The tumour cells and the reaction cells in Sarcoma 37 ascitic fluids have been studied in thin sections with the electron microscope. The reaction cells were either leucocytes or much larger acidophilic peritoneal cells of the same dimensions as the tumour cells; the peritoneal cells formed as much as 20 per cent of the large cell population. The fine structure of the cells is described and some new observations recorded. It has been found that the cell membrane of eosinophil granulocytes has a laminated composition and the characteristic granules of these cells a double limiting membrane. The pores in the double nuclear membrane of the peritoneal cells have been observed to have a fine line running across them. In the tumour cells, a rounded granular body with a central dense area has been found in the region of the centrosome; these cells were also seen to contain rows of parallel smooth surfaced cisternae lying 150 mµ apart similar to those hitherto only observed in spermatids. There was a feltwork of fine filaments in the cytoplasm of the centrosome region of the tumour cells. The cytoplasmic fine structure underlying the basophilia of the tumour cells and the acidophilia of the peritoneal cells is compared and discussed.  相似文献

Summary The morphology and fine structure of aleurone cells of soybean [Glycine max (L.) Merr.] seed coats were analyzed with transmission electron microscopy for the period of rapid seed fill up to physiological maturity. Thin sections and freeze-fracture replicas were prepared for each stage. The aleurone is a tissue lining the embryo sac and consists of a single layer of cells attached to the aerenchyma of the seed coat proper. During seed fill, aleurone cells contained numerous Golgi-derived vesicles in the basal region of the cytoplasm that were either free or attached to the plasma membrane along the lateral and basal regions of the cell wall. Correspondingly, the Golgi apparatus were well developed with individual dictyosomes having 5 to 8, highly fenestrated stacked cisternae. The degree of fenestration along the periphery of each cisterna increased from the cis to trans region. Rough endoplasmic reticulum (RER) was also abundant, often consisting of up to 30, stacked swollen cisternae which occupied large regions of cytoplasm. Plasmodesmata which connected adjacent aleurone cells was not observed along the dorsal walls of aleurone cells that faced aerenchyma. At physiological maturity, dictyosome cisternae were less fenestrated and had fewer associated secretory vesicles. Stacked lamellae of RER were absent, being replaced by short tubular cisternae and small vesicles. At physiological maturity, the aleurone cells had thick walls, and contained numerous lipid bodies in apposition to the plasma membrane. The cytoplasm appeared densely stained in thin-sections and contained protein bodies and amyloplasts with large starch grains. We conclude that during the period of rapid seed fill aleurone cells produce, package, transport and secrete vesicular contents toward the embryo, that is followed at physiological maturity by the storage of lipid, protein and starch in the same cells. The embryo is the most likely destination for secretory products during the period of rapid seed fill. The fate of the stored food reserves in aleurone cells at physiological maturity may be analogous to that of aleurone tissue of grasses, being utilized during imbibition for processes important to germination.  相似文献

The fine structure of subsurface cisternae and lamellar bodies in the outer hair cells of the guinea-pig organ of Corti was studied with thin sections and freeze-fracture replicas. Subsurface cisternae in the outer hair cells consist of multilayers along the lateral plasma membrane of the cell. The outermost layer is a flattened cistern in the upper part of the supranuclear region, but comprises a series of tubules in the lower part. Deeper layers are fenestrated cisternae in which disc-like areas are found in the upper part of the supranuclear region. Lamellar bodies consist of concentric layers of fenestrated cisternae and are located in the apical cytoplasm beneath the cuticular plate. They are continuous with the subsurface cisternae. In the supranuclear cytoplasm, bulges of the subsurface cisternae and the lamellar bodies are found. Dilated cisternae are also present. Some dilated cisternae contain many small vesicles, which display acid phosphatase activity. The dilated cisternae are considered as forms of the bulges undergoing transformation into multivesicular bodies. The possible role of the lamellar bodies, and the origin and fate of the subsurface cisternae are discussed.  相似文献

Cellular autophagocytosis was observed in mouse seminal vesicle cells incubated in vitro up to 8 h in medium 199 or Krebs-Ringer bicarbonate buffer. During the first 2 h of incubation, early forms of autophagic vacuoles were seen in the cells, advanced forms containing degraded material began to cumulate later. After 6--8 h, early vacuoles occurred sparsely, while advanced forms were detected in a great number. During the first 2 h of incubation, we often observed smooth surfaced membrane pairs between the cisternae of rough surfaced endoplasmic reticulum resembling isolating membranes of autophagic vacuoles. They varied in size and shape from short, straight cisternae to long, curved ones, almost completely encircling areas of the cytoplasm. Based on these observations, we propose a tentative scheme of the formation of autophagic vacuoles, viz., the short, straight cisternae would represent the first stage in the development of an autophagic vacuole, while the curved sack-like forms are interpreted as successive steps leading to the complete sequestration of an area of the cytoplasm.  相似文献

An anatomical analysis of the chicken trigeminal ganglion was made using light microscopy on specimens prepared by usual chemical fixation or freeze-drying methods and by electron microscopy. Two types of neurons were consistently seen, dark and light cells. Dark cells contained a dense cytoplasm with Nissl substance distributed evenly throughout, whereas light cells had a less dense cytoplasm containing clumps of Nissl substance. The Nissl bodies in light cells contained only a few small cisternae of granular endoplasmic reticulum as compared with many stacked cisternae in Nissl bodies of dark cells. The ratio of dark to light cells was approximately 62:38 in all regions of the ganglion. Dark cells were consistently smaller than light cells. In the seven-day old chick, the mean diameters of the dark and light neurons were 21.4 μ and 29.5 μ respectively; in the adult the values were 29.9 μ and 39.7 μ respectively. It is concluded that the dark and light cells belong to two distinct neuronal cell populations.  相似文献

The lymphoma cells from a patient with leukemia lymphoblastic sarcoma (Kiel classification) were observed by scanning and transmission electron microscopy. These cells were also examined by E, EA, EAC rosette-formation tests and by the indirect immunofluorescence technique for surface immunoglobulins. The malignant cells showed failure of rosette-formation or absence of surface immunoglobulins. Scanning electron microscopy revealed that many uniform protrusions were present on the cell surfaces. These surface protrusions were different from those seen on E-or EAC-rosette-forming cells. Ultrastructurally, the malignant cells were characterized by long profiles of rough surfaced endoplasmic reticulum with regular, narrow cisternae which radiated from Golgi area to the periphery of cytoplasm. These appearances differed from those observed in T-or B-lymphoma cells.  相似文献

Summary Using 0.5 thick (i.e., semi-thin) and conventional thin sections, observations have been made on the localization of acid phosphatase in the Golgi apparatus and related structures in the pars recta of rat kidney. In thin sections one or two Golgi cisternae located at the concave (basilar) aspect of the stack had enzymic activity. The periphery of these cisternae may be fenestrated. Coated vesicles were seen apparently free in the cytoplasm and in continuity with both reactive Golgi cisternae and smooth tubular elements. Smooth vesicular profiles with electron-lucent matrices and low enzymic activity were seen, apparently free, in the central Golgi zone. Semi-thin sections demonstrated more fully the extent of the reactive Golgi elements, their architecture and their relationships with other organelles. Within a stack the reactive Golgi cisternae were continuous with one another. A network of anastomosing tubular elements formed the periphery of the cisternae and linked some adjacent cisternae. Tubules extended considerable distances from this network in apical and lateral directions. Vesicular protuberances formed ends to the tubular extensions but free vesicles were not obvious. Apparent continuity was seen between reactive tubules and dense bodies (secondary lysosomes). Vesicular profiles with electron-lucent matrices andlow enzymic activity appeared to be continuous with the periphery of reactive Golgi cisternae and may represent the formation of primary lysosomes. This study demonstrates that semi-thin sections could be used to great advantage in the study of organelle interactions in both normal and pathological states.  相似文献

The ultrastructure of the storage parenchyma cells of the cotyledons of developing bean (Phaseolus vulgaris L.) seeds was examined in ultrathin frozen sections of specimens fixed in a mixture of glutaraldehyde, formaldehyde and acrolein, infused with 1 M sucrose, and sectioned at-80° C. Ultrastructural preservation was excellent and the various subcellular organelles could readily be identified in sections which had been stained with uranyl acetate and embedded in Carbowax and methylcellulose. The cells contained large protein bodies, numerous long endoplasmic reticulum cisternae, mitochondria, dictyosomes, and electron-dense vesicles ranging in size from 0.2 to 1.0 m. Indirect immunolabelling using rabbit immunoglobulin G against purified phaseolin (7S reserve protein), and ferritin-conjugated goat immunoglobulin G against rabbit immunoglobulin G was used to localize phaseolin. With a concentration of 0.1 mg/ml of anti-phaseolin immunoglobin G, heavy labeling with ferritin particles was observed ober the protein bodies, the cisternae of the endoplasmic reticulum, and the vesicles. The same structures were lightly labeled when the concentration of the primary antigen was 0.02 mg/ml. Ferritin particles were also found over the Golgi bodies. The absence of ferritin particles from other organelles such as mitochondria and from areas of cytoplasm devoid of organelles indicated the specificity of the staining, especially at the lower concentration of anti-phaseolin immunoglobulin G.Abbreviations ER endoplasmic reticulum - IgG immunoglobulin G  相似文献

An attempt is made to correlate fine structure with the histochemical reactions of the epidermis in the synbranchiform fish Monopterus cuchia. Three sources of mucus are identified. Superficial epithelial cells produce weakly acidic glycoprotein which is secreted at the surface as the external mucous layer or cuticle. Numerous large unicellular mucous glands have a secretion which is strongly acidic and sulphated, although the basal and peripheral parts of these cells, which contain most of the rough endoplasmic reticulum, react strongly for neutral glycoprotein; Golgi cisternae appear to be involved in a change of histochemical reaction from neutral to strongly acidic as the secretion is formed. A second, slender, type of mucous gland cell, not previously reported, gives a weaker reaction for sulphated acidic glycoprotein and has cytoplasm with numerous Golgi cisternae and free ribosomes, producing electron–dense secreted drops. Sacciform cells, with a protein–aceous secretion, have a characteristic fine structure with membranous "bubbles" at the surface of the cytoplasm. Free alveolar macrophages of normal mouse lung have been studied in the electron microscope. The tissue was obtained from several young adult white mice. One other animal was instilled intranasally with diluted India ink 1½ hours prior to the removal of the lung. Thin sections of the osmium-fixed, methacrylate-embedded tissue were examined either in an RCA EMU 2 electron microscope or in a Siemens and Halske Elmiskop I b. A few thick sections obtained from the same embeddings were stained for iron. The normal alveolar macrophages, which are usually in contact with the alveolar epithelium, were found to contain a variety of inclusion bodies, along with the usual cytoplasmic components like mitochondria, endoplasmic reticulum, and Palade granules. Another typical component of the cytoplasm of these cells which appears as small (~6 mµ) very dense granules of composite fine structure is interpreted as ferritin. It is assumed that this ferritin is formed from red blood cells ingested by the alveolar macrophages. The macrophages in the alveoli were found to phagocytize intranasally instilled India ink particles. Such cells, with engulfed India ink particles, were often of more rounded form and the particles were frequently seen lying inside membrane-bound vacuoles or vesicles of the cytoplasm. The membrane of a few vesicles containing India ink particles was seen as the invaginated portion of the cell plasma membrane, and in one instance these same vesicles were seemingly interconnected with a rough surfaced cisterna of the endoplasmic reticulum. The process of phagocytosis is recognized as related to the "normal" process of pinocytosis.  相似文献

High voltage electron microscopy and computer axial tomography have been used to study the 3-D structure of trans-Golgi cisternae and trans- Golgi networks (TGNs) in NRK cells. Both structures were specifically labeled by photoconversion of a fluorescent analogue of ceramide using a modification of the techique of Pagano et al. (J. Cell Biol. 1991. 113: 1267-1279). Regions of the Golgi ribbon in fixed, stained cells were cut in 250-nm sections and analyzed by tilt series microscopy and subsequent tomographic reconstruction. Resolution of the reconstructions ranged from 6 to 10 nm. The size and structure of the TGN varied considerably throughout the Golgi ribbon; all reconstructions were made from regions with pronounced TGN. Most regions analyzed contained multiple (2-4) Golgi cisternae that stain with ceramide. These "peel off" from the closely stacked cisternae and are continuous at their ends with tubules that contribute to the TGN. Most vesicular profiles visualized in the TGN are connected to TGN tubules. The budding of vesicles appears to occur synchronously along the length of a TGN tubule. Two distinct coats were visualized on budding vesicles: clathrin cages and a novel, lace-like structure. Individual TGN tubules produce vesicles of only one coat type. These observations lead to the following predictions: (a) sorting of molecules must occur prior to the formation of TGN tubules; (b) vesicle formation takes place almost synchronously along a given TGN tubule; and (c) lace-like coats form an exocytic vesicles.  相似文献

Summary The ultrastructure of coleoptile cells of ungerminated rice grains has been examined following fixation in glutaraldehyde and osmium tetroxide. In many respects the cell structure resembles that reported for other dormant seed tissues: the cells contain protein bodies and lipid droplets, mitochondria and plastids show little internal structure but cytoplasm invaginates into many plastids; golgi cisternae cannot be discerned. Rough ER is present as cisternae surrounding protein bodies, as occasional regions of parallel layers, and in concentric whorls where it alternates with smooth paired membranes of an unknown nature. The ribosomes on the ER are at least partly arranged into regular rows. Various crystalline, presumably proteinaceous, inclusions lie in the groundplasm, plastids and nuclei.  相似文献

Summary Morphology, occurrence, and distribution of dilated cisternae of the endoplasmic reticulum (ER) were studied by electron microscopy. The cisternae which contained an electron-dense matrix were intimately associated with the granular ER membranes appearing as tubular necks at the edges of the ER profiles. After budding off from the ER the cisternae still had ribosomes attached to the outside of the bounding membranes. The accumulations were variable in shape, being 0.4 to 1.5 in width and 4 to 5 in length.The cisternae were found to be unique for plants of theCruciferae and could not be observed in species from related families such asPapaveraceae andResedaceae.The dilated cisternae were a common component of the cytoplasm in root tips, stems, and leaves. In meristematic cells the number of accumulations was small but increased in older differentiating cells of the root cap. The similarity to microbodies described by previous authors from other plants is discussed.  相似文献

Fine structure and stereo-images of the Golgi apparatus and endoplasmic reticulum (ER) in the subcommissural organ (SCO) cells were visualized by the application of zinc-iodide osmium tetroxide (ZIO) impregnation, conventional electron microscopy and high voltage electron microscopy (HVEM). The Golgi apparatus in the SCO cells of rats, gerbils and hamsters consisted of flattened saccules stacked in parallel array. It showed a selective staining toward ZIO mixture and might form a complex network of tubular structures because of the presence of numerous fenestrations in the flattened Golgi saccules. The cytoplasm of the SCO cells in the rat and gerbil was crowded by dilated cisternae of the ER with a few flattened profiles. In the hamster SCO cells, however, the dilated cisternae of the ER were not observed. Flattened cisternae of ER in all species studied showed a positivity for ZIO impregnation and formed a complex tubular network, whereas dilated cisternae of the ER in the rats and gerbils did not show any reactivity. It was thus determined that the observation of thin and thick sections selectively stained with appropriate reagent for defined cellular organelles under conventional electron microscopy and HVEM offered valuable information about three-dimensional organization of the cell. A definite species-specific variation of SCO ultrastructure and cytochemistry was also demonstrated.  相似文献

Summary Yoshida sarcoma cells in four days after inoculation have been studied with the electron microscope in thin sections. The mitochondria found attached to the nuclear membrane in the prophase look to serve a removal of nuclear membrane. In the telophase, the mitochondria are not found in the juxtanuclear region of the cytoplasm, while the smooth-surfaced endoplasmic reticulum makes its appearance surrounding the surface of nucleus. These findings suggest that the smooth surfaced endoplasmic reticulum participates directly in the development of the nuclear membrane. Single or double particles 23–60 m in diameter are found at the nuclear terminal of the double layer of endoplasmic reticulum. The development of the nuclear membrane may be ascribed to such structural elements.  相似文献

Summary The connections and structural inter-relations of dictyosomes and endoplasmic reticulum (ER) in cotyledons of germinating mung beans were studied using thick (0.3 m) sections of aldehyde fixed, zinc iodide-osmium tetroxide (ZIO) impregnated tissue. The sections were examined by conventional (100 kV), rather than high voltage, transmission electron microscopy.Continuity of cisternal ER with tubular ER was confirmed and a direct connection of tubular ER totrans dictyosome cisternae was observed as were GERL networks associated withtrans dictyosome cisternae.Dictyosomes also gave rise to an extensive system of very fine tubules (10–20 nm diam) which have not been described previously in plant tissue. These tubules, which originated at thetrans dictyosome face, extended throughout the cytoplasm and were found connected to cisternal ER and tubular ER.The implications of these observations are discussed with regard to present ideas concerning endomembrane flow and protein sorting by the Golgi apparatus.  相似文献

The ultrastructure and histochemistry of the refractile, vesiculate cells (“blasenzellen,”“cellules secretrices,”“gland cells”) of Antithamnion defectum Kylin were examined. The refringent vacuolar contents disclosed two components of differing density: an electron opaque, proteinaceous matrix material surrounding cores of irregularly shaped, less opaque material. The cores contain less protein and more unknown material than the matrix. Part or all of the vacuolar material is synthesized by abundant rough endoplasmic reticulum (ER) and deposited in smooth surfaced cisternae that swell to form vesicles. Mitochondria are usually associated with stacks of the swelling cisternae. The vesicles enlarge by continued deposition of synthesized material and coalescence with other vesicles. All vesicles eventually coalesce to form the mature vacuole. A crystalline array of fibrils develops in the cytoplasm during later stages of vacuole enlargement. The crystal contains a sulfated, acidic polysaccharidic material. The chloroplasts, if present, and nucleus degenerate at vacuole maturity. Active release of the vacuolar material does not occur, and organelles for extracellular secretion are not present. Structural evidence suggests a storage, rather than secretory, function for the cells.  相似文献
