The cartilaginous fish Chimaera monstrosa swims slowly by means of pectoral fin movements, and fast by undulations of the tail. In order to compare the fibres in the corresponding muscles, they were studied by histochemistry and electron microscopy. Three fibre types were identified by microphotometry and morphometry. Most of the axial muscles are white fibres, containing little mitochondria and glycogen. Red fibres, with glycogen and about 5 % mitochondria constitute a thin sheet in the axial muscles, composed of one fibre layer only. Pink fibres, with intermediate amounts of glycogen and mitochondria are situated between these two types, but are often not covered by red fibres. Pectoral muscles contain numerous red and intermediate fibres, partially mixed, superficially, and white fibres deeper. Pectoral muscle red fibres contain about 25 % mitochondria, half of which are situated in subsarcolemmal accummulations. The sarcotubular system has T-tubules at the Z discs, and the terminal cisternae are partially divided by regularly spaced clefts.  相似文献

Summary Sharks and skates (Chondrichthyes: Elasmobranchii) have a glial blood-brain barrier, while all other vertebrates examined so far have an endothelial barrier. For comparative reasons it is desirable to examine the blood-brain barrier in species from the other subclass of cartilaginous fish, the holocephalans. The ultrastructure of cerebral capillaries in the chimaera (Chondrichthyes: Holocephali) is described in the present study. The endothelial cells are remarkably thick. Fenestrae and transendothelial channels were not observed. The endothelial cells are joined by elaborate tight junctions. The perivascular glial processes are separated by wide spaces (15–60nm) without obliterating junctional complexes. These findings indicate that the chimaera has an endothelial blood-brain barrier.  相似文献

Summary The ultrastructure of the lateral-line neuromasts in the ratfish, Chimaera monstrosa is described. The neuromasts rest at the bottom of open grooves and consist of sensory, supporting, basal and mantle cells. Each sensory cell is equipped with sensory hairs consisting of a single kinocilium and several stereocilia. There are several types of sensory hair arrangement, and cells with a particular arrangement form patches within the neuromast. There are two types of afferent synapse. The most common afferent synapse has a presynaptic body and is typically associated with an extensive system of anastomosing tubules on the presynaptic side. When the tubules are absent, vesicles surround the presynaptic body. These synapses are often associated into synaptic fields, containing up to 35 synaptic sites. The second type of afferent synapse does not have a presynaptic body and is not associated with the tubular system. The afferent synapses of the second type do not form synaptic fields and are uncommon. The efferent synapses are either associated with a postsynaptic sac or more commonly with a strongly osmiophilic postsynaptic membrane. The accessory cells are similar to those in the acoustico-lateralis organs of other aquatic vertebrates. A possibility of movement of the presynaptic bodies and of involvement of the tubular system in the turnover of the transmitter is discussed. A comparison of the hair tuft types in the neuromasts of Ch. monstrosa with those in the labyrinth of the goldfish and of the frog is attempted.  相似文献

Summary The ultrastructure of the heart in Chimaera monstrosa L. is described. The endocardial and the epicardial cells are similar in the three cardiac regions. Myocardial cells show small variations.The myofibre, 4–6 m thick, contains one or a few myofibrils. Each myosin filament is surrounded by six actin filaments. The sarcomere banding pattern includes the Z-, A-, I-, M-, N-, and H-band. End-to-end attachments between myofibres are composed of alternating desmosomes and fasciae adhaerentes. Desmosomes and nexuses occur between longitudinally oriented cell surfaces. The sarcoplasmic reticulum is poorly developed but well defined. Peripheral coupling-like structures are common, T-tubules are absent. Membrane bound dense bodies occur in all regions. Areas with ribosomes and single myosin filaments are often seen.The epicardial cells have a regular hexagonal surface and are much thicker than the endocardial cells. Numerous short and a few longer cytoplasmic extensions face the pericardial cavity.The fiat endocardial cells contain a large nucleus and small amounts of cytoplasm.  相似文献

In the present ultrastructural study we have extended previous reports on the histological organization and cell components of the lymphohaemopoietic masses occurring in the cranium, mainly in the orbit, the preorbital canal, and the suprapalatal and coiacoid areas of the holocephalan Chimaera monstrosa. Mature and developing granulocytes, monocytes, macrophages, lymphocytes and plasma cells occur in a reticular and/or fibroblastic supporting stroma inside the cartilaginous skeleton. Heterophils, which are the most abundant granulocytes in the cranial tissue, contain two distinct cytoplasmic granular populations, whereas eosinophils show one uniformly electron dense granule type. Heterophils and eosinophils may differentiate from a common precursor producing granules of each cell type in relation to the activity of rough endoplasmic reticulum and Golgi complex. The presence of macrophages, lymphocytes and developing and mature plasma cells suggests an important role of the cranial lymphohaemopoietic tissue in eliciting the immune responses. A phylogenetical relationship between this tissue and the higher vertebrate bone marrow is proposed on the basis of histological similarities between the cell microenvironments governing haemopoietic differentiation in these organs.  相似文献

Comprehensive analyses have been made of the plasma and parietal muscle of the marine cyclostome Myxine glutinosa and of the chimaeraoid Chimaera monstrosa , and the sum of some 17 constituents compared with the directly determined osmotic concentration of plasma. Both Myxine and Chimaera plasmas are isosmotic with sea water within 2%. Ions account for at least 96% of the total concentration of the plasma in Myxine and about 60% in Chimaera, but the blood of the latter contains 332 mM urea, compared with 3 mM in Myxine . In muscle a similar divergence exists, the urea concentrations per kilogram muscle water being practically the same as in the respective plasmas of the two species. The mean values in millimoles of the other nitrogenous constituents of muscle are for Myxine and Chimaera (figures for latter in brackets), trimethylamine oxide 87 (189), betaine 65 (38), creatine and creatinine 30 (37), amino acids 291 (43). Using osmotic coefficients the calculated osmotic concentration of muscle exceeds that of plasma in Myxine and possible reasons for this are discussed.
Estimates of inulin space in Myxine muscle have enabled intracellular concentrations of ions to be calculated. Analysis of Myxine bile shows it to have much higher concentrations of sodium, calcium and magnesium, and much lower chloride, than plasma. The resulting large anion deficit is presumably made up chiefly by the bile salt myxinol disulphate.  相似文献

The presence of the opal chimaera, Chimaera opalescens, is reported for the first time in the deep waters of the Azores, with the capture of four specimens by fishermen and the video recording of an additional five individuals. Species identification was supported by the 646 bp sequenced fragment of the mitochondrial gene cytochrome oxidase subunit I. Because C. opalescens is a recently recognised species that had been recurrently misidentified as rabbitfish, Chimaera monstrosa, the historical data of C. monstrosa in the Azores were reviewed to assess the possible presence of both Chimaera species in the region. Although several authors have reported the occurrence of C. monstrosa in the Azorean waters since the 1800s, the majority of these are based on only three specimens caught during the late 1800s. The investigation performed using literature and examination of the museum specimens still available concluded that the most likely scenario is that C. monstrosa is absent from the Azores and past records of that species in the region are most likely misidentifications of C. opalescens.  相似文献

Recent studies have revealed the 3D morphology and collagen fiber architecture of myosepta in teleostome fishes. Here we present the first data set on the myoseptal structure of a representative of the chondrichthyan clade. We investigate the series of myosepta in the ratfish Chimaera monstrosa (Holocephali) from the anterior to the posterior body using microdissections of cleared and stained specimens, polarized light microscopy of excised myosepta, and histology. The features of the myoseptal system of Chimaera are compared to data from closely related vertebrate groups and are mapped onto a phylogenetic tree to further clarify the characteristics of the myoseptal series in the gnathostome ancestor. The 3D morphology and collagen fiber architecture of the myoseptal series in C. monstrosa resembles that of Teleostomi (Actinopterygii+Sarcopterygii) with regard to several features. Our comparative analysis reveals that some of them have evolved in the gnathostome stem lineage. (1) A series of epineural and epaxial lateral tendons (LTs) along the whole body, and a series of epipleural and hypaxial LTs in the postanal region evolved in the gnathostome stem lineage. (2) The LTs increase in length towards the posterior body (three-fold in Chimaera). Data on Chimaera and some comparative data on actinopterygian fishes indicate that LTs also increase in thickness towards the posterior body, but further data are necessary to test whether this holds true generally. (3) Another conspicuous apomorphic gnathostome feature is represented by multi-layer structures of myosepta. These are formed along the vertebral column by converging medial regions of successive sloping parts of myosepta. (4) The dorsalmost and ventralmost flanking parts of myosepta bear a set of mediolaterally oriented collagen fibers that are present in all gnathostomes but are lacking in outgroups. Preanal hypaxial myosepta are clearly different from epaxial myosepta and postanal hypaxial myosepta in terms of their collagen fiber architecture. In Chimaera, preanal hypaxial myosepta consist of an array of mediolaterally oriented collagen fibers closely resembling the condition in other gnathostome groups and in petromyzontids. Only one series of tendons, the myorhabdoid tendons of the flanking parts of myosepta, have evolved in the stem lineage of Myopterygii (Gnathostomata+Petromyzontida). Similar to LTs, the tendons of this series also increase in length towards the posterior body. In combination with other studies, the present study provides a framework for the design of morphologically based experiments and modeling to further address the function of myosepta and myoseptal tendons in gnathostomes.  相似文献

Extracts from preparations of partially purified Anaplasma marginale revealed low levels of lactate dehydrogenase (LDH). Enzyme inhibition by immune sera further indicated that A. marginale possesses a protein moiety the same as that of the normal red blood cell (RBC), although data suggested an alteration of LDH(1) from that observed in normal RBC. Bimodal isozyme distribution was detected after electrophoresis of the extracts. One isozyme approached the cathode and the other the anode, and both appeared to be nicotinamide adenine dinucleotide-dependent. Heterogeneity of parasite and host cell isozymes was established on the basis of zone electrophoresis on cellulose acetate strips.  相似文献

为了探讨藏羚羊(Pantholops hodgsonii)对低氧环境的适应机制。以生活在同海拔(4 300 m)的藏绵羊(Tibetan Sheep)为对照,用分光光度法测定2种动物心肌、骨骼肌中肌红蛋白(myoglobin,Mb)含量、乳酸(lactic acid,LD)含量及乳酸脱氢酶(lactate dehydrogenase,LDH)活力。结果显示,藏羚羊心肌和骨骼肌中Mb含量明显高于藏绵羊(P0.05),但心肌和骨骼肌的Mb含量无差别(P0.05),而藏绵羊心肌Mb含量明显高于骨骼肌(P0.05);藏羚羊心肌和骨骼肌中LD含量及LDH活力明显低于藏绵羊(P0.05),且2种动物心肌中的LDH活力均明显低于其骨骼肌(P0.01)。结果表明,藏羚羊尽管生活在高寒缺氧地区,其心肌和骨骼肌细胞仍然能得到丰富的氧供应,并非处于缺氧状态,这可能是通过增加心肌和骨骼肌中Mb的含量,提高其在低氧环境获取和储存氧的能力,从而提高有氧获能水平。与之相反,藏绵羊尽管也生活在高寒缺氧地区,但其心肌和骨骼肌中Mb含量相对于藏羚羊较低,且LD含量和LDH活力较高,说明其心肌和骨骼肌细胞内氧供不如藏羚羊丰富,提示藏绵羊可能主要以糖酵解获能。我们推测这种差异可能与两种动物不同的运动习性密切相关,且认为藏羚羊较高的Mb含量可能是其适应高原缺氧条件的分子基础之一。  相似文献

The tetrameric heart isozyme of lactate dehydrogenase (H4) is modified by p-chloromercuribenzoate (PCMB) to produce the inactive tetramer and then hybridized with native tetrameric muscle isozyme (M4). The hybrid mixture was isolated by polyacrylamide gel electrophoresis (PAGE) and then stained for enzyme activity and with Coomassie brilliant blue. Only three bands were found on the gels in either case. The hybrid enzymes as isolated by PAGE have half the specific activity of the native muscle enzyme. The electrophoresis properties of HM3 are very similar to those of HM3, while the electrophoresis properties of are very similar to those of H2M2. The above results strongly suggest that the tetramer having enzymatic activity contains at least two native subunits, and the di-subunit in the tetrameric enzyme is the minimal functional unit.  相似文献

Regulation of Staphylococcus aureus Lactate Dehydrogenase   总被引:8,自引:4,他引:4  
The effect of growth conditions on the specific activity of nicotinamide adenine dinucleotide (NAD)-linked lactate dehydrogenase (LDH) in extracts of Staphylococcus aureus strain SG 511A was examined. Kinetic and electrophoretic experiments, with extracts prepared from aerobically and anaerobically grown cells, provided evidence for only one physiologically significant enzyme. The aerobic level of NAD-linked LDH of S. aureus remained constant and was independent of the carbon source. In contrast, the level of LDH produced in an anaerobic environment was variable and was dependent on the carbon source. Growth anaerobically on pyruvate, as the sole fermentable carbon source, resulted in a maximal level of LDH activity, a value about eightfold greater than the aerobic level. Anaerobic growth either on pyruvate plus glucose or on glucose alone, however, resulted in approximately a threefold decrease in this maximum. Experiments with a heme-requiring auxotroph derived from S. aureus demonstrated that the aerobic level of LDH activity was dependent on a functional respiratory chain.  相似文献

In elasmobranchs (sharks, rays) the total weight of lymphomyeloid structures isolated by dissection varied between about 0.6 and 1.0% of the body weight. In a holocephalan (Chimaera) the corresponding value was 1.7%. Reciprocal size relations may exist between different types of lymphomyeloid organs. A thymus was found in adult specimens of rays (Raja) and the rabbit-fish (Chimaera). The lymphomyeloid organs supposedly are active in haemopoiesis and the immune responses.  相似文献

The feeding ecology of Chimaera monstrosa at a depth of 550?m was studied off S??ac?k Bay, Eastern Mediterranean, in 2008. Forty males and 57 females with a length between 7.80 and 45.5?cm were caught. 33 stomachs contained food items. The diet was composed of 11 prey items belonging to 6 groups: Bryozoa, Cnidaria, Crustacea, Mollusca, Tunicata, and Teleostei, of which Crustacea (IRI% [Index of Relative Importance]=80) and Mollusca (IR%=16) were the most abundant prey groups. The trophic level was estimated at 3.50, which is similar to the results from other studies in the western and northern part of the Mediterranean and the Atlantic Ocean but is higher than in the North Sea.  相似文献
