Karyotype, sex chromosome system and cytogenetics characteristics of an unidentified species of the genus Apareiodon originating from Piquiri River (Paraná State, Brazil) were investigated using differential staining techniques (C-banding and Ag-staining) and fluorescent in situ hybridization (FISH) with 5S and 18S rDNA probes. The diploid chromosome number was 2n = 54 with 25 pairs of meta- (m) to submetacentric (sm) and 2 pairs of subtelocentric (st) chromosomes. The major ribosomal rDNA sites as revealed by Ag-staining and FISH with 18S rDNA probe were found in distal region of longer arm of st chromosome pair 26, while minor 5S sites were observed in the interstitial sites on chromosome pairs 2 (smaller cluster) and 7 (larger one). The C-positive heterochromatin had pericentromeric and telomeric distribution. The heteromorphic sex chromosome system consisted of male ZZ (pair 21) and female middle-sized m/st Z/W chromosomes. The pericentric inversion of heterochromatinized short arm of ancestral Z followed by multiplication of heterochromatin segments is hypothesized for origin of W chromosome. The observed karyotype and chromosomal markers corresponded to those found in other species of the genus.  相似文献

Sixteen species of Jaborosa (Solanaceae), including eight endemic to Argentina, were studied karyologically. The numbers n  = 12 and/or 2n  = 24 were found in all species, the majority of the cases being new reports. Mitotic chromosomes were small- to medium-sized, the average length varying from 2.95 to 4.93 µm. All species had one to three chromosome pairs with satellites. The karyotypes, obtained for 13 species, were slightly asymmetrical: A 1 ranged from 0.228 to 0.483, A 2 ranged from 0.095 to 0.182, and Paszko's asymmetry index ranged from 0.84 to 3.47. In all species, metacentric chromosomes were the most common, followed by submetacentrics, but subtelocentrics were rare. Morphological similarities and sectional arrangements were not reflected in either a principal components analysis plot or asymmetry index plot, but the species could be singled out by their karyotype formulae and the different karyotype parameters taken. In Jaborosa , a notably diversified genus, exo-morphological evolution has taken place, together with evident chromosome rearrangements, whose disposition is different and not as clear as in related genera.  © 2008 The Linnean Society of London, Botanical Journal of the Linnean Society , 2008, 156 , 467–478.  相似文献

Until recently, Sellocharis paradoxa Taubert, the only species of this Genisteae genus, was known solely by the isotypes. Recent new collections in Rio Grande do Sul, southern Brazil, have enabled data on chromosome number and karyotype morphology to be obtained for the first time. S. paradoxa has 2 n  = 20 chromosomes, and a bimodal asymmetrical karyotype, composed of one pair of long ( c . 6.3 µm) metacentric, five pairs of shorter acrocentric, and four pairs of shorter telocentric chromosomes ranging from c . 3.7 to 2.7 µm. The chromosome number and karyotype morphology of S. paradoxa do not fit into the Genisteae pattern. Existing information is so far insufficient to answer evolutionary questions about its origin and phylogenetic relationships, but the uniqueness of this taxon, first indicated by its peculiar leaf arrangement, and now supported by its uncommon karyological constitution, strongly suggests that the suprageneric taxonomic position of S. paradoxa should be re-evaluated.  © 2007 The Linnean Society of London, Botanical Journal of the Linnean Society , 2007, 155 , 223–226.  相似文献

Chromosome numbers and measurements were recorded in 47 individuals of ten taxa of Primula , representing eight species and two subspecies from ten populations. The basic numbers of chromosomes were x  = 8, 9 or 11, and they were mostly metacentric, medium-long to medium-small, ranging in length from c . 3.6 µm to 1.6 µm. Diploid chromosome numbers of 2 n  = 2 x  = 24 and 22 were scored for the first time in P. loeseneri Kitag. and P. prenantha Balf. & Sm., respectively. A new ploidy level of 2 n  = 4 x  = 44 was found in P. burmanica Balf. & King. The recorded diploid numbers of the remaining species confirmed earlier reports. Interspecific variability in chromosome numbers was correlated with heterogeneity in their mean length. Comparison of the data with those in the literature revealed that the observed variability of chromosomal characters was compatible with the other taxonomic criteria and supports the current taxonomic delimitation. Chromosomal variation at the diploid level is the predominant feature in Primula evolution, and allopatric speciation has played a major role in its specific diversity. Subgen. Aleuritia could represent the main clade from which the other subgenera have evolved. The present paper reports the chromosome numbers and karyotypes of eight species of Sect. Rhiziridium in Allium (Liaceae). The materials were all collected from their natural populations in east Inner Mongolia, China. The karyotype analysis is made on the basis of Li et al. (1985).The results are as follows (for chromosomes parameters, voucher specimens and localities, see Table 1 and Plate 1--2 the idiograms of the eight species in Fig. 1): (1) Auium leucocephalum Turcz. The somatic chromosome number and karyotype of this species is 2n=16=12m=2sm+2st (2SAT), in Stebbinsl(1971) kayotype classification, which belongs to 2A (Plate 1: 1; Fig. 1: 1). The range of chromosome relative length varies between 8.90--15.55%. Two small satellites are attached to the short arms of the 8th pair of chromosomes. (2) A. strictum Schrader has 2n (4x) =32=16m+4sm+12st, belonging to 2B (Plate 1: 2 & Fig. 1: 2). Satellites were not observed., and the range of chromosome relative length is between 3. 67-11.00%. (3) A. ramosum L. 2n=16=14m+ 2st (2SAT), belonging to 2A (Plate 1: 3 & Fig. 1: 3), Two small satellies are attached to the short arms of the 8th pair of chromosomes. The range of chromosome relative length is between 9.17-16.39%. The chromosome number and karyotype of this species are in accordancewith those reported by Li et al. (1982) with the material from Jinshan, Beijing. (4) A. bidentatum Fisch. ex Prokh. 2n (4x) =32=24m+4sm+4T, belonging to 2B (Plate 1: 4 & Fig. 1: 4). Satellites were not observed. A small median B-chromosome was found in root-tip cells of the population growing in sandy soil, and it is the first discovery (Plate 2: 9). The species has terminal chromosomes, which are seldom seen in Sect. Rhiziridium. The range of chromosome relative length is between 3.32—9.06%. (5) A. tenuissimu L. 2n=16= 10m+4sm+2st(2SAT), belonging to 2B(Plate 1:5 & Fig. 1:5). Two large satellites are attached to the short arms of the 8th pair of chromosome. The range of chromosome relative length is between 8.27--17.56%. (6)A. anisopodium Ledeb. 2n = 16 = l2m +2sm + 2st (2SAT), belonging to 2A (Plate 2:7 & Fig. 1: 7). Two large satellites are attached to the short arms of the 8th pair of chromosomes. In somatic cells of some plants of this species, a small submedian B-chromosome was found (Plate 2: 10, 11). The range of chromosome relative length is between 8.05-17.08 %. (7) A. anisopodium Ledeb. var. zimmermannianum (Gilg) Wang et Tang 2n (4x)=32=24m+4sm+4st( 4SAT), belonging to 2A (Plate 1: 6 & Fig. 1: 6). Four large satellites are attached to the short arms of the 15 and 16th pairs of chromosomes. The range of chromosome relative length is between 4.45--8.35%. This variety is similar to A. anisopodium Ledeb. in morphological characters, and their karyotype formulas are also very similar. The present authors consider that the variety is an allotetraploid derived from A. anisopodium Ledeb. (8) A. condensatum Turcz. 2n=16=14m+2st (2SAT), belonging to 2B (Plate 2:8 & Fig. 1:8). Two. small satellites are attached to the short arms of the 6th pair of chromosomes. In a few individuals of this species median (M) B-chromosome was discovered, and the number is stable (Plate 2: 12). The range of chromosome relative length is between 7.64--17.07%. In short, the chromosome numbers of the species studied in the present work are found to be 2n=16 or 32, and the karyotypes belong to 2A or 2B, highly symmetrical. The karyotypes of Chinese materials of these species are mostly reported for the first time. Threespecies have B-chromosomes.  相似文献

The chromosome numbers and karyotypes of 7 species of Smilax L. in Liliaceae (s. 1.) are cytotaxonomically studied in this work. Their karyotypic characters, distinction between the species and the chromosomal basis of sexual differentiation are discussed. The karyotypes of most species are first reported. The results are shown as follows (see Tables 1-4 for the chromosome parameters and the karyotype constitution; Fig. 1 for their idiograms): 1. Smilax nipponica Miq. The species is one of the herbaceous species distributed in East Asia. Two karyotypes, 2n = 26(type A) and 2n = 32 (type B), are found in the species (Plate 1: 1-7). The karyotype of No. 88032 (uncertain of -L--M--S- sexuality) is 2n = 26 = 2m + 6st + 6m + 4sm + 6sm + 2st. The karyotype has 4 pairs of L chromosomes, of which the first three pairs are subterminal, and the 4th is median. The karyotype belongs to 3B. No. 88045 (the male) and No. 88046 (the female) have 2n = 32. Their karyotypes are basically uniform, and both are -L--M-- S 2n=32= 2m+4sm+ 2st+ 2m+4sm+ 6m+ 10sm + 2st, also with 4 pairs of L chromosomes, but the 2nd pair is median, and thus different from the type A. The karyotype belongs to 3B. The first pair of chromosomes of the male are distinctly unequal in length, with the D. V. (0.93) of relative length between them obviously greater than that of the female (0.1). The pair seems to be of sex-chromosomes. Sixteen bivalents (n= 16) were observed at PMCs MI of No. 88045 (Plate 1: 4). The major difference between the karyotypes A and B are greater relative length of L chromosomes in the type A than in the type B, and the increase of chromosome number in the karyotype B mainly due to the increase of st chromosomes. Nakajima (1937)reports 2n= 30 for S. hederacea var. nipponica (=S. nipponica, Wang and Tang, 1980). 2. S. riparia A. DC. This species is also herbaceous, distributed in East Asia. Thirty chromosomes were found in root-tip cells (uncertain of sexuality). The kar -L--M--S-yotype is 2n = 30 = 8st + 6sm + 2st + 6m + 6sm + 2st (Plate 3: 1, 5), consisting mainly of sm and st chromosomes. There are 4 pairs of L chromosomes which are all subterminal and the m chromosomes appear to fall all into S category. Though the karyotype belongs to 3B, it is less symmetrical than that of S. nipponica. The species is karyologically rather different from S. nipponica, therefore. The first pair of chromosomes of this material are unequal in length, and it may be a male. The karyotype of this species is first reported. 3. S. sieboldii Miq. The species is a thorny climbing shrub, distributed in East Asia. At PMCs All, 16 chromosomes (n= 16) were found (Plate 2: 6), in accordance with Nakajima's (1933) report for a Japanese material. 4. S. china L. This species, a thorny climbing shrub, is of a wide distribution range mainly in East Asia and Southeast Asia. Two karyotypes were observed in different populations. (1) The population from Xikou has 2n = 96(6x) = 20st+L- -M- 6t + 6sm + 12st + 52(S) (Plate 3:7), of which the first three pairs of chromosomes are terminal, different from those in the other species. The arm ratios of both L and M chromosomes are larger than 2.0, which resembles those of S. davidiana. (2) PMCs MI of the population from Shangyu shew 15 chromosomes (n 15). The hexaploid of the species is recorded for the first time. Hsu (1967,1971) reported 2n = 30 from Taiwai and Nakajima (1937) recorded n = 30 from Japan, which indicates that the karyotype of the species varies not only in ploidy, but also in number. 5. S. davidiana A. DC. The somatic cells were found to have 32 chromosomes, and PMCs MI shew 16 bivalents (Plate 2: 1-5). The karyotype is 2n = 32=-L- -M- -S 8st + 4sm + 4st + 8sm + 8st. The karyotype belongs to 3B, and is less symmetrical than those in herbaceous species. The D. V. (0.20) of relative length between the two homologues of the first pair is slightly larger in the male than in the female (0.14), and it is thus difficult to determine whether they are sexual chromosomes or not. 6. S. glabra Roxb. The species is a non-thorny climbing shrub, distributed in East Asia and Southeast Asia. 32 chromosomes were found in somatic cells. The -L- -M- - Skaryotype is 2n= 32= 8st + 10st+6sm+8st (Plate 3: 2, 6),with only 3 pairs of sm chromosomes (12, 13 and 16th). The karyotype is more asymmetric than that of S. davidiana, although it is also of 3B (Table 1). The karyotype is first reported for the species. 7. S. nervo-marginata Hay. var. liukiuensis (Hay.) Wang et Tang The variety has a relatively narrow distribution range, mainly occurring in eastern China. The chromosomal number of somatic cells is 2n= 32 (Plate 3: 3-4). The karyotype is -L- -M- -S 2n = 32 = 2sm + 6st + 2sm + 2st + 2m + 6sm + 12st, evidently different from that of S. glabra. The first pair of chromosomes are submedian, and much longer than the 2nd to 4th pairs. The ratio in length of the largest chromosome to the smallest one is 4.3. The symmetric degree is of 3C, a unique type. The karyotype of the species is reported for the first time. In Smilax, the known basic numbers are 13, 15, 16 and 17. The two herbaceous species distributed in East Asia have three basic numbers: 13, 15 and 16, while the woody species studied mainly have 16, with no 13 recorded. Mangaly (1968) studied 8 herbaceous species in North America and reported 2n=26 for them except S. pseudo-china with 2n=30. Mangaly considered that a probably ancestral home of Smilax, both the herbaceous and woody, is in Southeast Asia and the eastern Himalayas, and speculated that the ancestral type of Sect. Coprosmanthus is possibly an Asian species, S. riparia. The karyotypes of the two herbaceous species in East Asia consist mostly of sm and m chromosomes, whereas those for the North American species are all of st chromosomes. Based on the general rule of karyotypic evolution, i.e. from symmetry to asymmetry, his speculation seems reasonable. Researches on sex-chromosomes of Smilax have been carried out since 1930 (Lindsay, 1930; Jensen, 1937; Nakajima, 1937; Mangaly, 1968), and they are generally considered to be the largest pair, but there is still no adequate evidence. The result of our observation on S. nipponica may confirm that the first pair of chromosomes of this species is XY type of sex-chromosomes. Chromosomes of the genus are small and medium-sized, varying between 1-6 μm, slightly larger in herbaceous species than in woody ones, larger in the karyotype of 2n=26 than in that of 2n=32. Based on karyotype constitution of the above 5 species, the karyotype in the genus is characterized by 4 pairs of L chromosomes and 2-5 pairs of M chromosomes, and mostly st and sm chromosomes, and by rather asymmetrical 3B type. The degree of symmetry in the above 5 species is from Sect. Coprosmanthus to Sect. Coilanthus, and herbaceous species towoody ones.  相似文献

The karyotype of the Mediterranean species Anemone hortensis L. (Ranunculaceae) was characterized with emphasis on heterochromatin distribution and localization of ribosomal (18S−5.8S−26S and 5S rDNA) and telomeric repeats (TTTAGGG). Diploid chromosome complement, 2 n  = 2 x  = 16, common to all investigated populations, consisted of three acrocentric, one meta-submetacentric and four metacentric chromosomes ranging in size from 6.34 to 10.47 µm. Fluorescence in situ hybridization (FISH) with 18S and 5S rDNA probes revealed two 18S−5.8S−26S rDNA loci on a satellite and secondary constriction of acrocentric chromosome pair 2 and terminally on acrocentric chromosome pair 3, and two 5S rDNA loci in the pericentromeric region of meta-submetacentric chromosome pair 4 and in the proximity of the 18S−5.8S−26S rDNA locus on chromosome pair 2. The only GC-rich heterochromatin, as revealed by fluorochrome Chromomycin A3 staining, was that associated with nucleolar organizer regions, whereas AT-rich heterochromatin, stained with 4,6-diamino-2-phenylindole (DAPI), was distributed intercalarly and terminally on the long arm of all three acrocentric chromosomes, and terminally on chromosomes 4 and 5. FISH with Arabidopsis -type telomeric repeats (TTTAGGG) as a probe revealed two classes of signals, small dot-like and large bands, at chromosome termini exclusively, where they corresponded to terminal DAPI-stained heterochromatin. Heteromorphism of chromosome pair 4, which refers to terminal DAPI bands and FISH signals, was observed in populations of Anemone hortensis . In Myrtaceae (Myrteae), the diploid chromosome number 2 n  = 2 x  = 22 is the most common, although variations of ploidy level occur, with some triploid (2 n  = 3 x  = 33) and tetraploid (2 n  = 4 x  = 44) records. Karyotype details in this group are scarce because the chromosomes are small (< 2 μm). In this work, we carried out a karyotypic analysis of 15 species of Myrtaceae grouped in different subtribes and genera. Measurements of chromosome length (long arm, L ; short arm, S ) were taken and several karyotypic parameters were calculated for each species. 以45S r DNA和拟南芥型端粒序列为探针对慈姑(Sagittaria trifolia L.)有丝分裂中期染色体进行单色和双色荧光原位杂交分析,并用银染方法检测慈姑45S r DNA位点的表达,最后结合染色体测量数据和45S r DNA杂交信号建立慈姑的核型。结果显示,慈姑的单倍基因组总长度为76.9±1.38μm,最长染色体为11.55±0.10μm,最短染色体为4.54±0.27μm;慈姑的核型公式为:2n=22=2m+2sm+14st+4t,核型不对称性参数CI、A1、A2、As K(%)、AI分别为19.86±11.06、0.72、0.27、78.82、15.29,核型属于Stebbins类型中的3B型。慈姑具有3对45S r DNA位点,分别位于第8、9、10号染色体的短臂末端。拟南芥型端粒序列的杂交信号出现在慈姑每一条染色体的长、短臂末端。银染检测到6个Ag-NOR和6个核仁,表明3对45S r DNA位点在间期核都有表达。本研究结果为药食兼用植物慈姑提供了分子细胞遗传学基础资料。  相似文献

The chromosome number and morphology of eight species belonging to the section Santolinoideae of the genus Achillea L. (Asteraceae) were investigated using karyological techniques. The species studied were Achillea cucullata (Hausskn.) Bornm., A. vermicularis Trin., A. monocephala Boiss. & Bal., A. boissieri (Hausskn.) Bornm., A. santolina L., A. gypsicola Hub.-Mor., A. goniocephala Boiss. & Bal., and A. spinulifolia Fenzl ex Boiss. In these species, the chromosome numbers were 2 n  = 18 in A. vermicularis , A. monocephala , and A. boissieri , 2 n  = 18A + 1B in A. spinulifolia , 2 n  = 36 in A. cucullata and A. santolina , and 2 n  = 54 in A. gypsicola and A. goniocephala . All of the chromosomes had median point (M), median region (m), or submedian (sm) centromeres. Archolaemus, one of the five genera of Neotropical freshwater fish of the family Sternopygidae (Gymnotiformes), was long considered a monotypic genus represented by Archolaemus blax. Currently, it consists of six species, most of them occurring in the Amazon region. There are no cytogenetic data for species of this genus. In the present study, we used classical cytogenetics (conventional staining and C‐banding) and molecular cytogenetics (probes of telomeric sequences and multigenic families 18S rDNA, 5S rDNA, and U2 snDNA) to study the karyotype of Archolaemus janeae from Xingu and Tapajós rivers in the state of Pará (Brazil). The results showed that the two populations have identical karyotypes with 46 chromosomes: four submetacentric and 42 acrocentric (2n = 46; 4m/sm + 42a). Constitutive heterochromatin occurs in the centromeric region of all chromosomes, in addition to small bands in the interstitial and distal regions of some pairs. The 18S rDNA occurs in the distal region of the short arm of pair 2; the 5S rDNA occurs in five chromosome pairs; and the U2 snDNA sequence occurs in chromosome pairs 3, 6, and 13. No interstitial telomeric sequence was observed. These results show karyotypic similarity between the studied populations suggesting the existence of a single species and are of great importance as a reference for future cytotaxonomic studies of the genus.  相似文献

The present study provides specific cytogenetic information on prometaphase chromosomes of the alpaca (Lama pacos, fam. Camelidae, 2n = 74) that forms a basis for future work on karyotype standardization and gene mapping of the species, as well as for comparative studies and future genetic improvement programs within the family Camelidae. Based on the centromeric index (CI) measurements, alpaca chromosomes have been classified into four groups: group A, subtelocentrics, from pair 1 to 10; group B, telocentrics, from pair 11 to 20; group C, submetacentrics, from pair 21 to 29; group D, metacentrics, from pair 30 to 36 plus sex chromosomes. For each chromosome pair, the following data are provided: relative chromosome length, centromeric index, conventional Giemsa staining, sequential QFQ/C-banding, GTG- and RBG-banding patterns with corresponding ideograms, RBA-banding and sequential RBA/silver staining for NOR localization. The overall number of RBG-bands revealed was 391. Nucleolus organizer-bearing chromosomes were identified as pairs 6, 28, 31, 32, 33 and 34. Comparative ZOO-FISH analysis with camel (Camelus dromedarius) X and Y painting probes was also carried out to validate X-Y chromosome identification of alpaca and to confirm close homologies between the sex chromosomes of these two species.  相似文献

In this paper 7 species of wildflowers were collected from Beijing suburb and Jilin Province. They are all common sightly and hardy perennials in their localities (See the Appendix for detail of the materials). The micrographs of their somatic metaphases are shown in Plate 1; the karyotype formulae, ranges of chromosome length and classification of karyotypes according to Stebbins (1971) are shown in Table 1; the idiograms of 5 species in Figs. 1-5. The karyotype analysis is made on the basis of Li and Chen (1985)(1). The essential points are as follows; (1) Ten pairs of chromosomes of Achyrophorus ciliatus are all submetacentric (sm). (2) Twelve pairs of chromosomes are all metacentric (m), and the short arms of the seventh pair of chromosomes with a pair of satellites in Orychophragmus violaceus. (3) The seventh and nineth pairs of chromosomes are sm and the short arms of latter with satellites in Silene repens var. angustifolia. It is reported for the first time. (4) In Scabiosa tschiliensis. the first, fourth, fifth and eighth pairs of chromosomes are sm, the sixth is terminal (t). The second and seventh are subterminal (st), the third is m. There are satellites on the short arms of third and seventh pairs. It is reported for the first time. (5) The eleventh pair of chromosomes is sm and the others are all m. The short arms of the twelfth pairs with satellites in Lychnis fulgens. (6) The chromosome number (2n) is 42, with a pair of satellites in Papaver pseudo-radicatum. It is also reported for the first time. (7) The chromosome number is2n=56 with two pairs of satellites in Rehmannia glutinosa.  相似文献

Cytogenetic characteristics of the Caspian Pond Turtle, Mauremys caspica, in Golestan and Mazandaran provinces in the northern part of Iran show that the chromosome number is 2n?=?52 and the arm number NF?=?78. The karyotype consisted of 9 metacentric (M), 1 submetacentric (SM), 3 subtelocentric (ST) and 13 telocentric (T) chromosome pairs. The Centromeric Index ranges from 11.79 to 45.68, the arm ratio between 1.18 and 7.47, the relative length between 1.60 and 11.46, and the length variation between 1.05 and 7.48. Average total length of the chromosomes is 65.27 µm. The largest chromosome in this species is a pair of the metacentric chromosome. Location of NOR was determined on chromosome pair no. 10.  相似文献

Eleocharis subarticulata is recorded as the third species of Cyperaceae with a reduced chromosome number ( n  = 3), following reports on Rhynchospora tenuis ( n  = 2) and Fimbristylis umbellaris ( n  = 3). For Eleocharis, the numbers recorded to date vary from 2 n  = 10 to 2 n  = c. 196, with x  = 5 as the possible basic number. The karyotype of E. subarticulata was studied using conventional staining (mitosis and meiosis), C-CMA3/DAPI banding, and FISH with 45S rDNA and telomere probes. The chromosomes showed no primary constrictions, as expected in the holocentric chromosomes of Cyperaceae. The meiotic behaviour was abnormal, with a single multivalent ring of six chromosomes at metaphase I, resulting from multiple translocations. At anaphase I six chromatids migrated to each pole, evidencing the inverted meiosis, and these groups were also visible at metaphase II. The C-CMA3/DAPI banding technique showed only four terminal GC-rich blocks. FISH with 45S rDNA probes revealed four terminal signals, probably associated with GC-rich blocks. The telomeric probe located terminal signals in all the chromosomes, besides a hybridization site in the middle of the large pair. The occurrence of ectopic telomeric sites has not been described previously for plants with holokinetic karyotypes and with reduced chromosome numbers. Mitotic chromosomes of eight populations of six species of Vernonia sensu Baker (Asteraceae, Vernonieae, section Lepidaploa , group Axilliflorae ) were studied to assess the validity of maintaining the genus as proposed currently ( sensu Baker) or of splitting it into several smaller genera ( sensu Robinson). The species were collected in areas of 'cerrado' and 'campo rupestre' in southern and central-western Brazil. The chromosome numbers were 2 n  = 20–40 and karyotypic analysis showed a predominance of metacentric chromosomes, with some submetacentrics. B chromosomes were seen in one population of V. geminata . The chromosomes varied in length from 0.9 to 4.6 µm, with a total chromatin length of 29.7–45.0 µm and a symmetry index of 41.2–46.9%. The intrachromosomal asymmetry index varied from 0.13 to 0.29, and the interchromosomal asymmetry index varied from 0.15 to 0.21. The dispersion diagram showed that population 1 of V. geminata had the most asymmetrical karyotype. These results do not provide sufficient evidence for a conclusive cytotaxonomic discussion of Vernonia , mainly because of the impossibility of associating any infrageneric group with specific karyotypic characters, and also because of the small number of species analysed so far.  © 2007 The Linnean Society of London. In order to analyze chromosome numbers and karyotypes of intestinal trematodes belonging to the genus, Metagonimus, the gonad tissues of M. takahashii, M. miyatai, and M. yokogawai were prepared and examined. The number of bivalents in the first meiotic division of M. takahashii was nine (n = 9). The diploid number of M. miyatai was observed to be eighteen (2n = 18) and their chromosomes consisted of one pair of metacentric, 7 pairs of submetacentric, and one pair of telocentric chromosomes. The diploid number of M. yokogawai was thirty-two (2n = 32) and the chromosome complements were composed of two pairs of metacentric, 11 pairs of submetacentric, and three pairs of subtelocentric chromosomes. These results could be a supporting evidence for the validity of the new species, M. miyatai, distinct from M. yokogawai.  相似文献

With the help of Computer Aided Karyotyping procedures, Ag-NOR staining and C-banding techniques, the karyotype of Pyrrhopappus carolinianus (Asteraceae, Lactuceae) has been studied. The species has 2n=12 chromosomes. Silver staining reveals that the two shortest pairs of chromosomes possess NOR's. On the basis of chromosome length and centromere position, only the longest chromosome pair and the satellite chromosomes can be identified. Two types of C-banding can be obtained, dependent on the temperature of the hydrochloric acid hydrolysis of the root tips. Hydrolysis at 60°C results exclusively in centromeric bands, whereas a treatment at room temperature reveals a pattern of intercalary bands. A computer assisted analysis of the intercalary banding pattern resulted in the construction of schematic representation of the average C-banding pattern. This banding pattern allows an easy identification of each of the chromosome pairs.  相似文献

Two populations of Sphaerium corneum were sampled from River Vilnelė and small pond in Vilnius, Lithuania. The chromosomes were studied using conventional Giemsa staining and karyometric analysis. Inter- and intra-individual variation in the diploid chromosome numbers was revealed and two different sources of chromosome variability were identified: B chromosomes and the structural changes of chromosomes of the basic (A) set. The chromosome set of the more common karyotypic form, 2 n  = 30, found in both populations, consists of all biarmed metacentric and meta-submetacentric chromosomes of gradually decreasing size. Small, biarmed, mitotically unstable B chromosomes were found in the cells of this karyotypic form. Specimens with 2 n  = 36 were found only in pond. No B chromosomes were detected in their cells. The karyotype is characterized by presence of two pairs of medium telocentrics and four pairs of small subtelocentrics. The remaining chromosomes are biarmed. Robertsonian fusions appear to be involved in formation of two karyotypic forms of S. corneum . DNA sequence analyses showed that ITS1 is identical in both karyotypic forms. On the other hand, differences in 16S sequence were revealed and two haplotypes, corresponding to two karyotypic forms, were identified. The present study opens new perspectives in establishing species-specific characters for confident identification of Sphaerium species and provides insights to the genetic intraspecific variability and possible mechanisms of speciation.  © 2006 The Linnean Society of London, Biological Journal of the Linnean Society , 2006, 89 , 53–64.  相似文献
