Kjellmaniella crassifolia, the edible macro-brown alga in Japan contained nearly 27% of alginates of which nearly 7% was extractable from the fronds with boiling water for 6 h and the residual alginates in the frond were almost exhaustively extracted with a dilute alkali at 60 °C for 6 h. The alginates dissolved in all these extracts with both boiling water and dilute alkali were purified by fractionation with MgCl2 and alcohol.The content of MM blocks in the boiling water-soluble alginate sample increased remarkably during heating for 6 h while that of GG blocks from the same sample decreased. In contrast, MM blocks in the alkali-soluble alginate sample decreased during 6 h heating while GG blocks continued to increase. Since the amounts of MG blocks showed slight fluctation, the M:G ratio of alginates extracted with boiling water increased towards the end of extraction whereas the reverse is true for the alkali-soluble alginates.  相似文献

Biocompatibility of biomaterials and biomaterial-based medical devices is a critical issue for the long-term function on multiple therapeutic systems. In the past few years, there has been an increasing interest in producing more biocompatible biomaterials and in developing novel assays to analyze the quality of the products. In this study, a battery of in vitro techniques to assess the biocompatibility of alginates with different compositions and purities and alginate-based microcapsules is presented. Study of the protein and polyphenol content of the alginates revealed clear differences between the nonpurified and the purified alginates. A similar behavior was observed when the mitogenic activity and the tumor necrosis factor-alphasecretion induced by the alginates were assessed. Interestingly, when the latter two techniques were adapted to evaluate the different alginate microcapsules, a correlation with the results obtained for the alginate samples was observed. These results reinforce the idea of using the full battery of assays here reported to screen alginates and alginate-based microcapsules before implantation.  相似文献

Alginic acid and metal alginates are prepared from fresh algae using extraction method. FTIR spectra indicate that alginic acid is converted into metal alginate. Asymmetric stretching of free carboxyl group of zinc alginate at 1596 cm−1 is shifted to 1582 cm−1 in cadmium alginate, due to the change of charge density, radius and atomic weight of the cation. Surface morphology changes by changing the cross-linker and cross-linker concentration at same magnification. Total intrusion volume, porosity (%) and pore size distribution also changes by changing cross-linker and cross-linker concentration. Thermal degradation results reveals that zinc and cadmium alginates started decomposing at 100 °C, but rapid degradation started around 300 °C and showed a stepwise weight loss during thermal sweep, indicating different types of reactions during degradation. Kinetic analysis was performed to fit with TGA data, where the entire degradation process has been considered as two or three consecutive 1st order reactions.  相似文献

This review summarizes data on alginates, polysaccharides isolated from brown algae. Their raw sources, chemical structure, and physical–chemical properties are discussed briefly. This paper is mainly focused on the physiological activity and usage of alginates in food and pharmaceutical industries. The data on alginate toxicity and necessary safety measures are also presented. The capability of alginates to bind and eliminate heavy metals and radionuclides from animal and human organisms is discussed. This paper also deals with the effects of alginates on the metabolism of lipids, carbohydrates, etc., and their usage in medicine, dietetics, and biotechnology.  相似文献

Alginate is an important polysaccharide used widely in the food, textile, printing and pharmaceutical industries for its viscosifying, and gelling properties. All commercially produced alginates are isolated from farmed brown seaweeds. These algal alginates suffer from heterogeneity in composition and material properties. Here, we will discuss alginates produced by bacteria; the molecular mechanisms involved in their biosynthesis; and the potential to utilize these bacterially produced or modified alginates for high-value applications where defined material properties are required.  相似文献

A chemotaxonomic study on the marine brown alga Cystoseira schiffneri collected from the Tunisian marine coast allowed us to identify kjellmanianone ( 1 ) and a new isololiolide derivative named schiffnerilolide ( 2 ). The structure elucidation and the assignment of relative configurations of the isolated natural products were based on advanced mass spectrometric and nuclear magnetic resonance techniques. This outcome suggested a close phylogenetic relationship of C. schiffneri with brown algae belonging to genus Sargassum C. Agardh . Molecular characterization using the nuclear small subunit rRNA (SSU rRNA) gene (18S) sequence as genetic marker was made. Pigment analysis showed a significant seasonal change of carotenoids, in particular of fucoxanthin and fucoxanthinol. Also galactolipids, the main constituents of the thylakoid membranes, showed remarkable seasonal changes.  相似文献

A micellar electrokinetic chromatography method was developed for the determination and quantification of sodium alginate. The alginate peak migrated in the very short time of 1.33 min and calibrated easily though the polydisperse properties of alginates. The minimum detection limit (LOD) of the method was calculated as 0.393 mg/ml. This analysis method was successfully applied to the alginate quantification in an antacid pharmaceutical formulation. Precise and reproducible analysis results were obtained, with liquid formulations injected directly without any pre-separation process.  相似文献

A bacterium (strain Al) isolated from a ditch produces three kinds of intracellular alginate lyases [Al-I (molecular weight: M.W. 60,000), Al-II-1 (M.W. 60,000) and Al-II-2]; the former two lyases have been purified and characterized (Yonemoto et al., J. Ferment. Bioeng., 72, 152–157, 1991). As part of a series of studies, Al-II-2 lyase was purified from cell-free extract of the bacterium. The lyase, with a M.W. of 25,000, depolymerized sodium-, potassium- and propyleneglycol alginates most efficiently at pH 8.0, 70°C, but it was inactive toward bacterial alginates with O-acetyl groups.  相似文献

A series of alginates isolated from the stem and leaf of a brown algae (Laminaria hyperborea), bacterial mannuronan, in vitro epimerized mannuronans, and periodate oxidized alginates were analyzed by size-exclusion chromatography (SEC) combined with online multiangle laser light scattering (MALS) and viscometry (collectively abbreviated SMV). Selected samples were also analyzed off-line using low-angle laser light scattering and capillary viscometry. Excellent agreement between the two methods was obtained for properly purified samples. In contrast, abnormal results were obtained for some industrial samples due to the presence of particulate material. Naturally occurring alginates and in vitro epimerized mannuronans were found to obey essentially the same RG-M and [eta]-M relations, and hence, the same Mark-Houwink-Sakurada (MHS) equations (valid for I = 0.10 M): 20 000 g/mol < M < 100 000 g/mol, [eta] = 0.0054 .M(1.00); 100 000 g/mol < M < 1 000 000 g/mol, [eta] = 0.071 .M(0.89). Application of the wormlike chain model to the [eta]-M data obtained by SMV yielded persistence lengths (q) of 15 nm for all alginates at an ionic strength of 0.17 M. Intrinsic viscosities corresponding to infinite ionic strength were estimated on the basis of Smidsr?d's B-parameter, and the wormlike chain model then yielded q = 12 nm. Periodate oxidized alginates showed, in contrast, a pronounced decrease in persistence length with increasing degree of oxidation, reaching values below 4 nm at 44% oxidation. Periodate oxidation also resulted in some depolymerization, even in the presence of a free-radical scavenger.  相似文献

Alginate may be considered as a block co-polymer of D-mannuronic and L-guluronic acids, and consists of three types of blocks: homopolymeric blocks of mannuronic acid (MM) and of guluronic acid (GG), and blocks with an alternating sequence (MG). The block composition of alginates has been characterized by a simple chemical method involving partial hydrolysis with acid, followed by fractional precipitation of the acid-resistant part of the alginate. Alginates from eleven different species of brown algae have been examined and, for five species, alginates from different tissues have been compared. The results indicate that young tissue is rich in MM blocks, and that the difference between the alginates from different species is mainly due to the alginates from the older parts of the plants. Extracellular alginates from two types of bacteria have been examined.  相似文献

I.r. absorption bands associated with the functional groups of carboxylic acid derivatives are useful for the analysis of alginates and pectins. The ester, amide, and uronate contents of pectins and the uronate content of alginates were determined, respectively, from the ester-carbonyl stretching band (1740 cm- minus 1), the amide I band (1650 cm- minus 1), and the carboxylate antisymmetric stretching band (1607 cm- minus 1) obtained from the spectra of solutions in D2O-phosphate buffer. The results are accurate to within plus or minus 2-4%, are self consistent, and agree well with the few reliable results that are available. The method should be applicable for the determination of carboxylic acid derivatives in other polysaccharides.  相似文献

Summary A simple method of producing ultrafiltration membranes, using calcium and barium alginates, has been developed. Although restricted in their flux and rejection characteristics these membranes offer a cheap model system for studying the fouling of hydrophilic membrane surfaces. The method is reproducible, and with suitable modification allows the membranes to be dried and reconstituted with little effect on performance.  相似文献

Further development of the poly(hexamethylenebiguanidinium chloride) assay method previously reported by these authors for the determination of alginates has resulted in a rapid procedure to determine this polysaccharide in liquors obtained by alkali extraction of brown seaweeds. The method involves neutralisation of the liquor followed by a simple alginate precipitation procedure employing the organic cation poly(hexamethylenebiguanidinium chloride) which possesses a UV absorption at 235 nm and can therefore be used in the quantitative step of the assay. Results produced by the assay for determination of alginate in liquors are in good agreement with another recognised method. The method is insensitive to the presence of numerous substances in the liquors in addition to alginate. In contrast to many other existing methods, alginate materials with various d-mannuronic acid/l-guluronic acid ratios show essentially identical responses to the assay.  相似文献

A ‘lignin’ fraction isolated by the Björkman method from the marine brown alga Fucus vesiculosus has been examined by 13C NMR and degradative analysis, and shown to consist of polyphloroglucinols identical to those characterized earlier from Fucus. Two new meronorsesquiterpenes (cystoazorones A and B) and two new meroditerpenes (cystoazorols A and B), along with benzoic acid were isolated from the brown macroalga Cystoseira abies-marina. The structures of the new compounds were established by 1D and 2D NMR as well as HRMS spectral analysis. The in vitro cytotoxicity and antioxidant activity of the isolated compounds were also evaluated. Cystoazorones A and B, and cystoazorol A exhibited in vitro growth inhibitory activity against HeLa cells. The HeLa cell line in log phase was found to be more sensitive to cystoazorol A than when it was in lag phase. Cystoazorol A also showed a selectivity index higher than taxol, which was used as a positive control. Cystoazorols A and B were found to be the strongest antioxidants among the compounds tested.  相似文献

News on both the structure and the periodism of the communities with Cystoseira from the Mediterranean Sea based on literature data, are presented.

In order to a better knowledge of such communities, the opportunity of a standardized methodology based on a preliminary study of the qualitative minimal area, on monthly samples to be carried out throughout a year as well as on observations on the vertical distribution of species within the community, is suggested.  相似文献
