Background

Dilated cardiomyopathy (DCM) is characterized by idiopathic dilation and systolic contractile dysfunction of the cardiac chambers. The present work aimed to study the alterations in gene expression of ion channels involved in cardiomyocyte function.

Methods and Results

Microarray profiling using the Affymetrix Human Gene® 1.0 ST array was performed using 17 RNA samples, 12 from DCM patients undergoing cardiac transplantation and 5 control donors (CNT). The analysis focused on 7 cardiac ion channel genes, since this category has not been previously studied in human DCM. SCN2B was upregulated, while KCNJ5, KCNJ8, CLIC2, CLCN3, CACNB2, and CACNA1C were downregulated. The RT-qPCR (21 DCM and 8 CNT samples) validated the gene expression of SCN2B (p < 0.0001), KCNJ5 (p < 0.05), KCNJ8 (p < 0.05), CLIC2 (p < 0.05), and CACNB2 (p < 0.05). Furthermore, we performed an IPA analysis and we found a functional relationship between the different ion channels studied in this work.

Conclusion

This study shows a differential expression of ion channel genes involved in cardiac contraction in DCM that might partly underlie the changes in left ventricular function observed in these patients. These results could be the basis for new genetic therapeutic approaches.  相似文献

目的:观察心肌解偶联蛋白2(uncoupling protein 2,UCP2)在酒精性心肌病(alcoholic cardiomyopathy,ACM)时表达变化及对心肌能量代谢的影响.方法:将Wistar大鼠分为三组,酒精组(A组,10只)、少量饮酒组(B组,7只)和对照组(C组,7只),三组给予相同饮食,酒精组通过采取逐渐增加饮用酒精浓度并长期定量摄入的方法建立ACM模型,少量饮酒组长期饮用少量低浓度酒精,对照组以水代酒.6个月后心脏彩超测定心功能;计算左室/体重指数;RT-PCR法测定心肌组织UCP2 mRNA表达;Western blot法测定心肌UCP2蛋白表达;高效液相色谱分析法测定心肌三磷酸腺苷酸(ATP)、二磷酸腺苷酸(ADP)、单磷酸腺苷酸(AMP)和磷酸肌酸(PCr)含量.结果:酒精组左心室射血分数和左心室短轴缩短率均低于对照组和少量饮酒组(P均<0.01),左心室舒张末期内径则高于对照组和少量饮酒组(P<0.01),左室/体重指数明显增加(P<0.01);酒精组UCP2 mRNA及蛋白表达高于对照组和少量饮酒组(P均<0.01);酒精组ATP、ADP、AMP和PCr较对照组和少量饮酒组明显减少(P均<0.01),相关性分析显示心肌组织ATP水平与UCP2蛋白表达呈显著负相关(r=0.896,P<0.01).结论:ACM时心肌UCP2表达明显增加,导致心肌能量代谢障碍,恶化心脏功能.  相似文献

目的利用cTnT^R141W转基因扩张型心肌病小鼠,研究人参皂甙Rb1对遗传性扩张型心肌病心功能及心脏重构的作用及其可能机制。方法将cTnT^R141W转基因小鼠随机分为模型组和人参皂甙Rb1治疗组(70 mg/kg/d),连续给药7个月,取野生型小鼠作为对照组。心脏超声检测心脏功能及几何构型。HE染色观察心肌细胞变化。透射电镜分析心肌超微结构。RT-PCR检测心肌粘附蛋白的表达。免疫荧光激光共聚焦观察心肌粘附分子Itga8的表达与分布。结果Rb1长期给药能显著改善该模型的心脏功能及几何构型。光镜和透射电镜观察显示Rb1能减轻心肌细胞排列紊乱及超微结构的破坏。RT-PCR结果显示,在模型中Cx40表达降低,E-cad、itga8和itgb1bp3表达升高,但在Rb1组中接近正常水平。免疫荧光激光共聚焦结果显示Rb1可降低Itga8的表达量并调节其分布。结论Rb1可改善扩张型心肌病模型的心功能,抑制心脏重构,其作用可能部分通过调节粘附蛋白的表达而实现的。  相似文献

While considerable evidence supports the causal relationship between increases in c-Myc (Myc) and cardiomyopathy as a part of a “fetal re-expression” pattern, the functional role of Myc in mechanisms of cardiomyopathy remains unclear. To address this, we developed a bitransgenic mouse that inducibly expresses Myc under the control of the cardiomyocyte-specific MHC promoter. In adult mice the induction of Myc expression in cardiomyocytes in the heart led to the development of severe hypertrophic cardiomyopathy followed by ventricular dysfunction and ultimately death from congestive heart failure. Mechanistically, following Myc activation, cell cycle markers and other indices of DNA replication were significantly increased suggesting that cell cycle-related events might be a primary mechanism of cardiac dysfunction. Furthermore, pathological alterations at the cellular level included alterations in mitochondrial function with dysregulation of mitochondrial biogenesis and defects in electron transport chain complexes I and III. These data are consistent with the known role of Myc in several different pathways including cell cycle activation, mitochondrial proliferation, and apoptosis, and indicate that Myc activation in cardiomyocytes is an important regulator of downstream pathological sequelae. Moreover, our findings indicate that the induction of Myc in cardiomyocytes is sufficient to cause cardiomyopathy and heart failure, and that sustained induction of Myc, leading to cell cycle re-entry in adult cardiomyocytes, represents a maladaptive response for the mature heart.  相似文献

Background

The endoplasmic reticulum (ER) is a multifunctional organelle responsible for the synthesis and folding of proteins as well as for signalling and calcium storage, that has been linked to the contraction-relaxation process. Perturbations of its homeostasis activate a stress response in diseases such as heart failure (HF). To elucidate the alterations in ER molecular components, we analyze the levels of ER stress and structure proteins in human dilated (DCM) and ischemic (ICM) cardiomyopathies, and its relationship with patient''s functional status.

Methods and Results

We examined 52 explanted human hearts from DCM (n = 21) and ICM (n = 21) subjects and 10 non-failing hearts as controls. Our results showed specific changes in stress (IRE1, p<0.05; p-IRE1, p<0.05) and structural (Reticulon 1, p<0.01) protein levels. The stress proteins GRP78, XBP1 and ATF6 as well as the structural proteins RRBP1, kinectin, and Nogo A and B, were upregulated in both DCM and ICM patients. Immunofluorescence results were concordant with quantified Western blot levels. Moreover, we show a novel relationship between stress and structural proteins. RRBP1, involved in procollagen synthesis and remodeling, was related with left ventricular function.

Conclusions

In the present study, we report the existence of alterations in ER stress response and shaping proteins. We show a plausible effect of the ER stress on ER structure in a suitable sample of DCM and ICM subjects. Patients with higher values of RRBP1 had worse left ventricular function.  相似文献

We aim to assess the effectiveness of feedback-controlled resonant drift pacing as a method for low energy defibrillation. Antitachycardia pacing is the only low energy defibrillation approach to have gained clinical significance, but it is still suboptimal. Low energy defibrillation would avoid adverse side effects associated with high voltage shocks and allow the application of implantable cardioverter defibrillator (ICD) therapy, in cases where such therapy is not tolerated today. We present results of computer simulations of a bidomain model of cardiac tissue with human atrial ionic kinetics. Reentry was initiated and low energy shocks were applied with the same period as the reentry, using feedback to maintain resonance. We demonstrate that such stimulation can move the core of reentrant patterns, in the direction that depends on the location of the electrodes and the time delay in the feedback. Termination of reentry is achieved with shock strength one-order-of-magnitude weaker than in conventional single-shock defibrillation. We conclude that resonant drift pacing can terminate reentry at a fraction of the shock strength currently used for defibrillation and can potentially work where antitachycardia pacing fails, due to the feedback mechanisms. Success depends on a number of details that these numerical simulations have uncovered.  相似文献

An experimentally naïve 37.7-kg Yorkshire-crossbred gilt died unexpectedly 2 d after arrival. Necropsy revealed severe dilated cardiomyopathy characterized grossly by markedly dilated ventricles and thinned ventricular walls and interventricular septum. Histologically there was multifocal myofiber attenuation and patchy loss of myofiber cross striations. The liver contained submassive to massive, diffuse hepatic centrilobular hemorrhage and degeneration. These lesions supported a diagnosis of dilated cardiomyopathy with right heart failure and secondary hepatic degeneration due to marked acute passive congestion. To our knowledge, this case is the first report of spontaneous dilated cardiomyopathy in swine and represents a potential diagnostic challenge regarding the differentiation of the cardiac-associated liver lesion from hepatosis dietetica. The diagnosis of dilated cardiomyopathy and right-sided heart failure was supported by the character of the hepatic lesion, absence of typical gross or histologic lesions of mulberry heart disease, and normal selenium levels.Abbreviations: DCM, dilated cardiomyopathyPigs are a common biomedical research model of cardiovascular disease, due to their large size, which accommodates surgical procedures, as well as their similarities to humans in regard to coronary vasculature and hemodynamics.15 However, compared with developmental (atrial or ventricular septal defects),10 nutritional (vitamin E–selenium deficiency),24 and infectious (pericarditis or endocarditis) etiologies, primary cardiomyopathies are not recognized as an important underlying etiology in swine.17 Aside from a documented familial preponderance for the development of hypertrophic cardiomyopathy in specific herds,4,11 other primary cardiomyopathies documented in humans and veterinary species have not been identified in pigs. In humans and dogs, dilated cardiomyopathy is the most prevalent cardiomyopathy reported and in humans is one the leading conditions necessitating a heart transplant.12,22 The current report describes the clinical presentation, gross necropsy findings, and histopathologic characterization of a case of spontaneous dilated cardiomyopathy and right-sided congestive heart failure in a production pig, with some potentially confounding similarities to hepatosis dietetica.  相似文献

Using a polarographic technique, we studied the peculiarities of energy metabolism in neurons of the rat brainstem structures related to normal physiological aging. Experiments were carried out under in vitro conditions on mitochondrial (MCh) suspensions prepared from the brainstem cells of young and old rats. In addition, we examined, using the same technique, the parameters of oxidative phosphorylation in analogous MCh suspension under conditions of experimental MCh dysfunction induced by single systemic injection of rotenone into young animals. In the case where we used a succinate + rotenone mixture as the substrate for oxidation, the intensity of ADP-stimulated respiration (V3) in preparations from brainstem neurons of old animals was significantly smaller (against the background of a decrease in the efficacy of respiration control, V3/V4). If a mixture glutamate + malate was used as the substrate for oxidation, the V3 and the efficacy of phosphorylation (ADP/O) decreased significantly. The experimental MCh dysfunction resulted in the lowering of practically all parameters of oxidation and phosphorylation under conditions of oxidation of glutamate + malate, as well as V3, V3/V4, and ADP/O, in the case where we used succinate + rotenone as the substrate for oxidation. Less expressed changes in the recorded indices upon oxidation of succinate + rotenone were indicative of activation of the succinate oxidase pathway; this preserved the electrotransport function of the respiratory chain in the MCh on a certain level and the ability of the latter to provide oxidative phosphorylation.  相似文献

摘要 目的:研究扩张型心肌病(DCM)患者左心室球形指数(LVSI)、单核细胞计数与高密度脂蛋白胆固醇(HDL-C)比值(MHR)和血清半乳糖凝集素-3(Gal-3)的变化,并分析其与DCM患者心功能的关系。方法:选择2018年1月-2022年10月在我院接受治疗的DMC患者68例作为研究组,并选取同期50例健康体检者作为对照组。研究组患者根据出院6个月份是否发生主要不良心血管事件分为MACE组和No-MACE组。比较各组间左心室射血分数(LVEF)、左心室舒张末期内径(LVEDD)、左室收缩末期内径(LVESD)、LVSI、单核细胞计数、HDL-C、MHR以及血清Gal-3。结果:(1)研究组患者LVEF显著低于对照组,而研究组患者LVEDD、LVESD以及LVSI均显著高于对照组(P<0.05)。(2)研究组患者单核细胞计数、HDL-C、MHR以及Gal-3均显著高于对照组(P<0.05)。(3)不同心功能分级DCM患者LVEF、LVEDD、LVESD、单核细胞计数和HDL-C组间比较无显著差异(P>0.05),但LVSI、MHR和血清Gal-3组间比较差异显著(P<0.05)。(4)MACE组患者LVSI、MHR和Gal-3均显著高于No-MACE组患者(P<0.05)。结论:LVSI、MHR和血清Gal-3在扩张型心肌病患者中升高,并与患者心功能分级有关,可作为DCM患者心功能恶化评价指标。  相似文献

应用凝胶内差异显示电泳技术研究肾阳虚大鼠肝线粒体蛋白质组,并从肝线粒体蛋白质组角度阐述肾阳虚与能量代谢的关系.8个分别来自于肾阳虚大鼠和正常大鼠的肝线粒体蛋白质样品(各4个)分别用荧光染料Cy3、Cy5标记,以及8个样品等量混合物用Cy2标记作为内标,每一Cy3、Cy5标记样品与Cy2标记的内标等量混合后在同一胶中进行电泳分离,经不同光激发后扫描得到不同样品的蛋白质组图谱.经DeCyder软件结合内标分析,以肾阳虚组动物与正常组动物肝线粒体蛋白质相差1.2倍以上的蛋白作为差异蛋白,实验共获得16个差异蛋白质,经质谱测定和与蛋白质文库比对,鉴定11个蛋白质.其中,肾阳虚动物热休克蛋白60和70、肌氨酸脱氢酶、氨甲酰磷酸合成酶、亚硫酸盐氧化酶、ATP合酶、醛脱氢酶和NADH脱氢酶表达量增加,而丙酮酸脱氢酶、α酮戊二酸脱氢酶、脂酰辅酶A脱氢酶和鸟氨酸氨基转移酶表达量降低.实验表明,肾阳虚动物能量代谢相关酶的变化与肾阳虚的临床虚寒症状有关.  相似文献

Morphine belongs among the most commonly used opioids in medical practice due to its strong analgesic effects. However, sustained administration of morphine leads to the development of tolerance and dependence and may cause long-lasting alterations in nervous tissue. Although proteomic approaches enabled to reveal changes in multiple gene expression in the brain as a consequence of morphine treatment, there is lack of information about the effect of this drug on heart tissue. Here we studied the effect of 10-day morphine exposure and subsequent drug withdrawal (3 or 6 days) on the rat heart proteome. Using the iTRAQ technique, we identified 541 proteins in the cytosol, 595 proteins in the plasma membrane-enriched fraction and 538 proteins in the mitochondria-enriched fraction derived from the left ventricles. Altogether, the expression levels of 237 proteins were altered by morphine treatment or withdrawal. The majority of changes (58 proteins) occurred in the cytosol after a 3-day abstinence period. Significant alterations were found in the expression of heat shock proteins (HSP27, α-B crystallin, HSP70, HSP10 and HSP60), whose levels were markedly up-regulated after morphine treatment or withdrawal. Besides that morphine exposure up-regulated MAPK p38 (isoform CRA_b) which is a well-known up-stream mediator of phosphorylation and activation of HSP27 and α-B crystallin. Whereas there were no alterations in the levels of proteins involved in oxidative stress, several changes were determined in the levels of pro- and anti-apoptotic proteins. These data provide a complex view on quantitative changes in the cardiac proteome induced by morphine treatment or withdrawal and demonstrate great sensitivity of this organ to morphine.  相似文献

The innate antiviral response is mediated, at least in part, by Toll-like receptors (TLRs). TLR3 signaling is activated in response to viral infection, and the absence of TLR3 in mice significantly increases mortality after infection with enteroviruses that cause myocarditis and/or dilated cardiomyopathy. We screened TLR3 in patients diagnosed with enteroviral myocarditis/cardiomyopathy and identified a rare variant in one patient as well as a significantly increased occurrence of a common polymorphism compared with controls. Expression of either variant resulted in significantly reduced TLR3-mediated signaling after stimulation with synthetic double-stranded RNA. Furthermore, Coxsackievirus B3 infection of cell lines expressing mutated TLR3 abrogated activation of the type I interferon pathway, leading to increased viral replication. TLR3-mediated type I interferon signaling required cellular autophagy and was suppressed by 3-methyladenine and bafilomycin A1, by inhibitors of lysosomal proteolysis, and by reduced expression of Beclin 1, Atg5, or microtubule-associated protein 1 light chain 3β (MAP1LC3β). However, TLR3-mediated signaling was restored upon exogenous expression of Beclin 1 or a variant MAP1LC3β fusion protein refractory to RNA interference. These data suggest that individuals harboring these variants may have a blunted innate immune response to enteroviral infection, leading to reduced viral clearance and an increased risk of cardiac pathology.  相似文献

The most common cause of dilated cardiomyopathy and heart failure (HF) is ischemic heart disease; however, in a third of all patients the cause remains undefined and patients are diagnosed as having idiopathic dilated cardiomyopathy (IDC). Recent studies suggest that many patients with IDC have a family history of HF and rare genetic variants in over 35 genes have been shown to be causative of disease. We employed whole‐exome sequencing to identify the causative variant in a large family with autosomal dominant transmission of dilated cardiomyopathy. Sequencing and subsequent informatics revealed a novel 10‐nucleotide deletion in the BCL2‐associated athanogene 3 (BAG3) gene (Ch10:del 121436332_12143641: del. 1266_1275 [NM 004281]) that segregated with all affected individuals. The deletion predicted a shift in the reading frame with the resultant deletion of 135 amino acids from the C‐terminal end of the protein. Consistent with genetic variants in genes encoding other sarcomeric proteins there was a considerable amount of genetic heterogeneity in the affected family members. Interestingly, we also found that the levels of BAG3 protein were significantly reduced in the hearts from unrelated patients with end‐stage HF undergoing cardiac transplantation when compared with non‐failing controls. 白藜芦醇具有多种生物学功能和药用价值,例如抗炎、抗衰老、抗病毒、抗肿瘤等。为了探讨白藜芦醇在外源性抗氧化剂方面的开发价值。本研究建立了跑步力竭SD大鼠模型,应用不同浓度的白藜芦醇处理大鼠4周。研究显示,白藜芦醇处理可以剂量依赖性方式提高大鼠的跑步力竭时间(p<0.05)。白藜芦醇处理以剂量依赖方式降低大鼠血清乳酸和尿素氮水平并升高游离脂肪酸水平(p<0.05)。白藜芦醇处理以剂量依赖方式升高大鼠体内超氧化物歧化酶和过氧化氢酶水平,并降低丙二醛水平(p<0.05)。白藜芦醇处理以剂量依赖方式降低血清肌酸激酶、天冬氨酸转氨酶和丙氨酸转氨酶水平(p<0.05)。此外,白藜芦醇明显减轻了大鼠骨骼肌的病理改变。因此,白藜芦醇可提高跑步力竭大鼠的抗疲劳能力,改善能量代谢方式,提高机体抗氧化能力,减少运动损伤。  相似文献

We developed a Raman spectroscopy-based approach for simultaneous study of redox changes in c-and b-type cytochromes and for a semiquantitative estimation of the amount of oxygenated myoglobin in a perfused rat heart. Excitation at 532 nm was used to obtain Raman scattering of the myocardial surface of the isolated heart at normal and hypoxic conditions. Raman spectra of the heart under normal pO2 demonstrate unique peaks attributable to reduced c-and b-type cytochromes and oxymyoglobin (oMb). The cytochrome peaks decreased in intensity upon FCCP treatment, as predicted from uncoupling mitochondrial respiration. Conversely, transient hypoxia causes the reversible increase in the intensity of peaks assigned to cytochromes c and c1, reflecting electron stacking proximal to cytochrome oxidase due to the lack of terminal electron acceptor O2. Intensities of peaks assigned to oxy- and deoxyhemoglobin were used for the semiquantitative estimation of oMb deoxygenation that was found to be of approximately 50 under hypoxia conditions.  相似文献

Resveratrol (RSV), polyphenol from grape, was studied to evaluate its effects on calorimetric parameters, energy metabolism, and antioxidants in the myocardium of diabetic rats. The animals were randomly divided into four groups (n = 8): C (control group): normal rats; C-RSV: normal rats receiving RSV; DM: diabetic rats; and DM-RSV: diabetics rats receiving RSV. Type 1 diabetes mellitus was induced with administration of streptozotocin (STZ; 60 mg−1 body weight, single dose, i.p.). After 48 hours of STZ administration, the animals received RSV (1.0 mg/kg/day) for gavage for 30 days. Food, water, and energy intake were higher in the DM group, while administration of RSV caused decreases (p<0.05) in these parameters. The glycemia decreased and higher final body weight increased in DM-RSV when compared with the DM group. The diabetic rats showed higher serum-free fatty acid, which was normalized with RSV. Oxygen consumption (VO2) and carbon dioxide production (VCO2) decreased (p<0.05) in the DM group. This was accompanied by reductions in RQ. The C-RSV group showed higher VO2 and VCO2 values. Pyruvate dehydrogenase activity was lower in the DM group and normalizes with RSV. The DM group exhibited higher myocardial β-hydroxyacyl coenzyme-A dehydrogenase and citrate synthase activity, and RSV decreased the activity of these enzymes. The DM group had higher cardiac lactate dehydrogenase compared to the DM-RSV group. Myocardial protein carbonyl was increased in the DM group. RSV increased reduced glutathione in the cardiac tissue of diabetic animals. The glutathione reductase activity was higher in the DM-RSV group compared to the DM group. In conclusion, diabetes is accompanied by cardiac energy metabolism dysfunction and change in the biomarkers of oxidative stress. The cardioprotective effect may be mediated through RVS''s ability to normalize free fatty acid oxidation, enhance utilization glucose, and control the biomarkers'' level of oxidative stress under diabetic conditions.  相似文献

In light of the increasing incidence of renal cell carcinoma (RCC), its molecular mechanisms have been comprehensively explored in numerous recent studies. However, few studies focus on the influence of multi‐factor interactions during the occurrence and development of RCC. This study aims to investigate the quantitative global proteome and the changes in lysine succinylation in related proteins, seeking to facilitate a better understanding of the molecular mechanisms underlying RCC. LC‐MS/MS combined with bioinformatics analysis are used to quantitatively detect the perspectives at the global protein level. IP and WB analysis were conducted to further verify the alternations of related proteins and lysine succinylation. A total of 3,217 proteins and 1,238 lysine succinylation sites are quantified in RCC tissues, and 668 differentially expressed proteins and 161 differentially expressed lysine succinylation sites are identified. Besides, expressions of PGK1 and PKM2 at protein and lysine, succinylation levels are significantly altered in RCC tissues. Bioinformatics analysis indicates that the glycolysis pathway is a potential mechanism of RCC progression and lysine succinylation may plays a potential role in energy metabolism. These results can provide a new direction for exploring the molecular mechanism of RCC tumorigenesis.  相似文献
