通过对东亚七筋姑(Clintoniaudensis Trautv.etMey.)19个居群的cpDNA基因间隔区(cpDNA intergenic regions)rpl20-rps12和trnL-F进行序列分析,结果表明,当空位(Gap)作缺失处理时,trnL-F全序列排序后的长度为754个位点,G C含量为35.2%~35.8%,rpl20-rps12序列排序后长度为814bp,G C含量为34.1%~34.3%.聚类结果显示四倍体居群没有单独聚为一支,其多倍体可能是同源多倍体,起源方式属于异地多起源.  相似文献

Fifteen species in six genera of the family Liliaceae were karyomorphologically studied. They share the complex chromocenter type of the resting nuclei and the interstitial type of the prophase chromosomes in somatic cells except that Clintonia udensis Trautv. et Mey is of the densely diffuse type and gradient type respectively. 用扫描电镜对七筋姑属北美全部4个种和东亚七筋姑不同产地的11个居群进行了种皮微形态特征的观察和研究,结果表明,北美4种中,Clintonia umbellulata以其种皮近于平滑、表面分布有大小不等的凹坑而不同于其它各种具条纹状或网穴状的表皮纹饰。北美西部的2个种C.uniflora和C.andrewsiana种皮纹饰相似,关系密切;而北美东部的C.borealis与东亚种C.udensis长白  相似文献

Summary At the second generation of the interspecific cross between the cultivated hexaploid (2n = 42) oat A. Sativa and the wild tetraploid (2n = 28) A. Murphyi, a plant having hexaploid and diploid (2n = 14) sectors was selected. Meiosis was highly regular in the diploid sector but the tillers failed to proceed beyond the boot stage and no seeds were produced. It is suggested that this diploid sector represents an entire genome of one of the diploid progenitors of the hexaploid oat.  相似文献

The efficiency of an original approach to involvement of the valuable genetic pool of wild diploid potato species from Mexico is estimated. The essence of this method is in generation of dihaploids (2n = 2x = 24) of tetraploid somatic hybrids (2n = 4x = 48) followed by backcrossing with dihaploids of Solanum tuberosum. A haploid producer, S. phureja IvP35, was used to generate ten dihaploids of S. tuberosum + S. pinnatisectum, all of which crossed with fertile S. tuberosum dihaploids and developed plump viable seeds. This gives the possibility of an efficient introgression of the genes valuable for breeding from wild species to the bred plants at a diploid level, which has several advantages compared with the corresponding procedure at a tetraploid level. A part of the dihaploids produced was compatible (the pollen tubes reached the ovary) with diploid and tetraploid forms of S. pinnatisectum; however, no viable seeds were developed. The attempt to generate the dihaploids of S. tuberosum + S. bulbocastanum somatic hydrides using the haploid producer S. phureja IvP35 was unsuccessful.  相似文献

The gametic chromosome numbers of sevenHymenasplenium (Aspleniaceae) species from Xishuangbanna, Yunnan Prov., China, were investigated. All the examined individuals ofH. obscurum, H. cheilosorum andH. latipinnum were sexual diploids with n=39 chromosomes. Intraspecific cytological variation was found inH. excisum, which has a sexual diploid (n=39) and a tetraploid (n=78). Only a triploid apogamous cytotype (n=ca.117) was found inH. laterepens. Hymenasplenium apogamum showed the most complicated intraspecific variation and included a sexual diploid (n=39), a sexual tetraploid (n=78) and an apogamous triploid (n=ca.117). This work reports for the first time the sexual diploids ofH. cheilosorum andH. apogamum, which are only apogamous elsewhere in east Asia, Himalayas and Indochina. These results may indicate that this area is one of the diversity centers ofHymenasplenium. Most of the above species have chromosome numbers based on x=39. In contrast,H. costarisorum contains a sexual diploid (n=36) and a sexual tetraploid (n=72), indicating that its basic number is x=36.  相似文献

六倍体普通小麦(Triticum aestivum L.)是由四倍体小麦(T.turgidum L.)与二倍体节节麦(Aegilops tanschii Coss.)天然杂交然后通过染色体自然加倍形成的异源多倍体.这一起源过程是自然条件下天然发生的,它的发生需要具备一个条件:四倍体小麦与节节麦的天然杂交种子在自然条件(没有幼胚培养等)下能够正常发芽出苗.我们从22份节节麦中发现来自中东的节节麦AS60在不采用幼胚培养等人工辅助条件下,仍然很容易与四倍体小麦和普通小麦产生有生活力的杂种植株.AS60与四倍体小麦的杂交种子有50.0%(反交)及57.1%(正交)的种子,而AS60与六倍体普通小麦的杂交种子则有45.5%不需幼胚培养等措施能够正常发芽、生长.AS60的这一特征正是普通小麦起源过程需要的条件.最后探讨了这一发现对小麦遗传改良和对普通小麦起源演化研究的意义.  相似文献

Chromosome pairing in females and males of diploid (2n = 22) and tetraploid (2n = 44) Odontophrynus americanus and diploid Ceratophrys cranwelli (2n = 26) and tetraploid C. ornata (2n = 104) showed that diploid females formed more chiasmata per paired arm than diploid males and polyploids of both sexes. There was a reduction in the level of recombination in female polyploids by forming multivalents with terminal chiasmata. The reduction reflected a change in the genetic control of pairing in females after polyploidization.  相似文献

Polyploidy is an important factor shaping the geographic range of a species. Clintonia udensis (Clintonia) is a primary perennial herb widely distributed in China with two karyotypic characteristics—diploid and tetraploid and thereby used to understand the ploidy and distribution. This study unraveled the patterns of genetic variation and spatiotemporal history among the cytotypes of C. udensis using simple sequence repeat or microsatellites. The results showed that the diploids and tetraploids showed the medium level of genetic differentiation; tetraploid was slightly lower than diploid in genetic diversity; recurrent polyploidization seems to have opened new possibilities for the local genotype; the spatiotemporal history of C. udensis allows tracing the interplay of polyploidy evolution; isolated and different ecological surroundings could act as evolutionary capacitors, preserve distinct karyological, and genetic diversity. The approaches of integrating genetic differentiation and spatiotemporal history of diploidy and tetraploidy of Clintonia udens would possibly provide a powerful way to understand the ploidy and plant distribution and undertaken in similar studies in other plant species simultaneously contained the diploid and tetraploid.  相似文献

对鸡导山地区7个多星韭种群的生物生态学特性进行了比较全面的研究。结果表明;鸡足山地区的多星韭有二倍体和四倍体2种倍性。多星韭二倍体比四倍体更能忍受较恶劣的环境条件。二倍体多星韭生境单一,种群分化不明显;四倍体多星韭生境多样,种群间变异丰富,分化强烈。  相似文献

A tetraploid (2n = 28) clone of Fragaria from SW Finland is described. The clone is true-breeding with respect to tetraploidy. The tetraploid strawberry shows RFLP similarity with F. vesca , a common species, and, to a lesser extent, with F. viridis , a southern species, whose nearest reported occurrence was about 80 km away during the 1940s. The following alternatives are considered as possible reasons for the tetraploidy: unreduced gametes in an interspecific hybrid or hybrid derivative; hybridization followed by induction of tetraploidy either by viruses, or by industrial pollutants, e.g. chlorophenols and their congener compounds in the soil. The original seed might also have been brought from abroad unintentionally with other seeds for field trials which have been conducted since 1928 in the vicinity. A hexaploid ( F. moschata )× diploid hybrid leads directly to tetraploidy and a tetraploid derivative from an octoploid ( F . × ananassa ) × diploid hybridization can be obtained but the current tetraploid has different morphology.  相似文献

In the loach, or Oriental weatherfish Misgurnus anguillicaudatus (Teleostei: Cobitidae), diploid (2n = 50) and tetraploid individuals (4n = 100) are often sympatric in central China. The evolutionary mechanism of this tetraploidization was analyzed with the observation of meiotic behavior of chromosomes in both the germinal vesicles of mature oocytes and the primary spermatocytes in diploid and tetraploid loaches. Whereas diploid specimens usually showed 25 bivalents in meiotic cells, tetraploid loaches exhibited 0-6 quadrivalents and 38-50 bivalents in both sexes, with the modal number of quadrivalents as three in females and four in males. In the diploid specimens, the two largest metacentric chromosomes bearing nucleolar organizing regions (NORs) identified by chromomycin A(3) staining and fluorescence in situ hybridization with a 5.8S + 28S rDNA probe formed one bivalent with terminal association. In the tetraploids, four NOR-bearing chromosomes never formed a quadrivalent, but were organized into two terminally-associated bivalents. These findings suggest an autotetraploid origin of the natural tetraploid loach and subsequent rediploidization of whole genome. The latter process, however, seems still in progress as inferred from the concurrence of up-to several quadrivalents and the majority of bivalents.  相似文献

Few studies of cpDNA have provided evolutionary and/or phylogenetic information at the intraspecific level. We analyzed restriction site variation using 19 endonucleases in 37 populations representing both diploid (2n = 14) and autotetraploid (2n = 28) Tolmiea menziesii. Seven restriction site mutations and five length mutations were observed. Although diploid and tetraploid Tolmiea have been intensively studied using nuclear markers, cpDNA variation provided additional evolutionary insights not revealed previously. The chloroplast genomes of diploid and tetraploid Tolmiea are as distinct as those of many pairs of congeneric species of angiosperms. Based on outgroup comparisons, the primitive chloroplast genome is present in tetraploid rather than diploid Tolmiea. These findings suggest that either: (1) diploid and tetraploid Tolmiea may have diverged since the origin of the autotetraploid, (2) the original diploid donor of the cytoplasm present in the tetraploid subsequently became extinct, or (3) the diploid was actually derived from the tetraploid via polyhaploidy. cpDNA variation also revealed that despite their close geographic proximity, diploid and tetraploid Tolmiea do not experience cytoplasmic gene flow. Last, three cytoplasmically distinct groups of diploid populations exist, two of which occupy distinct geographic areas. These findings demonstrate that, at least in some plant species, restriction fragment analysis of cpDNA can provide important evolutionary and phylogenetic information at low taxonomic levels.  相似文献

Nuclear DNA contents were determined by flow cytometry for 20 populations of the perennial C4 shrub Atriplex halimus L. (Chenopodiaceae) originating from the Mediterranean basin and Fuerteventura (Canary Islands). Two populations were also analysed for chromosome number: one (from Ibiza, Spain), with a 2C nuclear DNA content of 2.40 pg, was shown to be diploid (2 n  = 2 x  = 18), whilst the other (from Sicily, Italy), with 5.11 pg, was tetraploid (2 n  = 2 x  = 36). With respect to nuclear DNA content, two groups of populations were detected, diploids with 2.40–2.44 pg and tetraploids with 4.77–5.13 pg. The diploid populations were mainly from the western Mediterranean (Spain and France) and Fuerteventura, whereas tetraploids were generally, but not exclusively, from more arid areas in North Africa and the eastern Mediterranean. In general, the diploid and tetraploid populations corresponded to the subspecies halimus and schweinfurthii , respectively. For certain populations having morphologies intermediate between those considered typical of these two subspecies, nuclear DNA contents showed them to be tetraploid. The karyotypes of 3 species of Ceratoides (C. arborescens, C. ewersmanniana and C. latens) were studied. The results show that C. arborescens is a diploid, with karyotype formula 2n=2x=18=16m+2sm, C. ewersmanniana is also a diploid, with 2n=2x=18=14m+4sm, while C. latens is a tetraploid, with 2n=4x=36=24m+12sm. According to the karyotypes, the morphological features and geographical distribution, C. arborescens seems to be a primitive species and it might have originated in northern China. C. arborescens and C. ewersmanniana are similar to each other, not only in morphology but also in karyotype, which shows that they are closely related and that C. arborescens might be the progenitor of C. ewersmanniana. The karyotypes of the 3 speciesof Ceratoides are basically identical, with only minor differences.  相似文献

The present papar is a report on cytotaxonomical studies of Allium Sect. Caloscordum(Liliaceae)from China. Six populations of A. neriniflorum and A.tubiflorum were introduced from Shaanxi,Hebei(Beijing),Liaoning and Heilongjiang.Based on the introduction and cultivated experiment, the authors made a karyotype analysis and a morphological comparison of the six populations. The results are as follows. Except the population 5 (Baihuashan,Beijing),which was found to be tetraploid 2n=4x=32,all the populations were dipliod 2n=2x=16. There were two types of satellites: terminal and intercalary. The basic number,the karyotypes as well as the types of satellites in Sect. Caloscordum were the same as those in the other sections of Allium. In the populations 1, 2, 3 and 4(Daheshan,Jinxia County,Liaoning;Taikang County, Heilongjiang; Chengde to Huaqing, Hebei; Liulimao, Huairou,Beijing.), the sixth pair of chromosomes had a pair of intercalary satellites,the mean length of pedicels is about 7 cm and the plant had no two-growth cycle.In the populations 6 (Xianyunei, Huashan, Shaanxi): the eighth chromosome pair had a pair of terminal satellites, the pedicel was shorter,with the mean length 2.39 cm,and the plant had two-growth cycle. In the tetraploid population 5; fourteenth and sixteenth chromosome pairs both had a pair of terminal satellites,the mean length of pedicel was 2.39 cm and the plant had the two-growth cycle.According to the results,the authors keep Sect. Caloscordum in the genus Allium and recognize two species in China in the section; the populations 1, 2, 3 and 4 belong to A.neriniflorum,and the populations 5 and 6 to A.tubiflorum.Our observation on dissecting ovary showed that there were 2~11 ovules per locule;the diploid populations mainly had 5 ~6 ovules per locule,while the tetraploid ones mainly had 4 ovules per locule.Therefore, the number of ovules per locule is not a reliable character for classifiction but it may be useful in distinguishing diploid from tetraploid.However,we consider that the satellite type,the pedicel length and whether the plant has two-growth cycle are the diagnostic features for the species of Sect.Caloscordum.  相似文献

BACKGROUND AND AIMS: Nuclear DNA amounts of 12 diploid and one tetraploid taxa and 12 natural interspecific hybrids of Cirsium from 102 populations in the Czech Republic, Austria, Slovakia and Hungary were estimated. METHODS: DAPI and PI flow cytometry were used. KEY RESULTS: 2C-values of diploid (2n = 34) species varied from 2.14 pg in C. heterophyllum to 3.60 pg in C. eriophorum (1.68-fold difference); the 2C value for the tetraploid C. vulgare was estimated at 5.54 pg. The DNA contents of hybrids were located between the values of their putative parents, although usually closer to the species with the smaller genome. Biennial species of Cirsium possessed larger nuclear DNA amounts than their perennial relatives. Genome size was negatively correlated with Ellenberg's indicator values for continentality and moisture and with eastern limits of distribution. A negative relationship was also detected between the genome size and the tendency to form natural interspecific hybrids. On the contrary, C-values positively corresponded with the spinyness (degree of spinosity). AT frequency ranged from 48.38 % in C. eriophorum to 51.75 % in C. arvense. Significant intraspecific DNA content variation in DAPI sessions was detected in C. acaule (probably due to the presence of B-chromosomes), and in tetraploid C. vulgare. Only the diploid level was confirmed for the Pannonian C. brachycephalum, generally considered to be tetraploid. In addition, triploidy was discovered for the first time in C. rivulare. CONCLUSIONS: Considerable differences in nuclear DNA content exist among Central European species of Cirsium on the diploid level. Perennial soft spiny Cirsium species of wet habitats and continental distributions generally have smaller genomes. The hybrids of diploid species remain diploid, and their DNA content is smaller than the mean of the parents. Species with smaller genomes produce interspecific hybrids more frequently.  相似文献

We present a new method to produce tetraploid Crassostrea gigas by cytochalasin B inhibition of polar body 2 expulsion in diploid females crossed with tetraploid males. This offers a means of direct introgression of genetic characters from selected diploid to tetraploid lines, avoiding a triploid step. Offspring larval ploidy shifted over time and depended on size, with tetraploids more frequent among the smaller larvae and triploids among the large. Viable tetraploids were found at 4 and 6 months, indicating the technique was successful. The possibility that gynogenesis occurred was tested by microsatellite analysis to confirm the presence of paternally inherited alleles. These were present in all animals of the 2n × 4n + CB (female first) cross. However, a 4n × 2n + CB cross produced triploids, including some gynogens. Our method illustrates for the first time that diploid C. gigas eggs, if selected for large size, can give viable tetraploid offspring.  相似文献

Comparisons of the chromosome numbers, 2C nuclear DNA amounts and karyomorphology were made in explant cultures of diploid (2n = 2x = 14) and autotetraploid (2n = 4x = 28) Phlox drummondii. In 6–36 week old calli derived from diploid internodal segment explants, and in cells of root tips regenerated from such callus, marked differences were observed in chromosome number. The chromosome numbers ranged from 2n = 14 to 2n = 100 and DNA amounts from 8.20 to 63.20 pg in the diploid derived callus, while the extent of variation was much reduced in the regenerated roots. In contrast, the autotetraploid cultures were characterised by the maintenance of the same chromosome number and DNA amounts as the mother plant. Modified chromosome structures were not apparent in any of the cultures. The possible reasons for the chromosomal instability at the diploid level and stability at the tetraploid level are discussed.  相似文献
