The ability of certain theta-defensins, including retrocyclin-1, to protect human cells from infection by HIV-1 marks them as potentially useful molecules. Theta-defensins composed of L-amino acids are likely to be unstable in environments that contain host and microbial proteases. This study compared the properties of two enantiomeric theta-defensins, retrocyclin-1, and RC-112. Although these peptides have identical sequences, RC-112 is composed exclusively of D-amino acids, whereas retrocyclin-1 contains only L-amino acids. We compared the ability of these peptides to protect JC53-BL human cells from infection by 30 primary HIV-1 isolates. JC53-BL cells are modified HeLa cells that express surface CD4, CXCR4, and CCR5. They also contain reporter cassettes that are driven by the HIV-1 LTR, and express beta-galactosidase and luciferase. The HIV-1 isolates varied in co-receptor specificity and included subtypes A, B, C, D, CRF01-AE, and G. RC-112 was several fold more potent than retrocyclin-1 across the entire HIV-1 panel. Although RC-112 bound immobilized gp120 and CD4 with lower affinity than did retrocyclin-1, surface plasmon resonance experiments performed with 1 microg/mL of RC-112 and retrocyclin-1 revealed that both glycoproteins were bound to a similar extent. The superior antiviral performance of RC-112 most likely reflected its resistance to degradation by surface-associated or secreted proteases of the JC53-BL target cells. Theta-defensins composed exclusively of D-amino acids merit consideration as starting points for designing microbicides for topical application to the vagina or rectum.  相似文献

Although there appear to be no exceptions to the rule that proteins are composed solely of the L-isomers of amino acids, D-amino acids and derivatives of them do occur rather widely in living organisms. In some cases they have well-understood functions, but in other cases their occurrence raises interesting questions. Several peptide antibiotics contain D-amino acids (1). The peptido-glycans of Gram-positive bacterial cell walls contain D-glutamic acid, D-alanine, and D-asparagine (2). D-amino acids are also found in animals, chiefly annelids and insects (3). In this paper some aspects of D-amino acids in higher plants will be reviewed.  相似文献

A method for the quantitative determination of serum D-amino acids in the range 0.5-20 nmol is described. In the method alpha-keto acids, derived from D-amino acids by D-amino acid oxidase, are measured as hydrazones. The method is unresponsive to the presence of a large excess of L-amino acids. It allows a fast assay in a small amount of specimen (0.1 ml), with good reproducibility and accuracy.  相似文献

Here we report experimental evidence that the translation initiation apparatus accepts D-amino acids ((D)aa), as opposed to only L-methionine, as initiators. Nineteen (D)aa, as the stereoisomers to their natural L-amino acids, were charged onto initiator tRNA(fMet)(CAU) using flexizyme technology and tested for initiation in a reconstituted Escherichia coli translation system lacking methionine, i.e., the initiator was reprogrammed from methionine to (D)aa. Remarkably, all (D)aa could initiate translation while the efficiency of initiation depends upon the type of side chain. The peptide product initiated with (D)aa was generally in a nonformylated form, indicating that methionyl-tRNA formyltransferase poorly formylated the corresponding (D)aa-tRNA(fMet)(CAU). Although the inefficient formylation of (D)aa-tRNA(fMet)(CAU) resulted in modest expression of the corresponding peptide, preacetylation of (D)aa-tRNA(fMet)(CAU) dramatically increased expression level, implying that the formylation efficiency is one of the critical determinants of initiation efficiency with (D)aa. Our findings provide not only the experimental evidence that translation initiation tolerates (D)aa, but also a new means for the mRNA-directed synthesis of peptides capped with (D)aa or acyl-(D)aa at the N terminus.  相似文献

Determination of the D-amino acid content in foods and in biological samples is a very important task. In order to achieve this goal we developed a biosensor employing the flavoenzyme D-amino acid oxidase from the yeast Rhodotorula gracilis. To produce a device in which the D-amino acid composition does not alter the results, both the wild-type and a number of mutants obtained by rational design and directed evolution approaches were used. An analysis of D-amino acid oxidase mutants activity on D-amino acid mixtures containing various ratios of neutral, acidic, and basic substrates identified the Amberzyme-immobilized T60A/Q144R/K152E and M213G mutants as the best choice: their response shows an only limited dependence on the solution composition when at least 20% of the D-amino acid is made up of D-alanine (standard error is approximately 5-9%). This is the first report, to our knowledge, demonstrating that the entire D-amino acid content can be determined by using a screen-printed electrode amperometric biosensor, with a detection limit of 0.25 mM and a mean response time of 10-15 min. The D-amino acid assay based on R. gracilis DAAO-biosensor is inexpensive, simple to perform, and rapid: the D-amino acid concentration of a variety of biological samples can be investigated using this assay.  相似文献

Semi-metals (boron, silicon, arsenic and selenium) form organo-metal compounds, some of which are found in nature and affect the physiology of living organisms. They include, e.g., the boron-containing antibiotics aplasmomycin, borophycin, boromycin, and tartrolon or the silicon compounds present in "silicate" bacteria, relatives of the genus Bacillus, which release silicon from aluminosilicates through the secretion of organic acids. Arsenic is incorporated into arsenosugars and arsenobetaines by marine algae and invertebrates, and fungi and bacteria can produce volatile methylated arsenic compounds. Some prokaryotes can use arsenate as a terminal electron acceptor while others can utilize arsenite as an electron donor to generate energy. Selenium is incorporated into selenocysteine that is found in some proteins. Biomethylation of selenide produces methylselenide and dimethylselenide. Selenium analogues of amino acids, antitumor, antibacterial, antifungal, antiviral, anti-infective drugs are often used as analogues of important pharmacological sulfur compounds. Other metalloids, i.e. the rare and toxic tellurium and the radioactive short-lived astatine, have no biological significance.  相似文献

Our previous research has shown that many red algae metabolize polyunsaturated fatty acids to oxidized products resembling the eicosanoid hormones from mammals. We have extended these studies to members of the Phaeophyceae and Chlorophyta and find they also possess similar biosynthetic pathways. From several we have identified novel prostaglandin-like substances. Studies of the molecular mechanisms by which some of these marine oxylipins are formed have revealed that novel oxidative reactions are utilized. Understanding of these biosynthetic pathways in detail has allowed their utilization to produce research biochemicals of high value, such as 12S-hydroperoxyeicosatetraenoic acid (12S-HPETE). Because of their biological properties, seaweed-derived oxylipins have potential utility as pharmaceuticals and research biochemicals.  相似文献

Complete oxidation of some reportedly “unreactive” d-amino acids—aspartic acid, threonine, lysine—is possible by appropriate adjustments of enzyme concentration (units of activity) and reaction time, using a commercially available enzyme. Under the same conditions, less than 35% oxidation of d-glutamic acid is achieved. To avoid potential bacterial contamination and false negative as well as false positive results for certain compounds, long incubations should be minimized. Corroborative assays such as dual ninhydrin and radioactivity determinations are recommended.  相似文献
