Mangosteen extracts (ME) contain high levels of polyphenolic compounds and antioxidant activity. Protective effects of ME against β-amyloid peptide (Aβ), induced cytotoxicity have been reported. Here, we further studied the protective effects of ME against oxidative stress induced by hydrogen peroxide (H2O2) and polychlorinated biphenyls (PCBs), and demonstrated the protection against memory impairment in mice. The cytoprotective effects of ME were measured as cell viability and the reduction in ROS activity. In SK-N-SH cell cultures, 200 μg/ml ME could partially antagonize the effects of 150 or 300 µM H2O2 on cell viability, ROS level and caspase-3 activity. At 200, 400 or 800 µg/ml, ME reduced AChE activity of SK-N-SH cells to about 60% of the control. In vivo study, Morris water maze and passive avoidance tests were used to assess the memory of the animals. ME, especially at 100 mg/kg body weight, could improve the animal’s memory and also antagonize the effect of scopolamine on memory. The increase in ROS level and caspase-3 activity in the brain of scopolamine-treated mice were antagonized by the ME treatment. The study demonstrated cytoprotective effects of ME against H2O2 and PCB-52 toxicity and having AChE inhibitory effect in cell culture. ME treatment in mice could attenuate scopolamine-induced memory deficit and oxidative stress in brain.  相似文献

Objectives are to examine the effects of sleep deprivation (SD) on spatial learning and memory in mice, to determine how SD effects the expression of phosphorylated cyclic AMP responsive element binding protein (pCREB) in mouse hippocampus, and to explore the mechanism of influence of sleep deprivation on cognitive function. Twenty, 3-month-old female C57BL/6J mice were randomly assigned into two groups, the sleep deprivation group (SD, n = 10) and control group with normal sleep (CC, n = 10). The mice in SD group were deprived sleep by “gentle touch” for 20 days and then all the mice were subjected for Morris Water Maze test to determine the mean latency of escape (LE). Percentage of time spent in the target quadrant was calculated. Mouse hippocampus pCREB levels were quantified by western blot. Compared with CC group, SD mice had a significantly longer mean LE time (P < 0.05) and spent less time in the target quadrant (P < 0.05). Western blot revealed that hippocampus pCREB levels in the SD group were significantly lower than that in control group (0.71 ± 0.03 vs 0.82 ± 0.06, P < 0.01). The impairment in spatial learning and memory in sleep-deprived animals may be associated with the reduction of pCREB in the hippocampus.  相似文献

Previously, we identified methoxsalen (8-methoxy-2′,3′,6,7-furocoumarin) as the bioactive compound probably responsible for acetylcholinesterase (AchE) inhibition achieved by feeding crude extract of Poncirus trifoliate. To confirm the activity of methoxsalen, Institute of Cancer Research (ICR) mice were fed a control or a methoxsalen-supplemented diet for 4 weeks, and then learning and memory enhancing effects with respect to trimethyltin (TMT)-induced neurotoxicity were evaluated. The brain tissues of ICR mice were dissected after completion of the behavioral tests for biochemical analysis. Methoxsalen effectively reversed TMT-induced memory impairment on both Y-maze and passive avoidance tests. Brain AchE activity was inhibited by the oral consumption of all concentrations of methoxsalen. Moreover, the level of oxidative stress was significantly ameliorated in the groups on methodsalen containing diets. This is the first in vivo study conducted with methoxsalen in the field of AD research, and it indicates that further investigation of methoxsalen is warranted.  相似文献

研究了罗格列酮对链脲佐菌素(streptozotocin, STZ)脑室内注射的阿尔茨海默病(AD)模型小鼠学习记忆减退的影响及机制.在小鼠脑室内注射STZ建立AD模型,治疗组小鼠采用罗格列酮灌胃给药30天.Morris水迷宫实验检测小鼠学习记忆能力,免疫印迹和免疫荧光检测Tau蛋白的磷酸化、神经丝(NFs)蛋白的磷酸化及糖基化、JNK和ERK蛋白的表达,微管结合实验检测Tau蛋白与微管的组装功能,荧光染料Fluoro-Jade B检测小鼠脑内退变神经元.结果显示,相比对照组,模型组小鼠平均逃避潜伏期和路径长度明显增加、穿越隐匿平台次数明显减少、Tau和NFs蛋白表达过度磷酸化、NFs蛋白糖基化减弱,而用罗格列酮干预的小鼠学习记忆改善并且Tau和NFs蛋白的磷酸化水平降低、NFs蛋白糖基化水平增加,Tau蛋白与微管结合能力改善,模型组JNK的磷酸化高于对照组和治疗组、模型组ERK1的磷酸化低于对照组和治疗组、各组在ERK2磷酸化无明显差异,模型组小鼠脑中FJB标记的退化神经元明显多于对照组和治疗组.结果说明,罗格列酮能改善STZ脑室内注射引起的小鼠学习记忆减退,其机制可能与改善胰岛素信号通路、降低Tau和NFs蛋白的过度磷酸化、减少神经退行性变有关.  相似文献

Dictyostelium discoideum amoebae have been used extensively to study the structure and dynamics of the endocytic pathway. Here, we show that while the general structure of the endocytic pathway is maintained in starved cells, its dynamics rapidly slow down. In addition, analysis of apm3 and lvsB mutants reveals that the functional organization of the endocytic pathway is profoundly modified upon starvation. Indeed, in these mutant cells, some of the defects observed in rich medium persist in starved cells, notably an abnormally slow transfer of endocytosed material between endocytic compartments. Other parameters, such as endocytosis of the fluid phase or the rate of fusion of postlysosomes to the cell surface, vary dramatically upon starvation. Studying the endocytic pathway in starved cells can provide a different perspective, allowing the primary (invariant) defects resulting from specific mutations to be distinguished from their secondary (conditional) consequences.Dictyostelium discoideum is a widely used model organism for studying the organization and function of the endocytic pathway. In Dictyostelium, the organization of the endocytic pathway is similar to that in higher eukaryotes. The pathway in Dictyostelium can be divided into four steps (see Fig. S1 in the supplemental material): uptake at the plasma membrane of particles and medium, transfer through early acidic endocytic compartments (lysosomes), passage into less acidic postlysosomes (PLs), and finally, exocytosis of undigested materials (17, 20). Thus, Dictyostelium recapitulates many of the functions of the endocytic pathway in mammalian cells, including some features observed in most cell types (lysosome biogenesis) and some observed only in specialized cells (phagocytosis, macropinocytosis, and lysosome secretion).Dictyostelium amoebae live in the soil, where they feed by ingesting and digesting other microorganisms. In addition, axenic laboratory strains can macropinocytose medium to ensure their growth. Accordingly, both in natural situations and in laboratory settings, the endocytic pathway plays a key role in the acquisition of nutrients by Dictyostelium cells. In agreement with this notion, several observations suggest that the physiology of the endocytic pathway is sensitive to nutrient availability. In particular, starvation induces secretion of lysosomal enzymes by an unknown mechanism (11). The morphology of the endocytic pathway is also sensitive to nutritional cues, as shown for example by the observation that formation of multilamellar endosomes is enhanced in cells fed with bacteria (18).Here, we analyzed the effect of starvation on the organization as well as the dynamics of the endocytic pathway. We found that, while the overall organization was not extensively modified in starved cells, the dynamics of endocytic compartments were altered. Moreover, analysis of two specific knockout mutants, the apm3 (6) and lvsB (8) strains, revealed that their phenotype was profoundly altered upon starvation, providing further insight about the role of Apm3 and LvsB in the endocytic pathway.  相似文献

In the present study, we identified the effects of inhaled Pimpinella peregrina essential oil (1 and 3 %, for 21 continuous days) on scopolamine-induced memory impairment, anxiety, and depression in laboratory rats. Y-maze and radial arm-maze tests were used for assessing memory processes. Also, the anxiety and depressive responses were studied by means of the elevated plus-maze and forced swimming tests. The scopolamine alone-treated rats exhibited the following: decrease of the spontaneous alternation percentage in Y-maze test, increase of the number of working and reference memory errors in radial arm-maze test, along with decrease of the exploratory activity, the percentage of the time spent and the number of entries in the open arm within elevated plus-maze test and decrease of swimming time and increase of immobility time within forced swimming test. Inhalation of the P. peregrina essential oil significantly improved memory formation and exhibited anxiolytic- and antidepressant-like effects in scopolamine-treated rats. Our results suggest that the P. peregrina essential oil inhalation ameliorates scopolamine-induced memory impairment, anxiety, and depression. Moreover, studies on the P. peregrina essential oil may open a new therapeutic window for the prevention of neurological abnormalities closely related to Alzheimer’s disease.  相似文献

Endogenous microbiota play essential roles in the host’s immune system, physiology, reproduction and nutrient metabolism. We hypothesized that a continuous administration of an exogenous probiotic might also influence the host’s development. Thus, we treated zebrafish from birth to sexual maturation (2-months treatment) with Lactobacillus rhamnosus, a probiotic species intended for human use. We monitored for the presence of L. rhamnosus during the entire treatment. Zebrafish at 6 days post fertilization (dpf) exhibited elevated gene expression levels for Insulin-like growth factors -I and -II, Peroxisome proliferator activated receptors -α and -β, VDR-α and RAR-γ when compared to untreated-10 days old zebrafish. Using a gonadotropin-releasing hormone 3 GFP transgenic zebrafish (GnRH3-GFP), higher GnRH3 expression was found at 6, 8 and 10 dpf upon L. rhamnosus treatment. The same larvae exhibited earlier backbone calcification and gonad maturation. Noteworthy in the gonad development was the presence of first testes differentiation at 3 weeks post fertilization in the treated zebrafish population -which normally occurs at 8 weeks- and a dramatic sex ratio modulation (93% females, 7% males in control vs. 55% females, 45% males in the treated group). We infer that administration of L. rhamnosus stimulated the IGF system, leading to a faster backbone calcification. Moreover we hypothesize a role for administration of L. rhamnosus on GnRH3 modulation during early larval development, which in turn affects gonadal development and sex differentiation. These findings suggest a significant role of the microbiota composition on the host organism development profile and open new perspectives in the study of probiotics usage and application.  相似文献

为观察延衰合剂(Yanshuai mixture,YSM)的延缓衰老作用,采用D-半乳糖连续颈背部皮下注射诱导亚急性衰老小鼠模型,造模同时给予YSM低、中、高剂量灌胃,连续10周。观察小鼠衰老征象,Moriss水迷宫检测学习记忆功能,测定血清及脑组织中总抗氧化能力(T-AOC)、超氧化物歧化酶(SOD)、丙二醛(MDA)。结果显示,与模型组比较,YSM各剂量组均能使小鼠逃避潜伏期明显缩短,原平台象限游泳时间明显延长(P<0.05);YSM高剂量组T-AOC、SOD活性显著升高(P<0.01),MDA水平显著下降(P<0.01)。表明YSM可明显改善衰老小鼠学习记忆功能,提高抗氧化能力,从而可能具有较好的延缓衰老的作用。  相似文献

目的 :探讨磁场作用对小鼠学习记忆能力的影响。方法 :应用水迷宫学习模型测定并对比 30分钟磁场处理组、1 5分钟磁场处理组和非磁场处理的正常对照组动物的空间学习记忆能力。结果 :水迷宫学习训练的实验表明 30分钟磁场处理组与正常对照组比较 ,动物到达水下平台的时间延长 ;游程增加 ;平均游速减慢 ,且均具有显著性差异 (p <0 .0 5)。 1 5分钟磁场处理组与正常对照组比较 ,动物到达水下平台的时间延长 ,且具有显著性差异 (p <0 .0 5) ;游程和平均速度与正常对照组相比无显著性差异 (p >0 .0 5)。结论 :磁场处理 30分钟或 1 5分钟损伤小鼠的空间学习记忆能力 ,且以 30分钟的磁场处理作用较强  相似文献

给小鼠断绝食物,观察小鼠学习记忆能力的变化及生理应激反应。结果表明,饥饿后小鼠的体重明显减轻,学习能力明显增强,记忆能力和大脑5-羟色胺含量无明显改变,心肌丙二醛含量下降,肝组织超氧化物歧化酶(SOD)活性无明显改变,血浆SOD活性提高,血浆SOD同工酶变带无明显改变。提示饥饿胁迫使小鼠产生适应性的生理反应,使小鼠学习能力提高,加强延缓机体衰退的生理功能;这些应激反应有利于小鼠在逆境下的生存。  相似文献

The therapeutic effects of bacteriophage (phage) KPP12 in Pseudomonas aeruginosa keratitis were investigated in mice. Morphological analysis showed that phage KPP12 is a member of the family Myoviridae, morphotype A1, and DNA sequence analysis revealed that phage KPP12 is similar to PB1-like viruses. Analysis of the phage KPP12 genome did not identify any genes related to drug resistance, pathogenicity or lysogenicity, and so phage KPP12 may be a good candidate for therapeutic. KPP12 showed a broad host range for P. aeruginosa strains isolated from clinical ophthalmic infections. Inoculation of the scarified cornea with P. aeruginosa caused severe keratitis and eventual corneal perforation. Subsequent single-dose administration of KPP12 eye-drops significantly improved disease outcome, and preserved the structural integrity and transparency of the infected cornea. KPP12 treatment resulted in the suppression of neutrophil infiltration and greatly enhanced bacterial clearance in the infected cornea. These results indicate that bacteriophage eye-drops may be a novel adjunctive or alternative therapeutic agent for the treatment of infectious keratitis secondary to antibiotic-resistant bacteria.  相似文献

目的:研究红楝子提取物对小鼠耐力、记忆能力的影响并寻找它们的最高活性部分。方法:采用转棒仪和避暗仪分别测试小鼠的耐力、记忆能力。研究红楝子的乙醇提取物、石油醚提取物、氯仿提取物和水提取物对小鼠的耐力、记忆能力的活性,找出其活性最高的部分。结果:1、红楝子粗提物对小鼠耐力有降低作用,在红楝子提取物的不同溶剂萃取部分中,石油醚萃取部分对小鼠耐力影响最大,应为其对降低小鼠耐力活性部位。2、红楝子氯仿提取物有明显的提高小鼠记忆能力的作用,为活性最高的部分。  相似文献

Breast cancer is one of the most invasive cancers with high mortality. The immune stimulating Propionibacterium acnes is a Gram positive bacterium that has the ability to cause inflammation and activate Th1-type cytokine immune response. Antitumor response was associated with the inflammation induced by P. acnes, but the antitumor effect of this bacterium was not evaluated in combination with other agents. The aim of this study was to test the antitumor potential of a combination of melatonin and P. acnes against breast cancer implanted in mice. Balb/C mice were transplanted with EMT6/P cell line and in vivo antitumor effect was assessed for P. acnes, melatonin, and a combination of melatonin and P. acnes. Tumor and organs sections were examined using hematoxylin/eosin staining protocol, and TUNEL colorimetric assay was used to detect apoptosis. The expression of vascular endothelial growth factor (VEGF) was measured in tumor sections and serum levels of INF-γ, and IL-4 were measured to evaluate the immune system function. To evaluate the toxicity of our combination, AST and ALT levels were measured in the serum of treated mice. The combination of melatonin and P. acnes has high efficiency in targeting breast cancer in mice. Forty percent of treated mice were completely cured using this combination and the combination inhibited metastasis of cancer cells to other organs. The combination therapy reduced angiogenesis, exhibited no toxicity, induced apoptosis, and stimulates strong Th1-type cytokine antitumor immune response. The combination of melatonin and P. acnes represents a promising option to treat breast cancer. However, carful preclinical and clinical evaluation is needed before considering this combination for human therapy.  相似文献

Genetic dissection of disease susceptibility in Arabidopsis to powdery and downy mildew has identified multiple susceptibility (S) genes whose impairment results in disease resistance. Although several of these S-genes have been cloned and characterized in more detail it is unknown to which degree their function in disease susceptibility is conserved among different plant species. Moreover, it is unclear whether impairment of such genes has potential in disease resistance breeding due to possible fitness costs associated with impaired alleles. Here we show that the Arabidopsis PMR4 and DMR1, genes encoding a callose synthase and homoserine kinase respectively, have functional orthologs in tomato with respect to their S-gene function. Silencing of both genes using RNAi resulted in resistance to the tomato powdery mildew fungus Oidium neolycopersici. Resistance to O. neolycopersici by SlDMR1 silencing was associated with severely reduced plant growth whereas SlPMR4 silencing was not. SlPMR4 is therefore a suitable candidate gene as target for mutagenesis to obtain alleles that can be deployed in disease resistance breeding of tomato.  相似文献

探讨游泳运动对APP/PS1转基因小鼠学习记忆能力的影响。选择11月龄的雄性APP/PS1转基因小鼠,随机平均分为对照组和游泳组,对照组常规饲养而游泳组进行1个月的游泳运动训练。分别采用刚果红染色、Tunel检测、Western Blot和Morris水迷宫等实验方法观察小鼠大脑皮层Aβ斑形成、神经元凋亡、线粒体生成相关蛋白表达和学习记忆能力的变化情况。结果发现,游泳运动可以减少APP/PS1转基因小鼠大脑皮层Aβ斑的形成、抑制神经元凋亡、促进线粒体生成、增强小鼠的学习记忆能力。由此可见,游泳运动可作为一项防治阿尔茨海默病的行为治疗候选方案。  相似文献

Streptococcus sanguinis is a cause of infective endocarditis and has been shown to require a manganese transporter called SsaB for virulence and O2 tolerance. Like certain other pathogens, S. sanguinis possesses aerobic class Ib (NrdEF) and anaerobic class III (NrdDG) ribonucleotide reductases (RNRs) that perform the essential function of reducing ribonucleotides to deoxyribonucleotides. The accompanying paper (Makhlynets, O., Boal, A. K., Rhodes, D. V., Kitten, T., Rosenzweig, A. C., and Stubbe, J. (2014) J. Biol. Chem. 289, 6259–6272) indicates that in the presence of O2, the S. sanguinis class Ib RNR self-assembles an essential diferric-tyrosyl radical (FeIII2-Y) in vitro, whereas assembly of a dimanganese-tyrosyl radical (MnIII2-Y) cofactor requires NrdI, and MnIII2-Y is more active than FeIII2-Y with the endogenous reducing system of NrdH and thioredoxin reductase (TrxR1). In this study, we have shown that deletion of either nrdHEKF or nrdI completely abolishes virulence in an animal model of endocarditis, whereas nrdD mutation has no effect. The nrdHEKF, nrdI, and trxR1 mutants fail to grow aerobically, whereas anaerobic growth requires nrdD. The nrdJ gene encoding an O2-independent adenosylcobalamin-cofactored RNR was introduced into the nrdHEKF, nrdI, and trxR1 mutants. Growth of the nrdHEKF and nrdI mutants in the presence of O2 was partially restored. The combined results suggest that MnIII2-Y-cofactored NrdF is required for growth under aerobic conditions and in animals. This could explain in part why manganese is necessary for virulence and O2 tolerance in many bacterial pathogens possessing a class Ib RNR and suggests NrdF and NrdI may serve as promising new antimicrobial targets.  相似文献

Lentinula edodes-derived polysaccharides possess many therapeutic characteristics, including anti-tumor and immuno-modulation. The gut microbes play a critical role in modulation of immune function. However, the impact of Lentinula edodes-derived polysaccharides on the gut microbes have not yet been explored. In this study, high-throughput pyrosequencing technique was employed to investigate the effects of a new heteropolysaccharide L2 from Lentinula edodes on microbiota diversity and composition of small intestine, cecum, colon and distal end of colon (feces) in mice. The results demonstrated that along mouse intestine the microbiota exhibit distinctly different space distribution. L2 treatment reduced the diversity and evenness of gut microbiota along the intestine, especially in the cecum and colon. In the fecal microbial communities, the decrease of Bacteroidetes by significantly increasing Proteobacteria were observed, which were characterized by the increased Helicobacteraceae and reduced S24-7 at family level. Some OTUs, corresponding to Bacteroides acidifaciens, Alistipes and Helicobacter suncus, were found to be significantly increased in L2 treated-mice. In particular, 4 phyla Chloroflexi, Gemmatimonadetes, Nitrospirae and Planctomycetes are exclusively present in L2-treated mice. This is helpful for further demonstrating healthy action mechanism of Lentinula edodes-derived polysaccharide L2.  相似文献
