Summary Nuclei of various cell types in the eye of embryonal and adult Guinea pigs were studied by means of electron microscopy, cytophotometry and autoradiography. Striking differences in condensation and arrangement of chromatin were found between the different tissues and cells. Several nuclear types were analyzed quantitatively with regard to their content of condensed and decondensed chromatin by means of electron microscopic morphometry. Structural differences in chromatin organization coincided with different nuclear DNA contents in various cell types of the retina, such as bipolar cells, Müller cells, rods and cones, and the pigmented epithelium.The differences between DNA-Feulgen means obtained by cytophotometric analysis were highly significant. Template activity as shown by3H-uridine incorporation made evident that the rate of RNA synthesis is positively correlated with the quantity of decondensed chromatin. It is speculated that differentiation of the Guinea pig eye involves differential DNA synthesis, and that the extra-DNA could have some trigger function for the pattern of chromatin condensation and thus the pattern of gene expression.  相似文献

The replication fidelities of Pfu, Taq, Vent, Deep Vent and UlTma DNA polymerases were compared using a PCR-based forward mutation assay. Average error rates (mutation frequency/bp/duplication) increased as follows: Pfu (1.3 x 10(-6)) < Deep Vent (2.7 x 10(-6)) < Vent (2.8 x 10(-6)) < Taq (8.0 x 10(-6)) < < exo- Pfu and UlTma (approximately 5 x 10(-5)). Buffer optimization experiments indicated that Pfu fidelity was highest in the presence of 2-3 mM MgSO4 and 100-300 microM each dNTP and at pH 8.5-9.1. Under these conditions, the error rate of exo- Pfu was approximately 40-fold higher (5 x 10(-5)) than the error rate of Pfu. As the reaction pH was raised from pH 8 to 9, the error rate of Pfu decreased approximately 2-fold, while the error rate of exo- Pfu increased approximately 9-fold. An increase in error rate with pH has also been noted for the exonuclease-deficient DNA polymerases Taq and exo- Klenow, suggesting that the parameters which influence replication error rates may be similar in pol l- and alpha-like polymerases. Finally, the fidelity of 'long PCR' DNA polymerase mixtures was examined. The error rates of a Taq/Pfu DNA polymerase mixture and a Klentaq/Pfu DNA polymerase mixture were found to be less than the error rate of Taq DNA polymerase, but approximately 3-4-fold higher than the error rate of Pfu DNA polymerase.  相似文献

The genome of guinea pig cytomegalovirus (GPCMV) was analyzed and compared with that of human cytomegalovirus (HCMV). GPCMV and HCMV DNAs were isolated from virions and further purified by CsCl centrifugation. Purified GPCMV DNA sedimented as a single peak in a neutral sucrose gradient and was infectious when transfected into guinea pig embryo fibroblast cells. The cytopathology was characteristic of that seen after infection with GPCMV. Virus DNA purified from virions isolated from infected GPEF or 104C1 cells had a CsCl buoyant density of 1.713 g/cm3, which corresponds to a guanine plus cytosine content of 54.1%. The CsCl buoyant density of GPCMV DNA was slightly less than that of HCMV DNA (1.716 g/cm3), but sufficiently different so that the two virus DNA peaks did not coincide. GPCMV DNA cosedimented with T4 DNA in a neutral sucrose gradient. Restriction endonuclease cleavage of GPCMV or HCMV DNAs with HindIII, XbaI, or EcoRI yielded fragments easily separable by agarose gel electrophoresis and ranging from 1.0 X 10(6) to 25.8 X 10(6) daltons. The number, size, and molarity of GPCMV DNA fragments generated by restriction enzymes were determined. Hybridization of restriction endonuclease-cleaved GPCMV DNA with radioactively labeled HCMV DNA and, conversely, hybridization of restriction endonuclease-cleaved HCMV DNA with radioactively labeled GPCMV DNA indicated sequence homology between the two virus DNAs.  相似文献

The tissue localization patterns of radiolabeled dividing cells obtained from gut lamina propria (LP), mesenteric (MLN) and peripheral (PLN) lymph nodes, and Peyer's patches (PP) were studied in guinea pigs using an adoptive lymphocyte transfer method. Within 24 hr 125I-deoxyuridine-labeled cells from donor LP and MLN, but not PLN, selectively localized in recipient gut and MLN. In contrast, donor PLN lymphoblasts returned to their sites of origin while labeled PP donor cells exhibited no specific tissue localization. These findings suggest that the gut LP, like the MLN, contains a population of cells which, unlike those in the PP and PLN, has the capacity to selectively localize in mucosal tissues. From this and other published work, we conclude that within the LP there is a population of cells at different stages of differentiation with a propensity to populate mucosae.  相似文献

Leukemic guinea pig lymphocytes (L2C) have ten times as many low-density lipoprotein (LDL) receptors as healthy lymphocytes, but LDL accounts for only 38% of the cholesterol in L2C cells, compared to more than 95% in normal cells. Our data show that LDL fails to regulate cholesterol biosynthesis and that there is a defect in LDL internalization and receptor turnover in L2C cells. We also demonstrate that the degradation of LDL is not a limiting process. By discriminating between binding and internalization, we show that internalization in L2C is much slower than in normal cells and that the decrease in metabolism is related to the slow turnover of the LDL receptors.  相似文献

The fidelity of DNA synthesis with purified DNA polymerase alpha and beta from human placenta has been studied. With poly[d(A-T)] as the template-primer and Mg2+ as the metal activator, DNA polymerase alpha incorporates 1 mol of dGMP for every 6,000 to 12,000 mol of complementary nucleotides polymerized. Under the same conditions, DNA polymerase beta is more accurate, the error rate being 1/20,000 to 1/60,000. This greater accuracy of DNA polymerase beta is observed with a variety of homopolymer templates. With both enzymes, substitution of Mg2+ with activating concentrations of Mn2+ or Co2+ enhances the frequency of misincorporation. At greater than activating concentrations of Mn2+ and Co2+, there is an inhibition of complementary nucleotide incorporation, further increasing the frequency of misincorporation. Nearest neighbor analysis of the products synthesized with both enzymes indicates that the noncomplementary nucleotides are incorporated predominantly as single base substitutions. The greater accuracy of DNA polymerase beta over DNA polymerase alpha should be considered in relationship to their possible roles in DNA replication and repair.  相似文献

1. The metabolism of flavone has been studied in the guinea pig; after administration of the compound either orally or intraperitoneally, 4'-hydroxyflavone is excreted in the urine. 2. Small amounts of 3',4'-dihydroxyflavone were also detected in the urine after oral administration of flavone. 3. Hydroxyflavone formation is not suppressed by administering high doses of the antibacterial compounds aureomycin and phthaloylsulphathiazole. 4. Although salicylic acid appeared in trace quantities in the urine after administration of flavone, no other aromatic acid metabolites of flavone were detected.  相似文献

The guinea pig plantar epidermis was examined by light-microscopical histochemical methods and by transmission electron microscopy. Autolysis of cell structure was much less complete in guinea pig plantar horny layer than in the back, and stainable cytoplasm was retained in keratinized cells but organelles were lost except for some degraded ultrastructural remnants. By light microscopy the whole thickness of the horny layer showed bound phospholipid and bound cysteine, and there was a weak cystine reaction at the peripheries of the keratinized cells. In ultrastructure the keratohyalin contained slightly larger subparticles than in the back skin. The horny layer was not divisible into basal, intermediate and superficial regions as in hairy skin. The stratum lucidum of light microscopy was not defined in electron micrographs. Osmium-stained cytoplasmic material was retained in horny cells about to be desquamated, in contrast to the empty appearance of these cells in hairy skin. Epidermal cells in plantar skin have ultrastructural cytoplasmic processes which are longer than they are broad. In the horny layer these interdigitate with those of neighbouring cells and are held together by lateral demonsomal junctions. Probably this gives mechanical strength against shearing forces experienced by the plantar horny layer.  相似文献

Studies on DNA polymerases and topoisomerases in archaebacteria   总被引:1,自引:0,他引:1  
We have isolated DNA polymerases and topoisomerases from two thermoacidophilic archaebacteria: Sulfolobus acidocaldarius and Thermoplasma acidophilum. The DNA polymerases are composed of a single polypeptide with molecular masses of 100 and 85 kDa, respectively. Antibodies against Sulfolobus DNA polymerase did not cross react with Thermoplasma DNA polymerase. Whereas the major DNA topoisomerase activity in S. acidocaldarius is an ATP-dependent type I DNA topoisomerase with a reverse gyrase activity, the major DNA topoisomerase activity in T. acidophilum is a ATP-independent relaxing activity. Both enzymes resemble more the eubacterial than the eukaryotic type I DNA topoisomerase. We have found that small plasmids from halobacteria are negatively supercoiled and that DNA topoisomerase II inhibitors modify their topology. This suggests the existence of an archaebacterial type II DNA topoisomerase related to its eubacterial and eukaryotic counterparts. As in eubacteria, novobiocin induces positive supercoiling of halobacterial plasmids, indicating the absence of a eukaryotic-like type I DNA topoisomerase that relaxes positive superturns.  相似文献

Guinea pig insulin exhibits an unusually high degree of divergence from the conserved insulins of other mammals. cDNA clones encoding guinea pig preproinsulin were isolated, and their nucleic acid sequences were determined. Comparisons of the nucleic acid sequence and its predicted amino acid sequence with sequences encoding insulins of other species revealed that the gene encoding guinea pig preproinsulin evolved from the same ancestral mammalian gene as other known mammalian insulin genes.  相似文献

Bacteriophage T4 gene 43 codes for the viral DNA polymerase. We report here the sequence of gene 43 and about 70 nucleotides of 5'- and 3'-flanking sequences, determined by both DNA and RNA sequencing. We have also purified T4 DNA polymerase from T4 infected Escherichia coli and from E. coli containing a gene 43 overexpression vector. A major portion of the deduced amino acid sequence has been verified by peptide mapping and sequencing of the purified DNA polymerase. All these results are consistent with T4 DNA polymerase having 898 amino acids with a calculated Mr = 103,572. Comparison of the primary structure of T4 DNA polymerase with the sequence of other procaryotic and eucaryotic DNA polymerases indicates that T4 DNA polymerase has regions of striking similarity with animal virus DNA polymerases and human DNA polymerase alpha. Surprisingly, T4 DNA polymerase shares only limited similarity with E. coli polymerase I and no detectable similarity with T7 DNA polymerase. Based on the location of specific mutations in T4 DNA polymerase and the conservation of particular sequences in T4 and eucaryotic DNA polymerases, we propose that the NH2-terminal half of T4 DNA polymerase forms a domain that carries out the 3'----5' exonuclease activity whereas the COOH-terminal half of the polypeptide contains the dNTP-binding site and is necessary for DNA synthesis.  相似文献
