 0.6 days and a mean luteal phase of 18.6 ± 0.7 days, was found (n = 14 cycles). Following conception, both urinary steroid conjugate concentrations increased and elevated levels were maintained beyond the normal luteal phase length, allowing pregnancy to be determined at around day 25–30. During mid- to late pregnancy, PdG levels declined while E1C concentrations continued to be elevated until approximately 6 weeks before parturition when a decrease occurred. Both hormones showed a clear and rapid fall to follicular phase values following termination of pregnancy at either parturition or mid-term abortion. Post partum ovulations (n = 5) occurred on average 17–18 days following birth with four ovulations leading to conceptions. The results demonstrate the potential of urinary steroid conjugate analysis as a practical and reliable method for non-invasive monitoring of reproductive status in the female saddle-back tamarin. © 1995 Wiley-Liss, Inc.  相似文献

Lifespan in rodents is prolonged by caloric restriction (CR) and by mutations affecting the somatotropic axis. It is not known if CR can alter the age‐associated decline in growth hormone (GH), insulin‐like growth factor (IGF)‐1 and GH secretion. To evaluate the effect of CR on GH secretory dynamics; forty‐three young (36.8 ± 1.0 years), overweight (BMI 27.8 ± 0.7) men (n = 20) and women (n = 23) were randomized into four groups; control = 100% of energy requirements; CR = 25% caloric restriction; CR + EX = 12.5% CR + 12.5% increase in energy expenditure by structured exercise; LCD = low calorie diet until 15% weight reduction followed by weight maintenance. At baseline and after 6 months, body composition (DXA), abdominal visceral fat (CT) 11 h GH secretion (blood sampling every 10 min for 11 h; 21:00–08:00 hours) and deconvolution analysis were measured. After 6 months, weight (control: ?1 ± 1%, CR: ?10 ± 1%, CR + EX: ?10 ± 1%, LCD: ?14 ± 1%), fat mass (control: ?2 ± 3%, CR: ?24 ± 3%, CR + EX: ?25 ± 3%, LCD: ?31 ± 2%) and visceral fat (control: ?2 ± 4%, CR: ?28 ± 4%, CR + EX: ?27 ± 3%, LCD: ?36 ± 2%) were significantly (P < 0.001) reduced in the three intervention groups compared to control. Mean 11 h GH concentrations were not changed in CR or control but increased in CR + EX (P < 0.0001) and LCD (P < 0.0001) because of increased secretory burst mass (CR + EX: 34 ± 13%, LCD: 27 ± 22%, P < 0.05) and amplitude (CR + EX: 34 ± 14%, LCD: 30 ± 20%, P < 0.05) but not to changes in secretory burst frequency or GH half‐life. Fasting ghrelin was significantly increased from baseline in all three intervention groups; however, total IGF‐1 concentrations were increased only in CR + EX (10 ± 7%, P < 0.05) and LCD (19 ± 4%, P < 0.001). A 25% CR diet for 6 months does not change GH, GH secretion or IGF‐1 in nonobese men and women.  相似文献

We postulated that dietary ingestion of vitamin D may be used by some Alaskan Arctic marine mammal species in addition to, or instead of, cutaneous production to meet nutritional requirements. Zooplankton (n=5) sampled near Kaktovik, Alaska, contained no measurable vitamin D2 or D3, but did contain provitamin D (7‐dehydrocholesterol), the cutaneous precursor for previtamin D3 in mammals. Fillets and livers from five fish species were sampled near Barrow, Alaska, and evaluated for vitamin D3 content (no vitamin D2 was detected). Differences in vitamin D3 content appeared significant (P≤0.10) among fish livers (Kruskal‐Wallis [H test]=8.25, df=4, P=0.08) and among fish fillets (H=7.80, df=4, P=0.01). We also found significant differences in several pairwise comparisons (Mann‐Whitney U‐test) of vitamin D3 levels in fillets and livers. Blubber from six species of marine mammals had no detectable vitamin D2. The H test results for blubber vitamin D3 concentration were highly significant: 28.12, df=5, P<0.001. There were also significant differences in vitamin D3 content from blubber in pairwise comparisons of primarily invertebrate feeders (bowhead whale (Balaena mysticetus) [mean=4.20 SD±1.10 ng/g], and Pacific walrus (Odobenus rosmarus divergens) [5.43±2.82 ng/g]) vs. primarily piscivorous feeders (ringed seal (Phoca hispida) [746.57±493.00 ng/g] and beluga whale (Delphinapterus leucas) [426.00±174.92 ng/g]) and a semiaquatic terrestrial carnivore (polar bear (Ursus maritimus) [406.17±311.70 ng/g]). The bearded seal (Erignathus barbatus) had intermediate blubber vitamin D3 concentration (156.83±139.25 ng/g), which may reflect an intermediate‐type feeding strategy or an artifact of the small sample size. Eight species of pangolin have been identified in the world. However, understanding of pangolin reproductive biology has been limited to fragmentary records. In this study, the concentration of serum progesterone in three pregnant and two nonpregnant rescued female Formosan pangolins (Manis pentadactyla pentadactyla) was monitored using a commercial progesterone radioimmunoassay kit. During gestation, the serum progesterone of pregnant pangolins A, B, and C remained at 28.5–55 ng/ml (n = 31 samples), 10.9–50.1 ng/ml (n = 34), and 12.4 and 33.5 ng/ml with a peak at 47.6 ng/ml (n = 19), respectively, whereas the serum progesterone of nonpregnant pangolins D and E remained at 1.99 ± 1.62 ng/ml (n = 80) and 2.27 ± 1.64 ng/ml (n = 27), respectively. From this study, it was found that female pangolin weighing as low as 2.14 kg was already capable of reproduction. For pregnant pangolins to give birth to viable offspring, their body weight must increase significantly, 63.89 and 134.0% in the study, from the time of inception or early pregnancy until parturition. In addition, study has found that both viable offspring were born fully developed and exceeded 80 g in weight. The period of gestation was found to be as short as 318 or longer than 372 days. Therefore, the Formosan pangolin should only be able to reproduce once a year. This study is the first insight into hormone assay for determining the gestation period of pangolin. Further investigations on the same subject are necessary to establish criteria for the recognition of reproductive status in pangolins. Objective: To examine the effects of graded doses of hydrocortisone (HC) on leptin secretion, and determine the effect of fasting. Research Methods and Procedures: This was a randomized, placebo‐controlled, crossover study, with a 1‐week “washout” period between interventions. Eight healthy subjects [age = 36 ± 2.3 years (±SE), body mass index = 31.5 ± 1.6 kg/m2] completed the dose‐response study in which an intravenous infusion of saline (placebo) or HC (30 or 100 mg) was administered for 24 hours. Four healthy subjects (age = 35.2 ± 3.0 years, body mass index = 27.1 ± 2.1 kg/m2) completed the fasting study, which entailed continuous infusion of saline, HC (300 mg/24 hours) in the fed state, or HC (300 mg/24 hours) with total caloric deprivation for 24 hours. Blood sampling was performed every 1 to 2 hours for measurement of leptin, cortisol, insulin, and glucose levels. Results: Peak hyperleptinemia occurred after 16 hours of HC infusion; peak/baseline leptin levels were 129% (placebo), 140% (30 mg of HC for 24 hours, p = 0.05), and 185% (100 mg of HC for 24 hours, p < 0.01). During infusion of HC (300 mg/24 hours or placebo), the peak/baseline plasma leptin levels were 16.1 ± 5.8/12.8 ± 5.9 ng/mL (placebo with food, 126%), 14.6 ± 6.0/12.5 ± 6.5 ng/mL (HC fasting, 117%), and 32.5 ± 12.5/12.0 ± 8.4 ng/mL (HC with food, 271%, p < 0.001). Discussion: Leptin secretory responses occur at physiological doses of HC, are obliterated by fasting, and thus may be of metabolic significance.  相似文献

Abstract: We chemically restrained free-ranging coyotes (Canis latrans), red foxes (Vulpes vulpes), and raccoons (Procyon lotor) using medetomidine antagonized by atipamezole. All coyotes and 80% of red foxes were sedated with mean ± standard deviation doses of 0.12 ± 0.02 mg/kg and 0.14 ± 0.02 mg/kg medetomidine, respectively. Seventy-seven percent of raccoons were sedated with 0.21 ± 0.05 mg/kg medetomidine. In all species we observed occasional movement, muscle rigidity, and partial-arousal during sedation. Animals were alert within 4.3–8.6 ± 3.5–8.4 min following atipamezole at 0.4 mg/kg. Medetomidine and atipamezole provided safe handling in most animals and rapid recovery without use of a controlled substance. At these doses, biologists in the field should be prepared to administer a supplementary dose of medetomidine to some animals depending on ambient conditions and the objectives of the restraint event.  相似文献
