前列腺癌是全球发病率第二的男性恶性肿瘤,近年来我国前列腺癌的发病率显著上升。最近,大量研究发现,长非编码RNA表达水平的改变与前列腺癌的发生、发展、诊断、治疗和预后密切相关。该文就长非编码RNA与前列腺癌的关系及相关机理进行综述,有助于读者了解长非编码RNA在前列腺癌发病过程中的重要性,为前列腺癌的综合防治提供线索。  相似文献

Prostatic acid phosphatase (PAP) expression increases proportionally with prostate cancer progression, making it useful in prognosticating intermediate to high-risk prostate cancers. A novel ligand that can specifically bind to PAP would be very helpful for guiding prostate cancer therapy. RNA aptamers bind to target molecules with high specificity and have key advantages such as low immunogenicity and easy synthesis. Here, human PAP-specific aptamers were screened from a 2′-fluoropyrimidine (FY)-modified RNA library by SELEX. The candidate aptamer families were identified within six rounds followed by analysis of their sequences and PAP-specific binding. A gel shift assay was used to identify PAP binding aptamers and the 6N aptamer specifically bound to PAP with a Kd value of 118 nM. RT-PCR and fluorescence labeling analyses revealed that the 6N aptamer bound to PAP-positive mammalian cells, such as PC-3 and LNCaP. IMR-90 negative control cells did not bind the 6N aptamer. Systematic minimization analyses revealed that 50 nucleotide sequences and their two hairpin structures in the 6N 2′-FY RNA aptamer were equally important for PAP binding. Renewed interest in PAP combined with the versatility of RNA aptamers, including conjugation of anti-cancer drugs and nano-imaging probes, could open up a new route for early theragnosis of prostate cancer.  相似文献

长非编码RNA(long non-coding RNAs, lncRNAs)是一类转录本长度大于200个核苷酸,不具有蛋白质编码功能的非编码RNA(non-coding RNA, ncRNA)。人类基因组中,ncRNA基因占比超过90%,数量远大于蛋白质编码基因。作为生物大分子,lncRNA具有特定的初级和高级结构,在基因表达调控等生物学进程中发挥着特有的功能。lncRNA数量多,结构各异,因此鉴定和表征新的lncRNA,探索其结构和功能,是当前基因研究领域的热点之一。在临床疾病机制研究中,大量结果表明,lncRNA与临床疾病发生发展,特别是肿瘤的发生发展具有密切的相关性。伴随着后基因组学时代基因鉴定和功能探索方法的不断进步,探索lncRNA在疾病发生中的功能及表达变化,深入解锁lncRNA在疾病发生中涉及的分子机制,将为疾病早期预防、诊断和预后提供有效参考。基于以上的研究大背景,本文对lncRNA的定义、基因鉴定的策略和方法,高级结构检测及其对应的生物学功能,以及lncRNA的分类进行了阐述;另一方面,基于lncRNA与肿瘤发生发展的密切关系,本文以经典抑癌基因p53为切入点,对多种p53相关的lncRNA在结直肠癌(colorectal cancer, CRC)发生发展中的作用进行了归纳小结,阐述了lncRNA在结直肠癌中的表达变化、涉及的分子互作机制和信号通路,对其作为分子标志物在临床中的应用潜力进行了评估。我们乐观地认为,作为生物分子标志物,lncRNA将为包括癌症在内的疾病治疗提供全新、精准和个性化的分子靶点。  相似文献

目的:探讨前列腺癌细胞系中的环状RNA circRNA-1565的表达及鉴定。方法:培养前列腺正常上皮细胞(RWPE-1)、4种前列腺癌细胞(22RV1、LNCap、PC-3、DU145),提取总RNA,采用RT-PCR检测不同细胞系中circRNA-1565的表达,并对其进行成环性验证及细胞内亚定位实验,进行差异比较。结果:circRNA-1565在正常前列腺细胞系(RWPE-1)中表达量极低,在转移性前列腺癌细胞系中高表达,在非转移性前列腺癌细胞系中低表达。且circRNA-1565是一条主要定位于细胞质内的具有有效环状结构的RNA分子。结论:circRNA-1565在不同前列腺癌细胞系中存在差异表达,可能会通过mi RNA sponge途径发挥生物学作用。  相似文献

采用RNA-seq技术对志贺菌耐药性与非编码RNA之间的关系进行研究。首先采用梯度剂量的环丙沙星诱导福氏志贺菌2a标准菌株产生耐药性而获得耐药菌株,然后提取标准菌株与耐药菌株的总RNA进行RNA-seq分析。结果显示,在福氏志贺菌的2个样品中共发现100个候选sRNA,并与sRNA的数据库比对得到注释信息,每个候选sRNA都对应着很多个靶标基因,而且靶标基因的表达也有明显差异。预测这些sRNA可能会通过调控耐药基因和一些与细胞增殖或凋亡相关的基因发挥作用,因此还需要挑选合适的候选sRNA并对其用荧光定量PCR实验进行验证。  相似文献

Nontemplated 3'-end oligouridylation of RNA occurs in many species, including humans. Unlike the familiar phenomenon of polyadenylation, nontemplated addition of uridines to RNA is poorly characterized in higher eukaryotes. Recent studies have reported nontemplated 3'-end oligouridylation of small RNAs and mRNAs. Oligouridylation is involved in many aspects of microRNA biology from biogenesis to turnover of the mature species, and it may also mark long mRNAs for degradation by promoting decapping of the protective 5'-cap structure. To determine the prevalence of oligouridylation in higher eukaryotes, we used next-generation sequencing technology to deeply examine the population of small RNAs in human cells. Our data revealed widespread nontemplated nucleotide addition to the 3' ends of many classes of RNA, with short stretches of uridine being the most frequently added nucleotide.  相似文献

鸟氨酸脱羧酶基因反义RNA对前列腺癌细胞生长的抑制作用   总被引:4,自引:0,他引:4  
为研究鸟氨酸脱羧酶 (ODC)基因反义RNA对前列腺癌细胞的生长抑制作用 ,将表达ODC第 3外显子反义RNA的重组腺病毒rAd ODC Ex3as分别感染前列腺癌细胞株PC 3和LNCap .通过MTT法观察其对前列腺癌细胞增殖的影响 ,并确定不同细胞合适的感染滴度 ,再采用Western印迹和流式细胞术检测rAd ODC Ex3as对细胞中ODC表达的抑制作用、对细胞周期和凋亡的影响以及与CDK抑制物p2 1的关系 .实验显示 ,rAd ODC Ex3as分别以 5 0MOI、2 5MOI感染PC 3和LNCap细胞可明显抑制其生长增殖 ,而不引起细胞毒性作用 ;其对两种细胞中ODC表达的抑制作用分别为4 5 %和 5 9% .流式细胞DNA含量分析证实 ,rAd ODC Ex3as可引起PC 3和LNCap细胞周期G1期阻滞 ,但并未引起凋亡 .通过Western印迹发现 ,细胞中ODC表达的降低可诱导p2 1蛋白的过表达 .结果表明 ,rAd ODC Ex3as在体外能有效地干扰ODC基因的表达 ,并通过诱导p2 1的过表达使其细胞周期停于G1期 ,从而抑制前列腺癌细胞PC 3和LNCap的增殖 ,为其进一步基因治疗的研究打下基础 .  相似文献

乳腺癌是女性最常见的恶性肿瘤之一,且其发病率呈逐年上升的趋势,尽管随着诊疗技术的进步,乳腺癌的死亡率得到了实质性的降低,但它仍是女性肿瘤死亡的第二大原因,目前对乳腺癌的治疗还面临很大的挑战。长链非编码RNA(Longnon-codingRNA,LncRNA)是一类长度超过200个核苷酸的非编码RNA分子,缺乏开放的阅读框,无编码蛋白质的功能。近年来研究发现,LncRNA可能在多种恶性肿瘤的发生、发展、侵袭及转移过程中起调控作用,且目前对其与乳腺癌的关系研究较多,对LncRNA与乳腺癌相关性的研究有望为乳腺癌的诊治提供新思路。现就LncRNA在乳腺癌中的研究进展做一综述。  相似文献

Prostate cancer (PC) is the most common carcinoma among men worldwide which results in 26% of leading causes of cancer-related death. However, the ideal and effective molecular marker remains elusive. CircRNA, initially observed in plant-infected viruses and Sendai virus in 1979, is generated from pre-mRNA back-splicing and comes in to play by adequate expression. The differential expression in prostate tissues compared with the control reveals the promising capacity in modulating processes including carcinogenesis and metastasis. However, the biological mechanisms of regulatory network in PC needs to systemically concluded. In this review, we enlightened the comprehensive studies on the definite mechanisms of circRNAs affecting tumor progression and metastasis. What''s more, we validated the potential clinical application of circRNAs serving as diagnostic and prognostic biomarker. The discussion and analysis in circRNAs will broaden our knowledge of the pathogenesis of PC and further optimize the current therapies against different condition.  相似文献

生物标志物一直是医学领域的研究热点.目前主要以实时荧光定量PCR技术作为分子诊断的检测手段,用于大样本的核酸检测,但该技术具有限制性,会导致假阴性.作为第三代PCR的数字PCR技术,优化并提高了检测灵敏度,无需标准曲线实现绝对定量检测,大大提高了检测的精准度,尤其是对于低表达RNA的检测,显得更为出色.本研究以hsacirc0061276为例,利用微滴式数字PCR EvaGreen染料法对血浆中hsacirc0061276的拷贝数进行绝对定量地检测,成功检测出胃炎和胃癌患者血浆中hsacirc0061276的具体拷贝数.因此,数字PCR作为新型检测技术在临床实际应用中有较大推广价值.  相似文献

非编码RNA是一类没有开放阅读框、不能翻译成为蛋自质的RNA分子。在哺乳动物中,它们主要是指微小RNA、小干扰RNA、PIWI互作RNA和其他一些反义转录本等。它们在生物体内广泛存在,通过RNA干扰、基因沉默、基因印迹和DNA甲基化等机制调控着基因的表达。非编码RNA增加了真核细胞调控网络的复杂性,也为科学地解释一些现象提供了新的途径。  相似文献

SUMO (small ubiquitin-related modifier)是一种小泛素相关修饰物,能共价结合许多调控基因转录的重要蛋白,包括转录因子、转录辅助因子等.SUMO化修饰对蛋白-蛋白之间的相互作用、亚细胞定位、基因转录的活性以及靶蛋白的稳定性等具有重要的调节作用. SUMO化修饰是一个动态可逆的过程,将SUMO从靶蛋白上去除,称为去SUMO化(desumoylation),去SUMO化是SUMO特异蛋白酶(SUMO-specific proteases,SENPs)的主要功能.由于SUMO化是近几年才发现的一种新的蛋白质翻译后修饰系统,对其生物学功能还不十分清楚.前列腺癌是男性最常见的恶性肿瘤,最近的研究发现,SENP1在前列腺癌细胞中高表达,而且雄激素能诱导SENP1的表达,表明SENP1与前列腺癌的发生、发展密切相关.在本篇综述中,我们将就SENP1作一介绍.  相似文献

Thyroid cancer (TC) is one of the most common malignant tumors, with high morbidity and mortality rates worldwide. The incidence of TC, especially that of papillary thyroid carcinoma (PTC); has increased rapidly in recent decades. Autoimmune thyroid disease (AITD) is closely related to TC and has an estimated prevalence of 5%. Thus, it is becoming increasingly important to identify potential diagnostic biomarkers and therapeutic targets for TC and AITD. Circular RNAs (circRNAs) are a class of non-coding RNAs with covalently bonded circular structures that lack 5''-3'' polarity and polyadenylated tails. Several circRNAs play crucial roles in the development of various diseases, including TC and AITD, and could be important new biomarkers and/or targets for the diagnosis and therapy of such disorders. Although there are four subtypes of TC, research on circRNA has largely focused on its connection to PTC. Therefore, this review mainly summarizes the relationships between circRNAs and PTC and AITD, including the molecular mechanisms underlying these relationships. In particular, the functions of “miRNA sponges” and their interactions with proteins and RNA are discussed. The possible targeting of circRNAs for the prevention, diagnosis, and treatment of TC and AITD is also described. CircRNAs could be potential biomarkers of TC and AITD, although validation will be required before they can be implemented in clinical practice.  相似文献
