亮氨酸拉链肿瘤抑制因子2(leucine zipper tumor suppressor 2,Lzts2)是一种新发现的抑癌基因,位于人类染色体10q24.3,在多种肿瘤中异常表达,且对肿瘤的发生、增殖和迁移中均发挥重要作用。LZTS2拥有亮氨酸拉链结构域(LZ),可以通过该结构域结合DNA调节基因转录。LZTS2还可以参与Wnt/β-catenin信号通路调控β-catenin的表达及细胞内分布,与NF-κB亦拥有广泛的相互作用,从而调控细胞增殖和凋亡。在微管系统中,LZTS2也有重要的调节作用,可抑制细胞的有丝分裂和迁移,增加肿瘤细胞的药物敏感性。LZTS2作为新的抑癌基因,与肿瘤的大小和淋巴结转移等生物学特性密切相关,可能成为新的诊断标志物,判断肿瘤的预后,亦可能为肿瘤的基因治疗潜在方向。然而关于LZTS2及其作用机制的研究尚为不足,本文将对LZTS2在肿瘤中的作用机制作简要综述。  相似文献

Tumor microenvironment (TME) is a host for a complex network of heterogeneous stromal cells with overlapping or opposing functions depending on the dominant signals within this milieu. Reciprocal paracrine interactions between cancer cells with cells within the tumor stroma often reshape the TME in favor of the promotion of tumor. These complex interactions require more sophisticated approaches for cancer therapy, and, therefore, advancing knowledge about dominant drivers of cancer within the TME is critical for designing therapeutic schemes. This review will provide knowledge about TME architecture, multiple signaling, and cross communications between cells within this milieu, and its targeting for immunotherapy of cancer.  相似文献

With rare exceptions, spontaneous tumors originate from a single cell. Yet, at the time of clinical diagnosis, the majority of human tumors display startling heterogeneity in many morphological and physiological features, such as expression of cell surface receptors, proliferative and angiogenic potential. To a substantial extent, this heterogeneity might be attributed to morphological and epigenetic plasticity, but there is also strong evidence for the co-existence of genetically divergent tumor cell clones within tumors. In this perspective, we summarize the sources of intra-tumor phenotypic heterogeneity with emphasis on genetic heterogeneity. We review experimental evidence for the existence of both intra-tumor clonal heterogeneity as well as frequent evolutionary divergence between primary tumors and metastatic outgrowths. Furthermore, we discuss potential biological and clinical implications of intra-tumor clonal heterogeneity.  相似文献

To test if proteolysis is involved in tumor cell extravasation, we developed an in vitro model where tumor cells cross an endothelial monolayer cultured on a basement membrane. Using this model we classified the ability of the cells to transmigrate through the endothelial cell barrier onto the underlying matrix, and scored this invasion according to the stage of passage through the endothelium. Metalloproteinase inhibitors reduced tumor cell extravasation by at least 35%. Visualization of protease and cell adhesion molecules by confocal microscopy demonstrated the cell surface localization of MMP-2, MMP-9, MT1-MMP, furin, CD44 and αvβ3, during the process of transendothelial migration. By the addition of inhibitors and bio-modulators we assessed the functional requirement of the aforementioned molecules for efficient migration. Proteolytic digestion occurred at the cell-matrix interface and was most evident during the migratory stage. All of the inhibitors and biomodulators affected the transition of the tumor cells into the migratory stage, highlighting the most prevalent use of proteolysis at this particular step of tumor cell extravasation. These data suggest that a proteolytic interface operates at the tumor cell surface within the tumor-endothelial cell microenvironment.  相似文献

 Cell-based tumor vaccines have been developed on the basis of the hypothesis that tumor cells can be genetically modified to present antigen to T lymphocytes directly. Contrary to expectations, cross-priming is the predominant pathway for activation of tumor-specific CD8+ T cells, while direct presentation of antigen dominates activation of tumor-specific CD4+ T cells. These results pose interesting paradoxes for the generation of immune responses, and have definite implications for the development of anti-cancer vaccines.  Increasing the ability of tumor-reactive T cells to mediate tumor regression in vivo has been a major goal of tumor immunologists. Progress toward this goal has been aided by the identification of tumor-associated antigens on both experimental mouse tumors and human tumors. However, the self-like nature and low immunogenicity of these antigens has made it clear that other measures to enhance the effectiveness of the T cells reactive to these antigens are essential if immunotherapy is to be clinically effective. An increased understanding of antigen processing and presentation is an important step in this process, as is the use of cytokines to increase immune responsiveness. Despite recent advances, there is still much to be learned before the specificity of the immune system is safely harnessed to halt malignant cell growth effectively. Three models of tumor cell loss are described. the effects of cell loss on other cellular kinetic parameters are evaluated, and experiments which may distinguish among the models are discussed. Each model is based on a different cell-loss mechanism, and equations for the cell-cycle, cell-frequency distribution, the growth of both the proliferating and non-proliferating cell population, the growth fraction (GF), and the relative rate of volumetric growth, (dV/dt)/V, are derived. The following types of data are simulated for each model: the pulse labelling index, the mitotic index, and the labeling index as a function of time after a single or a series of 3H-TdR injections. the relative volumetric growth rate has the same mathematical form for each model. the PLM curves predicted by each model for the tumor lines studied (S102F and Slow) are not appreciably different. the predicted initial labeling index and mitotic index may differ significantly among the models depending upon the tumor line. the most striking difference among the models lies in the predictions regarding the labeling index as a function of time after a single or after a series of 3H-TdR injections. These types of labeling experiments should be valuable for distinguishing the different cell-loss mechanisms in solid tumors.  相似文献

干细胞是一种具有自我更新、无限增殖和多向分化能力的细胞.而多数肿瘤是由不同增殖潜能的不均一性细胞构成.随着对干细胞的研究不断深入,使人们对肿瘤的发生机制重新进行了审视,并在造血系统、脑、肺、乳腺等部位肿瘤中发现极少量的具有与干细胞非常类似生物学特性的细胞,称之为肿瘤干细胞,它们很可能是肿瘤细胞的起源.肿瘤干细胞的提出.使得靶向性杀伤肿瘤干细胞从而使根治肿瘤和防止肿瘤复发和转移成为可能.所以研究肿瘤干细胞的起源及其与肿瘤的发生关系,成为当前研究和治疗肿瘤领域的新热点.本文就肿瘤干细胞的存在证据、干细胞与肿瘤干细胞的异同点及它们与肿瘤发生之间的关系作简要的综述.  相似文献
