Olfactory receptor neuron (ORN) response was measured to assess why some males ("rare males") of the Asian corn borer (ACB), Ostrinia furnacalis, have a broad behavioral response to fly upwind to both the ACB and the European corn borer (ECB), Ostrinia nubilalis, pheromone blends. We performed single-sensillum electrophysiological recordings on ACB males that had been behaviorally assessed for upwind flight response to the ACB blend [60:40 (Z)-12-tetradecenyl acetate (Z12-14:OAc) to (E)-12-tetradecenyl acetate (E12-14:OAc)], as well as to ECB (Z-strain) and ECB (E-strain) blends [3:97 and 99:1 (Z)-11-tetradecenyl acetate (Z11-14:OAc) to (E)-11-tetradecenyl acetate (E11-14:OAc)]. Sensilla from all types of males had large- and small-spike-sized ORNs responding strongly to Z12- or E12-14:OAc, but weakly to Z11- and E11-14:OAc. In the majority of males ("normal males") that flew upwind only to the ACB blend, Z11-14:OAc elicited responses in an intermediate spike-sized ORN associated with behavioral antagonism that is mainly tuned to (Z)-9-tetradecenyl acetate (Z9-14:OAc). In the rare-type ACB males that flew to both the ACB and ECB pheromone blends, Z11-14:OAc did not stimulate this ORN. Increased responsiveness to ancestral pheromone components by ORNs associated with behavioral antagonism could be instrumental in reproductive character displacement, or in reinforcement and reproductive isolation during speciation by helping to increase assortative mating between males and females in derived populations that use novel sex pheromone blends.

Addition of (Z)-11-hexadecenyl acetate (Z11-16:Ac) into a normally attractive binary blend of Heliothis virescens pheromone components resulted in a suppression of upwind flight and source location by males. Male response was reduced even at the lowest dosages of Z11-16:Ac tested but upwind flight and source location were most clearly reduced when the loading of Z11-16:Ac reached 10% or more of the (Z)-11-hexadecenal (Z11-16:Ald) loading (the major component present in the binary blend). Similar patterns of suppression in response were noted when Z11-16:Ac was added to binary blends of pheromone components at both 10 and 100 μg loadings of Z11-16:Ald. Males in casting flight following upwind flight in a mechanically generated pulsed plume, responded to the interception of a subsequent, single binary-blend filament by making a toward-source upwind surge. Responses of males to a single filament that was tainted by a level of Z11-16:Ac that had allowed some reduced level of upwind flight and source location to occur in the previous plume experiments were diminished compared with their control counterparts. Analysis of the flight tracks revealed that the surges in response to single tainted filaments were stunted because males made fewer significant changes in course angles steered, airspeeds generated, and in the tempo of counterturns executed. Pheromone mixture discrimination by male cabbage loopers, Trichoplusiani (Hübner), was assessed in a wind tunnel by responsedifferences to paired stimuli. They discriminate differencesbetween mixtures that contain three and all six of the pheromonecomponents that are emitted by virgin females. The ability todiscriminate between these two mixtures is perplexing becauseonly three antennal pheromone specialist neurons respond atnatural stimulus intensities. The cabbage looper also discriminatedbetween mixtures whose component ratios differed slightly fromthat of the female. There was some interaction between the stimulusintensity, the distance between the stimulus sources, and possiblythe extent of the perceptual difference. The mixtures of pheromonecomponents failed to enhance the amount of upwind flight responseover that of Z7–12:Ac alone. Other evidence argues thatthe instantaneous release of pheromone may exceed time-averagedmeasures.

Female Asian elephants (Elephas maximus) release a pre-ovulatory urinary pheromone, (Z)-7-dodecenyl acetate (Z7--12:Ac), to signal males of their readiness to mate. Z7--12:Ac is quantitatively elevated during the follicular stage of estrus, reaching maximum concentrations just prior to ovulation, as demonstrated by two complementary headspace techniques: (i) evacuated canister capture followed by cryogenic trapping; (ii) solid phase microextraction (SPME), used prior to gas chromatography/ mass spectrometry (GC/MS). These patterns were coincident with observed male behaviors and were consistent with biochemical and binding properties of the active ligand, including optimal binding pH. To release maximum amounts of Z7--12:Ac for quantitation, serum and urine samples from three mature female Asian elephants in their luteal and follicular stages of several estrous cycles were subjected to heat and pH changes and were then treated with protease prior to SPME-GC/MS analyses. When the post-luteal serum progesterone concentrations declined to baseline levels, Z7--12:Ac became detectable in the female urine. Throughout the follicular stage pheromone concentrations increased linearly with no apparent relationship to the two serum luteinizing hormone peaks. Pre-ovulatory urine also contained related compounds, including (Z)-7--12-dodecenol. The relative amount of this alcohol increased relative to acetate during long-term storage, with a proportional reduction in bioactivity. Z7--12:Ac was not detected in mucus samples from the urogenital tract. A potential precursor of Z7--12:Ac was identified in liver homogenates from female elephants in the follicular stage.

Abstract. We investigated the behavioural response of male Helicoverpa zea (Boddie) to the fine-scale structure of an odour plume experimentally modified in a wind tunnel by using an air-pulsing device. Male H. zea flew upwind to pulsed filaments of a binary pheromone blend of (Z)-11-hexadecanal (Z11-16:Ald) and (Z)-9-hexadecanal (Z9-16:Ald) in the ratio of 20:1. Sustained upwind flight in experimentally altered intermittent plumes was dependent on concentration, as well as the frequency of generation of odour filaments. At a loading of 10μg of the major pheromone component, Zll-16:Ald, which gave an emission rate of approximately that released by a female H. zea , sustained upwind flight and source contact correlated positively with filament delivery rate, becoming significant at a minimum filament delivery rate of 2/s. Decreases in upwind progress and source location were recorded at a loading of 1 μg of Z11-16:Ald. At this suboptimal dosage, a high filament generation rate of 10/s was necessary for significant upwind progress and source contact. When an interspecific compound: (Z)-11-hexadecenyl acetate (Z11-16:OAc), was added to the attractive pheromone binary aldehyde blend of H. zea at a proportion of 10% of the major pheromone component, and pulsed from the same source, there was a significant reduction in sustained upwind progress and source location by males, indicating that Z11-16:OAc is antagonistic to the upwind progress of H. zea. However, Z11-16:OAc was less antagonistic when its filaments were isolated and alternated with pheromone filaments, indicating a strong effect of the synchronous arrival of odour filaments on the antenna needed for antagonism of upwind flight.

As the ratio of (E)-8-dodecenyl acetate (E8–12Ac) to (Z)-8-dodecenyl acetate (Z8–12Ac) increased past optimal low levels in the pheromone blend, fewer males were able to fly 2.5 m upwind to the source. The tracks of males that flew in plumes of such high-(E)off-blends were slower and narrower than those of males flying to lower-(E)blends. The tracks were narrower, first of all, because as the proportion of E8–12Ac increased, the males steered more into the wind. More of their thrust was directed upwind and therefore their groundspeed to either side of the windline was reduced. In addition, males also reduced their airspeeds to high-(E)blends, which contributed to the decreased groundspeeds and narrower tracks. No significant changes in the frequency of counterturning were found in response to increasing proportions of E8–12Ac. The inability to continue upwind flight in a plume of an off ratio was indicated by in-flight arrestment in the plume. Arrestment resulted from changes in the course angles steered by the males and the airspeeds flown.

The upwind flight and landing responses of male Mamestra suasa (Den. & Schiff.) (Lepidoptera; Noctuidae) to various synthetic chemicals were studied in a wind tunnel. A mixture of (Z)-11-hexadecenyl acetate (Z11-16: Ac) and (Z)-11-hexadecenal (Z11-16: Ald) elicited upwind flight responses past the midpoint of the wind tunnel, and both components were necessary for landing of the males to occur at the pheromone source point. The best response to the mixtures tested was to a ratio 100:1 (ng) of Z11-16: Ac and Z11-16: Ald (blend B).The addition of (Z)-9-tetradecenyl acetate (Z9-14: Ac) (10 ng) to blend B resulted in a significant decrease of upwind flight and landing responses. The inhibitory effect of this chemical when it is mixed with blend B, and the presence of a specific receptor for this compound in sensilla trichodea of male M. suasa, suggest that Z9-14: Ac is involved in the chemical communication between M. suasa and other sympatric species. The addition of (Z)-11-hexadecenol (Z11-16: OH) (1 or 10 ng) to blend B had no effect on male upwind flight and landing responses. This compound does not seem to be involved in the chemical communication of this species.

Abstract The sex attractant of the female redbanded leafroller moth, Argyrotaenia velutinana (Walker), is a blend of seven compounds. Specialized olfactory receptor neurones had been found for only two of the compounds, (Z)-11-tetradecenyl acetate (Z11-14:Ac) and (E)-11-tetradecenyl acetate (E11–14:Ac). These receptor neurones were always found in pairs within the long trichoid sensilla, which are the most abundant multi-pored sensilla on the male antenna. A systematic survey of all regions of the male antenna with standard extracellular recording techniques was undertaken to find receptor neurones responsive to the remaining five minor components of the female pheromone. Of the 113 long trichoid sensilla sampled, all contained two receptor neurones, one specialized for Z11–14:Ac and a second specialized for Ell –14:Ac. A comparable number of recordings were then obtained from the less abundant classes of multi-pored sensilla. Two new receptor neurone types were found, responsive to the minor pheromone components (E)-9-dodecenyl acetate (E9-12:Ac) and (Z)-9-dodecenyl acetate (Z9-12:Ac). Scanning electron micrographs indicated that these recordings were obtained from shorter, narrower trichoid sensilla. The majority of these sensilla appeared to contain three neurones capable of spontaneous action potential production. In each sensillum, only one receptor neurone appeared to respond to stimulation with a minor component of the female blend. The remaining two neurones did not respond to the chemical stimuli evaluated.

ABSTRACT. The flight response of individual male Oriental fruit moths, Grapholitha molesta (Busck), was observed in a sustained-flight tunnel to 100 blend–dosage combinations of the three sex pheromone components: (Z)- and (E)-8-dodecenyl acetate and (Z)-8-dodecen-l-ol (1, 3, 10, 30 and 100 μg of Z8-12: AC with, 2, 6, 10, 20% E and, 0, 3, 10, 30 or100% OH alcohol added). Complete flights to the source were observed only to blend combinations containing all three components. Males exhibited highest response levels to two dosages (3 and 10μg) of the natural 6% E blend and these levels were relatively unaffected by changes in the proportion of Z8-12: OH. Certain treatments surrounding the peak area also elicited high response levels compared to the 6% E treatments, but these were strongly dependent on the proportion of OH in the blend. Hierarchical cluster analysis was utilized to compare and group treatments that elicited similar levels of response over the entire flight sequence and to derive an area of blend-dosage combinations within the matrix tested that elicited peak levels of attraction. Analysis of the response patterns for suboptimal treatments adjacent to the area of optimal attraction showed that response specificity was controlled by two major effects on flight behaviour, one occurring early in the flight sequence affecting male orientation to the odour plume, and the other occurring later in the sequence as an arrestment of upwind flight. These effects were strongly influenced by changes in the OH component and the E isomer, with the latter playing the critical role in effecting flight behaviour. Temporal analysis of the flight response to treatments within the optimal area showed that whereas the % OH did not significantly affect the magnitude of response, increasing the level of Z8-12: OH in the blend did significantly increase the duration of each phase of the behavioural sequence. Considering both the magnitude and temporal aspects of male response, optimal attraction in male OFM was best characterized by a small area of treatments around the peak 6% E blends that contained 10% OH. Finally, field tests showed a high degree of correlation between trends in response with changing proportion of Z8-12: OH as observed in the flight tunnel. Peak dosages were generally higher in the field, however, compared to those in the flight tunnel.

Flight-tunnel experiments were conducted using Helicoverpa zea males to determine whether or not (Z)-11-hexadecen-1-ol (Z11-16:OH), a compound emitted by another heliothine moth species, Heliothis subflexa, is a behavioral antagonist when admixed with the two-component pheromone blend of H. zea. Males were less likely to fly upwind all the way to the source when 0.3% Z11-16:OH was present in the blend. Even 0.1% Z11-16:OH caused differences in the flight behavior of H. zea males; they steered more off the windline than males responding to the pheromone blend alone, resulting in more oblique track angles. Thus Z11-16:OH appears to act antagonistically, along with another compound, (Z)-11-hexadecen-1-ol acetate (Z11-16:Ac), when it is added to the H. zea pheromone blend.

ABSTRACT In field-trapping experiments the unsaturated analogues ( E )-6-, ( E )-12-, and ( Z )-12-tetradecenyl acetate were as attractive to male Yponomeuta rorellus Latr. as the native pheromone component tetradecyl acetate. All four analogues attracted more males than virgin females did, whereas ( Z )-6-, ( E )-11-, ( Z )-10- and ( Z )-11-tetradecenyl acetate were essentially non-attractive. Addition of 1–30% of ( Z )-11-tetradecenyl acetate to the pheromone tetradecyl acetate reduced the attraction to less than 2%. Flight tunnel experiments with Y. rorellus confirmed the activity of the ( E )-6- and ( E )-12-tetradecenyl acetates and demonstrated the activity of ( E )-7-tetradecenyl acetate as well. These analogues elicited orientation behaviour, upwind flight and landing at the odour source as frequently as the native pheromone did. Single sensillum recordings from male Y. rorellus showed two types of cells in most sensilla. A large spike amplitude cell was stimulated by tetradecyl acetate and the unsaturated analogues ( E )-11-, ( E )-6- and ( E )-12-tetradecenyl acetate, and to a lower extent by the ( Z )-6-, ( Z )-11- and ( Z )-12-isomers. A cell with medium spike amplitude was stimulated by ( Z )-9-tetradecenyl and ( Z )-11-hexadecenyl acetate. Some sensilla contained a third cell firing with a small spike amplitude which was activated by ( Z )-11-tetradecenol. Thus the tetradecyl acetate receptor was stimulated not only by the behaviourally active analogues, but also by behavioural antagonists. The interaction of ( E )-11-tetra-decenyl acetate and tetradecyl acetate with the same antennal receptor cell was also demonstrated in Y.cagnagellus . Electrophysiological discrimination between behavioural attractants and antagonists and the role of behavioural antagonists in the interspecific relations between Y.rorellus and sympatric closely related species are discussed.

The relative attractiveness of synthetic Choristoneura rosaceana (Harris) pheromone consisting of the major compound Z-11-tetradecenyl acetate (Z11-14:OAc), or the major compound plus one, two, or all three of the minor compounds E-11-tetradecenyl acetate (E11-14:OAc), Z-11-tetradecenol (Z11-14:OH), and Z-11-tetradecenal (Z11-14:Ald), in amounts similar to those in the natural pheromone, was compared in a flight tunnel and in apple orchards. In the flight tunnel, there was an increase in the proportion of moths that were activated when E11-14:OAc was combined with the main compound, but no detectible additional increases in response with the addition of Z11-14:OH and Z11-14:Ald. The time required for activation was twice as long when using the major compound than when using the major compound and one or more of the minor compounds. There was a trend of increase in the proportion of males initiating the take-off, lock-on, close-in, and touchdown phases of upwind flight when progressively more complete blends were used. The time required for the initiation of these behavioral phases was statistically similar for each treatment. The proportion of moths landing at the pheromone source was 10 times greater when using the complete blend than when using the main compound alone. Similar results were obtained in apple orchards, where the four-compound blend was 17-55 times more attractive than the main compound. The superior attractiveness of the four-compound pheromone compared with the major compound, or to the major compound plus the minor compound E11-14:OAc, suggests that it would be the most effective synthetic pheromone if competitive attraction were an important mechanism of disruption in this species.

Abstract. The neurophysiological bases for behaviourally expressed olfactory redundancy in the sex pheromone communication system of the cabbage looper moth, Trichoplusia ni (Hübner) (Lepidoptera: Noctuidae), were examined by coupling the cut-sensillum extracellular recording technique with a highly specific neuronal marking method for moth peripheral receptors. In seventy-two antennal sensilla, axonal pathways of cobalt-stained neurones could be traced into the male-specific macroglomerular complex in the antennal lobe. In T. ni males this comprises five glomeruli, two of which are subdivided into morphologically, and in some instances functionally identifiable, regions. Axonal arborizations of forty-eight neurones (single stainings) showed high fidelity (98%) for containment within a specific glomerulus or glomerular subdivision, and the neuropil targeted seemed to be related to the specificity of a neurone to a particular female-emitted sex pheromone component (27-12:Ac, Z7-14:Ac, Z9-14:Ac, 12:Ac, 11–12:Ac, Z5-12:Ac), or to a behavioural antagonist (Z7-12:OH). Double (twenty-one) and multiple stainings (three) showed axons projecting into two or more glomeruli, respectively, with 100% fidelity for the component-specific glomerulus or glomerular subdivision to be targeted. We suggest that the potential for a single minor component to cross-stimulate two or more neurones within a sensillum may enable partial blends to continue to provide sensory input into all of the pheromone-processing glomeruli of the complex. Our interpretation is that redundancy occurs at the receptor level on neighbouring dendrites, and thus allows various four-component partial blends to evoke full pheromone-mediated behaviour.

Six compounds were identified from gland extracts of the cotton bollworm, Heliothis armigera(Hubner): (Z)-11-hexadecenal (Z11-16:Ald), (Z)-9-hexa-decenal (Z9-16:Ald), hexadecanal, (Z)-11-hexadecenol (Z11-16:OH), (Z)-7-hexadecenal (Z7-16:Ald), and (Z)-9-tetradecenal (Z9-14:Ald). Each of the compounds that were identified was examined for its ability to elicit sexual responses from male moths in a flight tunnel. Males flew upwind to Z11-16:Ald alone, but greater levels of copulatory responses were evoked with the addition of 2.5% Z9-16:Ald to the Z11-16:Ald. Addition of hexadecanal to the binary mixture had no effect in raising the behavioral response of the males in the flight tunnel. The effect of Z7-16:Ald on male flight depended on the loading. The addition of 1% of this component to 2 mg of the binary mixture reduced levels of copulatory response, but the same addition (1 %) to 10 g of the binary mixture increased copulatory response. The addition of 79-14:Ald or Z11-16:OH to the binary mixture reduced behavioral responses of males. High loadings of the binary mixture (200–2000 g) were better than a low loading (10 g) in eliciting response of males.Contribution from the Agricultural Research Organization, The Volcani Center, Bet Dagan, Israel. Analysis of sex pheromone glands of the apple leafroller Bonagota cranaodes Meyrick by gas chromatography coupled with mass spectrometry or electroantennographic detection showed the presence of 14 structurally related acetates and alcohols of the chain length 10-18, including the main pheromone component (E,Z)-3,5-dodecadienyl acetate (E3,Z5-12Ac). Male antennae responded to the main compound, its Z,Z isomer, (E,Z) -3,5-tetradecadienyl acetate (E3,Z5-14Ac), and the monoenes (Z)-5-dodecenyl acetate (Z5-12Ac) and (Z)-9-hexadecenyl acetate (Z9-16Ac). Traps baited with a four-component blend of E3,Z5-12Ac, Z5-12Ac, E3,Z5-14Ac, and Z9-16Ac in a 100:5:5:100 ratio were significantly more attractive than the main compound alone. This improved trap lure is more suitable for monitoring population densities of B. cranaodes, and for detection of the onset of the seasonal flight period. A more complete pheromone blend is of importance also with respect to current attempts to develop mating disruption for control of this major pest of apple in Brazil.

Lutzomyia longipalpis adult males form leks on or near hosts and release (1S,3S,7R)-3-methyl-alpha-himachalene from their tergal glands to lure females to the same site for mating and feeding. Here we have examined whether the male-produced attractant could also serve as a male aggregation stimulus. High resolution chiral capillary gas chromatography analysis of male tergal gland extracts, synthetic (1S,3S,7R)-3-methyl-alpha-himachalene, and a synthetic mixture of all isomers of 3-methyl-alpha-himachalene, was coupled to electrophysiological recordings from ascoid sensillum receptor cells in antennae of male and female sandflies. Receptor cells of both sexes responded only to the main component of the male tergal gland extract that eluted at the same retention time as (1S,3S,7R)-3-methyl-alpha-himachalene. Furthermore, of the eight 3-methyl-alpha-himachalene isomers in the synthetic mixture only the fraction containing (1S,3S,7R)-3-methyl-alpha-himachalene, co-eluting with an isomer of (1S*,3S*,7S*)-3-methyl-alpha-himachalene, elicited an electrophysiological response from male and female ascoid sensillum receptor cells. Both males and females flew upwind in a wind tunnel towards a filter paper disk treated with either 4-6 male equivalents of the tergal gland extract, pure (1S,3S,7R)-3-methyl-alpha-himachalene or the synthetic mixture of eight isomers. This indicates that (1S,3S,7R)-3-methyl-alpha-himachalene derived from L. longipalpis males may have a dual function in causing male aggregation as well as serving as a sex pheromone for females.
