Laboratory studies were carried out in the Department of Biological Sciences, Ogun State University, Ago-Iwoye, southwestern Nigeria, to determine the extent of fungal deterioration of melon seeds stored in two types of storage bags viz; jute and polyethylene bags. Melon seeds of varieties Tc139 and V2 were stored in jute and polyethylene bags under ambient conditions using the 2 × 2 factorial design (variety vs type of bag) for 12 months. The moisture content (mc), incidence of visible mouldiness (ivm) and germinability of the stored seeds were determined monthly. The mc of Tc139 ranged from 6.1 to 6.7% in jute and 6.2 to 6.5% in polyethylene bags. The ivm which was initially 2.1% increased to 10.7% and 5.5% in jute and polyethylene bags respectively, after 12 months in storage. The germination percentage decreased from 96.3% to 28.7% and 45.3% in jute and polyethylene bags, respectively. The mc of V2 stored in jute and polyethylene bags varied from 5.9 to 6.4%, and 5.8 to 6.2%, respectively. The ivm increased from 1.8% before storage to 8.9% and 4.8% in jute and polyethylene bags, respectively, after 12 months. The percentage seed germination declined from 98.0%to 37.3% in jute and 48.7% in polyethylene bags after 12 months. Decreased incidence of field fungi namely: Alternaria, Botryodiplodia theobromae, Cladosporium, Fusarium and Macrophomina phaseolina was accompanied by simultaneous increase in storage fungi viz: Aspergillus, Penicillium, and Rhizopus with prolonged storage. Entomogenous Fungi as Promising Biopesticides for Tick Control   总被引:6,自引:0,他引:6  
When ticks were sealed in nylon tetrapacks and infected with the entomogenous fungi, Beauveria bassiana and Metarizium anisopliae and maintained in potted grass in the field, the fungal oil formulations (109 conidia per ml) induced 100% mortality in larvae of Rhipicephalus appendiculatus and Amblyomma variegatum, whereas mortalities in nymphs varied between 80–100% and in adults 80–90%. The aqueous formulations (109 conidia per ml) induced mortalities of 40–50% and reductions in egg hatchability of 68% (B. bassiana) and 48% (M. anisopliae) when sprayed on Boophilus decoloratus engorging on cattle. The strains of B. bassiana and M. anisopliae isolated from naturally infected ticks were also found to induce high mortalities in both R. appendiculatus and A.variegatum in tetrapacks placed in potted grass. Both aqueous and oil-based formulations were found to be effective, although the latter induced higher mortalities. These fungal strains in aqueous formulation (108 conidia per ml) suppressed on-host populations of adult R. appendiculatus by 80% (B. bassiana) and 92% (M. anisopliae) when sprayed on tick-infested grass once per month for a period of 6 months. The feasibility of using entomogenous fungi for tick control in the field is discussed.  相似文献

Fungal formulations are vital for effective biopesticide development. Good formulations help to optimise field efficacy, while poor formulations result in product failure. This study aimed to produce a hydrophobicity test that would be appropriate for fungal conidia produced to a commercial quality and determine relative hydrophobicity of fungi from four different genera by using laser diffraction. A particle size analyser was used to determine the hydrophobicity of: three Metarhizium acridum samples, M. anisopliae, Beauveria bassiana, Trichoderma stromaticum, T. harzianum, T. viride and Alternaria eichhorniae conidia by suspending the conidia in three different liquids: Shellsol T (a mineral oil), water and 0.05 % Tween 80. Hydrophobicity was determined by the size of the particles formed in each of the liquids. All the Metarhizium samples were the most hydrophobic followed by B. bassiana and A. eichhorniae. The Trichoderma samples were the least hydrophobic. As a comparison, a phase exclusion assay and a salt-mediated aggregation and sedimentation (SAS) test were performed. It was not possible to get a reliable reading for the B. bassiana, A. eichhorniae and T. viride samples using the phase exclusion assay. The addition of salt in the SAS test did not affect the rate of sedimentation. It was hypothesised that conidia size affected the results of the SAS test that made A. eichhorniae the most hydrophobic conidia. Particle size analysis (PSA) was a more accurate test for comparing fungi from difference genera compared to the SAS test and phase exclusion assay. PSA was also used to test three emulsions and demonstrated that different formulations had an effect on particle size.  相似文献

Mexican fermented maize dough, pozol, including traditional banana leaf-wrapped samples and material in plastic bags, was purchased. All samples were pH 4.7 to 5.7 approx. 12 h after preparation, pH declining to 3.6 to 3.9 after 6 to 9 days storage at ambient temperature. These latter samples had dry matter contents of 31% to 48% (w/w), 0.35% to 0.75% titratable acidity as lactic acid and lactic acid bacteria as predominant microbial flora at about 108 c.f.u./ml. The lactic acid bacteria included strains of Leuconostoc mesenteroides, Lactobacillus plantarum, Lactobacillus confusus, Lactococcus lactis and Lactococcus raffinolactis. Fungi were not found in the samples stored in plastic bags. The samples wrapped in banana leaf, however, developed a large surface mycoflora within 2 days. This included Geotrichum candidum, yeasts and moulds. The majority of the lactic acid bacteria and approx. 50% of yeasts hydrolysed starch to some extent. No Geotrichum isolate hydrolysed starch. Lactate was assimilated by all the Geotrichum isolates and by 17 of 39 yeast strains.  相似文献

In this study, the mobilization and further translocation of phosphorus from conidia of saprotrophic fungus Trichoderma virens into Pinus sylvestris seedlings by nondestructive measuring of 32P was assessed. The radioactive phosphorus flux from the conidia to the Scots pine seedlings forming mycorrhiza with Laccaria laccata and Suillus bovinus amounted up to 27.82% and 7.42%, respectively, on the 28th day of the experiment, while at the same time in nonmycorrhizal pine seedlings, the detected radioactivity reached only 0.56%. Our studies revealed that both ectomycorrhizal fungi: L. laccata and S. bovinus, mobilized the phosphorus from radioactive conidia of T. virens. On this basis, we conclude that activities of the mycosymbionts may facilitate absorption and further translocation of phosphorus from organic matter into the host plants.  相似文献

Wood-based composites such as oriented strand board (OSB) are principle framing elements in building construction in North America. However, these materials are often affected by moulds in wet or humid environmental conditions. A common control method for prevention of mould growth on panels is preservative treatment of panels with various pesticides or chemicals. In recent years, environmentally friendly pest control methods are required because of environmental issue. This research aimed to develop a biological technology to protect OSB against mould infection by post-treatment of panels with natural extracts from fungal antagonists. In this study, the culture metabolites of a fungal antagonist, Phaeotheca dimorphospora DesRochers & Ouellette, were extracted, and the antibiotic activity of the extracts was tested in Petri plates against various moulds and decay fungi. The OSB panels were then dip-treated with the extracts and exposed to a humid environment for mould growth testing in a period of 8 weeks. The results showed that the mycelia growth of all fungi tested (moulds, white-rot and brown-rot fungi) was inhibited by the extracts of P. dimorphospora on agar plates. Panel samples dipped with the fungal extracts in acetone got little mould growth on them, whereas untreated control panels and those samples treated with acetone alone were seriously affected by various moulds.  相似文献

The sterile insect technique has been used for more than 50 years to control a range of insects around the world. Sterile insect technique is rapidly becoming a major component of many area‐wide fruit fly management programmes. Irradiation of immature life stages induces sterility in adults, which are then distributed over large areas to mate with wild flies, resulting in no viable offspring. However, irradiation in normal air results in declining adult quality. To optimize the quality of sterile adult flies, several techniques are available to lower the levels of oxygen in fruit fly tissues prior to irradiation. The simplest method is to seal pupae in plastic bags and allow the oxygen consumption of pupae to minimize oxygen in both the air and pupal tissue. Some fruit fly species have rapid decreases in eclosion as a result of low oxygen atmospheres. We tested the tolerance of Queensland fruit fly, Bactrocera tryoni (Froggatt) (Diptera: Tephritidae), to low oxygen for the first time. In the first two experiments, unirradiated B. tryoni pupae were tested for different periods in sealed plastic bags at 17, 21, and 26 °C. Optimum eclosion occurred at 21 °C with the lowest eclosion at 26 °C. In general, mean full eclosion declined at ca. 0.1% eclosion per hour sealed in plastic bags during the first 96 h for all temperatures. In the third and fourth experiments at 17 °C, there was a decline in average eclosion for irradiated and unirradiated pupae of about 13.4% after they were sealed in plastic bags for 192 h. In general, B. tryoni eclosion declined at 0.1% per hour inside sealed plastic bags for periods up to 192 h at 17 °C. Queensland fruit flies can tolerate long periods of conditions found inside sealed plastic bags and current practices for sterile B. tryoni release programmes will result in minimum decrease in eclosion. The possible evolution of tolerance of these conditions is discussed.  相似文献

The variation in seed moisture, mould content and seed germination of two species of melon,Citrullus lanatus L. andCitrullus vulgaris Shrad. stored in jute bags was monitored at intervals of 2 months for one year. Seed moisture decreased in the first 6 months of storage followed by a gradual increase in the rainy months. The field fungi predominated immediately after harvest and disappeared completely after 6 months of storage. The storage fungi increased in both species and abundance throughout the storage period.Citrullus lanatus maintained high germination percentage for the first 4 months,C. vulgaris for the first 2 months, thereafter seed germinability decreased with prolonged storage.  相似文献

Aquatic hyphomycetes colonizing the submerged chirpine (pinus roxburghii SARG .) needle litter in a high altitude, Kumaun Himalayan stream were studied. 15 species belonging to different genera of aquatic Hyphomycetes have been recognized as the colonizers of chirpine needle litter. Clavariopsis aquatica, Heliscus lugdunensis, Lunulospora cymbiformis, Triscelophorus acuminatus and T. monosporus were found with a high frequency of occurrence. The conidial production was highest in Flagellospora penicillioides, however, Campylospora chaetocladia, L. cymbiformis and T. acuminatus had less number of conidia per unit area of pine needles. The chirpine needle litter decomposition in the freshwater habitat is also discussed.  相似文献

Our overall objectives were to prepare commercially acceptable formulations of the postharvest biological control yeasts, Metschnikowia pulcherrima and Pichia guilliermondii, which have a long storage life and to determine the effectiveness of these formulations to control postharvest green and blue moulds on citrus fruit. Yeasts, grown on a cane molasses-based medium, were combined with talc or kaolin carriers and various adjuvants and the viability of yeast in 12 formulations was determined over a 6 month period. Formulation no. 11, containing talc, sodium alginate, sucrose, and yeast extract, for both yeasts had a significantly higher viable yeast cell content over a 6 month storage period. Among the formulations, three formulations (formulations no. 5, 6, and 11) were selected for additional in vivo testing because they had higher levels of viability amongst yeast cell populations during storage and were easier to resuspend remained in suspension more easily. These formulations were tested on Satsuma mandarin and grapefruit to control green and blue moulds. Formulations no. 5, 6, and 11 for both yeasts effectively controlled green mould, while only formulation no. 11 with either yeast isolate M. pulcherrima (isolate M1/1) or P. guilliermondii (isolate P1/3) effectively controlled both blue and green moulds.  相似文献

Powder formulations were prepared with conidia of the entomopathogenic fungus Beauveria bassiana strain 447 originally isolated in Brazil from the red imported fire ant, Solenopsis invicta. The carrier materials used in formulations included talc hydrous magnesium silicate , silica gel, powdered rice and cornstarch. The formulations were stored in plastic containers under three conditions: a ambient temperature 15-38 C ; b refrigerator 6 2 C ; and c freezer 10- 7 C . Formulations stored under ambient temperature conditions completely lost viability after 1-8 months. Unformulated conidia stored under ambient conditions were totally unviable after just 2 months. All formulations stored under refrigerator and freezer conditions maintained 100 viability for 7 years. The virulences of the conidial preparations against adult workers of the fire ant, S. saevissima, and the sugarcane borer, Diatraea saccharalis, were higher for fresh conidia or conidia stored in the freezer than for conidia stored in the refrigerator. After 30 months of storage, the unformulated conidia and the formulations stored in the refrigerator showed slow germination and had low virulence. There was a strong correlation between the rate of germination and the virulence toward D. saccharalis, but not toward S. saevissima.  相似文献

Entomopathogenic fungi, such as Metarhizium anisopliae and Beauveria bassiana, have been shown to be efficacious in killing mosquito larvae of different mosquito species. The current study compared the pathogenicity and efficacy of two formulations of three fungal strains against different instars of three mosquito species with the aim of identifying the most virulent strain for use under field conditions. Three strains of Metarhizium, ARESF 4556, ARSEF 3297 and V275, were assayed against early (L2?3) and late (L3–4) instar larvae of Aedes aegypti, Anopheles stephensi and Culex quinquefasciatus. Two formulations of the fungi were tested, dry conidia and aqueous suspensions (i.e. ‘wet’ conidia). Effects of all combinations of conidia, mosquito species, instar, fungal strain and concentration on mosquito mortality were analysed using Cox regression and Kaplan–Meier analyses. Strain ARSEF 4556 was more virulent than ARSEF 3297 and V275, with LT50 values ranging from 0.3 to 1.1 days, with Anopheles and Culex being more susceptible than Aedes. Early and late instars were equally susceptible independent of species. Although the formulation did influence mortality rates, both ‘wet’ and ‘dry’ conidia applications were highly effective in killing mosquito larvae. Viable spores were more efficacious than heat killed spores. The latter did cause mortality but only at high concentrations. Metarhizium sp. has proved to be effective in reducing survivability of all larval stages of Aedes, Anopheles and Culex under laboratory conditions. Aedes larvae were generally more tolerant than Anopheles and Culex irrespective of fungal strain.  相似文献

Abstract. 1. Recent studies have demonstrated the existence of positive density dependence in the survival and development of Drosophila (the so‐called Allee effect); however the underlying mechanisms of such Allee effects have remained elusive. Competition with filamentous fungi have often been suggested to be involved in causing high mortality at low larval density, but it has not yet been explicitly tested if the well known spatial aggregation of insect eggs yields a fitness benefit for the developing larvae in the presence of noxious moulds. 2. Using Drosophila melanogaster, the present study tested whether larval survival is greater in aggregations when confronted with various combinations of three representative mould species (Aspergillus, Alternaria, and Penicillium) and a head start for fungal development. 3. High rates of fungal‐dependent mortality and significant positive density‐dependent larval survival (i.e. Allee effects) were observed when larvae were confronted with food resources containing established colonies of Aspergillus or Alternaria. Neither the simultaneous transfer of Aspergillus or Alternaria spores with larvae to food patches nor food infections with Penicillium affected insect larval development. 4. Significant correlations between mould growth and larval survival could be identified, although the patterns that emerged were shown to be inconsistent when the effects were compared between fungal species and fungal priority. Because mould growth only partly explained larval survival, the influence of other fungal‐borne factors, e.g. mycotoxins, needs to be elucidated in order to understand the mechanistic basis of insect–mould interactions. 5. These results are the first to argue convincingly for moulds being involved in mediating Allee effects for insects on ephemeral resources; however they also demonstrate an unexpected diversity in insect–mould interactions. Considering this diversity may be important in understanding insect spatial ecology.  相似文献

Summary The number of fungal species colonizing thermal and acoustic fiberglass insulations used in heating, ventilation, and air conditioning (HVAC) systems was fewer than that obtained from initial direct culture of these insulations. The colonization, determined by the microscopic observation of conidiophores with conidia, was primarily of acrylic-latex-facing material, but eventually the fungi permeated the fiberglass matrix. Isolates ofAspergillus versicolor were most often obtained from non-challenged insulation, whereasAcremonium obclavatum appeared to be the primary colonizing fungus in high-humidity (>90%) challenge chambers. At a lower humidity (about 70%)Aspergillus flavus was one of the more prominent fungi. Not all duct liner samples were equally susceptible to colonization and duct board appeared relatively resistant to colonization.  相似文献

One hundred and six strains ofA. fumigatus were isolated from 21 sawmills in Sweden, and 73 of these strains were examined for production of fumitremorgen B and verruculogen (tremorgenic mycotoxins) on YES-medium using thin layer chromatography (TLC). Twenty-three strains (32%) were tremorgen producers and 50 strains (68%) were non-producers. Tremorgenic mycotoxins were detected in conidia of sevenA. fumigatus strains. The amount of toxin varied between 0.6–8.0 µg/108 conidia (mean value 2.3 µg/108 conidia, equivalent with 0.18%). No production of the mycotoxin gliotoxin was detected in 6 strains ofA. fumigatus. No tremorgens were detected during mould growth on wood substrates, in spite of the use of different wood species (Scots pine,Pinus sylvestris; Norway spruce,Picea abies and birch,Betula spp.), dried versus non-dried wood, bark (pine), leached wood, and wood after various sterilization methods.  相似文献

The effect of formulation, fungal concentration, type and seasonal changes on the mortality of the tick Amblyomma variegatum was investigated. A previous study demonstrated high pathogenicity of strains of the fungi Beauveria bassiana and Metarhizium anisopliae against the ticks Rhipicephalus appendiculatus and Amblyomma variegatum (Kaaya et al. J Invertebr Pathol 1996; 67: 15–20). The present study was undertaken to explore possible additive or synergistic effects of the two fungi on A. variegatum. The effects of oil and water formulations at different concentrations of each fungus and combination of the two on the mortality of A. variegatum in the laboratory and in the field during the wet and dry seasons were determined and compared. The oil formulation performed better in all assays, with highest tick mortality of 92% occurring during the wet season at conidia concentration of 1 × 1010 conidia/ml of the mixed fungi compared to 49% for the water formulation at similar conidia concentration. However, at the same conidial concentration during the dry season, mortalities in the field were relatively low with the mixture of the fungi recording 24% and 17% tick mortality for the oil and water formulation respectively. The effect of infecting the ticks with a cocktail of the two fungi was inconclusive under more controlled conditions in the laboratory but field results under both wet and dry seasons indicated significant differences between the separate and mixed fungi infections. The results demonstrate a potential of cocktail formulations in the control of ticks and possibly of other arthropod pests.  相似文献

Abstract

The paucity of information on the moulds in Indian pearl millet (Pennisetum glaucum) led to the studies that were conducted at ICRISAT, India to evaluate (a) 447 germplasm accessions of 32 countries for mould reaction in rainy season, (b) threshed grain mould rating (TGMS) and mycoflora on grains of each accession, and (c) mould scores in field and in vitro. Post physiological maturity evaluation showed that 16% of the accessions secured a mould rating of 2. In TGMS, 18% were mould free and 57% secured a rating of 2 on a 1 – 9 scale. Assessment of twenty representative accessions in vitro against individual and mixed conidial suspensions (1 × 10(6) conidia ml(?1)) of Fusarium moniliforme, F. pallidoroseum and Curvularia pennisetti indicated significant correlation (r = 0.97) between the overall field and in vitro scores of mixed spores inoculations. The mycoflora for TGMS in blotter test revealed that Fusarium moniliforme, F. pallidoroseum, Curvularia pennisetti, Helminthosporium spp., Alternaria spp. and Colletotrichum spp. to be the major fungi affecting pearl millet grain. It is advisable to harvest panicles at the physiological maturity stage to obtain better quality grains. A strong negative correlation between TGMS and % GS (r = 0.4601) and positive correlation between TGMS and % UGS (r = 0.4654) indicated that, the lesser the threshed grain mould rating higher the % seed germination.  相似文献

Bacteriological surveillance is little applied in management of equine frozen semen but it is quite important to verify the microbial contamination in order to find out the chance of transmission of pathology to the mare in AI. Authors describe a qualitative and quantitative analysis for bacterial contamination on long time (3–17 years) equine frozen semen stored in liquid nitrogen. The semen checked, produced in Italy and in another Europe country, was cryopreserved in liquid nitrogen inside sealed plastic straws. One hundred and ten straws were checked out for pathogenic and no pathogenic bacteria, aerobes and anaerobes and fungi (moulds and yeasts). The Total Microbial Charge was quite variable with an average of about 1.4 × 105 CFU/ml. Mostly the microbial agents identified were fungi (17.5%), Enterobacter-coccus spp. (15%), Pseudomonas spp. (6.25%), Stenothophomonas maltophila (6.25%) and anaerobic bacteria like Propionibacterium granulosum (7.5%) and Clostridium spp. (3.75%). 3.75% were unidentified Gram-negative rod and cocci. Streptococcus spp., Staph. aureus, E. coli, Th. equigenitalis and Mycoplasma spp. were not detected. The most represented species were Enterobacter-coccus spp. (1.1 × 105 CFU/ml), St. maltophila (8 × 104 CFU/ml) and Pr. granulosum (7 × 104 CFU/ml) while yeast and even more moulds were little abundant (4.7 × 104 and 3.4 × 104 CFU/ml respectively). The knowledge of equine frozen semen microbial quality is essential to check out transmission of venereal disease and improve the quality of cryopreserved germplasm.  相似文献

An ultrastructural study was performed on Arthrinium aureum. The fungi were treated with glutaraldehyde and osmium tetroxide fixation. The hypha and conidia has a concentric membrane system which consisted of multiple membranes of a myelinoid appearance, and continued to the conidia and hypha plasma membrane. The fungi were also treated with periodic acid-alkaline bismuth (PABi) staining after glutaraldehyde and osmium tetroxide fixation. PABi positive materials were found on the marginal glycogen granules, the concentric membrane system and the conidia plasma membrane.  相似文献

Rates of decomposition, and soil faunal abundance and diversity associated with single-species and mixed-species litters were studied in a litter bag experiment in an oak–pine forest. We used two canopy species of leaf litter, pine and oak, and one shrub species, Sasa, and compared decomposition rates, and soil microarthropod abundance and community structure of oribatid mites in the litter bags. Mass loss of single species decreased in the order: oak > pine > Sasa. While the total mass loss rates of mixed litter were intermediate between those of the constituent species, enhancement of mass loss from the three-species mixture and from mixed slow-decomposing litters (pine and Sasa) was observed. Faunal abundance in litter bags was higher in mixed-species litter than in those with single-species litter, and species richness of oribatid mites was also higher in the three-species mixed litter. Faunal abundance in single-species litter bags was not correlated with mass loss, although enhancement of mass loss in mixed litter bags corresponded with higher microarthropod abundance. Habitat heterogeneity in mixed litter bags seemed to be responsible for the more abundant soil microarthropod community.  相似文献
