反转录转座子通过RNA中间体进行反转录而转座,广泛分布于各种植物基因组中,拷贝数多,异质性高,在种内和种间表现出较高的序列差异性和丰富的插入多态性。针对这些特点,开发出了几种基于反转录转座子的分子标记,如SSAP、RIVPI、RAP、REMAP和RBIP等。由于反转录转座子标记能揭示出丰富的多态性,因而在遗传多样性和系谱研究、遗传连锁图谱构建及性状基因定位等方面得到了应用。随着分离技术的不断改进,获取序列信息更加容易,反转录转座子作为分子标记用于作物遗传育种将具有广阔前景。  相似文献

Thirty-five grape varieties were evaluated for their molecular diversities based on inter-primer binding site (iPBS) markers. Fifteen selected iPBS primers generated a total of 99 polymorphic DNA bands with 86.3% polymorphism. The effectiveness of iPBS marker is comparable to or even more efficient than other retrotransposon-based markers in grape. The differentiation between cultivated and wild grape varieties were clearly showed by both UPGMA cluster analysis and PCoA analysis revealing that Chinese cultivated and wild grape germplasm are highly divergent and possess abundant genetic diversities. This study also confirmed that the iPBS marker is a simple, informative, reproducible and suitable method for grape genetic diversity evaluation.  相似文献

The variation in transposition history of different Ty1-copia group LTR retrotransposons in the species lineages of the Pisum genus has been investigated. A heterogeneous population of Ty1-copia elements was isolated by degenerate PCR and two of these (Tps12 and Tps19) were selected on the basis of their copy number and sequence conservation between closely related species for further in-depth study of their transpositional history in Pisum species. The insertional polymorphism of these elements and the previously characterised PDR1 element was studied by sequence-specific amplification polymorphism (SSAP). Each of these elements reveals a unique transpositional history within 55 diverse Pisum accessions. Phylogenetic trees based on the SSAP data show that SSAP markers for individual elements are able to resolve different species lineages within the Pisum genus. Finally, the SSAP data from all of these retrotransposon markers were combined to reveal a detailed picture of the intra and inter-species relationships within Pisum. Summary RFLP variability was studied in eight U.S. peanut cultivars, representing the four market types, and in 14 wild Arachis species accessions, using random genomic clones from a PstI library. Very low levels of RFLP variability were found among the allotetraploids, which included the U.S. cultivars and Arachis monticola, a wild species. The diploid wild species were very diverse, however. RFLP patterns of the allotetraploids were more complex than the diploids, and the two constituent genomes could usually be distinguished. On the basis of RFLP band sharing, A. ipaensis, A. duranensis, and A. spegazzinii appeared most closely related to the diploid progenitor species of the allotetraploids. A dendrogram of relationships among the diploid wild species was constructed based on band sharing.  相似文献

本文旨在探讨植物种质资源保存中由于人为和自然缘故导致遗传结构改变的评价指标和评价方法.在介绍植物种质资源保存研究一些基本概念的基础上,归纳了测度种质库(收集品)遗传潜势的6种遗传多样性统计指标,包括同一变异层次的类型数、类型分布均衡度、遗传相似性与遗传距离、遗传方差与遗传变异系数、多元变异指数以及亲本系数.指出若无遗传丰富度相伴,单有遗传离散度并未提供遗传多样性的完整测度.探讨了人为条件导致植物种质资源遗传结构改变的遗传流失、环境胁迫所致植物种质资源遗传结构改变的遗传脆弱性和种子扩繁所引发的植物种质资源遗传结构改变的遗传漂变和遗传漂移等的统计指标.文末给出了自花授粉植物和异花授粉植物群体适宜样本容量研究的个例.  相似文献

缢蛏种群遗传多样性的周年变异分析   总被引:1,自引:0,他引:1  
利用线粒体COI标记分析了福建省宁德市漳湾镇横屿村滩涂5个月份缢蛏群体(S3,S5,S7,S9,S11)的遗传结构,以期评估不同月份时期种群的遗传多样性差异。基于线粒体COI基因的结果表明,5个缢蛏群体的平均核苷酸差异数位于2.7836-3.6894之间,核苷酸多样性指数位于0.0050-0.0066之间,遗传多样性水平大致表现为S3和S5群体较高于S7和S9群体,明显高于S11群体。AMOVA分析结果显示,群体间遗传变异量占总变异的7.18%,而群体内变异达到了92.82%,说明遗传差异主要来自于群体内部。由此可见,从3月份到11月份,缢蛏群体的遗传多样性水平呈现出下降趋势,尤其是在11月份,差异最为明显。  相似文献

Amphitetranychus viennensis (Zacher) is a serious economic pest of various fruit trees. Due to its restricted geographic distribution in Asia and Europe, it is an important quarantine species in the North American countries. Despite its significance in agricultural damage and quarantine, the genetic diversity of this species has not been studied in detail. In this study, we determined the nucleotide sequences of mitochondrial cytochrome c oxidase subunit I (COI) gene and internal transcribed spacer 2 (ITS2) region of the nuclear ribosomal DNA in A. viennensis collected from various regions of Korea. Variations in the COI and ITS2 sequences of the Korean samples ranged up to 6.92 and 4.02 %, respectively. A sequence comparison showed that the Korean samples were divided into two groups, one comprising the Geoje and Jinju samples, and the other comprising all samples except for the Geoje/Jinju samples. In most samples, except for the Geoje/Jinju samples, variations in both COI and ITS2 sequences were less than 1.04 and 0.99 %, respectively. However, the variations between two groups were 6.23–6.92 % and 3.02–4.02 %, respectively. In addition, the phylogenic analysis performed with sequences in the GenBank database showed that the samples from Geoje/Jinju, which are located in the southern region of Korea, were most similar to the Japanese samples, but the others were most similar to those of China and Taiwan. Thus, our study suggests that two genetically different groups of A. viennensis exist in Korea.  相似文献

高等植物中的反转录转座子是构成植物基因组的重要成分之一.它分病毒家族和非病毒家族两类,病毒家族包括反转录病毒和类似于反转录病毒的非病毒转座子,病毒家族中的反转录转座子可再细分为Ty3-gypsy类和Ty1-copia类;非病毒家族可细分为LINE类和SINE类.正常情况下大部分反转录转座子不具有活性,某些生物或非生物因素胁迫可激活部分反转录转座子转座.反转录转座子自身编码反转录酶进行转录,以\"拷贝-粘贴\"的转座模式导致基因组扩增和进化.具有活性的反转录转座子通过插入产生新的突变,可作为一种基因标签技术,应用于功能基因组学研究,并成为研究植物基因功能和表达的重要技术平台.本文综述了近几年来在植物反转录转座子方面的研究进展,主要包括植物反转录转座子的结构、特征、活性及其对基因组的影响和它们在功能基因组学中的应用.  相似文献

In plant populations a positive correlation between population size, genetic variation and fitness components is often found, due to increased pollen limitation or reduced genetic variation and inbreeding depression in smaller populations. However, components of fitness also depend on environmental factors which can vary strongly between years. The dry grassland species Muscari tenuiflorum experiences long term habitat isolation and small population sizes. We analyzed seed production of M. tenuiflorum in four years and its dependence on population size and genetic variation. Genetic diversity within populations was high (AFLP: He = 0.245; allozymes: He = 0.348). An analysis of molecular variance revealed considerable population differentiation (AFLP: 26%; allozyme: 17%). An overall pattern of isolation by distance was found, which, however was not present at distances below 20 km, indicating stronger effects of genetic drift. Genetic diversity was positively correlated to population size. Self pollination reduced seed set by 24%, indicating inbreeding depression. Reproductive fitness was not correlated to genetic diversity and a positive correlation with population size was present in two of four study years. The absence of a general pattern stresses the importance for multi-year studies. Overall, the results show that despite long term habitat isolation M. tenuiflorum maintains seed production in many years independent of population size. The long term persistence of populations is thus expected to depend less on intrinsic genetic or demographic properties affecting seed production but on successful plant establishment and persistence, which latter are based on conservation and protection of suitable habitats.  相似文献

Shortleaf and loblolly pine trees (n = 93 and 102, respectively) from 22 seed sources of the Southwide Southern Pine Seed Source Study plantings or equivalent origin were evaluated for amplified fragment length polymorphism (AFLP) variation. These sampled trees represent shortleaf pine and loblolly pine, as they existed across their native geographic ranges before intensive forest management. Using 17 primer pairs, a total of 96 AFLPs between shortleaf pine and loblolly pine were produced and scored on the sample trees and two control-pollinated F1 interspecies hybrids and their parents. In addition, the well known isocitrate dehydrogenase (IDH) isozyme marker was scored for all trees. IDH detected two putative hybrids among the loblolly pine samples and two among the shortleaf pine samples, while either 13 or 12 putative hybrids were detected using all AFLP markers and IDH and either NewHybrids or Structure software, respectively. Results of this study show that later generation hybrids can be reliably identified using AFLP markers and confirmed that IDH is not a definitive marker for detecting hybrids; that is, at least in some seed sources, the alternative species’ IDH allele resides in the source species. Based on all the markers, hybridization frequency varied geographically, ranging from 30% in an Arkansas seed source to 0% in several other seed sources. The hybridization level was higher in populations west of the Mississippi River than in populations east of the river; the shortleaf pine hybridization rates were 16.3% and 2.4% and the loblolly pine rates were 4.5% and 3.3%, west and east of the river, respectively. The results suggest that hybridization between these two species is significant but varies by seed source and species, and the potential for the unintended creation of hybrids should be considered in forest management decisions regarding both natural and artificial regeneration.  相似文献

This paper presented a statistical and comparative analysis of common parameters of plant genetic diversity by using relevant data of 314 wild plant species from 235 published articles. The results indicated that the parameters of genetic diversity revealed by RAPD and AFLP are comparable, but all parameters of genetic variation detected by ISSR, allozyme and SSR are incomparable , which are not comparative with those by RAPD and AFLP. The genetic differentiation value Gst based on Hardy-Weinberg equilibrium is obviously lower than the value Φst based on AMOVA analysis, which showed that these two parameters are incomparable as well. Furthermore, the statistical and comparative results of genetic diversity of 179 plant species by RAPD and AFLP indicated that at population level: 1) the genetic diversity of gymnosperm is higher than those of both dicotyledon and monocotyledon of angiosperm, but lower genetic differentiation; 2) the genetic diversity of tree is higher than those of shrub and herb, but lower genetic differentiation; 3) the clonal plant has higher genetic diversity than those reproduce sexnally, and 4) the cross-breeding plant has higher genetic diversity than self- breeding plant; 5 ) the widespread plant species has higher genetic diversity than the rare, endangered or endemic species.  相似文献
