濒危植物木根麦冬的胚胎学研究   总被引:20,自引:0,他引:20  
木根麦冬的花药壁分为4层,即:表皮、药室内壁、中层与绒毡层。绒毡层为腺质型。大孢子母细胞减数分裂过程中的胞质分裂为连续型。小孢子四分体排列为左右对称型与四面体型。雌蕊子房壁在授粉后4~5天破裂。胚珠倒生,双珠被,厚珠心。大孢子四分体线形排列,胚囊发育为蓼型。极核在受精前融合成一个大的次生核。胚胎发育为紫菀型。核型胚乳。木根麦冬居群中存在雄性不育植株,约占55.8%。胚胎在其发育早期约有43.3%败育。根据本研究的实验观察,(1)雄性不育和自花授粉以及低效率的传粉机制而导致的异花传粉的失败;(2)部分胚胎的败育;(3)子房壁破裂而导致胚珠裸露等是木根麦冬有性生殖过程中的薄弱环节,也可能是导致该植物濒危的部分因素。  相似文献

为了揭示多拷贝基因的进化方式,对濒危植物木根麦冬(Ophiopogon xylorrhizus Wang et Dai)3个居群、6个个体的294个5S rRNA基因拷贝及其姐妹种林生麦冬(O.sylvicola Wang et Tang) 的45个拷贝进行了DNA测序和序列分析,并以这个迄今发表的最大的单个物种的5S rDNA基因数据,以PAUP程序重建了分子系统发育树。结果表明:1)所得序列呈高度多样性,长度变化在307-548碱基之间,仅13对(3.8%)相同,序列分化指数较高:木根麦冬是0.078,林生麦冬是0.032,两物种间是0.149;2)100%的统计值支持两物种的5S rRNA基因分别来自于祖先种的一个拷贝,即“建立者拷贝”,这个拷贝在物种形成之后进行了一系列连续的扩增,形成一个直系的基因家族,而祖先种的其他拷贝在物种形成后被丢失;3)不同拷贝是独立进化的,序列间的一致化过程很弱,这在串联重复的rRNA基因中是罕见的;4)木根麦冬居群间曾存在频繁的基因交流,使5S rRNA基因的许多拷贝扩散于不同居群,维持着种内较高的遗传多样性。可以认为是某些近期发生的变化,阻止了居群间的基因交流,导致该物种广泛的自交,发生自交衰退,并最后导致濒危。  相似文献

We assessed the utility of codominant allozyme markers for paternity analysis in natural populations of Ophiopogon xylorrhizus (Liliaceae s. l.) by means of likelihood‐based approach. In the three independent natural stands of 46 individual plants with 354 seeds of O. xylorrhizus (Liliaceae s. l), we assigned 319 seeds to a single individual with the highest LOD score. Among these, 179 seeds had a significant D‐value. An individual (acting either as female or male) mateds with an average of one other individual on average to reproduce, except when it self‐pollinated. If an individual that neither donated pollen to other individuals nor selfed, being strictly a pollen acceptor, the number of offspring produced was half that produced by as many offspring as individuals which both provided and accepted pollen (4.1 vs. 9.5). Significant positive relationships were found in all the three stands between the number of self‐pollen an individual produced and the number of seeds that individual set. Male outcrossing reproductive success was unevenly varied, both in amount and spatial location. The individuals located in the centre of the stand and the individuals closest to each other had higher male outcrossing reproductive success than those at the edge of the stand. Male selfing reproductive success showed a similar pattern, with an even distribution of female reproductive output. Most mating events were took place within 20 m. The distance between mates mirrored the active ranges of pollinators, and shaped the fine‐scale population spatial genetic spatial structure. The results indicate that selfing assures reproductive success in O. xylorrhizus.  相似文献

木根麦冬(Ophiopogonxylorrhizus)是我国珍稀濒危植物,分布仅限于云南西双版纳雨林,在植物系统学和保护生物学研究中具有独特的意义。随机扩增多态DNA(RAPD)方法是揭示群体遗传多样性的高效、简便方法,但一般均以新鲜材料提取总DNA,对一些分布边远地区物种难以采用此法。本文研究从木根麦冬干叶片中提取总DNA,进行RAPD分析。样品取自4个居群、49个个体。选取生长旺盛的叶片,在野外用硅胶快速干燥保存样品。采用高盐低pH值法提取总DNA,每克鲜重所得的干叶可得80~160μg。通过对模板DNA的各种处理和PCR扩增程序的调整,解决了扩增片段边缘弥散、界线模糊、产率低等问题,获得了理想的扩增带型。这一成果对其它从野外直接采样的干叶提取DNA进行RAPD研究具有指导意义  相似文献

冠果草花药壁的发育为单子口十型,绒毡层为周原质团型。小孢子母细胞减数分裂为连续型,四分体呈左右对称式排列,成熟花粉为三细胞型。双珠被,假厚珠心,倒生胚珠。胚囊发育为葱型,成熟胚囊的特点是两个极核分别位于中央细胞两端,不融合成次生核。受精过程中,一个精于与卵核融合形成合子,另一精子先与珠孔端极核融合,之后受精极核再移动到合点端与另一极核融合,形成初生胚乳核。胚的发育为石竹型。成熟胚呈马蹄形,具有2片真叶。胚乳发育为沼生目型。随着胚的发育,胚乳细胞逐渐解体,成熟种子中无胚乳。  相似文献

This paper deals with the embryological characteristics of Sagittaria guayanensis H. B.K. subsp. lappula (D. Don) Bojin. The anther wall development follows the Monocotyledonous type. The cytokinesis of microspore mother cell in meiosis is of the Successive type. The tetrads of microspores show an isobilateral arrangement, and the mature pollen grains are 3-celled. The ovule is bitegminous, pseudo-crassinucellate and anatropous. The megaspore mother cell originates directly from a single archesporial cell. The mature embryo sac consists of 7 cells including 8 nuclei and conforms to the Allium type. The two polar nuclei do not fuse into a secondary nucleus before fertilization. Instead, one sperm fuses with the micropylar end polar nucleus first , and the fertilized polar nucleus then migrates to the chalazal end, where it fuses with the second polar nucleus, forming the primary endosperm nucleus. The embryo development conforms to the Caryophyllad type. The mature embryo is U-shaped and forms the embryonic shoot apex accompanied by two leaves. The endosperm development corresponds to the Helobial type. The primary endosperm nucleus (invariably lying in the chalazal part of the embryo sac) divides and forms two chambers:large micropylar one and small chalazal one. The chalazal endosperm chamber remains binucleate, while, in the micropylar chamber free nuclear divisions occur and then cellnlarization takes place. During the embryo formation the endosperm gradually degrades and can not be found in the mature seed. The subgenus Lophotocarpus is different from the subgenus Sagittaria in some embryological aspects, especially in the structure of mature embryo sac and the double fertilization process.  相似文献

长喙毛茛泽泻Ranalisma rostratum Stapf的配子融合开始较早,但精核与极核融合速度较快。精核与珠孔极核融合后即移向胚囊合点端与另一极核再融合形成初生胚乳核。初生胚乳核第一次分裂后形成两个细胞,以后珠孔端胚乳细胞再进行游离核分裂,胚乳发育属于沼生目型。开花后两天,合子分裂,八分体呈4层,每层呈两个细胞方式排列胚胎发生属于石竹型。  相似文献

Ranalisma rostratum Stapf is a rare and endangered species. This paper deals with the fertilization and the development of embryo and endosperm in this plant.The embryogenesis is of Caryophyllad type and the development of endosperm belongs to Heobial type.Before fertilization,the two polar nuclei are located respectively at both ends of embryo sac. In most angiosperms with two polar nuclei,the polar nuclei may fuse eiher before fertilization to form a secondary nucleus or during fertilization called triple fusion. In Ranalisma rostratum Stapf, however, it is found that only in case when the micropylar polar nucleus is fertilized,it can move to the chalazal end and fuse with the chalazal polar nucleus.This phenomenon is very rare and the process must take more time to fulfil fertilization both polar nuclei. This feature of fusion of polar nuclei is therefore thought as a primitive character from the view of phylogeny.  相似文献

通过实地网格化调查、标本查阅及历史资料搜集对上海濒危植物现状及保护对策进行了研究,结果表明:上海共有国家重点保护和濒危野生植物23科27属28种,建议上海重点保护的野生植物有54科82属101种。上海原生植物减少、外来植物增多、生物多样性降低等是上海一些原生植物变为濒危的主要原因。今后应建立上海濒危植物保护体系,重点保护上海的植物多样性中心,并尽快立法公布上海地方重点保护野生植物名录;加强植物多样性保护宣传,保护上海的野生植物资源并防范外来植物入侵。  相似文献

Beneficial microbes such as plant mutualistic fungi, hold the promise of ameliorating challenges faced in native plant conservation such as disease management. As an alternative to costly chemical pest control, conservation efforts could potentially harness the benefits of plant mutualistic fungi to aid in defense and disease resistance, but there are few tests of this notion. We set out to test the efficacy of controlling a common foliar pathogen, the powdery mildew Neoerysiphe galeopsidis, by inoculating the endangered Hawaiian plant species Phyllostegia kaalaensis with potentially beneficial members of its wild-type mycobiome. We tested whether inoculating plants with above or belowground fungal mutualists, or both, led to increased disease resistance in the host. We found that while all treatments reduced average disease incidence, colonization by the foliar yeast Moesziomyces aphidis was the only treatment to do so significantly. These results provide an exciting new strategy for plant conservation practices.  相似文献

In-situ study on eleven commercially important species viz;Adiantum capillus-veneris L,Bergenia ciliata (Haw) Sternb,Colchicum luteum Baker,Polygonum amplexicaule D.Den,Cuminum cyminum L,Dioscorea deltoidea Wall Kunth,L Morchella esculenta L,Paeonia emodi Wall ex H Kf,Podophyllum hexandrum (Royle) Chatt & Mukh,Valeriana wallichii DC and Viola serpens Wall ex Roxb was conducted in four locations viz; Malam (1 400 to 2 000m),Bargin (1 700 to 2 300m),Biakand (1 500 to 2 100m) and Shinko (2 100 to 2 700m).The data was recorded from both protected and unprotected sites of each site.Each site had 3 altitudinal sampling point.The density, herbage coverage and fresh biomass were determined in each locations for every species.All the investigated parameters generally showed an increase of 3 to 6 times over unprotected sites in each locations.Morchella esculenta,Dioscorea deltoidea,Colchicum luteum and Podophyllum hexandrum were absent in all unprotected sites while other species had low values in these sites.The investigated parameters of Colchicum luteum,Bergenia ciliata,Paeonioa emodi,Dioscorea deltoidea and Podophyllum hexandrum generally increased with the increasing elevation.Soil analysis,soil and air temperatures were recorded for each site.The air and soil temperature were slightly higher in open areas than in the protected site and showed decrease with increasing elevation.While the soil fertility was relatively high in protected sites as compared to unprotected area.The study shows that protection promotes the growth, distribution and occurrence of medicinal plants.It is possible with the participation of local communities to conserve these resources.  相似文献

利用CART(分类和回归树Classification and regression tree)模型,采用A2和B2气候情景,模拟分析了气候变化对观光木(Tsoongiodendron odorum)、鹅掌楸(Liriodendron chinense)、独叶草(Kingdonia uniflora)、草苁蓉(Boschniakia rossica)和刺五加(Acanthopanax senticosus)分布范围及空间格局的影响。结果表明:与目前适宜分布范围相比,气候发生变化后,观光木的分布范围变化不大,其它植物则缩小;观光木的新适宜与总适宜分布范围扩大,而其它植物缩小;草丛蓉和刺五加的目前适宜、新适宜及总适宜分布范围缩小幅度较大,鹅掌楸和独叶草次之;观光木和鹅掌楸向目前适宜分布区北部区域扩展,独叶草向西南区域扩展,草丛蓉和刺五加的适宜范围在2081~2100年会散失。气候变化下,这些植物的适宜分布范围随年降水量和年均气温的变化不一致,观光木的目前适宜、独叶草的新适宜及总适宜分布范围与年降水量和年均气温,以及草苁蓉的分布范围与降水量的相关性不显著(P0.05)。这些植物的空间分布格局将随气候变化而发生变化。  相似文献

As herbivory usually leads to loss of photosynthesizing biomass, its consequences for plants are often negative. However, in favorable conditions, effects of herbivory on plants may be neutral or even beneficial. According to the compensatory continuum hypothesis plants can tolerate herbivory best in resource-rich conditions. Besides herbivory, also primarily positive biotic interactions like mycorrhizal symbiosis, bear carbon costs. Tritrophic plant–fungus–herbivore interaction further complicates plant's cost-benefit balance, because herbivory of the host plant is expected to cause decline in mycorrhizal colonization under high availability of soil nutrients when benefits of symbiosis decline in relation to costs. To gain insight into above interactions we tested the effects of plant size and resource manipulation (simulated herbivory and fertilization) on both above-ground performance and on root fungal colonization of the biennial Gentianella campestris.Clipping caused allocation shift from height growth to branches in all groups except in large and fertilized plants. For large plants nutrient addition may have come too late, as the number of meristems was most likely determined already before the fertilization. Clipping decreased the amount of DSE (dark septate endophytic) fungi which generally are not considered to be mycorrhizal. The effect of clipping on total fungal colonization and colonization by arbuscular mycorrhizal (AM) fungal coils were found to depend on host size and resource level. Dissimilar mycorrhizal response to simulated herbivory in small vs. large plants could be due to more intensive light competition in case of small plants. Carbon limited small plants may not be able to maintain high mycorrhizal colonization, whereas large clipped plants allocate extra resources to roots and mycorrhizal fungi at the expense of above-ground parts. Our results suggest that herbivory may increase carbon limitation that leads re-growing shoots and fungal symbionts to function as competing sinks for the limited carbon reserves.  相似文献

Translocation is a recognized means of rescuing imperiled species but the evidence for the long-term success of translocations is limited. We report the successful translocation of reproductive individuals of a critically endangered shrub Otostegia bucharica from a site facing imminent habitat destruction into a nearby natural population of the species. The relocated plants were visited the year after planting and 13 years later to assess short-and long-term plant survival. Significant percentage of plants that survived transplanting shock and very dry spring following transplanting(around 36%), and further decrease of this number in the next 12 years by only 14%, indicated that O. bucharica is amenable to translocation using reproductive plants. Based on results of species distribution modeling, and failed attempts of ex situ cultivation, we propose introduction of this species into areas with suitable climatic and soil conditions.However, because there is currently no nature reserve in Uzbekistan having suitable conditions for the species under the present climate and that expected in the near future, and because all known habitats of O. bucharica are exposed to the very strong anthropogenic pressure, establishment of a new protected area, awareness building and involvement of local community in conservation activities are required to prevent extinction of this extremely rare species.  相似文献

广东省国家I级重点保护野生植物资源现状及保护策略   总被引:1,自引:0,他引:1  
在查找史料的基础上,根据国家重点保护植物名录(第一批),确定了在广东有分布的仙湖苏铁、银杏、南方红豆杉、合柱金莲木、伯乐树、报春苣苔、水松、异形玉叶金花和台湾苏铁9种国家I级重点保护植物作为调查对象,在全省开展了较为全面的调查。结果只找到了前6种,植株数量合计为123468株。除仙湖苏铁和报春苣苔分布地点没有变化外,其他7种的分布地点丧失率在20%-100%之间。除南方红豆杉基本上属于正常种群外,其余8种分属于野外绝迹、濒临绝迹或濒危类物种,其资源状况堪忧。在对其资源现状进行详细分析的基础上,提出了相应的保护管理策略和发展措施。  相似文献
