The effects of water deprivation, rehydration and hyperhydration were investigated in the black Moroccan goat (Capra hircus). Mean daily water intake was 46 ± 5 ml/kg in lactating and 36 ± 4 ml/kg in non-lactating black Moroccan goats, and milk production 21 ± 1 ml/kg. Mean urine excretion was 8 ± 2 ml/kg body weight in both groups, and the daily water losses via evaporation and feces were estimated at 23 ± 3 ml/kg during lactation and 28 ± 4 ml/kg during non-lactation. Forty-eight hours of water deprivation caused a body weight loss of 9% and 6% in lactating and non-lactating goats, respectively, and a drop of 28% in milk production with only a slight decrease in food intake. After rehydration, the elevated plasma osmolality as well as Na and total protein concentrations returned to basal values within 2–3 hr, indicating a rapid absorption of the ingested water, but urine excretion did not increase. After hyperhydration (10% of body weight), 46% of the load was excreted by the kidneys within 6 hr. In conclusion, black Moroccan goats have a low water turnover, and they can retain water upon rehydration but not store excess water after hyperhydration.  相似文献

The objective of this study was to investigate the relationship between nitrogen (N) partitioning and isotopic fractionation in lactating goats consuming diets with a constant high concentration of N and increasing levels of water soluble carbohydrate (WSC). Eight lactating goats were offered four different ratios of WSC : N in the diet. A two-period incomplete cross-over design was used, with two goats assigned to each treatment in each period. N balance measurements were conducted, with measurement of feed N intake and total output of N in milk, faeces and urine. Treatment, period and infusion effects were tested using general ANOVA; the relationships between variables were analysed by linear regression. Dietary treatment and period had significant effects on dry matter (DM) intake (g/day). DM digestibility (g/kg DM) and N digestibility (g/kg N) increased as the ratio of WSC : N increased in the diet. No treatment effect was observed on milk urea N concentration (g/l) or urinary excretion of purine derivatives (mM/day). Although dietary treatment and period had significant effects on N intake, the change of N intake was small; no effect was observed for N partitioning among faeces, milk and urine. Milk, plasma and faeces were enriched in 15N compared with feed, whilst urine was depleted in 15N relative to feed. No significant relationship was established between N partitioning and isotopic fractionation. This study failed to confirm the potential to use N isotopic fractionation as an indicator of N partitioning in dairy goats when diets provided N in excess to requirements, most likely because the range of milk N output/N intake and urinary N output/N intake were narrow.  相似文献

Binding of [3H]R5020 (17,21-dimethyl-19-nor-4,9-pregnadiene-3, 20-dione) to bovine mammary cytosol indicated the presence of progestin binding sites of high-affinity and low-capacity in tissue from prepartum, nonlactating and from postpartum, lactating cows. To prevent binding of [3H]R5020 to glucocorticoid binding sites, a 200-fold molar excess of nonradioactive cortisol was included during all incubations, thus specific binding was limited to progestin binding sites. Nonradioactive R5020 and progesterone effectively inhibited [3H]R5020 binding to progestin binding sites, while estradiol-17β, dihydrotestosterone (17β-hydroxy-5α-androstan-3-one), dexamethasone (9-fluoro-11β, 17, 21-trihydroxy-16α-methyl-1,4-pregnadiene-3,20-dione) or additional cortisol were ineffective. Dissociation constants for specifically bound [3H]R5020 in cytosol from mammary tissue of nonlactating and lactating cows were nearly identical, averaging 1.9 ( ± 0.3) and 0.8( ± 0.2) × 10?9M, respectively. However, binding capacities (fmol/mg cytosolic protein) were greater in cytosol from prepartum, nonlactating (179 ± 53) than postpartum, lactating (41 ± 15) cows. Specific binding components in cytosol from lactating cows sedimented in the 6-7S region on linear sucrose density gradients. When subjected to isoelectric focusing, specific binders with isoelectric points (pI) of approximately 6.1, 7.9 and 8.3 were resolved. The decrease in number of binding sites during lactation was due to the virtual absence of the anionic binding species, suggesting that their presence is necessary for progesterone to inhibit milk secretion.  相似文献

Eighteen multiparous lactating grazing Holstein cows, 9 ruminally cannulated, average 136.1 ± 14.6 days in milk, were randomly assigned to three treatments consisting of water containing different levels of total dissolved solids (TDS; mg/l): Treatment 1 = 1,000; Treatment 2 = 5,000 and Treatment 3 = 10,000, at the Experimental Dairy Unit at Rafaela Experimental Station (31°11′S latitude) during summer 2005. Animals were arranged in a randomized complete block design with three 28-day experimental periods, with 3 weeks for water adaptation and 1 week for measurements. Feed and water intake, milk production and composition, body weight and condition score and rumen parameters were evaluated. No treatment effects were observed in any of the variables evaluated, with the exception of water intake, which was higher for animals receiving 10,000 mg/l TDS in the drinking water (189 l/day vs. 106 and 122 l/day for cows receiving water with 1,000 and 5,000 mg/l TDS, respectively). Water intake was significantly higher for animals in treatment 10,000 (P < 0.05). It was concluded that the rumen presents a surprising buffer capacity and that consideration of TDS alone is insufficient to characterize drinking water quality.  相似文献

Lactating female rats without their pups exhibit lower HPA responsiveness to stress than nonlactating females. However, responsiveness to stress is similar when lactating females are tested with their pups and the stressor involves a potential threat to the offspring. This study constitutes the first comparison of stress responsiveness in lactating and nonlactating female nonhuman primates. Subjects were 53 multiparous female free-ranging rhesus macaques. Approximately half of the females were lactating and half were nonpregnant/nonlactating. Blood samples were obtained after capture and after overnight housing in an individual cage. Lactating females were tested with their infants. Lactating females had significantly higher plasma cortisol levels than nonlactating females on both days. Having or not having an infant was also a better predictor of plasma cortisol levels among all females than their age, dominance rank, group of origin, time of day at which the sample was obtained, and time elapsed since beginning of the sampling procedure or since anesthesia. Plasma cortisol levels of lactating females were not significantly correlated with post-partum stage or with the cortisol levels of their infants. Capture, handling, and individual housing in a cage are powerful psychological stressors for free-ranging primates. We suggest that the higher plasma cortisol levels exhibited by lactating females reflect greater responsiveness to stress associated with perception of risks to infants. Hyporesponsiveness to stress may not be a general characteristic of lactation in all mammalian species, but a short-term effect of infant suckling that is most apparent with stressors unrelated to the offspring.  相似文献

We investigated the physiological and behavioural strategies by which lactating Ethiopian Somali goats endure repeated periods of water deprivation. The experiment lasted for 32 days and was divided into four periods of 8 days each. Measurements were taken during the first 4 days in each period. Seven does (W1) were watered once daily and seven does (W4) were watered once every 4th day. Rectal temperature was taken at 0800 and 1715 h. Blood samples were taken in the evening and milk samples in the morning. The goats were on pasture between 0900 to 1215 h and 1315 to 1630 h with behaviour recorded every 5 min. The does were supplemented with 300 g of concentrates per head per day. Plasma and milk osmolality were determined by freezing point depression. Plasma total protein was measured on a TS refractometer. Plasma vasopressin concentrations were analysed by radio-immunoassay. The mean daily water intake of W1 was 1897 ml compared with the calculated mean of 1075 ml in W4 (P < 0.001). The mean diurnal variation of the rectal temperature was 3.5°C in both groups. Afternoon rectal temperature in W4 during period 1 was higher than that in the W1 on the days of water deprivation (P < 0.05). With repeated periods, plasma osmolality in W4 increased less over the days of water deprivation. It was 336, 321, 311 and 306 mosm/l on the 4th day at periods 1, 2, 3 and 4, respectively. The corresponding vasopressin concentrations were 10.0, 9.2, 4.2 and 4.4 pmol/l. Total plasma protein concentration during period 1 on days 3 and 4 were higher in W4 than in W1 (P < 0.01). During the subsequent periods, it did not increase more in W4 compared with W1, but it was lower in W4 on the days after watering. W4 milk production decreased by 22% compared with W1 in all periods. With increasing days of water deprivation, the W4 goats spent less time in the sun, grazed shorter time and frequently ate cactus (Opuntia ficus-indica) as compared with W1. Results suggested that dehydrated lactating Ethiopian Somali goats economised on water by diurnal variations of rectal temperature, storing water in the extracellular fluid, by changing behaviour at grazing and by decreasing milk production.  相似文献

Twelve wethers (27–33 kg and 12–14 months of age) representing meat (Nubian), milk (Alpine) and mohair (Angora) producing goats were used to evaluate breed differences in protein utilization with diets containing 9 (L), 15(M) or 21% CP (H) and 2.4 Mcal ME/kg DM in a digestion trial. Fecal N, urine N and absorbed N as percent of N intake were not affected (P>0.05) by breed. Retained N as percent of absorbed N was not different (P>0.05) between breeds. Ruminal propionate (molar %) was lower (P<0.05) in Angora, but other ruminal VFA and ammonia-N were not affected (P>0.05) by diet or breed. Plasma urea-N increased (P<0.01) with dietary CP level (8.3, 22.0 and 33.3 mg/dl for L, M and H, respectively), and concentrations were lowest for Angoras and highest for Nubians (18.5 vs. 21.2 vs. 23.9 mg/dl) (P<0.01). Plasma total protein (mean 69.7 g/l), glucose (mean 83.1 mg/dl), non-esterfied fatty acids (mean 101.4 μEq/l) and cortisol (mean 10.8 ng/ml) were not affected (P>0.05) by breed or diet. Plasma glucagon concentrations increased (P<0.05) with increasing CP intake (72.4, 167.6 and 239.1 pg/ml for L, M and H, respectively). The study indicated that there was no apparent breed difference between Nubian, Alpine and Angora goats in nitrogen utilization when goats were fed pelleted diets containing 9 to 21% CP.  相似文献

Due to global climatic changes, water and soil salinization is an increasing worldwide phenomenon, thus creating new threats for farm animal production. The present study was designed to investigate the adaptation capacity of goats towards sodium chloride (NaCl) in drinking water. Twelve non-pregnant Boer goats with an average body weight of 50.5 ± 9.0 kg were kept in individual pens. The study was conducted in four phases applying a two-choice preference test. In the control phase (phase 1) only fresh water was supplied in two containers. In phase 2, water with different salt concentrations (0.25%, 0.5%, 0.75%, 1.0%, 1.25% and 1.5%) was offered in one container and tap water in the other (sensitivity test). During the third phase (adaptation), goats were stepwise habituated to saline water by offering only saline water with different increasing concentrations (between 0% and 1.5% NaCl) in both containers. Subsequently, in phase 4 (sensitivity re-test) the same treatment as in phase 2 was repeated. Goats had ad libitum access to hay, water and a mineral licking block. Individual water and feed intake were recorded daily, while body weight and body condition score were measured every 2nd week. Body weight was not affected by saline water intake, whereas dry matter intake and body condition scores decreased significantly during the experiment. Water intake was significantly (P<0.001) higher in phase 2 (sensitivity test) and phase 3 (adaptation), compared to phase 1 (control) and phase 4 (sensitivity re-test). Total sodium intake followed the same pattern. In phase 2, when goats had the choice between fresh and saline water for the first time they preferred higher salt concentrations and consumed significantly (P<0.001) higher amounts of saline water (75.4 ± 53.2 g/kg BW0.82 per day) than in the re-test (40.4 ± 34.0 g/kg BW0.82 per day) after the habituation period. Thus, salt discrimination rejection thresholds were lowered to 1.25% in phase 4 compared to 1.5% in phase 2. The results suggest that a stepwise adaptation to saline drinking water in goats is an effective method to habituate the animals to saline water intake when concentrations were below 1.5%. Goats reacted more sensitively to the salinity of drinking water after prolonged exposure to saline water indicating flexible regulation mechanisms depending on the total sodium balance of the animal.  相似文献

This study investigated if lactating and nonlactating rats presented differences in relation to hepatic sensitivity to HgCl2 and the potential preventive role of ZnCl2. Lactating (days 3–12 of lactation) and nonlactating rats received 27 mg/kg ZnCl2 for five consecutive days and 5 mg/kg HgCl2 for five subsequent days. Lactating and nonlactating rats exposed to HgCl2 presented a decrease in food intake, a decrease in plasma alanine aminotransferase (ALT), and an increase in hepatic Hg levels when compared to the control group. Only lactating rats exposed to HgCl2 presented an increase in hepatic δ-aminolevulinic acid dehydratase activity. On the other hand, only nonlactating rats exposed to HgCl2 presented an increase in plasma aspartate aminotransferase (AST). ZnCl2 pre-exposure partially protected the increase in plasma AST activity presented by nonlactating rats and potentiated the liver Hg accumulation in lactating rats. Pups from the Sal–Hg and Zn–Hg groups showed a decrease in absolute liver weight and an increase in liver Hg levels. Summarizing, this study demonstrated that lactating rats presented distinct biochemical responses compared to nonlactating rats exposed to HgCl2 when hepatic parameters were evaluated.  相似文献

Salinization of groundwater and soil is a prevalent global issue with serious consequences on animal health and production. The present study was conducted to investigate the capacity of Boer goats to adjust their salt intake from saline drinking water in a free-choice system. In total, 12 non-pregnant Boer goats aged between 1 and 8 years with an average BW of 46.4±8.3 kg were kept in individual pens for 4 weeks. In the control phase (1 week), only fresh water was supplied in five identical buckets for each pen. During the subsequent treatment phase (3 weeks), fresh water and four different concentrations (0.75, 1.0, 1.25 and 1.5% NaCl) of saline water were offered simultaneously in a free-choice system. The positions of the concentrations were changed daily at random. Cut hay and water were provided ad libitum, and a mineral supplement was allocated. Feed and water intake, mineral supplement intake, ambient temperature and relative humidity were recorded daily, whereas BW and body condition score were measured weekly. Dry matter intake, total water intake and total sodium intake were significantly (P<0.001) higher during the treatment phase. Body weight and body condition were not affected by saline water intake. Across the treatment phase, saline water consumption was significantly (P<0.001) lower in young (19.6±27.1 g/kg BW0.82 per day) than in adult goats (27.9±31.5 g/kg BW0.82 per day), indicating that young goats were more sensitive towards the saline water. All goats had a significant preference for fresh water (0% salt) over saline water. At the first offering of the simultaneous choice situation (week 2), animals did not differentiate between the salt concentration of 0.75% and 1.0%. However, with successive treatment (weeks 3 and 4), animals distinguished between saline water concentrations and preferred the 0.75% salt concentration. Salt concentrations of 1% to 1.5% were avoided. The total sodium intake of the goats ranged between 0.37 and 0.55 g /kg BW0.75 per day during the treatment phase, being 8- to 11-fold higher than the daily requirements of sodium for body maintenance. The results suggest that goats are able to differentiate between saline water concentrations and to adjust their sodium intake by quick adjustments in self-selection in a free-choice system. Compared with two-choice preference tests, the present free-choice situation allows evaluating changes in saline water acceptance with prolonged exposure.  相似文献

The effects of an intraruminal load of 3 litres of water on body water movements was compared in Sardi sheep during the last month of pregnancy, lactation and a non-pregnant, non-lactating control period. Before the water load, rumen fluid volume, estimated by polyethylene glycol was similar in pregnant, compared to control, animals and 27% higher in lactating sheep. After the water load, rumen volume returned to pre-hydration level in 1 h during pregnancy, after 3 h during lactation and in the control period. Rumen osmolality decreased by 40% and remained at this low level for 3 h after the water load in all physiological periods. When the water load was tritiated water (TOH), the rate of TOH transfer into plasma was faster during the last month of pregnancy than during the control period. Plasma osmolality and proteins decreased in response to the water load. No differences in these responses were observed between pregnancy, lactation and the control period. Water diuresis began in the first 30 min following the water load in pregnant ewes and in the second 30 min in lactating and control ewes. The diuresis was also more pronounced in pregnant, than in non-pregnant, states. These results indicate that water is more rapidly absorbed from the gastrointestinal tract in pregnant, rather than in non-pregnant, sheep. This may partly explain the increased water turnover seen during pregnancy.  相似文献

During roosting in summer, reproductive female bats appear to use torpor less frequently and at higher body temperatures (T b) than male bats, ostensibly to maximise offspring growth. To test whether field observations result from differences in thermal physiology or behavioural thermoregulation during roosting, we measured the thermoregulatory response and energetics of captive pregnant and lactating female and male long-eared bats (Nyctophilus geoffroyi 8.9 g and N. gouldi 11.5 g) during overnight exposure to a constant ambient temperature (T a) of 15°C. Bats were captured 1–1.5 h after sunset and measurements began at 21:22±0:36 h. All N. geoffroyi entered torpor commencing at 23:47±01:01 h. For N. gouldi, 10/10 males, 9/10 pregnant females and 7/8 lactating females entered torpor commencing at 01:10±01:40 h. The minimum T b of torpid bats was 15.6±1.1°C and torpid metabolic rate (TMR) was reduced to 0.05±0.02 ml O2 g−1 h−1. Sex or reproductive condition of either species did not affect the timing of entry into torpor (F=1.5, df=2, 19, P=0.24), minimum TMR (F=0.21, df=4, 40, P=0.93) or minimum T b (F=0.76, df=5, 41, P=0.58). Moreover, sex or reproductive condition did not affect the allometric relationship between minimum resting metabolic rate and body mass (F=1.1, df=4, 37, P=0.37). Our study shows that under identical thermal conditions, thermal physiology of pregnant and lactating female and male bats are indistinguishable. This suggests that the observed reluctance by reproductive females to enter torpor in the field is predominantly because of ecological rather than physiological differences, which reflect the fact that females roost gregariously whereas male bats typically roost solitarily.  相似文献

Three lactating and three non-lactating black Bedouin goats were subjected to four days of water deprivation or to hemorrhage. Four days of water deprivation caused body wt losses of 32 and 23% and plasma volume losses of 30 and 34% in lactating and non-lactating goats respectively. Plasma osmolality increased 17 and 15% in lactating and non-lactating goats. Plasma arginine vasopressin concentration rose from about 5 pg/ml to a mean of 36 pg/ml. Plasma renin activity increased from about 0.7 ng/ml/hr to a mean of 3.45 ng/ml/hr in lactating and to 3.15 ng/ml/hr in non-lactating goats. At 4.5 hr post-rehydration plasma osmolality and plasma vasopressin concentration were back to normal in non-lactating, but still elevated in lactating goats. Plasma renin activity increased after rehydration. Rapid blood volume loss of 21-28% increased plasma vasopressin concentration to 16-35 pg/ml in non-lactating and to 70 or greater than 500 pg/ml in lactating goats. It is concluded that black Bedouin goats are well adapted to endure severe dehydration and rapid rehydration, but that they (especially lactating animals) react strongly to rapid volume depletion.  相似文献

Onymacris plana, a tenebrionid beetle from the sand dunes of the Namib desert, lost weight very slowly during 12 days of dehydration at 26°C. Measurement of total lipid showed a gradual decline, the metabolic water produced being sufficient to maintain a constant water content. At the same time the haemolymph volume decreased by 66%. When given water the dehydrated beetles drank rapidly and their weight and haemolymph volume were restored to normal. Haemolymph osmolarity was closely regulated despite the changes in volume. Haemolymph potassium was also well regulated, but sodium was lost from the haemolymph during a cycle of dehydration and rehydration, even though sodium losses in the faeces were small. Water balance in Onymacris depends on efficient conservation of water in periods of drought and on water uptake by drinking during the coastal fogs of the Namib.  相似文献

To examine the role of cortisol in seawater osmoregulation in a euryhaline teleost, adult killifish were acclimated to brackish water (10 per thousand) and RU486 or vehicle was administered orally in peanut oil daily for five days at low (40 mg.kg(-1)) or high dose (200 mg.kg(-1)). Fish were transferred to 1.5 x seawater (45 per thousand) or to brackish water (control) and sampled at 24 h and 48 h after transfer, when Cl- secretion is upregulated. At 24 h, opercular membrane Cl- secretion rate, as Isc, was increased only in the high dose RU486 group. Stimulation of membranes by 3-isobutyl-1-methylxanthine and cAMP increased Isc in vehicle treated controls but those from RU486-treated animals were unchanged and membranes from brackish water animals showed a decrease in Isc. At 48 h, Isc increased and transepithelial resistance decreased in vehicle and RU486 groups, compared to brackish water controls. Plasma cortisol increased in all groups transferred to high salinity, compared to brackish water controls. RU486 treated animals had higher cortisol levels compared to vehicle controls. Vehicle treated controls had lower cortisol levels than untreated or RU486 treated animals, higher stimulation of Isc, and lower hematocrit at 24 h, beneficial effects attributed to increased caloric intake from the peanut oil vehicle. Chloride cell density was significantly increased in the high dose RU486 group at 48 hours, yet Isc was unchanged, suggesting a decrease in Cl- secretion per cell. Thus cortisol enhances NaCl secretion capacity in chloride cells, likely via glucocorticoid type receptors.  相似文献

Atlantic cod (0.8–2.5 kg) were transferred froni 8°C to 1°C seawater for 17 days. No fish died. Exposure to 1°C water produced no changes in hematocrit, or in plasma concentrations of Cl or Mg2+. Cold water exposure caused a marked increase in plasma cortisol and glucose concentrations. Fish in 1°C seawater had higher gill Na-K-ATPase activity than fish in 8°C seawater, whereas there were no differences in gill lipid class or fatty acid composition.  相似文献

Obese subjects may be more vulnerable to injury from heat stress, and appear to be less efficient at thermoregulation. Sweat rate, tympanic temperature and osmolality in obese subjects were investigated in Japan during two seasons. The purpose of this study was to examine the relationship between obesity, thermoregulatory response and season. Five obese (BMI, 32.0?±?4.9 kg/m2) and five non-obese (BMI, 23.2?±?2.9 kg/m2) men participated in this experiment at latitude 35°10′ N and longitude 136°57.9′E. The average atmospheric temperature was 29.1?±?1.0 °C in summer and 3.3?±?1.4 °C in winter. Tympanic temperature and sweat rate were measured during leg water immersion at 42 °C for 30 min. Blood samples were analyzed for plasma osmolality. The relationship between tympanic temperature and sweat rate decreased significantly in obese compared to in non-obese subjects in both seasons, there being a lowered sweat rate for any core temperature in obese subjects. Plasma osmolality was significantly higher in obese than in non-obese subjects in both seasons. Thermal sensation increased significantly in non-obese than in obese in winter but not in summer. Our data show that thermoregulatory responses are attenuated in obese subjects compared with controls, suggesting that obese people are at increased risk of heat-related illnesses.  相似文献

Individual drinking patterns are a potential tool for disease monitoring in pigs. However, to date, individual pig drinking behaviour has not been described, and effects of external factors have not been examined. The aim of this study was to perform detailed quantification of drinking behaviour of growing pigs and to examine effects of period of day and effects of competition for access to the drinking nipple on the drinking behaviour, amount of water used and water wastage. In all, 52 cross-bred castrated male pigs (live weight 20.5±1.7 kg; mean±s.d.) maintained as either 3 (N3) or 10 (N10) pigs per pen and water nipple (four groups/treatment) were used. All pigs were fitted with a transponder ear tag. A radio frequency identification reader recorded and time stamped visits at the nipple. In each pen, water flow was logged every second. The drinking behaviour was recorded for 4 consecutive days and analysed using a linear mixed model. Overall, the pigs spent 594 s at the nipple during 24 h distributed among 44 visits. During this period, 5 l of water were used, of which >30% was wasted. Social competition did not affect the drinking behaviour over 24 h, except for the proportion of interrupted visits where pigs, kept with recommended nipple availability (N10), showed an increased proportion of interrupted drinking bouts compared with pigs kept at very low level of competition (N3) (0.18±0.01 v. 0.11±0.01; P<0.01). However, splitting data into 8-h periods (P1, P2, P3) starting from 0600 h revealed differences between treatments, showing that in N3, water use per visit was lower in P1 than P2 and P3 (110±10 v. 126±7 and 132±7 ml; P<0.05), whereas in N10, the water used per visit was higher during P3 than during the other periods (P1: 107±14 ml, P2: 112±10 ml v. P3: 151±10 ml; P<0.001). A similar pattern was found for visit duration. In N3, fewer nipple visits were observed in P2 than P1 (15.6±1.2 v. 22.0±1.2; P<0.001), whereas no difference was found between P1 and P2 in N10. The results demonstrate that growing pigs at the two levels of competition maintained a comparable level of 24 h water intake by changing behavioural variables involved in drinking. This dynamic characteristic of drinking behaviour means that if individual drinking patterns are to be used as disease monitoring tools, it is important to consider effects of external factors and include data on period level to allow rapid detection of behavioural changes.  相似文献

This study investigated the effect of water restriction on wool and blood cortisol concentrations and water consumption patterns in heat-stressed sheep. Nine Corriedale female sheep (average BW=43±6.5 kg) were individually fed diets based on maintenance requirement in metabolic crates. They were assigned to three treatments according to a Latin square design (3×3) for three periods with a 21-day duration for each period (nine sheep per treatment). Treatments included free access to water (FAW), 2 h water restriction (2hWR) and 3 h water restriction (3hWR) after feeding. Average temperature–humidity index in the experimental room was 27.9 throughout the experiment that defines heat stress conditions. Wool samples were taken at the end of each period on day 21. No differences were found in cortisol concentration in each fragment (dried, washed and residual extract) of wool (P<0.05). Total wool cortisol concentration was higher in the 3hWR group than the other treatments (P<0.05). Blood cortisol was not different among the treatments (P>0.05) and resulted in higher variable data compared with wool cortisol. Blood neutrophils and neutrophil/lymphocyte ratio suppressed in FAW and 3hWR groups compared with the 2hWR group (P<0.05). The duration of water consumption recorded after feeding in the 3hWR group was higher than in the 2hWR group when recorded in the afternoon (P<0.01). Water consumption rate was higher in the 3hWR group than in the 2hWR group (P<0.01). However, total water consumed was lower in the 3hWR group compared with other treatments (P>0.05). It can be concluded that wool cortisol provides more precise and accurate data than blood cortisol during heat stress conditions. Water restriction for 3 h after feeding can act as a stressor and is critical for sheep during heat stress as the consumption of water decreases with restriction.  相似文献
