Over a three year period 8 cows and 2 heifers were administered testosterone via injections and/or silastic implants. During and subsequent to treatments, blood samples were collected for determination of testosterone and cows were observed for male sex behavior. Male sex behavior was induced by administering 200 mg of testosterone propionate every other day for 20 days, and once induced, male sex behavior was maintained by injections of 200 mg of testosterone propionate every 10 days or by two 15 cm silastic implants containing testosterone propionate. Male sex behavior was also induced and maintained by administering an injection of testosterone enanthate (1 gm) and testosterone propionate (200 mg) and two 15 cm silastic implants containing testosterone propionate at the same time. Cows which are administered testosterone in this manner detected 94% of 231 females that were in estrus 342 times.  相似文献

Two gilts were administered testosterone propionate and their subsequent plasma testosterone concentrations and male sex behavior were recorded. These were compared to testosterone concentrations and male sex behavior in boars. Testosterone propionate (75 mg) was administered to the gilts every other day for 20 days (induction scheme) and every 10 days there-after (maintenance scheme). Concentrations of testosterone in plasma were elevated to concentrations detected in the boars during the induction scheme. During the maintenance scheme, concentrations of testosterone appeared to be lower than in boars. At 20, 30 and 40 days following the first injection, sniffing, nosing and mating song behaviors were exhibited by the testosterone treated gilts similar in frequency to the boars. Mounting behavior was first detected 30 days following the first testosterone propionate injection, and by day 40, the frequency of mounting was greater than observed in boars.  相似文献

In the first of two experiments, ovariectomized ewes with silastic implants containing testosterone propionate had higher concentrations of plasma testosterone than ovariectomized ewes with silastic implants containing testosterone (4.0 +/- .2 vs .8 +/- .1 ng/ml). In the second experiment, one or two 10 cm silastic implants filled with testoster-one propionate increased concentrations of plasma testosterone and induced male sex behavior in ovariectomized ewes similar to plasma testosterone levels and male sex behavior in rams. The interval from initiation of the treatment to the first behavior test in which mounting was consistently observed was 14.4 +/- 2.9 days and once mounting behavior was consistently induced, the testosterone treated ewes mounted estrous ewes an average of 10.7 +/- 1.3 times during a 10-minute test.  相似文献

Two pony mares were administered 150 mg of testosterone propionate every other day for 20 days (ten injections) and every ten days there-after. An additional two mares and one stallion were not treated and served as controls. Testosterone propionate was dissolved in absolute ethanol and administered subcutaneously. Sex behavior tests were conducted 26 and 40 days after the first injection. Control mares exhibited very little male sex behavior. Both testosterone propionatetreated mares, however, exhibited mounting, sniffing, flehmen, biting and vocalization behavior in the presence of an estrous mare. The testosterone propionate-treated mares mounted and bit estrous mares more frequently than the stallion but exhibited less sniffing, flehmen and vocalization behavior in the presence of an estrous mare than the stallion. Testosterone propionate-treated mares and the stallion mounted an estrous mare 23.3 +/- 9.7 seconds and 172.5 +/- 22.5 seconds, respectively, after being introduced into the pen. Mares in estrus were mounted by the testosterone propionate-treated mares and the stallion an average of 4.0 +/- 1.3 and 1.0 +/- 0 times, respectively, during a ten-minute test. None of the non-estrous mares was ever mounted by the testosterone propionate-treated mares. In summary, testosterone propionate induced male sex behavior in intact mares and the testosterone propionate-treated mares effectively detected estrous mares.  相似文献

An experiment was conducted to evaluate the effectiveness of wethers and ewes treated with testosterone preparations to induce ovulation and breeding activity in anestrous ewes. The testosterone was administered three times at weekly intervals. Wethers and ewes treated with 105 mg testosterone propionate and wethers treated with 100 mg testosterone from testosterone cyclopentyl propionate were as successful as vasectomised rams in inducing ovulation and cyclic activity in ewes. Seven days after the last injection the concentrations of testosterone in peripheral bloods were not significantly different from that in the vasectomised rams. By day 28 the concentrations were significantly lower than in the rams. The testosterone preparations tested are suitable for the induction of male sexual behavior and are rapidly excreted by sheep.  相似文献

Six ovariectomized mares were divided into 3 groups to determine the effects of exogenous progesterone in oil and repositol progesterone on plasma progesterone concentrations. Progesterone in oil was administered in 7 daily injections in Exp. I. Progesterone concentrations were not maintained greater than 1.0 ng/ml for 24 h with 50 mg/day. However, they remained greater than 1.0 ng/ml during the last 4 days of 100 mg/day and greater than 1.5 ng/ml throughout the injection sequence of 200 mg/day. Repositol progesterone was administered on Days 1 and 7 in Exp. II. At 500 mg, progesterone concentrations peaked in 6 h but returned to near 1.0 ng/ml in 2 days. At 1000 mg and 2000 mg, plasma progesterone was maintained at approximately 2.0 and 4.0 ng/ml respectively for 7 days after injection on Day 1 and was 1.5 and 3.5 ng/ml respectively, 11 days after injection on Day 7. An indication of a cumulative effect on plasma progesterone was observed following repeated dosages of both progesterone in oil and repositol progesterone.  相似文献

The therapeutic effectiveness of intramuscularly administered testosterone esters and free testosterone in suppositories was investigated by the measurement of plasma testosterone and LH levels after administration to normal and hypogonadal men. Testosterone levels were elevated above the lower physiological limit for 1 day after 25 mg testosterone one propionate, for 2 days after 50 mg testosterone propionate and for 14 days after 250 mg testosterone oenanthate. LH levels were suppressed for the corresponding periods. Elevated plasma testosterone and suppressed LH levels were maintained by testosterone suppositories (3 x 20 mg for 5 days).  相似文献

Two studies were conducted to determine the passage of testosterone, testosterone propionate and testosterone enanthate through silastic implants and to determine the retention of the three hormones once they enter the blood. In the first experimental, ovariectomized ewes with implants containing testosterone propionate and ewes with implants containing testosterone enanthate had higher levels of plasma testosterone than ewes with implants containing testosterone. Testosterone enanthate implants released more hormone during the 13-day period than the testosterone propionate and testosterone implants and testosterone propionate implants released more hormone than testosterone implants. In the second experiment, concentrations of plasma testosterone were elevated longer for ovariectomized ewes intravenously administered testosterone propionate, than ewes receiving testosterone or testosterone enanthate intravenously.  相似文献

Estrous behavior in response to ambient and long-day photoperiods was evaluated in ewes developed by 10 years of selection for ability to lamb in autumn. Following October lambing, 67 ewes were moved indoors and exposed to long-day (16L:8D) or ambient photoperiods from February 2 until July 6. Two vasectomized rams with marking harnesses were housed with each group. Estrous behavior was monitored twice weekly. Ewes from the selection line were unresponsive to long days, with no effects on estrous behavior, frequency of ovulation, or circulating prolactin. Adult ewes were anestrus for only 34±3 d, but 2- and 3-years-old ewes were anestrus for 72±7 and 57±10 d, respectively. Frequencies of ovulation based on circulating progesterone concentrations in March, May, and June were 97%, 95% and 52%, respectively, indicating that many ewes that did not exhibit estrus still ovulated. Prolactin concentrations increased from 10 ng/ml in February to 27 ng/ml in March and 173 ng/ml in June but were not affected by light treatment. Ten ewes that failed to exhibit estrus behavior for at most 24 d during the main study were then monitored for 74 additional long days. Nine of 10 ewes did not exhibit estrus for periods similar to 1 or 2 estrus cycles during this period, but eight ewes re-initiated cycles by the end of the study on September 18. Selection for ability to lamb in autumn thus resulted in ewes with an abbreviated seasonal anestrus and reduced sensitivity to long days.  相似文献

Three experiments were conducted on Texel ewes to study the influence of prostaglandin F(2alpha) (PGF(2alpha)), prolactin (PRL), estradiol (E(2)), and gonadotrophin releasing hormone (GnRH) on postpartum reproductive activity. In Experiment 1, oral administration of indomethacin (25 to 50 mg/day/ewe) from Day 3 post partum to the first detected estrus inhibited plasma 13, 14-dihydro-15-keto, PGF(2alpha) (PGFM) concentrations (P < 0.0001). This treatment resulted in an earlier rise in the frequency and amplitude of luteinizing hormone (LH) pulses and a resumption of estrous behavior (P < 0.05), while ovarian activity estimated by progesterone (P(4)) concentrations resumed to the same extent in treated ewes and controls. Bromocriptine treatment (2.5 mg/day/ewe) reduced plasma PRL levels (P < 0.0001) but had no effect on ovarian activity as evidenced by P(4) and resumption of estrus or on either the frequency or amplitude of the LH pulse. In Experiment 2, a single injection of GnRH agonist (42 mcg of buserelin/ewe) on Day 16 post partum resulted in an abrupt elevation of plasma LH concentrations; mean LH values were 18 to 27 times higher when compared with those of the control ewes. Two days after this treatment, ovulations occurred in 5 of the treated ewes and in 2 of the control ewes. This induced ovarian activity was not associated with estrous behavior; however, after an adequate subsequent luteal phase all the treated ewes displayed estrus, the resumption of estrus thus being earlier in treated than in control ewes (P < 0.01). In Experiment 3, E(2) supplementation from Day 16 to Day 28 post partum increased the number of LH pulses per 6 hours in suckling ewes (P < 0.05) and induced earlier resumption of estrus in dry ewes but not in suckling ewes (P < 0.01). Luteal function was detected about 5 and 8 days after the insertion of E(2) implants in 4 dry ewes and in 2 suckling ewes, respectively.  相似文献

Adult hypophysectomized rats were maintained on different regimens of testosterone propionate (TP) treatment for 27 days (0.2, 0.4, 0.6 and 1 mg/day) and autopsied 16 hours after the last injection. Blood samples were taken, sex organs were weighed and one testis from each animal was fixed in Bouins fluid for histologic analysis. The other testis and blood were used for testosterone (T) determinations. Both testicular and plasma T were below detectable levels in hypophysectomized control rats. The plasma T level showed a dose response relationship with increasing dose of TP but such was not the case for intratesticular T concentrations. Qualitative and quantitative evaluation of testis sections showed that spermatogenesis was incomplete in rats receiving 0.2 mg TP/day characterized by the absence of step 15 to 19 spermatids, degeneration of some pachytene spermatocytes and a significantly lower yield of B type spermatogonia. Analysis of testis sections from animals treated with 0.4 to 1 mg TP/day showed complete maintenance and maturation of pachytene spermatocytes, meiosis and spermiogenesis. However, even with the highest dose of TP (1 mg/day) the total yield of B type spermatogonia was only about 58% of the intact controls. It is concluded that at least 0.4 mg/day of exogenous TP is essential for qualitative maintenance of spermatogenesis in hypophysectomized rats with an intratesticular T concentration of 17 to 18 ng/gm testis.  相似文献

The induction of estrus in 17 previously cycling nulliparous ewes, 9 to 10 months of age, was attempted with Medroxyprogesterone acetate (MAP) pessaries during the early anestrous period (March-April). Ewes were verified to be anestrous by the lack of estrous behavior in the presence of a vasectomized ram and by a radioimmunoassay for serum progesterone in two samples taken 7 days apart showing less than 1 ng/ml serum progesterone. Superovulation was attempted with injections of either FSH or FSH + LH. MAP vaginal pessaries remained in place for a period of 12 days and FSH was administered to all ewes (IM) at 12 hr intervals over a 3 day period; 5 mg was injected twice on day 11 after pessary insertion, followed by 4 and 3 mg injections twice daily on each succeeding day, for a total of 24 mg per ewe. Nine ewes were given 25 mg LH (IV) within 8 hrs after the onset of behavioral estrus in addition to FSH. Ewes were hand-mated to several rams at 12 hr intervals throughout the estrus period. Ovulation and fertilization rates were recorded for each ewe following midline laparotomy and embryo collection. All ewes were in estrus between 36 and 48 hrs after removal of the MAP pessaries. In ewes injected with FSH only, 8 of 8 ovulated with a mean ovulation rate of 6.0 +/- 4.4 and a fertilization rate of 70%. Nine of 9 ewes receiving both FSH + LH ovulated with a mean ovulation rate of 13.9 +/- 13.1 and a fertilization rate of 72%. Statistical analysis by Students t-test resulted in differences in number of ova recovered (P<.05) between FSH only and FSH + LH treated ewes and a trend towards increased ovulation rate in FSH + LH treated ewes. These results show that early seasonally anestrous ewes can be successfully induced and synchronized for estrus with MAP pessaries and the number of ova recovered is increased with the inclusion of LH in the superovulation regime.  相似文献

This study examines the effect of treatment with controlled-release poly(DL-lactide-coglycolide) microsphere formulations of the LH-RH agonist [D-Trp6, des-Gly-NH10(2)]-LH-RH ethylamide (LH-RH-A) designed to release about 100 or 200 micrograms of the peptide per day for 3, 5 or 6 months in male dogs. Plasma levels of testosterone and LH-RH-A were measured at 2-day intervals. After the first injection of the 100-micrograms/day formulation, plasma testosterone increased from 1.6 +/- 0.2 to 3.5 +/- 0.6 ng/ml for 5-7 days before decreasing and remaining at 0.05 +/- 0.008 ng/ml for approximately 150 days (5 months). After two months of recovery, microspheres designed to release 100 micrograms for 6 months of LH-RH agonist per day were then injected. Plasma testosterone levels showed an elevation from 1.5 +/- 0.5 to 4.7 +/- 2.0 ng/ml during the first few days before gradually decreasing to castration levels for 200 days (6 months). One month later, plasma testosterone had returned to normal levels. When microspheres designed to deliver an average of 200 micrograms per day of the peptide for 3 months were injected in another series of animals, castration levels of plasma testosterone were maintained for 95 days with a progressive increase to normal values at later time intervals. The animals of the first series of experiments were then sacrificed after 4 months of recovery following maintenance of plasma testosterone at castration levels for a total period of 11 months. The testes, prostate and pituitary gland were kept for histological examination which was completely normal in all tissues. The efficacy and excellent tolerance of the controlled-release form of LH-RH-A as inhibitor of the pituitary-gonadal axis strongly support the use of such long-term controlled-release formulations of LH-RH agonists for the treatment of sex steroid sensitive diseases.  相似文献

Naturally cycling white faced ewes were utilized to study the effects of continuously elevated environmental temperature and/or humidity on plasma concentrations of luteinizing hormone (LH), prolactin (PRL), progesterone (P4) and testosterone (TE) during the estrous cycle. Fourteen ewes were randomly allocated on the day of estrus (day 0) to either thermoneutral conditions (21.1 degrees C, 65% relative humidity) or elevated ambient temperature/humidity conditions (36.1 degrees C, 71% relative humidity) producing an average 1.4 degrees C hyperthermia. Animals remained in their respective environments and blood samples were collected daily until the next estrus or day 20, whichever occurred first. Starting at noon on day 14, blood was sampled every 2 hours. Concentrations of LH, PRL, P4 and TE were quantified using validated radioimmunoassays. Hyperthermic ewes exhibited 1) a significant decrease (P<0.05) in the incidence of behavioral estrus and a preovulatory LH surge at the expected time of the estrous cycle, 2) significantly lower (P<0.05) plasma P4 between days 7 and 13 of the cycle, 3) a six-fold increase of PRL levels (P<0.01). Plasma levels of TE were not significantly affected by hyperthermia. The only two experimental ewes which exhibited estrus and an LH surge also showed an unusual and significant peak in plasma P4 two days before estrus. These results confirm that elevated environmental temperatures that result in hyperthermia can induce endocrine imbalances in the ewe which may contribute to decreased reproductive efficiency in the heat-stressed female.  相似文献

To determine the threshold doses of testosterone propionate (TP) that cause clear-cut behavioral changes in the sexual behavior of castrated male cynomolgus monkeys, observations were made on three males during successive 5-week treatment periods while they received daily subcutaneous doses of 100 μg TP increasing in octaves to 25.6 mg TP. Males were tested with each of the same two ovariectomized, estrogen-treated females (6 pairs, 330 1-hr behavior tests). To mimic the diurnal plasma testosterone rhythm, TP injections were given at 1600 hr and blood samples were obtained at 0800 hr (141 samples). Male ejaculatory activity increased at the threshold dose of 200 μg TP per day giving plasma testosterone levels of 830 ng/100 ml, which is in the physiological range of 600–1600 ng/100 ml for intact males. This threshold dose was eight times higher than in rhesus monkeys on a dose per kilogram body weight basis. There was a further marked increase in ejaculatory performance at higher doses (6.4 to 25.6 mg) giving supraphysiological plasma levels of 4000–9000 ng/100 ml. There were individual differences in the behavioral changes occurring with TP treatment, and the female partner modulated the effects. These findings were generally similar to those obtained with male rhesus monkeys, but certain species differences were noted.  相似文献

At calving forty-eight Holstein and Guernsey cows were assigned according to age and breed to one of six postpartum periods (1 or 2, 3 or 4, 5 or 6, 7 or 8, 12 or 13 and 18 or 19 days postpartum). Thirty-six of the cows (6 cows per postpartum period) received a single intramuscular injection of 100 μg GnRH. The other twelve cows (2 cows per postpartum period) served as controls and received a single intramuscular injection of the carrier vehicle for GnRH.Four of 36 cows administered GnRH and three of the 12 control cows ovulated by the day following treatment. Four of the cows were 12 or 13 days postpartum (1 control and 3 GnRH treated) and three were 18 or 19 days postpartum (2 controls and 1 GnRH treated). Six of the seven cows that ovulated the day following treatment had a follicle > 1.0 cm the day prior to treatment. Follicular growth was detected in the earlier postpartum periods but ovulation the following day was not detected for either control or GnRH treated cows. Following estrus or silent estrus, plasma progesterone concentrations increased to about 4 ng/ml on day 13. However, in cows ovulating the day following GnRH treatment, plasma progesterone declined from about 3 ng/ml on day 9 to approximately 1 ng/ml on day 13 postestrus. In addition, LH in plasma was higher (P < .01) ? through 13 days following estrus or silent estrus in cows ovulating the day after GnRH treatment in comparison to cows during the first or subsequent postpartum estrous cycles.In summary, in addition to days postpartum other factors including follicular development and maturity are probably involved in GnRH induced ovulation.  相似文献

The objective of this study was to determine whether supplemental beta-carotene would influence reproductive function in mares maintained on spring and summer pastures and to characterize plasma carotene concentrations during the estrous cycle. Carotene concentrations in plasma did not vary with day of estrous cycle (P = 0.7455). Mares receiving every other day injections of beta-carotene (400 mg; n = 4) or saline (10 ml; n = 4) during proestrus/estrus did not differ in plasma estradiol (E(2)) concentrations (P = 0.6313), follicle development (P = 0.8068), or plasma progesterone (P(4)) concentrations during the following diestrus (P = 0.4954). Moreover, no differences in plasma P(4) concentrations (P = 0.9047) were detected between mares receiving every other day injections of beta-carotene (400 mg; n = 4) or saline (10 ml; n = 4) during diestrus. However, administration of beta-carotene raised plasma carotene concentrations relative to controls when injected during proestrus/estrus (P = 0.0096) and diestrus (P = 0.0099). Pregnancy rates (P = 0.4900) and number of cycles required for pregnancy (P = 0.2880) were similar for mares administered injections of saline (10 ml; n = 37), beta-carotene (400 mg; n = 37), vitamin A (160,000 IU; n = 38), or vitamin A + beta-carotene (160,000 IU + 400 mg; n = 43), on the first or second day of estrus and on the day of breeding. Therefore, these results collectively suggest that supplemental beta-carotene does not affect the reproductive function of mares fed adequate dietary carotene. Whether supplemental beta-carotene would enhance reproductive function in mares on low carotene diets warrants further investigation.  相似文献

The objective of our study was to determine the effect of chronic utero-ovarian vein catheterization in ewes on estrous cycle length, plasma progesterone (P) concentration, and myometrial electromyographic activity. Cyclic ewes with inferior vena cava catheters were used as controls. Estrus was synchronized in ten ewes and 10 to 12 d following estrus, the ewes were anesthetized, fitted with myometrial electromyograph leads and with utero-ovarian vein (n = 5) or inferior vena cava (n = 5) catheters. After surgery, ewes returned to estrus as expected (16 to 18 d interestrus interval). The second cycle of four of five ewes with utero-ovarian vein catheters were prolonged (40 to 58 d). The inferior vena cava catheterized ewes had normal length second cycles. Plasma P concentrations reflected the estrous cycles: low ( 0.05).  相似文献

Changes in LH, FSH, and testosterone concentrations, testicular firmness and resilience, and scrotal circumference were monitored in 16 Corriedale rams (8 experienced adult and 8 inexperienced young) for 20 days during which they were used to stimulate ewes. The experiment was conducted during November (mid-non-breeding season). Increases (P<0.05) were observed in LH and testosterone concentrations and in testicular firmness and resilience during the first 4 days when rams were in permanent contact with estrual ewes. During the following 13 days, when rams were in contact with non-estrual ewes (i.e. initially estrual ewes were no longer in estrus), LH and testosterone concentrations decreased. When initially anestrous ewes exhibited estrus 17 to 20 days later, concentrations of testosterone increased. Testicular firmness and resilience remained high throughout the period. We conclude that: (1) rams used to stimulate anestrous ewes show an increase in LH and testosterone concentrations beginning at 12 h after joining, and greater concentrations are maintained while estrual ewes and mating are allowed; and (2) estrous and mating activity are probably the most important stimuli for the increase in hormone concentrations.  相似文献

The objectives of this study were (1) to establish the characteristics of estrus behavior in mithun cows (n = 12) and (2) to determine the relationships between this behavior and the plasma concentrations of estradiol-17beta (E2), total estrogen, and progesterone. Estrus was detected by visual observations of estrus signs, per recta examination of genitalia and bull parading thrice a day for three consecutive cycles. Among the behavioral signs of estrus, the cow to be mounted by bull (100%) was the best indicator of estrus followed by standing to be mounted (92%). Per rectum examination of genital organs revealed relaxed and open os externa of cervix, turgid uterus, and ovaries having palpable follicles in all animals. The mean (+/-SEM) length of estrus cycle and duration of estrus were recorded to be 21.8 +/- 0.69 days and 12.6 +/- 1.34 h, respectively. Endocrine profiles during the peri-estrus period showed that the mean highest peak concentrations of E2 (27.29 +/- 0.79 pg/ml) and total estrogen (45.69 +/- 2.32 pg/ml) occurred at -3.90 +/- 2.27 and -3.89 +/- 2.26 h prior to the onset of estrus, respectively. Plasma progesterone concentration was basal (0.14 +/- 0.001 ng/ml) during the peri-estrus period. Plasma E2 and total estrogen were found to increase from 6 days before estrus to reach a peak level on the day of estrus and decline thereafter to basal level on day 3 of the cycle. The plasma progesterone concentration was the lowest on the day of estrus showing gradual increase to register a peak level on day 15 of the cycle. Estrus behavior was found to be positively correlated with the maximum peak concentration of E2 (r = 0.89; P < 0.0001) and total estrogen (r = 0.66; P = 0.019) during the peri-estrus period. The mean total estrogen concentration during the peri-estrus period was significantly correlated with estrus behavior (r = 0.60; P = 0.04). The correlations between the estrus behavior and E2:progesterone ratios at 6 days before the onset of estrus (r = 0.92) and on the day of estrus (r = 0.95) was significant. The total estrogen:progesterone ratios at 6 days before the onset of estrus and on the day of estrus were also positively correlated with the estrus behavior (r = 0.86 and 0.88). In conclusion, our results suggest that the maximum peak concentration of E2 and total estrogen and mean level of total estrogen during the peri-estrus period and the E2:progesterone and total estrogen:progesterone ratios on 6 days before the onset of estrus and on the day of estrus are the important factors contributing the behavioral manifestation of estrus in mithun cows.  相似文献
