烟草脱外壁花粉人工萌发与离体授粉实验系统的建立   总被引:4,自引:0,他引:4  
通过花蕾低温处理、花药漂浮培养与花粉短时酶解程序可脱去花粉外壁,分离出烟草(Nicotianatabacum L.)的脱外壁花粉。研究了分离过程中的酶液渗透压、培养基中聚乙二醇(PEG)与蔗糖以及添加水解乳蛋白等因素对脱外壁花粉人工萌发的影响。在含30%PEG-6000与0.1%水解乳蛋白的D_2培养基中,萌发率最高达57.8%;花粉管生长正常,培养24h后一半以上的花粉管中生殖细胞分裂成精子。用微滴和贴滤纸小片的方法将30~40粒脱外壁花粉授予柱头上,近一半能萌发花粉管并在花柱中生长。采取授粉后子房培养方法,获得了种子与幼苗。从而建立了脱外壁花粉离体授粉实验系统。讨论了脱外壁花粉人工萌发与离体授粉实验系统的建立对于研究外壁在花粉萌发中的生物学功能以及开拓新的转基因技术等方面的意义。  相似文献

脱外壁花粉的分离与人工萌发已在芸苔属(Brassica L.)中取得成功。由于其缺乏外壁与萌发沟这一特点,脱外壁花粉是研究花粉萌发的有用的实验系统。作者重点研究了脱外壁花粉分离与萌发过程中有关极性形成、萌发位点预定及新壁的合成等问题。结果表明在外壁脱离之前花粉已经活化并预先建立了极性;脱外壁后的萌发位点仍位于原萌发沟处;在萌发位点处合成的新壁可能起限定花粉管直径的作用。由此推论,脱外壁花粉可在花粉生物学研究中广泛加以应用。  相似文献

Studies on protoplasts isolation were carried out with mature pollen grains of 29 samples of species of Allium aflatunense, A. cepa, A. fistulosum, A. karataviense, A. longicuspis, A. nutans, A. odorum, A. sativum and A. schoenoprasum. Surface sterilized pollen grains drifted from crushed anthers were incubated in an enzyme solution containing 1% (w/v) cellulase Onozuka R-10, 1% (w/v) Macerozyme R-10, 0,5 mol l-1 sucrose and the basal salts of Nitsch medium. Protoplasts were released within 3 to 120 min, either from the pollen grain, through a slightly disturbed germination pore (narrow aperture), or through a wider aperture, when the exine surrounding the germination pore was disturbed. For the first time, protoplasts were obtained from 13 genotypes of 6 Allium species, at a rate of 1 to 30% of the digested intact pollen grains, depending on the genotype.  相似文献

 Gene constructs containing the β-glucuronidase (GUS) gene or green fluorescent protein (GFP) gene under the control of pollen-specific promoter Zm13-260 from maize were introduced by particle bombardment into de-exined pollen of Nicotiana tabacum. The de-exined pollen exhibited transient expression of the GUS or GFP gene as indicated by histochemical and fluorescent assay, respectively. The frequency of de-exined pollen transformation with the GUS or GFP gene was approximately 6 and 3 times higher, respectively, than that of pollen with intact walls, indicating that pollen deprived of the exine barrier responded better to foreign gene transfer than did the original. Cytological observation of GUS-expressing pollen grains showed that introduced gold particles were visible in the cytoplasm and vegetative nucleus as well as in the generative nucleus. A successful mass isolation of viable sperm cells from stored bicellular pollen of Lilium davidii Duch. was reported. When fresh pollen was cultured in BKS 15 medium, 87 % germinated in which the generative cells of the fresh pollen underwent mitosis and formed sperm cells within 28 hours. For pollen stored at -20℃ and -70℃ for 6 months, only less than 20 % germinated; but the germination percentage rose to 80 % after they have been hydrated and gradually warmed. Pollen grains of L. davidii which have been stored at -70 ℃ for 6 months, after being thawed were firstly germinated in a 15% sucrose medium for 28 hours, and then osmotically shocked with 10 % sucrose Solution. The solution was later adjusted to a final sucrose concentration of 15%. After density gradient centrifugation, 4 mL suspension of purified sperm cells with a density of 6 × 106 cells/mL were obtained at the interphase of 5%—15% percoll, with a 12% yield of viable sperm cells. The purified sperm cells had a diameter of 13—15 μm and reacted positive to fluorochrome, indicating that they have intact plasma membrane.  相似文献

以紫荆(Cercis chinensis Bunge)花粉为材料,采用人工培养法研究了紫荆花粉离体萌发的最适条件.结果表明:蔗糖、硼酸在一定质量浓度范围内能促进花粉的萌发,最适花粉萌发的质量浓度分别为蔗糖50 mg/L以及硼酸20 mg/L,最佳萌发温度20℃.正交试验设计结果表明:花粉萌发的最佳质量浓度分剐为70 mg/L蔗糖以及18mg/L硼酸,最佳萌发温度为22℃,此时的萌发率可达60.51%.  相似文献

以含笑(Michelia figo)花粉为试验材料,采用花粉人工培养法研究了含笑花粉离体萌发的最适条件。结果表明:蔗糖、硼酸在一定浓度范围内能促进花粉萌发,含笑花粉萌发的最适蔗糖和硼酸分别为50 mg.L-1和5mg.L-1,萌发率分别为21.51%和44.14%;花粉最佳萌发温度为25℃,萌发率达53.33%。正交试验设计结果表明,花粉萌发的最佳组合为70 mg.L-1蔗糖+5 mg.L-1硼酸+28℃,萌发率可达65.33%。  相似文献

Protoplasts were released in large quantities from mature pollen of Brassica napus L. and B. campestris var. purpurea for the first time, with a yield up to 66.7% and 70.4% respectively. Most of the pollen protoplasts were viable as tested by fluorochromatic reaction with fluorescein diacetate. The success of isolation of pollen protoplasts in these two Brassica species relied on a technique modified from the previous method developed for several monocotyledonous flowers. The procedure included two steps: First, the pollen was hydrated in 1 mol/L of sucrose solution at 28–30℃ for ca. 9h. During this process, the exine of most pollen dehisced and was detached from the pollen grains which were then covered by intine alone. Second, the hydrated pollen was transferred into an enzyme solution containing 1% cellulase, 1% pectinase, I mol/L mannitol, 0.5% potassium dextran sulphate and Ks medium salts. After 4–6 of enzymatic maceration, the intine was degradated resulting in the release of protoplasts. Factors affecting the two steps have been investigated.  相似文献

采用液体培养法研究不同培养基组分和培养条件对蜡梅花粉萌发和花粉管生长的影响。结果表明:(1)PEG-4000是蜡梅花粉离体培养所必需的培养基成分,当培养基中无PEG-4000时,花粉不能正常萌发。(2)培养基内低浓度蔗糖对花粉萌发和花粉管的生长无显著影响,但随着蔗糖浓度的升高,则对花粉萌发和花粉管生长表现出强烈的抑制作用,且浓度越高,抑制效应越强。(3)培养基内其它组分分别在一定浓度范围(0~250g/L PEG-4000、0~50mg/L硼酸、0~30mg/L硝酸钙)内对花粉萌发及花粉管生长有促进作用,但超过上述高限值时则起抑制作用。(4)培养基内镁和钾的浓度对花粉萌发及花粉管生长影响不显著。研究表明,蜡梅最适花粉液体培养基组分为250g/L PEG-4000+50mg/L H3BO3+30mg/L Ca(NO3)2.4H2O,且在pH 5.5、温度15℃和600lx的光照培养条件下蜡梅花粉萌发和花粉管生长最佳。  相似文献

A new method for isolation of quantities of mature pollen protoplasts in Nicotiana tabacum has been established. The first step was to germinate mature pollen in Brewbaker and Kwack medium containing 20% sucrose. When most of the pollen grains had just germinated short pollen tubes, they were transferred to an enzymatic solution for the second step. The enzymatic solution contained 1% pectinase, 1% cellulase, 0.5% potassium dextran sulfate, 1 mol/L mannitol, 0.4 mol/L sorbitol in Dx medium with or without 15% Ficoll. The enzymes firstly degraded the pollen tube wall and then the intine. As a result, intact pollen protoplasts were released with the isolation rate up to 50%-70%. Factors affecting pollen protoplast isolation during the germination and maceration of pollen grains were studied. The suceees depended on two key points:pollen germination duration and osmotieum concentration. The optimal germination duration was 30 rain at 30℃. When it was too long, long pollen tubes formed and subsequently, large number of subprotoplasts instead of whole protoplasts were yielded, as the case reported by previous investigators. The optimal concentration of mannitol and sorbitol in enzyme solution was as high as 1.4 mol/L in total. Lowering of the osmoticum concentration resulted in decrease of percentage of pollen protoplasts.  相似文献

山玉兰(Magnolia delavayi)是木兰科木兰属的常绿乔木或大型灌木,是重要的园林造景、庭院绿化素材,也是重要的育种资源。山玉兰花粉的研究对其杂交育种的成败具有重要影响,但目前尚未见其花粉活力的相关报道。该研究以新鲜的山玉兰花粉为对象,采用悬滴培养法分析了温度、光照以及培养液的蔗糖和硼酸浓度对山玉兰花粉萌发的影响。结果表明:(1)山玉兰花粉萌发时,最适宜的温度为27℃。(2)光暗条件下,山玉兰花粉以浓度为5%的蔗糖培养效果最佳,其萌发率在16%以上;而硼酸浓度则以0.001%的培养效果最佳。(3)蔗糖与硼酸共同作用可有效促进花粉萌发和花粉管生长。其中,在光照条件下,以5.0%蔗糖+0.001%硼酸为最适宜的培养液,花粉萌发率达41.27%,花粉管长达281.49μm;而在黑暗条件下,则以5.0%蔗糖+0.01%硼酸为最适宜的培养液,花粉萌发率达45.71%,花粉管长达254.00μm。该研究结果为进一步开展人工辅助授粉、发掘山玉兰的种质资源工作奠定了基础。  相似文献

I n vitro and in vivo rehydration and germination in Cupressus arizonica pollen were examined using light and scanning electron microscopy. Shed pollen has 12.6% water content, which reduced to 8.2% after dispersal, and this latter pollen survived for some months at room temperature and for years at −10 °C. Rehydration requires breaking of the sporoderm walls and depends on the composition and pH of the rehydration medium. Acidity restrains the breakage, while alkalinity promotes it. Pollen division follows exine shedding and requires the persistence of the mucilaginous layer; hence, pH values countering these outcomes prevent division. Division results in a large and a small cell separated by a callosic wall. A pollen tube develops from the innermost intine of the large cell, which is callosic, and extends into the mucilaginous middle intine. The percentage germination never exceeded 17% in all tested media. In vivo , pollen rehydrates and casts off the exine in the micropylar drop. Drop withdrawal brings pollen to the apical nucellar cells that degenerate in the meantime, and it leaves a deposit on the surface of the micropylar canal. After contaction of the nucellar cells, the pollen flattens and its mucilaginous layer shrinks and disappears. This occurs simultaneously with sealing of the micropylar canal. During this time, pollen divides asymmetrically without the callosic wall, and the larger cell develops a tube in the interface with the nucellus. Only some pollen grains accomplish adhesion to the nucellus and germinate. The in vitro and in vivo developmental stages are discussed.  相似文献

采用单因素实验设计对适宜于曼地亚红豆杉‘Hicksii’(Taxus media‘Hicksii')花粉活力检测的TTC染色法和离体培养法的实验条件进行了选择,并采用优化条件研究了在25℃~-196℃条件下储藏13周花粉活力的变化.结果表明:采用TTC染色法测定的花粉活力均高于离体培养法.在TTC染色法的3个影响因素(染液pH、TTC浓度和染色温度)中,染液pH对检测结果有极显著影响,而染色温度和TTC浓度则无显著影响,但温度对染色速率有影响.在离体培养法的3个影响因素(培养基中蔗糖添加量、H3BO3和Ca(NO3)2浓度及培养温度)中,蔗糖添加量对检测结果有极显著影响,在含质量体积分数15%蔗糖的培养基上花粉活力最高,而在含质量体积分数20%和25%蔗糖的培养基上花粉均不能萌发;在含100和200 mg ·L-1H3BO3的培养基中添加200 mg·L-1Ca(NO3)2均能显著提高花粉活力;培养温度对花粉萌发速率有影响但对花粉活力没有明显影响.TTC染色法的最优检测条件为:用5.0g·L-1TTC染液(pH 7.0)在35℃下染色2.0h;离体培养法的最优检测条件为:用含质量体积分数15%蔗糖、100 mg·L-1H3BO3和200 mg·L-1Ca(NO3)2的培养基暗培养4d.在25℃、4℃、0℃、-20℃、-80℃和-196℃条件下储藏13周,‘Hicksii'花粉活力和保持时间有明显差异,其中,于-80℃和-196℃储藏3d 花粉就丧失活力;于25℃和-20℃储藏7周、4 ℃储藏10周,花粉仍有一定的活力;而在0℃条件下花粉活力最高,且储藏11周花粉仍有活力.推测曼地亚红豆杉‘Hicksii’花粉对低温的抗性较差,0℃为其适宜的储藏温度.  相似文献

The effect of medium concentration, pollen grain concentration, pH of the media, light and temperature on the germination of Vin ca rosea pollen grains, and the growth of their pollen tubes in vitro have been studied. The pollen grains germinate best at a sucrose concentration between 14.2% and 30%; when the pollen grain concentration exceeds 800 per 0.0234 ml; at near neutral pH (6.5); in darkness and at a temperature close to 30°. Moreover buffering ions affect the growth of the pollen tubes. Pollen grains remain viable in a wide range of temperatures, and the wall of the pollen grain is capable of withstanding severe osmotic imbalance. Low temperature induces spherical swellings at the tips of the pollen tubes, followed by accumulation of a hyaline plug.  相似文献

(南京农业大学园艺学院,南京210095)摘要:研究培养基成分、pH值和培养方式对梅花花粉离体萌发和花粉管生长的影响。结果表明:不同品种梅花花粉离体萌发的最适培养基为ME3+200g.L-1PEG4000(pH5.0),品种‘淡丰后’、‘久观绿萼’、‘喧妍宫粉’和‘月光玉蝶’最高萌发率可分别达到58.6%、60.6%、85.6%和50.7%。PEG4000能显著促进梅花花粉萌发,在培养基各成分中作用最大,不可替代。低浓度(50g.L-1)蔗糖对梅花品种花粉萌发作用不显著,而高浓度(≥100g.L-1)蔗糖明显抑制花粉萌发和花粉管生长。固体和液体培养对梅花花粉离体萌发的影响差异不显著。  相似文献

马占相思(Acacia mangium)是含羞草科(Mimosaceae)金合欢属(Acacia Mill.)树种,在我国的广东、广西、福建以及海南等省(区)大面积种植推广。马占相思用途广泛,其杂交育种工作近年来逐渐受到重视,马占相思花粉研究对其杂交育种成败具有重要影响。该研究以新鲜的马占相思花粉为供试材料,开展不同蔗糖浓度(50、100、150、200、250和300 g·L~(-1))、不同硼酸浓度(100、200和300 mg·L~(-1))以及不同培养温度(26、28和30℃)对马占相思花粉离体萌发影响的研究,同时使用筛选出的马占相思花粉离体萌发的最佳处理,开展对另外3株与初试的马占相思生长发育状况接近的马占相思的花粉活力检测研究,并观察记录马占相思花粉离体萌发特征。结果表明:将刚开放的马占相思花序采回室内阴干,于次日10:00后,用毛笔刷法成功收集大量马占相思花粉,经显微镜观察检测,花粉纯度较高,可以保证后续研究备用;马占相思花粉离体萌发的最佳处理为200 g·L~(-1)蔗糖、100 mg·L~(-1)硼酸、28℃培养温度;马占相思花粉在培养24 h之后,花粉萌发率最高(为94.28%),花粉管最长(为6.0 D);检测3株马占相思的花粉萌发率分别为90.11%、82.31%、85.67%,具有显著差异。该研究结果为今后进一步开展人工控制授粉、选育优良的相思杂交新品种提供了基础。  相似文献

To study the cytological and biochemical characteristics of intine, pollen deprived of exine, or de-exined pollen, was isolated from the gymnosperms Pinus bungeana and Picea wilsonii. The factors influencing the isolation rate were examined. Cellulose, callose, pectin, and arabinogalactan proteins (AGPs) were localized in this material using fluorescent probes, and components of the isolated intine were further analyzed by Fourier transform infrared (FTIR) microspectroscopy. The isolation protocol was repeatable and reliable. Cellulose was found to be evenly distributed on the surface of the intine, as indicated by strong calcofluor White ST (CW) fluorescence, and aniline blue staining revealed that callose was present on the intine of P. bungeana but not on that of P. wilsonii. Immunolabeling revealed that acidic pectin epitopes recognized by the monoclonal antibody JIM5 were present on the pollen intine, as well as esterified pectin recognized by the monoclonal antibody JIM7, and AGPs recognized by the LM2 antibody. Two lectin binding sites, the concanavalin agglutinin (Con A) and soybean agglutinin (SBA) binding sites, were present on the intine surface, but no wheat germ agglutinin (WGA) binding sites were detectable. 采用花粉离体萌发法研究不同蔗糖浓度、硼酸浓度和不同温度对七叶树花粉萌发的影响及花粉活力测定,用联苯胺-过氧化氢法测定柱头可授性.结果表明:七叶树花粉萌发的最佳培养基是12%蔗糖+30 mg/L硼酸,花粉萌发的最适温度为25℃.雄花在开花当天花粉活力最高达75.69%,并在开花当天的上午10:00时,花粉活力最强,10:00~16:00花粉活力保持较高活力,是最佳授粉时段.两性花的柱头可授期持续时间较长,为8~9 d,开花3d达到最强,开花1~4 d柱头可授性保持较高水平,为授粉的最佳时间段.因此,从七叶树的花部特征、花粉活力与柱头可授性及花粉萌发的条件看,在长期的自然选择下七叶树在花部结构和开花生理上都是相配合的,以保障生殖成功.  相似文献
