Calli produced from stem segments of seedling of Coriandrum satwum which were cultured on MS agar medium containing NAA 1.0mg/L. The embryogenic cell colony suspension was estabilished on MS liquid medium containing NAA 1.0mg/L%2,4-D 0.2mg/L+BA 0.5 mg/L. The cell suspension culture was used for protoplast preparation. Protoplasts were obtained in the enzyme mixture containing 2.0% Onozuka R-10, 1.0% pectinase, 0.5% snailase, 0.5% dextran sulfate potassium Salt, 0.6mol/L mannital CPW solution at pH 5.8 and 25℃. Cultured in a KM8P liquid medium containing NAA 1.0mg/L+2,4-D 0.2mg/L+6-BA 0.5 mg/L, glucose 0.4mol/L and CM 20mi/L; the protoplasts entered the stage of derision after three days, cell clusters formed in 10 days and calli formed after about 50 days. When the calli were transferred to MS agar medium containing many growth substances, they differentiated into embryoids, and then developed into plantlet with many green leaves and roots on the 1/2 MS agar medium.  相似文献

Calli were induced and suspension cell lines were set up from ovule of Malus pumila Mill. Protoplasts (5.40 × 106/g fr. wt) were isolated from suspension cell lines in enzyme mixture solution containing 2.0% Onozuka R-10, 0.5% pectinase, 0. 65 mol/L mannital, 0.01 mol/L CaC12, 0.7 mmol/L KH2 PO4, 0.3% dextran sulfate potassium salt, at pH 5.8 for 6 h at 26℃. The cell clumps were formed from protoplasts cultured in modified MS, K8p, D2 media. Calli were formed on MS solid medium containing 2.0 mg/L IAA, 2.0 mg/L NAA, 0.1 mg/L BA. Shoots were differentiated on differentiated medium after several changes of the medium. Eventually, shoots rooted and developed into whole plantlets on a rooting medium.  相似文献

Calll with many embryogenic cell colonies were produced from segments of seedlling of Peucedanum terebinthaceum (Fisch.) Fisch. ex Turcz. which were cultured on the 1/2MS agar medium (with half quantity of macronutrients) containing 1 mg/l 2,4-D. Cell suspension culture with high percentage of embryogenic cell colonies was established from the calli shaking in liquid medium. The cell suspension culture was used for protoplast preparation. Protoplasts were obtained with the enzyme mixture containing 1.5% Onozuka R-10, 0.3% Macerozyme R-10, 0.5% Snailase, 5 mmol/l CeCl2, 1 mmol/l KH2PO4, 0.6 mol/l mannital at pH 5.8 and 25℃. Cultured in a modified MS liquid medium containing 1 mg/l 2,4-D+ 0.5 mg/l zeatin, the protoplasts emered division after four days, and formed cell colonies of 0.5–1mm after about forty days. When transfered to 1/2 MS liquid medium supplemented with zeatin (0.5 mg/l), the cell colonies differentiated in to embryoids, then developed into plantlets with many green leaves and roots on the 1/2 MS agar medium devoid of phytohormones.  相似文献

Calll were initiated from the seedling segment of Peucedanum praeruptorum Dunn and subcultured on the MS agar medium with 0.5 mg/L 2,4-D. Cell suspension culture with a lot of embryogenic cell clumps was obtained in liquid medium. Protoplasts were isolated from the cell clumps in enzyme mixture solution containing 1.5% Onozuka R-10, 0.3% Macerozyme R-10, 0.5% helicase, 5 mmol/L CaCl2 and 0.6 mol/L mannital, at pH 5.6 and shaking for 5- hours at 25℃. Helicase is necessary for isolation. After purified by washing, the protoplasts were cultured in liquid medium containing 1 mg/L 2,4-D +0.5 mg/L zeatin. First cell division was observed after four days. Large cell clumps were formed after thirty days. Microcalli of 1 mm in size was formed after about fifty days, and continued to grow on the MS solid medium containing 0.5 mg/L 2,4-D and 200 mg/L casein hydrolysate, and later differentiated into embryoids when transferred to MS agar medium with 0.1 mg/L zeatin. Eventually, embryoids developed into whole plantlets on the MS solid medium without phytohormones.  相似文献

Protoplasts from 4-day-old embryogenic cell suspension cultures of Astragalus adsurgens, when cultured in KM8P medium which ammonium concentration was reduced to 2.5 mmol/L and supplemented with 0.5 mg/L NAA, 1.0 mg/L 2, 4-D, 0.7 mg/L BA and 0.4 mol/L glucose, underwent cell sustained divisions and formed cell colonies at a frequency of 16%-20%. Preplasmolysis or low temperature treatment of suspension cells prior to enzyme incubation enhanced colony formation. Following proliferation on MS medium containing 1.0 mg/L 2, 4-D and 0.5 mg/L BA, cell colonies were cultured on MS medium containing 0.1 mg/L NAA and 1.0 mg/L BA, where approximately 40% of colonies produced somatic embryos ranging in number from 20 to 40 per colony. No significant decrease was found in the potential of somatic embryogenesis when protoplast colonies were obtained from long-term cell suspensions. On hormone-free 1/2 MS medium, somatic embryos developed into intact plants, which showed normal morphology and stable chromosome number.  相似文献

The hypocotyls of the embryoid derived plantlets of Ligusticum wallichii Franch were used for protoplast preparation. Protoplasts were obtained with the enzyme mixture containing 1.5% Onozuka R-10, 0.3% Macerozyme R-10, 0.5% Snailase, 5 mmol/l CaCl2, 1 mmol/l KH2PO4, 0.6 mol/l manitol, at pH 5.6–5.8 and 27℃. Protoplasts were cultured in a modified MS liquid medium containing 1 mg/l 2,4-D + 0.5 mg/l 6- BA. The first divisions were found after twelve days, and the dividing cells formed cell colonies of 0.5–1 mm after about fourty days. When they were transferred to MS agar medium (with half quantity of macronutrients) supplemented with 2,4-D (0.5mg/l) and 6-BA(0.5mg/l), they grew into calli. At last, on the medium without any phytohormones, the growing calli differentiated embryoids which developed into plantlets with many green leaves and roots.  相似文献

以继代培养5—24代的沙打旺(Astra-galus adsurgens Pall.)胚性悬浮系为材料,从生长4d的细胞培养物中可游离出大量有活力的原生质体。细胞预质壁分离或低温预处理对原生质体得率和活力没有影响,但可提高原生质体培养的植板率。预处理的原生质体培养在NH_4NO_3浓度降至2.5mmol/L,并附加0.5mg/L NAA、1.0mg/L 2,4-D、0.7mg/L BA和0.4mol/L葡萄糖的KM8P培养基中,经持续分裂形成细胞克隆,植板率高达16%—20%。细胞克隆在附加1.0mg/L 2,4-D和0.5mg/L BA的MS培养基中增殖后,转至含0.1mg/L NAA和1.0mg/L BA的MS分化培养基中可形成体细胞胚,平均体细胞胚发生频率约为40%,每个克隆产生20—40个体细胞胚。在无激素的1/2MS培养基中,体细胞胚发育成形态正常和染色体数稳定的小植株。  相似文献

Embryogenic cell line was established from hypocotyl segments of Codonopsis pilosula (Franch.)Nannf. 4--8 day old embryogenic callus was used to isolate protoplasts in an enzyme solution containing 1.5 % cellulase Onozuka R-10 and 3 % pectinase. Protoplasts were cultured in MS,C81V,DPD and KMSp basal medium supplemented with 1.2 mg/L 2,4-D, 0.2 mg/L NAA, 0. 2 mg/L BAP, 0. 1 mg/L ZT,and different combinations of glucose and mannitol . Protoplast-derived cells underwent sustained divisions in KM8p medium. As an osmoticum, glucose was more beneficial to protoplast division. A combination of 0. 30 mol/L glucose with 0.10 mol/L mannitol gave the best result. Under proper conditions , protoplasts underwent the first division on the 3rd day of culture,formed colonies within 30 days , and developed into microcalli in 6 weeks. Plantlets were regenerated from protoplast-derived calli through somatic embryogenesis. 0.2 % activated charcoal promoted embryoid formation and root development.  相似文献

Protoplasts isolated from suspension cell lumps of Medicago lupulina L. started to divide after 2 clays in K8p culture medium containing 0. 1~2.0 mg/L of 2, 4-D, with a maximum division frequency of 38. 35%. After S weeks of culture, the protoplast-derived cell lumps were transferred to liquid/solid double-layer media for microcallus regeneration, with a maximum frequency of 0.58%. The whole plants were regenerated from protoplastderived calli via somatic embryogenesis and organogenesis. In somatic embryogenesis, the embryoids were induced on MS and W14 media with rather wide range (1. 0420.0 mg/L) of 2, 4-D concentration. The highest induction frequency of embryoids was 71.0%. In organogenesis, the differentiation media containing lower concentration of 6-BA (0. 5~0. 7 mg/L) were suitable for adventitious bud formation. The highest frequency of adventitious bud formation from calli was 27. 8%. The mature protoplast-regenerated plants were obtained 3 months after transplanting the plantlets into soil.  相似文献

Embryogenic calli were produced from the segments of the young roots, hypocotyls or petioles of test-tube seedlings on MS agar medium containing 1 mg/1 2,4-D. When shaken in the MS liquid medium, the calli formed cell suspension with many embryogenic cell clumps.Using the enzyme mixture: Onozuka R-10 1.5%+MacerozymeR-10 0.3%+Snailase 0.5%+CaCl2 5 mmol/l + Mannitol 0.6 mol/1 (pH=5.8), protoplasts were obtained from the cell clumps which had been subcultured for three to' seven days. When cultivated, the protoplasts grew and began to divide after four days, and formed cell clumps about l—2 mm within fifty days. Protoplast-derived calli were formed from the cell clumps on the MS agar medium with 0.5 mg/l 2,4-D. When transferred onto the MS agar medium containing 0.1 mg/1 6-BA or 0.1 mg/1 2,4-D and 0.5 mg/1 6-BA, the calli differentiated into embryoids. Heracleum moellendorffiz Hance is a herb belonging to Umbelliferae used in traditional medicine in China. The young stem-nodes were induced for callus formation on MS medium containing 1 mg/L 2,4-D. After subcultured for about five months, the embryogenic calli were used for cell suspension culture. The protoplasts were prepared from this suspension by digestion with enzyme mixture containing 1. 5% cellulase Onozuka R-10 +0. 3% macerozyme R-10 + 0. 5% snailase + 5 mmol CaCl2 + 0. 6 mol/L mannitol, at pH 5.8, and cultured in modified MS and modified N6 media with 0.3 % agarose. They divided after 3 days and developed into small cell colonies after about 2 weeks. From this time on, the glucose concentration in the culture media was decreased to 0. 2 mol/L,which led to futher growth of the colonies to small calf . After a period of proliferation on solid medium with 0. 5 mg/L 2,4-D, the calli were transferred to a medium with 0. 1 mg/L zeatin on which somatic embryos differentiated and developed to plantlets  相似文献

Protoplasts were successfully isolated from internodal callustissues of both Oxalis glaucifolia and O. rhombeo-ovata whenthey were digested in a solution containing 0.1% (w/v) MacerozymeR-10, 0.5% (w/v) cellulase Onozuka R-10 and 0.3 mmol m–3sucrose. Protoplasts proliferated to give cell colonies on Gamborget al.'s B5 medium supplemented with 0.3 mmol m–3 mannitol,0.5 mg dm–32, 4-D, and 2.0 mg dm–3 kinetin. Calluswas produced upon transfer of cell colonies to Murashige andSkoog medium containing 2.0 mg dm–3 l-naphthaleneaceticacid (NAA) and 0.1 mg dm–3 kinetin for O. glaucifolia,or with 5.0 mg dm–3 NAA and 0.5 mg dm–3 6-benzylaminopurine,for O. rhombeo-ovata. Plants were regenerated from O. glaucifoliaprotoplasts on a medium containing 0.1 mg dm–3 NAA, 1.0mg dm–3 kinetin and 1.0 mg dm–3 gibberellic acid,but only vascular nodules were differentiated by O. rhombeo-ovataprotoplast-derived calli. 外植体类型和光照条件决定沙打旺胚性愈伤组织的形成。用生长10d的胚性愈伤组织可分离到1.2×106个/g(原生质体/细胞),活力超过80%。当原生质体以1.0×105/mL的植板密度培养在含0.6%琼脂糖附加1.5mg/L 2,4-D、0.5mg/L BA和0.5mol/L葡萄糖的培养基(无机盐降为1/4)中,植板率为16.8%。条件培养基显著促进原生质体的生长发育。长大的细胞克隆经2周4℃低温处理后转到含0.1mg/L NAA和1.0mg/L BA分化培养基上,体细胞胚胎发生频率高达70%,每克细胞产生的体细胞胚数在200个以上。成熟的体细胞胚转到无激素的1/2MS培养基中即分化成苗,再生植株为正常的二倍体。  相似文献

Protoplasts were isolated from hypocotyls of 7-d-old seedlings of three genotypes of Brassica carinata after enzymatic digestion in cellulase R-10 (0.5%) and pectolyase Y-23 (0.025%). The protoplasts were stabilized with 0.4 M mannitol used as osmoticum, and were cultured in darkness in Kao's liquid medium containing 0.4 M glucose and the growth regulators 2,4-D (1.0 mg/l), NAA (0.1 mg/l) and zeatin riboside (0.5 mg/l). Protoplasts were transferred to 16 h photoperiod conditions after 3 d of dark culture, and the medium was diluted to reduce the osmoticum on the seventh and tenth days of culture. Microcolonies were thus obtained which, upon transfer to MS agarose medium with 2,4-D (0.1 mg/l), BAP (1 mg/l) and 0.1 M sucrose, proliferated further to produce callus clumps. The plating efficiency of the three genotypes varied from 1 to 2%. Calli 2–3 mm in diameter were transferred to MS agarose plates with zeatin (2 mg/l) where they produced shoot buds and shoots with frequencies ranging from 22.5 to 74.2% for the three genotypes. The shoots were rooted in medium with IBA (1 mg/l) and were then established in soil. Protoplasts isolated from hypocotyls of three-day-old seedlings of Brassica carinata (Braun) cv R-2128 were cultured in a modified Nitsch and Nitsch liquid medium containing 13% sucrose, 0.4% Ficoll, 0.25 mg/l BA, 0.5 mg/l NAA and 0.5 mg/l 2,4-D. The density of medium caused the protoplasts and the developing microcalli to float on the surface of the liquid medium whereas all debris and lysed cells sank to the bottom of the culture plate. After 4–6 weeks developing microcalli were approximately 0.5 mm in diameter and were transferred onto MS medium containing 3% sucrose, 0.4% agarose, 200 mg/l casein hydrolysate, 5 mg/l BA and 0.5 mg/l NAA, pH 5.7. 埃斯基红豆幼苗的下胚轴切段在附加2,4-D0.5mg/L,KT1mg/L的MS中形成胚性愈伤组织。来自11-13个月龄、继代6-15天的愈伤组织的原生质体,在改良的V-KM液体培养基中可持续分裂形成细胞团,培养10天时的分裂率和克隆率分别为65.88%和53.38%周后就可将将原生质体形成的小愈伤组织转于培养基上。原生质体在改良的B5液体培养基也可以分裂形成小愈伤组织,但分裂率低于V-KM。来自原  相似文献

Results of observation showed that the female gametophyte of Ginkgo biloba was at the coenocytic stage from March 30ty to May 30ty. A density of 6-8 x 10(5) protoplasts/ml with a viability of 87.3% was achieved when the female gametopytes collected from May 8th to 15th were treated with 0.5% cellulase Onzuka R-10, 0.5% Pectolyase for 4-5 hours. The thin-layer liquid Murashige and Tuker medium modified by omitting ammonium ions and supplementing with glutamine 1000 mg/L, Vc 5 mg/L, benzyladenine 1.0 mg/L and naphthaleneacetic acid 3.0 mg/L was used for the protoplast culture and multiple cell colonies were obtained.  相似文献

Protoplasts were prepared from exponentially growing cell suspension cultures derived from Pinus taeda seedlings. The cell suspension was derived from callus that had been initiated on solid Murashige and Skoog medium supplemented with 2mg/1 2,4-D, and maintained in liquid LM medium (D.C. Verma et al, Proceedings of the 5th International Congress of Plant Tissue and Cell Culture, Tokyo, Japan, 1982, p. 59–60). Evaluation of a range of enzyme formulations showed that almost all cells could be converted to viable protoplasts in 9 hours with the combination of 0.5% each of Onozuka R-10, Macerozyme R-10, Hemicellulase, Driselase, and bovine serum albumin in osmoticum (0.4 M mannitol, 0.2 M sorbitol, 1.5 mM CaCl2, 0.7 mM KCl). Protoplast purification required only simple washing which was achieved without damaging centrifugation. When cultured in LM medium supplemented with 1.5mM CaCl2, 10 mM arginine, 10mM glutamine and 5 mM asparagine, cell wall formation was observed after 48 hours. The cultured protoplasts produced numerous colonies in 3 weeks which continued to grow in liquid medium.  相似文献
