利用氨基酸分析方法,对甘蓝(BrasicaoleraceaL.var.capitata)自交不亲和系与亲和系的柱头和花粉蛋白质进行了研究。结果表明,自交不亲和系与亲和系的柱头和花粉氨基酸组分有显著差别。柱头中苏氨酸和酪氨酸含量以及花粉中甘氨酸和丙氨酸含量可以作为评价自交不亲和性的指标。鉴定标准:自交不亲和系柱头中苏氨酸含量高于0.223%,酪氨酸含量高于0.385%;花粉中甘氨酸含量高于1.593%,丙氨酸含量高于1.464%。亲和系柱头中苏氨酸含量低于0.185%,酪氨酸含量低于0.164%;花粉中甘氨酸含量低于1470%,丙氨酸含量低于1006%  相似文献

In the pollen-pistil system of petunia (Petunia hybrida L.) self-compatible and self-incompatible clones within 7 h after self-pollination, we determined the content of ACC (1-aminocyclopropane-1-carboxylic acid), the activity of two enzymes (ACC synthase and ACC oxidase), and the rate of ethylene production. Depending on the type of pollination, germination of pollen on the stigma surface and the pollen tube growth in the tissues of style were accompanied by different levels of ACC and ethylene release. The pollen-pistil system of the self-compatible clone contained twice more ACC than in the self-incompatible clone, whereas the pollen-pistil system in the self-incompatible clone produced 4–5 times more ethylene than in the self-compatible clone. For both types of pollination, ACC and ethylene were predominantly produced in the stigma tissues. The rate of ethylene production therein was 50 times greater than in the styles and ovaries, and the content of ACC was 100 times higher than in the styles and ovaries. Germination of male gametophyte after both types of pollination was accompanied by elevated ACC synthase activity (especially in the case of compatible pollination), whereas notable increase in ACC oxidase activity was manifested in growing pollen tubes after self-incompatible pollination  相似文献

In cruciferous plants, self-pollination is prevented by the action of genes situated at the self-incompatibility locus or S-locus. The self-incompatibility reaction is associated with expression of stigma glycoproteins encoded by the S-locus glycoprotein (SLG) gene. Only a few cases of self-compatible plants derived from self-incompatible lines in the crucifer Brassica have been reported. In these cases, self-compatibility was generally ascribed to the action of single genes unlinked to the S-locus. In contrast, we report here a line of Brassica oleracea var acephala with a self-compatible phenotype linked to the S-locus. By means of both biochemical and immunochemical analyses, we showed that this self-compatible (Sc) line nonetheless possesses stigmatic SLGs (SLG-Sc) that are expressed with a similar spatial and temporal pattern to that described for the SLGs of self-incompatible Brassica plants. Moreover, the SLG-Sc products segregate with the self-compatibility phenotype in F2 progeny, suggesting that changes at the S-locus may be responsible for the occurrence of the self-compatibility character. A cDNA clone encoding the SLG-Sc product was isolated, and the deduced amino acid sequence showed this glycoprotein to be highly homologous to the pollen recessive S2 allele glycoprotein. Hence, self-compatibility in this Brassica Sc line correlates with the expression of a pollen recessive-like S allele in the stigma.  相似文献

We investigated dynamics of the content of 1-aminocyclopropane-1-carboxylic acid (ACC) and ethylene production in male gametophyte development and germination in fertile (self-compatible and selfincompatible) and sterile clones of petunia. Fertile male gametophyte development was accompanied by two peaks of ethylene production by anther tissues. The first peak occurred during the microspore development simultaneously with the degeneration of both the tapetal tissues and the middle layers of the anther wall. The second peak coincided with dehydration and maturation of pollen grains. In the anther tissues of the sterile line of petunia, tenfold higher ethylene production was observed at the meiosis stage compared with that in fertile male gametophytes. This fact correlated with the degeneration of both microsporocytes and tapetal tissues. Exogenously applied ethylene (1–100 ppm) induced a degradation of the gametophytic generation at the meiosis stage. According to the obtained data, ethylene synthesis in germinating male gametophyte is provided by a 100-fold ACC accumulation in mature pollen grains. The male gametophyte germination, both in vitro, on the culture medium, and in vivo, on the stigma surface, was accompanied by an increase in ethylene production. Depending on the type of pollination, germination of pollen on the stigma surface and the pollen tube growth in the tissues of style were accompanied by various levels of ACC and ethylene release. The male gametophyte germination after self-compatible pollination was accompanied by higher content of ACC as compared with the self-incompatible clone, whereas, after the self-incompatible pollination, we observed a higher level of ethylene production compared with compatible pollination. For both types of pollination, ACC and ethylene were predominantly produced in the stigma tissues. Inhibitor of ethylene action, 2,5-norbornadiene (NBN), blocked both the development and germination of the male gametophyte. These results suggest that ethylene is an important factor in male gametophyte development, germination, and growth at the progamic phase of fertilization.  相似文献

Pollen-ovule ratio, pollen grains per flower, pollen grain volume, pollen biomass, ovules per ovary, seeds per fruit, seed volume, seed biomass, and corolla size were studied to reveal patterns of variation within a monophyletic group. Solanum sect. Basarthrum includes 22 neotropical species of two clades, one having small, few-seeded red fruits, and the other having larger, many-seeded green fruits. The former includes self-incompatible species and a dioecious species, while the latter includes self-incompatible species, self-compatible (and autogamous) species, and a domesticate (the “pepino”). Although the pollen quantity and ovule quantity of the self-incompatible species are significantly higher in the green-fruited subgroup than in the red-fruited subgroup, the pollen-ovule ratios of the self-incompatible species are not significantly different between the two subgroups, suggesting parallel evolution of the pollen-ovule ratio. Furthermore, the pollen-ovule ratio tracks the breeding system: self-incompatible species have significantly higher pollen-ovule ratios than self-compatible species, resulting both from fewer pollen grains and more ovules of the latter. The pollen-ovule ratio of the dioecious species is among the highest of all, resulting only from fewer ovules. The pepino is self-compatible but has a pollen-ovule ratio like its wild self-incompatible progenitors and shows wide variation in seed production and pollen quality, but not in pollen quantity. This paper reports on a survey of self-incompatibility in 110species of 22 genera in the family Commelinaceae. Genera fromboth tribes, Tradescantieae and Commelineae are included. Fifty-fivespecies were found to be self-incompatible, 50 species self-compatible,and five species comprised individuals which were self-incompatibleand individuals which were self-compatible. This variabilityand its possible evolutionary significance are discussed. Self-incompatible species had actinomorphic flowers and themajority of these were in the Tradescantieae. Species with zygomorphicflowers which were more commonly found in the Commelineae wereself-compatible. The ubiquitous presence of binucleate pollengrains supports previous data that self-incompatibility is ofthe gametophytic type. The site of pollen tube arrest, however,was on the stigma at or near the base of the stigma papillacells. There were two exceptions to this viz. an unnamed Dichorisandraspecies and Siderasis fuscata where pollen tube arrest was stylar. The significance of these data to taxonomy also receives comment. The hormonal status of the pollen-pistil system in Petunia hybrida L. during the progamic phase of fertilization was investigated. The contents of indolyl-3-acetic acid (IAA), abscisic acid (ABA), and cytokinins, as well as the rate of ethylene production in the pistils and their parts (stigma, style, and ovary) were measured over an 8-h period following compatible and self-incompatible pollination. In both pollinations, the phytohormones were present in various proportions in the stigma, style and ovary: the stigma was the main site of ethylene synthesis and contained 90% of the ABA, while the style contained 80% of the total cytokinin content in the pollinated pistil. Relatively low levels of hormones in the ovary did not influence the hormonal status of the pollen-pistil system. The interaction of the male gametophyte with the stigmatic tissues was accompanied by a 7- to 10-fold increase in ethylene production and a 1.5- to 2.0-fold increase in IAA content in the pollen-pistil system over 0–4 h. Pollen tube growth after self-incompatible pollination, in contrast to compatible pollination, was accompanied by a 3-fold increase in the ABA content in the stigma and style and by a 5-fold higher cytokinin content in the stylar tissues. Thus, the ethylene/ABA status of the stigma may play a role in controlling the processes of adhesion, hydration, and germination of pollen grains during pollination while the auxin/cytokinin status of the style may be involved in controlling pollen tube growth.  相似文献

Hydrolysis of acetylthiocholine and butyrylthiocholine has been observed in aqueous extracts from petunia pollen and pistils. The reproductive organs of self-compatible clone showed a higher rate of choline ester hydrolysis than those of self-incompatible clone. The highest rate of acetylthiocholine hydrolysis blocked by the cholinesterase inhibitors (physostigmine and neostigmine) was characteristic for the pollen of self-compatible clone. The incomplete (25 - 40 %) inhibition of hydrolysis in pistil extracts of self-compatible clone suggests the presence of unspecific esterases. The eight-fold lower hydrolysis was observed in the pistils of self-incompatible clone as compared to the pistils of compatible clone; neostigmine completely blocked this low hydrolytic activity. The treatment of flower buds with physostigmine and neostigmine (10-5 - 10-3 M) decreased the seed production by 10 - 20 % in compatible clone. When the surfaces of pistil stigmae were treated with physostigmine and neostigmine (10-5 - 10-3 M) before pollination, the seed formation was inhibited by 95 % after both self- and cross-pollination. Pollination is one of the most important steps during fertilization and sexual reproduction in plants, and numerous cell-cell interaction events occur between the pistil and the pollen grain/tube during this process. The pollen-stigma interaction is a highly selective process which leads to compatible or incompatible pollination. Previous studies in Solanaceae, Papaveraceae, and Brassicaceae provided some important insights into pollen-stigma recognition in self-incompatible systems. In recent years, considerable data have been available regarding pollen-stigma interaction during self-compatible pollination. In this review, we focus on discussing current knowledge on stigma factors that regulate pollen-stigma interaction in self-compatible systems in comparison with self-incompatible systems.  相似文献

研究了陆地棉洞A型核雄性不育系(抗A1,MA)不育和可育花药不同发育时期可溶性碳水化合物、游离氨基酸和IAA、GA3、ABA含量的动态变化。结果表明,不育花药中可溶性糖含量偏高,缺乏淀粉积累;有4种游离氨基酸含量在可育与不育花药问存在显著差异,其中不育花药天门冬氨酸含量偏高,这可能是其败育的原因之一,而脯氨酸、精氨酸、苯丙氨酸含量异常,则是不育花药败育的结果;在花药主要败育时期之前,不育花药中ABA含量极显著偏高,IAA、GA3含量极显著偏低。不育花药中IAA、GA3、ABA含量变化与洞A型核雄性不育系花药败育密切相关。  相似文献

Six plants of a self-incompatible population ofLeavenworthia çrassa were grown from seed collected in nature and cross-pollinated in all combinations. The incompatibility relationships between sibs were determined in eleven of the F1 families. A one-locus sporophytic incompatibility system was established. None of the parents was homozygous at the S locus. At least five, and possibly all six, of the parents did not share an S allele. Only one pair of alleles was shown to have different interactions in the pollen and stigmata. The identity and expression of the S alleles were determined in six families. Eight pairs of alleles were independently expressed in both the pollen grains and the stigmata. Sixteen pairs of alleles showed dominance of one allele over the other in the pollen grains or the stigmata or both.F1 plants of two crosses between different self-incompatible races were self-incompatible. F1 plants of six crosses between self-incompatible and self-compatible races were self-incompatible; in five of the families, the frequency of pseudo-compatibility was higher than in the self-incompatible parent. Self-incompatible hybrids from a cross between a self-incompatible and a self-compatible population provide a method for rapidly determining allelic interactions in plants with a sporophytic incompatibility system.The research was carried out at the Biological Laboratories, Harvard University, Cambridge, Massachusetts, U.S.A.  相似文献

Background and Aims

Selection may favour a partial or complete loss of self-incompatibility (SI) if it increases the reproductive output of individuals in the presence of low mate availability. The reproductive output of individuals varying in their strength of SI may also be affected by population density via its affect on the spatial structuring and number of S-alleles in populations. Modifiers increasing levels of self-compatibility can be selected when self-compatible individuals receive reproductive compensation by, for example, increasing seed set and/or when they become associated with high fitness genotypes.

Methods

The effect of variation in the strength of SI and scrub density (low versus high) on seed set, seed germination and inbreeding depression in seed germination (δgerm) was investigated in the partially self-incompatible species Flourensia cernua by analysing data from self-, cross- and open-pollinated florets.

Key Results

Examination of 100 plants in both high and low scrub densities revealed that 51% of plants were strongly self-incompatible and 49 % varied from being self-incompatible to self-compatible. Seed set after hand cross-pollination was higher than after open-pollination for self-incompatible, partially self-incompatible and self-compatible plants but was uniformly low for strongly self-incompatible plants. Strongly self-incompatible and self-incompatible plants exhibited lower seed set, seed germination and multiplicative female fitness (floral display × seed set × seed germination) in open-pollinated florets compared with partially self-incompatible and self-compatible plants. Scrub density also had an effect on seed set and inbreeding depression: in low-density scrubs seed set was higher after open-pollination and δgerm was lower.

Conclusions

These data suggest that (a) plants suffered outcross pollen limitation, (b) female fitness in partially self-incompatible and self-compatible plants is enhanced by increased mate-compatibility and (c) plants in low-density scrubs received higher quality pollen via open-pollination than plants in high-density scrubs.Key words: Flourensia cernua, population density, seed set, seed germination, female fitness, partial self-incompatibility, Mapimí Biosphere Reserve  相似文献

Summary A self-incompatible canola-quality Brassica napus ssp. oleifera line (W1) was generated by introgressing the S-locus from a self-incompatible B. campestris plant into the Westar cultivar. Using the polymerase chain reaction (PCR) with primers derived from conserved regions in S-locus glycoprotein (SLG) alleles, the central region of the active SLG gene (910) was obtained. The remaining portions of the cDNA for this 910 gene were subsequently cloned using the PCR-rapid amplification of cDNA ends (RACE) procedure. Sequence analysis revealed that the 910 cDNA show a high degree of sequence similarity to SLG alleles associated with Class I self-incompatible lines. The 910 gene was found to be absent in the original self-compatible cv. Westar (B. napus) and segregated with self-incompatibility in a mixed population generated from a cross between self-incompatible W1 and self-compatible Westar. RNA blot analysis indicated that high levels of 910 mRNAs were present in the stigma as buds approached anthesis. Thus, the SLG allele of W1 transferred from B. campestris via backcrosses to a line of cv. Westar has been identified.  相似文献

Self-compatibility segregation was assessed in two successive backcross progenies originating from an interspecific cross between Coffea canephora (self-incompatible) and Coffea heterocalyx (self-compatible). After self- and cross-pollination, pollen tube behaviour in styles was observed under ultraviolet fluorescence microscopy and fruit-set was determined at harvesting time. Segregation ratios in the two progenies were consistent with monofactorial control of self-compatibility. Self-compatible plants exhibited higher fruit-set than self-incompatible ones in open-pollination conditions. Segregation of AFLP markers was scored in the first backcross progeny. By molecular linkage analysis, the S locus could be mapped to a short linkage group.  相似文献

An S-receptor kinase (SRK) cDNA, SRK-910, from the active S-locus in a self-incompatible Brassica napus W1 line has been isolated and characterized. The SRK-910 gene is predominantly expressed in pistils and segregates with the W1 self-incompatibility phenotype in an F2 population derived from a cross between the self-incompatible W1 line and a self-compatible Westar line. Analysis of the predicted amino acid sequence demonstrated that the extracellular receptor domain is highly homologous to S-locus glycoproteins, whereas the cytoplasmic kinase domain contains conserved amino acids present in serine/threonine kinases. An SRK-910 kinase protein fusion was produced in Escherichia coli and found to contain kinase activity. Phosphoamino acid analysis confirmed that only serine and threonine residues were phosphorylated. Thus, the SRK-910 gene encodes a functional serine/threonine receptor kinase.  相似文献

Fumana ericifolia belongs to the family Cistaceae, in which all perennial species previously studied have been reported to be self-incompatible and xenogamous. Here we show that F. ericifolia is self-compatible and autogamous. Its flowers last only four to eight hours and produce a small amount of pollen and ovules. Self-pollination depends on changes in the relative position of the stigma and the anthers, triggered by the abscission of the petals; this in turn causes closing of the sepals, which push the anthers onto the stigma. Pollen remains highly viable and germinable, and the stigma keeps its receptivity several hours after the loss of the petals. Hand pollination treatments revealed that fruits are produced both in selfed and crossed flowers. The germination of pollen grains on the stigma, the rate of pollen tube growth along the style, and the penetration into the ovules were similar in self-and cross-pollinations; also, there were no significant differences in fruit set, seed set and weight of seeds. Thus, we report the first evidence of autogamy in a woody Cistaceae and suggest that selfing may ensure reproductive success when pollinators are limited.  相似文献

用350Gy剂量的Co^60-γ射线照射水稻三系保持系813B种子,获得巨胚突变体,命名为巨胚813B(简称813geB)。用巨胚813B回交813A育成特种稻不育系——巨胚813A(简称813geA)。对巨胚813B及巨胚813A的生物学特性进行研究,结果表明:巨胚813A与813A的主要农艺性状、雄性不育性及产量构成因素均无明显差异。巨胚813B与813B具有相似的农艺性状,但绝对胚重由0.61mg提高到1.36mg,相对胚重由2.70%提高到6.96%。巨胚813B糙米的蛋白质、粗脂肪、粗纤维及必需氨基酸含量均比813B高。其中蛋白质含量9.77%,比813B提高了8.80%;粗脂肪含量5.28%,比813B提高了87.23%;8种必需氨基酸含量均有提高。利用巨胚恢复系与巨胚813A可纽配成巨胚杂交稻。  相似文献

采用盐酸水解法分析了福建省建阳主栽的7个山药品种的氨基酸含量及组成。得出供试材料中都含有17种氨基酸,总氨基酸质量分数在2.86~6.64%,平均4.95%,其中以JY-4品种总氨基酸质量分数最高,JY-2最低;各种氨基酸质量分数高低顺序基本相似,以精氨酸、谷氨酸、天冬氨酸居前三位,胱氨酸质量分数最低,必需氨基酸质量分数的高低依次是亮氨酸>苯丙氨酸>赖氨酸>缬氨酸>异亮氨酸>苏氨酸>蛋氨酸;鲜味氨基酸在各品种间的差异显著,以JY-4最高,达3.41%,依次是JY-3、JY-6、JY-1、JY-5、JY-7、JY-2。再运用聚类分析方法从氨基酸的组成及含量角度对7个品种进行分类可分为三类;相关分析表明:山药总氨基酸含量愈高,必需氨基酸和鲜味氨基酸含量亦愈高。  相似文献

To develop inbred lines from self-incompatible, cultivated diploid potatoes, an S-locus inhibitor (Sli) gene derived from a self-compatible variant of a wild potato species, Solanum chacoense, was incorporated into various cultivated diploid potatoes. The progeny was selfed twice by the action of the Sli gene to obtain 74 S2 inbred clones belonging to 8 families. More than 40% of them were either non-flowering or pollen sterile. Among the pollen fertile clones, self-compatible clones occurred with a much lower frequency (20.9%) than expected (83.3%). The result demonstrated that self-compatibility was introduced and expressed in the gene pool of cultivated diploid potatoes by an action of the Sli gene, although serious inbreeding depression associated with selfing occurred. The genotypes of S2 inbreds were surveyed using 46 S. chacoense-specific RFLP (restriction fragment length polymorphism) markers covering the whole potato genome. More than half of the markers (67.4%) showed distorted segregation. Particularly, all markers on chromosome 12 were overrepresented in the S2 inbreds. This confirms our earlier finding that the Sli gene locates on chromosome 12 and the alleles linked with this gene are preferentially transmitted because of its essential requirement for selfing.  相似文献

Self-incompatibility in Solanaceae is controlled by a single multiallelic locus, the S-locus. The S-allele associated ribonucleases (S-RNases) in the pistil are involved in pollen rejection. In this work, we analyzed two newly isolated lines of Petunia hybrida, termed PB and PF. They both had the same set of S-RNases (SB1- and SB2-RNases), however the PB was a self-incompatible diploid while PF was a self-compatible tetraploid. Cross pollination tests between PB and PF indicated diploid pollen from PF lost the incompatibility phenotype. In order to clarify the effects of polyploidy on pollen phenotypic change, we artificially induced tetraploid plants from a diploid SB1SB2 heterozygote (= PB) and a diploid SB1SB1 homozygote. The obtained SB1 SB1SB1SB1 homoallelic tetraploid remained self-incompatible, whereas the SB1SB1SB2SB2 heteroallelic tetraploid became self-compatible. These data suggested that the diploid heteroallelic pollen lost the incompatibility phenotype and had the characteristics of self-compatibility with SB1SB2 style.  相似文献
