以豌豆(Pisum sativum L.)和绿豆(Phaseolus radiatus L.)为材料证明了IAA极性运输的自动抑制现象。用改进的“供体-受体技术”证明:受体中的IAAA可抑制~3H-IAA的极性运输,抑制程度随受体中加入IAA浓度的增加而增强;在“Y”型外植体一侧切口施用的IAA也可抑制~3H-IAA在另一侧的极性运输,并导致~3H-IAA在该侧组织中的积累以及代谢或钝化的增强。此外,组织中游离~3H-IAA的比例也随运输时间的延长而不断减小。  相似文献

Regeneration of whole plants of marigold (Tagetes erecta L.) was achieved by organogenesis using leaf explants. Leaf segments about 0.25 cm2 were taken from 3-week-old in vitro plantlets and cultured on MS basal medium containing BA with different auxins (NAA, 2,4-D and IAA). The exposure time of the explants on the regeneration medium was tested. The highest values for regeneration were obtained with BA (13.3 M) and IAA (17.1 M). Thirteen days was the best time of exposure of the explant to the regeneration medium for shoot induction.  相似文献

This study determined the effects of a static liquid culture system on shoot regeneration from citrus epicotyl explants. A mixture-amount experiment was used to determine the effects of zeatin riboside (ZR), 6-benzylaminopurine (BA), and indole-3-acetic acid (IAA) on two citrus types—citrange (Citrus sinensis ‘Washington’ L. Osbeck. × Poncirus trifoliata L. Raf var. Carrizo) and sweet orange (Citrus sinensis L. Osbeck var. ‘Ridge Pineapple’). A liquid culture system comprising a Petri dish, cellulose filter paper, and liquid culture medium was used. Shoot regeneration experiments were conducted over 6 wk that included 2 wk in the dark followed by 4 wk in the light. Three responses were measured: (1) number of explants forming buds and/or shoots, (2) number of explants with shoots >?2 mm, and (3) overall explant and shoot quality. The effects of paper disc number, liquid medium volume, and explant size on shoot regeneration were determined. High-quality shoots were produced from explants cultured in 5.25 to 12 mL medium volume and explant sizes ranging from 2 to 15 mm. The effects of the plant growth regulators were similar for the two citrus types and were as follows: (1) use of ZR or BA resulted in high-quality shoot production; (2) ZR and BA were not synergistic; (3) culture in 20 μM ZR resulted in the highest shoot production; (4) BA and IAA were strongly synergistic, with the greatest production with BA when IAA was included in the mixture; and (5) ZR and IAA were antagonistic, particularly with Ridge Pineapple.  相似文献

Levels of endogenous ABA and IAA were quantified during the first week of in vitro rooting of Wild Cherry (Prunus avium L.) using IBA in the culture medium. Hormones were measured in the apical, median and basal parts of the explants using an avidin-biotin based enzyme linked immunosorbent assay (ELISA), after a purification of the methanolic extracts by high-performance liquid chromatography (HPLC).Root primordia started to differentiate from day 5 at the basal part of the explants. ABA and IAA showed considerable changes and high levels were detected during the first week of culture. ABA levels increased transiently mainly in the apical part during root formation. Exogenous IBA was possibly transformed into IAA mainly in the basal part of the explants.  相似文献

The Malnad region located in the Western Ghats of Karnataka is known for the cultivation of indigenous rain fed land race cultivar of rice. The present study was to investigate the callogenic and caulogenic potentialities of the two indigenous rice cultivar namely Karimundaga and Kanadatumba using dehusked mature embryo explants. For callus and shoot bud differentiation, the explants were cultured on Murashige and Skoog (MS) medium supplemented with 2,4-D (1-3 mg/L), IAA (1-2 mg/L), Kn (1-4 mg/L) and BAP (1-4 mg/L). The morphogenic potentialities of the two rice cultivar differed in texture of callus. In both the cultivar callogenic frequency was optimized at 1 mg/L 2,4-D concentration, it was 94% in Karimundaga and 58% in Kanadatumba. Supplementation of IAA either alone (1-2 mg/L) or in combination with Kn or BAP at 1 to 4 mg/L concentration of each induces shoot bud differentiation from the calli. In the cultivar Karimundaga caulogenic frequency was highest (10.60 +/- 2.55) at 1.0 mg/L IAA and 4.0 mg/L BAP concentration. While in the cultivar Kanadatumba highest number of shoot buds (7.90 +/- 2.69) was differentiated at 1.0 mg/L IAA and 4.0 mg/L Kn concentration. The calli derived regenerants were successfully acclimatized in the greenhouse and agro-morphological variations were evaluated. The growth characteristics and yield related parameters exhibited by in vitro plants were lower than the in vivo plants.  相似文献

Each of four amino acid conjugates of IAA was able to replacethe IAA requirement for xylogenesis in lettuce pith explants,when supplied at concentrations ten to 100 times those optimalfor IAA. Tracheary development induced by these conjugates tendedto be slightly slower and less in amount than with IAA, andthe tracheary strands shorter and less regular. Responses differedsomewhat among the four conjugates: IAA-D, L-aspartate gavedevelopment most like that with free IAA, and IAA-D, L-phenylalanineoften yielded the weakest tracheary development, while responsesto IAA-L-alanine and IAA-glycine were intermediate. The resultsare interpreted in terms of the ‘bound’ IAA conjugatesdiffusing into the pith explants and becoming xylogenic onlyon hydrolysis to ‘free’ IAA. As tracheary strandformation is believed to result from IAA fluxes, it seems thatthe free IAA also moved through the discs, presumably towardsthe surfaces where it degrades rapidly. Tracheary strand formationin these explants can be compared with vascular strand formationin the normal shoot tip, where IAA conjugates (auxin ‘precursors’)move acropetally and are hydrolysed to free IAA especially inthe young leaf primordia, we suggest, yielding local sourcesof IAA which may contribute both to the phyllotactic spacingof primordia and, moving basipetally, to the definition of theauxin pathways that develop as procambial strands behind individualleaf primordia. Callus induction and thallus regeneration in some species of red algae   总被引:1,自引:0,他引:1  
Callus induction was obtained from axenic explants of 14 species of red algae. ASP12NTA solid medium (1.5% agar) supplemented with indole-3-acetic acid (IAA) and 6-benzylaminopurine (BAP) was used for callus induction. In most of the species, addition of IAA or BAP at 0.1 mg/L or 1.0 mg/L enhanced callus induction rate or callus size. The combination of IAA (0.1 mg/L) and BAP (0.1 mg/L) was more effective among eight species, while high concentrations of IAA (10 mg/L) showed an inhibitory effect. Great variation in callus form, source tissue, and color of the induced callus were observed. The callus mainly originated from medullary and cortical tissue of the explant. Callus with filamentous, oval and spherical cell chains or disorganized cell mass was observed. The excised calluses from the explants of six species showed sustained growth on subculture. On transfer of the subdivided callus mass of seven species to PES liquid medium, shoot formation and thallus regeneration were observed.  相似文献

Since in tobacco thin layers exogenous putrescine alters the physiological and mor-phogenic responses induced by IAA (indole-3-acetic acid) and/or BA (benzylade-nine), the effect of this polyamine on protease activity and on the formation of meristemoids and vegetative primordia was studied during morphogenesis. Superficial thin layer explants, excised from the stem of tobacco (Nicotiana tabacum L. cv. Samsun) plants in the vegetative stage, were cultured under various hormonal conditions (IAA, IAA+BA, BA) and in a hormone-free medium, in the presence or absence of 100 μM putrescine. Histological analysis showed that no meristemoids were formed on the control medium or with putrescine alone and only a few were formed on IAA-treated explants with or without putrescine. An increasing number of meristemoids was observed in IAA+BA and BA treatments during culture; in both cases this number was enhanced by the presence of exogenous putrescine. Protease activity was evaluated spectrophotometrically using two synthetic substrates, azocasein and N-benzoyl-DL-arginine-p-nitroanilide (BAPNA). In the former, maximum protease activity was observed in IAA+BA- and BA-treated explants on days 10 and 15, respectively, while with IAA activity was lowest, the maximum occurring on days 5–10. In this case exogenous putrescine enhanced protease activity in the presence of IAA alone or with BA, while it decreased it in the presence of BA. BAPNA-mediated proteolytic activity (serine-proteases) was highest in IAA+BA-treated explants, intermediate in BA- and not different from controls in IAA-treated explants. Putrescine only affected proteolytic activity in IAA+BA treatments. The use of specific inhibitors of protease activities indicated that these enzymes belong to two main classes of proteases, that is serine- and thiol-proteases. The pattern of proteolytic activities during culture appeared to be related to the differentiation of meristemoids into vegetative primordia. The effect of exogenous putrescine on protease activity was different depending on different synthetic substrates, developmental patterns, pH and ionic strength.  相似文献

AIMS: The main aim of this study was to investigate the possible role of indoleacetic acid (IAA) from bacteria to induce in vitro adventitious shoots in internodal explants of Brassica oleracea L. METHODS AND RESULTS: Culture supernatant of Halomonas sp. RE1 and Halomonas sp. HT1 that contain 21 and 40 microg ml(-1) IAA, respectively, was used to supplement Murashige and Skoog (MS) medium. Two combinations that were supplemented with bacterial supernatant (BS) are MS + BS and MS + BS + 10%CW (coconut water) while basal MS medium was used as control. The amounts of BS used in this experiment were 50, 100, 150 and 200 microl in 5 ml MS medium in each combination. In vitro-grown internodal explants of B. oleracea were inoculated on these media combinations and incubated in a growth chamber at 25 +/- 1 degrees C and exposed to 16-h cool fluorescent light. After 5-6 weeks of incubation adventitious shoot induction was observed in all treatments that were supplemented with BS as compared with the controls where very low response was observed. The frequency of shoot induction was high in media that were supplemented with 10%CW in the presence of bacterial auxin. CONCLUSIONS: It was concluded that IAA of microbial origin has the potential to induce adventitious shoots in internodal explants. SIGNIFICANCE AND IMPACT OF THE STUDY: IAA from bacteria can be effectively used in plant tissue culture; especially a combination of MS + BS + 10%CW is very cost-effective as compared with synthetic phytohormones for in vitro studies.  相似文献

Efficient callus and explant regeneration medium, using microbial extract (SPE purified) or supernatant has been formulated for Brassica oleracea L. var. capitata. Two cyanobacterial strains (Anabaena sp. Ck1 and Chroococcidiopsis sp. Ck4) and two bacterial strains, (Pseudomonas spp. Am3 and Am4) known to produce a number of cytokinins, tZ, cZ, ZR, DHZR and IAA were selected for the media formulation. Supernatant from strains with high cytokinin to IAA ratio, including Pseudomonas aeruginosa Am3 (2.08) and Chroococcidiopsis sp. Ck4 (0.8) efficiently induced compact calli which were turned green upon exposure to light. The strains producing lower cytokinins to IAA ratio (0.11–0.13) on the other hand induced friable callus which were unable to regenerate on the selected media combinations. Leaf, stem and root explants of Brassica oleracea L. regenerated on MS medium supplemented with phytohormones from microbial origin with efficiency comparable to standard cytokinins and IAA. Supplements from cyanobacterial origin proved to be the best for induction of adventitious roots and shoots on internodal and petiolar segments. Hypocotyl explants however, responded well on MS supplemented with bacterial metabolites. Induction of adventitious shoots on root explants was supported by phytohormones from both origin equally well. Callus induction on the seeds and regeneration of shoots on calli was also observed. Cyanobacteria based media were more efficient to induce calli capable of regeneration upon exposure to light. Internodal explants were highly amenable to regenerate shoot and roots simultaneously. Root explants were the less successful to regenerate shoots.  相似文献

The petiole abscission induced by deblading cotyledonary leavesof cotton (Gossypium hirsutum L. cv. Delta Pine) was acceleratedby the presence of the intact shoot apex or, in decapitatedplants and explants, by application to the stem (proximal application)of indol-3yl-acetic acid (IAA) or 1-aminocyclopropane-l-carboxylicacid (ACC). IAA and ACC accelerated the abscission of debladedpetioles whether applied above or below the cotyledonary node.Transport of IAA to the node was not required for the responseto proximal IAA. [2,3-14C]ACC was readily transported to thenodal region whether applied to the stem above or below thenode. Application of IAA or ACC to the stem did not induce theabscission of intact leaves or of debladed petioles treateddistally with IAA The acceleration of abscission by proximal IAA, but not thatcaused by ACC, was prevented if explants were treated with a-aminooxyaceticacid (AOA), an inhibitor of ACC-synthase. AOA also preventedthe acceleration of abscission caused by the shoot apex. Theprogress of abscission in debladed explants was greatly delayedby silver thiosulphate (STS—an inhibitor of ethylene action),whether or not the explants were treated with IAA or ACC. Itis suggested that the speeding effects of the shoot apex andof proximal auxin on the abscission of debladed petioles requiresauxin-induced ACC synthesis. Major gibberellins (GAs) in lettuce (Lactuca sativa L. cv Romaine) pith explants have been identified by gas chromatography-mass spectrometry (GC-MS) or GC-selected ion monitoring (GC-SIM) as GA1, 3-epi-GA1, GA8, GA19, and GA20. Treatment of pith explants with indole-3-acetic acid (IAA) (57 micromolar) plus kinetic (0.5 micromolar) induced xylogenesis. In this xylogenic treatment, the concentration of a biologically active, polar GA-like substance(s) increased during the first 2 days of culture, although all of the above GAs decreased (as measured by GC-SIM). In non-xylogenic treatments, where explants were cultured without exogenous hormones, or with IAA or kinetin alone, the concentration of the biologically active, polar GA-like substance(s) decreased during the first two days of culture, as did all of the above GAs (as measured by GC-SIM). Treatment of pith explants with exogenous GA1 alone did not induce xylogenesis, but GA1 at very low concentrations (0.0014 and 0.003 micromolar) synergized xylogenesis in the IAA plus kinetin-treated cultures. These results suggest that changes in the concentration of certain endogenous GAs may be involved in xylogenesis mediated by IAA plus kinetin in lettuce pith cultures.  相似文献

Hypericum perforatum L. (St. John’s wort) produces a number of phytochemicals having medicinal, anti-microbial, anti-viral and anti-oxidative properties. Plant extracts are generally used for treatment of mild to medium cases of depression. Plant regeneration can be achieved in this species by in vitro culture of a variety of explants. However, there are no reports of regeneration from petal explants. In this report plant regeneration from petal explants of St. John’s wort was evaluated. Petals of various ages were cultured on agarized Murashige and Skoog 1962 (MS) medium supplemented with auxin and cytokinin (kinetin), maintained in the dark and callus and shoot regeneration determined after 28 days. At an auxin to cytokinin ratio of 10:1, callus and shoot formation were induced by all levels of indole-3-acetic acid (IAA), indole-3-butyric acid (IBA) and 1-naphthaleneacetic acid (NAA), while 2,4-dichlorophenoxyacetic acid (2,4-D) induced only callus formation. The optimum level of auxin for shoot regeneration was 1.0 and 0.1 mg/l kinetin, where the regeneration frequency was 100 percent for all three auxins. The highest number of shoots per explant (57.4 and 53.4) was obtained with IAA and IBA, respectively. In the absence of auxin, kinetin levels of 0.1 and 0.25 mg/l induce callus and shoot formation at low frequency but not at lower levels. Callus and shoot formation did not occur in the absence of growth regulators. Petal-derived shoots were successfully rooted on half-strength MS medium without a requirement for exogenous auxin and flowering plants were established under greenhouse conditions. From these results it can be concluded that auxin type is a critical factor for plant regeneration from petal explants of Hypericum perforatum and there is no absolute requirement for high levels of cytokinin.  相似文献

In this report, rapid and effective shoot as well as root regeneration system through direct multiplication was successfully developed for Cichorium intybus L. Furthermore, the effect of exogenous growth regulators (TDZ and IAA) at different concentrations on the regulation process of the plant was also studied. Enhanced production of esculin in developed C. intybus L. was evaluated using leaf extract. Only on the expense of 20 days, regeneration was seen and very low dose of TDZ was seen to be more effective. When 0.02 mg/L of TDZ was combined with 1.5mg/L of IAA, nearly 100% of explants produced shoots with the highest number of regenerated shoots (85.37). With further increase in concentration (≥ 0.05 mg/L), the number of shoots per explants get decreased. A lower NAA to IBA ratio (1.0mg/L of IBA and 0.5mg/L of NAA) seemed to be more effective for root generation and considered to be the most effective combination among the tried groups. IBA was more effective in root development than NAA, but both were comparatively effective. On quantitative analysis by RP-HPLC, the 76.23% of Esculin were found in leaf extract of the in vitro developed C. intybus L. This amount was 26.77% higher than normal grown plants.  相似文献

对扁桃优良砧木“Hansen”侧芽进行离体快繁研究,结果表明:外植体取材时间不同,成活率差异明显,以春季芽即将萌动的3月下旬为最佳时间,在MS 6-BA1.0mg/L GA30.2mg/L培养基上诱导芽萌发,萌发率在75%以上;以MS 6-BAl.5mg/L IAA0.1mg/L进行芽的增殖培养效果最好;1/2MS IBA0.5mg/L诱导生根,并给以黑暗预处理可使生根率达85%以上,每株生根数量多,根系质量好;采用分步炼苗移栽,提高了小苗成活率,有利于小苗后期生长。  相似文献

Summary The net uptake and movement of radioactivity by 12-mm root segments of Zea mays have been studied as a function of time at 5, 15 and 25° C. Segments were supplied with an agar donor block containing 1 M IAA-1-14C or IAA-2-14C continuously or for a limited period of time (pulse-labelling). In the latter case the original donor block was replaced either by a plain agar block or by one containing 1 M unlabelled IAA. Receiver blocks were placed at the other end of the segments.The net uptake of radioactivity from the donor block at 15° C was greater at the basal end than at the apical end of the segment. At 5 and 15° C, the net uptake from a basal donor was virtually linear with time but at 25° C the rate of net accumulation decreased after about 10 h. Decarboxylation of IAA undoubtedly occurred at 15 and 25° C when the concentration in the tissue attained a high value.An acropetally polarised movement of radioactivity into the receiver blocks occurred regardless of whether the results were based on the actual amounts of radioactivity in the receiver block, or on the amounts in the receiver block expressed as a percentage of the net total radioactivity accumulated from the donor block. Only one radioactive substance was present in the receiver block and it ran to the same Rf as IAA in the isopropanol: ammonium: water solvent system.The amounts of radioactivity moving into that part of the root segment at least 6 mm distant from the end in contact with either an apical or a basal donor block were assessed. An acropetal polarity in the movement of radioactivity was observed on the basis of the actual amounts of radioactivity in these distal parts of the segments, but no such polarity was evident when the amounts of radioactivity were expressed as a percentage of the net total accumulated from the donor block. At least 3 radioactive substances were present in the tissue in addition to the substance running to the same Rf as IAA. The distribution of radioactivity in the segment cannot therefore be used to assess the distribution of IAA.Acropetal movement of radioactivity into an apical receiver block is not dependent upon the continued uptake of IAA at the basal end of the segment. No distinct pulses of radioactivity were detected moving through the root segments.Only a small part of the radioactivity in the root segment appears to be located in the polar transport system, while the bulk is not. The polarity found in the movement of the bulk radioactivity within the segment seems to be related to the polarity in IAA uptake from the donor blocks.  相似文献

本文以“京欣1号”母本和“伊选”西瓜4天苗龄子叶为外植体,研究离体培养植株高频率再生体系。结果表明:“伊选”子叶远轴端外植体的再生频率仅为10%,子叶近轴端外植体在5mg/LBA 0.1mg/L IAA的激素组合下植株再生频率为100%,平均每个外植体的丛生芽数在所有组合中最多,为10.3个;“京欣1号”母本子叶近轴端外植体在2mg/LBA 0.5mg/L IAA激素组合下植株再生频率为100%,平均每个外植体的丛生芽数在所有组合中最多,达6.9个。本试验条件下,子叶近轴端外植体接种4天即分化出不定芽,至再生苗的移栽仅需40天,在MS 0.1mg/L NAA的生根培养基上的生根率为97.3%,移栽成活率达98.5%。  相似文献

Auxin-calcium interaction has been studied to understand their involvement in adventitious root initiation from the hypocotyl explants of sunflower (Helianthus annuus L.). When hypocotyl explants were cultured on MS medium (containing calcium), 1 mg l-1 IAA was found to be optimal for root induction. However, the hypocotyl explants washed in EGTA (10-5M) solution for the removal of extracellular calcium, when cultured on medium containing IAA and calcium, exhibited enhanced rooting response. When EGTA-washed explants were cultured on the medium supplemented with lanthanum chloride (10-6 and 10-5M), it resulted in the inhibition of the rooting response and this inhibitory effect could be alleviated by the simultaneous addition of IAA. Similar observations have been made by using calcium channel blockers, verapamil and TMB-8, and also a calmodulin inhibitor, trifluoperazine. A net influx of extracellular calcium in the differentiating cells is thus presumed to accompany the auxin-induced response. These results have been discussed in light of initial lack of polarity in the decapitated hypocotyl segments subjected to auxin treatment.  相似文献

Hormone levels were measured in the apical, median and basal parts of the explants using an avidin-biotin based enzyme-linked immunosorbent assay (ELISA) after a purification of the extracts by high performance liquid chromatography (HPLC). All hormones showed rapid and considerable changes during the first eight days of growth. Exogenous IBA was probably transformed into IAA mainly in the basal part of the explant, and BA penetrated quickly. ABA levels were transiently enhanced in the apical part of the explants bearing young leaves. These phenomena are discussed in connection with the axillary bud reactivation.  相似文献

Cichorium intybus L. (cv. Bea) root explants grown in continuous light had a higher fresh weight, lignin and chlorophyll content than explants grown in darkness. Intermediary values were found when the light conditions were switched after 6 days. Peroxidase activities (EC 1.11.1.7) were measured in the apoplastic fluid with guaiacol, indoleacetic acid (IAA) and syringaldazine as substrates. There was an inverse relationship between specific IAA oxidase activity and explant growth. Specific syringaldazine oxidase activity (SSO) correlated with the lignin content. Analysis by isoelectric focusing (IEF) showed that the apoplastic fluid contained both cationic and anionic peroxidase isoforms, whose expression differed according to culture conditions. Isoform isoelectric point (pI) 7.0 was only detectable when the explants were cultured for 12 days in darkness. When explants were grown for the first 6 days in light, SSO and lignin content were high and the isoforms pI 4.0, 6.6, 7.6 and 8.1 were highly expressed. Conversely, when the first 6 days were in darkness, specific IAA oxidase activity was high and the isoforms pI 4.5, 6.7 and 6.8. were most strongly expressed. The isoperoxidases pI 7.8 and 7.9 were strongly expressed when the explants were cultured for at least 6 days in darkness.  相似文献
