The Liaodong oak ( Quercus liaotungensis Koidz. ) is a close relative of the Mongolia oak ( Q. mongolica Fisch. ) which were separated by some morphological characters, such as the number of leaf lobes and the squamate form of cupula. Recently some authors observed that morphological diversity made the Liaodong oak unable to separate clearly from the Mongolia one, therefore, plant materials have been collected from Maoer Mountain of Heilongjiang province as a typical Q. mongolica population, and from Guandi Mountain of Shanxi Province which represented the typical Liaodong oak population to compare isozyme and DNA diversity between both species. The winter bud samples were also analysed from Dongling Mountain near Beijing City since Dongling population is an intermediate form between the Maoer population and the Guandi population morphologically. Statistics of 13 putative loci belonging to 5 enzymes showed a high level of diversity within all populations. The value of genetic distance among populations was low, and showed that the Dongling population genetically located at the middle of both typical species. DNA data also showed that both typical oak populations shared similar variation with the Dongling population. Among 172 polymorphic RAPD and DAF loci, no population-specific band has been found. A significant difference in frequency of amplified products existed in 26 loci. Except for 3 irregular ones, frequency distribution of 23 loci seems clinal. The Dongling population also genetically located at the middle of both flanking populations. It was worth to note that the OPD-08434 was probably unique to the Dongling oak since its frequency in the Guandi population has been estimated up to 100% and decreased to 0 in the Maeer population. The Dongling population received this unique DNA from the Guandi population probably by introgression. The strong gene flow in both directions implied a long history of distributive continuity for both oak species. High levels of morphological, isozymatic and DNA diversity supplied enough genetic basis for reconstruction of degraded oak ecosystems.  相似文献

为研究辽宁沿海弯棘斜棘(鱼衔)(Repomucenus curvicornis)自然群体的遗传多样性及遗传结构,采用PCR扩增获得辽宁沿海弯棘斜棘(鱼衔)辽东湾群体(n=22)及黄海北部群体(n=18)线粒体的COⅠ及控制区(CR)部分DNA序列片段,进行序列比较及遗传多样性分析.获得弯棘斜棘(鱼衔)COⅠ基因片段624 bp,其A、T、C、G平均含量分别为24.09%、31.04%、25.28%和19.59%;CR片段460 bp,其A、T、C、G平均含量分别为32.96%、32.80%、14.86%和19.38%.基于COⅠ基因和CR序列得到的两群体变异位点数、平均核苷酸差异数、单倍型多样性指数以及核苷酸多样性指数分别为:38、4.67、0.96±0.02和0.0075±0.0042;26、3.35、0.97±0.02和0.0073±0.0043.序列分析结果均显示,辽东湾群体的遗传多样性低于黄海北部群体.分子方差(AMOVA)分析结果显示,基于COⅠ基因片段辽东湾与黄海北部群体间无明显遗传分化(Fst=0.0091,P=0.25)而基于CR序列两群体间具有较小但接近显著的遗传分化(Fst=0.0264,P=0.09).研究表明,线粒体CR序列与COⅠ基因均可作为检测弯棘斜棘(鱼衔)群体遗传多样性的有效分子标记,但CR序列遗传分化的敏感度要高于COⅠ基因,更适合作为弯棘斜棘(鱼衔)群体遗传研究的分子标记.  相似文献

The genetic structure of two Liaodong oak (Ouercus liaotungensis Koiz. ) populations whose habitats represent respectively moist and dry donditions at Dongling Mountain region, a suburb of Beijing City, was studied by using isozyme analysis. Thirty putative loci from thirteen enzyme systems were analyzed. The result showed very high genetic variability in the two populations, with a percentage of polymorphic loci of 86.6%, and an average number of alleles per locus of 2.25. There still existed obvious differences in a few loci although high levels of genetic similarity was observed, indicated by low value of both distance (D= 0. 029) and coefficient of gene differentiation (GsT = 0. 048). The dynamic changes of the gene frequencies of different age groups of oak trees may imply that the oak populations can be adaptive to different conditions.  相似文献

应用同工酶分析方法,测定北京市东灵山区两个分别代表干旱和湿润生境的辽东栋(QuercusliaotungensisKoiz.)群体的遗传结构。共分析统计了13 个酶系统30 个位点。结果表明:辽东栎群体内部存在丰富的遗传变异(多态位点百分率为86.6% ,等位基因平均数为2.25)。两群体遗传结构的相似性程度很高(D= 0.029, GST= 0.048);但在个别位点上仍存在较大差异,这些差异的产生可能与对小生境的适应有关。对不同年龄段的初步分析结果显示,基因频率的动态变化可能有其适应意义  相似文献

Miscanthus sinensis is a dominant perennial C4 grass with the potential to being a feedstock crop in North America, Europe, and China. Variation in chloroplast DNA sequence was used to obtain information regarding the genetic diversity and structure of populations of M. sinensis in southwest China. Chloroplast DNA, trnL-F and rpl20-rps12 sequences from seventy-five individuals representing 14 populations of M. sinensis were used to study the sequence variation. Seven haplotypes and 16 polymorphic sites (2.7%) were identified. Five substitutions, 6 indels, and 5 existing substitutions and indels sites, were detected through splicing these two gene segments. The genetic diversity within the studied populations (diversity of haploids, h = 0.561, nucleotide diversity, π = 0.00504) was low, this may be affected by the relatively larger effect of genetic drift on the chloroplast DNA, reflecting smaller effective populations than nuclear DNA. Genetic variance within the populations was higher than that between the populations, suggesting that higher gene flow may exist within these populations. The results of parsimony network in seven haplotypes indicated that H1 and H2 may be ancient haplotypes, which may help guide future research on the origin of M. sinensis. Our results provide information on the genetic diversity and structure of M. sinensis and may assist future studies on the phylogeography of M. sinensis.  相似文献

The genetic diversity and genetic structure of Potamogeton maackianus A. Benn. in seven lakes of the middle reaches of the Yangtze River were studied using random amplified polymorphic DNA (RAPD). The gene flow and genetic relationships between populations were analyzed in combination with the geographic distribution and the river system of the lakes. A total of 112 bands were amplified and 59 bands (52.7%) were polymorphic. Each of the 86 individuals investigated exhibited a unique genotype. The Shannon index was used to measure genetic diversity, and the total genetic diversity was 0.414 and the mean genetic diversity of populations was 0.148. P. maackianus showed a relatively high level of genetic diversity. Analyses of molecular variance (AMOVA) revealed that 63.8% of the total genetic diversity existed among populations and 36.2% within them, which was consistent with the genetic structure computed by the Shannon index: among-population variation and within population variation accounted for 64.4 and 35.7%, respectively. The gene flow among populations was very limited, and genetic isolation among populations occurred even though they were connected through the Yangtze River. Cluster analysis divided the seven populations into two groups, and the genetic relationships among the populations had no obvious association with their geographic distribution, or the historical relations with the river system of the lakes where they occurred. Mantel tests revealed that distance was an important factor affecting the genetic structure in populations. The development history of P. maackianus populations in Honghu Lake had an obvious effect on its genetic structure.  相似文献

利用核基因和叶绿体基因对台湾水韭(Isoetes taiwanensis Devol)2个居群20个样本的遗传多样性和遗传结构进行了分析,并对台湾水韭的保育提出了建议。核基因的检测结果显示,2个居群共存在18个单倍型,单倍型多样性为0.9842,核苷酸多样性为0.00215,居群间基因流Nm为4.26,居群间分化系数Gst值为0.05543;叶绿体DNA的检测结果显示,单倍型数目为6,单倍型多样性为0.6211,核苷酸多样性为0.00326。其中台北居群的单倍型多样性和核苷酸多样性均比金门居群高。核基因和叶绿体基因的AMOVA分析显示,居群内的遗传差异远高于居群间。台湾水韭的核苷酸多样性相对其他水韭属植物较低,可能与台湾水韭的染色体倍型以及狭域分布有关。居群结构的形成可能和基因流以及奠基者效应有关。宜采用原地和迁地保护的方法对居群进行保护。  相似文献

运用ISSR-PCR分子标记技术对邓氏马先蒿(Pedicularis dunniana)云南中甸以及四川木格措两个自然居群的遗传分化以及遗传多样性进行研究。结果表明:物种水平上邓氏马先蒿遗传多样性水平较高(PPB=62%),但居群平均水平很低,云南中甸居群多态性位点百分率只有23%。四川居群也仅仅是27%,AMOVA分析结果表明,居群间基因分化系数Fst=0.7462,即遗传变异绝大部分来源于两居群间,居群间表现出强烈的遗传分化。自交的交配方式以及小居群产生的遗传漂变效可能是造成邓氏马先蒿目前遗传结构的主要原因。  相似文献

利用相关序列扩增多态性SRAP(Sequence-related amplified polymorphism)分子标记首次对4个暗纹东方鲀(Takifugu obscurus)群体(1个长江捕捞群体、1个放流群体和2个养殖群体)进行了遗传多样性分析。从49对引物组合中筛选出18对扩增条带清晰、稳定的引物组合对4个群体进行扩增,共获得231个位点,其中多态位点数为156个,总多态位点百分率为67.53%。4个群体内多态位点比例为54.98%—58.87%,群体的Nei’s基因多样性指数(H)为0.1992—0.2005,群体Shannon多样性指数(I)为0.2953—0.3016,群体间基因流(Nm)为4.1291。长江捕捞群体的多态位点比例、基因多样性指数、Shannon多样性指数均略高于其他三个群体。采用UPGMA法对4个群体进行聚类分析显示,上海养殖群体单独聚为一类,其余3个群体聚为另一类。AMOVA结果表明遗传变异主要来源于群体内,占总遗传变异的87.40%。以上结果表明,4个暗纹东方鲀群体具有较高的遗传多样性,群体间相似性较大,并且存在一定的基因交流。  相似文献

用随机扩增多态DNA (RAPD)标记对 5个三棱栎 (Trigonobalanusdoichangensis)居群共 99个个体进行遗传多样性和居群遗传结构分析。 16个引物共检测到 15 7个位点 ,其中多态位点 83个 ,占 5 2 87%。物种水平Shannon多样性指数I =0 2 4 31,Nei基因多样度h =0 15 95 ,种内总遗传变异量Ht=0 16 0 0 ,居群内遗传变异量Hs =0 0 74 9,居群间变异量大于居群内变异量 ,表明三棱栎的遗传变异主要存在于居群之间。与同科植物相比 ,三棱栎遗传多样性较低 ,遗传分化系数Gst =0 5 32 0 ,说明居群间的遗传变异占 5 3 2 0 % ,居群间已出现强烈的遗传分化。当地人的强烈活动造成的生境破碎化和居群隔离 ,以及三棱栎演化过程中的地史变化对其种群发展的影响等 ,可能是造成其居群间强烈的遗传分化和较低遗传多样性的原因。基于本研究结果 ,提出了三棱栎遗传多样性的保护策略。  相似文献

采用DALP (Direct amplification of length polymorphism) 分子标记技术, 对产自云南的药用植物丽江山慈菇Iphigenia indica (L.) Kunth的9个居群进行DNA指纹检测。筛选出5个引物组合, 扩增共产生131条DNA片段, 其中104 条谱带具有遗传多态性, 约占79 39%, 平均每组引物扩增所得多态条带为20 8, 9个居群平均多态百分率为42 21%。9个居群平均观察等位基因数Na为1 4224, 总Na为1 7939; 平均有效等位基因数Ne 为1 3141, 总Ne 为1 4810; 平均遗传多样性指数H为0 1745, 总H为0 2831; 平均Shannon 多样性指数I 为0 2527, 总I为0 4231; 总基因多样性Ht为0 2831, 居群内多样性Hs 为0 1745, 居群间基因分化系数Gst为0 3834, 即丽江山慈菇有61 66%的遗传变异来自居群内, 38 34%来自居群间, 居群间存在较高水平的遗传分化。滇西北居群的遗传多样性明显高于滇中居群的遗传多样性, 这与滇中地区丽江山慈菇野生资源被大规模挖掘有着直接的关系。  相似文献

基于mtDNA Cyt b序列分析齐口裂腹鱼群体遗传多样性   总被引:2,自引:0,他引:2  
研究测定了长江上游4个齐口裂腹鱼(Schizothorax prenanti)野生群体(重庆巫溪、重庆城口、四川雅安、四川阿坝)共104个个体的线粒体Cyt b基因部分序列, 以探讨齐口裂腹鱼野生群体的遗传多样性和遗传结构。结果表明: 在104个个体Cyt b序列中共检测到43个多态性位点, 25个单倍型。4个齐口裂腹鱼群体的单倍型多样性介于0.704—0.884, 核苷酸多样性介于0.007—0.012。群体间Kimura双参数遗传距离介于0.008—0.017, 其中四川雅安群体与四川阿坝群体间遗传距离最近, 基因交流频繁。重庆城口群体与四川雅安群体间遗传距离最远, 基因交流受阻。AMOVA分析表明, 齐口裂腹鱼的遗传分化主要来自群体内部, 且组群间、组群内群体间和群体内存在显著的遗传分化。中性检验得到Tajima’s D和 Fu’s Fs的值不显著, 且歧点分布图呈多峰, 表明长江上游4个齐口裂腹鱼野生群体未经历过种群扩张。研究旨为齐口裂腹鱼野生资源保护提供必要参考意见, 同时为齐口裂腹鱼种质资源合理开发和利用提供理论依据。  相似文献

为了更有效地保护和合理开发大别山山核桃(Carya dabieshanensis)资源,该文利用RAPD分子标记技术,对3个天然大别山山核桃种群的90个单株的遗传多样性、种群内和种群间的遗传变异进行了研究,结果表明:20对10 bp随机引物共检测到238条谱带,其中多态带为162条,占68.1%。遗传多样性分析结果显示: Shannon多样性指数为0.476 1,58.18%的变异分布于群体内,而种群间变异占了41.82%;Nei指数群体总基因多样度为0.314 5,群体内平均基因多样度(HS)为0.186 5,群体间的基因多样度(HST)为0.128 0,群体Nei基因分化系数(GST)为0.406 7,说明40.67%的变异存在于种群间,群体内的变异占了总变异的59.33%,与Shannon多样性指数相比基本一致,均表明种群内有较丰富的遗传变异,这为优良品种选育提供广阔前景;种群间的基因流(Nm)为0.730 6,证明种群间遗传交换较小,这与环境适应性和高山阻隔有一定的关系。  相似文献

Sawtooth oak (Quercus acutissima) is a predominant tree species in the deciduous broad-leaved forest in China. It distributes in a large landscape area and can disperse in various ecology types. Molecular study on sawtooth oak can provide valuable information about the genetic diversity level and genetic relatedness on this important tree species. Insight into the genetic structure also provides resources of a species with its current feature and future evolutionary potential. The genetic structure of sawtooth oak was investigated by randomly amplified polymorphic DNA (RAPD). Twelve RAPD markers were used to assess genetic diversity of 408 individuals from 17 provenances enveloping most of the current distribution area of sawtooth oak. A total of 66 amplification products were detected, of which 49 bands (74.24 %) were polymorphic. Nei’s gene diversity, 0.2409, indicated a relatively high level of genetic variation in sawtooth oak germplasm. Analysis of molecular variance showed that most of the genetic diversity (87 %) was allocated within provenances. A combination of UPGMA dendrogram and STRUCTURE analysis was employed to estimate the genetic relationships of sawtooth oak germplasm; interestingly, the two methods presented similar grouping pattern with few discrepancies. Results revealed that 16 out of 17 provenances were clustered into one group, while the other 1 (LQ provenance) constituted a separate cluster. The data presented in this study suggested that the RAPD method was a valuable tool for estimation of genetic diversity and genetic relatedness of sawtooth oak germplasm. The present study also gave useful implications for germplasm conservation and new cultivar development for this promising energy tree species.  相似文献

用RAPD标记对来自不同产地的24份金莲花种质从分子水平开展遗传多样性研究,为综舍评价我国金莲花资源现状及植物资源的合理保护提供参考。结果显示,30条RAPD引物共扩增获得194条清晰可辨条带,其中多态性条带108条,多态性位点百分率为55.67%,金莲花总多样性指数为0.1460,居群内遗传多样性为0.0591,Shannon’s多样性指数为0.2248;通过UPGMA进行聚类分析,将24份金莲花种质划分为3个大类,根据总遗传多样性和居群内遗传多样性计算不同居群间遗传分化系数为0.6902,基因流估计平均值为0.2244。研究结果表明,目前我国金莲花种质资源的遗传多样性指教偏低,应该尽快采取相应措施,对不同金莲花资源进行合理保护。  相似文献

Genetic diversity within populations of Fusarium pseudograminearum isolated from wheat grains from the Canadian provinces of Alberta and Saskatchewan was investigated. Three restriction enzymes (EcoRI, HaeIII, and PstI) were used to carry out restriction analysis of the nuclear ribosomal DNA (nrDNA) intergenic spacer region (IGS region) and eight primers were used to generate inter-simple sequence-repeat (ISSR) molecular markers. Our study indicated substantially high genetic diversity within these two populations, but low genetic differentiation and frequent gene flow among populations. The IGS data showed no genetic distinction between the two Alberta populations and only minor genetic differentiation between the Saskatchewan and Alberta populations. Analysis of molecular variance indicated that most genetic variability resulted from differences among isolates within populations. Multilocus linkage disequilibrium analysis suggested a panmictic population genetic structure and the occurrence of significant recombination in F. pseudograminearum. Regular gene flow and random mating between isolates from different populations could result in novel genotypes with both improved pathological and biological traits.  相似文献

用40个10碱基随机引物对福建2个生态类型的7个地方鸭种的基因组池DNA进行了RAPD分析,从扩增产物表现为多态的引物筛选出9个特异性强的引物进行个体DNA的RAPD分析,并利用Shannon指数估算了2个生态区域鸭群的遗传多样性,探讨了它们与生态环境之间的关系及其变化规律.结果表明,福建境内的地方鸭种具有较高的遗传多样性,闽东沿海地区鸭群有67.97%的遗传变异来自群体内,32.03%来自于群体间;闽西山地丘陵地区鸭群有59.05%的遗传变异来自群体内,40.95%来自于群体间;闽东地区鸭群的遗传变异明显高于闽西地区鸭群.同时,用非加权组平均法(UPGMA)对各种群的聚类分析表明,闽西地区的连城白鸭、泰宁麻鸭、龙岩山麻鸭和三明麻鸭归为一类;闽东地区的龙海金定鸭和莆田黑鸭聚为一类.不同生态区域内鸭群的遗传变异与地理位置存在一定的相关性.  相似文献

Dams have the potential to affect population size and connectivity, reduce genetic diversity, and increase genetic differences among isolated riverine fish populations. Previous research has reported adverse effects on the distribution and demographics of black redhorse (Moxostoma duquesnei), a threatened fish species in Canada. However, effects on genetic diversity and population structure are unknown. We used microsatellite DNA markers to assess the number of genetic populations in the Grand River (Ontario) and to test whether dams have resulted in a loss of genetic diversity and increased genetic differentiation among populations. Three hundred and seventy-seven individuals from eight Grand River sites were genotyped at eight microsatellite loci. Measures of genetic diversity were moderately high and not significantly different among populations; strong evidence of recent population bottlenecks was not detected. Pairwise FST and exact tests identified weak (global FST = 0.011) but statistically significant population structure, although little population structuring was detected using either genetic distances or an individual-based clustering method. Neither geographic distance nor the number of intervening dams were correlated with pairwise differences among populations. Tests for regional equilibrium indicate that Grand River populations were either in equilibrium between gene flow and genetic drift or that gene flow is more influential than drift. While studies on other species have identified strong dam-related effects on genetic diversity and population structure, this study suggests that barrier permeability, river fragment length and the ecological characteristics of affected species can counterbalance dam-related effects.  相似文献
