SUMMARY.
  • 1 The origin of vertical temperature stratification in Erstwhile Water (Lake District, Cumbria, England) was observed.
  • 2 At the first stage of the spring warming of the whole water column some discontinuity in the vertical temperature distribution appears in the surface and bottom layers.
  • 3 Persistent summer stratification occurred for the first time at the lee end of the lake immediately after a change of wind direction, when the cumulative potential energy deficit dominated the cumulative kinetic energy by about 110-8 Jcm-2.
  • 4 The results show the importance of the proper choice of initial conditions in computations of total energy, as well as the necessity to consider the spatial distribution of potential and mechanical energy responsible for the summer stratification in the lake.

The effects of fumonisin on development of Syrian hamster fetuses were studied using fumonisin B1 and B2 extracted fromFusarium moniliforme corn-culture and purified fumonisin B1. A significant increase in litters with fetal deaths occurred with the high doses of purified (18 mg FB1/kg) and culture-extracted (18 mg FB1 plus 4.5 mg FB2) fumonisin. It is concluded that prenatal exposure to fumonisin on days 8 and 9 of gestation is detrimental to fetal hamster survivability but does not induce clinical maternal intoxication at these doses. Equivalent doses of fumonisin B1, whether from culture-extract or pure solution produced similar results.  相似文献

Coccolith malformations occur more frequently in cultured specimens than in specimens from natural samples, a phenomenon commonly termed ‘culture artefacts’. The causes of culture artefacts are unknown. Here, we tested the effect of culture flask shape, mixing, and cell density on the morphology of Emiliania huxleyi coccoliths. While there was no effect of different culture flask types typically used in coccolithophore culturing, continuous mixing reduced the percentage of malformations by ca. 11 % in exponential-phase cells (cell density ca. 80 × 103 cells per ml) and ca. 17 % in stationary-phase cells (cell density ca. 2 × 106 cells per ml). Stationary-phase cells displayed 19 % more malformations than mid-exponential-phase cells when not mixed at all and 20 % more malformations when continuously mixed. It is concluded that the lack of mixing and unnaturally high cell densities, typical for coccolithophore stock cultures, are partly responsible for culture artefacts.  相似文献

Arthropod cuticles observed in section generally present alternating clear and dark bands. These have often been interpreted in terms of superimposed layers of different structure or composition. It has been shown, however that this material is homogeneous, and is formed by a twisted arrangement of microfibrils. The dense bands correspond actually to a microtomy artefact and they form dark single spirals in certain distorted areas of the cuticle. A model was proposed, involving the interaction between knife motion and microfibrils; it will be referred to as the stepped model, since the proposed mechanism results in the formation of steps at the surface of sections, on both faces. These steps are limited by structures resembling crests or cliffs, whose regular distribution produces alternating thick and thin bands in the section. This explains the observed contrast (Bouligand, (1972)). Two very interesting models were proposed later (Gordon and Winfree, (1978)) and are referred to as the cos F model and the sand model, but steps and crests are absent in these models. However, Giraud-Guille ((1986)), has shown very clearly the existence of these crests, which seem to be quite essential in this microtomy artefact. To clarify the debate, the texts defining the initial and the two new models are reproduced here and the difficulties encountered by each model are discussed. A mathematical formulation of this artefact is presented in an appendix to the present article; this leads to a more complete discussion of the possible models. Other factors are also taken into consideration: microfibril orientation and staining. The main factor of contrasts is undoubtedly the variation in thickness over a single section, as proposed in the stepped model.  相似文献

Developmental and toxic effects of aqueous extracts of F. moniliforme culture material containing known levels of fumonisin B1 were recently reported in mice and included maternal hepatotoxicity and lethality, maternal body weight gain reduction, increased embryonic resorptions, reduced offspring body weights, and fetal malformations including cleft palate, hydrocephalus, malformed ribs and incomplete digital and sternal ossification. These studies also suggested that the effects of the fungal extract on the mouse offspring may be mediated via maternal effects. The contribution of fumonisin B1 (FB1), a major toxic metabolite of F moniliforme, in the induction of these effects was evaluated in this study by administering 0 to 100 mg pure FB1/kg of body weight on gestational days (GD) 7 through 15 to pregnant Charles River CD1 mice and assessing maternal health and fetal development till the end of gestation. Doses of 25 mg/kg or higher of pure FB1 induced maternal liver lesions (mostly necrotic changes), associated with ascites and increased hepatocytic nuclear diameter. Fumonisin doses of 50 mg/kg or higher also resulted in significantly increased maternal ALT on GD12, and reduced offspring bodyweights on GD 18. Increased resorptions and decreased numbers of live offspring were only evident at 100 mg FB1/kg body weight. Offspring exhibited dose-dependent increase in the incidence and severity of hydrocephalus of both the lateral and third ventricles at doses of 25 mg/kg or higher. Doses of 25 mg/kg or higher also increased the sphinganine/sphingosine (Sa/So) ratios in maternal but not fetal livers. These results suggest that FB1 may be a developmental toxicant accounting for most but not all earlier reported effects of F. moniliforme culture extract. Association of FB1 effects on the offspring with maternal hepatoxicity and with alteration of Sa/So ratio in maternal but not fetal liver supported the earlier claim that FB1 effects on the mouse offspring are mediated by maternal hepatotoxicity.  相似文献

Using monoclonal anti-fumonisin B1 antibody (anti-FB1) and avidin-biotin-peroxidase system, liver and kidneys of broiler chicks were evaluated for the detection and distribution of fumonisins (FBs). One hundred and fifty micrograms of FB1 or culture extract of Fusarium moniliforme str. 113F containing 150 microg of FB1 and 4 microg of FB2 were administered into the vitelline sac of 1-day old, specific pathogen-free chicks. The animals were killed 24 h after injection, and renal and hepatic tissues submitted for immunohistochemical analysis. FBs were detected in the epithelial cells of convoluted distal and proximal tubules of the kidneys, as well as in the cytoplasm of hepatocytes. This novel immunohistochemical method developed is expected to be an efficient way for monitoring the target of the FB toxins in tissues.  相似文献

Summary The precipitation patterns of the following ultracytochemical methods in rat muscle cells were compared and examined critically: the potassium pyroantimonate method for calcium demonstration; the calcium phosphate technique for the Ca2+ — ATPase reaction; the formazan reaction for the demonstration of creatine kinase activity (all performed on heart muscle); and the lead phosphate technique for the Mg2+ — ATPase reaction in skeletal muscle. Using X-ray microanalysis, it was found that the antimonate precipitate contains only calcium as the precipitated ion in the vast majority of cases. Most probably it consists of pure calcium pyroantimonate. However, in myocytes showing the well-established precipitation pattern, the concentration of calcium was estimated to be about two orders of magnitude higher than the native concentration of total intracellular calcium. It is concluded that calcium ions diffuse freely from the extracellular space and from adjacent cells into cells containing antimonate and are precipitated mostly at sites where heterogeneous nucleation is facilitated by intracellular catalysts (biopolymers).As shown by the similar precipitation patterns for the four reactions compared, these catalysts are not specific to any of these reactions and are most probably neither calcium-binding sites nor sites of any one of the enzymes examined in the native cell.  相似文献

Fumonisin B1, a fungal mycotoxin that grows on corn and other agricultural products, alters sphingolipid metabolism by inhibiting ceramide synthase. The precise mechanism of fumonisin B1 toxicity has not been completely elucidated; however, a central feature in the cytotoxicity is alteration of sphingolipid metabolism through interruption of de novo ceramide synthesis. An affinity column consisting of fumonisin B1 covalently bound to an HPLC column matrix was used to isolate a rat liver protein that consistently bound to the column. The protein was identified as argininosuccinate synthetase by protein sequencing. The enzyme-catalyzed formation of argininosuccinic acid from citrulline and aspartate by recombinant human and rat liver argininosuccinate synthetase was inhibited by fumonisin B1. Fumonisin B1 showed mixed inhibition against citrulline, aspartate, and ATP to the enzyme. Fumonisin B1 had a Ki' of approximately 6 mM with the recombinant human argininosuccinate synthase and a Ki' of 35 mM with a crude preparation of enzyme prepared from rat liver. Neither tricarballylic acid nor hydrolyzed fumonisin B1 inhibited recombinant human argininosuccinate synthetase. This is the first demonstration of fumonisin B1 inhibition of argininosuccinate synthethase, a urea cycle enzyme, which adds to the list of enzymes that are inhibited in vitro by fumonisin B1 (ceramide synthase, protein serine/threonine phosphatase). The extent of the inhibition of argininosuccinate synthetase in cells, and the possible role of this enzyme inhibition in the cellular toxicity of FB1, remains to be established.  相似文献

Cordia nodosa Lamark (Boraginaceae) is a myrmecophyte (i.e., plants housing ants in hollow structures) that provisions associated ants with food bodies (FBs) produced 24 h a day. Distributed over all the young parts of the plants, they induce ants to forage continually and so to protect the plants. Metabolites are stored in the inner cells of C. nodosa FBs as they form. In addition the peripheral cells have an extrafloral nectary-like function and secrete a substance that covers the FBs. The amalgam of these two functions, distinct in other known cases, is discussed taking into account the origin of FBs and extrafloral nectaries.  相似文献

Landraces of maize (Zea mays ssp. mays) and its wild teosinte relatives (Zea mays spp. parviglumis and mexicana) were surveyed for sensitivity to fumonisin B(1), a phytotoxin produced by the maize pathogen Gibberella moniliformis. Only two of 42 Z. mays samples were highly insensitive to FB(1) (ED(50) = ca. 200 microM). The teosintes and 76% of the maize landraces were moderately or highly sensitive to FB(1) (ED(50) < or = 30 microM), which indicates that FB(1) sensitivity is likely to be an ancestral trait in Z. mays. F(1) generations derived from crosses between FB(1)-sensitive maize inbred B73 and insensitive landraces were significantly less sensitive than B73. Thus, our data indicate that FB(1)-insensitivity is a relatively rare but heritable trait in maize. We also report the sensitivity of maize to other Gibberella toxins - beauvericin, diacetoxyscirpenol, and moniliformin.  相似文献

Fumonisins B1 (FB1) and B2 (FB2), two structurally related mycotoxins with cancer-promoting activity, were recently isolated from corn cultures of Fusarium moniliforme MRC 826. These toxins have been reported to be produced also by isolates of F. proliferatum. Contamination of foods and feeds by F. moniliforme has been associated with human esophageal cancer risk, and FB1 has been shown to be the causative agent of the neurotoxic disease leukoencephalomalacia in horses. Because of the toxicological importance of the fumonisins, the potential to produce FB1 and FB2 was determined in a study of 40 toxic Fusarium isolates representing 27 taxa in 9 of the 12 sections of Fusarium, as well as two recently described species not yet classified into sections. With the exception of one isolate of F. nygamai, fumonisin production was restricted to isolates of F. moniliforme and F. proliferatum, in the section Liseola. The F. nygamai isolate produced 605 micrograms of FB1 g-1 and 530 micrograms of FB2 g-1, and the identity of the toxins was confirmed by capillary gas chromatography-mass spectrometry. This is the first report of the production of the fumonisins by F. nygamai.  相似文献

(Tsel)、热耐受性和体温对食物同化的影响。Tsel无显著的日时间变化,两性个体的Tsel、临界低温(CTMin)和临界高温(CTMax)无显著的差异。Tsel、CTMin和CTMax的平均值分别为30.9℃、3.2℃和43.3℃。体温对动物食物通过时间有显著的影响。在25℃-33℃范围内,食物通过时间随体温升高而缩短;体温超过33℃后,食物通过时间随体温升高而延长。温度对原尾蜥虎的摄食量、表观消化系数(ADC)和同化效率(AE)有显著的影响。在25℃-37℃范围内,动物在低体温下(25℃和27℃)的摄食量、ADC和AE小于更高体温下动物的对应数值。种间比较结果显示,原尾蜥虎是生活于南方的蜥蜴中具有较强耐受极端体温的种类。  相似文献
