Summary Acetylene was reduced to ethylene by effective white clover nodules and by fully and partially effective intact nodules, nodule homogenates, and bacteroids of soybeans. Succinate and several amino acids markedly stimulated the reduction by effective soybean bacteroids, but the stimulation was slight with partially effective bacteroids. Acetylene metabolism by effective soybean bacteroids was also enhanced by excretions of in vitro-grown Rhizobium japonicum, excretions of bacteria derived from effective and ineffective nodules, and the soluble fraction from these nodules. Inhibitors of nitrogen fixation were not found in ineffective nodules. Ineffective soybean and white clover nodules and homogenates or isolated bacteria from ineffective soybean nodules did not reduce acetylene. Additions of succinate, amino acids, the soluble fraction of effective nodules, or excretions of effective bacteroids or of in vitro-grown cells of an effective R. japonicum strain did not promote nitrogen fixation by bacterial cells obtained from ineffective soybean nodules.  相似文献

We examined the effect of drought stress on proline accumulation,nitrogenase activity and activities of enzymes related to prolinemetabolism in soybean (Glycine max [L.] Merr.) nodules. Nitrogenase(C2H2 reducing) activity was inhibited 90% or more as a resultof drought stress. This inhibition was substantially reversedafter a 4 h recovery period. Pyrroline-5-carboxylate reductaseactivity in extracts of drought-stressed nodules from 25-d-oldplants was 55% higher than in unstressed nodules, but the sameactivity in preparations from 55-d-old plants was similar tothat of control plants. Extracts of recovering nodules on plantsof both ages had activities near those of controls. Droughtstress increased the activity of the pentose phosphate pathwayby about 65% in extracts of nodules from 55-d-old plants, butthere was no effect in extracts of nodules from younger plants(25-d-old). Proline dehydrogenase activity was 3.7 and 1.6 timeshigher in bacteroids isolated from nodules taken from 25- and55-d-old stressed plants, respectively, than in comparable controlbacteroids. This activity remained high in bacteroids from bothsets of recovering nodules. The amount of proline in extractsfrom stressed nodules was 3- to 4-fold higher than in unstressednodules, despite increased proline dehydrogenase activity andremained high in nodules collected 4 h after rewatering. Thisincrease was observed in both cytoplasmic and bacteroid fractions.The possible physiological significance of these results isdiscussed. Two different types of nitrogen-fixing root nodules are known — actinorhizal nodules induced byFrankia and legume nodules induced by rhizobia. While legume nodules show a stem-like structure with peripheral vascular bundles, actinorhizal nodule lobes resemble modified lateral roots with a central vascular bundle. To compare carbon metabolism in legume and actinorhizal nodules, sucrose synthase and enolase cDNA clones were isolated from a cDNA library, obtained from actinorhizal nodules ofAlnus glutinosa. The expression of the corresponding genes was markedly enhanced in nodules compared to roots. In situ hybridization showed that, in nodules, both sucrose synthase and enolase were expressed at high levels in the infected cortical cells as well as in the pericycle of the central vascular bundle of a nodule lobe. Legume sucrose synthase expression was studied in indeterminate nodules from pea and determinate nodules fromPhaseolus vulgaris by usingin situ hybridization.  相似文献

Antiserum was prepared in rabbits against purified alfalfa (Medicago sativa L.) nodule phosphoenolpyruvate carboxylase (PEPC). Immunotitration assays revealed that the antiserum recognized the enzyme from alfalfa nodules, uninoculated alfalfa roots, and from soybean nodules. Tandem-crossed immunoelectrophoresis showed that the PEPC protein from alfalfa roots and nodules was immunologically indistinguishable. The 101 kilodalton polypeptide subunit of alfalfa nodule PEPC was identified on Western blots. The PEPC polypeptide was detected in low quantities in young alfalfa roots and nodules but was present at increased levels in mature nodules. Senescent nodules appeared to contain a reduced amount of the PEPC polypeptide. PEPC was also detected by western blot in some plant- and bacterially-conditioned ineffective alfalfa nodules but was not detected in bacteroids isolated from effective nodules. Alfalfa nodule PEPC is constitutively expressed in low levels in roots. In nodules, expression of PEPC polypeptide increases several-fold, resulting in increased PEPC activity. Antiserum prepared against the C4 PEPC from maize leaves recognized the PEPC enzyme in all legume nodules and roots tested, while the antiserum prepared against alfalfa nodule PEPC also recognized the leaf PEPC of several C4 plant species. Neither antiserum reacted strongly with any C3 leaf proteins. The molecular weight of the PEPC polypeptide from C4 leaves and legume nodules appears to be similar.  相似文献

Trehalose and trehalase in root nodules from various legumes   总被引:4,自引:1,他引:3  
Nitrogen-fixing (effective) nodules from various legume- Rhizobium combinations were analyzed for trehalose and other soluble carbohydrates using gas chromatography and for trehalase activity using biochemical assays. Whereas the bacterial disaccharide trehalose was present only in the minority of the nodules, trehalase activity was found in all of them. Extracts from determinate nodules had a higher trehalase activity than extracts from indeterminate nodules. More detailed studies were done on soybean nodules formed in interactions with two effective and 5 ineffective Bradyrhizobium japonicum strains. Only in effective soybean nodules colonized by the strain 61-A-101 was trehalose a major soluble carbohydrate. Irrespective of the wildtype strains used. effective soybean nodules contained about 10 nkat trehalase g−1 fresh weight, whereas the ineffective nodules colonized by mutant strains derived from these wildtype strains contained 2 to 30 times less trehalase. However, a clear correlation between trehalose content and trehalase activity could not be established.  相似文献

Characterization of the proteolytic compartment in rat hepatocyte nodules   总被引:1,自引:0,他引:1  
Persistent liver nodules (hepatocyte nodules, neoplastic nodules) were produced in rat liver by intermittent feeding with 2-acetylaminofluorene. Dense bodies (secondary lysosomes) were purified and characterized from the nodules. The purity of the dense body fraction was 90%. The levels of various lysosomal enzyme activities were lower in these dense bodies in comparison with dense bodies from control liver. Similarly, protein degradation was 50% lower in dense bodies from liver nodules than in control liver. The number of autophagic vacuoles (AVs) in the nodular tissue increased considerably after 3 h vinblastine treatment. We have taken advantage of this expansion in an effort to isolate these organelles from liver nodules. Autophagic vacuoles have been isolated recently from liver and kidney but not from putatively premalignant liver nodules. Fraction purity of AVs from liver nodules was 95%. As with dense bodies, AVs from nodular tissue displayed lower activities of proteinases and lower rates of protein degradation when compared with their counterparts from normal liver tissue. Accordingly, the lower rate of overall protein degradation in liver nodules can be ascribed to a decrease in lysosomal activity. A diminished autophagic sequestration capacity is the most plausible explanation for the decreased rate of proteolysis in cells. This could conceivably give these nodular cells a growth advantage and assist in their selective outgrowth as well as in their transformation from neoplastic into true cancer cells.  相似文献

本文报道大叶相思的结瘤固氮和氢酶活性的研究结果。大叶相思结瘤状况与其他含羞草亚科的树种相似,刚形成的幼瘤为单生球状或椭圆形,以后顶端伸长或分叉,呈分叉状、姜状和扇状。同一植株的不同成熟度根瘤的固氮活性也不同,成熟壮瘤固氮活性最高,幼瘤次之,衰老瘤最低。不同立地条件下种植的大叶相思根瘤固氮活性虽有差异,但生长在pH4.7的酸性红壤中的大叶相思,根瘤固氮活性仍具有较高水平。大叶相思根瘤固氮活性也有明显的季节变化,夏秋较高,春冬较低。根瘤离体后7小时内固氮活性变化不大,甚至在离体后21小时内仍维持一定水平的固氮活性。大叶相思根瘤具有吸氢酶,其吸H_2活性在最初3小时内随时间延长而升高,3—7小时内仍维持一定水平。在根瘤固氮系统中注入外源分子H_2,可提高固氮酶活性,外源H_2最适浓度为7.5%。由于其根瘤具有催化吸收分子H_2的氢酶系统,能吸收利用固氮反应所放出的大量H_2,因而能更有效地利用光合产物于固氮过程。大叶相思根瘤离体后能较长时间维持高的固氮活性水平,可能与其吸H_2酶系统有关。  相似文献

In red alder (Alnus rubra Bong.) the nitrogen fixing actinomycete Frankia can be divided into strains which form spores (sp+) within nodules and those that do not. Red alder nodules were collect from 42 sites in southwestern British Columbia and their spore type was determined. Sp+ nodules were found in 18 of the sites, where their proportion ranged from 4 to 93%. The distribution of sites with sp+ nodules was clumped, with significant autocorrelation at the 0–10 km interval. No sp+ nodules were found in any of the eight stands in the submaritime region, indicating a possible continentality effect on spore type distribution. There was a significant positive association between the presence of Rubus spectabilis and the presence sp+ nodules. Stand and soil chemical variables were generally poor predictors of the proportion of sp+ nodules, although total soil nitrogen and exchangeable phosphorus accounted for 36.4 and 28.5% respectively, of the variation in the proportion of sp+ nodules on sites where they were present. There was also a small but significant negative relation between soil pH and the proportion of sp+ nodules and young stands generally did not have any sp+ nodules.  相似文献

Dicarboxylic acid transport mutants of Rhizobium species are usually deficient in their ability to fix atmospheric dinitrogen. We report here a study comparing the physiology of root nodules on Phaseolus vulgaris L. cv. Goldie induced by an effective strain of Rhizobium leguminosarum biovar phaseoli and a C4-dicarboxylic acid utilization mutant. The mutant, while able to form nodules, was ineffective in N2 fixation. Carbohydrates and organic acids of roots and nodules formed by the 2 strains were monitored at 3-day intervals from 13 to 34 days after inoculation. Both carbohydrates and organic acids accumulated in ineffective nodules in comparison with the effective nodules. The concentration of malic acid was tenfold higher in ineffective nodules than in effective nodules. Other organic acids, i.e., lactate, malonate, ascorbate and gluconate, were also detected. Lactate and ascorbate were the only other organic acids accumulating in ineffective nodules. The most prevalent carbohydrates found in both types of nodules were sucrose, glucose and fructose. Myo-inositol was the only cyclitol detected in both types of nodules. Carbohydrates and organic acids were present in lower concentration in roots than in nodules, except for lactate. These compounds were not consistently detected in higher concentration in roots from plants inoculated with the mutant strain, as was the case in nodules.  相似文献

Root nodules were collected from the existing plantation of Casuarina equisetifolia from three different agroclimatic zones of Tamil Nadu, India. Morphological variation, histology, histochemistry and nutrient contents of the nodules were studied. The nodules were found to be simple, dichotomously branched and coralloid. The stele in simple and branched nodules reached up to the tip of the nodules. Structure of Frankia nodules was described using histochemical methods to elucidate the nature of micro-symbiont within the nodules. The nodule consisted of a distinct periderm enclosing the cortex. The cortex showed patches of infected cells interspersed among the uninfected ones. Inside the cortex was a stele bound by an endodermis, which contained high amount of tannin. The micro-symbiont was observed in the nodule tisues in hyphal form and spore packed sporocorp was also seen in the cortex. The histochemical tests revealed the presence of proteins, insoluble polysaccharides and polyphenols.  相似文献

Abstract Nodulation ability was tested for Frankia strains HFPCcI3 and ELI, and Frankia sources A.t. and G.a. from Allocasuarina torulosa and Gymnostoma australianum , respectively, on A. torulosa Miq., Casuarina cunninghamiana Miq., G. australianum L. Johnson and Elaeagnus triflora Roxb. It was shown that A. torulosa and C. cunninghamiana formed nodules only with the Frankia sources obtained from their own host plant, while E. triflora formed nodules with three of the four Frankia sources tested. All nodules formed were effectively fixing nitrogen. Specific nitrogenase activity was highest in E. triflora inoculated with the Frankia strain isolated from nodules of the same species. Identification of Frankia sources in the nodules was performed by use of PCR amplification of DNA with a random primer. PCR amplification of DNA isolated from nodules of G. australianum and E. triflora inoculated with Frankia strain EL1 revealed, when compared with DNA amplified from free living Frankia strain EL1, that there was only one Frankia strain causing the observed nodules.  相似文献

Ononitol (4-O-methyl-myo-inositol) was identified as a major carbohydrate in Pisum sativum nodules, comprising 25–34% of the total mono- plus disaccharides in nodules formed by two Rhizobium leguminosarum strains. Ononitol was purified from Glycine max nodules and was found to be a minor carbohydrate in these nodules. The distribution of ononitol in bacteroids and cytosol from soybean nodules suggests that it is not synthesized by bacteroids.  相似文献

Callus tissue cultures were developed from apical meristem regions of tumor-like ineffective root nodules of alfalfa. Callus growth was a function of tissue source and hormone composition and concentration. Callus derived from ineffective nodules also were shown not to contain Rhizobium meliloti.

Glutamate dehydrogenase, glutamine synthetase, glutamate synthase, glutamate oxaloacetate transaminase and phosphoenolpyruvate carboxylase activities were present in callus cultures and in the respective nodule source used for callus induction. The mean specific activity of all enzymes evaluated was higher in callus cultures than in ineffective nodules. Quantitative but not qualitative differences in enzyme activities were evident between ineffective nodules and callus derived from these nodules. Tissue cultures derived from ineffective nodules may provide a model system to evaluate host plant-Rhizobium interactions.

Uricase (nodulin-35) cDNA, LjUr, was isolated from nodules of a model legume, Lotus japonicus. LjUr expression was most abundant in nodules, although it was detected in nonsymbiotic tissues as well, particularly in roots. Expression in nodules was detected in uninfected cells, nodule parenchyma, and, more intensely, in vascular bundles. Phylogenetic analysis of uricase sequences from various legumes indicated that uricases of amide- and ureide-transporting legumes form two distinct clades. LjUr is in the cluster of amide-transport legumes even though L. japonicus bears determinate nodules.  相似文献

The malate dehydrogenase present in the cytoplasmic fraction of plant origin and bacteroids from yellow lupine root nodules was investigated. The plant enzyme was 14 times more active in nodules than in roots and it contained 6 molecular forms in nodules compared with 3 forms detected in roots. The highest malate dehydrogenase activity in plant fraction and bacteroids was noted in 50-day old plants. Changes in the isoenzymatic patterns of malate dehydrogenase in plant fraction and bacteroids accompanying ageing of the lupine root nodules were observed. Possible physiological role of malate pathway in metabolism of lupine root nodules is discussed.  相似文献

Summary Among plants native to Japan, nodule formation is confirmed in 14 species and varieties of Alnus, 10 of Elacagnus, 2 of Myrica and 1 of Coriaria, in a number of instances for the first time. Plants of 20 foreign species, in 8 genera, which bear nodules in their native countries, were raised in the nursery in Tokyo; only species of Alnus, Myrica and Ceanothus formed nodules. No nodules were found on native plants ofDryas octopetala var.asiatica. In a trial extending over 12 years evidence was obtained that the growth ofPinus thunbergii was benefited by underplanting withMyrica rubra, a result attributed to nitrogen fixation in the root nodules of the latter species. In the attempted isolation of the endophytes from the nodules of Alnus and other non-legume Angiosperms, although actinomycetes peculiar to the host species were usually obtained from the nodules, none of the isolates induced nodules in re-inoculation tests. Also from Podocarpus nodules actinomycetal and bacterial strains were commonly isolated; re-inoculation tests with these are in progress.  相似文献

目的:研究甲状腺结节良恶性的彩色多普勒超声特征及其诊断价值。方法:选取从2016年3月~2019年2月于我院接受手术治疗的甲状腺疾病患者300例作为研究对象,均予以彩超检查,比较甲状腺良恶性结节的超声特征(主要包括直径、钙化情况、边界、回声、血流状况)。以病理活检结果为金标准,分析彩超诊断甲状腺结节良恶性的敏感性、特异性以及准确度。对比甲状腺良恶性结节的血流分型情况以及各项血流动力学参数。结果:恶性结节超声特征直径≥2 cm、有钙化、边界模糊、无回声/低回声、血流丰富人数占比均高于良性结节(均P<0.05)。以手术病理组织活检结果作为金标准,彩色多普勒超声诊断甲状腺结节的敏感性、特异性以及准确度分别为97.73%、86.11%、96.33%。甲状腺良性结节血流分型为Ⅰ型、Ⅱ型人数占比高于恶性结节,而Ⅲ型、Ⅳ型人数占比低于恶性结节(均P<0.05)。甲状腺良性结节的收缩期峰值血流速度(PSV)、阻力指数(RI)均低于恶性结节,而舒张末期血流速度(EDV)高于恶性结节(均P<0.05)。结论:彩色多普勒超声对甲状腺结节良恶性的鉴别价值较高,且具有较高的敏感性、特异性以及准确度,可为甲状腺良恶性结节的早期诊断、临床治疗提供重要的参考依据。  相似文献
