变应性哮喘是一种由辅助性T细胞(T helper cell,Th cell)调节的慢性炎症性疾病。Th1/Th2的失衡一直被认为是变应性哮喘的发病机制,Th2细胞及其分泌的细胞因子白介素4(interleukin 4,IL-4)、IL-5以及IL-13在变应性哮喘特异性症状的发病中发挥重要作用。最近研究发现Th17细胞及其分泌的IL-17参与变应性哮喘的发展过程,IL-23在Th17细胞维持生存和功能成熟中发挥重要作用,并参与抗原诱导的气道炎症反应。该文对目前IL-23/Th17轴在变应性气道炎症反应中的研究进展作一综述。  相似文献

Th17细胞是近年发现的一种新型CD4^+效应性T细胞,以其特异性的分泌IL-17而命名。介导免疫耐受的Treg细胞和介导炎症反应的Th17细胞间功能和分化过程相互对抗,在正常状态下,两者保持平衡,但机体发生功能异常时常表现出Treg/Th17失衡,引起炎性反应、自身免疫性疾病、移植物抗宿主病等的发生和发展,并决定疾病的转归和预后,在肿瘤免疫中亦发挥了重要作用。该文就Treg/Th17失衡在肿瘤,尤其是子宫颈癌发生发展中的作用进行综述。  相似文献

探讨IL-21/IL21R信号在炎症性肠病病变形成中对肠固有层辅助T细胞应答的调控机制。利用IL-21R基因敲除小鼠(IL-21R KO),建立TNBS诱导的炎症性肠病动物模型。监测小鼠体重及生存率;HE染色观察小鼠肠组织形态结构及炎症反应情况;分离和提取肠固有层淋巴细胞(lamina propria lymphocyte,LPL),应用流式细胞仪检测Th1、Th2、Th17、Treg细胞比例和绝对数;ELISA法检测LPL培养上清中的细胞因子(IFN-γ、IL-4、IL-17A、IL-10)的分泌。结果显示:与WT小鼠相比,IL-21R KO小鼠体重下降更快,生存率明显降低;HE染色显示IL-21R KO小鼠肠管上皮损伤严重、隐窝大面积消失、大量炎性细胞浸润,并侵犯到黏膜肌层;IL-21R KO小鼠肠固有层Th1细胞应答显著增强,相反Th2、Th17和Treg应答明显抑制。因此,IL-21/IL-21R信号通过调节小鼠肠固有层Th细胞应答,在TNBS诱导的肠炎病变形成中发挥重要的保护性作用。  相似文献

Th17细胞和Treg细胞是CD4+T细胞在不同细胞因子环境中分化出的新亚群,发挥不同的生物学效应,使机体的免疫系统处于平衡状态.Th17/Treg细胞失衡可引起一系列自身免疫性疾病.银屑病是与遗传、免疫异常有关的皮肤炎症性疾病,其发病机制尚不清楚.越来越多的研究发现,Th17细胞增多和Treg细胞减少及其分泌的细胞因子在银屑病的发病中有着重要作用.本文围绕这一机制综述了近年来有关Th17细胞、Treg细胞在银屑病发病机制中作用的研究,帮助我们更深入地了解银屑病的发病机制并为今后临床诊断和治疗提供依据.  相似文献

目的:探讨糖尿病患者外周血Th17细胞和Treg细胞数量以及相关细胞因子表达的变化。方法:以本院2013年1月至2015年12月收治的50例糖尿病患者为研究对象,其中1型糖尿病患者25例,2型糖尿病患者25例,同时以25例健康体检人群为正常对照,检测各组外周血Th17/Treg细胞数量以及血清IL-17、IL-10和TGF-β水平。结果:糖尿病患者外周血中Th17细胞比例均显著高于正常对照组(P0.05)而Treg细胞的比例均显著低于正常对照组(P0.05),且1型糖尿病患者Treg细胞比例显著低于2型糖尿病患者(P0.05)。糖尿病患者血清中IL-10和TGF-β水平均显著低于正常对照组(P0.05)而IL-17水平均显著高于正常对照组(P0.05),且1型糖尿病患者的IL-10水平显著低于2型糖尿病患者(P0.05)。结论:糖尿病患者体内Treg细胞数量及相关细胞因子低于正常而Th17细胞数量及相关细胞因子高于正常,这可能与患者体内的自身免疫失调有关。  相似文献

近几年来以分泌白介素17(interleukin 17,IL-17)为特征的辅助性T细胞Th17(T help cell 17,Th17)细胞被认为是有区别于Th1(T help cell 1,Th1)、Th2(T help cell 2,Th2)新型的细胞亚群,它的发现改变了以往人们只将Th细胞分为Th1、Th2的传统分类认识。Th17细胞参与了自身免疫疾病、肿瘤的发生及机体各种炎症的发病机制,其分泌的细胞因子在生物学功能中发挥了极其重要的作用。同时Th17细胞的活化需要各种转化生长因子、IL-6(interleukin 6,IL-6)、IL-23(interleukin 23,IL-23)等细胞因子的参与,活化的Th17细胞同时再进一步的促进各种细胞因子的分泌,以通过分泌IL-17、IL-21(interleukin 21,IL-21)、IL-22(interleukin22,IL-22)、IL-26(interleukin 26,IL-26)、肿瘤坏死因子(tumor necrosis factor,TNF)α等细胞因子导致机体炎症等各种疾病的发生。  相似文献

【目的】观察嗜酸乳杆菌完整肽聚糖(Whole peptidoglycan,WPG)对致敏脾淋巴细胞Th1/Th2及Treg/Th17平衡的体外调节作用。【方法】通过腹腔注射β-乳球蛋白(β-Lg)建立BALB/c小鼠牛乳过敏模型。造模成功后,分离致敏小鼠的脾淋巴细胞并分别与不同剂量的WPG共同孵育,酶联免疫法检测细胞上清液中抗体(总IgE和特异性IgE),Th1/Th2及Treg/Th17相关细胞因子(IFN-γ,IL-4,TGF-β,IL-17)水平,流式细胞术检测脾淋巴细胞中CD3+、CD4+和CD8+的百分含量,荧光定量PCR法检测过敏小鼠脾细胞中Th1/Th2及Treg/Th17相关转录因子T-bet、GATA-3、Foxp3和RORγt mRNA的表达量。【结果】WPG体外刺激致敏脾细胞后可显著抑制IgE的产生,上调CD3+、CD4+细胞数及CD4+/CD8+比值,下调Th2型因子(IL-4,GATA-3mRNA)和Th17型因子(IL-17,RORγt mRNA)的表达,且与过敏组相比,中、高剂量WPG的作用效果显著(P0.05);另外,WPG体外作用还上调了Th1型因子(IFN-γ,T-bet mRNA)及Treg型因子(TGF-β,Foxp3 mRNA)的表达,且具有剂量依赖性。【结论】嗜酸乳杆菌WPG体外刺激可有效纠正致敏脾淋巴细胞的Th1/Th2及Treg/Th17失衡。  相似文献

目的:探讨复发性自然流产患者外周血中Th17、Treg细胞以及相关细胞因子的变化。方法:选择复发性自然流产患者25例,正常妊娠妇女25例以及正常非妊娠育龄妇女25例,流式细胞术测定外周血中Th17和Treg细胞数量,ELISA法测定血清IL-10、TGF-β及IL-17的浓度。结果:复发性自然流产患者外周血中Th17细胞百分率显著高于正常妊娠以及正常非孕妇女(P0.05),Treg细胞百分率显著低于正常非孕妇女(P0.05),但与正常妊娠妇女相比无显著性差异(P0.05);复发性自然流产患者外周血IL-10及TGF-β水平显著低于正常妊娠妇女以及正常非孕妇女(P0.05),而IL-17水平显著高于正常妊娠妇女以及正常非孕妇女(P0.05)。结论:外周血Th17细胞数和IL-17水平的升高以及抑制性细胞因子IL-10和TGF-β水平的下降可能是复发性自然流产发生的重要原因。  相似文献

目的:Th17/Treg免疫失衡在原发性胆汁性胆管炎(PBC)的发病机制中起到关键作用。自噬是调节T细胞稳态的重要环节。本研究旨在探索自噬对PBC患者Th17/Treg免疫失衡的影响。方法:选取20例初次诊断的PBC患者,20例健康对照者,收集其外周血单个核细胞(PBMCs)。利用流式细胞术检测PBC患者与健康对照者PBMCs中Th17和Treg细胞分布及其胞内自噬的水平,进一步体外利用氯喹抑制自噬,证实自噬对PBC患者Th17/Treg平衡的影响。结果:相较于健康对照者,PBC患者Th17细胞占CD4~+T比例上升(P0.05),且血清中IL-17含量较高(P0.01);Treg细胞比例下降(P0.05),且血清中TGF-β含量较低(P0.01)。而PBC患者Th17细胞和Treg细胞内自噬水平均升高。另外,体外实验结果证实,自噬抑制剂氯喹能够有效地恢复PBC患者PBMCs中Th17/Treg平衡。结论:异常活化的自噬可导致PBC患者Th17/Treg免疫失衡,抑制自噬能够在体外恢复此平衡。因此,对异常自噬的干预可能将成为PBC疾病治疗的新方法。  相似文献

复发性流产作为困扰育龄期女性的常见病,病理机制十分复杂,其中,约有50%原因不明,为不明原因复发性流产,多与免疫因素相关。Th17与Treg细胞作为CD4+T细胞的重要亚群,是维持妊娠母胎免疫耐受平衡的重要因子。肠道菌群广泛参与机体的内分泌、免疫反应和营养代谢等,近年来作为新兴领域已被许多学者证明与妊娠相关疾病的发生密切相关。研究表明,肠道菌群可以调控Th17/Treg细胞的分化与平衡,而肠道菌群失调则会导致Th17/Treg失衡,进而影响正常妊娠,导致不良的妊娠结局。目前,基于肠道菌群调控下的Th17/Treg细胞失衡与不明原因复发性流产的相关研究较少,现从肠道菌群对Th17/Treg细胞平衡的调节和Th17/Treg细胞失衡导致不明原因复发性流产出发,提出Th17/Treg细胞失衡相关的不明原因复发性流产可能与肠道菌群存在密切联系的假说,为免疫相关不明原因复发性流产的病因学研究和治疗提供新的靶向。  相似文献

BackgroundTh1/Th17 imbalance had been indicated to mediate several kinds of inflammatory diseases. We deduce that Th1/Th17 imbalance might also contribute to the pathogenesis of acute graft-versus-host disease (GVHD). This study is to investigate the relation between Th1/Th17 imbalance and acute GVHD.MethodsWe applied a murine GVHD model of C57BL/6 (H-2b) donor to BALB/c (H-2d) recipient by treating the recipients with low dose of halofuginone (HF), which is competent in selectively inhibiting Th17 differentiation and facilitating Th1 differentiation. Recipient mice were monitored for survival rate, body weight change, clinical symptoms and pathological evidence of acute GVHD. We also measured the proportions of Th1 and Th17 cells in circulation and expression levels of IFN-γ and IL-17A in tissues involved in GVHD.ResultsFirstly, we confirm the existence of Th1/Th17 imbalance in acute GVHD and Th1/Th17 imbalance positively correlates with severity of acute GVHD. Secondly, low dose of HF augments Th1/Th17 imbalance by driving the Th1/Th17 balance to a Th1-dominant reaction. Finally, augmented Th1/Th17 imbalance leads to aggravated systemic GVHD. An increased Th1-type reaction results in aggravated hepatic and intestinal GVHD, and inhibiting Th17 differentiation is sufficient to alleviate pulmonic impairment.ConclusionOur study is indicative for a critical role of Th1/Th17 imbalance in the pathogenesis of murine GVHD.  相似文献

Autoimmune diseases, including arthritis, often result from an imbalance between regulatory T (Treg) cells and IL-17-producing (Th17) cells. Dozens of studies in mice and humans have shed light on the pathological significance of T cells in RA. Since Th17 cells play an important role in the exacerbation of inflammation and bone destruction in joints, it has been an important issue how arthritic Th17 cells arise. Th17 cells are generated in the local inflammatory milieu via cytokines produced by macrophages or synovial fibroblasts, while it is reported that Th17 cells are generated in the gut in the presence of specific commensal bacteria. A recent report showed a pathogenic Th17 cell subset with a distinct pattern of gene expression and a potent osteoclastogenic ability are converted from Foxp3+ T cells in arthritic joints. Since Foxp3+ Treg cells contain T cells which recognize self-antigens, the fate of plastic Foxp3+ T cells can be a critical determinant of autoimmunity or self-tolerance. Further analysis on the molecular basis and antigen-specificity of arthritogenic Th17 cell subsets will be helpful to establish novel therapeutic approaches and clarify how self-tolerance breaks down in autoimmune arthritis.  相似文献

目的探究子宫内膜异位症患者免疫调节Th17细胞及Treg细胞的表达意义。 方法选取2017年1月至2018年12月青岛大学附属医院收治患有子宫内膜异位症的患者,为子宫内膜异位组(EMT组),选取同一时期在医院因不孕不育进行腹腔镜检查的患者,为正常组(NM组),两组分别56例。EMT组和NM组患者在一般资料上差异无统计学意义。通过流式细胞仪、HE染色法、qRT-PCR法、ELISA法分析EMT组和NM组患者Th17、Treg细胞所占比例、子宫内膜组织病变情况、ROR-γt、Foxp3 mRNA表达含量的差异性来探究子宫内膜异位症患者Th17细胞及Treg细胞变化。实验结果用 ±s表示,并采用独立样本t检验进行比较。 结果EMT组患者CD4+ T细胞中Th17所占比例为5.48±2.81,Treg所占比例为4.22±1.04,NM组Th17所占比例为2.34±1.01,Treg所占比例为6.14±1.52,差异均有统计学意义(t = 7.869,3.015,P = 0.014,0.026)。EMT组患者血清中IL-17水平为(256.38±34.15)?pg/ ml、IL-22为(67.48±10.89)?pg/ml,NM组患者血清中IL-17水平为(198.04±27.59)?pg/ml、IL-22为(43.78±6.92)?pg/ml,差异均有统计学意义(t = 9.944,4.689,P = 0.008,0.017)。EMT组患者血清中IL-10水平为(18.56±4.77)?pg/ml、TGF-β为(148.28±40.52) pg/ ml,NM组患者血清中IL-10水平为(28.35±6.07)pg/ml、TGF-β为(204.78±19.87)pg/ml,差异均有统计学意义(t = 9.491,2.849,P = 0.012,0.034)。EMT组患者子宫内膜组织形态不规则,多数细胞不完整,破损或缺失,且炎性细胞增多,在其周围聚集。NM组患者子宫内膜组织形态规则,细胞没有明显破损或缺失,未见细胞周围炎性因子增多。qRT-PCR检测结果显示,EMT组和NM组ROR-γ mRNA分别为2.89±0.76、1.71±0.26,EMT组和NM组Foxp3 mRNA分别为2.25±0.34、1.13±0.18,两组差异均有统计学意义(t = 10.996,6.759,P = 0.006,0.011)。 结论子宫内膜异位症患者外周血免疫调节细胞Th17/Treg平衡失调,免疫调节紊乱与子宫内膜异位发生、发展有密切关系。  相似文献

Purpose: Sulfur mustard (SM) lung is a heterogeneous disease associated with abnormal inflammatory immune responses. The Th17/Treg axis imbalance is associated with the pathogenesis of chronic inflammatory pulmonary disease. We aimed to determine the distribution of different Th17 and Treg cells in patients with SM lung and chronic obstructive pulmonary disease (COPD) and evaluate the clinical implications in this homeostasis. Methods: In this analytical cross-sectional study, CD4?+?Foxp3+?Treg and CD4+?IL-17+?Th17 cells were measured in peripheral blood mononuclear cells (PBMCs) and transbronchial biopsy (TBB) samples of 15 SM-exposed patients, 12 COPD and 13 healthy controls (HCs). The potential correlation between the ratio of Th17/Tregs and lung function was evaluated with multivariate logistic regression (MLR) analysis. Results: The frequency of CD4?+?FoxP3+?Tregs and CD4?+?IL-17+?Th17 was increased ~1.7-fold (8.71/4.95) and ~2.7-fold (1.028/0.371) respectively, in the PBMC of SM patients compared with the health controls (p?<?0.001). The results indicated that there were increases in the frequency of Th17 and Tregs cells in the patients with COPD versus the HC, that is, ~2.6-fold (0.987/0.371) and ~1.4-fold (7.12/4.95), respectively; but they did not reach to SM level (p?≥?0.05). Moreover, in the TBB samples, the CD4?+?IL-17+?Th17 and CD4+?FoxP3+?Tregs numbers were significantly higher in SM and COPD patients than HC (p?<?0.05). The Th17 and Treg cells were inversely correlated with forced expiratory volume in 1s (FEV1%) (r?=??0.351, p =?0.001; r?=??0.344, p?=?0.021) and FEV1/FVC (r?=??0.44, p?=?0.001; r?=??0.302, p?=?0.011), respectively. Instead, positive correlations were found between Treg/Th17 ratios and forced FEV1%pred (r?=?0.156, p?=?0.007), as well as FEV1/FVC ratio (r?=?0.334, p?=?0.006). Conclusions: The imbalance of Th17/Treg has a key role in immunopathogenesis of chronic phase of mustard lung disease.  相似文献

CD4+ T cells differentiate into subsets that promote immunity or minimize damage to the host. T helper 17 cells (Th17) are effector cells that function in inflammatory responses. T regulatory cells (Tregs) maintain tolerance and prevent autoimmunity by secreting immunosuppressive cytokines and expressing check point receptors. While the functions of Th17 and Treg cells are different, both cell fate trajectories require T cell receptor (TCR) and TGF-β receptor (TGF-βR) signals, and Th17 polarization requires an additional IL-6 receptor (IL-6R) signal. Utilizing high-resolution phosphoproteomics, we identified that both synergistic and additive interactions between TCR, TGF-βR, and IL-6R shape kinase signaling networks to differentially regulate key pathways during the early phase of Treg versus Th17 induction. Quantitative biochemical analysis revealed that CD4+ T cells integrate receptor signals via SMAD3, which is a mediator of TGF-βR signaling. Treg induction potentiates the formation of the canonical SMAD3/4 trimer to activate a negative feedback loop through kinases PKA and CSK to suppress TCR signaling, phosphatidylinositol metabolism, and mTOR signaling. IL-6R signaling activates STAT3 to bind SMAD3 and block formation of the SMAD3/4 trimer during the early phase of Th17 induction, which leads to elevated TCR and PI3K signaling. These data provide a biochemical mechanism by which CD4+ T cells integrate TCR, TGF-β, and IL-6 signals via generation of alternate SMAD3 complexes that control the development of early signaling networks to potentiate the choice of Treg versus Th17 cell fate.  相似文献
